The activity of yeast invertase as a function of oxidation-reduction potential has been investigated using a large number of oxidants and reductants. The activity is constant over the range of Eh from –270 to +600 mv., but above Eh = +600 mv. there is a sharp decrease in activity reaching 0 at Eh = +1,000 mv. The inhibiting action of strong oxidants is upon the enzyme rather than on the substrate and appears to be essentially irreversible Experiments indicate that the inhibiting action of strong oxidants on invertase is primarily related to their high oxidation-reduction potential rather than to a specific toxic action unrelated to Eh. The effects of oxidation-reduction potential upon invertase activity are independent of the purity of the enzyme, since they are the same for commercial invertases, fresh bakers'' yeast, powdered bakers'' yeast, brewers'' yeast, and highly purified invertase. Possible mechanisms involved in the inactivation of invertase by oxidants are discussed.  相似文献

The objective of this research was to develop a system in which the direction of fructan metabolism could be controlled. Three-week-old wheat seedlings (Triticum aestivum L. cv Caldwell) grown at 25°C were transferred to cold temperature (10°C) to induce fructan synthesis and then were transferred to continuous darkness at 25°C after defoliation and fructan degradation monitored. The total fructan content increased significantly 1 day after transferring from 25°C to 10°C in both leaf blades and the remainder of the shoot tissue, 90% of which was leaf sheath tissue. Leaf sheaths contained higher concentrations of fructan and greater portions of high molecular weight fructan than did leaf blades. Fructan content in leaf sheaths declined rapidly and was gone completely within 48 hours following transfer to 25°C in darkness. In leaf blades the invertase activity fluctuated during cold treatment. The activity of sucrose:sucrose fructosyl transferase increased markedly during cold treatment, while fructan hydrolase activity decreased slightly. In leaf sheaths, however, the activity of invertase decreased rapidly upon transfer to cold temperature and remained low. Trends in sucrose:sucrose fructosyl transferase and hydrolase activity in sheaths were the same as those of leaf blades. Sheath invertase and hydrolase activity increased when plants were transferred back to darkness at 25°C, while sucrose:sucrose fructosyl transferase activity decreased. These results indicate that changing leaf sheath temperature can be utilized to control the direction of fructan metabolism and thus provide a system in which the synthesis or degradation of fructan can be examined.  相似文献

The speed of translatory movement of Beggiatoa alba is governed by temperature in such a way that between 5° and 33° the temperature characteristics µ = 16,100 and µ = 8,400 respectively obtain for the temperature ranges 5° to 16.5° and 16.5° to 33°. The "break" at 16°–17° is emphasized by the occurrence of a wider latitude of variation in speed above this temperature. Above 16° the progression of Thiothrix yields µ = 8,300. The possible relation of these values to that previously obtained for similar movement in (photosynthetic) Oscillatoria is commented upon.  相似文献

1. Mackerel egg development was followed to hatching at constant temperatures of 10°, 11°, 12°, 13°, 14°, 15°, 16°, 17°, 18°, 19°, 20°, 21°, 22°, and 24°C. Experiment showed that typical development could be realized only between 11° and 21°. 2. The length of the developmental period increases from 49.5 hours to 207 hours when the temperature is lowered from 21° to 10°C. 3. The calculated µ for the development of the mackerel egg is about 19,000 at temperatures above 15° and approximately 24,900 for temperatures below 15°C. 15° is, apparently, a critical temperature for this process. 4. The calculated values of µ for eight stages of development preceding hatching, i.e. 6 somites, 12 somites, 18 somites, 24 somites, three-quarters circles, four-fifths circles, five-sixths circles, and full circles, are essentially the same as the µ''s for hatching, indicating that the rate of differentiation up to hatching is governed by one process throughout. Critical temperatures for these stages approximate 15°. 5. The total mortality during the incubation period was least at 16°C. where it amounted to 43 per cent. At temperatures above and below this there was a steady increase in the percentage of mortality which reached 100 per cent at 10° and 21°.  相似文献

The growth response of tomato (Lycopersicon esculentum L.) to temperature and irradiance may be related to carbohydrate concentration. Plants in the exponential phase of vegetative growth were grown under temperatures ranging from 9 to 36°C and under low or high irradiances of approximately 110 or 370 microeinsteins per square meter per second photosynthetically active radiation for a 12 hour photoperiod. The relative growth rate, leaf area ratio, net assimilation rate and whole plant carbohydrate levels were measured. At high irradiance, relative growth rate was 43% faster and total nonstructural carbohydrate concentration was 41% greater than at low irradiance. The change in carbohydrate with irradiance could explain the growth response. Plant growth was fastest at 25°C and decreased parabolically at lower and higher temperatures with a half-maximal rate at 13 and 36°C. Total nonstructural carbohydrate decreased between 13 and 23°C and remained constant at higher temperatures. Soluble sugar concentrations varied little with temperature above 13°C except for sucrose, whose level rose above 30°C. The change in carbohydrate with temperature could not explain the growth response. Above 23°C tomato plants appeared to regulate growth rate to maintain a relatively constant nonstructural carbohydrate concentration.  相似文献

