Intense and very intense reactions were obtained for acid phosphatase, calcium activated ATP-ase (pH 9.4), magnesium activated ATP-ase (pH 7.2) and glucose-6-phosphatase in the cytoplasms of the myenteric plexus nerve cells of the small intestine of Macacus rhesus and rabbit. Nucleotidase activity was moderate or slight and unspecific alkaline phosphatase activity absent. Both ATP-ases presented an intense activity in the myenteric plexus nerve cells of human fetuses 30, 33, and 34 weeks old; 5-nucleotidase activity, slight in the 30-week-old fetuses became more intense in the 33- and 34-week-old fetuses. The satellite neuroglial cells, nerve fibers and blood capillaries presented negative alkaline phosphatase reactions and intense or very intense activities of the other phosphatases.  相似文献

Some hydrolases are localized cytochemically in the pollen and pollen tubes of Amaryllis vittata Ait. The function of different enzymes is discussed in relation to pollen tubes morphogenesis. Activity of most of the enzymes was confined to colpus region, pollen wall and general cytoplasm of pollen and pollen tube. The activity of hydrolytic enzymes like acid monophosphoesterase and lipase and was nil in the exine of both germinated and ungerminated pollen, whereas intense reaction for esterase was observed in exine. Enzyme activity increased after germination which suggest the hydrolysis of stored metabolites and synthesis of proteins and other metabolites for the active growth of pollen tube. Intense reaction for enzymes like alkaline phosphomonoesterase, ATP-ase, 5-nucleotidase etc. at the tip region of pollen tube suggest their role in physiological processes associated with exchange of materials through intercellular transport during tube wall polysaccharide biogenesis.  相似文献

The effect of lipolytic, glycolytic and proteolytic enzymes on the activities of plasma membrane enzyme activities in rat liver and kidney has been investigated by a pretreatment of tissue sections with the lytic enzymes. 2. The action of the proteolytic enzymes causes a very strong decrease of leucyl-beta-naphthylamidase activity, whereas the activities of ATP-ase, 5'-nucleotidase and alkaline phosphatase show a lesser decrease. This indicates a different membrane anchorage of leucyl-beta-naphthylamidase as compared to that of the phosphatases. 3. Treatment with glycolytic enzymes results in a decrease of 5'-nucleotidase and ATP-ase activity, whereas liver alkaline phosphatase and leucyl-beta-naphthylamidase show an increase in activity. 4. Treatment with phospholipase C gives about the same results. The very strong decrease of 5'-nucleotidase activity indicates a great dependence on phospholipids.  相似文献

High doses of propranolol give rise to a liver disenzymic injury by depression of its clearance and colloidopexic functions, but without any histological change. The molecular target responsible for this disturbance seems to lie at the level of ATP-ase, because propranolol stimulates the membrane activity and supresses the citochondrial one. There are described some effects of this drug and its pharmocological opposite, isopropylenorepinephrine, on the liver cell enzymes, which may serve as a morphological proof of the antagonism between the mitochondrial and plasmalemal ATP-ase activity, on the one hand, and of the membrane ATP-ase and AMP-cyclase, on the other hand.  相似文献

A new enzymatic method based on the stoichiometric reaction of oxygen dissolved in water is presented. This method is very sensitive, reproducible, and is not influenced by the presence in the solution of oxidoreducing substances. This is due to the enzyme specificity toward ascorbic acid. The method has been used in order to determine the contents of oxygen and ascorbic acid in canned green beans. We have so demonstrated that, after 36 months of conservation, oxygen reacts much quicker with oxidoreducing substances present in the cans than with ascorbic acid.  相似文献

Summary Carbonic anhydrase (CAH) activity was histochemically demonstrated in various parts of the alimentary canal of rat and in the stomach of man using the method of Waldeyer and Häusler (1959). The most intense histochemical reaction was observed in the parietal cells of the rat stomach, and reactions of decreasing intensity in the epithelial cells of the colon, appendix, jejunoileum, duodenum and oesophagus in the order mentioned. An intense reaction was also observed in the parietal cells of the stomach of man and a weak activity in the pyloric glands. After electrophoresis on cellulose acetate film the CAH activity in human and rat stomach mucosa showed one band with the same migration rate as the fastest moving band of the erythrocyte CAH.  相似文献

It was shown that intravenous injection of vasopressin in a dose of 5 pressor units per 1 kg of body weight led to changes in the ATP-ase activity of the heart and liver microsomes in one hour. These changes coursed in a different direction, i.e. ATP-ase activity of the heart microsomes increased, and ATP-ase activity of the liver microsomes decreased.  相似文献

