【目的】利用N-甲基吡咯烷酮(N-methylpyrrolidone, NMP)作为电子供体进行反硝化实验,以实现废水资源化。【方法】分别将NMP废水和葡萄糖作为电子供体加入到模拟的城市污水处理尾水中进行反硝化,比较2种电子供体去除硝酸盐的规律。同时考查NMP在反硝化过程中的氮素释放规律,并对所释放的氮素进行后续处理。最后再对它们作为电子供体时的反硝化污泥采用高通量测序,从微生物群落的角度分析NMP作为电子供体时其作用机理是否相同。【结果】当以NMP为电子供体时,硝酸盐氮的去除速率比葡萄糖为电子供体时要快67%。在8 h的反硝化结束后,剩余的硝酸盐氮、累积的亚硝酸盐氮和NMP本身所释放氨氮之和的总氮,与葡萄糖为电子供体时相近。【结论】NMP废水可以作为电子供体用于城镇污水处理厂的深度脱氮。对2种碳源所驯化的反硝化污泥样品进行高通量分析表明,NMP与葡萄糖作为电子供体用于反硝化反应时,相关的作用机理是不同的。该项研究结果对利用含氮杂环化合物作为电子供体进行反硝化具有重要的理论指导意义。  相似文献

Biological denitrification typically requires the addition of a supplemental electron donor, which can add a significant operating expense to wastewater treatment facilities. Most common electron donors are organic, but reduced inorganic sulfur compounds (RISCs), such as sulfide (HS?) and elemental sulfur (S0), may be more cost-effective. S0 is an inexpensive and well characterized electron donor, but it provides slow denitrification rates due to its low solubility. A lesser-known RISC is sulfite (\({\text{SO}}_{3}^{2 - }\)), which can be easily produced from S0 by a simple combustion process. Unlike S0, \({\text{SO}}_{3}^{2 - }\) is highly soluble, and therefore may provide higher denitrification rates. However, very little is known about microbial denitrification with \({\text{SO}}_{3}^{2 - }\). Also, \({\text{SO}}_{3}^{2 - }\) is a strong reductant that reacts abiotically with oxygen and has toxic effects on microorganisms. This paper reviews \({\text{SO}}_{3}^{2 - }\) in the environment, \({\text{SO}}_{3}^{2 - }\) chemistry, microbiology, toxicity, and its potential use for denitrification. Since \({\text{SO}}_{3}^{2 - }\) is an intermediate in the sulfur oxidation pathway of most sulfur-oxidizing microorganisms, it is an energetic electron donor and it should select for a \({\text{SO}}_{3}^{2 - }\)-oxidizing community. Our review of the literature, as well as our own lab experience, suggests that \({\text{SO}}_{3}^{2 - }\) can effectively serve as an electron donor for denitrification. Further research is needed to determine the kinetics of \({\text{SO}}_{3}^{2 - }\)-based denitrification, its toxic threshold for sulfur-oxidizing microorganisms, and its potential inhibition of sensitive species such as nitrifying microorganisms and potential formation of nitrous oxide. Its effect on sludge settling efficiency also should be explored.  相似文献

The potential for PHB (poly-beta-hydroxybutyrate) to serve as the electron donor for effective simultaneous nitrification and denitrification (SND) was investigated in a 2-L sequencing batch reactor (SBR) using a mixed culture and acetate as the organic substrate. During the feast period (i.e., acetate present), heterotrophic respiration activity was high and nitrification was prevented due to the inability of nitrifying bacteria to compete with heterotrophs for oxygen. Once acetate was depleted the oxidation rate of PHB was up to 6 times slower than that of soluble acetate and nitrification could proceed due to the decreased competition for oxygen. The slow nature of PHB degradation meant that it was an effective substrate for SND, as it was oxidised at a similar rate to ammonium and was therefore available for SND throughout the entire aerobic period. The percentage of nitrogen removed via SND increased at lower DO concentrations during the famine period, with up to 78% SND achieved at a DO concentration of 0.5 mg L(-1). However, the increased percentage of SND at a low DO concentration was compromised by a 2-times slower rate of nitrogen removal. A moderate DO concentration of 1 mg L(-1) was optimal for both SND efficiency (61%) and rate (4.4 mmol N x Cmol x(-1) x h(-1)). Electron flux analysis showed that the period of highest SND activity occurred during the first hour of the aerobic famine period, when the specific oxygen uptake rate (SOUR) was highest. It is postulated that a high SOUR due to NH(4) (+) and PHB oxidation decreases oxygen penetration into the floc, creating larger zones for anoxic denitrification. The accumulation of nitrate towards the end of the SND period showed that SND was finally limited by the rate of denitrification. As PHB degradation was found to follow first-order kinetics (df(PHB)/dt = -0.19 x f(PHB)), higher PHB concentrations would be expected to drive SND faster by increasing the availability rate of reducing power and reducing penetration of oxygen into the floc, due to the corresponding increased SOUR. Process control techniques to accumulate higher internal PHB concentrations to improve PHB-driven SND are discussed.  相似文献

