The milk production conditions on 44 farms (18 designated and 26 non-designated) have been assessed by score-card marks for visual conditions, facilities, equipment, and methods. Over a period of one year fortnightly samples from these farms were subjected to routine resazurin tests and laboratory pasteurized counts. The results were compared with the total score-card marks and the equipment assessments of the farms. It was found that neither test alone sufficed to detect unsatisfactory production, but failure on the two tests combined agreed well with the production conditions and facilities.
The relation of season and mean shade atmospheric temperature to the thermoduric bacterial content of the milk was studied but no clear association was observed.  相似文献

During centrifugation operation, the major challenge in the recovery of extracellular proteins is the removal of the maximum liquid entrapped within the spaces between the settled solids–dewatering level. The ability of the scroll decanter centrifuge (SDC) to process continuously large amounts of feed material with high concentration of solids without the need for resuspension of feeds, and also to achieve relatively high dewatering, could be of great benefit for future use in the biopharmaceutical industry. However, for reliable prediction of dewatering in such a centrifuge, tests using the same kind of equipment at pilot‐scale are required, which are time consuming and costly. To alleviate the need of pilot‐scale trials, a novel USD device, with reduced amounts of feed (2 mL) and to be used in the laboratory, was developed to predict the dewatering levels of a SDC. To verify USD device, dewatering levels achieved were plotted against equivalent compression (Gtcomp) and decanting (Gtdec) times, obtained from scroll rates and feed flow rates operated at pilot‐scale, respectively. The USD device was able to successfully match dewatering trends of the pilot‐scale as a function of both Gtcomp and Gtdec, particularly for high cell density feeds, hence accounting for all key variables that influenced dewatering in a SDC. In addition, it accurately mimicked the maximum dewatering performance of the pilot‐scale equipment. Therefore the USD device has the potential to be a useful tool at early stages of process development to gather performance data in the laboratory thus minimizing lengthy and costly runs with pilot‐scale SDC. © 2013 American Institute of Chemical Engineers Biotechnol. The Canadian Association for Health, Physical Education and Recreation fitness test (CAHPER test) composed of six items was compared to two laboratory tests of endurance fitness, physical working capacity at a minute pulse rate of 170 (PWC170) and maximum oxygen uptake (Vo2 max.) in over 500 Winnipeg school children of both sexes aged 6 to 17 years. CAHPER test results were similar to the national average published by CAHPER in a test booklet. Correlation coefficients (r) of Vo2 max. for boys with the CAHPER tests were: sit-ups .42, broad jump .69, shuttle run .50, arm hang .43, 50-yard dash .60, 300-yard run .65; for girls the r values were about half the values for the boys. Much of the correlation between CAHPER tests and Vo2 max. or PWC170 depended on the association of each test with body size. When multiple correlations were obtained including surface area as the first variable, the only significant factor correlating with the endurance tests was the arm hang; none of the other tests showed a significant correlation. “Physical fitness” is task-specific, so that a subject''s position in the scoring scale of a fitness test depends entirely upon the test. The CAHPER test for physical fitness shows little or no correlation with standard laboratory measures of endurance in average children.  相似文献

Abstract

Measurements of extremely low frequency (ELF) magnetic fields were conducted in the environment of commercial laboratory equipment in order to evaluate the possible co-exposure during the experimental processes on cell cultures. Three types of device were evaluated: a cell culture CO2 incubator, a thermostatic water bath and a laboratory shaker table. These devices usually have electric motors, heating wires and electronic control systems, therefore may expose the cell cultures to undesirable ELF stray magnetic fields. Spatial distributions of magnetic field time domain signal waveform and frequency spectral analysis (FFT) were processed. Long- and short-term variation of stray magnetic field was also evaluated under normal use of investigated laboratory devices. The results show that the equipment under test may add a considerable ELF magnetic field to the ambient environmental magnetic field or to the intentional exposure to ELF, RF or other physical/chemical agents. The maximum stray magnetic fields were higher than 3?µT, 20?µT and 75?µT in the CO2 incubator, in water bath and on the laboratory shaker table, respectively, with high variation of spatial distribution and time domain. Our investigation emphasizes possible confounding factors conducting cell culture studies related to low-level ELF-EMF exposure due to the existing stray magnetic fields in the ambient environment of laboratory equipment.  相似文献

