The reaction of the superoxide radical anion (O2), with the semi-oxidized tryptophan neutral radical (Trp·) generated from tryptophan (Trp) by pulse radiolysis has been observed in a variety of functionalized Trp derivatives including peptides. It is found that the reaction proceeds 4–5 times faster in positively charged peptides, such as Lys-Trp-Lys, Lys-Gly-Trp-Lys and Lys-Gly-Trp-Lys-O-tert-butyl, than in solutions of the negatively charged N-acetyl tryptophan (NAT). However, the reactivity of O2 with the Trp· radical is totally inhibited upon binding of these peptides to micelles of negatively charged SDS and is reduced upon binding to native DNA. By contrast, no change in reactivity is observed in a medium containing CTAB, where the peptides cannot bind to the positively charged micelles. On the other hand, the reactivity of the Trp· radical formed from NAT with O2 is reduced to half that of the free Trp· in buffer but is markedly increased in CTAB micelles. The models studied here incorporate elements of the complex environment in which Trp· and O2 may be concomitantly formed in biological system and demonstrate the magnitude of the influence such elements may have on the kinetics of reactions involving these two species.  相似文献

Electrospray ionisation (ESI) mass spectrometry was used to examine the reactions of the clinically used antiarthritic agent [Au(S2O3)2]3−, and AuPEt3Cl, a derivative of another clinically used agent auranofin, with human serum albumin (HSA) obtained from a human volunteer. Both compounds reacted readily with HSA to form complexes containing one or more covalently attached gold fragments. In the case of AuPEt3Cl, binding was accompanied by the loss of the chloride ligand, while for [Au(S2O3)2]3− the mass spectral data indicated binding of Au(S2O3) groups. Experiments performed using HSA with Cys34 blocked by reaction with iodoacetamide were consistent with reaction of both gold compounds with this amino acid. Separate blocking experiments using diethylpyrocarbonate and AuPEt3Cl also provided evidence for histidine residues acting as lower-affinity binding sites for this gold compound. ESI mass spectra of solutions containing [Au(S2O3)2]3− or [Au(CN)2], and HSA, provided evidence for the formation of protein complexes in which intact gold molecules were non-covalently bound. In the case of [Au(S2O3)2]3−, these non-covalent complexes proved to be transitory in nature. However, for [Au(CN)2] a non-covalent complex containing a single gold molecule bound to HSA was found to be stable, and constituted the main adduct formed in solutions containing low-to-medium Au-to-HSA ratios. Evidence was also obtained for the formation of a covalent adduct in which a single Au(CN) moiety was bonded to Cys34 of the protein. AuPEt3Cl reacted to a much lower extent with HSA that had Cys34 modified by formation of a disulfide bond to added cysteine, than with unmodified HSA. Chlorite dismutase (Cld) is a heme enzyme capable of rapidly and selectively decomposing chlorite (ClO2 ) to Cl and O2. The ability of Cld to promote O2 formation from ClO2 is unusual. Heme enzymes generally utilize ClO2 as an oxidant for reactions such as oxygen atom transfer to, or halogenation of, a second substrate. The X-ray crystal structure of Dechloromonas aromatica Cld co-crystallized with the substrate analogue nitrite (NO2 ) was determined to investigate features responsible for this novel reactivity. The enzyme active site contains a single b-type heme coordinated by a proximal histidine residue. Structural analysis identified a glutamate residue hydrogen-bonded to the heme proximal histidine that may stabilize reactive heme species. A solvent-exposed arginine residue likely gates substrate entry to a tightly confined distal pocket. On the basis of the proposed mechanism of Cld, initial reaction of ClO2 within the distal pocket generates hypochlorite (ClO) and a compound I intermediate. The sterically restrictive distal pocket probably facilitates the rapid rebound of ClO with compound I forming the Cl and O2 products. Common to other heme enzymes, Cld is inactivated after a finite number of turnovers, potentially via the observed formation of an off-pathway tryptophanyl radical species through electron migration to compound I. Three tryptophan residues of Cld have been identified as candidates for this off-pathway radical. Finally, a juxtaposition of hydrophobic residues between the distal pocket and the enzyme surface suggests O2 may have a preferential direction for exiting the active site.  相似文献

