The activity of the substance(s) which are contained in the cephalic endocrine organs of the locust which induce egg diapause in Bombyx mori was examined by implantation and injection of saline extracts of these organs. Extracts from the median and lateral neurosecretory parts of the locust brain were not effective in inducing egg diapause. Extracts of the corpora cardiaca, corpora allata, and suboesophageal ganglion of the locust induced diapause eggs in Bombyx pharate adults from which the suboesophageal ganglion had been removed. The first two extracts could induce egg diapause even in isolated abdomens of pharate adults of Bombyx. In the locust corpora cardiaca, the activity was present only in the glandular lobe and not in the nervous region. This activity decreased when the nervi corporis cardiaci I and II and of nervi corporis allati I were cut. Allatectomy also brought about a decrease in the activity in the glandular lobe which could not be restored by the injection of juvenile hormone. The activity in the corpora allata was enhanced slightly by the disconnection though not significantly.From these results, it is assumed that the corpora cardiaca, corpora allata and suboesophageal ganglion of the locust contain and active principle(s) capable of inducing egg diapause in Bombyx mori. The nervous connections between the brain, corpora cardiaca, and corpora allata are essential for the accumulation of the active substance(s) in the glandular lobes of the corpora cardiaca.  相似文献

Diapause egg production was examined in non-diapause egg producers by implantation of various cephalic organs into pharate adults 4 days after larval-pupal ecdysis. The implantation of five pairs of corpora cardiaca or corpora allata induced a great amount of egg diapause. Implantation of these organs was effective in inducing egg diapause even when the suboesophageal ganglion of the recipients had been removed, although the effect of the corpora allata decreased moderately. The injection of juvenile hormone into 4-day-old pharate adults did not greatly increase production of diapause eggs.  相似文献

The suboesophageal ganglion of the silkworm, Bombyx mori synthesizes sufficient diapause hormone to produce diapause eggs, regardless of the photoperiodic conditions experienced during the larval stages. When larvae destined to produce non-diapause eggs are implanted with the brain-suboesophageal ganglion complex from larvae which have been reared under short-day conditions, the resulting adults lay diapause eggs. The larvae receiving the complex from larvae reared under long-day conditions gave rise to adults which did not produce any diapause eggs. The brains from pupae which have been reared under long-day conditions show an activity inhibiting the secretion of diapause hormone by the suboesophageal ganglion. The mechanism through which the brain controls the secretion of diapause hormone from the suboesophageal ganglion can be modified by photoperiodic conditions during the larval stages.  相似文献

Developing ovaries from pharate adults of the silkworm, Bombyx mori, were incubated in a medium containing 14C-trehalose or 14C-glucose, and the effects of diapause hormone on the incorporation of these isotopes into ovary glycogen were studied. The rates of incorporation of 14C-trehalose remained unchanged in ovaries incubated for 36 hr when the medium was renewed at intervals of 12 hr, and showed saturation kinetics against the concentration of the sugar in the medium, giving apparent Km values of 6·0 mM for trehalose.There was no difference in 14C-glucose incorporation by ovaries grown in the presence (+SG) and absence (?SG) of the suboesophageal ganglion (SG). However, when 14C-trehalose was used as a substrate for glycogen synthesis, there was a marked difference in the incorporation between them, i.e. the incorporation was more than 60 per cent higher in +SG ovaries than that in ?SG ovaries. Increased incorporation of 14C-trehalose was also observed in ovaries from SG-removed pharate adults which received an injection of diapause hormone preparations. Maximum stimulation rates (about twofold) appeared 36 hr after the injection. Further, comparable effects on 14C-trehalose incorporation were observed in ovaries which were incubated with diapause hormone preparations added in vitro.These data are discussed in relation to the hormonal regulation of trehalase activity in ovaries.  相似文献

The haemolymph protein concentration in Bombyx mori decreases normally by about one-fourth during pharate adult development. In females homozygous for the small egg gene, the concentration of haemolymph protein remained constant throughout the pupal and pharate adult stages. The sm gene does not influence the synthesis of vitellogenic female protein of pupal and pharate adult haemolymph (FP). Normal ovaries transferred to the haemocoele of sm females undergo normal vitellogenesis. In the absence of normal alleles of sm, the ovaries encounter difficulties in the incorporation of FP into their oöcytes from pharate adult haemolymph. These results suggest that an active translocation mechanism is involved in the transfer of haemolymph protein into the ovaries.  相似文献

Cephalic nervous connexions were found necessary for oviposition and normal egg maturation in the two species studied. Neurosecretory cells of A type from the brain, suboesophageal, thoracic, and four abdominal ganglia seem to elaborate a stimulating substance for egg laying. This neurosecretion type is not present in corpora cardiaca, corpora allata, the four last abdominal ganglia, or perisympathetic organs which have no clear-cut effect on oviposition. The circadian rhythm of egg laying appears to be entirely controlled by the liberation of this neurosecretion and by an inhibitory factor localized in the head.In Carausius, egg deposition by the valvulae of the ovipositor is regulated by these two factors whereas egg transit through the common oviduct is a distinct preliminary step under nervous control. This nervous effect from the posterior region does not exist in Clitumnus.  相似文献