Background

In patients with COPD, both laboratory exercise tests and field walking tests are used to assess physical performance. In laboratory tests, peak exercise capacity in watts (W peak) and/or peak oxygen uptake (VO2 peak) are assessed, whereas the performance on walking tests usually is expressed as distance walked. The aim of the study was to investigate the relationship between an incremental shuttle walking test (ISWT) and two laboratory cycle tests in order to assess whether W peak could be estimated from an ISWT.

Methods

Ninety-three patients with moderate or severe COPD performed an ISWT, an incremental cycle test (ICT) to measure W peak and a semi-steady-state cycle test with breath-by-breath gas exchange analysis (CPET) to measure VO2 peak. Routine equations for conversion between cycle tests were used to estimate W peak from measured VO2 peak (CPET). Conversion equation for estimation of W peak from ISWT was found by univariate regression.

Results

There was a significant correlation between W peak and distance walked on ISWT × body weight (r = 0.88, p < 0.0001). The agreement between W peak measured by ICT and estimated from ISWT was similar to the agreement between measured W peak (ICT) and W peak estimated from measured VO2 peak by CPET.

Conclusion

Peak exercise capacity measured by an incremental cycle test could be estimated from an ISWT with similar accuracy as when estimated from peak oxygen uptake in patients with COPD.

We propose a second method at an implementation level with cost and complexity reductions for monitoring HIV patients in resource-limited settings. The model is SMARThivPack, a "three tests" combo kit. Cost and complexity reductions of HIV patient monitoring technologies were described in a first model (at a technology development stage) proposed earlier. In developed countries, HIV patient clinical management programs comport three laboratory tests containing CD4 count, viral load measurement and pharmaco-resistance testing. In most developing countries only CD4 count is performed without the other two tests. Cost and complexity of required technologies are the major challenges to the developing world to meeting the "three tests" standard. SMARThivPack model achieves the "three tests" standard in the developing world by optimizing technology choice as to favour cross-use of equipments, thereby reducing equipment cost at the laboratory implementation level. Equipment cross use also reduces complexity by reducing training time, promoting specialization and substantiating experience in equipment use. The second level cost and complexity reductions model sets the grounds for a third level, global coordination cost and complexity reductions model.  相似文献

The current rearing technique forErigone atra (Blackwall) andOedothorax apicatus (Blackwall) (Araneae, Erigonidae) was improved. To reduce time spent rearing on live fruit flies the spiders were kept on a culture of the Collembola speciesLepidocyrtus lanuginosus (Gmelin) (Entomobryidae). Side-effects on spiders of two pyrethroid insecticides (fenvalerate and lambda-cyhalothrin) and one carbamate insecticide (pirimicarb) were tested. Sensitivity of adults of both sexes and juveniles to insecticides and their influence on the rate of emergence of spiderlings from cocoons were investigated using topical application, spraying or residual contact. LD50 values for adults ranged from 0.49 to 2.52 ng a.i./spider for lambda-cyhalothrin and from 5.75 to 98.20 ng a.i./spider for fenvalerate. Topical application also resulted in up to a week's delay of web-building. A moving laboratory spraying equipment was used to spray spiders with different insecticide dosages and water volumes. Pyrethroids sprayed onto adults in webs had stronger effects than pyrethroids sprayed onto sitting or walking spiders on the soil surface. Residual contamination caused higher mortality of spiders after contact with lambda-cyhalothrin than fenvalerate. In all tests, males were more susceptible to pyrethroids than females; this difference was related to body weight. Mortality rate was higher forE. atra than forO. apicatus. Both pyrethroids were also toxic to spiderlings. Lambda-cyhalothrin inhibited emergence ofE. atra spiderlings from cocoons. Pirimicarb was harmless to both spider species.  相似文献

