Two genetically distinct habitat races of Drosophila melanogaster coexist in Brazzaville (Congo). One is the typical field type of Afrotropical populations, the other mainly breeds in beer residues in breweries. These two populations differ in their ethanol tolerance, in their allelic frequencies at several enzyme and microsatellite loci and in the composition of their cuticular hydrocarbons. The brewery population is quite similar to European temperate populations with regard to all these traits. Previous investigations of two morphological traits (ovariole number and sternopleural bristle number) failed to detect any difference between the two habitat races. Here we investigated other morphological traits (wing and thorax length, thorax pigmentation and female abdomen pigmentation). The reaction norms of these traits according to growth temperature were compared in the two Afrotropical habitat races and in a French temperate population. As expected, the French population was very different from the field African population: as a general rule, the brewery population (Kronenbourg) was intermediate in several aspects between the other two. We conclude that the strong selective forces that maintain the genetic divergence between the two habitat races also act on morphometrical traits, and the possible selective mechanisms are discussed.  相似文献

Variation in Adh and Gpdh-1 gene frequencies has been used to check for microdifferentiation in Spanish samples of Drosophila melanogaster inside and outside a wine cellar. Flies were collected after vintage and after overwintering respectively; within each period samples were taken on up to five consecutive days each month. Variation of gene frequencies of Adh and Gpdh-1 can be considered random when samples collected each month are taken into account. When mean monthly frequencies are considered, Gpdh-1 does not show any significant variation all over the year; yet, variation of the frequency of Adh S shows a cyclical pattern, its frequency being maximum at the end of the summer and minimum after overwintering. Due to the parallel change of the frequency of the inversion In(2L)t and the Adh S allele, no decision can be made whether the Adh locus itself or the inversion are responsible for the changes.  相似文献

Determining the genetic basis of environmental adaptation is a central problem of evolutionary biology. This issue has been fruitfully addressed by examining genetic differentiation between populations that are recently separated and/or experience high rates of gene flow. A good example of this approach is the decades-long investigation of selection acting along latitudinal clines in Drosophila melanogaster. Here we use next-generation genome sequencing to reexamine the well-studied Australian D. melanogaster cline. We find evidence for extensive differentiation between temperate and tropical populations, with regulatory regions and unannotated regions showing particularly high levels of differentiation. Although the physical genomic scale of geographic differentiation is small--on the order of gene sized--we observed several larger highly differentiated regions. The region spanned by the cosmopolitan inversion polymorphism In(3R)P shows higher levels of differentiation, consistent with the major difference in allele frequencies of Standard and In(3R)P karyotypes in temperate vs. tropical Australian populations. Our analysis reveals evidence for spatially varying selection on a number of key biological processes, suggesting fundamental biological differences between flies from these two geographic regions.  相似文献

While pre-reproductive isolations are more and more frequently described between closely related species or within species, very little is known about their conditions of emergence. In Brazzaville, two populations (Kronenbourg and Loua) of Drosophila melanogaster show a premating isolation. Two hypotheses were proposed to explain such a situation: a local sympatric differentiation or an allopatric divergence followed by a secondary contact. A microsatellite analysis, using markers on all chromosomes, strongly suggests that the Kronenbourg population has a European origin. Therefore, the allopatric divergence between Kronenbourg and Loua populations is probably responsible for the sexual isolation observed today in sympatry, after a recent introduction of a European propagule in Brazzaville.  相似文献

Using multilocus (RAPD) markers, variation and divergence of genomic DNA was examined in two Drosophila melanogaster populations from Russia and three populations from Ukraine. The populations were found to exhibit high polymorphism at RAPD markers. Estimation of genetic distances between the populations showed low differentiation of geographically distant populations of D. melanogaster. Significant gene flow between the D. melanogaster populations was found, which depended on the geographical distance between them.  相似文献

To study the genetic differentiation and inversion clines in Indian natural populations of Drosophila melanogaster, 14 natural populations (6 from the north and 8 from the south) were screened for chromosome inversions. The chromosomal analysis revealed the presence of 23 paracentric inversions, which include 4 common cosmopolitan, 4 rare cosmopolitan, 2 recurrent endemic, and 13 unique endemic (new inversions detected for the first time) inversions. The difference in karyotype frequencies between populations from the north and south were highly significant and the level of inversion heterozygosity was higher in populations from the south. Statistically significant negative correlations were found between each of the four common cosmopolitan inversions and latitude. These findings are in accord with results from other worldwide geographic regions and show that Indian populations of D. melanogaster have undergone considerable genetic differentiation at the level of inversion polymorphism.  相似文献

