The content of phosphor-containing metabolites as regards inorganic phosphate of tissues of the mouse liver after injection of zinc highly dispersed powder at the dose of 5 mg/kg has been established with quantitative calculation of 31P-NMR spectra. A decrease in the relative level of phosphor-containing metabolites in the liver regeneration on the first day after partial hepatectomy has been observed. It has been shown that the relative level of phosphor-containing metabolites after injection more decreased during the first two days after operation. The observed changes of P-compounds metabolites in the liver after injection of zinc highly dispersed powder are related to its stimulation effect on metabolism of sugar and phospholipids and on the cell respiratory process.  相似文献

It was revealed that an involuntary rats' intoxication with 10% ethanol solution during 8 months caused a reduction of zinc content in the brain. A subcutaneous depot administration of highly dispersed zinc powder in a dose of 5 mg/kg reduced ethanol consumption under conditions of free choice for two weeks. This effect of the preparation was associated with normalization of zinc content and increase of a malondialdehyde level in the brain.  相似文献

Administration of highly dispersed copper powder in a dose 0.2 mg/kg three days before modelled coronary-occlusion myocardial infarction caused transitory increase in activity of antioxidant enzymes--superoxide dismutase and glutathione peroxidase in the necrotic zone of myocardium of rats, and also staunch increase in the activity of these enzymes in noninfarction region.  相似文献

The present study was designed to investigate the effect of mercuric chloride administration on copper, zinc, and iron concentrations in the liver, kidney, lung, heart, spleen, and muscle of rats. The results showed that after dose and time exposure to mercuric chloride, the concentration of mercury in the six tissues was significantly elevated. Data showed that there were no interaction between mercury and tissue iron. There was a considerable elevation of the content of copper in the kidney and liver. The most significant changes in the copper concentration took place in the kidneys. About a twofold increase in the copper content of the kidney was noted after exposure to mercuric chloride (3 mg and 5 mg/kg). Only slight elevations in the copper content occurred in the liver, especially in high dose and longer exposure time. In the remaining organs, the copper content was not changed significantly (p>0.05). The most significant changes in the zinc concentration took place in liver, kidney, lung, and heart (5 mg/kg). Marked changes in kidney zinc concentrations were observed at any of the specified doses. Zinc concentrations were significantly increased in kidney of rats sacrificed 9–48 h after sc injection of HgCl2 (5 mg/kg); in liver obtained from rats at 18, 24, or 48 h after injection; and in lung after 24 or 48 h of treatment. The heart and spleen zinc concentrations were elevated at 24 and 48 h after injection of HgCl2 (5 mg/kg), respectively. The results of this study implicate that effects on copper and zinc concentrations of the target tissues of mercury may play an important role in the pathogenesis of acute mercuric chloride intoxication.  相似文献

Summary Laboratory studies to determine the tolerance of an isolate ofAlternaria tenuis causing a black core rot of mandarin oranges (Citrus reticulata) have shown that presence of low amounts of manganese, zinc, iron, copper, boron, and molybdenum in a synthetic medium stimulated growth and sporulation of the fungus. Concentrations of 20 ppm or more of copper sulphate, boric acid, and ammonium molybdate, however, caused significant reduction of growth. Conidia of the fungus were very sensitive to even low amounts of copper, zinc, and iron. Copper sulphate was most toxic to spore germination followed, in order of inhibitory effect, by zinc sulphate, ferrous sulphate, and boric acid. Ammonium molybdate and manganese sulphate were stimulatory to spore germination at 5 ppm. There was a positive correlation between percentage of spore germination and average length of germ tubes in different concentrations of various chemicals. The possibility of using copper sulphate, zinc sulphate, and ferrous sulphate as spray materials for trees and as dip for ripe fruits to prevent black core rot are discussed.Assoc. The interaction of injected zinc and cadmium with metallothionein was investigated in newborn rats. Tissues of 5-day-old rats were removed 24 h after a single injection (Sc) of saline or zinc (20 mg/kg, body wt.) or cadmium (1 mg/kg, body wt.) with 2.5 μCi of 65Zn or 109Cd or 5 μCi of [35S]cysteine. Injection of zinc resulted in a 75% increase in the hepatic zinc concentration with a concomitant elevation of metallothionein (P < 0.001), zinc in metallothionein increased by 45% (P < 0.05); [35S]cysteine incorporation indicated the induced synthesis of metallothionein. Injection of cadmium did not alter either metallothionein or zinc levels in liver, but cadmium in cytosol was preferentially bound to metallothionein. Neither treatment altered hepatic copper metabolism and copper in metallothionein, nor renal zinc and metallothionein levels. These data indicate that zinc injection can elevate hepatic zinc levels and induce metallothionein synthesis in newborn rats despite high basal levels; cadmium injection does not induce metallothionein synthesis, though cadmium is avidly sequestered by pre-existing metallothionein. The differences in the induction of metallothionein by these divalent cations can be explained by the differences in their binding affinities for thiol groups in intracellular metallothionein.  相似文献

