Plasma levels of 22 endogenous amino acids were measured by ion-exchange chromatography in four species of snakes: Thamnophis sirtalis, T. radix, Aipysurus laevis, and Python molurus. Despite considerable interspecific variation in the amino acid composition, all species showed relatively high plasma concentrations of histidine, a feature apparently unique to reptiles. The renal handling of these amino acids was studied by renal clearance methods. As in other vertebrates, net tubular absorption of filtered amino acids predominated. However, net tubular secretion of taurine, cysteic acid and/or phosphoserine and beta-alanine was observed, with taurine being the predominant amino acid secreted. The percentage reabsorption of the total amino acids filtered by the snake kidneys ranged from 79 to 95%. Evidence for the postrenal absorption of amino acids in these reptiles is presented. In species that normally undergo hibernation (Thamnophis spp.), the ability of the kidney to reabsorb amino acids was depressed by cold acclimation. Cold acclimation significantly decreased plasma levels of all amino acids except taurine, whose concentration increased. The increase in plasma taurine level may have resulted from cellular osmoregulation. Under these conditions, renal excretion of taurine increased concomitantly with the increase in plasma taurine concentration.  相似文献

Uptake of amino acids by cultured neuroblastoma and astrocytoma cells was studied in the presence and absence ofl-histidine. Intracellularly accumulated histidine was assumed to induce accumulation of radioactively labeled amino acids from medium by means of exchange transport. Neuroblastoma cells accumulated more histidine than astrocytoma cells and were more sensitive to the enhancement of the uptake of other large neutral amino acids by histidine. Histidine also increased glutamic acid uptake in astrocytoma cells, but reduced it in neuroblastoma cells. The greatest differences between the cell lines in amino acid uptake without histidine were found with acidic amino acids (astrocytoma cells accumulated them more than neuroblastoma cells) and with taurine (the reverse was found). The uptake and exchange mechanisms for some neutral and acidic amino acids may thus be dissimilar in the plasma membranes of cultured cells of neuronal and glial origin.  相似文献

The effects of palmitate on intracellular and extracellular amino acid concentrations of cultured astrocytes was studied. Exposure of astrocytes to either 0.72 mM or 0.36 mM palmitate was associated with a significant reduction in the intracellular pool of glutamine and taurine. In contrast, the intracellular concentration of histidine, glycine, citrulline, isoleucine and leucine were increased in the presence of 0.72 mM palmitate. Comparable changes in the extracellular amino acid pool were not observed. The data suggest that palmitic acid, which accumulates in the brain during periods of anoxia, alters the metabolism of several amino acids in cultured astrocytes. (1) The free amino acids in human CSF from eighteen subjects have been determined. The analyses were performed on 0-75 ml of CSF by an ion exchange chromatographic method which is capable of detection to the 10?10 mole level. (2) The amino acids always found in readily detectable amounts were: taurine, threonine, serine, glutamine, glutamic acid, citrulline, glycine, alanine, α-NH2-n- butyric acid, valine, methionine, isoleucine, leucine, tyrosine, phenylalanine, ethanolamine, ornithine, lysine, histidine and arginine. Urea was present. Aspartic acid and cystine, though always present, occurred in small or trace amounts. Proline was found in four cases and tryptophan in thirteen cases. In addition, twelve unknown peaks were nearly always evident in every chromatogram. (3) Filtrates 10 times more concentrated than those used regularly were prepared from pooled CSF and analysed. These analyses clearly confirmed the presence of those amino acids which were normally in very low concentration and they also served to distinguish the twelve unknown compounds from confusion with baseline artifacts. (4) The distribution of free amino acids in CSF was different from their distribution in blood plasma. (5) Despite a variety of neurological conditions and a wide age span few marked deviations were found in any of the amino acid concentrations.  相似文献

Summary We studied the plasma amino acid profiles in four models of hepatic injury in rats. In partially hepatectomized rats (65% of liver was removed) we observed significant increase of aromatic amino acids (AAA; i.e. tyrosine and phenylalanine), taurine, aspartate, threonine, serine, asparagine, methionine, ornithine and histidine. Branched-chain amino acids (BCAA; i.e. valine, leucine and isoleucine) concentrations were unchanged. In ischemic and carbon tetrachloride acute liver damage we observed extreme elevation of most of amino acids (BCAA included) and very low concentration of arginine. In carbon tetrachloride induced liver cirrhosis we observed increased levels of AAA, aspartate, asparagine, methionine, ornithine and histidine and decrease of BCAA, threonine and cystine. BCAA/AAA ratio decreased significantly in partially hepatectomized and cirrhotic rats and was unchanged in ischemic and acute carbon tetrachloride liver damage. We conclude that a high increase of most of amino acids is characteristic of fulminant hepatic necrosis; decreased BCAA/AAA ratio is characteristic of liver cirrhosis; and decrease of BCAA/AAA ratio may not be used as an indicator of the severity of hepatic parenchymal damage.Abbreviations BCAA branched-chain amino acids (i.e. valine, leucine and isoleucine) - AAA aromatic amino acids (i.e. tyrosine and phenylalanine)  相似文献

