Total S concentration in the top 35 cm of Big Run Bog peat averaged 9.7 mol·g — wet mass–1 (123 mol·g dry mass–1). Of that total, an average of 80.8% was carbon bonded S, 10.4% was ester sulfate S, 4.5% was FeS2­S, 2.7% was FeS­S, 1.2% was elemental S, and 0.4% was SO4 2–­S. In peat collected in March 1986, injected with35S­SO4 2– and incubated at 4 °C, mean rates of dissimilatory sulfate reduction (formation of H2S + S0 + FeS + FeS2), carbon bonded S formation, and ester sulfate S formation averaged 3.22, 0.53, and 0.36 nmol·g wet mass–1·h–1, respectively. Measured rates of sulfide oxidation were comparable to rates of sulfate reduction. Although dissolved SO4 2– concentrations in Big Run Bog interstitial water (< 200 µM) are low enough to theoretically limit sulfate reducing bacteria, rates of sulfate reduction integrated throughout the top 30–35 cm of peat of 9 and 34 mmol·m–2·d–1 (at 4 °C are greater than or comparable to rates in coastal marine sediments. We suggest that sulfate reduction was supported by a rapid turnover of the dissolved SO4 2– pool (average turnover time of 1.1 days). Although over 90% of the total S in Big Run Bog peat was organic S, cycling of S was dominated by fluxes through the inorganic S pools.  相似文献

Methane production and sulfate reduction in two Appalachian peatlands   总被引:9,自引:7,他引:2  
Anaerobic carbon mineralization was evaluated over a 1-year period in two Sphagnum-dominated peatlands, Big Run Bog, West Virginia, and Buckle's Bog, Maryland. In the top 35 cm of peat, mean rates of methane production, anaerobic carbon dioxide production, and sulfate reduction at Big Run Bog were 63,406 and 146 mol L-1 d-1, respectively, and at Buckle's Bog were 18, 486 and 104 mol L-1 d-1. Annual anaerobic carbon mineralization to methane and carbon dioxide at Big Run Bog and Buckle's Bog was 52.8 and 57.2 mol m-2, respectively. Rates of methane production were similar to rates reported for other freshwater peatlands, but methane production accounted for only 11.7 and 2.8%, respectively, of the total anaerobic carbon mineralization at these two sites. Carbon dioxide production, resulting substantially from sulfate reduction, dominated anaerobic carbon mineralization. Considerable sulfate reduction despite low instantaneous dissolved sulfate concentrations (typically < 300 mol L-1 of substrate) was apparently fueled by oxidation and rapid turnover of the reduced inorganic sulfur pool.The coincidence of high sulfate inputs to the Big Run Bog and Buckle's Bog watersheds through acid precipitation with the unexpected importance of sulfate reduction leads us to suggest a new hypothesis: peatlands not receiving high sulfate loading should exhibit low rates of anaerobic decomposition, and a predominance of methane production over sulfate reduction; however, if such peatlands become subjected to high rates of sulfur deposition, sulfate reduction may be enhanced as an anaerobic mineralization pathway with attendant effects on carbon balance and peat accumulation.  相似文献

The mineralization of organic carbon to CH4 and CO2 inSphagnum-derived peat from Big Run Bog, West Virginia, was measured at 4 times in the year (February, May, September, and November) using anaerobic, peat-slurry incubations. Rates of both CH4 production and CO2 production changed seasonally in surface peat (0–25 cm depth), but were the same on each collection date in deep peat (30–45 cm depth). Methane production in surface peat ranged from 0.2 to 18.8 mol mol(C)–1 hr–1 (or 0.07 to 10.4 g(CH4) g–1 hr–1) between the February and September collections, respectively, and was approximately 1 mol mol(C)–1 hr–1 in deep peat. Carbon dioxide production in surface peat ranged from 3.2 to 20 mol mol(C)–1 hr–1 (or 4.8 to 30.3 g(CO2) g–1 hr–1) between the February and September collections, respectively, and was about 4 mol mol(C)–1 hr–1 in deep peat. In surface peat, temperature the master variable controlling the seasonal pattern