The cytochrome b 6 f complex occupies a central position in photosynthetic electron transport and proton translocation by linking PS II to PS I in linear electron flow from water to NADP+, and around PS I for cyclic electron flow. Cytochrome b 6 f complexes are uniquely located in three membrane domains: the appressed granal membranes, the non-appressed stroma thylakoids and end grana membranes, and also the non-appressed grana margins, in contrast to the marked lateral heterogeneity of the localization of all other thylakoid multiprotein complexes. In addition to its vital role in vectorial electron transfer and proton translocation across the membrane, cytochrome b 6 f complex is also involved in the regulation of balanced light excitation energy distribution between the photosystems, since its redox state governs the activation of LHC II kinase (the kinase that phosphorylates the mobile peripheral fraction of the chlorophyll a/b-proteins of LHC II of PS II). Hence, cytochrome b 6 f complex is the molecular link in the interactive co-regulation of light-harvesting and electron transfer. The importance of a highly dynamic, yet flexible organization of the thylakoid membranes of plants and green algae has been highlighted by the exciting discovery that a lateral reorganization of some cytochrome b 6 f complexes occurs in the state transition mechanism both in vivo and in vitro (Vallon et al. 1991). The lateral redistribution of phosphorylated LHC II from stacked granal membrane regions is accompanied by a concomitant movement of some cytochrome b 6 f complexes from the granal membranes out to the PS I-containing stroma thylakoids. Thus, the dynamic movement of cytochrome b 6 f complex as a multiprotein complex is a molecular mechanism for short-term adaptation to changing light conditions. With the concept of different membrane domains for linear and cyclic electron flow gaining credence, it is thought that linear electron flow occurs in the granal compartments and cyclic electron flow is localised in the stroma thylakoids at non-limiting irradiances. It is postulated that dynamic lateral reversible redistribution of some cytochrome b 6 f complexes are part of the molecular mechanism involved in the regulation of linear electron transfer (ATP and NADPH) and cyclic electron flow (ATP only). Finally, the molecular significance of the marked regulation of cytochrome b 6 f complexes for long-term regulation and optimization of photosynthetic function under varying environmental conditions, particularly light acclimation, is discussed.Abbreviations Chl chlorophyll - cyt cytochrome - PS Photosystem  相似文献

Coccolithophores impact the ocean carbon cycle principally through the generation of CO2 during CaCO3 production. Coccolithophore biomineralization has been examined most extensively in Pleurochrysis carterae and Emiliania huxleyi both of which produce mineralized scales—coccoliths—composed of elaborate calcite crystals attached to an underlying organic base plate. Calcification of preformed base plates is mediated by acidic polysaccharides and occurs in Golgi-derived structures known as mineralizing vesicles. In Pleurochrysis a high capacity calcium-binding polysaccharide PS2 is required for efficient nucleation of calcitic protocrystals. A galacturonomannan PS3 is required for the growth and transformation of the protocrystals into a massive double disc of calcite. The genes that regulate expression of the glycans have not yet been identified. In addition to the coccolith-bearing diploid phases, Pleurochrysis and Emiliania possess both haploid and diploid non-calcifying stages, which are self-perpetuating via binary fission. One non-calcifying Pleurochrysis phase fails to synthesis PS2 and spontaneously reverts to the mineralizing morphotype in laboratory cultures. As yet, there is little information on environmental factors that effect the expression or silencing of calcifying genes or favor the growth of calcifying over non-calcifying phases. These issues will need extensive investigation, if we are to appreciate the role of coccolithophores in the regulation of atmospheric CO2 levels.  相似文献