In experiments with whole animals infested with a highly malignant strain of Ehrlich ascites tumor cells, serial concentrations of amino acids were determined for host plasma, ascitic fluid, and tumor cells, throughout tumor development. Concentration gradients of glutamine, asparagine, valine, leucine, isoleucine, phenylalanine, tyrosine, histidine, tryptophan, arginine, serine, methionine, and taurine from the host plasma toward the ascitic liquid were established; while on the other hand, concentration gradients from the ascitic liquid toward the plasma were established for glutamate, aspartate, glycine, alanine, proline, and threonine. With the exception of aspartate the concentrations of these amino acids were highest inside the cells. Arginine was the only amino acid not detected in tumor cells. In vitro incubations of tumor cells in the presence of glutamine and/or glucose, as the energy and nitrogen sources, confirmed the amino acid fluxes previously deduced from the observed relative concentrations of amino acids in plasma, ascitic liquid, and tumor cells, suggesting that glutamate, alanine, aspartate, glycine, and serine can be produced by tumors. These findings support that changes in amino acid patterns occurring in the host system are related to tumor development.  相似文献

—During postnatal growth the free amino acids pattern of rat retina differs at various developmental stages. The adult level for individual amino acids is reached on the 30th day of maturation. During differentiation the taurine, glutamic acid, GABA, glutamine, aspartic acid, glycine arginine, methionine and histidine levels increase while proline. alanine, ornithine and tyrosine decrease.  相似文献

In order to deplete tissue taurine, 2-guanidinoethanesulphonate, a structural analogue of taurine was administered in drinking water with taurine-free diet to adult rats for four weeks. As a consequence the taurine concentrations in the blood serum, liver, kidney, spleen, intestine, lung, heart, muscle and cerebellum fell by nearly one half. Threonine, serine, glycine, alanine, methionine, tyrosine, lysine and histidine concentrations increased in blood plasma. Similar changes were also discernible in the heart and muscle. In the kidney and the lung the concentrations of several other amino acids fell as well, though increments occurred in the threonine content in the kidney and in threonine, serine and methionine contents in the lung. Taurine was practically the only amino acid the level of which fell in the liver, spleen, intestine and cerebellum. These findings indicate that 2-guanidinoethanesulphonate combined with taurine-free diet effectively lowers tissue taurine levels, but its action is not specific to taurine. It may be used as a tool to elucidate the physiological functions of taurine in the body.  相似文献

The levels of free amino acids in the cerebral cortex of acute and chronic uremic rats were examined. Amino acids significantly elevated were aspartate, glutamine, glycine, histidine, ornithine, phenylalanine, phosphoethanolamine and taurine, whereas 1-methyl histidine and 3-methyl histidine were specifically detected in uremic rats. Glutamate, arginine and carnosine disclosed a significant reduction. There was no change in the concentrations of γ-aminobutyrate and alanine. The above findings were essentially identical in both acute and chronic uremia. It was conjectured that these changes of amino acid levels in the brain might participate in the progress of uremic encephalopathy.  相似文献

The free amino acid content in the cerebral cortex of rats administered caffeine orally, and with automutilation behavior similar to that observed in the Lesch-Nyhan syndrome, have been measured. Amino acids significantly elevated were taurine, histidine and aspartic acid, whereas tyrosine showed a significant reduction. There was no change in the concentration of -aminobutyric acid and glutamic acid. It has been conjectured that changes in amino acids levels in the cortex might be responsible for the pharmacological action of caffeine and for the progressive behavior abnormalities observed in these rats. Interestingly these results are similar to these found recently in experimental uremia.  相似文献

Free amino acid contents in skin extracts and influence of food and starvation on free amino acid content in skin mucus were analysed in sexually immature goldfish. Free amino acid concentration in skin mucus (91.1 mumol/g dry wt) was higher than in deep skin (54 mumol/g) or in whole skin (56.6 mumol/g) extracts. Free amino acid compositions were very similar in the latter extracts. They both differed from skin mucus extract in taurine, glutamic acid, glycine and histidine relative contents. Free amino acid composition in zooplankton used to feed goldfish was close to the composition found in corresponding skin mucus extracts, except in taurine content. Goldfish weighing 3 g (6 months old) and 17 g (1 year old) reared on zooplankton showed similar patterns of free amino acid composition in skin mucus. Comparison with free amino acid composition in skin mucus from goldfish fed on commercial food had big differences in glutamic acid, valine, methionine and lysine relative contents. During fasting, we observed an increase in the amount of mucus secreted and a concomitant decrease of the free amino acid concentration in the secretion. The origin of free amino acids found in skin mucus and their possible role in pheromonal and allelochemical communications of goldfish are discussed.  相似文献

