A technique of repeated differential centrifugation for the extraction of microorganisms from soil has been adapted for use with grass silage and compared with the classical stomaching technique. Differential centrifugation proved much more efficient as determined by viable counting procedures. The actual recoveries of cells added to silage were 109 and 9% for the differential centrifugation and stomaching techniques, respectivly  相似文献

Simultaneous and independent release of vasopressin and oxytocin in the rat   总被引:3,自引:0,他引:3  
The relative dependence or independence of the secretion of the neurohypophysial hormones, arginine vasopressin and oxytocin, was investigated using a wide variety of stimuli reported to cause the secretion of one or the other hormone. Differences in species, animal preparations, sampling techniques, assays, and other factors make comparison of many previous studies difficult. The aim of this study was to overcome these problems by using the same methodology, animal species, and assays to compare vasopressin and oxytocin release. To further strengthen the analysis, determinations of vasopressin and oxytocin were done in the same blood samples. The results demonstrated that during simultaneous release of both hormones, vasopressin is released in greater proportion following restraint stress, hemorrhage, isotonic hypovolemia, and nicotine, whereas oxytocin is released in greater proportion following endotoxin or hypertonic saline. Vasopressin was released without oxytocin following diethylstilbestrol. Oxytocin was released without concomitant vasopressin release following exercise, hypothermia, hyperthermia, labour, and lactation. Neither oxytocin nor vasopressin release was observed following thyroid-releasing hormone or insulin-induced hypoglycemia. These data illustrate the marked flexibility of the hypothalamo-neurohypophysial system that regulates secretion of vasopressin and oxytocin.  相似文献

We have investigated the effect of increasing concentrations of methylamine (5, 10, and 25 mM) on the removal of iron from the two iron-binding sites of transferrin during endocytosis by human erythroleukemia (K562) cells. The molecular forms of transferrin released from the cells were analyzed by polyacrylamide gel electrophoresis in 6 M urea. Endocytosis of diferric transferrin was efficient since greater than 10% of surface-bound protein escaped endocytosis and was released in the diferric form. Although transferrin exocytosed from control cells had been depleted of 80% of its iron and contained 65-70% apotransferrin, iron-bearing species were also released (15% C-terminal monoferric; 10% N-terminal; 10% diferric). The ratio of the two monoferric species (C/N) was 1.32 +/- 0.12 (mean +/- SD; n = 4), suggesting that iron in the N-terminal site was more accessible to cells. In the presence of methylamine there was a concentration-dependent increase in the proportion of diferric transferrin release (less than 80% at 25 mM) and a concomitant decrease in apotransferrin. Small amounts of the iron-depleted species, especially apotransferrin, appeared before diferric transferrin, suggesting that these were preferentially released from the cells. The discrepancy between the proportions of the monoferric transferrin species noted with control cells was enhanced at all concentrations of methylamine, most markedly at 10 mM when the C/N ratio was 2.4. The N-terminal site of transferrin loses its iron at a higher pH than the C-terminal site, and so by progressively perturbing the pH of the endocytic vesicle we have increased the difference between the two sites observed with control cells.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

A comparison was made between blending in 2% sodium citrate and stomaching in 0.1% peptone or 0.1% peptone - 1% Tween 80 for the enumeration of Escherichia coli in naturally contaminated cheeses. Statistical analysis of the results from 25 samples of cheese showed that there were no significant differences in recovery by the three methods at the 95% confidence level.  相似文献

Purified phosphatidylinositol-specific phospholipase C from Staphylococcus aureus released a substantial proportion of the total alkaline phosphatase activity from a wide range of tissues from several mammalian species. Co-purification of the phospholipase C and alkaline phosphatase-releasing activities and the inhibition of both these activities by iso-osmotic salt solutions suggested that the releasing effect was unlikely to be due to a contaminant.  相似文献

