Selection response of a modified recurrent full-sib (FS) selection scheme conducted in two European flint F2 maize (Zea mays L.) populations was re-evaluated. Our objectives were to (1) determine the selection response for per se and testcross performance in both populations and (2) separate genetic effects due to selection from those due to random genetic drift. Modified recurrent FS selection was conducted at three locations using an effective population size N e = 32 and a selection rate of 25% for a selection index, based on grain yield and grain moisture. Recombination was performed according to a pseudo-factorial mating scheme. Selection response was assessed using a population diallel including the source population and advanced selection cycles, as well as testcrosses with unrelatesd inbred line testers and the parental F1 generation. Selection response per cycle was significant for grain yield and grain moisture in both populations. Effects of random genetic drift caused only a small reduction in the selection response. No significant selection response was observed for testcrosses, suggesting that for heterotic traits, such as grain yield, a high frequency of favorable alleles in the elite tester masked the effects of genes segregating in the populations. We conclude that our modified recurrent FS selection is an alternative to other commonly applied intrapopulation recurrent selection schemes, and some of its features may also be useful for increasing the efficiency of interpopulation recurrent selection programs.  相似文献

Summary Vegetative segregation of a mixed plastid population in protoplast fusion-derived cell lines can be directed by a selection favouring the multiplication of one of the parental plastid types. This report defines some of the critical conditions leading to a homogeneous plastid population in cybrid plants generated by protoplast fusion between Nicotiana plumbaginifolia and an albino and streptomycin-resistant N. tabacum plastid mutant. Light (1,500 lx) conferred a strong selective advantage to chloroplasts versus albino plastids, while the lack of this effect in dim light (300 lx) indicated that a sufficient light intensity is essential to the phenomenon. Selection on streptomycin-containing medium in the dark, however, led to the preferential multiplication of resistant plastids. Streptomycin selection of resistant chloroplasts in the light, consequently, results in a plastid selection of doubled stringency. In another experiment a definite, but leaky, selection for chloroplast recombination (selection for greening on streptomycin-containing medium in dim light) was used to reveal various recombination products. Protoplast fusion in fact resulted in cybrid plants showing only simple chimeric segregation of unchanged parental plastids. These results demonstrate the essential requirement for stringent plastid selection, as defined by cell culture conditions, to precede the formation of shoots expected to possess the desired plastid genetic composition.  相似文献

Dispersal ability is an important fitness component in most plant species. Therefore, some phenotypic traits can be selected due to their effect on dispersal. In this study I determine the potential for dispersal-mediated selection on plant height in an autochorous plant, Erysimum mediohispanicum (Brassicaceae). Selection was quantified by selection gradients, structural equation modeling and generalized additive models. I detected significant dispersal-mediated linear selection gradient on plant height, taller plants dispersing seeds farther. Nevertheless, the generalized additive models suggest that the selection on stalk height was non linear. Indeed, it detected a threshold in the effect of stalk height on dispersal ability; plants shorter than that threshold had an extremely short dispersal, whereas plants taller than that threshold dispersed the seeds very far. Furthermore, the structural equation modeling showed that stalk height indirectly affected dispersal distance through its significant effect on one reproduction-related fitness component, taller plants having greater fecundity. Selection on E. mediohispanicum stalk height occurs through two simultaneous paths, one via producing many seeds and the other through increasing probability of dispersing them far away.  相似文献

Selection for reduced fiber concentration in forage crops is considered to be an effective approach to improve the voluntary intake potential of forages. The purpose of this study was to determine the effectiveness of several modifications to phenotypic recurrent selection for reducing neutral detergent fiber (NDF) concentration and its effect on correlated variables of smooth bromegrass (Bromus inermis Leyss). The selection methods were based on differences in the growth stage sampled (vegetative vs heading), the method of determining NDF in the laboratory (wet-laboratory vs near-infrared reflectance spectroscopy), and the method of intercrossing selected individuals (in situ vs replicated polycross). Selection at the vegetative growth stage was most effective, probably, due to minimal sampling variation within plants. Polycrossing generally increased gains due to more effective interpollination, but increased cycle time by 50%, resulting in similar gains per year for in situ pollination vs polycrossing. Selection for reduced NDF did not generally affect the digestibility of the NDF fraction. Selection for reduced NDF led to reduced forage yield for all selection methods, due partly to a genetic correlation with NDF and partly to inbreeding depression. Three potential solutions were proposed to break the apparent association between reduced NDF and forage yield: increase effective population size, practice combined selection for both traits, and/or make chance hybrids between genetically divergent low-NDF strains. Summary The response to selection for leaf proteins was studied during three selection cycles. Selection for high total nitrogen content showed 75% heritability, and the levels of both ribulose 1,5-bisphosphate carboxylase oxygenase (Rubisco) and cytoplasmic protein were strongly under nuclear DNA control. High and low protein content were correlated with chloroplast area. Although the amounts of nuclear DNA were similar, the ratio of Rubisco/DNA and chlorophyll/DNA changed during the selection process. It can be concluded that the levels of Rubisco achieved in mature plants of M. sativa are under nuclear DNA control. The possible involvement of small subunit (SSU) genes in controlling these levels is discussed.  相似文献