A novel denitrifying bacterium was isolated using bicarbonate as the sole carbon source in a defined medium. Strain W3 was isolated from deep sediments of East Lake (Wuhan, China). In this study, analysis of 16S rRNA genes showed that strain W3 was affiliated with Microbacterium sp. When using Fe2+ as the only electron donor, this strain could convert 88.6 % of NO3 −-N to N2, corresponding to an Fe2+ oxidation rate of 80 %. Meanwhile, neither NO2 −-N nor NH4 +-N was accumulated after the experiment. In similar experiments with Fe(II)-EDTA, cell encrustations did not occur and supplementary substrates were consumed. The accumulated NO2 −-N was below 2.5 mg L−1. In addition, PCR revealed five kinds of key denitrifying genes: narG, napA, nirS, norB and nosZ. These results indicated that strain W3 could be used as an alternative autotrophic denitrifier for the treatment of groundwater and low C/N ratio wastewater.  相似文献

The heavy use of fertilizers in agricultural lands can result in significant nitrate (NO3) loadings to the aquatic environment. We hypothesized that biological denitrification in agricultural ditches and streams could be enhanced by adding elemental sulfur (So) to the sediment layer, where it could act as a biofilm support and electron donor. Using a bench-scale stream mesocosm with a bed of So granules, we explored NO3 removal fluxes as a function of the effluent NO3 concentrations. With effluent NO3 ranging from 0.5 mg N L−1 to 4.1 mg N L−1, NO3 removal fluxes ranged from 228 mg N m−2 d−1 to 708 mg N m−2 d−1. This is as much as 100 times higher than for agricultural drainage streams. Sulfate (SO42−) production was high due to aerobic sulfur oxidation. Molecular studies demonstrated that the So amendment selected for Thiobacillus species, and that no special inoculum was required for establishing a So-based autotrophic denitrifying community. Modeling studies suggested that denitrification was diffusion limited, and advective flow through the bed would greatly enhance NO3 removal fluxes. Our results indicate that amendment with So is an effective means to stimulate denitrification in a stream environment. To minimize SO42− production, it may be better to place So deeper in the sediment layer.  相似文献

Spent sulfidic caustic was applied to sulfur utilizing autotrophic denitrification as the simultaneous source of electron donor and alkalinity. The two experiment set-up of upflow anoxic hybrid growth reactor (UAHGR) and upflow anoxic suspended growth reactor (UASGR) was adopted and nitrate removals were similar in both reactors. Approximately 90% of the initial nitrate was denitrified at nitrate loading rate of 0.15∼0.40 kgNO3 /m3·d. The experimental stoichiometric ratio of sulfate production to nitrate removal was ranged from 1.5 to 2.1 mgSO4 2−/mgNO3 . During the operation period, denaturing gradient gel electrophoresis (DGGE) analysis of polymerase chain reaction (PCR)-amplified 16S rDNA fragments for the sludge sample of both reactors showed the change of microbial communities. Thiobacillus denitrificans-like microorganism occupied 28.5% (18 clones) of the 63 clones by cloning the PCR products from the sludge sample of UAHGR. Acidovorax avenae, which can reduce nitrate to nitrogen gas while oxidizing phenol (heterotrophic denitrifier), was also found in 7 clones (11.1%). Although an organic carbon source was not added to the medium, a microorganism (Kaistella koreensis) capable of oxidizing organic compounds was found in 7 clones (11.1%). Therefore, the microbial community of spent sulfidic caustic applied autotrophic denitrification process well corresponds to the substrate components of spent sulfidic caustic. Through the batch cultivation of microorganisms in UAHGR, the microbial kinetic coefficients of spent sulfidic caustic applied autotrophic denitrification were estimated to be μ max = 0.097 h−1, k d = 0.0021 h−1, K s = 200 mgNO3 /L, and Y = 0.31 mgMLVSS/mgNO3 .  相似文献

