The formation of biomimetic lipid membranes has the potential to provide insights into cellular lipid membrane dynamics. The construction of such membranes necessitates not only the utilization of appropriate lipids, but also physiologically relevant substrate/support materials. The substrate materials employed have been shown to have demonstrable effects on the behavior of the overlying lipid membrane, and thus must be studied before use as a model cushion support. To our knowledge, we report the formation and investigation of a novel actin protein-supported lipid membrane. Specifically, inner leaflet lateral mobility of globular actin-supported DMPC (1,2-dimyristoyl-sn-glycero-3-phosphocholine) bilayers, deposited via the Langmuir-Blodgett/Langmuir Schaefer methodology, was investigated by z-scan fluorescence correlation spectroscopy across a temperature range of 20–44°C. The actin substrate was found to decrease the diffusion coefficient when compared to an identical membrane supported on glass. The depression of the diffusion coefficient occurred across all measured temperatures. These results indicated that the actin substrate exerted a direct effect on the fluidity of the lipid membrane and highlighted the fact that the choice of substrate/support is critical in studies of model lipid membranes.  相似文献

In some species, female mate choice is non‐independent as, under certain circumstances, females may copy the mate choice of other nearby females. One standard experimental protocol used to test for mate‐choice copying is the mate‐choice ‘reversal’ protocol. In this protocol, a focal female is allowed to choose between two males as potential mates and then is presented with an opportunity to see another female (i.e. the model female) choose the male that she did not initially choose. The focal is subsequently allowed to again choose between the same set of males. An observed reversal of her initial choice in this second preference test has been previously interpreted as evidence for mate‐choice copying. Alternatively, it has recently been proposed that environmental events, such as seeing the mate choice of nearby females that occur within the visual field of a female actively engaged in mate assessment, may ‘disrupt’ her decision‐making behavior and consequently alter the consistency of her mating preference, and may thus cause mate‐choice reversals. The disruption hypothesis predicts that if a model female is placed near the male that the focal female initially chose, the latter's mate preference would be disrupted and she would subsequently and consistently prefer the male that she initially rejected. Here we examined whether the disruption hypothesis explains mate‐choice copying in the guppy (Poecilia reticulata). Our results do not support this hypothesis, but rather provide further support for mate‐choice copying in the guppy.  相似文献

Many studies demonstrate that partner choice has played an important role in the evolution of human cooperation, but little work has tested its impact on the evolution of human fairness. In experiments involving divisions of money, people become either over-generous or over-selfish when they are in competition to be chosen as cooperative partners. Hence, it is difficult to see how partner choice could result in the evolution of fair, equal divisions. Here, we show that this puzzle can be solved if we consider the outside options on which partner choice operates. We conduct a behavioural experiment, run agent-based simulations and analyse a game-theoretic model to understand how outside options affect partner choice and fairness. All support the conclusion that partner choice leads to fairness only when individuals have equal outside options. We discuss how this condition has been met in our evolutionary history, and the implications of these findings for our understanding of other aspects of fairness less specific than preferences for equal divisions of resources.  相似文献

Purpose

The main goal of this work is to evaluate the environmental impact of a 63-m blade for wind generators. The embodied energy and the carbon footprint are used as supporting tools for material selection in the initial project stages.

Methods

Real industrial data regarding the most used materials for wind turbine blade construction are used. Two eco-parameters, embodied energy and carbon footprint, were calculated from each selected material together with values of manufacture, transport, use, and final disposal. The blades must be built to have a mechanical strength high enough to withstand vibrations caused by manufacturing flaws, turbulence, or irregular loading. In this sense, Young’s modulus, yield strength, and density were compared to the environmental footprint data to support the final material choice. This evaluation process of the possible materials to be used in the blade manufacture was carried out in the initial stages of the project.

Results

Composite materials such as glass fiber-reinforced polymer (GFRP) and carbon fiber-reinforced polymer (CFRP), bonded together with an adhesive material, are used to build modern wind turbine blades. Those composites comprise a considerable number of different materials that can be mixed to reach adequate performance. Comparisons were made with 46 pre-selected materials, considering the mechanical behavior and environmental impacts. The final selected materials have better properties than the reference material. Finally, two materials with the desired mechanical properties and with a potential lower negative environmental impact than the reference material were selected.