The photosynthetic activities of three planktonic desmid species (Staurastrum brachiatum, Staurodesmus cuspidatus var. curvatus, and Staurastrum chaetoceras) were compared after adaptation to medium enriched with either a 20 mM Na+-phosphate (P) or HEPES buffer. Incubations up to 2 d were carried out at pH 6 or 8 under normal air or air enriched with 5 % CO2. Gross maximum photosynthetic rate (P max) and growth rate were decreased in both S. brachiatum and Std. cuspidatus at higher pH when using the HEPES buffer and this effect was independent of CO2 concentration, indicating that pH had an inhibitory effect on photosynthesis and growth in these species. The P-buffer at pH 8 caused a large decrease in P max and quantum yield for charge separation in photosystem 2 (PS2), compared to HEPES-buffered algae. This effect was very large in both S. brachiatum and Std. cuspidatus, two species characteristic of soft water lakes, but also significant in S. chaetoceras, a species dominant in eutrophic, hard water lakes. The decreased P max in P-buffer could not be related to a significant increase in cellular P content known to be responsible for inhibition in isolated chloroplasts. Experiments at pH 6 and 8 showed that two conditions, high pH and high Na+ concentration, both contributed to the decreased P max and quantum yield in the desmids. Effects of a P-buffer were less pronounced by using K+-P buffer. The use of P-buffer at pH 8 possibly resulted in high irradiance stress in all species, indicated by damage in the PS2 core complex. In the soft water species pH 8 resulted in increased non-photochemical quenching together with a high de-epoxidation state of the xanthophyll cycle pigments.  相似文献

Effects of salt and alkali stresses on growth, osmotic adjustment and ionic balance of Suaeda glauca (Bge.), an alkali-resistant succulent halophyte, were compared. The results showed that alkali stress clearly inhibited the growth of S. glauca. Moreover, the concentrations of Na+ and K+ both increased with increasing salinity under both stresses, suggesting no competitive inhibition between absorptions of Na+ and K+. The mechanism underlying osmotic adjustment during salt stress was similar to alkali stress in shoots. The shared essential features were that organic acids, betaine and inorganic ions (dominated by Na+) mostly accumulated. On the other hand, the mechanisms governing ionic balance under both stresses were different. Under salt stress, S. glauca accumulated organic acids and inorganic anions to maintain the intracellular ionic equilibrium, but the anion contribution of inorganic ions was greater than that of organic acids. However, the concentrations of inorganic anions under alkali stress were significantly lower than those under salt stress of the same intensity, suggesting that alkali stress might inhibit uptake of anions, such as NO3 and H2PO4 . Under alkali stress, organic acids were the dominant factor in maintaining ionic equilibrium. The contribution of organic acids to anions was 74.1%, while that of inorganic anions was only 25.9%. S. glauca enhanced the synthesis of organic acids, dominated by oxalic acid, to compensate for the shortage of inorganic anions.  相似文献

Cross stress of heat and high irradiance (HI) resulted in the accumulation of active oxygen species and photo-oxidative damage to photosynthetic apparatus of wheat leaves during grain development. Pre-treatment with calcium ion protected the photosynthetic system from oxidative damage by reducing O-. 2 production, inhibiting lipid peroxidation, and retarding electrolyte leakage from cell. Therefore, high Fv/Fm [maximal photochemical efficiency of photosystem 2 (PS2) while all PS2 reaction centres are open], Fm/F0 (another expression for the maximal photochemical efficiency of PS2), ΦPS2 (actual quantum yield of PS2 under actinic irradiation), qP (photochemical quenching coefficient), and P N (net photosynthetic rate) were maintained, and lower qNP (non-photochemical quenching coefficient) of the leaves was kept under heat and HI stress. EGTA (a chelant of calcium ion) and LaCl3 (a blocker of Ca2+ channel in cytoplasmic membrane) had the opposite effect. Thus Ca ion may help protect the photosynthetic system of wheat leaves from oxidative damage induced by the cross stress of heat and HI.  相似文献

Two green algal species, Chlamydomonas reinhardtii and Scenedesmus obliquus, exhibited a relative maximum during the decay of luminescence, when adapted to low CO2 conditions that was not observed in high CO2 adapted cells. From the kinetics of transient changes in the level of dark fluorescence, after illumination and parallel to the luminescence maxima, it was concluded that the maximum in Scenedesmus was mainly related to a decrease in nonphotochemical quenching, whereas in Chlamydomonas the maximum was mainly related to a dark reduction of the primary PS II acceptor QA.ATP/ADP ratios from low CO2 adapted Scenedesmus showed transient high levels after a dark/light transition that was not observed in high CO2 adapted cells. After 30 s of illumination the ATP/ADP ratios however stabilized at the same steady state level as in high CO2 adapted cells.Dark addition of HCO3 - to low CO2 adapted cells of Chlamydomonas resulted in a rapid transient quenching of luminescence that was not observed in low CO2 adapted cells of neither species. It is concluded that the luminescence maxima present in both low CO2 adapted Scenedesmus and Chlamydomonas reflect adaptation of the cells to low CO2 conditions. It is further suggested that the difference in mechanistic origin of luminescence maxima in the two species reflects differences in adaptation.Abbreviations ADP adenosine-diphosphate - ATP adenosine-triphosphate - Ci inorganic carbon - FD dark fluorescence recorded under dark adapted conditions - F0 fluorescence with all reaction centers open - FV variable fluorescence - PS I photosystem I - PS II photosystem II - QA the first quinone acceptor of PS II  相似文献