Summary The efficiency of selection for yield and fruit size using parts of the complete-season record was evaluated using data collected for individual strawberry seedlings (Fragaria x annanasa) in four genetic tests. Part-records were generated for each trait using cumulative weekly sums, and these were compared with complete records to assess the value of extended data collection. In addition, part-records were generated using data from alternate weeks of harvest. Scoring separate sets of seedlings in alternate weeks was assumed to allow a doubling of the unselected population size, halving the selection proportion and increasing selection intensity. Heritabilities estimated for part-records were often larger for mid-season cumulative part-records than for the complete-season records. Seasonal patterns for broad-sense and narrow-sense heritabilities differed, suggesting temporal expression of genes that condition yield and fruit size. Predicted genetic gains were largest using cumulative part-records between 4 and 12 weeks of the 15-week season, depending on trait and breeding objective (breeding value versus clonal value), with 8–9 weeks as a reasonable compromise. Selection using alternate-weeks records was identified as an efficient alternative to selection using total complete-season records. Superiority of the alternate-weeks option depends on the balance between increased selection intensity and decreased individual genetic information, and will differ for specific traits and program designs.  相似文献

Associations between neutral genetic markers and genes under selection have been suggested to explain the population genetic structure of neutral genes in cyclically parthenogenetic freshwater invertebrates. A simulation model was constructed in order to analyse the extrapolated consequences of observed fluctuations in genotype frequencies in Daphnia, in the presence of egg banks. When of sufficient depth and magnitude, egg banks in combination with fluctuating selection were shown to maintain genetic variation within populations indefinitely. The level of equilibrium diversity increased with the depth and magnitude of the banks, and with intensity of selection. The same threshold was responsible for genetic differentiation between populations, which was independent of migration rate, and which was attained very rapidly following initial Hardy–Weinberg equilibrium. In the absence of selection, egg banks increased the effective size of local populations, thereby decreasing genetic differentiation at migration-drift equilibrium. These results suggest that egg banks are crucial to the genetic structure in the presence of fluctuating selective pressures, but more data are needed if this knowledge is to be used in an improved general understanding of the genetic structure of cyclically parthenogenetic species.  相似文献

In order to improve the efficiency of cassava (Manihot esculenta Crantz) transformation, two different selection systems were assessed, a positive one based on the use of mannose as the selective agent, and a negative one based on hygromycin resistance encoded by an intron-containing hph gene. Transgenic plants selected on mannose or hygromycin were regenerated for the first time from embryogenic suspensions cocultivated with Agrobacterium. After the initial selection using mannose and hygromycin, 82.6% and 100% of the respective developing embryogenic callus lines were transgenic. A system allowing plant regeneration from only transgenic lines was designed by combining chemical selection with histochemical GUS assays. In total, 12 morphologically normal transgenic plant lines were produced, five using mannose and seven using hygromycin. The stable integration of the transgenes into the nuclear genome was verified using PCR and Southern analysis. RT-PCR and northern analyses confirmed the transgene expression in the regenerated plants. A rooting test on mannose containing medium was developed as an alternative to GUS assays in order to eliminate escapes from the positive selection system. Our results show that transgenic cassava plants can be obtained by using either antibiotic resistance genes that are not expressed in the micro-organisms or an antibiotic-free positive selection system.  相似文献

The effects of population size and selection intensity on the mean response was examined after 14 generations of within full-sib family selection for postweaning gain in mice. Population sizes of 1, 2, 4, 8 and 16 pair matings were each evaluated at selection intensities of 100% (control), 50% and 25% in a replicated experiment. Selection response per generation increased as selection intensity increased. Selection response and realized heritability tended to increase with increasing population size. Replicate variability in realized heritability was large at population sizes of 1, 2 and 4 pairs. Genetic drift was implicated as the primary factor causing the reduced response and lowered repeatability at the smaller population sizes. Lines with intended effective population sizes of 62 yielded larger selection responses per unit selection differential than lines with effective population sizes of 30 or less.  相似文献

