Matric bound water was measured as water retained by frozen and thawed tissue after desorption on a pressure membrane filter under 20 bars nitrogen gas pressure. Central water-storage tissue and peripheral chlorenchyma from leaves or stems of 15 taxonomically diverse non-halophytic succulent species were investigated. Matric bound water as a per cent of the dry weight averaged higher in water storage than in chlorenchyma tissue but lower than values reported for many mesophytic leaves. Matric bound water as a proportion of the total water held, however, was lower in water tissues. Osmotic potentials were generally high (solute contents low). It is concluded that matric or osmotic forces cannot account, in any unique way, for the high water content of water tissues. This appears to depend, instead, on the enormous ability of the thin-walled cells to take up available water and expand.

A thermocouple psychrometer method, previously described foruse in determining tissue and sap water potentials, has beenadapted for determining matric potentials. Matric values ranging from approximately zero to –10 x105 Pa were observed with wilting wheat leaves. Mean valuesof total tissue water potential and of its solute (osmotic),matric, and cell wall pressure components were –16.6,–18.3, –30 and +4.7 Pa x 105, respectively. Matric potentials are often ignored in investigations of plantwater relations. This practice is shown to result in underestimationof wall pressures (in this case by a mean of 64 per cent) andsometimes in spuriously negative values.

Seedling Traits Determine Drought Tolerance of Tropical Tree Species
Water availability is the most important factor determining tree species distribution in the tropics, but the underlying mechanisms are still not clear. In this study, we compared functional traits of 38 tropical tree species from dry and moist forest, and quantified their ability to survive drought in a dry‐down experiment in which wilting and survival were monitored. We evaluated how seedling traits affect drought survival, and how drought survival determines species distribution along the rainfall gradient. Dry forest species tended to have compound leaves, high stem dry matter content (stem dry mass/fresh mass), and low leaf area ratio, suggesting that reduction of transpiration and avoidance of xylem cavitation are important for their success. Three functional groups were identified based on the seedling traits: (1) drought avoiders with a deciduous leaf habitat and taproots; (2) drought resisters with tough tissues (i.e., a high dry matter content); and (3) light‐demanding moist forest species with a large belowground foraging capacity. Dry forest species had a longer drought survival time (62 d) than moist forest species (25 d). Deciduousness explained 69 percent of interspecific variation in drought survival. Among evergreen species, stem density explained 20 percent of the drought survival. Drought survival was not related to species distribution along the rainfall gradient, because it was mainly determined by deciduousness, and species with deciduous seedlings are found in both dry and moist forests. Among evergreen species, drought survival explained 28 percent of the variation in species position along the rainfall gradient. This suggests that, apart from drought tolerance, other factors such as history, dispersal limitation, shade tolerance, and fire shape species distribution patterns along the rainfall gradient.

Maize (Zea mays L.) hybrids varying in drought tolerance were treated with water stress in controlled environments. Experiments were performed during vegetative growth and water was withheld for 19 days beginning 17 days after sowing. Genotypic comparisons used measured changes of leaf water potential or results were expressed by time of treatment. Total dry matter of the drought tolerant hybrid on the final harvest was 53% less than that of the intermediate and susceptible maize hybrids when plants were water sufficient. This showed that maize hybrids selected for extreme drought tolerance possessed a dwarf phenotype that affected soil water contents and leaf water potentials. Changes of shoot and root growth, leaf water potential, net photosynthesis and stomatal conductance in response to the time of water stress treatment were diminished when comparing the drought tolerant to the intermediate or susceptible maize hybrids. Genotypic differences were observed in 26 of 40 total foliar metabolites during water stress treatments. Hierarchical clustering revealed that the tolerant maize hybrid initiated the accumulation of stress related metabolites at higher leaf water potentials than either the susceptible or intermediate hybrids. Opposite results occurred when changes of metabolites in maize leaves were expressed temporally. The above results demonstrated that genotypic differences were readily observed by comparing maize hybrids differing in drought tolerance based on either time of treatment or measured leaf water potential. Current findings provided new and potentially important insights into the mechanisms of drought tolerance in maize.