The acclimation of the photosynthetic apparatus of Palmaria palmata (L.) to light intensity was examined in the field and under laboratory conditions. Algae from 3 different shore levels and from laboratory cultures adapted to 6 different photon flux densities were compared. This was done on the basis of light doses, which were delivered by different light regimes in the field and in the laboratory. Laboratory samples were adjusted to constant photon flux densities between 7 and 569 μmol photons·m ? 2·s ? 1 in a 16:8 light:dark photoperiod. Under field conditions the daily amplitudes reached up to approximately 2000 μmol photons·m ? 2·s ? 1 within a natural daily light course. Over the course of 14 days the light doses resulting from those different regimes are similar for both treatments. An increasing growth rate per day with increasing light doses was observed in the laboratory. Growth was saturated at 113 mol photons·m ? 2·14 d ? 1. Light saturation points (Ek) of photosynthesis increased with increasing light doses for both field and laboratory samples, and all Ek values were significantly related to the growth light dose. A correlation between fresh weight‐related lutein content and growth light dose was found for laboratory samples only, whereas the lutein:chlorophyll a (chl a) ratio was strongly correlated with Ek for laboratory and field samples. The content of chl a and phycoerythrin (PE) per fresh weight decreased significantly with increasing light doses under field conditions. Simultaneously, the PE:chl a ratio increased, whereas this ratio was not influenced by laboratory treatments. The correspondence of Ek values for field and laboratory treatments indicated that they were affected mainly by light dose. However, the variability in pigmentation was mainly dependent on temporal variability in light intensity (the amplitude of variations in incident light).  相似文献

Some studies have found intermediate heritabilities for fluctuating asymmetry (FA) in traits, but almost all of these are flawed and/or based on laboratory experiments. We therefore tested if there was heritable variation for FA in bristle and wing traits in three field collections of Drosophila melanogaster by rearing F1s from field flies under laboratory conditions. One of the collections was reared to the F2 generation in the laboratory to compare heritability estimates from the laboratory with those from the field-laboratory comparison. Trait means indicated an increase in size under laboratory rearing. FAs increased in one collection, decreased in another collection, and showed no changes in the third collection under laboratory rearing. FAs from the collections tended to converge under laboratory conditions. Morphological traits were heritable under field conditions. However, FA was not significantly heritable for any of the individual traits or when FA was determined by combining traits. Comparisons of the two laboratory generations showed that FA heritability was low under laboratory conditions, in contrast to the morphological traits themselves. These findings suggest a very low heritability for FA in field and laboratory Drosophila. FA in bristle and wing traits may therefore be a poor indicator of genetic quality in Drosophila.  相似文献

Premating isolation between incipient species is rarely studied in nature, even though mating tests in captivity may give an inaccurate picture of natural hybridization. We studied premating barriers between the warningly colored butterflies Heliconius erato and H. himera (Lepidoptera) in a narrow contact zone in Ecuador, where hybrids are found at low frequency. Eggs obtained from wild-mated females, supplemented with eggs and young larvae collected from the wild, were reared to adulthood. Adult color patterns of these progeny were then used to infer how their parents must have mated. Likelihood was used to estimate both the frequencies of potential parental genotypes from adult phenotypes collected in the wild, and the degree of assortative mating from the inferred parents. The frequencies of parental genotypes varied across the hybrid zone, but our statistical method allowed estimates of hybrid deficit and assortative mating to be integrated across all sites sampled. The best estimate of the frequency of F1 and backcross hybrid adults in the center of the hybrid zone was 10%, with support limits (7.1%, 13.0%; support limits are asymptotically equivalent to 95% confidence limits). Mating was highly assortative: in the center of the hybrid zone the cross-mating probability between H. erato and H. himera was only 5% (0.3%, 21.4%). Wild hybrids themselves mated with both pure forms, and the probabilities that they mated in any direction were not significantly lower than those among conspecifics. These results are consistent with earlier laboratory studies on mate choice, and suggest that selection against hybrids must be strong to prevent formation of a hybrid swarm. Unfortunately, the wide support limits on mating behavior precluded a measure of the strength of selection from these data alone. Our statistical approach provides a useful general method for estimating mate choice in the wild.  相似文献