Habitat fragmentation is considered to be one of the major threats to biological diversity worldwide. To date, however, its consequences have mainly been studied in an ecological context, while little is known about its effects on evolutionary processes. In this study we examined whether habitat fragmentation affects selection on plant phenotypic traits via changes in plant-pollinator interactions, using the self-incompatible perennial herb Phyteuma spicatum. Specifically, we hypothesized that limited pollination service in small or low-density populations leads to increased selection for traits that attract pollinators. We recorded mean seed production per capsule and per plant as a measure of pollination intensity and assessed selection gradients (i.e., trait-fitness relationships) in 16 natural populations of varying size and density over 2 years. Mean seed production was not related to population size or density, except for a marginal significant effect of density on the mean number of seeds per capsule in 1 year. Linear selection for flowering time and synchrony was consistent across populations; relative fitness was higher in earlier flowering plants and in plants flowering synchronously with others. Selection on inflorescence size, however, varied among populations, and linear selection gradients for inflorescence size were negatively related to plant population size and density in 1 year. Selection for increased inflorescence size decreased with increasing population size and density. Contrary to our expectation this appeared not to be related to changes in pollination intensity (mean seed production was not related to population size or density in this year), but was rather likely linked to differences in some other component of the abiotic or biotic environment. In summary, our results show that habitat fragmentation may influence selection on plant phenotypic traits, thereby highlighting potential evolutionary consequences of human-induced environmental change.  相似文献

Summary Six replicate lines of Drosophila melanogaster, which had been selected for increased abdominal bristle number for more than 85 generations, were assayed by hierarchical analysis of variance and offspring on parent regression immediately after selection ceased, and by single-generation realised heritability after more than 25 generations of subsequent relaxed selection.Half-sib estimates of heritability in 5 lines were as high as in the base population and much higher than observed genetic gains would suggest, excluding lack of sufficient additive genetic variance as a cause of ineffective selection in these lines. Also, there was considerable diversity among the six lines in composition of phenotypic variability: in addition to differences in the additive genetic component, one or more of the components due to dominance, epistasis, sex-linkage or genotype-environment interaction appeared to be important in different lines.Even after relaxed selection, single-generation realised heritabilities in four lines were as high as in the base population. As a large proportion of total genetic gain must have been made by fixation of favourable alleles, the compensatory increase of genetic variability has been sought in a genetic model involving genes at low initial frequencies, enhancement of gene effects during selection and/or new mutations.  相似文献

The amount of genetic variation for resistance to foot rot caused by Pseudocercosporella herpotrichoides, Fusarium spp., and Microdochium nivale and for resistance to head blight caused by Fusarium culmorum are important parameters when estimating selection gain from recurrent selection in winter rye. One-hundred and eighty-six full-sib families of the selfincompatible population variety Halo, representing the Petkus gene pool, were tested for foot-rot resistance at five German location-year combinations (environments) and for head-blight resistance in three environments with artificial inoculation in all but one environment. Foot-rot rating was based on 25 stems per plot scored individually on a 1–9 scale. Head-blight resistance was plotwise scored on a 1–9 scale and, additionally, grain-weight per spike was measured relative to the non-inoculated control plots. Significant estimates of genotypic variance and medium-sized heritabilities (h 2=0.51–0.69) were observed in the combined analyses for all resistance traits. In four out of five environments, the amount of genetic variance was substantially smaller for foot-rot than for head-blight rating. Considerable environmental effects and significant genotype-environment interactions were found for both foot-rot and head-blight resistance. Coefficients of error-corrected correlation among environments were considerably closer than phenotypic correlations. No significant association was found between the resistances to both diseases (r=-0.20 to 0.17). In conclusion, intra-population improvement by recurrent selection should lead to substantial higher foot-rot and head-blight resistances due to significant quantitative genetic variation within Halo. Selection should be carried out in several environments. Lack of correlation between foot-rot and head-blight resistance requires separate infection tests for improving both resistances.  相似文献