The objective of this study was to examine the role of NADPH oxidase on superoxide radical production under waterlogging in mung bean (Vigna radiata) cvs. T 44 (tolerant) and Pusa Baisakhi (PB) (susceptible), and wild species Vigna luteola. Two days of waterlogging caused decline in superoxide radical (O2 ·−) contents in all the genotypes, however, further waterlogging up to 8 d caused significant increase in O2 ·− contents. In control and revived plants O2 ·− contents were higher in PB, while under waterlogging stress T 44 and V. luteola showed greater increases in the O2 ·− contents. During waterlogging the increase in O2 ·− content was found to be due to the diphenylene iodonium chloridesensitive NADPH oxidase (NOX). This was further confirmed by the waterlogging induced increase in NOX activity, which was higher in tolerant genotypes T 44 and V. luteola compared with PB. Gene expression studies showed enhanced expression of NOX in the roots of waterlogged V. luteola and T 44, while little expression was observed in control or treated plants of PB. PCR band products were cloned and sequenced, and partial cDNAs of NOX was obtained. Results suggest that increase in O2 ·− content during waterlogging could be due to the induction of membrane linked NOX.  相似文献

The generation of superoxide anion radical (O2 ·−) in the cytochrome b 6 f complex (Cyt b 6 f) of spinach under high-light illumination was studied using electron paramagnetic resonance spectroscopy. The generation of O2 ·− was lost in the absence of molecular oxygen. It was also suppressed in the presence of NaN3 and could be scavenged by extraneous antioxidants such as ascorbate, β-carotene, and glutathione. The results also indicate that O2 ·−, which is produced under high-light illumination of the Cyt b 6 f from spinach, might be generated from a reaction involing 1O2, and the Rieske Fe-S protein could serve as the electron donor in the O2 ·− production. The mechanism of photoprotection of the Cyt b 6 f complex by antioxidants is discussed.  相似文献

 Using the tetrazolium salt XTT (Na,3′-[(phenylamino)-carbonyl]-3,4-tetrazolium]-bis(4-methoxy-6-nitro)benzenesulfonic acid hydrate) as a sensitive and physiologically compatible probe for the determination of superoxide (O2 ·−) production in vivo, we have shown that maize (Zea mays L.) coleoptiles possess the capacity of generating O2 ·− in the apoplastic space. Our results are in agreement with the notion that this activity is localized at the plasma membrane and can be attributed to an O2 ·−-synthesizing enzyme with catalytic and kinetic properties similar to that of the NADPH oxidase of mammalian phagocytes, with the important exception that it utilizes NADH instead of NADPH as electron donor. When applied to the apoplastic space, NADH strongly increased the O2 ·−-producing activity of coleoptiles. The maize NADH-dependent O2 ·−-synthase activity could clearly be differentiated from peroxidase-mediated O2 ·−-synthesizing activity by its insensitivity to cyanide and azide, as well as by its much higher affinity to O2. Formation of O2 ·−, and concomitantly appearing H2O2, was preferentially localized in the outer epidermis of the coleoptile. The physiological significance of O2 ·− and H2O2 production in relation to the growth-controlling function of the epidermal cell wall is discussed. On electron pulse irradiation of N2 saturated deoxynucleotide aqueous solution containing 20 mmol/L K2S2O8, 200 mmol/L f-BuOH and rutin or quercetin, the transient absorption spectra of the deoxynucleotide radical cations decayed quickly. At the same time, the spectra of flavonoid phenoxyl radicals formed within several dozen microseconds. The results indicated that deoxynucleotide radical cations can be repaired by flavonoids. The rate constants of the repair reactions were 3.8 × 108-4.4×108 mol-1 · L · s-1 and 1.3×108-1.8×108 mol-1 · L · s-1 for dAMP and dGMP radical cations, respectively.  相似文献