Conclusions

Replacing the reference resin—epoxy/E-glass fiber—with the epoxy resin with the lowest environmental impact—epoxy/S-glass fiber—will reduce the total value of the environmental load to 102 GJ of energy and 3.4 t of CO2. As important as the material selection in the early stages of product development is the end of life (EoL) choice. In this case, the glass fiber has an EoL potential of 370 GJ of energy and 460 t of CO2 in the remanufacturing option, against zero for the landfill. This work shows that carefully selected raw materials and EoL alternatives for WTB can significantly reduce the environmental impact of this component.

Abstract Restoration practitioners have long been faced with a dichotomous choice of native versus introduced plant material confounded by a general lack of consensus concerning what constitutes being native. The “restoration gene pool” concept assigns plant materials to one of four restoration gene pools (primary to quaternary) in order of declining genetic correspondence to the target population. Adaptation is decoupled from genetic identity because they often do not correspond, particularly if ecosystem function of the disturbed site has been altered. Because use of plant material with highest genetic identity, that is, the primary restoration gene pool, may not be ultimately successful, material of higher order pools may be substituted. This decision can be made individually for each plant species in the restored plant community in the scientific context that ecosystem management demands. The restoration gene pool concept provides a place for cultivars of native species and noninvasive introduced plant material when use of native‐site material is not feasible. The use of metapopulation polycrosses or composites and multiple‐origin polycrosses or composites is encouraged as appropriate. The restoration gene pool concept can be implemented as a hierarchical decision‐support tool within the larger context of planning seedings.  相似文献

Viruses are used to test the barrier properties of materials. Binding of virus particles during passage through holes in the material may yield misleading test results. The choices of challenge virus and suspending medium may be important for minimizing confounding effects that might arise from such binding. In this study, different surrogate viruses, as well as different support media, were evaluated to determine optimal test parameters. Two membranes with high-binding properties (nitrocellulose and cationic polysulfone) were used as filters to compare binding activities of different surrogate challenge viruses (MS2, phi X174, T7, PRD1, and phi 6) in different media. The media consisted of buffered saline with surfactants, serum, or culture broth as additives. In addition, elution rates of viruses that bound to the membranes were determined. The results suggest that viruses can bind by hydrophobic and electrostatic interactions, with phi X174 displaying the lowest level of binding by either process. The nonionic detergents Triton X-100 and Tween 80 (0.1%) equally minimized hydrophobic interactions. Neither anionic nor cationic surfactants were as effective at nontoxic levels. Serum was effective at reducing both hydrophobic and electrostatic binding, with 2% being sufficient for eliminating binding under our test conditions. Thus, phi X174 remains the best choice as a surrogate virus to test barrier materials, and Triton X-100 (0.1%) remains a good choice for reducing hydrophobic binding. In addition, binding of viruses by barrier materials is unlikely to prevent passage of blood-borne pathogens.  相似文献

Several studies have shown that mate choice based on condition leads to higher reproductive success of the choosing individual. Yet, a growing body of literature has failed to find support for mate choice based on mate condition, even when the choosing individual would clearly benefit from such a choice. This indicates that animals’ mate choice is often more complex than currently appreciated and that even well-founded expected preferences cannot be taken for granted. Using the broad-nosed pipefish, Syngnathus typhle, we manipulated male condition experimentally to explore whether it affects female mate choice. In this sex-role-reversed species, males care for the offspring in a specialised brood pouch. Males are the choosier sex, but given the opportunity, females are selective as well. During brooding, males can both provide embryos with nutrients and take up nutrients that originate from eggs deposited in the pouch, and embryo survival correlates positively with male condition. Together, this suggests that it would be beneficial for females to mate with males in high condition. However, we found no female preference for males in better condition. Thus, this study adds to the literature of mate choice that is unaffected by mate condition. Possible reasons for our result are discussed.  相似文献

The ‘Support Problem’ is a benchmark test to investigate the understanding of spatial relationships between objects. We tested kea parrots'' performance in a paradigm that has previously been studied in primates. Kea perform comparably well to tamarins when they are confronted with a choice between two support devices, one of which has a reward resting on it and the other slightly next to it, or when given a choice between a continuous and a disrupted support. Kea did better than chimpanzees in some tasks in which the perceptual connection of the food to the support was altered. The results indicate that kea are capable of assessing the spatial means–end relationships of this problem spontaneously and in a way that is comparable with primates.  相似文献

