Ear implants that contained 3 mg Norgestomet or vaginal pessaries that contained 40 or 45 mg fluorogestone acetate were used to induce estrus in dairy goats in three herds in May. Ear implants or vaginal pessaries were left in place for 11 d. Cloprostenol (50 mug) and PMSG (500 IU) were administered i.m. 24 h prior to removal of ear implants or vaginal pessaries. After removal of vaginal pessaries, onset of standing estrus occurred in 22 23 goats (96%) at 20 +/- 4.7 h, in 19 20 goats (95%) at 22 +/- 6.3 h, and in 16 16 goats (100%) at 19 +/- 1.2 h in Herds A, B and C, respectively. After removal of ear implants, onset of standing estrus occurred in 25 25 goats (100%) at 19 +/- 4.9 h, in 20 22 goats (91%) at 22 +/- 7.0 h, and in 15 15 goats (100%) at 18 +/- 2.2 h in Herds A, B and C, respectively. Does were bred by natural service in Herds A and B, and by artificial insemination 28 h after vaginal pessary or ear implant removal in Herd C. Pregnancy rates were determined 39 to 53 d post breeding by real-time ultrasound. Pregnancy rates in goats with vaginal pessaries were 32, 55 and 6%; and in goats with ear implants they were 56, 67 and 27% in Herds A, B and C, respectively. Problems encountered included poor libido in some bucks, abortions in undersized yearling does, and loss of ear implants by three does (not included in the data). Statistically there was no difference in pregnancy rates between goats receiving vaginal pessaries or ear implants (P>0.10).  相似文献

Fifteen does received intravaginal pessaries impregnated with 20 mg fluorogestone acetate (Cronolone) at various times after the end of estrus for a period of fourteen days. The does were then treated on days 2, 4, 5, 12, or 16 after onset of estrus following the progestin treatment, with 15 mg of prostaglandin F2α (Lutalyse) injected intramuscularly. Seventy three percent of the does showed estrus within 2 days after withdrawal of progestin treatment. Ten out of 11 does treated with prostaglandins 4 to 16 days after the end of estrus exhibited estrus within 44 to 72 hours after treatment. Breeding following the use of prostaglandin in these does resulted in a high pregnancy rate. It is concluded that a sequential use of progestin and prostaglandin may be a useful method for achieving estrus synchronization associated with high fertility in does.  相似文献

Finewooled Rambouillet range ewes were used in a study to determine the feasibility of using a progesterone ear implant to synchronize estrus. In addition, some of the ewes were further treated with injections of follicle stimulating hormone-pituitary (FSH-P) to induce superovulation. Five days following estrus detection and breeding, FSH-P-treated ewes were laparotomized and surgically flushed to recover embryos. The number of corpora lutea (CL), the total number of embryos and the number of transferable embryos recovered were recorded along with the number and size of follicles present on both ovaries. Ewes synchronized as recipients were laparotomized for surgical transfer of embryos 5 d following estrus. The number of CL and follicles were recorded. Response to superovulation by FSH-P did not differ (P>0.05) between age groups of ewes when the number of CL present was counted. However, the total number of embryos flushed and good embryos was lower (P<0.05) among the oldest (7 yr) ewes. The number of follicles present showed little variation between age groups. Recipient ewes (No FSH-P) were similar in the number of CL with 6-yr-old ewes, having fewer (P>0.05) CL, than 3-, 4- or 7-yr-old ewes. Only slight variation was boserved in the number of follicles in recipient ewes. Among donor ewes receiving FSH-P in addition to Synchro-Mate B, 71% were detected in estrus within 48 h of implant removal vs 55% of the recipients.  相似文献

Estrus was synchronized in 64 dairy goats in July with norgestomet ear implants. Half the does received ear implants that contained 6 mg norgestomet and the remaining does received implants that contained 3 mg. Implants were left in place for 11 days. Each doe received i.m. injections of 400 IU PMSG and 50 mug cloprostenol 24 hours prior to implant removal. Twenty-eight of 32 does (87.5%) that received 6 mg or 3 mg norgestomet exhibited onset of estrus within 24 hours of implant removal. All does had exhibited onset of standing estrus by 43 hours after implant removal. Does were hand-mated to fertile bucks twice daily while in standing estrus. There were no differences between does implanted with 6 mg or 3 mg in fertility to the induced estrus (74.2% vs 75% kidding), mean length of gestation (151.0 +/- 3.2 vs 151.6 +/- 2.0 days), mean number of kids per doe (2.1 +/- 0.8 vs 2.3 +/- 0.7) or in mean kid weights (3.10 +/- 0.80 vs 3.06 +/- 0.86 kg) (6 mg vs 3 mg, respectively). It was concluded that ear implants that contained 3 mg of norgestomet were equally as effective as implants that contained 6 mg for synchronization of estrus in dairy goats.  相似文献

