This study investigated how CO2and temperature affect dry weight(d.wt) accumulation, total nonstructural carbohydrate (TNC)concentration, and partitioning of C and N among organs of twoimportant grasses of the shortgrass steppe,Pascopyrum smithiiRydb. (C3) andBouteloua gracilis(H.B.K.) Lag. ex Steud. (C4).Treatment combinations comprised two temperatures (20 and 35°C)at two concentrations of CO2(380 and 750 µmol mol-1),and two additional temperatures of 25 and 30°C at 750 µmolmol-1CO2. Plants were maintained under favourable nutrient andsoil moisture and harvested following 21, 35, and 49d of treatment.CO2-induced growth enhancements were greatest at temperaturesconsidered favourable for growth of these grasses. Comparedto growth at 380 µmol mol-1CO2, final d.wt of CO2-enrichedP.smithiiincreased 84% at 20°C, but only 4% at 35°C. Finald.wt ofB. graciliswas unaffected by CO2at 20°C, but wasenhanced by 28% at 35°C. Root:shoot ratios remained relativelyconstant across CO2levels, but increased inP. smithiiwith reductionin temperature. These partitioning results were adequately explainedby the theory of balanced root and shoot activity. Favourablegrowth temperatures led to CO2-induced accumulations of TNCin leaves of both species, and in stems ofP. smithii, whichgenerally reflected responses of above-ground d.wt partitioningto CO2. However, CO2-induced decreases in plant tissue N concentrationswere more evident forP. smithii. Roots of CO2-enrichedP. smithiihadgreater total N content at 20°C, an allocation of N below-groundthat may be an especially important adaptation for C3plants.Tissue N contents ofB. graciliswere unaffected by CO2. Resultssuggest CO2enrichment may lead to reduced N requirements forgrowth in C3plants and lower shoot N concentration, especiallyat favourable growth temperatures. The influence of hydrogenase in Bradyrizobum-Phaseoleae symbioseswas studied ex-planta and in-planra in soybean (Glycine max)and cowpea (Vigna unguiculata). The hydrogenase was activatedby the addition of hydrogen in the incubation gas phase whichmodified the response of nitrogenase activity of Hup+ (hydrogenuptake positive) symbiosis to the external oxygen partial pressure.For bacteroids the hydrogenase expression increased nitrogenaseactivity at supraoptimal pO2, acting possibly as a respiratoryprotection of nitrogenase. However, at suboptimal pO2, nitrogenaseactivity of Hup+ bacteroids decreased with hydrogen, a phenomenonattributed to the lower efficiency of ATP synthesis from hydrogenthan from carbon substrates oxidation. For undisturbed nodules,the hydrogenase expression in soybean increased the optimalpO2 for ARA (COP), from 35.3 to 40.3 kPa O2, and the ARA atsupraoptimal pO2; at suboptimal PO2 there was a negative effectof hydrogenase on ARA, although this inhibition was less thanon bacteroids and was not detected if plants were grown at 15°C rather than 20 °C root temperature. No H2 effectwas detected on cowpea nodules. The results on soybean nodulesare consistent with the concept that symbiotic nitrogen fixationis oxygen-limited and that hydrogenase activity has no beneficialeffect on nitrogen fixation in O2 limitation. Pascopyrum smithii (C3) andBouteloua gracilis (C4) are importantforage grasses native to the Colorado shortgrass steppe. Thisstudy investigated photosynthetic responses of these grassesto long-term CO2enrichment and temperature in relation to leafnonstructural carbohydrate (TNC) and [N]. Glasshouse-grown seedlingswere transferred to growth chambers and grown for 49 d at twoCO2concentrations (380 and 750 µmol mol-1) at 20 and 35°C, and two additional temperatures (25 and 30 °C) at750 µmol mol-1CO2. Leaf CO2exchange rate (CER) was measuredat a plant's respective growth temperature and at two CO2concentrationsof approx. 