Copper absorption was measured at two levels of dietary zinc in six healthy young men who were confined to a metabolic unit for a 75 d study of zinc utilization. A diet of conventional foods was fed, providing either 16.5 or 5.5 mg zinc and 1.3 mg copper daily. Copper absorption was determined by feeding65Cu, a stable isotope of copper, once during the 16.5 mg Zn diet and near the beginning and end of the 5.5 mg Zn diet. Apparent copper absorption averaged 48.1% when the 16.5 mg Zn diet was fed. This was significantly higher than the averages of 37.2 and 38.5% when the 5.5 mg Zn diet was fed. Absorption also differed significantly among subjects. Fecal copper did not differ between diets or among subjects. All subjects were in positive copper balance at both levels of dietary zinc. These results suggest that a dietary zinc intake slightly above the Recommended Dietary Allowance of 15 mg/d does not increase fecal copper loss and does not interfere with copper absorption.  相似文献

The antifungal protection afforded by streptomycin-copper chelate in vivo in greenhouse tests against Phytophthora infestans on tomato was found to be of the order of six times greater than that of streptomycin sulphate. The copper chelate also showed much greater resistance to simulated rainfall. Similar tests showed that streptomycin-copper chelate (in terms of streptomycin content) was from 2 1/2 to 5 times more effective as an antifungal prophylactic spray than zinc ethylene-bis(dithiocarbamate) (zineb), and some fifty times more active than copper oxychloride formulated as a commercial 50% wettable powder. These three materials showed roughly similar resistance to weathering by 'rainfall' and it was assumed therefore that 250 i.u./ml. streptomycin as streptomycin-copper chelate might be expected to give the same degree of protection against Phytophthora infestans in the field as would the usual field rates of usage of zineb and copper oxychloride.  相似文献

The anticancer drug, cis-diamminedichloroplatinum, was administered to male rats at a dose of 0.5, 1.0, 2.0 or 4.0 mg platinum/kg body weight. After 48 hrs, the animals were killed and platinum, zinc, copper and metallothionein were measured in renal and hepatic tissue and platinum, zinc and copper were measured in the plasma. Administration of this platinum-containing compound did not induce hepatic or renal metallothionein and did not significantly alter the concentrations of copper or zinc in hepatic or renal tissue or in the plasma.  相似文献

The effect of copper on the release of cyclooxygenase metabolites from starch elicited, rat, peritoneal macrophages was investigated. Copper sulphate, in the range 10(-6)-10(-5) M, inhibited the formation of prostaglandin (PG) E2 and thromboxane (Tx) B2, the stable metabolite of TxA2, in a dose dependent manner but had no effect on the production of 6-keto-PGF1 alpha, the stable product of prostacyclin. At higher concentrations (5 x 10(-5) and 10(-4) M) the synthesis of all three metabolites of arachidonic acid (AA) was stimulated as was the release of radioactivity from macrophages prelabelled with 14C AA. Copper had no effect on the metabolism of exogenous AA however. At 10(-4) M copper also stimulated secretion of the lysosomal enzyme, beta-glucuronidase (GUR). Copper nitrate (10(-4) M), but not zinc sulphate, also stimulated eicosanoid formation and lysosomal enzyme release. Our results are consistent with the idea that copper stimulates eicosanoid formation via an effect on PL activity.  相似文献