AMINO ACIDS IN SYNAPTIC VESICLES FROM MAMMALIAN CEREBRAL CORTEX: A REAPPRAISAL   总被引:12,自引:11,他引:1  
Synaptic vesicles were prepared from rat cerebral cortex and separated by gel filtration from small molecular weight compounds contaminating this fraction. Electron microscopy of the vesicle suspension showed that vesicles were by far the most abundant morphological entities. The amino acid content of the purified vesicle fraction was examined and the two amino acids appearing in the most significant amounts were found to be taurine and glutamate. This amino acid pool was not osmotically sensitive as is the vesicular pool of ACh and remained attached to the vesicular protein after passage through Sephadex columns equilibrated in water. However, amino acids added to the vesicle fraction prior to passage through Sephadex did not become associated with this pool and this indicated that the vesicular pool was not likely to be an artifact due to the vesicular protein non-specifically adsorbing amino acids. The release of taurine from incubated synaptosome beds was studied and elevated medium K+ (56 mm ) was found to cause a small increase (36 per cent) in the amount of the taurine released to the medium. During the same experiments another physiologically active amino acid, glutamate, was released in more significant amounts, increasing in the medium by 186 per cent. The possible significance of the presence of taurine is discussed.  相似文献

Amino acid analyses of mitochondrial membranes are compared with the amino acid composition of whole mitochondria (Alberti, 1964) and found to be very similar except in the cystine content. The composition of the endogenous amino acids found in freshly prepared mitochondria has been established and shown to differ considerably from the amino acid composition of membranes or whole mitochondria. The amino acids produced during anaerobic incubation of mitochondria at pH7.4, on the other hand, resemble the membrane in composition, supporting the view that neutral proteinase activity is responsible for their appearance. Aerobic incubation produces a similar pattern of amino acids except that amino acids such as proline, serine, asparagine, glutamic acid and glutamine, which can be metabolically utilized under aerobic conditions, are present to a smaller extent. The presence of large relative concentrations of endogenous taurine, cysteic acid and oxidized glutathione and the accumulation of taurine during incubation is found. The selective retention of taurine and cysteic acid within the mitochondria is established. It is proposed that the first step in the degeneration of isolated mitochondria results from lipid hydroperoxide accumulation caused by the lack of glutathione reductase in isolated mitochondria.  相似文献

Summary The amino acid taurine plays an important trophic role during development and regeneration of the central nervous system. Other amino acid systems, such as those for glutamate and gamma-aminobutyric acid (GABA), are modified during the same physiological and pathological processes. After crushing the optic nerve, goldfish retinal explants were plated in the absence and in the presence of different amino acids and amino acid receptor agonists. The length and the density of the neurites were measured at 5 days in culture. Taurine increased the length and the density of neurites. Glutamate and glycine increased them at low concentration, but were inhibitors at higher concentration. The combination of N-methyl-D-aspartate (NMDA) and glycine produced a greater inhibitory effect than NMDA alone. NMDA or alpha-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid (AMPA) added simultaneously with taurine impaired the stimulatory effect of the latter. GABA stimulated the emission of neurites in a concentration dependent manner. Hypotaurine also elevated the length of neurites, but cysteinesulfinic acid did not produce a significant effect. The concentrations of taurine, glutamate and GABA were determined by HPLC with fluorescent detection in the retina of goldfish at various days post-crushing the optic nerve. The levels of taurine were significantly increased at 48 h after the crush, and were elevated up to 20 days. Glutamate level decreased after the lesion of the optic nerve and was still low at 20 days. GABA concentration was not significantly different from the control. The freshwater fish gobi Preccottus glehni survives after wintering in ponds frozen in winter till the very bottom. In adaptation of poikilothermal animals to wintering at near-zero temperatures, an essential role is played by free amino acids; accumulation of a large amount of some particular acid at the period of preparation to the state of hibernation can indicate the protective role of this acid in the low-temperature adaptation. In the present work it has been shown that as soon as by the end of August, in the gobi muscle, the taurine concentration increases three times as compared with that in July, the taurine pool after this reaching 50% of the total pool of free amino acids in the muscle tissue. During December and after the 3-month hibernation in ice, it exceeds the April and July levels 8 and 4 times, respectively, and amounts to 50% of the total free amino acid pool for muscle and to 40% for blood. Several days prior to the beginning of winter hibernation under natural conditions, both in blood and in muscle, there is revealed disappearance or a sharp fall of concentration of essential amino acids. An essential peculiarity of the change in the free amino acid composition after hibernation was a significant rise of alanine concentration in muscle—3.5 times as compared with July and 1.4–1.8 times as compared with changes in December. The total free amino acid pool in muscle in December as compared with that in July increased almost 1.5 times (34.76 ± 1.12 μmol/g wet weight), while after hibernation—2 times. Peculiarities of taurine accumulation long before the beginning of winter cold allow suggesting that role of taurine consists in providing a possibility of existence of eurythermal fish at near-zero temperature.  相似文献

