Suspension cultured cells of the liverwort, Reboulia hemisphaerica and of the moss, Barbula unguiculata were independently subcultured in the medium containing 2% glucose in the dark or in the light for more than one year, and the photosynthetic activities of the final cultures were determined. Throughout the culture period light-grown cells of both species contained high amount of chlorophyll (4 to 34 g mg–1 dry weight) and showed a high photosynthetic activity (10 to 84 mol O2 mg–1 chlorophyll h–1). Dark-grown cells of R. hemisphaerica showed the same level of chlorophyll content and photosynthetic O2 evolving activity as light-grown cells. Although chlorophyll content in dark-grown B. unguiculata cells was ten-fold lower than that in light-grown cells, the photosynthetic activity of these dark-grown cells was higher than that of light-grown cells based on chlorophyll content.  相似文献

Summary The linear growth phase in cultures limited by intracellular (conservative) substrate is represented by a flat exponential curve. Plant regeneration from callus cultures of Piper longum was achieved through organogenesis. In vitro grown shoots were used as explants for callus induction. Competent callus was initiated around the nodal ring of tissue using Murashige and Skoog medium supplemented with 1.0 mg.l–1- naphthaleneacetic acid and 0.2 mg.l–1 N6-benzyladenine. Optimum growth regulator concentrations for shoot induction and shoot elongation were found to be 0.5 mg.l–1 indole-3-acetic acid with 1.5 mg.l–1 benzyladenine, and 0.1 mg.l–1 indole-3-acetic acid with 0.2 mg.l–1 benzyladenine, respectively. Elongated shoots were rooted on half-strength Murashige and Skoog medium having 0.1 mg.l–1 indole3-acetic acid. A heterotrophic cotton (Gossypium hirsutum L. cv. Stoneville 825) cell suspension culture was adapted to grow photoautotrophically. After two years in continuous photoautotrophic culture at 5% CO2 (balance air), the maximum growth rate of the photoautotrophic cell line was a 400% fresh weight increase in eight days. The Chl concentration was approximately 500 g per g fresh weight.Elevated CO2 (1%–5%) was required for culture growth, while the ambient air of the culture room (600 to 700 ul CO2 1–1) or darkness were lethal. The cell line had no net photosynthesis at 350 ul 1–1 CO2, 2% O2, and dark respiration ranged from 29 to 44 mol CO2 mg–1 Chl h–1. Photosynthesis was inhibited by O2. The approximate 1:1 ratio of ribulose 1,5-bisphosphate carboxylase (RuBPcase) to phosphoenolpyruvate carboxylase (PEPcase) (normally about 6:1 in mature leaves of C3 plants) was due to low RuBPcase activity relative to that of C3 leaves, not to high PEPcase activity. The PEPcase activity per unit Chl in the cell line was identical to that of spinach leaves, while the RuBPcase activity was only 15% of the spinach leaf RuBPcase activity. RuBPcase activity in the photoautotrophic cells was not limited by a lack of activation in vivo, since the enzyme in a rapidly prepared cell extract was 73% activated. No evidence of enzyme inactivation by secondary compounds in the cells was found as can be found with cotton leaves. A procedure is described for the rapid establishment of photoautotrophic protoplast-derived cultures ofNicotiana plumbaginifolia. Photoautotrophic growth was induced by lowering the glucose concentration to 2.5 g.l–1 in the protoplast culture medium and by omitting glucose from the subsequent dilution medium. Four week-old highly viable suspensions were plated on an agar-medium without glucose in unsealed Petri dishes and kept in illuminated chambers flushed with 0.05 % or 2 % CO2. Air-grown calli had net photosynthesis rates of 1.8 and 17 moles CO2.g–1 fresh wt.h–1 in air at 0.034 % CO2 and in air enriched with 1 % CO2, respectively. Calli grown in 2 % CO2 exhibited lower rates of net photosynthesis at the two CO2 concentrations tested (0 and 7.5 moles CO2.g–1 fresh wt.h–1, respectively). The contribution of photosynthesis to