Ultracytochemical investigation of ATP-ase activity was carried out in parietal cells of the mucosa and in cancer cells of human stomach carcinoma possessing a similar ultrastructure. In parietal cells the reaction product of ATP-ase was observed on the membranes of microvilli of intracellular canaliculi, on the membranes delineating the lateral intercellular space, on the basal plasmolemma and in the nucleoli. The reaction product was absent on the membranes of tubuvesicles and on the apical surface of the plasmalemma. In cancer cells the reaction product was found on the membranes of the microvilli of the intracellular canaliculi, basal plasmolemma and in the nucleoli. Comparative examination of ATP-ase activity in these cells implies that at least the part of the mechanism of hydrochloric acid secretion which is involved in the transfer of H+ and Cl- is retained in cancer cells. A steady decrease in hydrochlorid acid secretion observed in the stomach mucosa in cancer as well as in the tumour itself seems to be associated with other mechanisms.  相似文献

Summary The distribution of ATP-ase and other nucleotide phosphatases has been studied in young root tips of maize, barley and broad bean using frozen and paraffin sections stained by standard lead sulphide precipitation procedures. High ATP-ase activity was found at the root and cell surface which is in agreement with previous biochemical studies using excised roots and cell wall preparations. Staining was also found in the nuclei and at particulate sites in the cytoplasm. Differences were observed between the present work and the staining pattern obtained for -glycerophosphatase, and between ATP-ase staining in the three roots studied. The protracted effect of ultrasound treatment on the parenchymatous organs of rats has been studied histochemically at 1 to 10-day intervals. As a result of the altered redox conditions it is the oxidative enzymes in the first place that react to ultrasound treatment and their normalization takes a long time. Exposure decreased the activity of lysosomal enzymes and in the cell membrane the alkaline phosphatase and ATP-ase too. The activity of hydrolases returned to normal parallel with the normalization of the microscopic structure.  相似文献

1. Pretreatment of frozon cryostat sections with formaldehyde or calcium ions inhibits diffusion of the plasma membrane enzymes 5' nucleotidase, ATP-ase and alkaline phosphatase during incubation. 2. Treatment of fixed sections with different kinds of buffer at 37 degrees C induces diffusion of enzyme activity from the plasma membrane to other sites of the section and into the incubation medium. This buffer influence depends on temperature: at 4 degrees C only a slight diffusion occurs. Addition of phospholipase C, digitonin or taurocholate to the buffer opposes the buffer effect. 3. Pretreatment of frozen cryostat sections with a mixture of equal parts of chloroform and acetone give a good fixation of the plasma membrane enzymes 5'-nucleotidase, ATP-ase, alkaline phosphate and leucyl-beta-naphthylamidase. During this treatment the different kinds of lipids present in the membrane are ex-racted equally. After this fixation buffer treatment does not cause a visible diffusion of enzyme activity in the section. Only a slight diffusion (1 till 7 percent) into the buffer solution takes place. 4. The mentioned treatments open up possibilities to get insight into the membrane anchorage of plasma membrane enzymes.  相似文献

The present work describes histological and histochemical observations made on the neoplastic liver of Indian silver bills, Uroloncha malabarica. The histology of neoplastic tissue as well as liver has been discussed. Further, a few enzymes like alkaline phosphatase, acid phosphatase, 5-nucleotides and non-specific esterase have been localized in the diseased liver. The occurrence of lymphocytoma caused a marked change in the localization of the enzymes. Sometimes total inhibition of the enzyme was encountered. Damaged sinusoid cells and bile canaliculi of the neoplasm as well as liver lobules show no reaction for alkaline phosphatase. However, its counterpart, acid phosphatase, exhibits intense activity in both neoplastic tissue and liver cells. Aggregates of neoplastic tissue give moderate 5-nucleotidase reaction while it gives poor activity in hepatic tissue of the diseased liver. Parenchymatous cells are able to give some activity for the non-specific esterase while it is very dull in the neoplastic tissue.  相似文献

The protein component of light meromyosin [LMM-1] was shown earlier to relax glycerinated muscle fibres and actomyosin. Presently its influence on ATP-ase activity of myofibrils, actomyosin, myosin and heavy meromyosin has been studied. LMM-1 decreases Mg-ATP-ase activity of myofibrils and of reconstructed actomyosin by 25-- 30% and does not change [or slightly increases] Ca-ATP-ase activity of this protein and of myosin; besides LMM-1 is able to increase Mg-ATP-ase of HMM substantially. LMM-1 markedly inhibits [preliminary data] the activation of ATP-ase activity of HMM by actin. It is suggested that LMM-1 protein interacts with myosin and decreases the actin-myosin affinity, displacing actin out of the complex. It reacts only with one of the heads of myosin. Probably this suggestion can account for a relatively slight inhibition of ATP-ase activity of complex by LMM-1. LMM-1 represents a natural and specific inhibitor of Mg-AM-ATP-ase activity, included in the structure of myosin protofibrils and interacting with the myosin active site region.  相似文献

