Summary Auxin-induced cell elongation necessitates plasma membrane enlargement. The effect of auxin (10 M 2,4-dichlorophenoxyacetic acid) treatment on amount, composition, and rate of synthesis of plasma membrane lipids was examined. Auxin-treated and control soybean (Glycine max L.) hypocotyl segments were incubated with [14C]acetate for times ranging from 0.5 to 18 h, prior to isolation of plasma membrane by aqueous two-phase partitioning. The composition of individual plasma membrane lipids in elongating segments did not differ from the composition in treatment time-matched control segments, except that after longer auxin treatments, phospholipids had more unsaturated fatty acids. Plasma membrane phospholipid and free sterol content both increased in elongating segments. The relative proportion of sterols and phospholipids in the plasma membrane primarily depended on time after segment excision, for both auxin-treated and control segments. Soybean seedlings treated with ethylene exhibited small increases in ribonucleic acid content in the elongating section of the hypocotyl. Chromatin isolated from the elongating section of ethylene-treated seedlings showed a 35 to 60% increase in the capacity for RNA synthesis. The ethylene-induced response was saturated at 1 microliter/liter of ethylene and was fully expressed after 3 hours. Auxin caused marked accumulation of RNA and DNA in the elongating and basal tissue of the hypocotyl. Chromatin isolated from these auxin-treated tissues showed an 8- to 10- fold increase in RNA synthetic capacity as measured in vitro. Ethylene added with auxin reduced the auxin enhancement of nucleic acid synthesis in the elongating and basal tissues. Both ethylene and auxin treatment of the seedlings inhibited nucleic acid accumulation and chromatin activity in the apical tissue. Ethylene did not appear to mediate the auxin effects on nucleic acid synthesis in soybean hypocotyl with the possible exception of inhibition in the apical tissue.  相似文献

Carrier-mediated uptake of indole-3-acetic acid (IAA) by microsomal vesicles from Cucurbita pepo L. hypocotyls was strongly inhibited by 2,4-dichlorophenoxyacetic acid (2,4-D; i 50= 0.3 M) but only weakly by 1-naphthylacetic acid (NAA). The fully ionised auxin indol-3-yl methanesulphonic acid also inhibited (i 50=3 M). The same affinity ranking of these auxins for the uptake carrier, an electroimpelled auxin anion-H+ symport, is demonstrable in hypocotyl segments. The specificity of the auxin-anion eflux carrier was tested by the ability of different nonradioactive auxins to compete with [3H]IAA and reduce the stimulation of net radioactive uptake by N-1-naphthylphthalamic acid (NPA), a noncompetitive inhibitor of this carrier. By this criterion, NAA and IAA had comparable affinities, with 2,4-D interaction more weakly. Stimulation of [3H]IAA uptake by NAA, as a result of competition for the efflux carrier, could also be demonstrated when a suitable concentration of 2,4-D was used selectively to inhibit the uptake carrier. However, when [3H]NAA was used, no stimulation of its association with vesicles by NPA, 2,3,5-triiodobenzoic acid, or nonradioactive NAA was found. In hypocotyl segments, [3H]NAA net uptake was much less sensitive to NPA stimulation than was [14C]IAA uptake. The apparent contradictions concerning NAA could be explained by carrier-mediated auxin efflux making a smaller relative contribution to the overall transport of NAA than of IAA. Summary The types and amount of plasma membrane proteins synthesized during cell elongation in response to auxin (2,4-dichlorophenoxyacetic acid) treatment were investigated. Auxin-treated and control soybean (Glycine max L.) hypocotyl segments were incubated with [35S]methionine for various times, ranging from 0.5 to 18 h, prior to isolation of plasma membrane by aqueous two-phase partitioning. Protein accumulated in the plasma membrane after auxin treatment. Despite this accumulation, the protein incorporation rate, estimated by the amount of label in the plasma membrane following a 0.5 h [35S]methionine pulse, was unaffected by auxin treatment at both 0.5 and 18 h of treatment. Protein apparently accumulated by a mechanism distinct from enhanced incorporation. The plasma membrane proteins synthesized by elongating segments differed from controls at 18 h, as evidenced by the pattern of fluorographs following a 0.5 h radiolabelling. Antisera were raised to a 70-kD (kilodalton) soybean (Glycine max) protein encoded by a 2,4-dichlorophenoxyacetic acid (2,4-D) inducible mRNA, GH3. These antisera have been used to probe protein blots to study the kinetics and specificity of the GH3 induction response as well as the species specificity and intracellular location of the protein. Detectable levels of the GH3 protein are induced by 2,4-D within 2 h in elongating hypocotyl sections, root sections, and etiolated plumules, and within 30–60 min in soybean suspension cells. Synthesis of the GH3 protein is induced by a variety of auxins. Other plant hormones such as gibberellic acid, cytokinin and ethylene added with or whithout 2,4-D do not alter the level of GH3 protein induction. The GH3 protein is found only in the S100 fraction and is not associated with the nucleus or cell wall. This antiserum also reacts with a 2,4-D-inducible 70-kD protein in other dicots.  相似文献