in CO2 production, but the rate of CH4 production still had the lowest values in the February collection even when the peat was incubated at 19°C. The addition of glucose, acetate, and H2 to the peat-slurry did not stimulate CH4 production in surface peat, indicating that CH4 production in the winter was limited by factors other than glucose degradation products. The low rate of carbon mineralization in deep peat was due, in part, to poor chemical quality of the peat, because adding glucose and hydrogen directly stimulated CH4 production, and CO2 production to a lesser extent. Acetate was utilized in the peat by methanogens, but became a toxin at low pH values. The addition of SO4 2– to the peat-slurry inhibited CH4 production in surface peat, as expected, but surprisingly increased carbon mineralization through CH4 production in deep peat. Carbon mineralization under anaerobic conditions is of sufficient magnitude to have a major influence on peat accumulation and helps to explain the thin (< 2 m deep), old (> 13,000 yr) peat deposit found in Big Run Bog.  相似文献

A eubacterium producing a blue pigment was isolated from a drinking water filter, and subsequently identified as Vogesella indigofera. This bacterium was further investigated for its morphological and biochemical characteristics after exposure to hexavalent chromium, Cr6+. The threshold Cr6+ concentration inhibiting the pigment production by V. indigofera was 200–300 g ml–1 in liquid cultures of nutrient broth and 100–150 g ml–1 on nutrient agar plates. The Cr6+ concentration preventing V. indigofera growth was 300–400 g ml–1 in liquid cultures, but greater than 150 g ml–1 on agar plates. Moreover, rugose colonies without the blue pigmentation were observed on agar plates amended with 150 (g Cr6+) ml–1. The biochemical utilization profiles of the colonies without pigmentation did not differ from the original pigment-producing ones, indicating phenotypic plasticity of this bacterium. The difference of phenotypic expression of V. indigofera under various Cr6+ concentrations might have potential application as a pollution bioindicator for heavy metals.  相似文献

Biochemical and biophysical parameters, including D1-protein turnover, chlorophyll fluorescence, oxygen evolution activity and zeaxanthin formation were measured in the marine seagrassZostera capricorni (Aschers) in response to limiting (100 mol·m–2·–1), saturating (350 mol·m–2·s–1) or photoinhibitory (1100 mol·m–2·s–1) irradiances. Synthesis of D1 was maximal at 350 mol·m–2·s–1 which was also the irradiance at which the rate of photosynthetic O2 evolution was maximal. Degradation of D1 was saturated at 350 mol·m–2·s–1. The rate of D1 synthesis at 1100 mol·m–2·s–1 was very similar to that at 350 mol·m–2·s–1 for the first 90 min but then declined. At limiting or saturating irradiance little change was observed in the ratio of variable to maximal fluorescence (Fv/Fm) measured after dark adaptation of the leaves, while significant photoinhibition occurred at 1100 mol·m–2·s–1. The proportion of zeaxanthin in the total xanthophyll pool increased with increasing irradiance, indicative of the presence of a photoprotective xanthophyll cycle in this seagrass. We studied the effect of a calcite (CaCO3) treatment on peat and pore water chemistry in poor fen and conifer swamp wetlands next to Woods Lake and its tributaries to evaluate the role of wetlands in an Experimental Watershed Liming Study (EWLS). Peat was characteristically organic rich and nutrient poor, with exchangeable Ca concentrations of < 13 cmolckg–1. We estimated that between 0.4 to 4 Mg (CaCO3) ha–1 fell directly on three study sites; however, one year after the treatment the increase in Ca concentration (0–8 cm depth) was equivalent to a (CaCO3) dosage of 3 Mg ha–1 with an additional 2–4 Mg ha–1 of undissolved (CaCO3) still present, suggesting the peat retained Ca supplied from uplands. Most aspects of peat chemistry including