The aims of the present study were: (1) to assess aerobic metabolism in paraplegic (P) athletes (spinal lesion level, T4–L3) by means of peak oxygen uptake (O2peak) and ventilatory threshold (VT), and (2) to determine the nature of exercise limitation in these athletes by means of cardioventilatory responses at peak exercise. Eight P athletes underwent conventional spirographic measurements and then performed an incremental wheelchair exercise on an adapted treadmill. Ventilatory data were collected every minute using an automated metabolic system: ventilation (l · min−1), oxygen uptake (O2, l · min−1, ml · min−1 · kg−1), carbon dioxide production (CO2, ml · min−1), respiratory exchange ratio, breathing frequency and tidal volume. Heart rate (HR, beats · min−1) was collected with the aid of a standard electrocardiogram. O2peak was determined using conventional criteria. VT was determined by the breakpoint in the CO2O2 relationship, and is expressed as the absolute VT (O2, ml · min−1 · kg−1) and relative VT (percentage of O2peak). Spirometric values and cardioventilatory responses at rest and at peak exercise allowed the measurement of ventilatory reserve (VR), heart rate reserve (HRr), heart rate response (HRR), and O2 pulse (O2 P). Results showed a O2peak value of 40.6 (2.5) ml · min−1 · kg−1, an absolute VT detected at 23.1 (1.5) ml · min−1 · kg−1 O2 and a relative VT at 56.4 (2.2)% O2peak. HRr [15.8 (3.2) beats · min−1], HRR [48.6 (4.3) beat · l−1], and O2 P [0.23 (0.02) ml · kg−1 · beat−1] were normal, whereas VR at peak exercise [42.7 (2.4)%] was increased. As wheelchair exercise excluded the use of an able-bodied (AB) control group, we compared our O2peak and VT results with those for other P subjects and AB controls reported in the literature, and we compared our cardioventilatory responses with those for respiratory and cardiac patients. The low O2peak values obtained compared with subject values obtained during an arm-crank exercise may be due to a reduced active muscle mass. Absolute VT was somewhat comparable to that of AB subjects, mainly due to the similar muscle mass involved in wheelchair and arm-crank exercise by P and AB subjects, respectively. The increased VR, as reported in patients with chronic heart failure, suggested that P athletes exhibited cardiac limitation at peak exercise, and this contributed to the lower O2peak measured in these subjects. Hypoxia caused by eutrophication occurs over large areas in aquatic systems worldwide. Common carp (Cyprinus carpio) exposed to hypoxia (1 mg · O2 · l−1 and 2 mg · O2 · l−1) for 1 week showed a significant reduction in feeding rate, respiration rate, faecal production and nitrogenous excretion compared to those maintained at normoxia (7 mg · O2 · l−1). Fish exposed to hypoxia showed negative scope for growth (SfG), but no significant difference in the specific growth rate was revealed after 1 week in both hypoxic groups. A significant reduction in RNA/DNA ratio was, however, clearly evident in the white muscle of the 1 mg · O2 · l−1 treatment group, but not in the 2 mg · O2 · l−1 treatment group. Both specific growth rate and RNA/DNA ratio were significantly reduced when fish were exposed to severe hypoxia (0.5 mg · O2 · l−1) for 4 weeks. At all levels of hypoxia, growth reduction was accompanied by a significant decrease in RNA/DNA ratio in white muscle. Covariance analysis showed no significant difference between the slope of RNA/DNA ratio and growth rate under normoxic conditions and 0.5 mg · O2 · l−1 for 4 weeks (F=1.036, P > 0.326), as well as 1.0 mg · O2 · l−1 and 2.0 mg · O2 · l−1 for 1 week (F = 0.457, P > 0.5), indicating that the RNA/DNA ratio serves as a biomarker of growth under all oxygen levels, at least under controlled experimental conditions. Inoculation of bioreactors with shake-flask cultures present the organism with an immediate shift from an environment with little O2 to one in which O2 is typically at 100% saturation. The inoculation of such shake-flasks cultures into bioreactors sparged with 1 vvm air or 1 vvm air/O2 mix i.e. 50% O2 enrichment is an oxidatively stressful event, as judged by immediate increases in the intracellular concentrations of superoxide anion radical (O2·−) (from 4,600 to 11,600 RLU mg DCW−1 and 5,500 to 23,000 RLU mg DCW−1 respectively) and changes in the activities of the major antioxidant enzymes superoxide dismutase and catalase in all cultures. There are further effects on metabolic indices, particularly decreased nutrient consumption in oxygenated cultures (from 0.16 to 0.12 g starch g DCW h−1) and decreased protein production, indicating that inoculation of the bioreactor exerts a global burden on the cellular metabolic networks.  相似文献