Whitefish eggs incubated in aerated lake water at controlled tempera tures of 0°, 0.5°, 2°, 4°, 6°, 8°, 10°, and 12°C., failed to hatch at either 0° or 12°C. 0.6 per cent hatched alive at 10°C., 72.67 per cent hatched alive at 0.5°C., and an intermediate proportion hatched at intermediate temperatures. 2. The percentage of abnormal embryos which developed to the hatching stage varied directly with temperature between 4° and 12°, all embryos being abnormal at 12°C.; but none were abnormal at either 0.5°, or 2°C. Normal development predominated from 0.5 to 6°C. The highest proportion of embryos to hatch alive was 72.67 per cent at 0.5°C., which is, hence, the optimum temperature. 3. Total incubation time ranged from 29.6 days at 10°C. to 141 days at 0.5°C. 4. The time (T) required to attain any given stage of development is expressed in equations See PDF for Equation where temperature, t, is a negative exponent of the constant, A, whose value differs above or below 6°C., a critical temperature. Values of A above 6° fluctuate about 1.13; those of A below 6° fluctuate about 1.19 as a mean. 5. Applying Arrhenius'' equation µ values for the total incubation period are 27,500 below 6° and 27,100 above it. 6. The relative magnitude of A values of the exponential equation and µ values of Arrhenius'' equation show corresponding changes from one developmental period to another. 7. When plotted, thermal increments show cyclic variations, with maxima during periods of cleavage and of organogenesis. These may indicate the interaction of two separate sets of embryonic processes, which give a maximal response to temperature differences during these two separate periods. 8. Above 6°, µ values during the hatching process are distinct from those of developmental stages and are regarded as being due to the action of hatching enzymes.  相似文献

A method is given whereby the course of hydrolysis of sucrose by live yeast cells may be followed with precision equal to that found when invertase solutions prepared from autolyzed yeast are used to cause inversion. 2. The practical value of the equation of Nelson and Hitchcock as a means of following the course of enzymic hydrolysis of sucrose is hereby extended. 3. The inversion of sucrose by live yeast cells and by extracted invertase has been quantitatively compared. 4. Comparative studies of the fermentation of cane molasses into ethanol by Saccharomyces cerevisiae in the presence or absence of fungal invertase were performed. When cane molasses was fermented by the yeast at 30°C and pH 5.0, the presence of the enzyme had no effect on ethanol production. At pH 3.5, ethanol production was increased by the addition of invertase. At 40°C, the addition of invertase increased ethanol production by 5.5% at pH 5.0 and by 20.9% at pH 3.5.  相似文献

When wheat (Triticum aestivum) seedlings are exposed to a cold temperature (2-4°C) above 0°C, sucrose accumulates and sucrose synthase activity increases. The effect of a cold period on the level of sucrose synthase (SS) was investigated. Using antibodies against wheat germ SS, Western blots studies showed that the amount of the SS peptide increased during 14 days in the cold, when plants were moved from 23°C to 4°C. The level of SS diminished when plants were moved back to 23°C. Northern blots of poly(A)+ RNA, confirmed a five- to sixfold induction of SS in wheat leaves during cold acclimation. These results indicate that SS is involved in the plant response to a chilling stress.  相似文献

Most invertases identified to date have optimal activity at acidic pH, and are intolerant to neutral or alkaline environments. Here, an acid invertase named uninv2 is described. Uninv2 contained 586 amino acids, with a 100 amino acids N-terminal domain, a catalytic domain and a C-terminal domain. With sucrose as the substrate, uninv2 activity was optimal at pH 4.5 and at 45°C. Removal of N-terminal domain of uninv2 has shifted the optimum pH to 6.0 while retaining its optimum temperaure at 45°C. Both uninv2 and the truncated enzyme retained highly stable at neutral pH at 37°C, and they were stable at their optimum pH at 4°C for as long as 30 days. These characteristics make them far superior to invertase from Saccharomyces cerevisiae, which is mostly used as industrial enzyme.  相似文献