microbial respiration and SO4 reduction did not respond to the treatment.Peat pore water (5 and 20 cm depths) had a mean pH of 4.82 before treatment with high concentrations of dissolved organic carbon (DOC mean of 790 mol C/l) and low Ca2+ concentration (mean of 32 mol/l). The (CaCO3) treatment increased concentrations of Ca2+ to a mean of 87 mol/l and dissolved inorganic carbon (DIC) from 205 to a mean of 411 mol/l, whereas it decreased monomeric Al concentration from 19 to 10 mol/l. Otherwise, pore water chemistry showed little response to the treatment, at least within natural spatial and temporal variation of solute concentrations. The results suggest that liming watersheds with the relatively low (CaCO3) dosage applied in this study can benefit acidic waters downstream by exporting more Ca and DIC and less monomeric Al, with otherwise little effect on the peat itself.  相似文献

Synopsis Arsenic persists in Chautauqua Lake, New York waters 13 years after cessation of herbicide (sodium arsenite) application and continues to cycle within the lake. Arsenic concentrations in lake water ranged from 22.4–114.81 g l–1, = 49.0 ag l–1. Well water samples generally contained less than 10 g l–1 arsenic. Arsenic concentrations in lake water exceeded U.S. Public Health Service recommended maximum concentrations (10 g l–1) and many samples exceeded the maximum permissible limit (50 g l–1). Fish accumulated arsenic from water but did not magnify it. Fish to water arsenic ratios ranged from 0.4–41.6. Black crappie (Pomoxis nigromaculatus) contained the highest arsenic concentrations (0.14–2.04 g g–1 ), X = 0.7 g g–1) while perch (Perca flavescens), muskellunge (Esox masquinongy) and largemouth bass (Micropterus salmoides) contained the lowest concentrations (0.02–0.13 g g–1). Arsenic concentrations in fish do not appear to pose a health hazard for human consumers.  相似文献

Summary In vivo electrolyte transport and water absorption from the caeca of dehydrated, low-NaCl diet hens are reported. In the absence of luminal glucose or acetate, net electrolyte transport rates and water absorption are small. When physiological concentrations of acetate (40 mM) are included in the perfusate, Na+ transport and water absorption increase significantly (P<0.01): 38±7 eqNa+/caecum kg·h and 256±33 l H2O/caecum · kg · h.A similar increase in water absorption occurs with the inclusion of 15 mM glucose in the perfusate (219±30 l H2O/caecum · kg · h), however both net Na+ and Cl absorption increase: 28±6 eq Na+/caecum · kg · h and 21±5 eq Cl/caecum kg · h.These pronounced increases in electrolyte and water absorption are not accompanied by any significant increase in transmural potential difference.The data presented establish caeca as important sites in the recuperation of water and electrolytes in dehydrated, low-NaCl diet hens.Abbreviations ECPD electrochemical potential difference - PD (transmural) potential difference - PEG polyethylene glycol  相似文献

Summary Basal oxygen consumption, ventilatory frequency, and heart rate were recorded at four different times during the unusually protracted 15–16-month spawning run of the Southern Hemisphere lamprey Geotria australis. At 15°C, the mean basal oxygen consumption of G. australis caught immediately after they had left the sea and embarked on the spawning run (45 l · g-1 · h-1) was less than in young adults about to commence their marine feeding phase (64 l · g-1 · h-1), but greater than in large ammocoetes (26.5 l · g-1 · h-1). Basal oxygen consumption fell progressively during the spawning-run of to 33 l · g-1 · h-1 after 5 months and 25 l · g-1 · h-1 after 10 months, before rising to 35 l · g-1 · h-1 after 15 months when the animals were approaching sexual maturity. The downwards trend in basal oxygen consumption contrasts with that recorded during the spawning run of Lampetra fluviatilis. Furthermore, these values for spawning-run of G. australis are far lower than those