The influence of arbuscular mycorrhizal fungi on drought tolerance and recovery was studied in safflower (Carthamus tinctorius L.) and wheat (Triticum aestivum L.). Plants were grown with and without the mycorrhizal fungus, Glomus etunicatum Becker & Gerd., in nutrient-amended soil under environmentally-controlled conditions to yield mycorrhizal and nonmycorrhizal with similar leaf areas, root length densities, dry weights, and adequate tissue phosphorus. When drought stress was induced, mycorrhizal infection did not affect changes in leaf water, osmotic or pressure potentials, or osmotic potentials of leaf tissue rehydrated to full turgor in either safflower or wheat. Furthermore, in safflower, infection had little effect on drought tolerance as indicated by the level of leaf necrosis. Mycorrhizal wheat plants, however, had less necrotic leaf tissue than uninfected plants at moderate levels of drought stress (but not at severe levels) probably due to enhanced phosphorus nutrition. To determine the effects of infection on drought recovery, plants were rewatered at a range of soil water potentials from –1 to –4 MPa. We found that although safflower tended to recover more slowly from drought after rewatering than wheat, mycorrhizal infection did not directly affect drought recovery in either plant species. Daily water use after rewatering was reduced and was correlated to the extent that leaves were damaged by drought stress in both plant species, but was not directly influenced by the mycorrhizal status of the plants.

A pressure chamber was used to measure matric potentials of frozen and thawed leaves. Significant matric potentials were demonstrated in sunflower (Helianthus annuus L.), yew (Taxus cuspidata Sieb. and Zucc.), and rhododendron (Rhododendron roseum Rehd.). Matric potentials were particularly negative in rhododendron and were correlated with the amount of cell wall present and with the volume of water outside the leaf protoplasts at comparable matric potentials. It was concluded that matric forces in leaves are associated mainly with cell walls, at least within the physiological range of water contents. Calculations indicated that the water potential of the solution in the cell wall could be estimated for living tissue from the sum of matric and osmotic potentials acting on water outside the protoplasts.

The effects of topographic position on water regime in a semideciduous tropical forest on Barro Colorado Island in Panamá were assessed by measuring soil matric potential using the filter paper technique and by using measured soil water release characteristics to convert a long-term (20 years) gravimetric water content data-set to matric potential. These were also compared against predictions from a simple water balance model. Soil matric potentials on slope sites were significantly higher than on plateau sites throughout the measurement interval and slopes experienced a shorter duration of drought during the annual dry-season. Measured values of matric potential agreed with those predicted from converting the gravimetric measurements using water release characteristics. Annual duration of drought predicted by the simple water balance model agreed with values determined from the converted long term water content data-set and was able to predict the annual duration of drought on plateau sites. On slope sites, the water balance systematically and significantly overestimated the duration of drought obtained from the water content data-set, suggesting that slope sites were supplied with water from upslope. Predictions of annual drought duration from sites with higher annual rainfall than Barro Colorado Island (BCI), suggest that while plateau sites on BCI experience a water regime consistent with annual rainfall, slopes experience a water regime more similar to that of forests with much higher rainfall. We conclude that such large variations in water regime over small spatial scales may play a role in maintaining high species richness through providing opportunities for niche specialisation and by buffering slopes against possible climate change.

A technique is described to induce slowly increasing water deficitsin wheat (Triticum aestivum L.) seedlings by rooting them indamp vermiculite and growing them in a humid atmosphere. Aftereight weeks, drought-resistant genotypes can be identified eitherby drought avoidance or by drought tolerance responses. Genotypeswith lesser drought resistance become dormant or produce sterilespikes. The test roughly predicts the relative yield performanceof cultivars in dry field conditions. Cell membrane stability (CMS) technique was used to screen for drought tolerance, salt tolerant accessions of three Aegilops species, Ae. tauschii, Ae. cylindrica, Ae. geniculata and two hexaploid wheat (Tricitum aestivum L.) cultivars comprising salt tolerant LU-26 and drought tolerant Chakwal-86. The objectives were to see how valid it is for a salt tolerant plant to be drought tolerant as well and to identify the character(s) that may contribute to drought tolerance. Three moisture levels equal to 100, 50 and 25% saturation capacity of the soil were used for plant cultivation. Injury percentage (IP) based on in-vitro desiccation induced by polyethylene glycol (PEG) in leaf tissue was measured through the conductivity of the electrolyte leakage. Injury percentage decreased in all the test material with decrease in soil moisture contents. Ae. cylindrica exhibited minimum injury at 100% soil moisture level followed by Ae. tauschii and Ae. geniculata while drought tolerant wheat cultivars exhibited the maximum. The wheat cultivar Chakwal-86 has been developed for dry areas, with low soil moisture levels, and high water potential enhances the injury percentage. Aegilops cylindrica is a salt tolerant species and can thus tolerate water deficit conditions created due to low osmotic potential. Potassium appeared to play an important role in drought tolerance which was evident from high K+ contents and low K+ leakage from Aegilops cylindrica and drought tolerant wheat cultivar Chakwal-86. It was inferred from the study that salt tolerant species might prove drought tolerant in the areas where water deficit prevails due to the ability to create low intracellular osmotic potentials.