The main objective of this study was to investigate the effectiveness of certain progestagen-gonadotrophin treatments on synchronization of estrus in sheep. In Experiment I, 30 Chios ewes were treated at the beginning of the breeding season with medroxyprogesterone acetate (MAP) intravaginal sponges for 12 days and a single i.m. treatment of either FSH (Group 1,10 IU, n = 8; Group 2, 5 IU, n = 8; Group 3, 2.5 IU, n = 8) or eCG (Group 4, 400 IU, n = 6) at the time of sponge removal. Ten days after sponge removal laparotomy was performed to record ovarian response. Clinical estrus was observed in more (though not at a significant level) FSH treated than eCG treated sheep (62.5% versus 33.3%). Administration of 400 IU eCG resulted in the highest mean number of CL perewe ovulating (2.8 +/- 0.2), with administration of 10 IU FSH producing the next best results (2.1 +/- 0.3). Statistically significant differences in the mean number of CL per ewe ovulating were found only between ewes in Group 3 (1.7 +/- 0.4) and Group 4 (2.8 +/- 0.2) (P < 0.05). In Experiment II, 53 Chios and 30 Berrichon ewes were treated during the mid-breeding season with MAP intravaginal sponges for 12 days and a single i.m. treatment of either 10 IU FSH (27 Chios and 16 Berrichon ewes) or 400 IU eCG (26 Chios and 14 Berrichon ewes), at the time of sponge removal. Ewes that were in estrus on Days 2-4 and 19-23 after sponge removal were mated to fertile rams. No significant differences were recorded between treatment or breed groups in the proportions of ewes observed in estrus after treatment. In the Berrichon breed, FSH administration resulted in higher lambing rates (93.8% versus 57.1%, P < 0.05) and higher mean number of lambs born per ewe exposed to rams (1.4 +/- 0.2 versus 0.8 +/- 0.2, P < 0.05) than that of eCG. After treatment with eCG, the mean number of lambs born per ewe exposed to rams was higher in the Chios than the Berrichon breed (1.4 +/- 0.2 versus 0.8 +/- 0.2, P < 0.05). After treatment with FSH, the lambing rate was higher in the Berrichon than the Chios breed (93.8% versus 63.0%, P < 0.05). In conclusion, a single FSH treatment (5 or 10 IU) at the end of progestagen treatment appears to be more effective than eCG for the induction of synchronized estrus in sheep at the beginning of the breeding season, with no cases of abnormal ovarian response observed. During the mid-breeding season FSH (10 IU) appears to be equally as effective as eCG (400 IU) in respect of lambing rate and mean number of lambs born per ewe.  相似文献

This experiment was conducted to define the temporal relationships among estrus, the LH surge and ovulation after estrus synchronization in dwarf goats and to assess the effect of season on these parameters. In November (breeding season), March (transition period) and July (non-breeding season), estrus was synchronized in 12 dwarf goats by means of intravaginal sponges containing 60 mg medroxyprogesterone acetate (MAP) for 10 d, coupled with 125 microg cloprostenol i.m. 48 h before sponge removal and 300 IU eCG i.m. at sponge removal. A different group of animals was used during each time period. Onset of estrus was monitored using two males, and blood samples for the measurement of plasma LH were collected at 2-h intervals from 24 to 60 h after sponge removal. Ovulation was confirmed by laparoscopy at 54 and 72 h after sponge removal. A seasonal shift was detected in the intervals to onset of estrus, LH surge, and ovulation after sponge removal (P<0.05), with sponge removal to onset of estrus being shorter (P<0.05) in November (25.0 +/- 1.56 h) and July (28.9 +/- 2.43 h) than in March (40.9 +/- 3.27 h). The intervals between onset of estrus and the LH surge and between the LH surge and ovulation were found to be constant throughout the different seasons. An optimal time for breeding, artificial insemination, oocyte and embryo recovery, and embryo transfer may be predicted using information gained from these studies.  相似文献