measured at any time during the upstream migration of L. fluviatilis or during the parasitic phase of landlocked Petromyzon marinus. A low and declining metabolic rate during much of the spawning run of G. australis would facilitate the conservation of energy reserves during this very long non-feeding period. Trends shown by ventilatory frequency and heart rate essentially parallel those of basal oxygen consumption. The Q10s for basal oxygen consumption, ventilatory frequency and heart rate over the temperature range 5–25°C were 1.6, 1.6, and 1.7, respectively. The trends shown by basal oxygen consumption during metamorphosis and the upstream migration did not parallel those exhibited by circulating thyroid hormones.  相似文献

An understanding of the mechanisms controlling nutrient availability and retention in and across ecosystems allows for a greater understanding of the role of nutrients in maintaining ecosystem structure and function. To examine the underlying mechanisms of phosphorus (P) cycling in northern peatlands, we compared the retention and movement of P across a natural hydrologic/pH gradient in nine peatlands by applying as a light rain an in situ tracer amount of 32PO4 –3 to track changes in P pools (vegetation, soil, microbial) over 30 days. The 31P concentrations of available P, microbial P, and the root P at 10–20 cm did not differ across the gradient, although total soil P and aboveground vegetation P content (g m–2) increased from bog to rich fen. Total retention of 32P in the first 24 hours of application was greatest in the bogs and intermediate fens (90–100%) and was very low (20–50%) in the rich fens. Retention of P in the different pools was dependent on the type of peatland and changed with time. In the first 24 hours in the bogs and intermediate fens, the microbial pool contained the largest amount of 32P, but by the seventh day, the aboveground vegetation contained the largest amount. In the rich fen, the recovered 32P was almost equally divided between the aboveground vegetation and the litter layer with very little in other pools. Therefore, although bogs and intermediate fens have a small total P pool, they have similar P availability to rich fens because of rapid cycling and efficient retention of P.  相似文献

Thylakoids isolated from cells of the red alga Porphyridium cruentum exhibit an increased PS I activity on a chlorophyll basis with increasing growth irradiance, even though the stoichiometry of Photosystems I and II in such cells shows little change (Cunningham et al. (1989) Plant Physiol 91: 1179–1187). PS I activity was 26% greater in thylakoids of cells acclimated at 280 mol photons · m–2 · s–1 (VHL) than in cells acclimated at 10 mol photons · m–2 · s–1 (LL), indicating a change in the light absorbance capacity of PS I. Upon isolating PS I holocomplexes from VHL cells it was found that they contained 132±9 Chl/P700 while those obtained from LL cells had 165±4 Chl/P700. Examination of the polypeptide composition of PS I holocomplexes on SDS-PAGE showed a notable decrease of three polypeptides (19.5, 21.0 and 22 kDa) in VHL-complexes relative to LL-complexes. These polypeptides belong to a novel LHC I complex, recently discovered in red algae (Wolfe et al. (1994a) Nature 367: 566–568), that lacks Chl b and includes at least six different polypeptides. We suggest that the decrease in PS I Chl antenna size observed with increasing irradiance is attributable to changes occurring in the LHC I-antenna complex. Evidence for a Chl-binding antenna complex associated with PS II core complexes is lacking at this point. LHC II-type polypeptides were not observed in functionally active PS II preparations (Wolfe et al. The effect of lead on the filtration rate of the zebra musselDreissena polymorpha was investigated, together with the accumulation of Pb in the soft tissues of the mussels. The NOEC-filtration was 116 g.l–1 (0,56 mol.l–1) and the EC50-filtration was 370 g.l–1 (1.79 mol.l–1). The NOEC-accumulation was the concentration found in the control water (1.4g.l–1). These experiments show that the EC50-filtration for Pb is similar to that for Cd, higher than that for Cu and lower than that for Zn. The water quality criteria for lead allow 25 g Pb.l–1 in surface water. This will not cause short-term effects. Long-term effects may, however, occur, since an accumulation of Pb as low as 16 g.l–1 was recorded in this study.  相似文献

