The nematocysts of Chiropsalmus quadrigatus (Cubozoa; Cubomedusa; Chirodropidae) were examined to determine if their composition changes with an increase in body size. Fixed tentacles of specimens collected in Okinawa, Japan, were homogenized and their nematocysts were observed under a differential interference contrast microscope. Six nematocyst types were observed in medusae of all sizes microbasic mastigophores (MM), large and small trirhopaloids (lTR and sTR), holotrichous isorhizas (HI), ellipsoidal isorhizas (eI), and ovoid isorhizas (oI). Two other nematocysts, large ovoid isorhizas (loI) and microbasic euryteles (ME), were observed only in small individuals. There was also marked difference in proportion of tentacular nematocysts between small and large individuals. HI was the dominant type in small specimens, while MM and eI were predominant in large specimens. Nematocyst composition in the bell and pedalia also differed between small and large individuals. Bells of small medusae contained oI and sTR, while only oI were observed in most large individuals. The pedalia of small medusae had clusters of MM, ME, sTR, and oI. Such single clusters on pedalium bases were characteristic of small individuals. The pedalia of large individuals contained scattered oI. Tentacles of medusae are used for prey capture, so the changes in the major type of nematocysts in tentacles may reflect changes in prey type.  相似文献

Nematocysts of the scyphozoans Pelagia noctiluca and Rhizostoma pulmo were examined. In R. pulmo 4 types of nematocyst were observed: heterotrichous microbasic euryteles; holotrichous isorhizas; atrichous a-isorhizas; and atrichous -isorhizas. In P. noctiluca 5 types of nematocyst were seen: heterotrichous microbasic euryteles; heterotrichous isorhizas (previously described as atrichous isorhizas); holotrichous O-isorhizas; atrichous a-isorhizas; and an undescribed type, which in its structure and discharge mechanism resembles microbasic p-mastigophores. The results show, in both P. noctiluca and R. pulmo, a greater variety of nematocysts than described in previous studies.  相似文献

Jellyfish have an increasing impact on marine ecology. Cnidocysts bearing stinging cells afford, amongst others, prey capture and defence. Several different types of stinging capsules are found in one species and they are supposed to have specific functions, e.g. paralysing prey or adhering to it. Due to these assumed different roles of the capsules, it is suggested that toxins, which are contained in the capsules, differ in composition. Analysis of distinct types of nematocysts requires an appropriate method for the separation of the different types. Mixtures of types of nematocysts were obtained of two species of jellyfish, Aurelia aurita and Cyanea lamarckii, by maceration of the tissue. These mixtures were treated with a method called laser microdissection and pressure catapulting (LMPC). Optimized maceration methods, which were firstly introduced as a method for this purpose, in conjunction with optimized LMPC parameters lead to sufficient amounts of separated capsules of individual types for subsequent mass-spectrometric analyses. In case of A. aurita, the resulting mass spectra had some constituents in common, whereas in the overall pattern, the two distinct nematocyst types differed.  相似文献

Abstract. We examined the cnidomes (total complement of nematocysts) of medusae of the zooxanthellate and azooxanthellate jellyfishes Phyllorhiza punctata and Catostylus mosaicus (Rhizostomeae, Scyphozoa), and compared the assemblage of zooplankton captured on the oral arms of each species to determine whether differences in the types or amount of zooplankton captured were consistent with possible differences in the cnidomes. Cnidomes were described using light and scanning electron microscopy. Each species had a distinct cnidome and, in general, specimens of P. punctata appeared to have far fewer nematocysts than those of C. mosaicus. Four types of nematocysts were identified in medusae of C. mosaicus; 2 types of holotrichous isorhizae, rhopaloids, and birhopaloids. In C. mosaicus, the oral arms and bell margins possessed all of these types, but the cnidomes of the 2 regions differed in relative abundances and sizes of isorhizae and rhopaloids. Five types of nematocysts were identified in medusae of P. punctata, although not all types were found in all specimens. Round holotrichous isorhizae were found only in the bell, while oval holotrichous isorhizae, rhopaloids of 2 distinct size ranges, and birhopaloids were found in the bell and oral arms. Cnidomes of the bell and oral arms in specimens of P. punctata also differed in the relative abundance and sizes of oval isorhizae and rhopaloids. Although there were clear differences in the overall cnidomes and absolute abundances of nematocysts in each species, the oral arms (feeding appendages) of specimens of both C. mosaicus and P. punctata had similar types and relative abundances of nematocysts. Zooplankton sampled from the oral arms of each species showed that both species preyed predominantly on copepod nauplii and larvae of gastropods and bivalves. Medusae of C. mosaicus captured ~10 × more gastropod larvae and 5 × more bivalve larvae than those of P. punctata. Specimens of P. punctata captured approximately twice as many copepod nauplii as those of C. mosaicus. Differences in the relative abundance of types of zooplankton captured by each species could not be adequately explained by differences in the cnidomes of the oral arms.  相似文献