The developmental expressions of the mRNA of JH synthetic enzymes have been studied using homogenates of the corpora cardiaca-corpora allata (CC-CA) complexes in Bombyx mori [Kinjoh, T., Kaneko, Y., Itoyama, K., Mita, K., Hiruma, K., Shinoda, T., 2007. Control of juvenile hormone biosynthesis in Bombyx mori: cloning of the enzymes in the mevalonate pathway and assessment of their developmental expression in the corpora allata. Insect Biochemistry and Molecular Biology 37, 808-818]. The in situ hybridization analyses in the CC-CA complex showed that the distribution of the mRNAs of all the mevalonate enzymes and juvenile hormone (JH) acid O-methyltransferase occurred only in the CA cells, indicating that the fluctuations of the enzyme mRNA amounts in the CC-CA complexes were derived solely from the CA. In addition, the size of the CA and their nuclei was not associated with the JH synthetic activity by the CA until the pharate adult. Only female adult CA synthesized JH in B. mori, and the CA and the nuclei were significantly larger than those of male CA which do not synthesize JH.  相似文献

In the suboesophageal ganglion of Bombyx mori the diapause-regulator producing cells which may give information to the diapause-factor cells were found by means of electron microscopy.The diapause-regulator producing cells had larger granules (200 to 500 mμ dia.) than did the diapause-factor cells which were partially surrounded by the former. Highly electron-dense material of lysosome in the diapause regulator producing cells was observed in the diapause-egg producer, but such lysosomes were not present in the non-diapause-egg producer. It was found that many cytoplasmic granules fuse with lysosome, and smaller granules arise from lysosomes. Some implications regarding the diapause-factor cell and the diapause regulator producing cell are discussed.  相似文献

Electron micrographs of the larval brain of the silkworm, Bombyx mori, show that the largest median neurosecretory cell of the diapause egg producer may participate in the production of lipo-granules, and that of the non-diapause egg producer in the production of electron-lucent vesicles. It was found that there were ribosome-like particles in the diapause median neurosecretory cell, and they gradually developed into highly dense particles and lined up along the smooth-surfaced endoplasmic reticulum. The frontal ganglion of the tobacco hornworm, Manduca sexta (L.), was found to contain two neurosecretory (NS) cells (max dia = 40–45 μm). The cytoplasmic inclusions of the NS cells were stained purple with paraldehyde fuchsin, and marked fluctuations in amounts of NS material in the perikarya were observed, depending upon the developmental status of the insect. The perikarya of NS cells in the frontal ganglia of starved larvae and diapause pupae contained large accumulations of NS material, whereas feeding larvae and developing pharate adults showed relatively low amounts of neurosecretion. Electron microscopy revealed large accumulations of NS granules (dia = 80–240 nm) in the frontal ganglia of diapause pupae, but only slight accumulations of granules were seen in the NS cells of developing larvae and pharate adults.It was concluded that axonal transport and release but not synthesis is shut down during starvation and diapause, leading to accumulation of NS material in the perikarya. It is also suggested that the failure of many investigators to differentiate NS cells in the frontal ganglion of various insects may have been due to the selection of very active stages when the amount of available NS material was too low to be visualized by conventional staining techniques.  相似文献

The ultimate timing of hatching in the silkworm, Bombyx mori, is controlled by a circadian oscillator. The presence of eclosion hormone in developing embryos of the silkworm is demonstrated. Eclosion hormone activity first becomes detectable in embryos which have developed almost to the stage of the differentiation of the neuroendocrine system. Hormonal activity increases sharply to a maximum level 1 day before hatching and falls by about a half in the newly hatched larvae. Eclosion hormone was partially purified from the pharate first-instar larvae and approx, a 2100-fold purification was achieved. The molecular weight of the embryo eclosion hormone is estimated to be 7000 ~ 9000 Daltons by gel-filtration on Sephadex G-50 (superfine). The role of eclosion hormone on hatching behaviour of the silkworm, Bombyx mori, is discussed.  相似文献

Leucania separata larvae show various degrees of darkening depending on the population density. A ligature applied behind the thorax of crowded or yellow solitary larvae caused black or reddish brown pigmentation in the anterior part after the larval ecdysis. Extirpation of the brain, the corpus cardiacumcorpus allatum complexes, or the suboesophageal ganglion reduced the degree of melanization in the crowded larvae, lack of the suboesophageal ganglion having a particularly striking effect. Transplantation of 3 complexes of brain-corpora cardiaca-corpora allata-suboesophageal ganglion induced intense black pigmentation in the isolated abdomens of crowded larvae and reddish brown pigmentation with some melanization in the isolated abdomens of yellow solitary larvae, though the melanization in the latter was weaker than in the former. Implantation of these organs or of the suboesophageal ganglia into yellow solitary larvae caused black and reddish brown pigmentation after a larval ecdysis. In the pieces of integument implanted into the body cavity of crowded larvae, melanization occurred after ecdysis, whereas it did not occur in most of the fragments implanted in yellow solitary larvae. Transplantation of corpora allata and other organs from solitary larvae or injection of juvenile hormone into crowded larvae did not inhibit melanization.  相似文献