AIMS: To develop a laboratory-scale autotrophic membrane-immobilized biofilm reactor to remove nitrogen from drinking water. METHODS AND RESULTS: A polyvinyl alcohol (PVA) immobilized biofilm, attached to the surface of a silicone tube, was used as the basis of a bioreactor for simultaneous nitrification and denitrification of water. The bioreactor was aerated with air to supply oxygen for nitrification. Pure hydrogen was supplied to the silicone tube and diffused through the membrane wall to feed the biofilm for autotrophic denitrification. The bioreactor was effective for the simultaneous nitrification and denitrification of water after a short period of acclimation, while the biofilm exhibited good resistance to the inhibition of denitrification by dissolved oxygen; the denitrification rate decreased by only 8% as the dissolved oxygen increased from 2 mg l(-1) to saturation. CONCLUSIONS: By using PVA crosslinked with sodium nitrate to entrap nitrifying and denitrifying sludge on the surface of a silicone tube, a novel bioreactor for simultaneous nitrification and denitrification was developed. In addition to performing as an immobilizing agent to strengthen the biofilm, PVA protected the denitrifying microorganisms to reduce the inhibition by dissolved oxygen under aerobic condition. Therefore, nitrification and denitrification occurred simultaneously within the biofilm. Furthermore, the immobilization technique shortened the acclimation period of the bioreactor. SIGNIFICANCE AND IMPACT OF THE STUDY: The described space saving and simple to operate bioreactor for nitrogen removal performed autotrophic denitrification to solve the problem of residual carbon in heterotrophic denitrification, and thus is suitable for removing nitrogen from drinking water.  相似文献

Summary The resting cells of methanogen strain HU could be used as biocatalyser for converting exoge nous NADP+ into NADPH, using either formate or hydrogen as electron donor. To enhance the conversion efficiency of NADPH from NADP+, several inhibitors of methylcoenzyme M reductase were used in order to avoid further oxidation of NADPH to CH4. When methyl viologen (7.5 mol ml-1) was added to the reaction mixture (17 mg of dry cells, 2 mg Triton X-100, 294 mol of Na-formate and 12 mol of NADP+ per ml reaction mixture), 9.6 mol ml-1 NADPH (80% yield) could be produced in a 2-h reaction, compared with 7.2 mol ml-1 NADPH (60% yield) in a 6-h reaction in the absence of methyl viologen. Molecular hydrogen istead of formate also served as electron donor to convert NADP+ into NADPH. A gas mixture of H2/N2 (75/25) yielded 9.8 mol ml-1 NADPH (82% yield) in a 3-h reaction in the absence of formate, suggesting that H2 might be a promising, inexpensive electron donor for this reaction system.  相似文献

To preserve organic materials in the sludge required for denitrification, a study was made with a contact process. The contactor, when operated with a short retention time (30 min), gave incomplete metabolism of organic matter. With 64% of the influent soluble COD (SCOD) was adsorbed to activated sludge within 30 min. The specific mass of organic matter uptaken was 55 mg SCOD/g Mixed Liquor Suspended Solids that enhanced the denitrification efficiency up to 63% in the following denitrification step.  相似文献

An activated sludge reactor fed with thiocyanate and/or thiosulfate was operated to examine the characteristics of its microbial community. Terminal-restriction fragment length polymorphism analyses were conducted to detect shifts in the microbial community structure corresponding to influent conditions. Then, clone library analyses and RNA-based stable-isotope probing were conducted to identify sulfur-oxidizing bacteria (SOB) responsible for the degradation of each substrate. The results suggested that there were two types of SOB: thiocyanate-degrading bacteria (that can utilize both thocyanate and thiosulfate) and thiosulfate-specific bacteria (that cannot utilize thiocyanate). Thiocyanate-degrading SOB, however, were outcompeted by thiosulfate-specific SOB when the influent contained only thiosulfate. Of the sequenced clones, Marinicella-related (with 98.7% identity) and Methylobacter-related (with 91.3% identity) bacteria were identified as thiocyanate-degrading SOB, whereas Thiomicrospira thermophila-related (with 100% identity over 903 bp) bacteria were identified as thiosulfate-specific SOB.  相似文献

Sulfur-driven autotrophic denitrification refers to the chemolithotrophic process coupling denitrification with the oxidation of reduced inorganic sulfur compounds. Ever since 1904, when Thiobacillus denitrificans was isolated, autotrophic denitrifiers and their uncultured close relatives have been continuously identified from highly diverse ecosystems including hydrothermal vents, deep sea redox transition zones, sediments, soils, inland soda lakes, etc. Currently, 14 valid described species within α-, β-, γ-, and ε-Proteobacteria have been identified as capable of autotrophic denitrification. Autotrophic denitrification is also widely applied in environmental engineering for the removal of sulfide and nitrate from different water environments. This review summarizes recent researches on autotrophic denitrification, highlighting its diversity, metabolic traits, and engineering applications.  相似文献