During C4 photosynthesis, CO2 is released in bundle-sheath cells by decarboxylation of C4 acids and then refixed via ribulose-1,5-bisphosphate carboxylase. In this study we examined the efficiency of this process by determining the proportion of the released CO2 that diffuses back to mesophyll cells instead of being refixed. This leak of CO2 was assessed by determining the amount of 14CO2 released from leaves during a chase in high [12CO2] following a 70-s pulse in 14CO2. A computer-based analysis of the time-course curve for 14CO2 release indicated a first-order process and provided an estimate of the initial velocity of 14CO2 release from leaves. From this value and the net rate of photosynthesis determined from the 14CO2 fixed in the pulse, the CO2 leak rate from bundle-sheath cells (expressed as a percentage of the rate of CO2 production from C4 acids) could be deduced. For nine species of Gramineae representing the different subgroups of C4 plants and two NAD-malic enzyme-type dicotyledonous species, the CO2 leak ranged between 8 and 14%. However, very high CO2 leak rates (averaging about 27%) were recorded for two NADP-malic enzyme-type dicotyledonous species of Flaveria. The results are discussed in terms of the efficiency of C4 photosynthesis and observed quantum yields.  相似文献

The occurrence, size and maturity changes of Eubothrium fragile have been studied in postlarvae, juveniles and adult twaìte shad, Alosa fallax , from several locations in the River Severn. Parasites were only found in adult shad and not in post-larvae or juveniles. No juvenile or recently acquired cestodes were identified as such, but adults were present in shad throughout the whole period of their spawning migration. A large proportion of the parasites were gravid upon arrival in the river and, although eggs were subsequently released into fresh water, there was no loss of cestodes from the fish. It was concluded that E. fragile is a marine species, that the parasites found in adult shad in fresh water were the residue of a marine life cycle and that the eggs released in fresh water were part of the parasite's natural reproductive wastage. The distribution and biology of E. fragile were discussed and it is considered that it is typical of the marine species of the genus.  相似文献

The proportions of Type I and Type III collagen were evaluated from gestational, postpartum-retained, and released bovine placental membranes. Placentomes were excised at 90, 150, 210, and 270 days of gestation (n = 32) and from postpartum-retained (2 and 12 h, n = 8) and released (2 h, n = 4) membranes. Placentome components were processed for collagen, hydroxyproline, protein, and dry weight determination. Collagen extracts were separated by SDS-PAGE. Densitometry was used to establish the proportions of collagen alpha chains (Type I = 2 alpha 1 + 1 alpha 2; Type III = 3 alpha 1). No difference in the proportion of maternal caruncular Type I and Type III collagen was found. The proportion of Type I fetal cotyledonary collagen was lowest (p less than 0.05) at Day 90 of gestation but did not differ between Days 150, 210, 270, or between retained and released fetal membranes. The proportion of Type III fetal cotyledonary collagen was greatest (p less than 0.05) at Day 90. Retained fetal cotyledons had a greater (p less than 0.05) proportion of Type III collagen than did released fetal cotyledons. Therefore, although hydroxyproline content was not different between retained and released fetal membranes, the retained bovine fetal cotyledon was characterized by disproportionate amounts of Type III collagen as compared to the fetal cotyledon that was not retained.  相似文献

In many species, males can rapidly adjust their ejaculate performance in response to changing levels of sperm competition, an ability that is probably mediated by seminal fluid adaptive plasticity. In the black goby, Gobius niger, territorial males attach viscous ejaculate trails to the nest roof, from which sperm are slowly released into the water during the long-lasting spawning events. Sneaker males release their sperm in the vicinity of the nest, and territorial males try to keep them at a distance by patrolling their territory. We show here that territorial males'' ejaculate trails released a higher proportion of their sperm in the presence of a single sneaker, but this proportion decreased when there were three sneakers, an effect that is most likely mediated by a change in the seminal fluid composition. Field observations showed that when multiple sneaking attempts occurred, territorial males spent more time outside the nest, suggesting that ejaculation rate and territory defence are traded-off. Altogether, these results suggest that the adjustment of sperm release from the ejaculate may be strategic, guaranteeing a more continuous concentration of the territorial male''s sperm in the nest, although at a lower level, when he is engaged in prolonged territory defence outside the nest.  相似文献