380 and 700 µmol mol-1. Long-term CO2enrichmentstimulated CER in both species, although the response was greaterin the C3,P. smithii . Doubling the [CO2] from 380 to 750 µmolmol-1stimulated CER ofP. smithii slightly more in plants grownand measured at 30 °C compared to plants grown at 20, 25or 35 °C. CO2-enriched plants sometimes exhibited lowerCER when compared to ambient-grown controls measured at thesame [CO2], indicating photosynthetic acclimation to CO2growthregime. InP. smithii , such reductions in CER were associatedwith increases in TNC and specific leaf mass, reductions inleaf [N] and, in one instance, a reduction in leaf conductancecompared to controls. InB. gracilis , photosynthetic acclimationwas observed more often, but significant changes in leaf metabolitelevels from growth at different [CO2] were generally less evident.Temperatures considered optimal for growth (C3: 20 °C; C4:35 °C) sometimes led to CO2-induced accumulations of TNCin both species, with starch accumulating in the leaves of bothspecies, and fructans accumulating only inP. smithii. Photosynthesisof both species is likely to be enhanced in future CO2-enrichedand warmer environments, although responses will sometimes beattenuated by acclimation. The two parameters of the hyperbolic tangent equation, Pm and, were estimated from in situ vertical profiles of primary productionusing mesocosm data along a nutrient gradient. The parameters,derived from 4-h (around noon) 14C incubations, were used togetherwith the photosynthesis-light curve and hourly solar radiationdata to calculate daily primary production rates (Pd). Approximately40% of the daily production occurred in the 4 h around noon.Considering parameter uncertainty, there was no indication ofan increase in variation in production with increased nutrientloading, nor did biomass-specific P-I parameters increase. Annualproduction ranged from 82 to 901 g C m–2 year–1and was highest in the highest nutrient treatment tank. Dailyproductivity ranged from 0.02 to 9.1 g C m–2 day–1and was significantly correlated, in all treatments, with acomposite parameter BI0/k (where B is phytoplankton biomass;I0 is daily radiation and k is the extinction coefficient).Linear regressions of Pd against BI0/k indicated that much ofthe variability (86%) in productivity was explained by lightavailability and phytoplankton biomass. Two approaches for predictingproductivity were compared: (i) predicting production directlyfrom environmental variables (i.e. BI0/k) and (ii) predictingthe parameters of the P-I curve from environmental variablesand using these to calculate daily production.  相似文献

The uptake of N by phytoplankton is generally estimated usingthe 15N technique and, under some circumstances, the uptakeof C is estimated using 13C. Rigorous examination of formulaefor computing net transport rates leads to several interestingand even unexpected conclusions. These are that the 15N or 13Ctechnique formula for computing net transport rates () is identicalto that of the 14C technique, in spite of apparent dissimilaritieswhich reflect differences in equipment used for determiningnon-radioactive and radioactive isotopes; the so-called specificuptake rates (V) should not be used with natural samples, exceptas a step in the calculation of transport rates (); estimationof is unaffected by the presence/absence of non-phytoplanktonicpaniculate organic matter (POM) in the incubated sample; thepractice of adding the concentration of tracer to the denominatorof expression representing the concentration of tracer in thedissolved phase at the beginning of incubation should be discontinued;and the concentration of POM should be determined from the inoculatedsample at the end of incubation (or, alternatively, from a sampleincubated in parallel) and not from a water sample taken atthe beginning of the incubation.  相似文献