A challenge for photobiological production of hydrogen gas (H(2)) as a potential biofuel is to find suitable electron-donating feedstocks. Here, we examined the inorganic compound thiosulfate as a possible electron donor for nitrogenase-catalyzed H(2) production by the purple nonsulfur phototrophic bacterium (PNSB) Rhodopseudomonas palustris. Thiosulfate is an intermediate of microbial sulfur metabolism in nature and is also generated in industrial processes. We found that R. palustris grew photoautotrophically with thiosulfate and bicarbonate and produced H(2) when nitrogen gas was the sole nitrogen source (nitrogen-fixing conditions). In addition, illuminated nongrowing R. palustris cells converted about 80% of available electrons from thiosulfate to H(2). H(2) production with acetate and succinate as electron donors was less efficient (40 to 60%), partly because nongrowing cells excreted the intermediary metabolite α-ketoglutarate into the culture medium. The fixABCX operon (RPA4602 to RPA4605) encoding a predicted electron-transfer complex is necessary for growth using thiosulfate under nitrogen-fixing conditions and may serve as a point of engineering to control rates of H(2) production. The possibility to use thiosulfate expands the range of electron-donating compounds for H(2) production by PNSBs beyond biomass-based electron donors.  相似文献

1. The effect of ampicillin on cytochrome c reduction and on the superoxide production of human neutrophils stimulated by phorbol myristate acetate (PMA) was investigated. 2. Ampicillin did not stimulate the superoxide production of intact (resting) neutrophils and not amplify the superoxide production of neutrophils stimulated by phorbol myristate acetate (PMA). 3. However, ampicillin dose-dependently increased the reduction of cytochrome c. 4. In addition, 50 mM ampicillin stimulated a superoxide dismutase-inhibitable reduction of cytochrome c by 0.70 +/- 0.02 (mean +/- SD) nmol/min and a superoxide dismutase-noninhibitable reduction of cytochrome c by 2.08 +/- 0.03 (mean +/- SD) nmol/min. 5. These results suggest that ampicillin serves as an electron donor and/or a superoxide generator.  相似文献

A new method based on sulfide utilizing autotrophic denitrification was adopted to remove nitrate from wastewater and to reuse spent sulfidic caustic containing high sulfide and alkalinity levels. The experiments were performed using a bench-scale upflow anoxic hybrid growth reactor (UAHGR) and an upflow anoxic suspended growth reactor (UASGR) to characterize the stoichiometric relationship between sulfur and nitrate in the process as well as the performance of the reactors. The level of nitrate removal from the UAHGR and UASGR were maintained at over 90% at a nitrate loading rate ranging from 0.15∼0.40 kgNO3 /m3·d and no significant nitrite accumulation was observed in either reactor. Although the influent pH values were higher than the optimum range of autotrophic denitrification at 8.7∼10.1, the effluent pH was stable at 7.2∼7.9 due to the production of hydrogen ions during operation. The stoichiometric ratio of sulfate production to nitrate removal was 1.5∼2.1 mgSO4 2−/mgNO3 in both reactors. A comparison of the reactor performance revealed that the chemical parameters of the UAHGR operation corresponded to a plug flow like type reactor while the chemical parameters of the UASGR operation corresponded to a completely stirred tank reactor like type reactor. UAHGR did not require sludge recycling due to the packed media while UASGR required 300∼700% sludge recycling. Therefore, spent sulfidic caustic could be used in the sulfur utilizing autotrophic denitrification processes as substrate and alkalinity sources.  相似文献

Sulfur-utilizing autotrophic denitrification relies on an inorganic carbon source to reduce the nitrate by producing sulfuric acid as an end product and can be used for the treatment of wastewaters containing high levels of nitrates. In this study, sulfur-denitrifying bacteria were used in anoxic batch tests with sulfur as the electron donor and nitrate as the electron acceptor. Various medium components were tested under different conditions. Sulfur denitrification can drop the medium pH by producing acid, thus stopping the process half way. To control this mechanism, a 2:1 ratio of sulfur to oyster shell powder was used. Oyster shell powder addition to a sulfurdenitrifying reactor completely removed the nitrate. Using 50, 100, and 200 g of sulfur particles, reaction rate constants of 5.33, 6.29, and 7.96 mg(1/2)/l(1/2)·h were obtained, respectively; and using 200 g of sulfur particles showed the highest nitrate removal rates. For different sulfur particle sizes ranging from small (0.85-2.0 mm), medium (2.0-4.0 mm), and large (4.0-4.75 mm), reaction rate constants of 31.56, 10.88, and 6.23 mg(1/2)/l(1/2)·h were calculated. The fastest nitrate removal rate was observed for the smallest particle size. Addition of chemical oxygen demand (COD), methanol as the external carbon source, with the autotrophic denitrification in sufficiently alkaline conditions, created a balance between heterotrophic denitrification (which raises the pH) and sulfur-utilizing autotrophic denitrification, which lowers the pH.  相似文献