In situ photosynthetic activity in the green algal and the cyanobacterial photobionts of Placopsis contortuplicata was monitored within the same thallus using chlorophyll a fluorescence methods. It proved possible to show that the response to hydration of the green algal and the cyanobacterial photobionts is different within the same thallus. Measurements of the photochemical efficiency of PS II, Fv/Fm, reveal that in the dry lichen thallus photosynthetic activity could be induced in the green algal photobiont by water vapour uptake, in the cyanobacterial photobiont only if it was hydrated with liquid water. However, rates of apparent electron flow through PS II as well as rates of CO2 gas exchange were suboptimal after hydration with water vapour alone and maximum rates could only be observed when the thallus was saturated with liquid water. The differences in the waterrelated photosynthetic performance and different light response curves of apparent electron transport rate through PS II indicate that the two photobionts act highly independently of each other. It was shown that the cyanobacteria from the cephalodia in P. contortuplicata act as photobiont. The rate of electron flow through PS II was found to be saturated at 1500 mol photon m–2 s–1, despite a considerable increase of non-photochemical quenching in the green algal photobiont which is lacking in the cyanobacterial photobiont. No evidence of photoinhibition could be found in either photobiont. Pronounced competition between the green algal and the cyanobacterial thallus can be observed in the natural habitat, indicating that the symbiosis in P. contortuplicata should be regarded as a very variable adaptation to the extreme environmental conditions in the maritime Antarctic.Abbreviations DR dark respiration - ETR apparent rate of electron flow of PS II (=F/Fm×PFD) - F difference in yield of fluorescence and maximal Fm and steady state Fs under ambient light - Fo minimum level of fluorescence yield in dark-adapted state - Fo minimum level of fluorescence yield after transient darkening and far-red illumination - Fm maximum level of dark-adapted fluorescence yield - Fm maximum yield of fluorescence under ambient light - Fs yield of fluorescence at steady state - Fv difference in minimum fluorescence and maximum fluorescence in dark-adapted state - NP net photosynthesis - NPQ coefficient for non-photochemical quenching - PAR photosynthetically active radiation (400–700 nm) - PFD photon flux density in PAR - PS II photosystem II - qN coefficient for non-photochemical quenching - qP coefficient for photochemical quenching  相似文献

The lower limits of photosynthetically useable radiation at which growth and photosynthesis can occur establish the lower boundaries for the extent of photolithotrophy in the biosphere. Photolithotrophic growth denotes the capacity to grow with photons as the sole energy input. Slippage in terms of photosynthetic energy conversion implies a less than theoretical stoichiometry of energy-transduction process(es) such as the dissipation of intermediates of O2 evolution and of ATP synthesis (H+/e and H+/ATP ratios). Slippage is particularly important in limiting the growth of photolithotrophic organisms at very low photon fluence rates. We found that Dunaliella tertiolecta and Phaeodactylum tricornutum avoid such reductions in photon use efficiency by increasing the size and number of their photosynthetic units, respectively, and by altering QA reduction kinetics on the reducing side of PS II. P. tricornutum is also less susceptible to slippage in terms of the breakdown of intermediates in its O2 evolution pathway than D. tertiolecta. Minimizing H+ leakage through the CF0–CF1 ATP synthetase (and other H+ porters) is also discussed briefly. In combination, strategies employed by P.␣tricornutum effectively allow it to grow and photosynthesize at lower rates of energy input than D. tertiolecta, consistent with our observations. Differences in the responses of the photosynthetic apparatus of these two marine microalgae are mechanistic and probably representative of evolutionary divergences associated with strategies for dealing with environmental perturbations.  相似文献