We examined transepithelial transport of Ca2+ across the isolated opercular epithelium of the euryhaline killifish adapted to fresh water. The opercular epithelium, mounted in vitro with saline on the serosal side and fresh water (0.1 mmol·l–1 Ca2+) bathing the mucosal side, actively transported Ca2+ in the uptake direction; net flux averaged 20–30 nmol·cm–2·h–1. The rate of Ca2+ uptake varied linearly with the density of mitochondria-rich cells in the preparations. Ca2+ uptake was saturable, apparent K 1/2 of 0.348 mmol·l–1, indicative of a multistep transcellular pathway. Ca2+ uptake was inhibited partially by apically added 0.1 mmol·l–1 La3+ and 1.0 mmol·l–1 Mg2+. Addition of dibutyryl-cyclic adenosine monophosphate (0.5 mmol·l–1)+0.1 mmol·l–1 3-isobutyl-l-methylxanthine inhibited Ca2+ uptake by 54%, but epinephrine, clonidine and isoproterenol were without effect. Agents that increase intracellular Ca2+, thapsigargin (1.0 mol·l–1, serosal side), ionomycin (1.0 mol·l–1, serosal side) and the calmodulin blocker trifluoperazine (50 mol·l–1, mucosal side) all partially inhibited Ca2+ uptake. In contrast, apically added ionomycin increased mucosal to serosal unidirectional Ca2+ flux, indicating Ca2+ entry across the apical membrane is rate limiting in the transport. Verapamil (10–100 mol·l–1, mucosal side), a Ca2+ channel blocker, had no effect. Results are consistent with a model of Ca2+ uptake by mitochondria rich cells that involves passive Ca2+ entry across the apical membrane via verapamil-insensitive Ca2+ channels, intracellular complexing of Ca2+ by calmodulin and basolateral exit via an active transport process. Nitrate induction in spruce: an approach using compartmental analysis   总被引:6,自引:0,他引:6  
Using 13NO 3 -efflux analysis, the induction of nitrate uptake by externally supplied nitrate was monitored in roots of intact Picea glauca (Moench) Voss. seedlings over a 5-d period. In agreement with our earlier studies, efflux analysis revealed three compartments, which have been identified as surface adsorption, apparent free space, and cytoplasm. While induction of nitrate uptake was pronounced, NO 3 fluxes in induced plants were decidedly lower and the induction response was slower than in other species. Influx rose from 0.1 mol·g–1·h–1 (measured at 100 M [NO 3 o) in uninduced plants to a maximum of 0.5 mol·g–1h–1 after 3 d of exposure to 100 M [NO 3 o and declined to 0.3–0.4 mol·g–1h–1 at the end of the 5-d period. Efflux remained relatively constant around 0.02-0.04 mol·g–1h–1, but its percentage with respect to influx declined from initially high values (around 30%) to steady-state values of 4–7%. Cytoplasmic [NO 3 ] ranged from the low micromolar in uninduced plants to a maximum of 2 mM in plants fully induced at 100 M [NO 3 ]o. In-vivo root nitrate reductase activity (NRA) was measured over the same time period, and was found to follow a similar pattern of induction as influx. The maximum response in NRA slightly preceded that of influx. It increased from 25 nmol·g–1·h–1 without prior exposure to NO 3 to peak values around 150 nmol· g–1h–1 after 2 d of exposure to 100 M [NO 3 ]o. Subsequently, NRA declined by about 50%. Exchange of CO2 and O2 and chlorophyll fluorescence were measured in the presence of 360 1 · 1–1 CO2 in nitrogen in Helianthus annuss L. leaves which had been preconditioned in the dark or at a photon flux density (PFD) of 24 mol · m–2 · s–1 either in 21 or 0% O2. An initial light-dependent O2 outburst of 6 mol · m–2 was measured after aerobic dark incubation. It was attributed to the reduction of electron carriers, predominantly plastoquinone. The maximum initial rate of O2 evolution at PFD 8000 mol · m–2 · s–1 was 170 mol · m–2 · s–2 or about four times the steady CO2-and light-saturated rate of photosynthesis. Fluorescence measurements showed that the rate was still acceptor-limited. Fast O2 evolution ceased after electron carriers were reduced in the dark-adapted leaf, but continued for a short time at the lower rate of 62 mol · m–2 · s–1 in the light-adapted leaf. The data are interpreted to show that enzymes involved in 3-phosphoglycerate reduction are dark-inhibited, but were fully active in low light. In a dark-adapted leaf, respiratory CO2 evolution continued under nitrogen; it was partially inhibited by illumination. Prolonged exposure of a leaf to anaerobic conditions caused reducing equivalents to accumulate. This was shown by a slowly increasing chlorophyll fluorescence yield which indicated the reduction of the PSII acceptor QA in the dark. When the leaf was illuminated, no O2 evolution was detected from short light pulses, although transient O2 production was appreciable during longer light pulses. Production of hydrogen peroxide has been found in Ulva rigida (Chlorophyta). The formation of H2O2 was light dependent with a production of 1.2 mol·g FW–1·h–1 in sea water (pH 8.2) at an irradiance of 700 mol photons m–2·s–1. The excretion was also pH dependent: in pH 6.5 the production was not detectable (< 5 nmol·g FW–1·h–1) but at pH 9.0 the production was 5.0 mol·g FW–1·h–1. The production of H2O2 was totally inhibited by 3-(3,4-dichlorophenyl)-1,1 dimethylurea (DCMU). The ability of U. rigida growing in tanks (7501) under a natural light regime to excrete H2O2 was checked and found to be seven times higher at 08.00 hours than other times of the day. The H2O2 concentration in the cultivation tank (density: 2 g FW·l–1) reached the highest value (3 M) at 11.00 hours. Photosynthesis was not influenced by H2O2 formation. The H2O2 is suggested to come from the Mehler reaction (pseudocyclic photophosphorylation). With an oxygen evolution of 120 mmol·g FW–1·h–1 at pH 8.2 and 90 mmol·g FW–1·h–1 at pH 9.0, 0.5% and 2.7% of the electrons were used for extracellular H2O2 production. Kudzu (Pueraria lobata (Willd) Ohwi.) is a vine which forms large, monospecific stands in disturbed areas of the southeastern United States. Kudzu also emits isoprene, a hydrocarbon which can significantly affect atmospheric chemistry including reactions leading to tropospheric ozone. We have studied physiological aspects of isoprene emission from kudzu so the ecological consequences of isoprene emission can be better understood. We examined: (a) the development of isoprene emission as leaves developed, (b) the interaction between photon flux density and temperature effects on isoprene emission, (c) isoprene emission during and after water stress, and (d) the induction of isoprene emission from leaves grown at low temperature by water stress or elevated temperature. Isoprene emission under standard conditions of 1000 mol photons·m-2·s-1 and 30°C developed only after the leaf had reached full expansion, and was not complete until up to two weeks past the point of full expansion of the leaf. The effect of temperature on isoprene emission was much greater than found for other species, with a 10°C increase in temperature causing a eight-fold increase in the rate of isoprene emission. Isoprene emission from kudzu was stimulated by increases in photon flux density up to 3000 mol photons·m-2·s-1. In contrast, photosynthesis of kudzu was saturated at less than 1000 mol·m-2·s-1 photon flux density and was reduced at high temperature, so that up to 20% of the carbon fixed in photosynthesis was reemitted as isoprene gas at 1000 mol photons·m-2·s-1 and 35°C. Withholding water caused photosynthesis to decline nearly to zero after several