植物结构蛋白质组学研究目的在不断变化。早期旨在比较不同基因型和株系,估测系统发育距离。随后是用蛋白N端Edman测序法或氨基酸分析法鉴定蛋白。如今,应用双向凝胶电泳和质谱法,已能联手成功地解决蛋白分离和鉴定问题。这将有助于对不同突变系与野生型作比较和对不同的表达基因鉴定,应用数据库和作图软件有可能获得功能蛋白构象。 相似文献

目的:对分化抑制蛋白(Id)家族的N端序列进行保守性结构分析,并构建其基因突变体。方法:用ClustalX(1.81)软件对Id蛋白家族中的3个成员(Id1~Id3)的N端序列进行同源性分析,用Swiss-PdbViewer3.7(SP5)软件模拟高同源区域中关键性氨基酸突变前后的三维结构模型;用PCR方法将突变点引入Id序列,再通过重叠PCR方法扩增出全长编码序列,酶切与测序确证突变序列的准确性。结果:Id1~Id3蛋白的N端存在一个由11个氨基酸残基形成的高度保守的环-螺旋(Loop-Helix)结构,将其中最保守的丝氨酸与亮氨酸分别突变为甘氨酸与缬氨酸,将突变后的Id基因序列重组到pGEX原核表达载体中。结论:在Id1~Id3蛋白N端识别了一个保守的Loop-Helix结构,为深入研究其协同的功能特征提供了结构依据;突变其中的丝氨酸为研究Id蛋白潜在的磷酸化修饰及相应功能特征的改变奠定了基础。 相似文献

氨基酸突变能够改变蛋白的结构和功能,影响生物体的生命过程.基于串联质谱的鸟枪法蛋白质组学是目前大规模研究蛋白质组学的主要方法,但是现有的质谱数据鉴定流程为了提高鉴定结果的灵敏度往往会有意压缩数据库中的氨基酸突变信息.因此,如何挖掘数据中的氨基酸突变信息成为当前质谱数据鉴定的一个重要部分.当前应用于氨基酸突变鉴定的串联质谱鉴定方法大致可以分为3大类:基于序列数据库搜索的方法、基于序列标签搜索的算法以及基于图谱库搜索的算法.本文首先详细介绍了这3种氨基酸突变鉴定算法,并分析了各种方法的特点和不足,然后介绍了氨基酸突变鉴定的研究现状和发展方向.随着基于串联质谱的蛋白质组学的不断发展,蛋白序列中的氨基酸突变信息将被更好地解析出来,从而得以深入探讨由氨基酸突变引起的蛋白结构和功能改变,为揭示氨基酸突变的生物学意义奠定基础. 相似文献

外源高表达PC-1蛋白N端43个氨基酸可加速人前列腺癌细胞系C4-2的生长 总被引:3,自引:0,他引:3
研究PC-1蛋白N端43个氨基酸表达对人前列腺癌细胞C4—2生长的影响。用DNA重组技术将含PC—1蛋白N端43个氨基酸的DNA序列正向克隆到真核表达载体pIRES2-EGFP中,采用脂质体法将重组质粒稳定转染进C4—2细胞中,RT—PCR分析外源序列的转录情况,固相ELISA法测定PC—1蛋白N端43个氨基酸的表达,MTT实验分析细胞的生长速度。结果获得了稳定转染PC—J基因N端43个氨基酸的前列腺癌细胞株,在该细胞株中外源PC—1蛋白N端43个氨基酸得到高表达,细胞生长速度较对照细胞加快了38%。结果表明外源PC—I基因N端43个氨基酸高表达可提高人前列腺癌细胞C4—2的生长速度,推论PC—J基因高表达可能在人前列腺癌的发展中起一定的促进作用。 相似文献

以H5N2亚型禽流感病毒毒株血凝素蛋白裂解位点碱性氨基酸为研究对象,对其密码子偏好性和对应mRNA序列的折叠二级结构特点进行研究和分析。旨在探讨裂解位点氨基酸对应mRNA核苷酸片段的二级结构与病毒致病力的关系,希望能对禽流感病毒的研究提供一些基础性信息。将mRNA样本按照序列等步长递增的方法,用RNAstructure 4.1程序预测这些样本的动态延伸折叠二级结构。序列和结构的分析结果:裂解位点的碱性氨基酸对富含腺嘌呤的密码子有强烈偏好;与碱性氨基酸对应的mRNA片段上的核苷酸主要位于折叠二级结构的单链环区,少数位于配对螺旋区。结果表明:裂解位点氨基酸对应的mRNA核苷酸形成发夹端环的大小与其碱性氨基酸的多少具有正相关性。 相似文献

利用PCR法 ,从人胎盘总cDNA中扩增得到Rab3acDNA的全编码区 .序列分析表明 ,扩增得到的Rab3acDNA有 5个核苷酸发生了变异 ,但翻译的氨基酸与发表的完全一致 .将扩增得到的Rab3acDNA克隆于原核融合表达载体pGEX 4T 1中 ,在E .coliBL2 1中经IPTG诱导表达 .为了进一步鉴定表达产物 ,对纯化后的Rab3a蛋白进行了SDS PAGE、N端氨基酸测序、质谱分子量测定及氨基酸组成分析鉴定 .结果显示 ,表达蛋白的分子量约 2 5kD ,N端氨基酸序列为MASATDSR ,氨基酸组成分析表明 ,Rab3a蛋白获得了正确表达 相似文献

Recent advances in the study of Kaposi's sarcoma-associated herpesvirus replication and pathogenesis
It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis. 相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera. 相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment. 相似文献
