Stem cells are the only proliferating cells in flatworms and can be eliminated by irradiation with no damage to differentiated cells. We investigated the effect of fractionated irradiation schemes on Macrostomum lignano, namely, on survival, gene expression, morphology and regeneration. Proliferating cells were almost undetectable during the first week post-treatment. Cell proliferation and gene expression were restored within 1 month in a dose-dependent manner following exposure to up to 150 Gy irradiation. During recovery, stem cells did not cross the midline but were restricted within lateral compartments. An accumulated dose of 210 Gy resulted in a lethal phenotype. Our findings demonstrate that M. lignano represents a suitable model system for elucidating the effect of irradiation on the stem cell system in flatworms and for improving our understanding of the recovery potential of severely damaged stem-cell systems.  相似文献

For Australian pilots, short layovers (<40 h) are a feature of many international patterns. However, anecdotal reports suggest that flight crew members find patterns with short slips more fatiguing than those with a longer international layover, as they restrict the opportunity to obtain sufficient sleep. The current study aimed to determine whether pilots operating international patterns with short layovers have sufficient opportunity to recover prior to the inbound flight. Nineteen international pilots (ten captains, nine first officers) operating a direct return pattern from Australia to Los Angeles (LAX) with a short (n = 9) 9+/-0.8 h (mean+/-S.D) or long (n = 10) 62.2+/-0.9 h LAX layover wore an activity monitor and kept a sleep/duty diary during the pattern. Immediately before and after each flight, pilots completed a 5 min PalmPilot-based psychomotor vigilance task (Palm-PVT). Flights were of comparable duration outbound (3.5+/-0.6 h) and inbound (14.3+/-0.6 h) and timing. The amount of sleep obtained in-flight did not significantly vary as a function of layover length. However, pilots obtained significantly more sleep during the inbound (3.7+/-0.8 h) than the outbound flight (2.2+/-0.8 h). Pilots with the shorter layover obtained significantly less sleep in total during layover (14.0+/-2.7 h vs. 19.6+/-2.5), due to significantly fewer sleep periods (3.0+/-0.7 vs. 4.0+/-0.9). However, neither mean sleep duration nor the sleep obtained in the 24 h prior to the inbound flight significantly differed as a function of layover length. Response speed significantly varied across the pattern, and a significant interaction was also observed. For pilots with a short layover, response speed was significantly slower at the end of both the outbound and inbound flight, and prior to the inbound flight (i.e., at the end of layover), relative to response speed at the start of the pattern (pre-trip). Similarly, response speed for the longer layover was slower at the end of the outbound flight compared to pre-trip (approaching significance, p = 0.073). However, response speed at the beginning of the inbound flight was significantly faster than pre-trip and did not significantly differ from pre-trip at the end of the inbound flight. The data suggest that short slips (<40 h) do not allow pilots the opportunity to obtain sufficient sleep to reverse the effects of fatigue accumulated during the outbound flight. As a result, their response speed prior to the inbound flight is substantially slower than the response speed of flight crew with a longer layover.  相似文献

When restoring ecosystems, the simple removal of stresses causing degradation may seem preferable over other more costly and time consuming approaches. However, some restoration techniques can be implemented at reasonable cost and with increased efficiency in certain cases. We examined the successional trajectories of vegetation within abandoned block-cut peatlands in a major peat-producing region of Eastern Canada to evaluate whether the use of rewetting as a restoration technique can assist in the recovery of a typical bog plant community dominated by Sphagnum compared to spontaneous recolonization alone. We surveyed a total of 55 trenches in 6 peatlands twice, ~25 and ~35 years after the cessation of peat extraction. Canonical ordinations evidenced a generalized process of afforestation during the decade studied, partially driven by agricultural drainage in the surrounding landscape. Plant communities were dominated by ericaceous shrubs that hampered the spontaneous recovery of a Sphagnum-dominated system typical of bogs in the short and medium-term. Three of the six peatlands surveyed were partially restored by blocking drainage ditches. There, we surveyed plant composition in rewetted (28) and non-rewetted (26) trenches and observed that rewetting mitigated the increase in tree dominance, decreased the dominance by ericaceous shrubs, and favored the spread of non-vascular species with a wet habitat preference (notably Sphagnum species from the Cuspidata section). We conclude that the use of low intervention restoration techniques in block-cut bogs, such as the blockage of former drainage ditches, can re-orient undesired vegetation trajectories driven by spontaneous recolonization alone.  相似文献

Aims South American Pampa grasslands are habitats of great conservation interest, with a distinct and rich flora, but have been intensely converted to other land uses, including tree plantations. While necessity for restoration grows, no information on restoration potential of grasslands after afforestation exists. Here, we aim at analyzing composition and structure of grassland vegetation with a history of eucalyptus plantations in order to assess recovery potential of these areas. We hypothesized that areas with history of eucalyptus would differ from reference grasslands with no history of land-use change in terms of floristic and functional composition and would present lower species richness.  相似文献