The induction of nephrotoxic nephritis in rats with rabbit antibodies preparation results in proteinuria, hypoproteinemia and hyperlipidemia with little glomerular lesions. A study of some hydrolases in cortex and medulla on one hand and glomerular and tubules on the other, showed changes in the activities of following enzymes. 1) A 20-30 % decrease in Na+, K+ dependent ATP-ase in whole kidney. 2) A 20 % decrease in beta-galactosidase activity in glomerular and medulla. 3) A 20 % increase of arylsulphatase A activity in tubules. These results are discussed in the light of the present knowledge of sulphatide metabolism in kidney.  相似文献

Summary The localisation of alkaline-, adenosine tri-, glucose-6- and acid phosphatase was studied in the juxtaglomerular complexes of rat, mouse and human kidneys. An alkaline-and adenosine triphosphatase active region was observed between the macula densa, Goormaghtigh cell group and in the interstitium of the latter. The adenosine triphosphatase activity extended into the lateral cell membranes of the macular cells and in properly incubated sections it did not appear among other distal tubular cells. The granular juxtaglomerular cells were ATP-ase negative. The cells of the human macula densa and the granular juxtaglomerular cells of the rat and mouse showed acid phosphatase activity. The glucose-6-phosphatase reaction, accomplished at acid and alkaline pH, was negative in the JG complex of all three species. The possible role of these enzymes in the function of the JG complex also has been discussed.  相似文献

Most of the biological processes in the living organisms of both animals and man are known to be of rhythmical nature. Variability of enzymatic activity in circadian cycle depends on many factors among other on age, sexual maturity, diet as well as medication. The results obtained in our studies indicate, that the activity changes of acid phosphatase and ATP-ase Mg++ dependent in the liver of all the examined age groups were of 24 hour circadian rythm. As to the acid phosphatase activity the results of this experiments showed that in circadian cycle in all examined age groups there is only one peak of elevated activity. ATP-ase Mg++ dependent showed two activity peaks appearing at the same hour both in 30 and 60 days old animals. It should be noticed that the activities of ATP-ase Mg++ dependent in 100 day old animals were two times higher than in 30 and 60 days old rats.  相似文献

A histochemical study has been carried out on the eye muscles of the carp. On the base of the ATP-ase and SDH activity and with regard to the localization and diameter of muscle fibres six types of muscle fibres situated in the defined zones can be distinguished. The type 1 and 2 fibres display a moderately high ATP-ase activity at pH 9.4 and rather low activity after alkaline and acid preincubation. Type 1 shows a high SDH activity in contrast to type 2 with a low SDH activity. The other four types of fibres have the high ATP-ase activity at pH 9.4. Type 3 contains fibres with a moderately high ATP-ase activity after alkaline preincubation with a rather low activity after acid preincubation and with a low SDH activity. The fibres of type 4 characterized by the high ATP-ase activity after alkaline and acid preincubation and by the high SDH activity. The fibres of type 5 display high ATP-ase activity after alkaline and acid preincubation and the low SDH activity. They are situated in the white and intermediate fibre zones. The fibres of type 6 are comparable to the fibres of type 5, however they differ diameter and localization, i.e. they are situated in small diameter fibre zone. Using the electron microscope four types of fibres (A, B, C, and D) are found. They vary in the localization of T system, the organization of Z-line and in M-line appearance. In the type B of muscle fibres two different localizations of T system have been discerned.  相似文献

Summary The multiple forms of ATP-ase activity in maize root tips have been examined by separation and staining using polyacrylamide gel electrophoresis. The band pattern obtained using a whole homogenate supernatant fraction was compared with the pattern obtained from various cell organelle fractions. The kinetic properties of the ATP-ase activity in various fractions were also examined. The results support the view that ATP-ase activity exists in a number of molecular forms in the same tissue and that these forms may show specific intracellular localizations.  相似文献

1. Stomach and pyloric caeca homogenates from the crown-of-thorns starfish hydrolysed p-nitrophenyl esters, alpha-naphthyl esters, cholesteryl oleate and tributyrin. The pyloric caeca contained the highest activities. 2. The p-nitrophenyl acetate hydrolytic activity eluted at 0.23 M NaCl on ion exchange chromatography while the p-nitrophenyl palmitate hydrolytic activity eluted between 0.2 and 1.0 M NaCl. 3. Polyacrylamide gel zymograms for alpha-naphthyl acetate hydrolytic activity revealed one major band and several minor bands of activity for both tissues. 4. Isoelectric focusing zymograms revealed one major band with a pI = 4.2 for both tissues, with an additional band at pI = 3.5 for pyloric caeca. 5. The pyloric caeca contained twice as much lipid as the stomach. Lipid extracts contained mixtures of steroids and steroid-esters; a cholesterol-like sterol was tentatively identified.  相似文献