A chlorophyll b-less mutant of Chlamydomonas reinhardtii (Pg 27) was isolated after UV irradiation of the wild type cells. This photosynthetically competent mutant totally lacks chlorophyll b and the CP2 chlorophyll-protein complex. However, SDS-PAGE, proteolytic digestions and immunodetections demonstrated that the 24–25 Kd apoproteins of the lacking CP2 complex are still present in thylakoids of the Pg27 mutant. It is concluded that this CP2-less mutant is affected in the biosynthesis pathway of chlorophyll b.This CP2-less mutant was crossed with a CP1-less mutant (Fl5) Fluorescence emission spectra and fluorescence inductions in the presence of DCMU were analysed in the resulting (cp 2 , cp 1 + ), (cp 2 + , cp 1 ), (cp 2 + , cp 1 + ), cp 2 , cp 1 )tetratype. Differences in PS 2 optical cross section and in the relative amplitude or localisation of fluorescence emission peaks fit well with a quadripartite model where PS1 and PS2 would each correspond to a reaction centre core complex (CP1 and CP2 respectively) associated to a light harvesting antenna (LHC1 and LHC2 respectively). The occurrence of energy transfers from PS1 peripheral antenna to PS2 in the Fl 5 mutant shows that, in absence of CP1, at least a part of its associated PS1 light harvesting antenna migrates in the PS2 containing appressed thylakoids.Abbreviations Chl Chlorophyll - LHC Light harvesting chl a/b complex - CP2 Predominant form of LHC or SDS polyacrylamide gels - WT Wild type - DM Double mutant (cp 1 , cp 2 ) - SDS-PAGE sodium dodecyl sulfate polyacrylamide gel electrophoresis - DOC-PAGE Deoxycholate polyacrylamide gel electrophoresis  相似文献

The photoprotection of energy dissipation and water-water cycle were investigated by comparing chilling sensitivity of photosystems 2 (PS2) and 1 (PS1) in two chilling-sensitive plants, cucumber and sweet pepper, upon exposure to 4 °C under low irradiance (100 μmol m−2 s−1) for 6 h. During chilling stress, the maximum photochemical efficiency of PS2 (Fv/Fm) decreased only slightly in both plants, but the oxidisable P700 decreased markedly, which indicated that PS1 was more sensitive to chilling treatment under low irradiance than PS2. Sweet pepper leaves had lower Fv/Fm, higher non-photochemical quenching (NPQ), and higher oxidisable P700 during chilling stress. Activity of superoxide dismutase (SOD) and ascorbate peroxidase (APX) in cucumber leaves was higher, but APX activity decreased apparently compared to that at room temperature. The productions of active oxygen species (H2O2, O2 ) increased in both plants, faster in cucumber leaves than in sweet pepper leaves. In sweet pepper leaves, a stronger de-epoxidation of the xanthophyll cycle pigments, a higher NPQ could act as a major protective mechanism to reduce the formation of active oxygen species during stress. Thus sensitivity of both plants to chilling under low irradiance was dominated by the protective mechanisms between PS1 and PS2, especially the energy dissipation and the water-water cycle. We examined photosynthetic activities and thermostability of photosystem 2 (PS2) in leaves of elm (Ulmus pumila) seedlings from initiation to full expansion. During leaf development, net photosynthetic rate (P N) increased gradually and reached the maximum when leaves were fully developed. In parallel with the increase of P N, chlorophyll (Chl) content was significantly elevated. Chl a fluorescence measurements showed that the maximum quantum yield of PS2 (ϕPS2), the efficiency a trapped exciton, moved an electron into the electron transport chain further than QA o), and the quantum yield of electron transport beyond QAEo) increased gradually. These results were independently confirmed by our low irradiance experiments. When subjected to progressive heat stress, the young leaves exhibited considerably lower ϕPS2 and higher minimal fluorescence (F0) than the mature leaves, revealing the highly sensitive nature of PS2 under heat in the newly initiating leaves. Further analysis showed that PS2 structure in the newly initiating leaves was strongly altered under heat, as evidenced by the increased fluorescence signals at the position of the K step. We therefore demonstrated an inhibition in the oxygen-evolving complex (OEC) in the young leaves. This resulted in decrease in amount of the functional PS2 reaction centres and relative increase in the PS2 reaction centres with inhibited electron transport at the acceptor side under heat. We suggest that the enhanced thermostability of PS2 during leaf development is associated with improved OEC stability.  相似文献