A greenhouse experiment was conducted to assess the effect of water stress on growth and metabolic changes in nodules and other plant parts of two leguminous species, Phaseolus vulgaris and Sesbania aculeata, with the major objective that nodules play a vital role in drought tolerance. Imposition of water deficit conditions for 45 days to 15-day-old plants of P. vulgaris and S. aculeata reduced shoot mass and nodule mass of both species, but the reduction was more pronounced in P. vulgaris than in S. aculeata. Nitrate reductase (NR) activity was reduced more in the leaves and nodules of P. vulgaris than in S. aculeata. Soluble proteins in the nodules of S. aculeata were more decreased as compared to that in P. vulgaris. Free amino acids increased in all parts of both species due to water deficit, but a higher increase was observed in leaf and nodules of P. vulgaris than in S. aculeata. Osmoprotectants such as proline and glycine betaine increased more in the nodules and other parts of S. aculeata under drought stress. In conclusion, S. aculeata (salt tolerant) showed a higher degree of drought tolerance than P. vulgaris (salt sensitive). Drought tolerance of S. aculeata was found to be associated with a smaller reduction in number and mass of root nodules, a high activity of nitrate reductase in leaves and nodules, high accumulation of free proline in roots and nodules, and high glycine betaine content in nodules.

Poor crop stand is a common problem in saline areas. Germination and seedling emergence may be depressed as a result of impeded aeration, saline or dry conditions. In this study, we examined the effects of salinity and moisture stress and their interactions on seed germination and seedling growth of carrots. Variable soil matric and osmotic potentials were either obtained by equilibrating soil salinized to different degrees on a 0.5 MPa ceramic plate soil moisture extractor or by adding different amounts of salt solutions to the same mass of air-dried soil, based on a previously determined soil moisture release curve, and allowing to equilibrate for 1 week. Germination decreased significantly in the investigated silty soil (Aquic Ustifluvent) at soil moisture potentials higher than −0.01 MPa, whereas osmotic potentials as low as −0.5 MPa did not influence germination. Matric potentials of −0.3 and −0.4 MPa, respectively, resulted in a strong decrease (35–95%) of germination and delayed germination by 2 to 5 days in the silty soil to which different amounts (18 and 36%, respectively) and sizes (0.8–1.2 mm and 1.5–2.2 mm, respectively) of sand particles had been added. No effect of sand and grain diameter was detected. Germination was not affected by comparable osmotic potentials. Seedling growth showed a much higher sensitivity than germination to decreasing matric potentials, but was not affected by osmotic potentials ranging from −0.05 to −0.5 MPa. Optimum shoot growth occurred at matric potentials between −0.025 and −0.1 MPa. Shoot and root growth decreased markedly at matric potentials higher than −0.01 MPa. Fresh weight of shoots decreased gradually at matric potentials lower than −0.2 MPa. Root growth was significantly increased at matric potentials of −0.1 to −0.3 MPa, whereas comparable osmotic potentials did not have equivalent effects. It is concluded that germination and seedling growth are differently affected by comparable matric and osmotic stresses and that water stress exerts a more negative effect than salt stress.

A common observation in tropical dry forests is the habitat preference of tree species along spatial soil water gradients. This pattern of habitat partitioning might be a result of species differentiation in their strategy for using water, along with competing functions such as maximizing water exploitation and tolerating soil water stress. We tested whether species from drier soil conditions exhibited a tolerance strategy compared with that of wet-habitat species. In a comparison of 12 morphophysiological traits in seedlings of 10 closely related dry and wet-habitat species pairs, we explored what trade-offs guide differentiation between habitats and species. Contrary to our expectations, dry-habitat species showed mostly traits associated with an exploitation strategy (higher carbon assimilation capacity, specific leaf area and leaf-specific conductivity and lower water-use efficiency). Strikingly, dry-habitat species tended to retain their leaves longer during drought. Additionally, we detected multiple strategies to live within each habitat, in part due to variation of strategies among lineages, as well as functional differentiation along the water storage capacity-stem density (xylem safety) trade-off. Our results suggest that fundamental trade-offs guide functional niche differentiation among tree species expressed both within and between soil water habitats in a tropical dry forest.