growth was estimated to be 80 % in air-grown calli and more than 90 % in calli grown in 2 % CO2. Summary The influence of temperature on the growth of the theromophilic Bacillus caldotenax was investigated using chemostat techniques and a chemically defined minimal medium. All determined growth constants, that is maximal specific growth rate, yield and maintenance, were temperature dependent. It was striking that the very large maintenance requirement was about 10 times higher than for mesophilic cells under equivalent conditions. A death rate, which was very substantial at optimal and supraoptimal growth temperatures, was estimated by comparing the maintenance for substrate and oxygen. In late March 1997, an open-top-chamber (OTC) CO2 enrichment study was begun in the Colorado shortgrass steppe. The main objectives of the study were to determine the effect of elevated CO2 (720 mol mol–1) on plant production, photosynthesis, and water use of this mixed C3/C4 plant community, soil nitrogen (N) and carbon (C) cycling and the impact of changes induced by CO2 on trace gas exchange. From this study, we report here our weekly measurements of CO2, CH4, NOx and N2O fluxes within control (unchambered), ambient CO2 and elevated CO2 OTCs. Soil water and temperature were measured at each flux measurement time from early April 1997, year round, through October 2000. Even though both C3 and C4 plant biomass increased under elevated CO2 and soil moisture content was typically higher than under ambient CO2 conditions, none of the trace gas fluxes were significantly altered by CO2 enrichment. Over the 43 month period of observation NOx and N2O flux averaged 4.3 and 1.7 in ambient and 4.1 and 1.7 g N m–2 hr –1 in elevated CO2 OTCs, respectively. NOx flux was negatively correlated to plant biomass production. Methane oxidation rates averaged –31 and –34 g C m–2 hr–1 and ecosystem respiration averaged 43 and 44 mg C m–2 hr–1 under ambient and elevated CO2, respectively, over the same time period.  相似文献

Summary A turbidostatic and oxystatic fermentation system was used to study the growth kinetic ofMethylococcus capsulatus (Bath). Dissolved oxygen and methane concentrations were measured continuously with membrane inlet mass spectrometry. The specific growth rate was found to increase from 0.25 h–1 to 0.37 h–1 and the saturation constant for methane was found to decrease from 71 M to 1.3 M as the copper content of the medium was varied from a very low to a high value.  相似文献

Somatic embryogenesis leading to plant regeneration has been achieved in the bamboo, Dendrocalamus strictus, by culturing seeds (caryopses) on B5 basal medium supplemented with 2,4-dichlorophenoxyacetic acid. Callus cultures obtained from the embryonal end of the seeds differentiated chlorophyllous embryoids. On transfer to a germination medium (B5 liquid, sucrose, indolebutyric acid, and -naphthaleneacetic acid) 40% of the embryoids developed into plantlets. Spectrophotometric titrations were conducted on the system horse heart ferricytochromec plus ferrohexacyanide in the pH range 5 to 7 and at temperatures 8, 18, 22 and 28°C. A difference extinction coefficient for reducedvs. oxidized cytochromec at 550 nm of 21 mmol–1cm–1 was used in part of the evaluations. On the assumption that only one electron-transferlinked proton dissociation is effective for both ferro- and ferricytochromec in this pH range, various possible models are developed with only three conforming with the experimental pH dependence of the spectrophotometric equilibrium constant. The data conform best to a model with protonic dissociation constants between pH 5 and 7 such that the reduced cytochromec species is at least a factor of 3 more acidic than the one for oxidized cytochromec (with pKH 6). This interpretation holds least for the data at 22°C, which points to a structural rearrangement at about this temperature (Czerlinski and Bracokova, 1973; Zabinski and Czerlinski, 1974; Zabinski, et al., 1974). While the extinction coefficient of ferrocytochromec shows no significant change with pH and temperature, the one for ferricytochromec does: it is about 5% larger at pH 5 than at pH 7 (550 nm). Graphs for the absorption change of ferricytochromec (pH 7 as reference) document the details over the wavelength range 500 to 750 nm.  相似文献