用刺激隐核虫(Cryptocaryon irritans)的幼虫对卵形鲳鲹(Trachinotus ovatus)进行腹腔注射和体表感染,然后每隔1周用阻动试验(Immobilization assay)检测免疫鱼的抗血清和皮肤培养液对激刺隐核虫幼虫的阻动效价,在第14周,分别用亚致死剂量和致死剂量的刺激隐核虫幼虫对免疫鱼攻毒以检测所产生的免疫保护力。实验结果显示:两种免疫方法都能让卵形鲳鲹的血清和皮肤生成阻动刺激隐核虫幼虫的特异性抗体,并能使被免疫鱼获得明显的免疫保护,但是体表感染免疫组的血清和皮肤培养液的阻动效价都要比腹腔注射免疫组高,所获得的免疫保护力也更强。同时还发现,免疫鱼血清和皮肤培养液中的抗体存在明显的差异:两者的最初生成时间、达到峰值的时间、变化规律以及阻动效价等都不一致。因此,我们推测鱼类的系统免疫应答和皮肤粘膜免疫应答有可能是相互独立的,或者是不同步的。鱼类的体液免疫应答,特别是粘膜免疫应答对抵御刺激隐核虫的感染起了重要的作用,采用刺激隐核虫虫体疫苗可能成为预防海水鱼类白点病的一种选择。  相似文献

The R- and S-enantiomers of racemic [2-14C]Me 1', 4'-cis-diolof abscisic acid have been separated by high performance liquidchromatography on an optically-active Pirkle column. R-[2-14C]-and S-[2-14C]abscisic acids, formed from the Me 1', 4'-cis-diolby oxidation and alkyline hydrolysis were fed to tomato shootsand the extracts analysed by reversed phase high performanceliquid chromatography. R-[2-14C]abscisic acid formed mainlythe abscisic acid glucose ester (ABAGE), abscisic acid l'-glucoside(ABAGS) and an uncharacterized conjugate. Dihydrophaseic acid4'-B-D-glucoside, the major metabolite of RS-abscisic acid intomato shoots, was found to be derived virtually exclusivelyfrom the natural, S-abscisic acid. Phaseic acid and conjugatesof abscisic acid were also found as products of the naturallyoccurring enantiomer. The resolution method was used to measurethe relative proportions of R and S enantiomers in the freeacid liberated from conjugates formed from RS-[2-14C]ABA fedto shoots. The ratios show an excess of the R-enantiomer: 5.8:1, ABAGE; 29.4: 1, ABAGE; 8.3: 1 for an uncharacterized conjugateand 6.1: 1 for the residual free [2-14C]ABA. Key words: ABA, HPLC, Tomato  相似文献

During the 1st GAP Workshop at Konstanz in April 1982 comparativemeasurements of phytoplankton primary production by severaltechniques were conducted simultaneously at an offshore stationin Lake Konstanz and an experimental algal pond. Suspended glassbottle exposure techniques using 14C and 13C uptake gave Pz(mg C m–3 h–1) values which varied considerablynear-surface, but estimates of areal rates for the euphoticzone Pcu(mg C m–3 h–1) which were reasonably close.In the lake, Pz, from a vertical tube exposure (with 14C uptake)was greater than rates derived for integrated bottle samples.The oxygen bottle method permitted a good estimate of compensationdepth, corresponding to in situ growth studies. There were difficultiesin direct comparison between O2 and carbon methods. Correlationbetween them for Pz was good in the lake but poor in the pond,both for suspended bottle and vertical tube methods. Phosphoenolpyruvate carboxylase (PEPC) (EC 4.1.1.31 [EC] ) assayedin extracts of Panicum maximum Jacq. loses up to 50% of itsactivity after incubation for 60 minutes at 0C while the enzymefrom P. miliaceum L. is completely stable under these conditions.Following dilution at room temperature the enzyme from P. maximumis labile, while that from P. miliaceum is stable. The P. maximumenzyme can be largely stabilized against dilution and againstcold-inactivation by D2O which stabilizes hydrophobic bondsand the compatible solutes proline, betaine and trimethylamine-N-oxide.Mineral ions, previously demonstrated to be protective againstcold inactivation of pyruvate, Pi dikinase from maize, provideno protection of P. maximum PEPC against either cold or dilution.The chaotropic ion SCN- causes partial inactivation of the enzymefrom P. miliaceum in the cold. The possible interrelationshipbetween inactivation by dilution and inactivation by cold isdiscussed. A study was made of the incorporation of 14C by intact leavesof Coffea arabica (cultivars Mundo Novo, Catuai, 1130–13,and H 6586–2) and Coffea canephora (cultivar Guarini)supplied with gas mixtures containing 14CO2 under controlledconditions. Samples of the leaves were combusted and the 14Cin the CO2 produced measured using a liquid scintillation counter.The results were used to estimate photosynthetic rates. Theeffects of changing the partial pressures of O2 and CO2 on thephotosynthetic