UV‐absorbing films and nets are frequently used as covering materials for netted greenhouses and film tunnels in protected cultivation systems. This study explored the effects of such materials on the dispersal behaviour of western flower thrips (WFT), Frankliniella occidentalis, in flight cages under greenhouse conditions with additional artificial UV‐A light sources. The study involved release–recapture experiments in choice and no‐choice layouts. Different trapping methods were compared (blue sticky cards, plants and transparent cards) for recapture of thrips. In choice experiments, insects were released from a black box compartment between two tunnels covered with either UV‐transmitting or UV‐absorbing materials. A significantly higher proportion of (82–98%) WFT was recaptured in UV‐transmitting tunnels compared with UV‐absorbing tunnels. In no‐choice experiments, WFT were found to infiltrate the tunnels at different rates depending on the trap type used and experimental layout. In small‐scale dispersal experiments using blue sticky cards and plants as traps, infiltration was not significantly different between UV‐absorbing and UV‐transmitting tunnels, whereas when using transparent cards, WFT penetrated further into the UV‐transmitting plastic film tunnels. In larger‐scale dispersal experiments, plants or blue sticky cards were arranged in concentric circles around a source plant at the release point. Dispersal was found to differ depending on the method of release, but WFT tended to exhibit reduced dispersal from source plants under UV‐deficient conditions. In conclusion, our data support the hypothesis that manipulation of spectral light properties using UV‐absorbing cladding materials for protected crop stands interferes with the orientation and host finding of WFT, resulting in reduced dispersal into and within plant stands in UV‐deficient environments.  相似文献

A hybrid waveguide, which consists of a dielectric wire above a dielectric-metal interface, has been previously proposed to achieve high confinement with low loss. By exciting this geometry with an aperture in the metal that takes advantage of the extraordinary transmission through subwavelength apertures, it is possible to strongly couple to multiple modes. The real part of the fundamental mode is in fact capable of exceeding the index of refraction of all the materials used while maintaining a manageable imaginary part, as a result of appropriate choice of materials for the dielectric wire and the metal. In addition, as the confinement of the second mode is comparable to that of the fundamental mode but has a much longer propagation length, this mode can be utilized in light-guiding applications where enhanced confinement and propagation is desired.  相似文献

The evolution of mate choice for genetic benefits has become the tale of two hypotheses: Fisher's 'run-away' and 'good genes', or viability indicators. These hypotheses are often pitted against each other as alternatives, with evidence that attractive males sire more viable offspring interpreted as support for good genes and with a negative or null relationship between mating success of sons and other components of fitness interpreted as favouring the Fisher process. Here, we build a general model of female choice for indirect benefits that captures the essence of both the 'Fisherian' and 'good-genes' models. All versions of our model point to a single process that favours female preference for males siring offspring of high reproductive value. Enhanced mating success and survival are therefore equally valid genetic benefits of mate choice, but their relative importance varies depending on female choice costs. The relationship between male attractiveness and survival may be positive or negative, depending on life-history trade-offs and mating skew. This relationship can change sign in response to increased costliness of choice or environmental change. Any form of female preference is subject to self-reinforcing evolution, and any relationship (or lack thereof) between male display and offspring survival is inevitably an indicator of offspring reproductive values. Costly female choice can be maintained with or without higher offspring survival.  相似文献

The mechanisms and temporal aspects of mate choice according to genetic constitution are still puzzling. Recent studies indicate that fitness is positively related to diversity in immune genes (MHC). Both sexes should therefore choose mates of high genetic quality and/or compatibility. However, studies addressing the role of MHC diversity in pre- and post-copulatory mate choice decisions in wild-living animals are few. We investigated the impact of MHC constitution and of neutral microsatellite variability on pre- and post-copulatory mate choice in both sexes in a wild population of a promiscuous primate, the grey mouse lemur (Microcebus murinus). There was no support for pre-copulatory male or female mate choice, but our data indicate post-copulatory mate choice that is associated with genetic constitution. Fathers had a higher number of MHC supertypes different from those of the mother than randomly assigned males. Fathers also had a higher amino acid distance to the females' MHC as well as a higher total number of MHC supertypes and a higher degree of microsatellite heterozygosity than randomly assigned males. Female cryptic choice may be the underlying mechanism that operates towards an optimization of the genetic constitution of offspring. This is the first study that provides support for the importance of the MHC constitution in post-copulatory mate choice in non-human primates.  相似文献