The species specific response of photosystem 2 (PS2) efficiency and its thermotolerance to diurnal and seasonal alterations in leaf temperature, irradiance, and water relations were investigated under alpine field conditions (1 950 m) and in response to an in situ long-term heat treatment (+3 K). Three plant species were compared using the naturally occurring microstratification of alpine environments, i.e. under contrasting leaf temperatures but under similar macroclimatic conditions. Thermotolerance of PS2 showed a high variability in all three species of up to 9.6 K. Diumal changes (increases or even decreases) in PS2 thermotolerance occurred frequently with a maximum increase of +4.8 K in Loiseleuria procumbens. Increasing leaf temperatures and photosynthetic photon flux density influenced thermotolerance adjustments. Under long-term heating (+3 K) of L. procumbens canopies with infra-red lamps, the maxima of the critical (Tc) and the lethal (Tp) temperature of PS2 increased by at least 1 K. Thermotolerance of the leaf tissue (LT50) increased significantly by +0.6 K. The effects of slight water stress on thermotolerance of PS2 were species specific. High temperature thresholds for photoinhibition were significantly different between species and increased by 9 K from the species in the coldest microhabitat to the species in the warmest. Experimental heating of L. procumbens canopies by +3 K caused a significant (p>0.01) upward shift of the high temperature threshold for photoinhibition by +3 K. Each species appeared to be very well adapted to the thermal conditions of its microhabitat as under the most frequently experienced daytime leaf temperatures no photoinhibition occurred. The observed fine scale thermal adjustment of PS2 in response to increased leaf temperatures shows the potential to optimise photosynthesis under varying environmental conditions as long as the upper thermal limits are not exceeded.  相似文献

In our search for new natural photosynthetic inhibitors that could lead to the development of “green herbicides” less toxic to environment, the diterpene labdane-8α,15-diol (1) and its acetyl derivative (2) were isolated for the first time from Croton ciliatoglanduliferus Ort. They inhibited photophosphorylation, electron transport (basal, phosphorylating and uncoupled) and the partial reactions of both photosystems in spinach thylakoids. Compound 1 inhibits the photosystem II (PS II) partial reaction from water to Na+ Silicomolibdate (SiMo) and has no effect on partial reaction from diphenylcarbazide (DPC) to 2,6-dichlorophenol indophenol (DCPIP), therefore 1 inhibits at the water splitting enzyme and also inhibits PS I partial reaction from reduced phenylmetasulfate (PMS) to methylviologen (MV). Thus, it also inhibits in the span of P700 to Iron sulfur center X (FX). Compound 2 inhibits both, the PS II partial reactions from water to SiMo and from DPC to DCPIP; besides this, it inhibits the photosystem I (PS I) partial reaction from reduced PMS to MV. With these results, we concluded that the targets of the natural product 2 are located at the water splitting enzyme, and at P680 in PS II and at the span of P700 to FX in PS I. The results of compounds 1 and 2 on PS II were corroborated by chlorophyll a fluorescence.  相似文献

We compared the sensitivity to cold stress, in terms of photosynthetic capacity and changes in chlorophyll fluorescence of photosystem 2 (PS2), of an evergreen and a deciduous oak species, which co-occur in the southeastern United States. We predicted that the evergreen species, Quercus virginiana, which must endure winter, is likely to have an inherently greater capacity for energy dissipation and to be less susceptible to chilling stress than the deciduous species, Quercus michauxii. Short-term cold stress in both species lead to greater than 50 % reduction in maximum photosynthetic rates, 60-70 % reduction in electron transport, and irreversible quenching of PS2 fluorescence. The kinetics of recovery in the dark after exposure to 1 h high irradiance (1000 μmol m-2 s-1) and chilling (5 °C) showed that the evergreen Q. virginiana exhibited more protective qE and less irreversible quenching (qI) than the deciduous Q. michauxii. The large qE which we observed in Q. virginiana suggests that the capacity for photoprotection at low temperatures is not induced by a long-term acclimation to cold but preexists in evergreen leaves. This capacity may contribute to the ability of this species to maintain leaves during the winter. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