Quiescent 2,4D-dependent pear fruit cells (Pyrus communis L.) were cultured in a bioreactor in the absence of 2,4-Dichlorophenoxyacetic acid. Effects on the ability to divide following transfer into different concentrations of auxin (0; 0.45; 2.3; 4.5; 6.8; 9 M) are studied and discussed. Changes in cell number and in cell viability during subculture were examined (at all 2,4-D concentrations). In the presence of 2.3, 4.5 and 6.8 M of 2,4-D, in the standard medium, pear cells undergo a classical growth cycle and cell mortality increased only after 16 days. The level of ploidy estimated by cytophotometry shows, after previous starvation, a significant shift only for 4.5 M 2,4-D concentration, the value of the usual culture medium of the studied strain.Abbreviations 2,4-D 2,4-Dichlorophenoxyacetic acid  相似文献

In vitro translation products of polyadenylated RNA from untreated and auxin-treated basal sections of soybean (Glycine max var. Wayne) hypocotyl were analyzed by two-dimensional polyacrylamide gel electrophoresis. Within one hour of 2,4-dichlorophenoxyacetic acid treatment, the translatable messenger RNAs for at least twelve in vitro translation products are modulated upward. In vitro translation products of polyadenylated RNA from untreated, auxin-treated and Ethephon-treated intact soybean hypocotyl were also analyzed. Within two hours of treatment with either 2,4-dichlorophenoxyacetic acid or Ethephon, the translatable messenger RNAs for a group of high molecular weight in vitro translation products are modulated upward. There is a particular set of translatable messenger RNA, encoding in vitro translation products in the 24,000 to 32,000 molecular weight range, that is specifically modulated upward by auxin treatment in intact soybean hypocotyl and in hypocotyl sections.  相似文献

Summary Twelve cultivars of Brassica juncea grown in different agroclimatic regions of the world were tested for their ability to regenerate in vitro from hypocotyl explants and, accordingly, were divided into three groups. One group of cultivars regenerated on MS medium supplemented with 2,4-D, BAP and with NAA, BAP combinations; another group regenerated only on MS with 2,4-D, BAP; and the third group showed very low regeneration on both of these combinations. Inclusion of silver nitrate in the medium was essential for high frequency of regeneration. In general, Indian cultivars were more responsive than the cultivars of CIS and Australian origin. Using the media optimal for regeneration and an Agrobacterium-based binary vector carrying hpt and gus-intron genes, conditions for genetic transformation of B. juncea hypocotyl explants were optimized. Transformation frequencies, identified by GUS staining at the initial stages of growth, were lower on MS medium with 2,4-D, BAP than on MS with NAA, BAP. Plants resistant to 20 g/ml hygromycin were regenerated at a frequency of 11–36% from hypocotyl explants and were shown to be transformed by Southern blotting, GUS staining and progeny analysis.  相似文献