The experiences in the production of CO2-incubators [1] have been the basis for the development of a system of gassing incubation cabinets, which, starting from a basic structure with temperature and humidity control (climatic test cabinets), consist of modular built up devices with regulating circuits for carbon dioxide and oxygen content of the incubation room atmosphere. The performance of the system is considerably extended by the equipment of the devices with a microcomputer system, by which the dynamic control of the parameters of the incubation room atmosphere is possible the first time. A column soil elutriator (the Wye Washer) for extracting cyst nematodes and other small invertebrates from soil samples is described. Laboratory tests have shown the new equipment to achieve extraction rates equal to or better than existing techniques and to offer operational advantages also. Soil in quantities of up to 1 kg may be processed without drying or subsampling, the equipment can be left unattended once set to work, and can be used to extract a range of target organisms. There is only one working part (a valve) and no electrical parts, but water use and price is high. The Wye Washer is now used in the advisory entomology laboratory at Wye as the standard equipment for extracting cysts of Heterodera and Globodera, for eggs of cabbage root fly (Delia radicum) and for cocoons of black currant leaf midge (Dasineura tetensi).  相似文献

ObjectiveTo review the effect of thyroid autoantibodies on thyroid function assays and to present a case in which thyroid autoantibodies resulted in spurious assay readings for both total thyroid hormone levels and thyroid hormone uptake measurements.MethodsWe present a detailed case, including serial laboratory data, and review the relevant literature.ResultsA 61-year-old man with a history of autoimmune disease presented for evaluation of abnormal results of thyroid function tests. The patient had been treated for hypothyroidism with levothyroxine and was noted to have an elevated total thyroxine (T4) level in the setting of a low total triiodothyronine (T3) value and a mildly elevated thyrotropin concentration. He had been referred for evaluation of a presumed deiodinase deficiency that impaired conversion of T4 to T3. During treatment with levothyroxine, these test results were confirmed, and the patient was also found to have an elevated T4 uptake. These findings were initially thought to be due to an excess of transthyretin; however, more extensive testing revealed that the patient had an autoantibody to T4 that interfered with the assays for both T4 and T4 uptake.ConclusionAutoantibodies to both T3 and T4 have been described. Such antibodies are not uncommon in patients with thyroid disease. On rare occasions, these antibodies may cause spurious assay readings and obscure the diagnosis. To our knowledge, this is the first report in which both the total T4 level and the T4 uptake were elevated because of the presence of autoantibodies. Thyroid hormone autoantibodies must be considered when clinicians encounter patients with unexplained abnormal results of thyroid function tests. (Endocr Pract. 2008; 14:738-742)  相似文献

Recent research on fabricating scaffold‐type perovskite solar cells on plastic substrates has reported noteworthy progress in replacing the high‐temperature processing of TiO2 scaffolds and compact layers with various low‐temperature processes. Herein, recent progress in the laboratory is reported regarding the development of electrodeposited TiOx compact layers and brookite TiO2 scaffolds, both of which can be processed under 150 °C without greatly sacrificing their photovoltaic performance. Through systematic characterization of device properties and careful optimization of the fabrication conditions, a record‐high 15.76% power conversion efficiency of a plastic TiO2 scaffold‐type perovskite solar cell is demonstrated. In addition, bending durability and preliminary stability tests on this plastic perovskite solar cell show promising results and indicate clear directions for future improvement.  相似文献