The objective of this study was to evaluate the thermoregulatory responses, acid-basic and electrolytic equilibrium of locally adapted goats under natural heat conditions in a semi-arid region. Ten (10) Canindé goats aged between 2 and 3 years, non-lactating, non-pregnant and having a body weight (BW) of 22.90 ± 2.70 kg were used in this study. Air temperature (AT) and relative humidity (RH) were measured, and the radiant heat load (RHL) was subsequently calculated. Rectal temperature (RT), respiratory rate (RR), sweating rate (SR) and heat shock (S) were recorded at 1-h intervals for 24 continuous hours. Hydrogen potential (pH), partial pressure of carbon dioxide (PCO2), partial pressure of oxygen (PO2), bicarbonate (HCO3), base excess (BE), total carbon dioxide concentration (TCO2), oxygen saturation (SO2), sodium (Na+) and potassium (K+) were recorded at three moments during the day (5 a.m.; 1 p.m.; 6 p.m.). There were also significant differences between the means of hours of the day for AT and RH. RR was the thermoregulatory response which most closely followed RHL, with important elevations in the periods between 10 a.m. to noon. It was observed that the goats activated their SR mechanism before RR, more precisely between the hours of 9 a.m. and 1 p.m. The acid-base and electrolytic equilibrium for the goats which showed great association with the first components contributed the most to the total variation of the data. The most important variables in the adaptive profile of these animals in order of importance were: SO2, PO2, RR, RT, SR, HCO3, BE, TCO2 and pH. An association between all variables grouped in each period was observed, where the thermoregulatory responses in the periods of 5 a.m. and 6 p.m. were closer than when compared to 1 p.m., showing a physiological return to the initial state. Therefore, the variation in thermoregulatory responses, acid-base and electrolytic equilibrium indicated that the goats have the ability to recover after a challenging environmental condition.  相似文献

Onabotulinum toxin A (BTX-A) is a frequently used treatment modality to relax spastic muscles by preventing acetylcholine release at the motor nerve endings. Although considered safe, previous studies have shown that BTX-A injections cause muscle atrophy and deterioration in target and non-target muscles. Ideally, muscles should fully recover following BTX-A treatments, so that muscle strength and performance are not affected in the long-term. However, systematic, long-term data on the recovery of muscles exposed to BTX-A treatments are not available, thus practice guidelines on the frequency and duration of BTX-A injections, and associated recovery protocols, are based on clinical experience with little evidence-based information. Therefore, the purpose of this study was to investigate muscle recovery following a six months, monthly BTX-A injection (3.5 U/kg) protocol. Twenty seven skeletally mature NZW rabbits were divided into 5 groups: Control (n=5), zero month recovery – BTX-A+0 M (n=5), one month recovery – BTX-A+1 M (n=5), three months recovery – BTX-A+3 M (n=5), and six months recovery – BTX-A+6 M (n=7). Knee extensor strength, muscle mass and percent contractile material in injected and contralateral non-injected muscles was measured at each point of recovery. Strength and muscle mass were partially and completely recovered in injected and contralateral non-injected muscles for BTX-A+6 M group animals, respectively. The percent of contractile material partially recovered in the injected, but did not recover in the contralateral non-injected muscles. We conclude from these results that neither target nor non-target muscles fully recover within six months of a BTX-A treatment protocol and that clinical studies on muscle recovery should be pursued.  相似文献

The problem tackled here concerns the feasibility of DNA sequencingusing hybridization methods. We establish algorithms for andcomputational limitations to the reconstruction of a sequencefrom all its subsequences having the same length: in other words,the building of a string that contains all the words of a givenset, and only these ones. Generally there are several possiblestrings. We refer to graph theory and propose an algorithm toenumerate all the strings that are solutions. We then carriedout stimulations using real DNA sequences. They provided somenecessary conditions and give some upper bounds to the lengthof the sequence to recover in relation with the length of oligonucleotides.To avoid limiting ourselves to problems that admit a uniquesolution, we introduce another algorithm that produces a signaturefor each solution string. Abstract In early 2003 a series of large, wildfire‐related sediment slugs occurred in streams in the south‐eastern Australian alpine region. Back‐pack and boat‐mounted electrofishing were used to measure changes in riverine fish fauna after one particularly large sediment slug which started in an upland stream and then travelled downstream through 200 km of third and fourth order stream. Twelve impact sites and eight control sites were surveyed where there were previous data on fish populations. The sites were surveyed directly after the sediment slug had passed and then 12, 24 and 36 months after. Immediately after the sediment slug, fish abundances fell by between 95–100% at four impact sites in the upper reaches of the study area, primarily due to the effect of low‐dissolved oxygen levels. Twelve months later fish numbers were still decreased in the upper catchment but showed signs of recovery after 24 months. Further downstream, where water quality was not as severely affected by the sediment slug, the effects on native fishes were less apparent. The circumstances of these events represented a unique opportunity to obtain baseline data regarding the effects of post‐fire disturbances on fish, and their time to recovery. If fire were to occur in catchments where endangered species exist, our results suggest that actions such as translocation may be required to ensure the long‐term survival of threatened species.  相似文献

Hundreds of studies have assessed variation in the degree to which people experience disgust toward substances associated with pathogens, but little is known about the mechanistic sources of this variation. The current investigation uses olfactory perception and threshold methods to test whether it is apparent at the cue-detection level, at the cue-interpretation level, or both. It further tests whether relations between disgust sensitivity and olfactory perception are specific to odors associated with pathogens. Two studies (N's = 119 and 160) of individuals sampled from a Dutch university each revealed that pathogen disgust sensitivity relates to valence perceptions of odors found in pathogen sources, but not to valence perceptions of odors not associated with pathogens, nor to intensity perceptions of odors of either type. Study 2, which also assessed olfactory thresholds via a three-alternative forced-choice staircase method, did not reveal a relation between pathogen disgust sensitivity and the ability to detect an odor associated with pathogens, nor an odor not associated with pathogens. In total, results are consistent with the idea that pathogen disgust sensitivity relates to how olfactory pathogen cues are interpreted after detection, but not necessarily to the ability to detect such cues.  相似文献