It has been hypothesized that plants cannot tolerate combined shade and drought, as a result of morphological trade-offs. However, numerous plant species are reportedly widespread in shaded forest understories that face drought, whether seasonal or occasional. We studied juveniles of six plant species that cope with strong summer drought in the understoreys of mixed Quercus forests in southern Spain: the tall-shrubs Phillyrea latifolia and Viburnum tinus, the perennial herb Rubia peregrina, the small shrub Ruscus aculeatus, and climbers Hedera helix and Smilax aspera. All of these species persist in evergreen shade (c. 3% daylight). Two other species were studied as comparators, Ruscus hypoglossum, less tolerant of drought, and Ceratonia siliqua, less tolerant of shade. Morphological and chemical variables relevant to shade and drought tolerance were measured for juveniles in a range of sizes, and also for the leaves of mature plants. The species converge in features that confer tolerance of shade plus drought by reducing demand for resources. Demand for water is reduced through a moderate to high below-ground mass fraction and low to moderate specific leaf area (respectively 0.22–0.52 and 112–172 cm2 g–1 at 1.00 g total dry mass). Demand for both irradiance and water is reduced through a low to moderate foliar nitrogen concentration and long-lived, physically protected leaves (2 yr). The species also converge in features that confer tolerance of either low irradiance or drought through specialized capture of resource, without precluding the other tolerance. These features include deep roots relative to shoot size, moderately higher specific leaf area in shade (1.2–2.0 × that in sun) and higher chlorophyll:nitrogen ratio in shade. Foliar chlorophyll per unit mass was higher in shade, but chlorophyll was not necessarily synthesized in greater amounts; rather, it was higher apparently due to shade effects on structural features linked with specific leaf area. In contrast, N per unit mass was higher in sun leaves independently of specific leaf area. Despite these convergences, the species diverge considerably in their root mass allocation and architecture, leaf saturated water content, density of stomata and guard cell size. No single narrowly defined functional type is needed for tolerance of shade plus drought.

In environments where light is not a limiting resource such as rangelands and grasslands, there is much disagreement regarding the benefits provided by rapid light capture during the growing season and the species’ ability to withstand drought during the dry period. In this study, we selected four perennial herbaceous species with contrasting resource-use strategies (acquisitive versus conservative), which were transplanted as monocultures into PVC pots to evaluate their species-specific responses to drought. The two main strategies of drought-survival (avoidance versus tolerance) were driven by distinct underlying mechanisms that allow the plant to delay or tolerate water deficit in leaves. On the one hand, plants that produced reduced leaves with lower surface area:mass ratio (lower SLA) exhibited higher values of leaf water potential (LWP) and leaf relative water content (LRWC), which could be associated to a higher ability to delay tissue dehydration in enlarged leaves. Regarding the below-ground compartment, dehydration avoidance was promoted by prolonged elongation rates of thinner roots that allow the plant to increase water uptake and accessibility during the dry period. On the other hand, dehydration tolerance was positively related with progressive foliage senescence under water deficit, which probably favored a longer survival of meristematic basal tissues. The results presented in this study suggest the existence of a trade-off between the traits favoring rapid light-acquisition and those enhancing the ability to delay leaf dehydration. Thus, the species related most closely with a resource-acquisition strategy (Bromus erectus and Potentilla neumanniana) could be considered less efficient to delay leaf dehydration than the others (Carex humilis and Festuca christiani-bernardii), as indicated by their lower values of leaf water potential (LWP) and leaf relative water content (LRWC) under identical conditions of water deficit. Our findings support evidence that there is not a single strategy to effectively cope with drought and reveal the diversity of adaptive mechanisms among coexisting species.