Many patients with Chagas disease live in remote communities that lack both equipment and trained personnel to perform a diagnosis by conventional serology (CS). Thus, reliable tests suitable for use under difficult conditions are required. In this study, we evaluated the ability of personnel with and without laboratory skills to perform immunochromatographic (IC) tests to detect Chagas disease at a primary health care centre (PHCC). We examined whole blood samples from 241 patients and serum samples from 238 patients. Then, we calculated the percentage of overall agreement (POA) between the two groups of operators for the sensitivity (S), specificity (Sp) and positive (PPV) and negative (NPV) predictive values of IC tests compared to CS tests. We also evaluated the level of agreement between ELISAs and indirect haemagglutination (IHA) tests. The readings of the IC test results showed 100% agreement (POA = 1). The IC test on whole blood showed the following values: S = 87.3%; Sp = 98.8%; PPV = 96.9% and NPV = 95.9%. Additionally, the IC test on serum displayed the following results: S = 95.7%; Sp = 100%; PPV = 100% and NPV = 98.2%. Using whole blood, the agreement with ELISA was 96.3% and the agreement with IHA was 94.1%. Using serum, the agreement with ELISA was 97.8% and the agreement with IHA was 96.6%. The IC test performance with serum samples was excellent and demonstrated its usefulness in a PHCC with minimal equipment. If the IC test S value and NPV with whole blood are improved, then this test could also be used in areas lacking laboratories or specialised personnel.  相似文献

Aflatoxin contamination of maize by Aspergillus flavus poses serious potential economic losses in the US and health hazards to humans, particularly in West Africa. The Southern Regional Research Center of the United States Department of Agriculture, Agricultural Research Service (USDA-ARS-SRRC) and the International Institute of Tropical Agriculture (IITA) initiated a collaborative breeding project to develop maize germplasm with resistance to aflatoxin accumulation. Resistant genotypes from the US and selected inbred lines from IITA were used to generate backcrosses with 75% US germplasm and F1 crosses with 50% IITA and 50% US germplasm. A total of 65 S4 lines were developed from the backcross populations and 144 S4 lines were derived from the F1 crosses. These lines were separated into groups and screened in SRRC laboratory using a kernel-screening assay. Significant differences in aflatoxin production were detected among the lines within each group. Several promising S4 lines with aflatoxin values significantly lower than their respective US resistant recurrent parent or their elite tropical inbred parent were selected for resistance-confirmation tests. We found pairs of S4 lines with 75–94% common genetic backgrounds differing significantly in aflatoxin accumulation. These pairs of lines are currently being used for proteome analysis to identify resistance-associated proteins and the corresponding genes underlying resistance to aflatoxin accumulation. Following confirmation tests in the laboratory, lines with consistently low aflatoxin levels will be inoculated with A. flavus in the field in Nigeria to identify lines resistant to strains specific to both US and West Africa. Maize inbred lines with desirable agronomic traits and low levels of aflatoxin in the field would be released as sources of genes for resistance to aflatoxin production.  相似文献

Wearable systems are becoming increasingly popular for gait assessment outside of laboratory settings. A single shoe-embedded sensor module can measure the foot progression angle (FPA) during walking. The FPA has important clinical utility, particularly in populations with knee osteoarthritis, as it is a target for biomechanical treatments. However, the validity and the day-to-day reliability of FPA measurement using wearable systems during over-ground walking has yet to be established. Two gait analysis sessions on 20 healthy adults were conducted. During both sessions, participants performed natural over-ground walking in a motion capture laboratory and on a 100 m linear section of outdoor athletics track. FPA was measured in the laboratory via marker trajectory data, while the sensor module measured FPA during the outdoor track walking. Validity was examined by comparing the laboratory- and sensor-measured average FPA. Day-to-day reliability was examined by comparing the sensor-measured FPA between the first and second gait analysis sessions. Average absolute error between motion capture and sensor measured FPA were 1.7° and 2.1° at session 1 and 2, respectively. A Bland and Altman plot indicated no systematic bias, with 95% limit of agreement widths of 4.2° – 5.1°. Intraclass correlation coefficient (ICC2k) analysis resulted in good to excellent validity (ICC = 0.89 – 0.91) and reliability (ICC = 0.95). Overall, the shoe-embedded sensor module is a valid and reliable method of measuring FPA during over-ground walking without the need for laboratory equipment.  相似文献