Studies on chromatin and solubilized RNA polymerase from control and 2,4-D treated whole hypocotyls indicate that the activity of RNA polymerase I is enhanced by the auxin treatment while the activity of polymerase II is essentially the same in control and treated tissue. However, different portions of the hypcotyl respond differently to the auxin treatment. The enhancement of solubilized polymerase I activity is greater in the lower half of the hypocotyl than it is in the upper half. In the hook region an inhibition of RNA solubilized polymerase activity is observed. Similarly, 2,4-D treatment results in an inhibition of chromatin bound RNA polymerase activity in nuclei isolated from first leaves. Chromatography on DEAE-Sephadex also reveals a selective enhancement of polymerase I in the upper and lower halves of the hypocotyl.  相似文献

When 2,4-dichlorophenoxyacetic acid (2,4-D)-dependent tobacco cell suspensions, one normal and one transformed by Agrobacterium tumefaciens, were subcultured on hormone-lacking medium the stationary phase of the cell cycle was reached earlier than on medium containing 2,4-D. Addition of the auxin 2,4-D could restore cell division activity within 10–12 h for the most rapidly reacting cell line. The cell-division response was characterized as being auxin-specific and optimal with 2,4-D at 2.2 10-6 M. Although the cell lines used showed different characteristics, both reacted with a rapid increase in at least three mRNA species within 1 or 2 h after 2,4-D application. Two, 2,4-D-induced protein spots, seen after in-vitro translation, had the same characteristics (MWs 35 kilodaltons (kDa) and 25 kDa with isoelectric points of 7.1 and 6.3, respectively) in both cell lines. Water-treated controls did not show alterations in the translatable mRNA populations. This indicates that the accumulation of the corresponding mRNAs is an early hormone-induced event. Five constructions containing deletions of the promoter from an auxin-inducible gene of Arabidopsis thaliana, AtAux2-11, were fused to the coding region of the reporter gene LacZ, which encodes -galactosidase, and a polyadenylation 3-untranslated nopaline synthase sequence from Agrobacterium. These chimeric genes were introduced into Arabidopsis by Agrobacterium tumefaciens-mediated transformation, and expression of the gene was examined by spectrophotometric and histochemical analyses. A 600 bp fragment from the AtAux2-11 promoter conferred histochemical patterns of staining similar to the longest 5 promoter tested, a 3.0 kb fragment. Localization of AtAux2-11/LacZ activity in the transgenic plants revealed spatial and temporal expression patterns that correlated with tissues and cells undergoing physiological processes modulated by auxin. LacZ activity was expressed in the elongating region of roots, etiolated hypocotyls, and anther filaments. Expression was detected in the vascular cylinder of the root and the vascular tissue, epidermis, and cortex of the hypocotyl, and filament. The AtAux2-11/LacZ gene was preferentially expressed in cells on the elongating side of hypocotyls undergoing gravitropic curvature. Expression of the chimeric gene in the hypocotyls of light-grown seedlings was less than that in etiolated seedling hypcotyls. The AtAux2-11/LacZ gene was active in the root cap, and expression in the root stele increased at sites of lateral root initiation. Staining was evident in cell types that develop lignified cell walls, e.g. trichomes, anther endothecial cells, and especially developing xylem. The chimeric gene was not expressed in primary meristems. While the magnitude of expression increased after application of exogenous auxin (2,4-D), the histochemical localization of AtAux2-11/LacZ remained unchanged.Transgenic plants with a 600 bp promoter construct (–0.6 kb AtAux2-11/LacZ) had higher levels of basal and auxin-inducible expression than plants with a 3.0 kb promoter construct. Transgenic plants with a –500 bp promoter had levels of expression similar to the –3.0 kb construct. The –0.6 kb AtAux2-11/LacZ gene responded maximally to a concentration of 5 × 10–6 to 5 × 10–5 M 2,4-D and was responsive to as little as 5 × 10–8 M. A system of synchronous cell division was established by starvation of auxin and its readdition to suspension cultures of cells of Catharanthus roseus L. cv. Little-Pinky. When cells in the stationary phase were transferred to fresh medium free of 2,4-dichlorophenoxyacetic acid (2,4-D), cells were arrested preferentially at the G1 phase. After cells had been cultured