Histochemical and atomic absorption spectrophotometric methods were used to study the effects of reserpine and d-amphetamine on the neuronal trace metal distribution in various regions of the central nervous system (hippocampus, parietal cortex, cerebellum). Six hours after single d-amphetamine treatment (15 mg/kg i.p.), the neuronal zinc level was significantly decreased in the hippocampus and in the parietal cortex. The intensity of sulphide silver staining was similarly greatly decreased in all layers of the hippocampus and the parietal cortex. Such a change was not observed when d-amphetamine was administered in a lower dose (5 or 10 mg/kg i.p.). Twenty hours after single reserpine treatment (10 mg/kg i.p.), there were no changes in the tissue levels and distribution of zinc, copper, iron and manganese. In animals treated with reserpine on five consecutive days, in a dose of 10 mg/kg/day i.p., the trace metal distribution twenty hours following the final treatment was essentially the same as in the control. The results strongly suggest that zinc does not play a direct role in vivo in the storage and mobilization processes of the catecholamines. A high dose of d-amphetamine, however, has a non-specific, toxic effect that is not interrelated with the catecholaminergic neuronal function; this effect is manifested in a diminished intensity of sulphide silver staining and in a reduction of the tissue zinc level.  相似文献

Acute phase changes in trace mineral metabolism were examined in turkey embryos. An endotoxin injection resulted in increased concentrations of serum copper and liver zinc and decreased concentrations of serum zinc in embryos incubated either in ovo or ex ovo. Changes in zinc and copper metabolism occurred when endotoxin either was injected intramuscularly, into the amnionic fluid, or administered onto the chorioallantoic membrane. Unlike poults, embryos did not respond to an inflammatory challenge with decreased serum iron concentrations. Acute phase changes in embryo serum zinc and copper as well as liver zinc concentrations were similar to those in poults. Increased liver zinc concentrations were associated with increased zinc in metallothionein (MT). An injection of a crude interleukin 1 preparation into embryos resulted in similar increases in hepatic zinc and MT concentrations as an endotoxin injection, suggesting a role for this cytokine in mediating the acute phase changes in embryonic zinc metabolism.  相似文献

The role of changes in incubation medium, connected with electrochemical dissolution of fine copper powder (FCP), in efficiency of copper inhibition of energy-dependent proton efflux from E. coli was studied. In conditions of aeration of bacterial suspension in water, toxic properties of FCP can be modelled by dispersed hydrogen peroxide. Addition of hydrogen peroxide to bacteria does not influence their sensitivity to copper chloride. Transfer of the bacteria to anaerobic conditions leads to complete inhibition of energy-dependent proton efflux from E. coli by various copper-containing compounds. The data obtained suggest that the higher efficiency of FCP action on energy processes in E. coli as compared to copper chloride is determined by alkalinization of medium and oxygen absorption in the process of electrochemical dissolution of the metal.  相似文献

It was found that the rate of the NaDPH-dependent lipid peroxidation in rat microsomes in vitro decreases 3 hours and increases 24, 48 and 72 hours after the injection of a highly dispersed iron powder (2 mg/kg). The number of readily oxidizable phospholipid fractions (phosphatidylethanolamine, phosphatidylinositol) was shown to decrease 3 hours and to increase 24 hours following the injection. A direct correlation was observed between the activity of the NADPH-dependent microsomal lipid peroxidation system and the level of natural antioxidants in the lipids.  相似文献

The role of metal sulphides vis-à-vis the availability of dietary copper in ruminant animals has been investigated using zinc sulphide as a model metal sulphide and a selection of copper complexes and copper containing proteins as models for sources of dietary copper. The extent of reactivity of zinc sulphide towards the copper complexes is dependent upon the type of donor atom co-ordinated to copper:
The order to reactivity is found to be CuO > CuN > CuS complexes and is in keeping with the reported values for the instability constant pKn of the complexes. In contrast, no reaction is observed between zinc sulphide and the copper containing proteins studied (azurin, superoxide dismutase and cerulophasmin) and is attributed to the protection of the copper centres by the protein backbone. The results facilitate an understanding of copper metabolism in ruminants and a mechanism is proposed for the removal of dietary copper sources in such species.Reactions between copper(II) sulphate solutions and samples of zinc sulphide having a range of specific surface areas (prepared by sintering at differing temperatures) have been studied. The fact that the reactivity is found to be highly dependent upon the specific surface area of the metal sulphide may well be of significance when considering the fate of copper in sulphur-rich biological systems.  相似文献