The mineralization of organic carbon to CH4 and CO2 inSphagnum-derived peat from Big Run Bog, West Virginia, was measured at 4 times in the year (February, May, September, and November) using anaerobic, peat-slurry incubations. Rates of both CH4 production and CO2 production changed seasonally in surface peat (0–25 cm depth), but were the same on each collection date in deep peat (30–45 cm depth). Methane production in surface peat ranged from 0.2 to 18.8 mol mol(C)–1 hr–1 (or 0.07 to 10.4 g(CH4) g–1 hr–1) between the February and September collections, respectively, and was approximately 1 mol mol(C)–1 hr–1 in deep peat. Carbon dioxide production in surface peat ranged from 3.2 to 20 mol mol(C)–1 hr–1 (or 4.8 to 30.3 g(CO2) g–1 hr–1) between the February and September collections, respectively, and was about 4 mol mol(C)–1 hr–1 in deep peat. In surface peat, temperature the master variable controlling the seasonal pattern in CO2 production, but the rate of CH4 production still had the lowest values in the February collection even when the peat was incubated at 19°C. The addition of glucose, acetate, and H2 to the peat-slurry did not stimulate CH4 production in surface peat, indicating that CH4 production in the winter was limited by factors other than glucose degradation products. The low rate of carbon mineralization in deep peat was due, in part, to poor chemical quality of the peat, because adding glucose and hydrogen directly stimulated CH4 production, and CO2 production to a lesser extent. Acetate was utilized in the peat by methanogens, but became a toxin at low pH values. The addition of SO4 2– to the peat-slurry inhibited CH4 production in surface peat, as expected, but surprisingly increased carbon mineralization through CH4 production in deep peat. Carbon mineralization under anaerobic conditions is of sufficient magnitude to have a major influence on peat accumulation and helps to explain the thin (< 2 m deep), old (> 13,000 yr) peat deposit found in Big Run Bog.  相似文献

An efficient system for gene transfer into plants of Brassica juncea var. India Mustard, mediated by Agrobacterium tumefaciens. was developed through the manipulation of the culture medium and the use of the appropriate Agrobacterium strain. High frequency shoot regeneration (90–100%) was obtained from hypocotyl explants grown on medium containing 0.9% agarose, 3.3 mg/L AgNO3 and 0.5–2 mg/L BA in combination with 0.01–0.05 mg/L 2,4-D or 0.1–1 mg/L NAA. Of all the Agrobacterium strains tested, A. tumefaciens A208-SE, carrying the disarmed Ti plasmid and a binary vector pROA93, was the most effective for B. juncea transformation. pROA93 carries the coding sequences of the NPTII and the GUS genes, both driven by a common CaMV 35S promoter in two divergent directions. Inoculated explants grown on the selection medium in the presence of 0.5 mg/L BA and 0.1 mg/L NAA gave rise to transgenic shoots at the highest frequency (9%). All Ro transgenic plants were phenotypically normal, but variation in expression patterns of the GUS gene occurred among the transgenic plants in an organ- and tissue-specific manner. The effect of the ethylene antagonists norbornadiene and silver nitrate and the ethylene precursor l-aminocyclopropane-l-carboxylic acid (ACC) on Zea mays plant regeneration was studied. A 12-fold increase in plant regeneration, as measured by number of plants obtained per gram fresh weight from callus cultures of maize inbreds Pa91 and H99, was obtained by 250 M norbornadiene and 100 M silver nitrate treatments. An increase in amout of nonregenerable tissue and a 68% decrease in plant regeneration