rate were studied and estimates made of the CO2compensation point and photorespiration. The data obtained show differences between the mean net photosyntheticrates of the C. arabica cultivars (6·14 mg CO2 dm–2h–1) and the mean rate for the C. canephora cultivar (3·96mg CO2 dm–2 h–1). The cultivar of the latter speciesphotorespired more rapidly than the cultivar Catuai of C. arabica.Rates of photosynthesis in coffee measured using the 14CO2 methodwere similar to rates obtained by others using an infrared gasanalyser. The 14CO2 method proved to be reliable for photosyntheticmeasurements and the apparatus is suitable for use in fieldconditions.  相似文献

A high-rate oxidation pond is studied as a model system forcomparing 14C and oxygen evolution methods as tools for measuringprimary productivity in hypertrophic aquatic systems. Our resultsindicate that at very dense algal populations (up to 5 mg chl.a l–1) and high photosynthetic rates, 14C based resultsmay severely underestimate primary productivity, unless a wayis found to keep incubation times very short. Results obtainedwith our oxygen electrode were almost an order of magnitudehigher than those obtained by all 14C procedures. These highervalues correspond fairly well with a field-tested computer-simulationmodel, as well as with direct harvest data obtained at the samepond when operated under similar conditions. The abundance, life span, growth and production of the mud snailsHydrobia minoricensis, H. ulvae and H. ventrosa in a semi-naturallagoon system were studied by taking monthly samples at threesites during 1991 and 1992. The most abundant species, H. minoricensisoccurred at mean densities of 12834 to 26264 snails m–2(10.7 to 25.8g dry weigh m–2), depending on the site.The least abundant species, H. ulvae, occurred at mean densitiesof 185 to 353 snails m–2 (3.2 to 2.2g dry weight m–2).The numerical abundance and biomass of the three Hydrobia specieswere positively related to the biomass of benthic macroalgae(P<0.01). Although H. ulvae egg capsules were recorded throughoutthe year, newly hatched snailsof this species were not observed,in contrast to the other two species. The early spring and summercohorts of H. minoricensis and H. ventrosa seemed to be themost numerous. The average life spans of these two species wereestimated to be about 18 and 13 months respectively. Annualproduction estimates for the whole lagoon system were 29.0 (6.3),5.5 (0.8) and 5.2 (1.0)g dry weight (ash-free dry weight) m–2yr–1 for H. minoricensis, H. ulvae and H. ventrosa respectively.The annual P/B ratio was about 2 for H. minoricensis and H.ventrosa. Deoxygibberellin C (DGC), a C/D ring-rearranged isomer of GA20,was shown to inhibit the conversion of [2,3-3H2]GA9 to [2-3H]GA4by gibberellin 3ß-hydroxylase from immature seedsof Phaseolus vulgahs. Deoxygibberellin C inhibited the promotionof growth by exogenously applied GA20 of rice (Oryza sativaL.) seedlings. Evidence is also presented that DGC is a competitiveinhibitor of the 3ß-hydroxylase from P. vulgaris.However, DGC only weakly inhibited the conversion catalyzedby the 3ß-hydroxylase from Cucurbita maxima at highconcentrations, and it did not inhibit the promotion of growthby exogenously applied GA9 of cucumber (Cucumis sativus) seedlings.These results suggest that the 3ß-hydroxylases fromP. vulgaris and C. maxima have different structural requirementswith respect to their substrates. 