There is debate over whether homing pigeons, Columba livia, use olfactory information as part of their navigational map. Antagonists of the theory argue that homing deficits noted in anosmic pigeons may be due to a non-specific impairment in general information processing. In Experiment I, we present data from a modest investigation describing the typical navigational deficits that occur following zinc sulphate-mediated anosmia. Our results are consistent with previous experiments that noted impairments in homing performance from unfamiliar locations of anosmic pigeons. Experiment II is a critical experiment that involved a spatial working memory paradigm; this paradigm consisted of testing zinc sulphate-treated birds in a forced-choice alternation task in a T-maze. This experiment allowed us to determine whether anosmic pigeons were impaired in memory performance, a robust measure of general information processing. There were no differences between the last day of training and a subsequent-test day when pigeons received an intranasal injection of zinc sulphate. This experiment suggests that zinc sulphate anosmia does not impair general information processing, supporting the hypothesis that homing pigeons use olfactory cues when homing from unfamiliar locations.  相似文献

The effect of 1,2-dimethyl-3-hydroxypyrid-4-one [deferiprone (DE)] on aluminum mobilization and elimination from tissues and serum as well as the influence on the excretion of trace elements, copper, zinc and manganese in rabbits was investigated. Sixteen New Zealand rabbits were randomly divided into three groups: control, Al-only and Al+DE. The Al-only and Al+DE animals received injections of Al2(SO43.18H2O 600 micromol Al/kg 5 days per week for 3 weeks. One week after the last Al injection the Al+DE rabbits were given deferiprone 750 micromol/kg/day intragastrically for 2 weeks. At the 42nd day the animals were sacrificed and the organs were taken and digested. Blood was taken from the ear artery three times (at the initiation of the experiment, before and after deferiprone administration). The aluminum and copper, zinc, manganese were determined by atomic absorption spectrophotometry. Our results showed that deferiprone could highly mobilize aluminum stores from tissues. At the end of experiment the aluminum contents of bone, kidney, liver and brain in Al+DE were significantly lower than that in Al-only rabbits. The copper, zinc, manganese contents were not affected by deferiprone administration.  相似文献

Bryophytes are often viewed as slowly evolving with little genetic variation within and among populations. A study of heavy-metal tolerance was initiated to test the capacity of bryophytes to undergo genetic differentiation in response to natural selection. Tolerance of Funaria hygrometrica to copper and zinc was greater in populations that originated on soil with high concentrations of these metals. Protonemal growth was more inhibited by the metals than was germination, and copper was more toxic than zinc. Zinc and copper tolerances were correlated, but so were the zinc and copper concentrations of native substrates. The pattern of population differentiation for heavy-metal tolerance in this species is much like that of flowering plants. Five populations of Physcomitrium pyriforme, which does not occur on metal-contaminated soil, were all highly tolerant of zinc but extremely intolerant of copper. This species seems to have an inherent tolerance to the former. Significant variation in tolerance to copper and zinc occurred among populations, but tolerance did not correlate with metal contents in native substrates. This pattern differs from that of flowering plants. Normal populations of species that colonize contaminated sites tended to be more tolerant than populations of species that do not colonize such sites. The extensive population differentiation in Funaria hygrometrica augments the evidence from electrophoretic data that there is genetic variation among populations of mosses and liverworts.  相似文献

The ionic events underlying gamma-aminobutyric acid (GABA) receptor activation on the cell body of a cockroach identified motor neuron were investigated by using current-clamp and voltage-clamp techniques. The reversal potential for GABA-induced hyperpolarization was -77.0 +/- 2.4 mV (mean +/- s.e.m.; n = 22). The reversal potential for GABA was highly sensitive to changes in external chloride, only weakly affected by changes in external potassium, and independent of changes in either sodium or calcium ion concentration. Intracellular ion-sensitive microelectrodes confirmed that an influx of chloride ions mediated the GABA response. Intracellular injection of acetate, citrate, sulphate, fluoride or ammonium caused no change in the reversal potential for GABA. Intracellular injection of chloride, bromide, chlorate, bromate, or methyl sulphate shifted the reversal potential for GABA to values more positive than resting membrane potential. Evidence for chloride accumulating and for extrusion mechanisms was examined by using putative inhibitors. However, internal application of ammonium ions, and external application of 4-acetamido-4'-isothiocyanatostilbene-2,2'-disulphonic acid (SITS), 4,4'-diisothiocyanatostilbene-2,2'-disulphonic acid (DIDS), acetazolamide, furosemide, ammonium, zinc and copper ions, were all without effect on the reversal potential for GABA.  相似文献