were associated with callus treated with 1 mM ACC. Ethylene emanation from 1 mM ACC treated callus reached a maximum of 170 nl g–1 h–1 after 3 days compared to 7 nl g–1 h–1 for the control. The free proline content was up to 80% lower in 1 mM ACC treated callus grown for 30 days on medium with or without 12 mM proline, respectively, as compared to each control. Phosphorus and nitrogen excretion rates by zooplankton communities from two eutrophic and shallow Dutch lakes were measured in laboratory. The variations in excretion rates in the lakes (May–October) were caused mainly by fluctuation in zooplankton biomass. Mean summer excretion rates (June–September) were 2.4 and 0.9 µg PO4P·1–1·d–1 in Lake Loosdercht and Lake Breukeleveen, respectively. This difference between the lakes was caused mainly by the lower zooplankton biomass in Lake Breukeleveen. The excretion of 2.4 µg PO4P·1–1·d compared with the calculated P-demand of phytoplankton of 8.0 µg PO4P·1–1·d–1 is substantial in the summer (June–September) and far more important than the external P-supply of 0.4 µg P·1–1·d–1 and sediment release of 0.5 µg P·1–1·d–1. Both temperature and composition of zooplankton affected the weight specific excretion rates of the zooplankton community. The weight specific community excretion rates of P and N increased with temperature (exponential model); 1–8 g PO4P·mg–1 zooplankton-C·d–1 and 5–42 µg NH3N·mg–1 zooplankton-C·d–1 (10°C–20°C).  相似文献

Summary Aspergillus sp NCIM 508 produced 22 U/L of extracellular -mannosidase activity in a medium containing 8 % brewer's yeast cells. The optimum period and pH range for maximum production of the enzyme were 7 days and 4.0–6.0, respectively. The optimum pH and temperature for enzyme activity were 6.0 and 50°C, respectively. The enzyme was stable for 24 h at 28°C, in the pH range 6.0–7.0. The enzyme retained 100 and 65 % of its original activity after heating for 15 min at 45 and 55°C, respectively. The Km and Vmax for p-nitrophenyl--D- mannoside (PNPM) were 71M and 7.5 × 10–2 moles/min/mg, respectively. Diurnal variation in hydrological variables and dissolved inorganic nutrients such as PO inf4 sup3– -P, N O inf2 sup– -N, NO inf3 sup– -N and NH inf4 sup+ -N were studied in three interconnected biotopes including freshwater, marine and mangrove brackish water of the Kakinada coastal zone, Andhra Pradesh. Samples were collected at intervals of 3 hours, for a period of 24 hours. In the marine environment salinity varied from 26 to 32 whereas in the mangrove waters it fluctuated from 12 to 20 and in both biotopes salinity showed bimodal type of oscillation. Dissolved oxygen content was high in the mangrove waters during day time but decreased rapidly during the night hours. In the marine environment POf4 p3–-P concentration varied from 0.345 to 1.195 g at l–1, NO inf3 sup– -N from 1.03 to 6.62 g at l–1 and NO inf2 sup– -N from 0.086 to 0.506 g at l–1. The highest and the lowest concentrations of PO inf4 sup3– -P, NO inf3 sup– -N, NO inf2 sup– -N recorded in the mangrove waters were 0.790 and 0.325 g at l–1, 7.10 and 1.60 g at l–1 and 0.278 and 0.060 g at l–1, respectively. The concentration of PO inf4 sup3– -P, NO inf3 sup– -N and NO inf2 sup– -N were high in the freshwater canal, the maximum and minimum values being 1.110 and 0.730 g at l–1, 26.40 and 9.98 g at l–1 and 0.520 and 0.252 g at l–1 respectively. The concentration of ammonia was relatively high in the mangrove water. Gross and net primary production in the mangrove water was 4 times higher than in the marine biotope. There was no export of dissolved nutrients from the mangrove environment to the adjacent marine waters.  相似文献