In this paper, we proposed a new method for the determination of either human serum albumin (HSA) or 5-Aminosalicylic acid (5-ASA) by synchronous fluorescence spectra and examined the interaction between them using the molecular modeling method under simulative physiological conditions. The optimum conditions of synchronous fluorometric determination of HSA were investigated and the method was successfully applied to the determination of 5-ASA added to serum, urine, and saliva samples. The linear range of the determination of HSA and 5-ASA were 1.60 – 414 μg mL−1 and 0.76 –22.95 μg mL−1, the detection limits were 0.552 μg mL−1 and 0.38 μg mL−1, respectively. In addition, the effect of various common ions on the determination of HSA with 5-ASA was also discussed at room temperature. Figure The salicylic acid moiety is located within the binding pocket. The ring of 5-ASA was inserted in the hydrophobic cavity of site I, and it is important to note that the residue ARG-218 and the trptophan residue of HSA (Trp214) are in close proximity to the ring of 5-ASA suggesting the existence of hydrophobic interaction between them.  相似文献

Ferryl compounds [Fe(IV)=O] in living organisms play an essential role in the radical catalytic cycle and degradation processes of hemeproteins. We studied the reactions between H2O2 and hemoglobin II (HbII) (GlnE7, TyrB10, PheCD1, PheE11), recombinant hemoglobin I (HbI) (GlnE7, PheB10, PheCD1, PheE11), and the HbI PheB10Tyr mutant of L. pectinata. We found that the tyrosine residue in the B10 position tailors, in two very distinct ways, the reactivity of the ferryl species, compounds I and II. First, increasing the reaction pH from 4.86 to 7.50, and then to 11.2, caused the the second-order rate constant for HbII to decrease from 141.60 to 77.78 M−1 s−1, and to 2.96 M−1 s−1, respectively. This pH dependence is associated with the disruption of the heme–tyrosine (603 nm) protein moiety, which controls the access of the H2O2 to the hemeprotein active center, thus regulating the formation of the ferryl species. Second, the presence of compound I was evident in the UV–vis spectra (648-nm band) in the reactions of HbI and recombinant HbI with H2O2, This band, however, is completely absent in the analogous reaction with HbII and the HbI PheB10Tyr mutant. Therefore, the existence of a hydrogen-bonding network between the heme pocket amino acids (i.e., TyrB10) and the ferryl compound I created a path much faster than 3.0×10−2 s−1 for the decay of compound I to compound II. Furthermore, the decay of the heme ferryl compound I to compound II was independent of the proximal HisF8 trans-ligand strength. Thus, the pH dependence of the heme–tyrosine moiety complex determined the overall reaction rate of the oxidative reaction limiting the interaction with H2O2 at neutral pH. The hydrogen-bonding strength between the TyrB10 and the heme ferryl species suggests the presence of a cycle where the ferryl consumption by the ferric heme increases significantly the pseudoperoxidase activity of these hemeproteins.  相似文献

Evolution of the neutral and charged particle compositions in the afterglow of a single streamer channel in atmospheric-pressure air is simulated numerically. It is shown that, in the course of charge-exchange reactions, O2 ions rapidly transform into ions with a larger electron bound energy (such as O3 and NO3); in humid air, they also transform into hydrated ions NO3(H2O) n . As a result, the efficiency of primary electron production in the streamer head due to destruction of residual negative ions generated in the previous pulse decreases substantially.  相似文献