In a mixed forest in Hong Kong, the foraging ecology of nine species of insectivorous birds was studied. Leaves and branches of diameters smaller than 2 cm were the most frequently searched microhabitats. Gleaning was the most frequently used foraging method. Apart from Blue-winged Minla and Japanese White-eye, no two species used similar proportions of vertical strata and microhabitats at the same time. Bird species using similar proportion of microhabitats were foraging in different proportion of vertical strata. This niche segregation enabled the bird species to coexist in the same habitat. Velvet-fronted Nuthatch differed from other species by its more frequent use of branches of diameters larger than 2 cm and tree trunks. This might be one of the reasons why this exotic species successfully established a breeding population in the study area.  相似文献

Aims Salt stress resulting from soil salinization is one of the driving forces of the land degradation throughout the world. The modern Yellow River delta is one of the most saline areas in China. Phytoremediation can be an effective way to restore the salinized ecosystems, which requires selecting appropriate plant species. This study explored the germination responses of common plant species from contrasting habitats in the Yellow River delta to varying salinity, offering experimental information for ecosystem restoration in the Yellow River delta.Methods In this study, 15 common plant species from the Yellow River delta were divided into two groups (high-salinity and low-salinity groups) by their natural habitats using Canonical Correlation Analysis. Seeds of each species were treated with five salinity levels (0, 5, 10, 20 and 30 ppt), using a randomized complete block design, and germinated seeds were counted and removed daily for 28 days to calculate the final germination proportion and mean time to germination. The germination responses of seeds to salinity treatments were compared between the two groups.Important findings In relation to salinity, seed germination behavior of the test species was closely related to the salinity level of the habitats over which they were distributed. Species from the habitats with higher salinity had generally higher final germination proportion but shorter mean time to germination than those from the habitats with lower salinity in all of five salinity treatments used. The final germination proportion and mean time to germination of low-salinity group species were more sensitive to salinity than those of high-salinity group species. Selecting the species with high final germination proportion and short mean time to germination is important for restoration of salinized land.  相似文献

The rodent neural cell adhesion molecule (NCAM) consists of three glycoproteins with Mr of 180,000, 140,000 and 120,000. The Mr 120,000 protein (NCAM-120) has been shown to exist in membrane-bound and soluble forms but the nature of its membrane association and release has remained obscure. We show here that phosphatidylinositol-specific phospholipase C (PI-PLC), but not a phospholipase C of different specificity, releases a substantial proportion of NCAM-120 from brain membranes and solubilizes almost quantitatively NCAM-120 present at the surface of C6 astroglial cells. The PI-PLC effect was highly selective since only one other protein species was detectably released from C6 cells. These results suggest that NCAM-120 is held in the membrane by covalently bound phosphatidylinositol or a closely related lipid in a way similar to several other surface proteins from eukaryotic cells. The presence of NCAM in a form which can be released from the cell surface by a highly selective mechanism raises additional possibilities for modulation and control of cell--cell adhesion.  相似文献

Limited digestion of trout testis nuclei with DNase I selectively degrades the protamine genes. Concomitant with the degradation of transcribed DNA sequences a series of chromosomal proteins are released; among these, the major species corresponds to the high mobility group protein H6. The amounts of H6 released from chromatin by limited DNase I action and that in the residual nuclear pellet have been determined. The species composition of the polychaetes derived from ichthyological and Sigsbee trawls in the Pechora and Kara seas in 2012 was studied and compared with the grab survey data of 1993–1995. The distribution of the large sabellidae, nektobenthic, and bathypelagic species that are poorly caught by a grab has been determined for the first time. Changes that were observed in the biogeographical polychaete structure in the Kara Sea (a higher proportion of the boreal species and a lower proportion of the Arctic species) may reflect a response of zoobenthos to the Arctic warming in the late 20th and early 21st centuries.  相似文献