Enzymatic catalysis has been pursued extensively in a wide range of important chemical processes for their unparalleled selectivity and mild reaction conditions. However, enzymes are usually costly and easy to inactivate in their free forms. Immobilization is the key to optimizing the in-service performance of an enzyme in industrial processes, particularly in the field of non-aqueous phase catalysis. Since the immobilization process for enzymes will inevitably result in some loss of activity, improving the activity retention of the immobilized enzyme is critical. To some extent, the performance of an immobilized enzyme is mainly governed by the supports used for immobilization, thus it is important to fully understand the properties of supporting materials and immobilization processes. In recent years, there has been growing concern in using polymeric materials as supports for their good mechanical and easily adjustable properties. Furthermore, a great many work has been done in order to improve the activity retention and stabilities of immobilized enzymes. Some introduce a spacer arm onto the support surface to improve the enzyme mobility. The support surface is also modified towards biocompatibility to reduce non-biospecific interactions between the enzyme and support. Besides, natural materials can be used directly as supporting materials owning to their inert and biocompatible properties. This review is focused on recent advances in using polymeric materials as hosts for lipase immobilization by two different methods, surface attachment and encapsulation. Polymeric materials of different forms, such as particles, membranes and nanofibers, are discussed in detail. The prospective applications of immobilized enzymes, especially the enzyme-immobilized membrane bioreactors (EMBR) are also discussed.  相似文献

A myriad of materials have been used for reestablishing continuity of the orbital floor following blunt facial trauma. Traditionally, autogenous grafts have been the material of choice for orbital floor reconstruction; however, alloplastic materials have gained popularity because of their availability and ease of use. A large clinical experience with long-term treatment results has never been reported for any substance used in orbital floor reconstruction. The purpose of this study was to review our long-term treatment results using Teflon for orbital floor reconstruction following blunt trauma, with emphasis on the incidence of infection, extrusion, and implant displacement. This report presents a 20-year review of 230 Teflon implants for reconstruction of traumatic orbital floor defects. With a mean follow-up period of 30 months, there was only one implant infection and no complications of extrusion or implant displacement. These findings support the use of Teflon as a safe and effective material for the reconstruction of orbital floor defects following blunt facial trauma.  相似文献

The problem of development of lateralization of motor systems in the course of early ontogenesis is important for understanding regularities of formation of functional characteristics of motor control. To solve these tasks, in the 8–74-day old rat pups the presence of lateralization was studied in two motor tests: the choice of the preferable rotation direction at holding the rat by the tail and the choice of one of the forelimbs in the test of initiation of the limb placement onto a support. It has been shown that in early ontogenesis, until the age of the sexual maturation, no constant lateralization is observed in realization of the unconditional motor reactions in any of the performed tests. The turning point of the establishment of the lateral preference has been revealed to correspond to the three-week age (the 21–23rd days), while the final lateralization both for individual animals and for the entire sample takes place at the age of 1.5–2 months. It is suggested that development of lateralization of the limb placement on a support occurs with the approximately two-week delay as compared with the process of formation of the preference in the choice of the rotation direction.  相似文献

The eukaryotic pathogen Leishmania donovani possesses a housekeeping protein Elongation-Factor-1alpha (EF-1alpha) which has been found to be unexpectedly involved in the pathogen's virulence. Because it is associated with virulence and essential for cell survival, this protein is an attractive choice for drug targeting; however, its sequence is highly similar (> 80% sequence identity) to that of its human homolog, rendering it a risky choice for a drug target. The chief difference between these two proteins has been found to be a 12 amino acid sequence present in human EF-1alpha but absent from leishmania EF-1alpha. Furthermore, it has been shown that this 12 amino acid insert in the human sequence corresponds to a hairpin loop on the surface of the protein. In this study, we searched for those spatial features in leishmania EF-1alpha that are impacted or obscured by the extra hairpin loop in the human counterpart. We have also conducted a large-scale in silico screening for small molecules that could plausibly bind to these protein features. While experimental evidence is required to verify our results, our findings thus far appear to support this approach as a new strategy for the development of antagonists against pathogenic targets having close human homologs.  相似文献