The potential of barley (Hordeum vulgare L.) and tomato (Lycopersicon esculentum Mill.) roots for net NO 3 - absorption increased two-to five fold within 2 d of being deprived of NO 3 - supply. Nitrogen-starved barley roots continued to maintain a high potential for NO 3 - absorption, whereas NO 3 - absorption by tomato roots declined below control levels after 10 d of N starvation. When placed in a 0.2 mM NO 3 - solution, roots of both species transported more NO 3 - and total solutes to the xylem after 2 d of N starvation than did N-sufficient controls. However, replenishment of root NO 3 - stores took precedence over NO 3 - transport to the xylem. Consequently, as N stress became more severe, transport of NO 3 - and total solutes to the xylem declined, relative to controls. Nitrogen stress caused an increase in hydraulic conductance (L p) and exudate volume (J v) in barley but decrased these parameters in tomato. Nitrogen stress had no significant effect upon abscisic acid (ABA) levels in roots of barley or flacca (a low-ABA mutant) tomato, but prevented an agerelated decline in ABA in wild-type tomato roots. Applied ABA had the same effect upon barley and upon the wild type and flacca tomatoes: L p and J v were increased, but NO 3 - absorption and NO 3 - flux to the xylem were either unaffected or sometimes inhibited. We conclude that ABA is not directly involved in the normal changes in NO 3 - absorption and transport that occur with N stress in barley and tomato, because (1) the root ABA level was either unaffected by N stress (barley and flacca tomato) or changed, after the greatest changes in NO 3 - absorption and transport and L p had been observed (wild-type tomato); (2) changes in NO 3 - absorption/transport characteristics either did not respond to applied ABA, or, if they did, they changed in the direction opposite to that predicted from changes in root ABA with N stress; and (3) the flacca tomato (which produces very little ABA in response to N stress) responded to N stress with very similar changes in NO 3 - transport to those observed in the wild type.Abbreviation and symbols ABA abscisic acid - Jv exudate volume - Lp root hydraulic conductance  相似文献

Barley (Hordeum vulgare L.) and tomato Lycopersicon esculentum Mill.) were grown hydroponically and examined 2, 5, and 10 d after being deprived of nitrogen (N) supply. Leaf elongation rate declined in both species in response to N stress before there was any reduction in rate of dryweight accumulation. Changes in water transport to the shoot could not explain reduced leaf elongation in tomato because leaf water content and water potential were unaffected by N stress at the time leaf elongation began to decline. Tomato maintained its shoot water status in N-stressed plants, despite reduced water absorption per gram root, because the decline in root hydraulic conductance with N stress was matched by a decline in stomatal conductance. In barley the decline in leaf elongation coincided with a small (8%) decline in water content per unit area of young leaves; this decline occurred because root hydraulic conductance was reduced more strongly by N stress than was stomatal conductance. Nitrogen stress caused a rapid decline in tissue NO 3 - pools and in NO 3 - flux to the xylem, particularly in tomato which had smaller tissue NO 3 - reserves. Even in barley, tissue NO 3 - reserves were too small and were mobilized too slowly (60% in 2 d) to support maximal growth for more than a few hours. Organic N mobilized from old leaves provided an additional N source to support continued growth of N-stressed plants. Abscisic acid (ABA) levels increased in leaves of both species within 2 d in response to N stress. Addition of ABA to roots caused an increase in volume of xylem exudate but had no effect upon NO 3 - flux to the xylem. After leaf-elongation rate had been reduced by N stress, photosynthesis declined in both barley and tomato. This decline was associated with increased leaf ABA content, reduced stomatal conductance and a decrease in organic N content. We suggest that N stress reduces growth by several mechanisms operating on different time scales: (1) increased leaf ABA content causing reduced cell-wall extensibility and leaf elongation and (2) a more gradual decline in photosynthesis caused by ABA-induced stomatal closure and by a decrease in leaf organic N.Abbreviation and symbols ABA abscisic acid - ci leaf internal CO2 concentration - Lp root hydraulic conductance  相似文献