1. The leaching rates of filterable reactive phosphorus (FRP) and dissolved organic carbon (DOC) from five leaf litter types commonly occurring in urban environments in Mediterranean regions of Southern Australia were compared. The relative composition, bioavailability and oxygen demand of this DOC were also assessed. Four tree species were assessed, including the native river red gum (Eucalyptus camaldulensis) and three introduced deciduous species, the English elm (Ulmus procera), London plane (Platanus acerifolia) and white poplar (Populus alba). Grass cuttings (mixed species) were selected as a common garden waste. 2. Except for English elm, the majority of FRP and DOC was released within the first 48 h. Grass cuttings released the highest amount of FRP with white poplar releasing the most DOC. Species that released relatively high amounts of DOC (white poplar, English elm, river red gum) released relatively low amounts of FRP. Conversely, species that released relatively low amounts of DOC (grass cuttings and London plane) tended to release relatively high amounts of FRP. 3. Analysis of DOC composition, combined with the differing oxygen demand and DOC depletion curves, demonstrated that there were substantial differences in the DOC leached from the leaf litter of the different species. Biochemical oxygen demand and the biodegradability of the DOC was positively correlated with the proportion of hydrophilic and hydrophobic acids present in the leachate. 4. These results demonstrate that simple measurements of nutrient release per gram of leaf litter would be insufficient to predict the ecological impact on receiving waters resulting from changes in dominant vegetation. Furthermore, the use of traps to prevent particulate leaf material from entering streams may have limited potential for reducing the load of dissolved nutrients. We conclude that any changes to vegetation type which substantially alter the timing of leaf fall or the composition of leaf leachates should be avoided.  相似文献

The oligosaccharide moiety of cell-surface mannose-labelled glycopeptides from a normal (WI38) and an SV40 transformed cell (W118Va) have been investigated using specific glycosidases. Partially purified mannose-containing glycopeptides were separated into acidic and neutral species by high voltage paper electrophoresis. Endo-beta-N-acetylgucosaminidase D, in the presence of three exoglycosidases, released from the acidic glycopeptides of non-growing cells a product completely absent in growing cells. However, the acidic species from growing WI18 Va and WI38 were found to be similar in the products released by enzyme digestion. The neutral species from growing normal cells contained a proportion of the glycopeptides resistant to endoglycosidase D while those from the non-growing cells were almost free of these resistant species. The SV40 transformed cells were further enriched, when compared to normal cells (WI38), in these neutral resistant species. We suggest that the oligomannosyl core of the majority of the susceptible species contains three mannose residues while that of the resistant species contains between six and eight.  相似文献

We measured production of reactive oxygen species by intact mitochondria from rat skeletal muscle, heart, and liver under various experimental conditions. By using different substrates and inhibitors, we determined the sites of production (which complexes in the electron transport chain produced superoxide). By measuring hydrogen peroxide production in the absence and presence of exogenous superoxide dismutase, we established the topology of superoxide production (on which side of the mitochondrial inner membrane superoxide was produced). Mitochondria did not release measurable amounts of superoxide or hydrogen peroxide when respiring on complex I or complex II substrates. Mitochondria from skeletal muscle or heart generated significant amounts of superoxide from complex I when respiring on palmitoyl carnitine. They produced superoxide at considerable rates in the presence of various inhibitors of the electron transport chain. Complex I (and perhaps the fatty acid oxidation electron transfer flavoprotein and its oxidoreductase) released superoxide on the matrix side of the inner membrane, whereas center o of complex III released superoxide on the cytoplasmic side. These results do not support the idea that mitochondria produce considerable amounts of reactive oxygen species under physiological conditions. Our upper estimate of the proportion of electron flow giving rise to hydrogen peroxide with palmitoyl carnitine as substrate (0.15%) is more than an order of magnitude lower than commonly cited values. We observed no difference in the rate of hydrogen peroxide production between rat and pigeon heart mitochondria respiring on complex I substrates. However, when complex I was fully reduced using rotenone, rat mitochondria released significantly more hydrogen peroxide than pigeon mitochondria. This difference was solely due to an elevated concentration of complex I in rat compared with pigeon heart mitochondria.  相似文献

The frequency of cyanogenesis of 48 species of vascular plants was examined by testing 30 individuals from five populations of each species for release of cyanide. The rate at which cyanide was released and the amount of cyanide released varied widely among individuals of a population and among populations of a species. For many taxa, the frequency of cyanogenesis was highly variable among populations. Of the species examined, 20 have not been reported previously as being cyanogenic.  相似文献