This study examined the effect of mild hypobaria (MH) on the peak oxygen consumption (O2peak) and performance of ten trained male athletes [ (SEM); O2peak = 72.4 (2.2) ml · kg−1 · min−1] and ten trained female athletes [O2peak = 60.8 (2.1) ml · kg−1 · min−1]. Subjects performed 5-min maximal work tests on a cycle ergometer within a hypobaric chamber at both normobaria (N, 99.33 kPa) and at MH (92.66 kPa), using a counter-balanced design. MH was equivalent to 580 m altitude. O2peak at MH decreased significantly compared with N in both men [− 5.9 (0.9)%] and women [− 3.7 (1.0)%]. Performance (total kJ) at MH was also reduced significantly in men [− 3.6 (0.8)%] and women [− 3.8 (1.2)%]. Arterial oxyhaemoglobin saturation (SaO2) at O2peak was significantly lower at MH compared with N in both men [90.1 (0.6)% versus 92.0 (0.6)%] and women [89.7 (3.1)% versus 92.1 (3.0)%]. While SaO2 at O2peak was not different between men and women, it was concluded that relative, rather than absolute, O2peak may be a more appropriate predictor of exercise-induced hypoxaemia. For men and women, it was calculated that 67–76% of the decrease in O2peak could be accounted for by a decrease in O2 delivery, which indicates that reduced O2 tension at mild altitude (580 m) leads to impairment of exercise performance in a maximal work bout lasting ≈ 5 min. Heavy metals (HMs) are toxic pollutants, which can negatively affect the physiological processes of plants; moreover, HMs can be present in the food chain endangering people’s health. The aim of this study was to investigate the early physiological events during HM exposure in the root tips of the food plant Pisum sativum L. Ten-day-old pea plants were treated with 100 μM CdCl2 or CuSO4, in nutrient solution for 48 h. We studied the rapid formation of different reactive oxygen species (hydrogen peroxide H2O2 and superoxide radical O2·−) and reactive nitrogen species (nitric oxide NO· and peroxynitrite ONOO) together with membrane damage and cell death in the meristem cells of pea roots using in vivo and in situ microscopic methods. In our experimental system, copper and cadmium induced the formation of H2O2 and NO. Two hours of heavy metal treatments resulted in an increased O2·− formation; however, later the level of this reactive molecule dramatically decreased. We found that high levels of NO were needed for ONOO production under HM exposure. A fast loss of membrane integrity and decreased cell viability were detected in root tips of copper-treated plants. The effects of cadmium seemed to be slower compared to copper, but this non-essential metal also caused cell death. We concluded that viability decreased when NO and H2O2 levels were simultaneously high in the same tissues. Using the NO scavenger it was also evidenced that NO generation is essential for cell death induction under copper or cadmium stress.  相似文献