days but had a much smaller effect on isoprene emission. Following the relief of water stress, photosynthesis recovered to the prestress level but isoprene emission increased to about five times the prestress rate. At 1000 mol photons·m-2·s-1 and 35°C as much as 67% of the carbon fixed in photosynthesis was reemitted as isoprene eight days after water stress. Leaves grown at less than 20°C did not make isoprene until an inductive treatment was given. Inductive treatments included growth at 24°C, leaf temperature of 30°C for 5 h, or witholding water from plants. With the new information on temperature and water stress effects on isoprene emission, we speculate that isoprene emission may help plants cope with stressful conditions.  相似文献

Summary Vasopressin-induced transformation of ridges to microvilli on the surface of granular cells of toad urinary bladder occurs in conjunction with induced alterations in the water permeability of the luminal membrane. This study was designed to establish the relationship between the time course for induction of microvilli and the time course for induction of increased water permeability after vasopressin stimulation. Hemibladders were examined at 2.5, 5, 10, 20 and 30 min following exposure to 20 mU/ml of vasopressin and at 5, 10, 20, 30, 40, 50 and 60 min after washout of vasopressin. Within 2.5 min, vasopressin initiated complete transformation of ridges to microvilli on approximately 13% of the granular cells, while osmotic water flow (Jv) was 0.31±0.10 l·min–1·cm–2. Five minutes following vasopressin stimulation, microvilli were present on approximately 30% of granular cells andJv was 2.27±0.13 l·min–1·cm–2. At 10 minJv was maximum at 4.03±0.15 l·min–1·cm–2 and 50% of the granular cells were covered with microvilli. This percentage increased to 70% at 20 min and was maintained at 30 min, althoughJv decreased to 3.9±0.35 l·min–1·cm–2 at 30 min. Five minutes following vasopressin washout, ridges interspersed with microvilli reappeared asJv fell to 1.10±0.30 l·min–1·cm–2. At 10 min after vasopressin washout,Jv approached basal levels, but the reversal of microvilli to ridges remained incomplete. At 60 min after vasopressin washout, the granular cells had regained their original ridgelike surface structures. Thus, these studies establish a temporal relationship between the induction and reversibility of vasopressin-induced microvillous formation and alterations in the osmotic water permeability of the apical plasmalemma.  相似文献

Using 13NH 4 + as a tracer, compartmental analyses for NH 4 + were performed in non-mycorrhizal roots of intact Picea glauca (Moench) Voss. seedlings at four different concentration regimes of external NH 4 + ([NH 4 + ]o), i.e. 0, 10, 100, and 1500 M. Three kinetically distinct compartments were identified, with half-lives of exchange of approximately 2 s, 30 s, and 14 min, assumed to represent surface adsorption, Donnan free space, and cytoplasm, respectively. No significant differences were found in half-lives of exchange with changes in [NH 4 + ]o. Influx was calculated to be 0.96 mol·g–1·h–1 in N-deprived plants (measured at 10 M [NH 4 + ]o), while under steady-state conditions it was 0.21 mol·g–1h–1 at 10 M [NH 4 + ]o, 1.96 mol·g–1–1 at 100 M [NH 4 + ]o, and 6.45 mol·g–1·h–1 at 1.5 mM [NH 4 + ]o. Efflux measured over the same range constituted approximately 9% of influx in N-deprived plants, 10% at 10 M, 28% at 100 M, and 35% at 1.5 mM [NH 4 + ]o. Cytoplasmic [NH 4 + ] was estimated at 6 m M in N-deprived plants, 2 mM at 10 M [NH 4 + ]o, 14 mM at 100 M, and 33 mM at 1.5 mM. Free-space [NH 4 + ] was 84 M, 50 M, 700 M, and 8 mM, respectively. In comparison with previously published data on fluxes and compartmentation of NO 3 in white-spruce seedlings, results of this study identify a pronounced physiological preference of this species for NH 4 + over NO 3 as an inorganic N source in terms of uptake and intracellular accumulation.