16-Deoxo-DGC also inhibitedcatalysis of the same conversion by 3ß-hydroxylasefrom P. vulgaris, and it slightly inhibited the conversion catalyzedby the enzyme from C. maxima. Several orphan G protein-coupled receptors, including GPR40, have recently been shown to be responsive to fatty acids. Although previous reports have suggested GPR40 detects medium- and long-chain fatty acids, it has been reported to be unresponsive to short chain fatty acids. In this study, we have heterologously expressed mouse GPR40 in Xenopus laevis oocytes and measured fatty acid-induced increases in intracellular Ca2+, via two electrode voltage clamp recordings of the endogenous Ca2+-activated chloride conductance. Exposure to 500 µM linoleic acid (C18:2), a long-chain fatty acid, stimulated significant currents in mGPR40-injected oocytes (P < 0.01, ANOVA), but not in water-injected control oocytes (not significant, ANOVA). These currents were confirmed as Ca2+-activated chloride conductances because they were biphasic, sensitive to changes in external pH, and inhibited by DIDS. Similar currents were observed with medium-chain fatty acids, such as lauric acid (C12:0) (P < 0.01, ANOVA), and more importantly, with short-chain fatty acids, such as butyric acid (C4:0) (P < 0.01, ANOVA). In contrast, no responses were observed in mGPR40-injected oocytes exposed to either acetic acid (C2:0) or propionic acid (C3:0). Therefore, GPR40 has the capacity to respond to fatty acids with chain lengths of four or greater. Respiration rate of the entire above-ground parts of field-grown8-year-old hinoki cypress [Chamaecyparis obtusa(Sieb. et Zucc.)Endl.] was measured at monthly intervals over a 5-year period,to evaluate the trend in proportion of maintenance and growthcomponents of respiration with stand development. Representativesample trees were selected for respiration measurements. Theannual respiration rates of individual sample trees were combinedand partitioned into maintenance and growth components by regressingspecific respiration rate on mean relative growth rate. Maintenanceand growth respiration coefficients generated in this way were5.2 mol CO2kg-1year-1and 39 mol  CO2kg-1, which are equivalentto 14.3 mg C kg-1C h-1(at mean annual air temperature of 15.1°C) and 0.94 kg C kg-1C. Considering the maintenance andgrowth respiration coefficients, and phytomass and phytomassincrement of individual trees in the stand, the maintenanceand growth respiration rates of the stand were estimated. This investigation included a 2-year monitoring program aimedat assessing the abundance and distribution of harmful marinephytoplankton along the Southern Adriatic coast of Italy. Monthlysampling was conducted from April 1995 to March 1997 at foursampling stations along four transects, to determine the temporaland spatial presence/abundance of the potentially toxic Pseudo-nitzschiaspecies. The study focussed on the most abundant taxa Pseudo-nitzschiacalliantha and Pseudo-nitzschia delicatissima, the identitiesof which were confirmed by TEM on cleaned net material. Thedistribution patterns of these potential Amnesic Shellfish Poisoning(ASP) toxin producers were statistically analysed by GeneralisedLinear Model ANalysis Of VAriance, Principal Component Analysis(GLM ANOVA, PCA) and Spearman’s correlation analysis inorder to address relationships between environmental variableand population dynamics. Abundances displayed horizontal andvertical structure in the study area. Inter-annual variabilitywas also observed for both species that appeared to responddifferently to the environmental factors investigated. Distributionsof P. calliantha showed a stronger seasonality and was morecorrelated with winter water conditions than P. delicatissima,which in turn exhibited a broader temporal distribution andappeared independent from major environmental constraints. Thisis the first report of the occurrence and dynamics of P. callianthaand P. delicatissima populations in Southern Adriatic coastalwaters. Bot. 38: 67–76 The permeabilities of the amines, ammonia (NH3), methylamine(CH3NH2) and ethylamine (CH3CH2NH2) in the giant-celled charophyteChara corallina (C. australis) R.Br. have been measured andcompared. The permeabilities were corrected for uptake fluxesof the amine cations. Based on net uptake rates, the permeabilityof ammonia was 