Can brain activity reveal a covert choice? Making a choice often evokes distinct emotions that accompany decision processes. Amygdala has been implicated in choice behavior that is guided by a prospective negative outcome. However, its specific involvement in emotional versus cognitive processing of choice behavior has been a subject of controversy. In this study, the human amygdala was monitored by functional magnetic resonance imaging (fMRI) while subjects were playing in a naturalistic choice paradigm against the experimenter. In order to win, players had to occasionally choose to bluff their opponent, risk "getting caught," and suffer a loss. A critical period, when choice has been made but outcome was still unknown, activated the amygdala preferentially following the choice that entailed risk of loss. Thus, the response of the amygdala differentiated between subject's covert choice of either playing fair or foul. These results support a role of the amygdala in choice behavior, both in the appraisal of inherent value of choice and the signaling of prospective negative outcomes.  相似文献

AIMS: To investigate the influence of the choice of yeast strain on the haze, shelf life, filterability and foam quality characteristics of fermented products. METHODS AND RESULTS: Twelve strains were used to ferment a chemically defined wort and hopped ale or stout wort. Fermented products were assessed for foam using the Rudin apparatus, and filterability and haze characteristics using the European Brewing Convention methods, to reveal differences in these parameters as a consequence of the choice of yeast strain and growth medium. CONCLUSIONS: Under the conditions used, the choice of strain of Saccharomyces cerevisiae effecting the primary fermentation has an impact on all of the parameters investigated, most notably when the fermentation medium is devoid of macromolecular material. SIGNIFICANCE AND IMPACT OF THE STUDY: The filtration of fermented products has a large cost implication for many brewers and wine makers, and the haze of the resulting filtrate is a key quality criterion. Also of importance to the quality of beer and some wines is the foaming and head retention of these beverages. The foam characteristics, filterability and potential for haze formation in a fermented product have long been known to be dependant on the raw materials used, as well as other production parameters. The choice of Saccharomyces cerevisiae strain used to ferment has itself been shown here to influence these parameters.  相似文献

Health-care research relies largely on human materials stored in highly specialised biorepositories. Medical translational research on tissues can be performed using a variety of resources in distinct situations. The best known is the secondary use of pathology archives where paraffin-embedded tissues are stored for diagnostic reasons. Another is collecting and storing frozen material obtained from leftover surgical diagnosis. Such residual tissues can either be used directly in research projects or used in the context of a clinical trial with new interventional medicinal products. The latter can make the regulations governing the use of these materials for medical research much more complicated. The use of residual materials is very distinct from biobanking projects for which tissue is specifically collected. This article describes the consequences of using residual human material from different sources in distinct situations and why signed informed consent is not always the preferred choice of individual countries regarding the use of residual material. In addition, signed informed consent is overdone when using residual tissues in medical research. We maintain that the opt-out system is a balanced choice if certain requirements are met, relating to sufficient transparency about using residual tissue for research, the purpose of such research and to the confidentiality of the data used in that research. Finally, the international exchange of samples can be based on the laws and regulations of the countries of origin. Respecting these form the basis of what can and cannot be done in the country where the research on the samples is being performed.  相似文献

The ecological forces determining where within a landscape plants recruit and grow has been termed proximal habitat choice. Habitat choice is imposed first by a heterogeneous pattern of seed dispersal across the patches that make up the landscape and second by environmental variation that favors plant survival in some patches more than in others. Seed-seedling conflicts can occur during both of these phases of habitat choice if conditions or traits that are favorable for seeds are unfavorable for seedlings. During the dispersal phase, smaller seeds may have a greater probability of dispersal than larger seeds, and thus a greater probability of escape from predation, but they contain fewer reserves for support of the establishing seedling. After dispersal, environmental characteristics of a given patch type that lead to disproportionately high seed survival may lead to disproportionately low seedling survival. Considering three hypothetical landscapes, each composed of five patch types, I demonstrate that seed-seedling conflicts can have a major impact on both the overall quantity of recruitment at the landscape level and on the distribution of recruitment among patches. Available empirical evidence suggests these conflicts may be widespread in natural systems. Given their potential importance and extent, seed-seedling conflicts may play a previously unrecognized role in habitat choice.  相似文献