for 2 days in medium without 2,4-D, readdition of 2,4-D induced the synchronous division of cells. In this system, 70–80% of cells divided synchronously within 3 to 4h, and the mitotic index increased sharply in parallel with the increase in cell number. Active synthesis of DNA was demonstrated by measurements of incorporation of [3H]-thymidine into the DNA fraction. Summary Using an Agrobacterium tumefaciens binary vector (pAL4404, pBI131), we have demonstrated the transfer of the -glucuronidase gene into the flax (Linum usitatissimum L.) cultivar Glenelg after selection for kanamycin resistance. The transformed lines were obtained by inoculation and subsequent regeneration of hypocotyl segments. The callus that formed on the cut surfaces of the hypocotyl segments was isolated three weeks after infection and was subsequently subcultured to yield shoots. This procedure generated a large number of transgenic shoots over a relatively short period of time. Three pumpkin embryogenic lines were initiated on wounded zygotic embryos cultured on medium with or without 2,4-dichlorophenoxyacetic acid (2,4-D). Somatic embryo development was controlled by the availability of various compounds in the medium: presence/absence of 2,4-D, nitrogen sources. The highest rate of DNA methylation was in the early embryo stages, predominantly on MSC medium with 2,4-D and on auxin-free medium supplemented with 1.0 mM NH4Cl. DNA methylation was correlated with early embryo development in a manner that was not exclusively dependent on the presence/absence of exogenous auxin. DNA methylation decreased during embryo maturation on auxin-free MSC medium and on auxin-free MSC supplemented with 12.3 M 5-azacytidine (5-azaC). The kinetics of induction of heat stability of cytoplasmic proteins and lipoproteins by auxin (2,4,-D) were determined for basal sections of soybean hypocotyl. Maximum heat stabilization occurred after 4 h of tissue incubation with 10-5M 2,4-D. The effect was less pronounced or absent with longer incubations. Membrane fractions sedimenting between 10,000 and 100,000 g and proteins of the 100,000 g supernatant were most affected. The auxin-induced protein aggregation response varied among experiments. With many tissue lots, the response was small or absent even though the tissue responded to the auxin uniformly by increased growth. The magnitude of response was proportional to the logarithm of auxin concentration but with low 2,4-D the portion of the homogenate protein coagulated by heat was increased and with supraoptimal concentrations it was decreased relative to the control. The smallest auxin-induced change in heat coagulability was observed at the auxin concentration nearest the optimum for growth. No direct correlation was found between the auxin-induced protein and lipo-protein aggregation phenomenon and total protein, chloroform-extractable lipid, residual lipid, growth or tissue deformability. Total sulfhydryl equivalent of the homogenates, however, did correlate with auxin effects on aggregation. This result, plus experiments where homogenates were exposed to oxidizing or reducing conditions, suggests that heat stabilization and associated protein aggregation phenomena are related to conversion of protein sulfhydryl to intramolecular disulfide bonds. No significance is attached to heat stabilization of cytoplasmic proteins as a requisite of auxin-induced growth.  相似文献

In order to understand better the relationship between auxin structure and activity on morphogenesis and cell elongation, six different auxins were tested on the regeneration of tomato (Lycopersicon esculentum Miller var. Alice) from cotyledons and on pea (Pisum sativum L. var. Alaska) stem elongation. The auxins were: indole-3-acetic acid (IAA), indole-3-butyric acid (IBA), 1, 2-benzisoxazole-3-acetic acid (BOA), 1,2-benzisothiazole-3-acetic acid (BIA), 1-naphthalenacetic acid (NAA), 2,4-dichlorophenoxyacetic acid (2,4-D). All these compounds obey the minimum requirement rules for auxin activity and all were effective on cell elongation. At the dose of 10 M and in the absence of cytokinin, they all, except 2,4-D, induced roots, while in the presence of cytokinin they induced shoots, roots, hairy root-like filaments (HRLF) or callus depending on their concentration. The morphogenetic pattern did not change by varying cytokinin concentration. We conclude that auxin structure plays a minor role in morphogenesis or cell elongation, because it is only responsible for variations in the level of auxin activity.  相似文献