The frequency of pulsation of the heart of Ciona intestinalis increases with temperature in both advisceral and abvisceral direction, according to the Arrhenius equation. The increase in pulsation is the same in both directions. The following µ values were obtained: 8,000–, 12,000+, 16,000, in several combinations, with critical temperatures at 10°, 15°, and 20°C. The values found are comparable with earlier findings for activity of the heart in different animals. This quantitative correspondence suggests anew the conception that temperature characteristics may be employed for recognition of controlling processes. The fact that the µ''s and the critical temperature are the same for advisceral and abvisceral beats, indicates that the general metabolic condition of the two ends of the heart is the same in any one individual.  相似文献

The rate of anaerobic production of CO2 by germinating seeds of Lupinus albus was studied as a function of temperature between 7.5° and 18°C. The mean value for the temperature characteristic was found to be 21,500± calories, which is slightly lower than that for the same process under aerobic conditions (23,500± calories). The values for the individual µ''s in the two cases overlap considerably. The possible identity of the processes underlying the production of CO2 aerobically and anaerobically is discussed.  相似文献

1. Extensive experimental data have been collected on the time required for the excystment process of the small ciliate Colpoda duodenaria throughout a range of temperatures of 8° through 32°C. and a range of concentrations of yeast extract excystment media of 0.08 through 22.4 gm./liter. 2. The excystment process has been separated into two periods, the first inversely proportional to the concentration of the yeast extract and the second independent of its concentration. 3. The first excystment period has been found to depend on the time for diffusion through the protoplasm of a compound from the yeast extract and on the time for a chemical reaction with the extremely high energy of activation of 220,000 calories/mole. 4. The changes in viscosity with temperature for this Colpoda, inferred from diffusion rate changes, have been found to be almost the same as those found by Heilbrunn for Amoeba dubia by the direct method of centrifuging granules. 5. The second excystment period is shown to be controlled by reactions whose apparent activation energies are 44,000 calories/mole below 15°C. and 18,000 calories/mole above 15°C.; above 25°C. this period is independent of temperature. 6. The distribution of the log excystment times of individual organisms about the mean log excystment time is found to be independent of temperature except in the range where the reaction with highest activation energy takes a significant length of time, and to increase rapidly with decreasing temperatures in this range.  相似文献

1. The investigations dealing with the properties of casein as an acid were reviewed. 2. The solubility of uncombined casein in water was measured at 5°C. and found to be 0.70±0.1 mg. of N per 100 gm. of water. 3. Robertson''s solubility measurements of casein in bases at various temperatures were recalculated and found to agree well with more recent measurements. 4. By combining the observations of several investigators, as well as the author''s measurements of the solubility of casein, in base, at various temperatures, the following conclusions were reached: (a) The solubility of casein in base is affected by the temperature in a discontinuous manner. (b) There exist two ranges of temperature, one, extending from about 21° to 37°C. and the other from about 60° to 85°C. where the solubility of casein in base is practically independent of temperature. (c) From 37° to 60° the equivalent combining weight of casein rises from the value 2100 to about 3700 gm. 5. By comparing the values of base bound by 1 gm. of casein at the two temperature ranges with a constant, the value of base necessary to saturate the same amount of casein, it was found that the latter value is a common multiple of the former values, indicating the stoichiometric nature of the effect of temperature.  相似文献

Because of reports of the potential risk of maternal hyperthermia to a developing embryo or fetus, studies were done to determine the length of time a woman must stay in a hot tub or sauna before her temperature reaches 38.9°C. The vaginal temperatures of 20 nonpregnant women of childbearing age were recorded while they sat in hot tubs set at 39.0°C or 41.1°C and in a sauna with an average temperature of 81.4°C. Five women were able to remain in the 39.0°C tub and six in the 41.1°C tub until their temperature reached 38.9°C, but in none did their temperature reach that level before 15 minutes in the 39.0°C tub or 10 minutes in the 41.1°C tub. The remainder left in discomfort while their body temperatures were lower. This indicates that the usual use of hot tubs is unlikely to raise a woman''s body temperature to potentially teratogenic levels, although prolonged use may. None of the women were able to remain in the sauna long enough for their temperature to reach 39.9°C.  相似文献