The field of Vitamin D assay technology has progressed significantly over the past 4 decades. Further, the clinical utility of these measurements has moved from esoteric into mainstream clinical diagnosis. This movement has been fueled by the realization that Vitamin D is involved in bodily systems beyond skeletal integrity. The clinical assay techniques for circulating 25(OH)D and 1,25(OH)2D have progressed away from competitive protein binding assay (CPBAs) that utilize tritium reporters to radioimmunoassay (RIAs) that utilize both I125 and chemiluminescent reporters. These advances have allowed direct serum analysis of 25(OH)D in an automated format that provides a huge sample throughput. Detection of circulating 25(OH)D can also be achieved utilizing direct high-performance liquid chromatographic (HPLC) or liquid chromatography coupled with mass spectrometry (LC–MS) techniques. These methods are accurate, however, they require expensive equipment and restrict sample throughput in the large clinical laboratory. Direct serum detection of 1,25(OH)2D is unlikely to occur for many reasons as a sample pre-purification will always be required. However, a semi-automated chemiluminescent detection system with automated sample preparation is in final development for the determination of circulating 1,25(OH)2D. These advances will allow both 25(OH)D and 1,25(OH)2D to be detected in an accurate, rapid fashion to meet the clinical demands we see emerging.  相似文献

An enzyme-linked lectinsorbent assay (ELLA) was developed for quantification of in situ biofilm produced by Staphylococcus epidermidis in polystyrene 96-well tissue culture plates with phosphatase-labeled wheat germ agglutinin (WGA) as a specific probe for the GlcNAcβ-1,4 n component of exocellular matrix material (EMM) that is responsible for intercellular adhesion and accumulation. The ELLA and the modified Christensen dye assay were used to test 13 laboratory strains of coagulase-negative staphylococci and 10 clinical isolates of S. epidermidis. Four biofilm-positive laboratory strains of S. epidermidis were positive by both tests, and six biofilm-negative strains were negative by both. One strain of S. hemolyticus was positive by the ELLA only. Two of the 10 clinical isolates of S. epidermidis were positive by both assays, two were negative by both, and the remaining were positive by the ELLA only. The ELLA was objective, reproducible, specific, sensitive, and useful for screening strains for their capacity to adhere to plastic, produce EMM, and form biofilm. Four populations of the cactophilous species D. buzzatii have been compared with respect to the phenotypic variation of thorax and wing length of wild versus laboratory reared flies. Three of the strains were intercrossed to provide parent, F1 and F2 comparisons as a test of co-adaptation. The genetic contribution to phenotypic variation of laboratory reared flies was estimated from the correlation between sibs derived from random pair mating and reared individually in separate cultures. The average natural temperature during development was estimated from the relations between the wing/thorax ratio and temperature in laboratory tests.The variance of thorax and wing length of wild flies was several times greater than that of laboratory reared flies and the increase was attributed primarily to variation in larval food supply although temperature fluctuation is also important. There was no evidence of heterosis or F2 break-down in the crosses. For two of the populations the heritability of thorax length was high, 60–70%, and substantially lower for the third. The average temperature estimated from the wing/thorax/temperature relationship differed between sites. The reduction of body size below the potential maximum averaged 30% for two and 20% for the other population, with a wide spread about these values. The evidence is discussed in relation to assessing the nature of ecological variation by comparing the variation of morphological traits in wild and laboratory reared flies.  相似文献