PS II-H is a small hydrophobic protein that is universally present in the PS II core complex of cyanobacteria and plants. The role of PS II-H was studied by directed mutagenesis and biochemical analysis in the thermophilic cyanobacterium Thermosynechococcus elongatus BP-1. The psbH disruptant could grow photoautotrophically; however, its growth was much slower than that of the wild type cell. Chromatography enabled the isolation of active oxygen-evolving PS II complexes from both the mutant and the wild type. The mutant yielded a relatively large amount of inactive PS II complex that lacked the following extrinsic proteins: the 33-kDa protein, the 12-kDa protein, and cytochrome c 550 . There were differences between the psbH disruptant and the wild type in terms of the oxygen evolution activities of the cells, thylakoids, and PS II complexes. At high concentrations of 2,6-DCBQ, the activity was much lower in the mutant than in the wild type. Gel filtration chromatography of the PS II complexes showed that both active and inactive PS II complexes isolated from the mutant were mostly in the monomeric form, while the active PS II complex from the wild type was in the dimeric form. The polypeptide composition of both active and inactive PS II complexes from the mutant showed the absence of another small polypeptide, PS II-X. These results suggest that the PS II-H protein is essential for stable assembly of native dimeric PS II complex containing PS II-X.  相似文献

Astaxanthin production is commonly induced under stress conditions such as nutrient deficiency (N or P), high light stress, and variations of temperature, high NaCl concentrations, and other factors. The objective of the present study is the analysis of the effect of oxidative stress by sodium orthovanadate (SOV), a nonspecific inhibitor of protein tyrosine phosphatases, on the cells growth and astaxanthin production of H. lacustris. In the presence of SOV (lower than 5.0 mM), maximum growth of H. lacustris obtained was 2.4 × 105 cells/mL in MBBM medium at 24°C under continuous illumination (40 μE/m2/s) of white fluorescent light, with continuous aeration of CO2 (0.2 vvm). Total carotenoids accumulated per cell biomass unit treated with 2.5 mM SOV has approximately shown 2.5 folds higher than the control after short period of SOV induction time as 2 days, despite that cells were grown under normal light. Meanwhile, maximal astaxanthin production from H. lacustris was 10.7 mg/g biomass in MBBM with 5 days of continuous illumination at 40 μE/m2/s, which has been established as optimal light intensity for the control culture of H. lacustris. Treating algae H. lacustris with sodium orthovanadate showed promoting the accumulation of astaxanthin by advancing either the inhibition of dephosphorylation or synthesis of ATP. Its potential role of PTPases in microalgae H. lacustris is discussed. The first two authors are equally contributed to this work.  相似文献

The biological activity of Bacillus thuringiensis (Bt) strain PS86Q3 against five Hymenopteran species was determined by means of bioassays adapted to each species. Four species of sawfly that are important pests of conifers (Diprion pini, Gilpinia hercyniae and Pristiphora abietina) or ornamental plants (Arge rosae), as well as the non-target honeybee, Apis mellifera, were studied. Two out of the four sawfly species tested were found to be sensitive to PS86Q3 crystals or spore/crystal suspensions. A sporulated culture of this strain was moderately active on D. pini, and a complete bioassay with solubilized crystals was performed to estimate the LC50 of 4.9 mg/ml. Pristiphora abietina was also found to be sensitive to PS86Q3, with an LC50 of 1.6 mg/ml. By contrast, at the concentrations tested, PS86Q3 did not prove active on the remaining sawflies, G. hercyniae and A. rosae. The strain was administered orally to check its effects on honeybees which were fed sucrose solutions supplemented with a PS86Q3 sporulated suspension, in a field assay using commercial beehives. No significant differences in larval mortality (as deduced by comparing the number of larvae, pupae and empty cells) were found between the Bt and control treatments. On the basis of the results presented here, the suitability of PS86Q3 for the control of Hymenopteran pests, particularly sawflies, in terms of both potency and environmental safety, is discussed.  相似文献