Thirty-four mixed breed cyclic does were randomly divided into two groups of 17 each. One group was synchronized for estrus using two i.m. injections of 8 mg PGF2alpha administered 11 days apart. The other group served as controls and was bred at the time of naturally occurring estrus. Both groups were bred by natural service. Ninety-four percent of the treated does came into estrus within a mean (+/- S.E.) of 53 +/- 3 hours after the second injection of 8 mg PGF2alpha. No differences (P > 0.10) in the first service conception rates based on radiography at mid-gestation were observed between the treated and control groups. It was concluded that the use of 8 mg injections of PGF2alpha 11 days apart had no detrimental effects on fertility of goats.  相似文献

There is still a lack of information on estrus synchronization in goats. Understanding the estrus synchronization protocols and the subsequent effects is important to improve the efficiency of assisted reproductive technologies (ARTs) and subsequently would improve the breeding procedures. This study will help in determining the most suitable estrus synchronization protocol and understand better the effect on the sexual behaviour and hormonal effects in goats. A total of 127 Boer does were used and divided into three groups with different duration of CIDR insertion intravaginally either for 14 (two groups) or 9 days (one group). Approximately 0.5 ml Estrumate® (PG) was administered intramuscularly to all groups at CIDR removal, and only groups PMSG14 and PMSG9 were administered with 200IU of Pregnant Mare Serum Gonadotropin (PMSG) intramuscularly. Estrus signs were observed at 4 h intervals and blood samples were collected for progesterone and luteinizing hormone determination. The percentage of does in estrus within 24 to 72 h post CIDR removal was significantly higher (P<0.05) in groups with PMSG compared to the group without the PMSG. The numbers of does display estrus signs within 24 to 28 h post CIDR removal were significantly higher (P<0.05) in group shorter period (9 days) compared to groups with 14 days CIDR. The P4 concentrations at 24 hours post CIDR removals and LH concentration was not significantly different (P>0.05) in all groups. The time of the LH peak in the group without the PMSG was significantly delayed (P<0.05) when compared to group 9 days CIDR and administered with PMSG. It is recommended to use the treatment for 9 days CIDR since the estrous cycle can be shortened.  相似文献

The induction of estrus in 17 previously cycling nulliparous ewes, 9 to 10 months of age, was attempted with Medroxyprogesterone acetate (MAP) pessaries during the early anestrous period (March-April). Ewes were verified to be anestrous by the lack of estrous behavior in the presence of a vasectomized ram and by a radioimmunoassay for serum progesterone in two samples taken 7 days apart showing less than 1 ng/ml serum progesterone. Superovulation was attempted with injections of either FSH or FSH + LH. MAP vaginal pessaries remained in place for a period of 12 days and FSH was administered to all ewes (IM) at 12 hr intervals over a 3 day period; 5 mg was injected twice on day 11 after pessary insertion, followed by 4 and 3 mg injections twice daily on each succeeding day, for a total of 24 mg per ewe. Nine ewes were given 25 mg LH (IV) within 8 hrs after the onset of behavioral estrus in addition to FSH. Ewes were hand-mated to several rams at 12 hr intervals throughout the estrus period. Ovulation and fertilization rates were recorded for each ewe following midline laparotomy and embryo collection. All ewes were in estrus between 36 and 48 hrs after removal of the MAP pessaries. In ewes injected with FSH only, 8 of 8 ovulated with a mean ovulation rate of 6.0 +/- 4.4 and a fertilization rate of 70%. Nine of 9 ewes receiving both FSH + LH ovulated with a mean ovulation rate of 13.9 +/- 13.1 and a fertilization rate of 72%. Statistical analysis by Students t-test resulted in differences in number of ova recovered (P<.05) between FSH only and FSH + LH treated ewes and a trend towards increased ovulation rate in FSH + LH treated ewes. These results show that early seasonally anestrous ewes can be successfully induced and synchronized for estrus with MAP pessaries and the number of ova recovered is increased with the inclusion of LH in the superovulation regime.  相似文献