Diapausing Bombyx mori eggs were treated with 20-hydroxyecdysone as follows: a 30 minute immersion in bleaching water was followed by a 24-hour soaking in a 10?4 M solution of the hormone. Of the treated embryos 85% developed to some extent and 25% of them terminated their embryonic development. The efficiency of the treatment rapidly decreased when eggs were allowed to hibernate at 7°C in order to break diapause.  相似文献

Incorporation of palmitic acid-1-14C into pharate adult tissues and their lipid components of Bombyx mori was investigated. Rapid incorporation of radioactivity took place predominantly in fat body and haemolymph lipids, and partially in ovarian lipids immediately after the injection at the middle stage of pharate adult development. The major parts of the radioactivities in fat body, haemolymph and ovary were distributed in triglycerides and phospholipids, diglycerides, and triglycerides, respectively. The patterns of time course of incorporation of radioactivity into lipid components of pharate adult tissues suggest that the major form of lipid released from fat body may be diglycerides and the diglycerides in haemolymph are probably the main source of ovarian triglycerides.  相似文献

Members of the neuropeptide family having Phe-X-Pro-Arg-Leu-NH(2) (FXPRLamide; X=Ser, Thr, Val, or Gly) at the C-terminus serve as regulators of oviduct and visceral muscle contraction, sex pheromone production, and diapause induction. Antibody raised against Bombyx mori diapause hormone recognized a variety of FXPRLamide peptides. Using this antibody, the antigen was immunocytochemically localized in the central nervous system (CNS) of the silkworm, Bombyx mori. Immunoreactive somata were observed in all ganglia of the CNS including the brain. Twelve somata localized at the midline of the suboesophageal ganglion (SG) were most intensely stained, and their neurite projections reached the retrocerebral complex. Thus, these cells in the SG exhibited typical features of neuroendocrine neurons. Marked reduction in immunoreactivity was observed in a pair of neurosecretory cells in the labial neuromere in SG of diapause type pupae, which indicates an active release of FXPRLamide peptides from these cells. No clear connection to neurohemal sites were observed in immunoreactive cells in the brain, thoracic or abdominal ganglia, suggesting that the immunoreactive peptides in these organs are likely to serve as neurotransmitters or neuromodulators.  相似文献

Ouabain, an inhibitor of the active transport of some inorganic cations, functioned to produce non-diapause eggs in diapause egg producers. The effectiveness of this chemical related to non-diapause egg production was evident in 40 nmole injection per 4-day-old pharate adult. Simultaneous injections of ouabain with diapause hormone preparation or K+-ion suppressed non-diapause egg production to as a great extent as though they functioned competitively. Other experiments indicate that ouabain does not affect the ovaries directly but appears to inhibit the release of endogenous diapause hormone from the suboesophageal ganglion by affecting the central nervous system. The shortage of 3-hydroxykynurenine in non-diapause eggs produced by ouabain is thought not to be the result of the blockage of active transport at the ovaries but the result of a lack of diapause hormone.  相似文献

The embryonic diapause of the silkworn,Bombyx mori, is induced by the diapause hormone (DH) which is secreted from the suboesophageal ganglion of pupae. The diapause nature of bivoltine strains uses environmental stimuli as the initial signal to determine the diapause nature. The experiments showed that DH gene expreon is a direct response to the environmend stimulus, such as high incubation temperature. The cDNA from the embryonic stage wa cloned and sequenm analysis showed the cDNA encoding DH. Expmion patterns of the DH gene in embryonic stage are different ar incubation temperatures 15°C and 25°C, suggesting that the incubation tempcreturt as an environmental signal is kept within the body to control the DH gene expmion at the pupal stage, so that the embryonic diapause of next generation can be determined.  相似文献

In vitro cultures of prothoracic glands of larvae of Periplaneta americana and of some Lepidoptera as biological tests are described. Incorporation of 3H-5-uridine in the RNA of the prothoracic glands represented the measure of the cellular activity of the glands.Activation factor I separated from extracts of corpora cardiaca of the cockroach Periplaneta americana by means of gel filtration techniques caused significant stimulation of RNA synthesis of the glands.  相似文献

The activity of ribosomal RNA (rRNA) genes as judged by nucleolar size and rRNA synthesis has been shown to depend upon the phase of diapause in the eggs of Bombyx mori. In the present study, we found that nucleolar size in diapausing eggs was enlarged at a very early stage during cold treatment, a procedure necessary for the termination of diapause. In contrast, the intrinsic capacity of ribosomal RNA synthesis in the chilled eggs, as examined at 25°C by radioactive precursor incorporation into rRNA, increased much later, in parallel with the break of diapause. The early phase of cold treatment is the period when the eggs undergo some important changes (the so-called diapause development), preparing for diapause termination. Thus we infer that the above mentioned increase in nucleolar size may be one of the features of diapause development.  相似文献