Milkfish (Chanos chanos) decreased their body weight from 47 to 28 g over the 60-day period of starvation. Starvation also resulted in the reduction of muscle lipid and protein, and hepatosomatic index. The predominant free amino acid (FAA) in white muscle of milkfish was histidine, followed by taurine and glycine. In the first 25 days of starvation, no significant change in histidine was found. After 40 days of starvation, however, the histidine concentration was significantly decreased by 46%, and remained unchanged thereafter. As compared to control group fish, the 60-day-starved fish possessed only half the amount of histidine. Taurine and glycine, on the other hand, showed no significant changes throughout starvation. Taurine became the most predominant in the FAA pool after 40 days of starvation, and the concentration of 60-day-starved fish was two times higher than that of control group fish without starvation. The ratios of histidine, taurine, and glycine to total FAAs remained approximately the same although the individual contributions varied considerably to the total FAAs during starvation. The results of this study suggested that a good strategy would be to keep taurine and glycine in milkfish muscle at relatively high levels for physiological function as histidine decreased drastically for energy source under conditions of food deprivation.  相似文献

A rapid, specific, and sensitive radioassay for measuring bile acid CoA:glycine/taurine: N-acyltransferase (EC 2.3.1) has been developed. In this assay, 3H-labeled amino acids (glycine or taurine) are conjugated with unlabeled bile acid CoA derivatives to form 3H-labeled bile acid amidates. Following incubation, the 3H-labeled bile acid amidate is separated from the unreacted amino acid by an n-butanol extraction method. The extraction procedure was developed by evaluating the effects of buffer concentration and pH on the recovery of radiolabeled bile acid amidate standards in the presence of human hepatic cytosol. Highest recovery (greater than 90%) of bile acid amidate standards occurred under acidic conditions (pH 2) in the presence of 1% (w/v) SDS. When the radioassay and accompanying n-butanol extraction procedure were utilized to study the amidation of glycine or taurine with cholic acid in human hepatic cytosol, a single peak of radioactivity corresponding with either authentic glycocholate or taurocholate was detected in the n-butanol phase by high-performance liquid chromatography. This assay for bile acid CoA:glycine/taurine: N-acyltransferase activity was linear with incubation time and protein concentration. This assay should be useful in the biochemical studies of this enzyme, as well as in the examination of bile acid amidation in clinical liver specimens.  相似文献

We examined the free amino acid composition of hemolymph from representatives of five spider families with an interest in knowing if the amino acid profile in the hemolymph of orb-web-building spiders reflects the high demands for small organic compounds in the sticky droplets of their webs. In nearly all analyses, on both orb and non-orb builders, glutamine was the most abundant free amino acid. Glycine, taurine, proline, histidine, and alanine also tended to be well-represented in orb and non-orb builders. While indications of taxon-specific differences in amino acid composition were observed, it was not apparent that two presumptive precursors (glutamine, taurine) of orb web sticky droplet compounds were uniquely enriched in araneids (orb builders). However, total amino acid concentrations were invariably highest in the araneids and especially so in overwintering juveniles, even as several of the essential amino acids declined during this winter diapause. Comparing the data from this study with those from earlier studies revealed a number of discrepancies. The possible origins of these differences are discussed.  相似文献

The first-pass metabolism of dietary sulfur amino acids by the liver and the robust upregulation of hepatic cysteine dioxygenase activity in response to an increase in dietary protein or sulfur amino acid level gives the liver a primary role in the removal of excess cysteine and in the synthesis of taurine. Hepatic taurine synthesis is largely restricted by the low availability of cysteinesulfinate as substrate for cysteinesulfinate decarboxylase, and taurine production is increased when cysteinesulfinate increases in response to an increase in the hepatic cysteine concentration and the associated increase in cysteine dioxygenase activity. The upregulation of cysteine dioxygenase in the presence of cysteine is a consequence of diminished ubiquitination of cysteine dioxygenase and a slower rate of degradation by the 26S proteasome.  相似文献