Selection on three phenotypic traits was estimated in a natural population of a fungus beetle, Bolitotherus cornutus. Lifetime fitness of a group of males in this population was estimated, and partitioned into five components: lifespan, attendance at the mating area, number of females courted, number of copulations attempted, and number of females inseminated. Three phenotypic characters were measured—elytral length, horn length, and weight; there were strong positive correlations among the three characters. Selection was estimated by regressing each component of fitness on the phenotypic traits. Of the three traits, only horn length was under significant direct selection. This selection was for longer horns and was due mainly to differences in lifespan and access to females. The positive selection on horn length combined with the positive correlations between horn length and the other two characters resulted in positive total selection on all three characters.  相似文献

Replicate harvest simulations were conducted in a large natural population of Panax quinquefolius L.␣(Araliaceae) to determine the selective effects of harvest. We investigated how minimum size requirements and the influence of size on apparency to human harvesters could result in preferential removal of large plants. To determine which plants were encountered in the large population, harvesters were tracked using GPS as they searched for every legally harvestable, adult plant they could find. Plants were assigned stage-specific fitness measures based on their contributions to population growth rate (λ) under three demographically based harvest regimes: no harvest, harvest and harvest removing seeds. Plant size was codified into a size-index equal to the product of total leaf area and stem height. Heterogeneity of slopes was tested to determine if the selection gradients (β) describing the relationship between fitness and size varied among the three harvest regimes. Harvest differentially reduced the fitness of larger plants in one of four individual harvest simulations. The combined harvest simulation significantly altered the selection regime for size in the population of juvenile and adult (harvestable) plants. Seed removal by harvesters intensified fitness declines for larger plants. Because larger plants contribute most to population growth, the selective effects of harvest could result in a shift in the evolutionary dynamics of this species with significant conservation implications.  相似文献

Selection and random genetic drift are the two main forces affecting the selection response of recurrent selection (RS) programs by changes in allele frequencies. Therefore, detailed knowledge on allele frequency changes attributable to these forces is of fundamental importance for assessing RS programs. The objectives of our study were to (1) estimate the number, position, and genetic effect of quantitative trait loci (QTL) for selection index and its components in the base populations, (2) determine changes in allele frequencies of QTL regions due to the effects of random genetic drift and selection, and (3) predict allele frequency changes by using QTL results and compare these predictions with observed values. We performed QTL analyses, based on restriction fragment length polymorphisms (RFLPs) and simple sequence repeats (SSRs), in 274 F2:3 lines of cross KW1265 × D146 (A × B) and 133 F3:4 lines of cross D145 × KW1292 (C × D) originating from two European flint maize populations. Four (A × B) and seven (C × D) cycles of RS were analyzed with SSRs for significant allele frequency changes due to selection. Several QTL regions for selection index were detected with simple and composite interval mapping. In some of them, flanking markers showed a significant allele frequency change after the first and the final selection cycles. The correlation between observed and predicted allele frequencies was significant only in A × B. We attribute these observations mainly to (1) the high dependence of the power of QTL detection on the population size and (2) the occurrence of undetectable QTL in repulsion phase. Assessment of allele frequency changes in RS programs can be used to detect marker alleles linked to QTL regions under selection pressure. Androgenesis is a phenomenon in which microspores are made to bypass the sexual pathway and follow the sporophytic mode of development to generate new plants without the intervention of fertilization under specialized in vitro conditions. Microspore culture provides an ideal system, with a large, relatively uniform population of haploid cells, for use in mutant selection, genetic transformation and in studies on the molecular mechanism of induction of androgenesis and embryogenesis. This paper involves a study on establishing a reproducible and efficient protocol for microspore embryogenesis in various varieties of Brassica juncea. The genotype had a pronounced effect on androgenic response in microspore cultures. The cultivar Rajat exhibited the most response, producing around 3500 embryos/100 buds. The microspores of B. juncea cv. PR-45 from ed plants maintained at a day/night temperature of 10 °C/5 °C form embryos with suspensors with varied morphology. The microspore embryos germinated to produce plants with frequencies. These plants exhibited 52% survival and 74% fertility.  相似文献