Genetic variation at 59 gene loci coding for enzymes (50) and larval proteins (9) has been studied in sympatric populations of Drosophila melanogaster and D. simulans from insular and continental origin. The average number of alleles per locus, the mean proportion of polymorphic loci and the mean heterozygosity are similar both within and between species. There are however some significant differences between D. simulans populations in the genotypic frequencies for four polymorphic loci.  相似文献

Drosophila melanogaster is widely used as a model in DNA variation studies. Patterns of polymorphism have, however, been affected by the history of this species, which is thought to have recently spread out of Africa to the rest of the world. We analyzed DNA sequence variation in 11 populations, including four continental African and seven non-African samples (including Madagascar), at four independent X-linked loci. Variation patterns at all four loci followed neutral expectations in all African populations, but departed from it in all non-African ones due to a marked haplotype dimorphism at three out of four loci. We also found that all non-African populations show the same major haplotypes, though in various frequencies. A parsimonious explanation for these observations is that all non-African populations are derived from a single ancestral population having undergone a substantial reduction of polymorphism, probably through a bottleneck. Less likely alternatives involve either selection at all four loci simultaneously (including balancing selection at three of them), or admixture between two divergent populations. Small but significant structure was observed among African populations, and there were indications of differentiation across Eurasia for non-African ones. Since population history may result in non-equilibrium variation patterns, our study confirms that the search for footprints of selection in the D. melanogaster genome must include a sufficient understanding of its history.  相似文献

Electrophoretic studies of the degree and pattern of polymorphism at two third-chromosome loci, esterase-6 (Est-6) and phosphoglucomutase (PGM), were carried out in three Drosophila melanogaster populations collected from different localities in Iraq: Mosul, Tuwaitha, and Basrah. The results show that only the Tuwaitha population was polymorphic for both loci; the other two populations were polymorphic for Est-6 and monomorphic for PGM. The allele frequency changes at both loci were followed for 20 generations in an experimental cage derived from the Tuwaitha population; it was found that there is a deviation from Hardy-Weinberg equilibrium at both loci toward the homozygote.  相似文献

Drosophila melanogaster shows clinal variation along latitudinal transects on multiple continents for several phenotypes, allozyme variants, sequence variants, and chromosome inversions. Previous investigation suggests that many such clines are due to spatially varying selection rather than demographic history, but the genomic extent of such selection is unknown. To map differentiation throughout the genome, we hybridized DNA from temperate and subtropical populations to Affymetrix tiling arrays. The dense genomic sampling of variants and low level of linkage disequilibrium in D. melanogaster enabled identification of many small, differentiated regions. Many regions are differentiated in parallel in the United States and Australia, strongly supporting the idea that they are influenced by spatially varying selection. Genomic differentiation is distributed nonrandomly with respect to gene function, even in regions differentiated on only one continent, providing further evidence for the role of selection. These data provide candidate genes for phenotypes known to vary clinally and implicate interesting new processes in genotype-by-environment interactions, including chorion proteins, proteins regulating meiotic recombination and segregation, gustatory and olfactory receptors, and proteins affecting synaptic function and behavior. This portrait of differentiation provides a genomic perspective on adaptation and the maintenance of variation through spatially varying selection.  相似文献