6?4?0?93 µm s–1 (n = 38). The permeabilitiesof methylamine and ethylamine were measured in net and exchangeflux experiments. The permeabilities of methylamine were notsignificantly different in net and exchange experiments, norto that of ammonia (Pmethylamine = 6?0?0?49 µm s–1(n = 44)). In net flux experiments the apparent permeabilityof ethylamine was slightly greater than that of ammonia andmethylamine (Pethylamine, net = 8?4?1?2 µm s–1 (n= 40)) but the permeability of ethylamine based on exchangeflux data was significantly higher (Pethylamine, exchange =14?1?2 µm s–1 (n = 20)). Methylamine can be validlyused as an ammonium analogue in permeability studies in Chara. The plasmalemma of Chara has acid and alkaline bands; littlediffusion of uncharged amines would occur across the acid bands.The actual permeability of amines across the alkaline bandsis probably about twice the values quoted above on a whole cellbasis i.e. the permeability of ammonia across the permeablepart of the plasmalemma is probably about 12 µm s–1. Biosynthetic pathways of acyl moieties of capsaicinoid in intactCapsicum fruits and spheroplasts prepared from placentas ofCapsicum fruits were examined using a radioisotopic technique.In intact Capsicum fruits, L-[U-14C] valine was incorporatedinto capsaicin and dihydrocapsaicin, the acyl constituents ofwhich are even-number branched chain fatty acids, while L-[U-14C]leucine was incorporated into nordihydrocapsaicin and homodihydrocapsaicin,which have odd-number branched chain facty acids as the acylmoieties. The intermediates of the odd- and even-number branchedchain fatty acids were identified with GLC/GPC after the spheroplastshad been incubated with L-[U-14C] valine or L-[U-14C] leucine.After incubation with L-[U-14C] valine, isobutyric acid and8-methyl nonanoic acid were detected, while isopentanoic acidand 9-methyl decanoic acid were found after incubation withL-[U-14C] leucine. The involvement of a-ketoisovalerate or a-ketoisocaproatein the biosynthesis of acyl moieties of capsaicinoid was alsodemonstrated in vitro using cell-free extracts of the placentasof Capsicum fruits. Plants of watermelon [Citrullus lanatus(Thunb.) Matsum. &Nakai, cv. Early Yates] were grown for up to 3 months aftergermination in controlled environment cabinets, and variousaspects of vegetative growth and fruit development were measured.Effects of light intensity were studied by comparing growthat 8, 16 and 32 klx at constant temperature and daylength (25°C, 14 h). Effects of daylength were studied by comparing8, 14 and 24 h at constant light intensity and temperature (32klx, 25 °C), and effects of tem perature were studied bycomparing 20°, 25°, 30°, 35° and 40 °C atconstant light intensity and day- length (32 klx, 14 h). Withincreasing light intensity and daylength lateral growth waspromoted whereas main shoots were less affected. Increase intemperature above 25 °C resulted in longer main shoots andprolific lateral growth, due both to more and to longer laterals.Environmental differences had little effect on internode lengthbut did affect the size of basal leaves. However, an increasein total leaf area at higher temperatures or with Continuouslight was mainly due to more leaves rather than larger leaves.The presence of developing fruit greatly reduced vegetativegrowth of plants. Main shoot length, lateral growth, numberof leaves, and even size of individual leaves, were all reduced.This reduction did not apply to d. wt of whole plants. Fruitingplants were very efficient, on a leaf area basis, in accumulatingd. wt. At 25 °C at the two higher light intensities with14 h days the presence of one developing fruit was inhibitoryto the setting of any subsequent fruit. With short days or lowlight, more fruits were set but they were small. With continuouslight or high temperature more than one fruit could developand they were large.  相似文献