Nitrification and denitrification rates were estimated simultaneously in soil-floodwater columns of a Crowley silt loam (Typic Albaqualfs) rice soil by an15N isotopic dilution technique. Labeled NO 3 was added to the floodwater of soil-water columns, half were treated with urea fertilizer. The (NO 3 +NO 2 )–N and (NO 3 +NO 2 )–N concentrations in the floodwater were measured over time and production and reduction rates for NO 3 calculated. Nitrate reduction in the urea amended columns averaged 515 mol N m–2h–1 and nitrification averaged 395 mol N m–2h–1 over the 35–153 d incubation. The nitrification rate for 4–19 d sampling period (1,560 mol N m–2h–1) in the urea amended columns was almost 9 times greater than the reduction rate (175 mol N m–2h–1) over the same period. Without the addition of urea the NO 3 production rate averaged 32 mol N m–2h–1 and reduction 101 mol N m–2h–1.  相似文献

Phytochrome contents have been assayed in vivo in cell suspension cultures of Petroselinum hortense, Daucus carota and Glycine max. After transferring the cells to fresh medium phytochrome increased in parallel with the increase in cell number, whereas the amount of phytochrome per cell remained constant. The rate of phytochrome reaccumulation after pretreatment with 15 h red light was very similar in all three systems (2.8–3.6 (e) 10–5/h). Dark reversion and a fast and slow Pfr destruction were observed in all systems. The rate constants of these reactions varied strongly between the systems. The phytochrome systems of the cell cultures were compared with those of etiolated and light-grown seedlings and it was concluded that the cell suspension cultures of Petroselinum hortense and Daucus carota behaved similarly to light-grown seedlings. By intracellular dialysis of isolated neurons of the mollusksHelix pomatia andLimnaea stagnalis and by a voltage clamp technique the characteristics of transmembrane ionic currents were studied during controlled changes in the ionic composition of the extracellular and intracellular medium. By replacing the intracellular potassium ions by Tris ions, functional blocking of the outward potassium currents was achieved and the inward current distinguished in a pure form. Replacement of Ringer's solution in the extracellular medium with sodium-free or calcium-free solution enabled the inward current to be separated into two additive components, one carried by sodium ions, the other by calcium ions. Sodium and calcium inward currents were found to have different kinetics and different potential-dependence: mNa=1±0.5 msec, mCa=3±1 msec, hNa=8±2 msec, hCa=115±10 msec (Vm=0), GNa=0.5 (Vm=–21±2 mV), GCa=0.5 (Vm=–8±2 mV). Both currents remained unchanged by tetrodotoxin, but the calcium current was specifically blocked by cadmium ions (2·10–3 M), verapamil, and D=600, and also by fluorine ions if injected intracellularly. All these results are regarded as evidence that the soma membrane of the neurons tested possesses separate systems of sodium and calcium ion-conducting channels. Total S concentration in the top 35 cm of Big Run Bog peat averaged 9.7 mol·g — wet mass–1 (123 mol·g dry mass–1). Of that total, an average of 80.8% was carbon bonded S, 10.4% was ester sulfate S, 4.5% was FeS2­S, 2.7% was FeS­S, 1.2% was elemental S, and 0.4% was SO4 2–­S. In peat collected in March 1986, injected with35S­SO4 2– and incubated at 4 °C, mean rates of dissimilatory sulfate reduction (formation of H2S + S0 + FeS + FeS2), carbon bonded S formation, and ester sulfate S formation averaged 3.22, 0.53, and 0.36 nmol·g wet mass–1·h–1, respectively. Measured rates of sulfide oxidation were comparable to rates of sulfate reduction. Although dissolved SO4 2– concentrations in Big Run Bog interstitial water (< 200 µM) are low enough to theoretically limit sulfate reducing bacteria, rates of sulfate reduction integrated throughout the top 30–35 cm of peat of 9 and 34 mmol·m–2·d–1 (at 4 °C are greater than or comparable to rates in coastal marine sediments. We suggest that sulfate reduction was supported by a rapid turnover of the dissolved SO4 2– pool (average turnover time of 1.1 days). Although over 90% of the total S in Big Run Bog peat was organic S, cycling of S was dominated by fluxes through the inorganic S pools.  相似文献