In five Indian localities, it was possible to collect D. melanogaster in two different types of habitats, namely ordinary domestic and alcohol rich ones. Tolerance and utilization capacity of several alcohols and of acetic acid were analyzed in a total of 10 local populations. Results on two other species (D. repleta and D. immigrans) were also available from one place. In each locality, the population from alcohol rich habitat proved to be more tolerant to all the investigated products and also to be more capable of using them as a resource. Alcohols toxicity increased with increasing carbon chain length and secondary alcohols were more toxic than primary ones. Utilization capacity of all products was relatively independent of their toxicity. Especially acetic acid, the toxicity of which was low and similar to that of ethanol, was always a fairly poor resource. From a genetic point of view, tolerance and utilization capacity appeared as two relatively independent traits. Natural selection, which is responsible for the genetic differentiation of local populations, is likely to act simultaneously on both traits. Drosophila melanogaster is an important model organism in evolutionary genetics, yet little is known about the population structure and the demographic history of this species within sub-Saharan Africa, which is thought to contain its ancestral range. We surveyed nucleotide variation at four 1-kb fragments in 240 individual lines representing 21 sub-Saharan and 4 Palearctic population samples of D. melanogaster. In agreement with recent studies, we find a small but significant level of genetic differentiation within sub-Saharan Africa. A clear geographic pattern is observed, with eastern and western African populations composing two genetically distinct groups. This pattern may have resulted from a relatively recent establishment of D. melanogaster in western Africa. Eastern populations show greater evidence for long-term stability, consistent with the hypothesis that eastern Africa contains the ancestral range of the species. Three sub-Saharan populations show evidence for cosmopolitan introgression. Apart from those cases, the closest relationships between Palearctic and sub-Saharan populations involve a sample from the rift zone (Uganda), suggesting that the progenitors of Palearctic D. melanogaster might have come from this region. Finally, we find a large excess of singleton polymorphisms in the full data set, which is best explained by a combination of population growth and purifying selection.  相似文献

When 41 populations from Africa (south of the Sahara) and Indian Ocean islands were analysed for their chromosomal inversion polymorphism, 34 rearrangements were found, including the four common cosmopolitans (In(2L)t, In(2R)NS, In(3L)P and In(3R)P), four rare cosmopolitans (In(2L)NS, In(3R)C, In(3R)Mo and In(3R)K) and six African polymorphic ('recurrent') endemics. Mean inversion frequencies per major autosome arm were positively and, generally, highly correlated to each other. There was no altitudinal nor latitudinal cline of inversion frequency, except for one African polymorphic endemic. Significant longitudinal clines were detected for In(2L)t, In(3L)P and In(3R)K; in all cases, inversion frequencies decreased eastward. Principal components analysis and ANOVA made it possible to distinguish three groups of populations. A high level of polymorphism was found in populations from west tropical Africa. The other low altitude populations from the mainland were moderately polymorphic, whereas the lowest levels of polymorphism were those of high altitude populations and of Indian Ocean islands. Moreover, some regional and local differentiation was also found. The frequency of unique autosomal inversions was not different from those found in Asia, Australia and America, but was significantly higher than that in Europe and North Africa. A West-East differentiation was also observed for the African polymorphic endemics. The present geographic pattern suggests a long, patchy evolution with restricted gene flow, followed by the modern period with numerous recent migrations linked to human transportation.  相似文献

Genetic rescue has been proposed as a management strategy to improve the fitness of genetically eroded populations by alleviating inbreeding depression. We studied the dynamics of genetic rescue in inbred populations of Drosophila. Using balancer chromosomes, we show that the force of heterosis that accompanies genetic rescue is large and allows even a recessive lethal to increase substantially in frequency in the rescued populations, particularly at stress temperatures. This indicates that deleterious alleles present in the immigrants can increase significantly in frequency in the recipient population when they are in linkage disequilibrium with genes responsible for the heterosis. In a second experiment we rescued eight inbred Drosophila populations with immigrants from two other inbred populations and observe: (i) there is a significant increase in viability both 5 and 10 generations after the rescue event, showing that the increase in fitness is not transient but persists long-term. (ii) The lower the fitness of the recipient population the larger the fitness increase. (iii) The increase in fitness depends significantly on the origin of the rescuers. The immigrants used were fixed for a conditional lethal that was mildly deleterious at 25°C but lethal at 29°C. By comparing fitness at 25°C (the temperature during the rescue experiment) and 29°C, we show that the lethal allele reached significant frequencies in most rescued populations, which upon renewed inbreeding became fixed in part of the inbred lines. In conclusion, in addition to the fitness increase genetic rescue can easily result in a substantial increase in the frequency of mildly deleterious alleles carried by the immigrants. This can endanger the rescued population greatly when it undergoes recurrent inbreeding. However, using a sufficient number of immigrants and to accompany the rescue event with the right demographic measures will overcome this problem. As such, genetic rescue still is a viable option to manage genetically eroded populations.  相似文献