Embryos from Bos indicus are more resistant to elevated culture temperature (i.e. heat shock) than embryos from some Bos taurus breeds. The present experiment was designed to determine if Brahman embryos have greater resistance to heat shock than Holstein embryos at a stage in development before the embryonic genome was fully activated. A second objective was to test breed effects on estrus synchronization and superovulation responses. A total of 29 Brahman and 24 Holstein cows were subjected to estrus synchronization using gonadotropin releasing hormone (GnRH) and prostaglandin F2alpha (PGF2alpha) superovulation. Embryos were collected at 48 h and day 5 after insemination. There was a tendency for a lower proportion of Brahmans to be detected in standing estrus than Holsteins. There were no differences between breeds in the proportion of cows detected in estrus using both tailpaint and standing estrus as criteria or in interval from PGF2alpha to estrus. The degree of synchrony in estrus was greater for Brahmans. Superovulation response was generally similar between breeds. At 48 h after insemination, there was a tendency for a greater proportion of Brahman oocytes to have undergone cleavage. Uncleaved oocytes were cultured for an additional 24 h-at this time, cleavage rate was similar between breeds. When embryos reached the 2-4-cell stage, they were heat-shocked for 4.5 h at 41 degrees C. This heat shock reduced the proportion of embryos that developed to the blastocyst stage but there was no breedxtreatment interaction. At day 5 after insemination, the number of embryos recovered was too low to allow comparison of breed effects. In conclusion, genetic effects on cellular thermotolerance that make Brahman embryos more resistant to heat shock are not expressed at the 2-4-cell stage. There were few differences between Brahman and Holstein in response to estrus synchronization and superovulation. The fact that cleavage tended to occur earlier in Brahman than Holstein embryos suggests breed differences in timing of ovulation, fertilization or events leading to cleavage.  相似文献

A study was conducted on 35 East African shorthorned female goats to determine if a combination of buck teasing and low doses of a prostaglandin (PGF(2) alpha) analogue, cloprostenol, given intravulvo-submucosally (i.v.s.m.) would be suitable for synchronization of estrus. Goats were allotted, with the onset of estrus, to seven groups (n = 5 goats per group). Five of the seven groups received varying doses of cloprostenol: Group 1 (125 mug cloprostenol i.m. per goat); Group 2 (62.5 mug cloprostenol i.v.s.m. per goat); Group 3 (62.5 mug cloprostenol i.v.s.m. per goat plus buck teasing); Group 4 (31.25 mug cloprostenol i.v.s.m. per goat); Group 5 (31.25 mug cloprostenol i.v.s.m. per goat plus buck teasing); Group 6 (buck teasing); Group 7, (2 ml physiological saline i.v.s.m. per goat, control group). Plasma progesterone concentration was measured on day of treatment and for 6 d thereafter. All goats in groups 1, 2, 3 and 5 exhibited estrus within 68 h. Thus, the number of goats receiving low doses of PG-cloprostenol intravulvo-submucosally observed in estrus increased (P < 0.05) with exposure to bucks. Exhibition of behavioral signs of estrus was maximal between 2 and 20 h after onset of signs of estrus. The exposure of females to males prior to intrauterine penetration was an advantage because copious mucus eased penetration.  相似文献

Two trials were conducted to examine the effects of estrus synchronization scheme, gonadotropin injection protocol and presence of a large ovarian follicle on response to superstimulation of follicular development and the ensuing superovulation. Estrus was synchronized with either a progestin compound (MGA) or by the use of a luteolytic agent (PGF). Superstimulation was induced with 280 mg equivalents of pFSH administered either by a single subcutaneous injection or by a series of 8 intramuscular injections over 4 d. Follicular development was followed for 5 d with real-time ultrasound, and the heifers were retrospectively classified as to the presence or absence of a large follicle (> or = 8 mm; morphologically dominant follicle) at the start of superstimulation. The 2 trials differed by season of the year and genetic origin of the heifers. In Trial I (20 heifers), the ovulation rate was influenced by the 3-way interaction of the synchronization scheme, injection protocol and morphologically dominant follicle (P = 0.05). The number of large follicles on Day 5 (Day 0 = day of start of superstimulation) and ovarian score (scale 1 to 5 based on extent of follicular development; 1 = least, 5 = most) on Day 5 were significantly correlated (P < 0.05) with ovulation rate. In Trial II (20 heifers), the ovulation rate, number of embryos recovered, number of transferable embryos and ovarian weights were all greater (P < 0.05 to P < 0.01) with the 8-injection protocol than the 1-injection protocol. The number of medium follicles (5 to 7 mm) on Days 2 and 3, number of large follicles (> or = 8 mm) on Days 3, 4 and 5 and ovarian scores on Days 4 and 5 were all significantly correlated (P < 0.05) with ovulation rate. In both trials, differences in follicle populations were not seen until Day 3 of the superstimulation procedure. Collectively, these trials do not provide strong support for a single injection of FSH, as used here, nor does it indicate a clear advantage for either MGA or PGF as a means of enhancing the ovulation rate or embryo quality.  相似文献