Summary Protoplasts were isolated from oat (Avena sativa L.) leaves by the combination of highly purified preparations of pectin lyase, xylanase, and cellulase C1. During the enzymic isolation, superoxide radical (O 2 ) was generated from the tissues. Both the protoplasts themselves and the cell walls, exposed to enzyme treatment, produced O 2 . Hydrogen peroxide (H2O2) apparently accumulated in the reaction mixture due to the spontaneous dismutation reaction of O 2 , while a part of H2O2 may have been produced directly from cell walls by the action of enzymes. Singlet molecular oxygen (1O2) generated in the reaction mixture was detected by cholesterol oxidation in small unilamellar liposomes. It seems likely that1O2 may be generated by the peroxidase-H2O2-halide system during enzymic treatment of the leaves. The present experiment was designed to study the importance of strength and muscle mass as factors limiting maximal oxygen uptake (O2 max ) in wheelchair subjects. Thirteen paraplegic subjects [mean age 29.8 (8.7) years] were studied during continuous incremental exercises until exhaustion on an arm-cranking ergometer (AC), a wheelchair ergometer (WE) and motor-driven treadmill (TM). Lean arm volume (LAV) was estimated using an anthropometric method based upon the measurement of various circumferences of the arm and forearm. Maximal strength (MVF) was measured while pushing on the rim of the wheelchair for three positions of the hand on the rim (−30°, 0° and +30°). The results indicate that paraplegic subjects reached a similar O2 max [1.23 (0.34) l · min−1, 1.25 (0.38) l · min−1, 1.22 (0.18) l · min−1 for AC, TM and WE, respectively] and O2 max /body mass [19.7 (5.2) ml · min−1 · kg−1, 19.5 (6.14) ml · min−1 · kg−1, 19.18 (4.27) ml · min−1 · kg−1 for AC, TM and WE, respectively on the three ergometers. Maximal heart rate f c max during the last minute of AC (173 (17) beats · min−1], TM [168 (14) beats · min−1], and WE [165 (16) beats · min−1], were correlated, but f c max was significantly higher for AC than for TM (P<0.03). There were significant correlations between MVF and LAV (P<0.001) and between the MVF data obtained at different angles of the hand on the rim [311.9 (90.1) N, 313.2 (81.2) N, 257.1 (71) N, at −30°, 0° and +30°, respectively]. There was no correlation between O2 max and LAV or MVF. The relatively low values of f c max suggest that O2 max was, at least in part, limited by local aerobic factors instead of central cardiovascular factors. On the other hand, the lack of a significant correlation between O2 max and MVF or muscle mass was not in favour of muscle strength being the main factor limiting O2 max in our subjects. The production of reactive oxygen species (ROS) and the ways by which ROS are generated are very important facts related to heavy metal toxicity in plants. In this work, superoxide anion (O2 ·−) generation diminished in cadmium treated sunflower (Helianthus annuus L.) leaf discs, and this reduction was time and Cd-concentration dependent. In line with these findings, we observed that NADPH-dependent oxidase activity was significantly inhibited by 0.1 and 0.5 mM Cd2+ treatments and the expression of the NADPH oxidase putative gene related to O2 ·− synthesis in sunflower leaves was 83 % inhibited by 0.1 mM CdCl2 and almost completely depleted by 0.5 mM CdCl2.  相似文献

The energy cost per unit of distance (C s, kilojoules per metre) of the front-crawl, back, breast and butterfly strokes was assessed in 20 elite swimmers. At sub-maximal speeds (v), C s was measured dividing steady-state oxygen consumption (O2) by the speed (v, metres per second). At supra-maximal v, C s was calculated by dividing the total metabolic energy (E, kilojoules) spent in covering 45.7, 91.4 and 182.9 m by the distance. E was obtained as: E = E an+O2max t pO2max(1−e−( t p/)), where E an was the amount of energy (kilojoules) derived from anaerobic sources, O2max litres per second was the maximal oxygen uptake, α (=20.9 kJ · l O2 −1) was the energy equivalent of O2, τ (24 s) was the time constant assumed for the attainment of O2max at muscle level at the onset of exercise, and t p (seconds) was the performance time. The lactic acid component was assumed to increase exponentially with t p to an asymptotic value of 0.418 kJ · kg−1 of body mass for t p ≥ 120 s. The lactic acid component of E an was obtained from the net increase of lactate concentration after exercise (Δ[La]b) assuming that, when Δ[La]b = 1 mmol · l−1 the net amount of metabolic energy released by lactate formation was 0.069 kJ · kg−1. Over the entire range of v, front crawl was the least costly stroke. For example at 1 m · s−1, C s amounted, on average, to 0.70, 0.84, 0.82 and 0.124 kJ · m−1 in front crawl, backstroke, butterfly and breaststroke, respectively; at 1.5 m · s−1, C s was 1.23, 1.47, 1.55 and 1.87 kJ · m−1 in the four strokes, respectively. The C s was a continuous function of the speed in all of the four strokes. It increased exponentially in crawl and backstroke, whereas in butterfly C s attained a minimum at the two lowest v to increase exponentially at higher v. The C s in breaststroke was a linear function of the v, probably because of the considerable amount of energy spent in this stroke for accelerating the body during the pushing phase so as to compensate for the loss of v occurring in the non-propulsive phase.