Fructans are the major storage carbohydrate in vegetative tissues of wheat (Triticum aestivum L.). Fructan:fructan fructosyl transferase (FFT) catalyzes fructosyl transfer between fructan molecules to elongate the fructan chain. The objective of this research was to isolate this activity in wheat. Wheat (cv Caldwell) plants grown at 25°C for 3 weeks were transferred to 10°C to induce fructan synthesis. From the leaf blades kept at 10°C for 4 days, fructosyl transferase activity was purified using salt precipitation and a series of chromatographic procedures including size exclusion, anion-exchange, and affinity chromatography. The transferase activity was free from invertase and other fructan-metabolizing activities. Fructosyl transferase had a broad pH spectrum with a peak activity at 6.5. The temperature optimum was 30°C. The activity was specific for fructosyl transfer from β(2→1)-linked 1-kestose or fructan to sucrose and β(2→1) fructosyl transfer to other fructans (1-FFT). Fructosyl transfer from oligofructans to sucrose was most efficient when 1-kestose was used as donor molecule and declined as the degree of polymerization of the donor increased from 3 to 5. 1-FFT catalyzed the in vitro synthesis of inulin tetra- and penta-saccharides from 1-kestose; however, formation of the tetrasaccharide was greatly reduced at high sucrose concentration. 6-Kestose could not act as donor molecule, but could accept a fructosyl moiety from 1-kestose to produce bifurcose and a tetrasaccharide having a β(2→1) fructose attached to the terminal fructose of 6-kestose. The role of this FFT activity in the synthesis of fructan in wheat is discussed.  相似文献

Fifty bacterial strains able to grow at pH 10 and 0°C were isolated from soils, and growth characteristics of three selected strains were investigated. Strain 207, which showed the best growth rate of all the isolates at the conditions described above, could grow at a temperature of −5 to 39°C at pH 8.5. The optimum pH for this strain changed from 9.5 at 10°C to 9.0 at 20°C.  相似文献

Carbon-starved cultures of strain Ant-300, a psychrophilic marine vibrio isolated from the Antarctic Convergence, were compared with their nonstarved counterparts for resistance to heat. Specifically, starved and unstarved cells were exposed to 17°C, which is 4°C above the maximum growth temperature, and compared with cells maintained at the optimum temperature (5 to 7°C). Total cell counts, direct viable-cell counts, and plate counts were monitored. At a temperature of 17°C, viability (as indicated by plate counts) was lost within 40 h, with direct viable-cell counts indicating less than 5% viability at this time. However, when cells were carbon starved for 1 week prior to heat challenge, significant plateability was maintained for more than 6 days; direct viable-cell counts of starved cells maintained at 17°C indicated the presence of viable cells for at least 12 days. Because starvation is the normal physiological state of copiotrophic, heterotrophic bacteria in oligotrophic marine waters, these data suggest that starvation conditions may be a significant factor in providing heat tolerance to psychrophiles.  相似文献

Candida ingens, a pellicle-forming yeast utilizing volatile fatty acids, grew over a pH range of 4.1 to 6.0 on nonsterile supernatants from anaerobically fermented pig wastes; growth was inconsistent between pH 4.1 and 4.6. When ambient temperature above the pellicle was 21°C and the temperature of the medium was 29 to 32°C, a pH range of 4.8 to 5.0 gave yields of 1.90 to 3.31 g of dry matter per liter, and 0.059 to 0.065 mol of volatile fatty acids was utilized per liter. There was no advantage in utilization of volatile fatty acids and yield of dry matter in keeping the pH constant during a 24-h growth period. C. ingens grew at pH 4.8 and 5.0 when both ambient and medium temperatures were 30°C. When ambient temperature was 10°C, maximum yield and utilization of volatile fatty acids occurred at a medium temperature of 28 to 30°C.  相似文献

This study was designed to compare the uptake and distribution of 14C among fructose, glucose, sucrose, and starch in the cob, pedicel, and endosperm tissues of maize (Zea mays L.) kernels induced to abort by high temperature with those that develop normally. Kernels cultured in vitro at 30 and 35°C were transferred to [14C]sucrose media 10 days after pollination. Kernels cultured at 35°C aborted prior to the onset of linear dry matter accumulation. Significant uptake into the cob, pedicel, and endosperm of radioactivity associated with the soluble and starch fractions of the tissues was detected after 24 hours in culture on labeled media. After 8 days in culture on [14C]sucrose media, 48 and 40% of the radioactivity associated with the cob carbohydrates was found in the reducing sugars at 30 and 35°C, respectively. This indicates that some of the sucrose taken up by the cob tissue was cleaved to fructose and glucose in the cob. Of the total carbohydrates, a higher percentage of label was associated with sucrose and a lower percentage with fructose and glucose in pedicel tissue of kernels cultured at 35°C compared to kernels cultured at 30°C. These results indicate that sucrose was not cleaved to fructose and glucose as rapidly during the unloading process in the pedicel of kernels induced to abort by high temperature. Kernels cultured at 35°C had a much lower proportion of label associated with endosperm starch (29%) than did kernels cultured at 30°C (89%). Kernels cultured at 35°C had a correspondingly higher proportion of 14C in endosperm fructose, glucose, and sucrose. These results indicate that starch synthesis in the endosperm is strongly inhibited in kernels induced to abort by high temperature even though there is an adequate supply of sugar.  相似文献