Indications for estrus induction in the bitch include missed breeding opportunities or conception failure, the treatment of primary or secondary anestrus and synchronization of ovulation for embryo transfer programs. Reported methods for canine estrus induction include the use of synthetic estrogens (diethylstilbesterol), dopamine agonists (bromocryptine and cabergoline), GnRH agonists (lutrelin, buserelin, fertirelin, deslorelin, and leuprolide) and exogenous gonadotropins (luteinizing hormone, follicle stimulating hormone, human chorionic gonadotropin, pregnant mare serum gonadotropin, and human menopausal gonadotropin). These methods vary widely in efficacy of inducing estrus, as well as in the fertility of the induced estrus. The applicability of some of these methods for clinical practice is questionable. This review will summarize published reports on estrus induction and synchronization in bitches and summarize preliminary results using a long-acting injectable preparation of deslorelin.  相似文献

Beef females (547) were included in three experiments to evaluate methods of identifying and inseminating nonpregnant beef females after synchronization of second estrus with norgestomet implants. In the first experiment, heifers not pregnant to the first insemination were identified for insemination via estrus (inseminated via the a.m./p.m. rule or 48 h after implant removal). In the second experiment, females not pregnant to the first insemination were identified for insemination via estrus (inseminated via the a.m./p.m. rule) or progesterone concentrations < 1.5 ng/mL at implant removal (inseminated 48 h after implant removal). In the third experiment, heifers not pregnant to the first insemination were identified for insemination via progesterone concentrations (as in experiment 2) or anterior vagina electrical resistance values < 81 ohm resistance 48 h after implant removal (inseminated after resistance measured). All methods of identifying and inseminating nonpregnant females were equally effective (P > 0.10) and did not effect (P > 0.10) calving rates from the first and second AI.  相似文献

The purpose of the experiment was to test the hypothesis that a variable and/or insufficient level of progestagen at the end of a treatment to synchronize estrus in goats could explain variability in the onset of estrus. The experiment was performed during the anestrous season on 2 herds, one of Alpine (n = 49) the other of Saanen (n = 53) dairy goats. The animals were allocated to 1 of 3 treatments: Group 1 received a vaginal sponge impregnated with 45 mg of fluorogestone acetate (FGA) on Day 0; Group 2 received a sponge on Day 0 plus a second sponge on Day 7; Group 3 received a sponge on Day 0 plus a second sponge on Day 9. The sponges were withdrawn on Day 11. All goats received 400 or 500 IU eCG and 50 mug PGF(2alpha) analog 48 h prior to sponge removal. They were inseminated with frozen-thawed semen 24 h after the onset of estrus. Among treatment groups no difference (P > 0.05) was observed for the following parameters: percentage of goats in estrus, percentage of goats ovulating, mean time and variability of onset of estrus. The fertility of Alpine goats in Group 3 was significantly decreased (P < 0.05). No effect on prolificacy was noticed. These observations show that to increase progestagen level at the end of treatment did not improve estrus synchronization. They provide further evidence that treatments with too high progestagen amounts can decrease fertility.  相似文献

该文探讨了小鼠发情周期中阴门状态、阴道脱落细胞类型变化规律、卵泡发育规律及其相互关系,并比较了发情周期不同阶段的超排效果。结果表明,采用阴门状态观察法和阴道脱落细胞涂片法,能有效判断小鼠发情周期阶段。卵巢组织切片观察结果表明,在发情周期不同阶段,小鼠的卵泡发育和黄体的生成与消退存在明显的规律性变化;小鼠发情周期中,其阴门状态、阴道脱落细胞种类及卵泡发育动态之间存在相关关系;发情周期不同阶段开始超排的小鼠,其配种见栓率和回收胚胎平均数均存在明显差异,发情前期显著优于发情后期与间情期(P<0.05),并高于发情期,但差异不显著(P>0.05),即阴门状态观察法与阴道脱落细胞涂片法均可用于小鼠发情周期阶段的判断,发情前期为最适宜的小鼠超排时期。  相似文献