Mast-seeding behaviour was monitored in 18 populations of eight species of the African cycad genus Encephalartos between 1988 and 1991. The coefficient of variation (V) in annual cone production for each population ranged between 88 and 200, indicating large fluctuations in reproductive effort between years. Data were collected to determine whether mast-seeding reduced levels of predispersal seed predation by satiating seed predators in mast years and whether it resulted in a reproductive advantage over plants which reproduced more frequently. Masting intensity was greatest in those populations in which individual plants suffered the highest levels of predispersal seed predation in years when only a few plants produced seeds. The principal seed predators were two congeneric weevil species, Antliarhinus zamiae and A. signatus, which develop exclusively on cycad seeds. The lowest intensity of mast-seeding was recorded for cycad populations with low levels of seed predation and in which A. zamiae and A. signatus occurred only in low numbers or were entirely absent. Larger seed crops appeared to result in lower levels of seed predation by A. zamiae and A. signatus in four populations of E. altensteinii, and differences in seed crop size accounted for 48–66% of variation in levels of seed predation in populations of five cycad species. In one population of E. altensteinii, lower levels of seed predation in plants reproducing periodically resulted in a reproductive advantage over plants reproducing more frequently. These results are consistent with the predator satiation hypothesis. However, in most cycad populations, numbers of seed predators did not appear to decrease significantly after a period of 2–8 years between reproductive episodes and, in two of three populations examined, periodic reproduction did not increase the number of seeds surviving to dispersal over a 4-year period. These results are interpreted to mean that periodic reproduction has not evolved in response to selection imposed by seed predators, but that selection may favour those plants which experience lower levels of seed predation by coning in synchrony with the majority of plants in the population.  相似文献

Summary The present study was undertaken to obtain information on average gene frequency in two heterotic populations of maize (Zea mays L.), Mezcla Amarillo Selection (MAS) and J607. Sixty-four male plants were taken in each of the populations and each of these were crossed to a different set of eight plants, four of which belonged to the same population and four to the other population. This resulted in two groups of intra-population (within MAS and within J607) and two groups of inter-population (MAS X J607 and J607 X MAS) progenies. Each group consisted of 256 full-sib progenies on the pattern of the North Carolina Design I mating system. The male plants were selfed to produce 64 S1 prgenies in each population. The materials were evaluated at two diverse locations, Ludhiana and Gurdaspur, for grain yield, ear length, ear girth, number of kernel rows, plant height, ear height and days to silk. An incomplete block design with two replications were used. The plot consisted of a 5 m long row. Ratios of estimated genetic components of variance and covariance were compared with corresponding theoretical ratios computed for a single locus for various gene frequencies and levels of dominance, and approximate ranges of the gene frequencies and their relative magnitude were worked out in the two populations. The average frequency of favourable genes for plant height was estimated as 0.6 in MAS and 0.8 in J607. For grain yield the average gene frequency was 0.8 to 0.9 in MAS and 0.7 to 0.8 in J607 whereas for ear height it was 0.5 to 0.7 in MAS and 0.4 to 0.6 in J607. The gene frequency in the two populations seemed to be similar for days to silk, ear length, ear girth and kernel rows.  相似文献

Summary Replicate lines, each initially with one hundred pairs of parents selected at 50% intensity, were derived from the Canberra strain. In later generations population size was reduced and selection intensity increased. Three lines were selected without irradiation and five with 1000 r X-rays per generation for thirty generations. Selection was continued until generation 66. Long-term responses were similar in unirradiated and irradiated lines, and there was evidence that genes with large effects influenced response patterns in both groups of lines.This work was carried out while B.H. was on leave from the New South Wales Department of Agriculture, and held a Commonwealth Research Studentship. Restriction fragment length polymorphism (RFLP) analysis was used to characterize variability in the Illinois Long-Term Selection Experiment oil strains. Considerable polymorphism was detected within each oil strain and among oil strains. Fifty-two individual plants from each of the Illinois High Oil (IHO), Illinois Low Oil (ILO), Reverse High Oil (RHO) and Reverse Low Oil (RLO) strains were sampled to determine RFLP allele/variant frequencies. Generation 90 was sampled for IHO, RHO, and RLO whereas generation 87 was sampled for ILO. Forty-nine RFLP probes distributed throughout the maize genome were used. Chi-square analysis was performed to determine if RFLP genotypes at each of the 49 RFLP loci were significantly different among strains. Oil strains that have been separated for 90 generations showed high levels of significantly-different RFLP genotypic frequencies. The comparison of ILO vs RHO gave only significant chi-square values while the comparisons of IHO vs RLO and RHO vs RLO had 111 ratios of significant to non-significant chi-square values. Strains that have been separated for only 42 generations showed a lower level of significantly-different RFLP genotypic frequencies. The comparisons of IHO vs RHO and ILO vs RLO both had only a 32 ratio of significant to non-significant chi-squares values. Detection of multiple RFLP alleles/variants among the oil strains was common with 59% of the RFLP loci examined exhibiting multiple variants. A number of RFLP loci in RHO (3) and RLO (11) were associated with a trend in RFLP allele/variant frequencies consistent with a response to reverse selection for oil concentration.  相似文献