Plant hydraulic architecture has been studied extensively, yet we know little about how hydraulic properties relate to species' life history strategies, such as drought and shade tolerance. The prevailing theories seem contradictory. We measured the sapwood (K(s) ) and leaf (K(l) ) hydraulic conductivities of 40 coexisting tree species in a Bolivian dry forest, and examined associations with functional stem and leaf traits and indices of species' drought (dry-season leaf water potential) and shade (juvenile crown exposure) tolerance. Hydraulic properties varied across species and between life-history groups (pioneers vs shade-tolerant, and deciduous vs evergreen species). In addition to the expected negative correlation of K(l) with drought tolerance, we found a strong, negative correlation between K(l) and species' shade tolerance. Across species, K(s) and K(l) were negatively correlated with wood density and positively with maximum vessel length. Consequently, drought and shade tolerance scaled similarly with hydraulic properties, wood density and leaf dry matter content. We found that deciduous species also had traits conferring efficient water transport relative to evergreen species. Hydraulic properties varied across species, corresponding to the classical trade-off between hydraulic efficiency and safety, which for these dry forest trees resulted in coordinated drought and shade tolerance across species rather than the frequently hypothesized trade-off.

A better understanding of plant water relations is needed for evaluating the suitability of plant species to site-specific reforestation programs in the heterogeneous karst landscapes in southwestern China that are characterized by temporary water deficit. During both wet and dry periods, leaf water potentials of 65 plant species from five different growth forms were studied at three representative sites (forest, shrubland and grassland), to compare their adaptive strategies against water stress and assess their suitability for reforestation programs. Herbs showed the highest predawn and midday water potentials and smallest diurnal ranges of water potential values at all the three sites, indicating that they follow water stress avoidance strategies. During the dry period, evergreen shrubs showed low water potentials, the largest diurnal ranges and highest soluble sugar contents. This indicates that they have a tolerance strategy responding to water stress. Deciduous shrubs and trees still showed relatively large diurnal ranges of water potential values and high soluble sugar contents, and did not shed leaves when experiencing the lowest midday water potentials during the dry period. They shed leaves only later in the dry winter period when even more serious drought was experienced. Their strategies seem to include both tolerance and avoidance mechanisms. Evergreen trees revealed relatively low water potentials with smallest diurnal range water potentials at the shrubland site, especially during the dry period, which indicated their weak ability to tolerate severe water stress. Increasing degradation of the vegetation clearly impacts negatively plant water relations. Using the ranges of leaf water potentials, the relative suitability of the plants for reforestation could be evaluated.

Plants overcome water deficit conditions by combining molecular, biochemical and morphological changes. At the molecular level, many stress-responsive genes have been isolated, but knowledge of their physiological functions remains fragmentary. Here, we report data for RD20, a stress-inducible Arabidopsis gene that belongs to the caleosin family. As for other caleosins, we showed that RD20 localized to oil bodies. Although caleosins are thought to play a role in the degradation of lipids during seed germination, induction of RD20 by dehydration, salt stress and ABA suggests that RD20 might be involved in processes other than germination. Using plants carrying the promoter RD20::uidA construct, we show that RD20 is expressed in leaves, guard cells and flowers, but not in root or in mature seeds. Water deficit triggers a transient increase in RD20 expression in leaves that appeared predominantly dependent on ABA signaling. To assess the biological significance of these data, a functional analysis using rd20 knock-out and overexpressing complemented lines cultivated either in standard or in water deficit conditions was performed. The rd20 knock-out plants present a higher transpiration rate that correlates with enhanced stomatal opening and a reduced tolerance to drought as compared with the wild type. These results support a role for RD20 in drought tolerance through stomatal control under water deficit conditions.

Leaf micromorphological traits and some physiological parameters with potential relevance to drought tolerance mechanisms were investigated in four selected winter wheat varieties. Plants were subjected to two cycles of drought treatment at anthesis. Yield components confirmed contrasting drought-sensitive and -tolerant behavior of the genotypes. Drought tolerance was associated with small flag leaf surfaces and less frequent occurrence of stomata. Substantial variation of leaf cuticular thickness was found among the cultivars. Thin cuticle coincided with drought sensitivity and correlated with a high rate of dark-adapted water loss from leaves. Unlike in Arabidopsis, thickening of the cuticular matrix in response to water deprivation did not occur. Water stress induced epicuticular wax crystal depositions preferentially on the abaxial leaf surfaces. According to microscopy and electrolyte leakage measurements from leaf tissues, membrane integrity was lost earlier or to a higher extent in sensitive than in tolerant genotypes. Cellular damage and a decline of relative water content of leaves in sensitive cultivars became distinctive during the second cycle of water deprivation. Our results indicate strong variation of traits with potential contribution to the complex phenotype of drought tolerance in wheat genotypes. The maintained membrane integrity and relative water content values during repeated water limited periods were found to correlate with drought tolerance in the selection of cultivars investigated.