Several species of scyphozoan medusae occur in river estuaries and other brackish waters but it is often unknown if the planulae settle and the scyphopolyps reproduce in those low-salinity waters. In the present study, scyphozoan species from the German Bight (North Sea) were tested in laboratory experiments to investigate their tolerance of low salinity. Planula larvae released from medusae in salinity 32 were still active after the salinity was reduced to 10 (Cyanea capillata, Cyanea lamarckii) and to 7 (Chrysaora hysoscella) in laboratory treatments. Planulae did not settle on the undersides of floating substrates when salinity was reduced to <20. By contrast, planulae released from C. capillata medusae in Kiel Bight (western Baltic Sea) in salinity 15 developed into polyps in laboratory cultures. Polyps reared from planulae in salinity 36 survived a reduction to 12 (C. capillata, C. lamarckii) and to 8 (Aurelia aurita). Polyps of all tested species strobilated and released young medusae (ephyrae) in salinity 12. These results show a high tolerance of planulae and polyps to low salinity, indicating their possible occurrence in estuaries and brackish waters. In addition to laboratory observations, young C. capillata ephyrae were collected in the western Baltic Sea (Kiel Bight) in salinity 15, which indicates that they were probably released by a local polyp population. We suggest that the polyps of the painfully stinging lion’s mane, C. capillata, may be more widespread in the Baltic Sea than previously assumed and that the occurrence of the medusae may not only depend on inflow of water masses from the North Sea.  相似文献

The study of hybrid zones is central to our understanding of the genetic basis of reproductive isolation and speciation, yet very little is known about the extent and significance of hybrid zones in marine fishes. We examined the population structure of cod in the transition area between the North Sea and the Baltic Sea employing nine microsatellite loci. Genetic differentiation between the North Sea sample and the rest increased along a transect to the Baltic proper, with a large increase in level of differentiation occurring in the Western Baltic area. Our objective was to determine whether this pattern was caused purely by varying degrees of mechanical mixing of North Sea and Baltic Sea cod or by interbreeding and formation of a hybrid swarm. Simulation studies revealed that traditional Hardy-Weinberg analysis did not have sufficient power for detection of a Wahlund effect. However, using a model-based clustering method for individual admixture analysis, we were able to demonstrate the existence of intermediate genotypes in all samples from the transition area. Accordingly, our data were explained best by a model of a hybrid swarm flanked by pure nonadmixed populations in the North Sea and the Baltic Sea proper. Significant correlation of gene identities across loci (gametic phase disequilibrium) was found only in a sample from the Western Baltic, suggesting this area as the centre of the apparent hybrid zone. A hybrid zone for cod in the ecotone between the high-saline North Sea and the low-saline Baltic Sea is discussed in relation to its possible origin and maintenance, and in relation to a classical study of haemoglobin variation in cod from the Baltic Sea/Danish Belt Sea, suggesting mixing of two divergent populations without interbreeding.  相似文献

Aphanizomenon Morren is an important member of the cyanobacterial community in the Baltic Sea, but studies of this genus have been hampered by the difficulty of growing it in laboratory culture. PCR amplification of DNA from colonies picked directly from water samples has circumvented this problem and made it possible to carry out an analysis of genetic diversity within the Baltic Sea and in two small North American lakes separated by just a few kilometers. The nucleotide sequence of the phycocyanin intergenic spacer and partial flanking coding regions of cpcB and cpcA was determined for 32 colonies of Aphanizomenon , 26 from the Baltic Sea, and 6 from the North American lakes. No variation was detected among the 26 Baltic Sea colonies, but two alleles, differing at 19 nucleotide positions (5.4%), were found in the North American lake colonies. Surprisingly, the two North American types were less closely related to each other than to the Baltic Sea genotype. The Baltic Sea Aphanizomenon is clearly distinct from A. flos-aquae at both the cpcB–cpcA and 16S rDNA loci, which lends phylogenetic support to their tentative separation based on ultrastructural analysis. We conclude that although there is significant genetic diversity in the genus Aphanizomenon , the population in the Baltic Sea is, in contrast to the Nodularia population, genetically homogeneous.  相似文献

To evaluate a rapid and cost-effective method for monitoring bacteria in ballast water, several marine bacterial isolates were characterized by matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS). Since International Maritime Organization (IMO) regulations are concerned with the unintended transportation of pathogenic bacteria through ballast water, emphasis was placed on detecting species of Vibrio, enterococci and coliforms. Seawater samples collected from the North Sea were incubated in steel ballast tanks and the presence of potentially harmful species of Pseudomonas was also investigated. At the genus-level, the identification of thirty six isolates using MALDI-TOF MS produced similar results to those obtained by 16S rRNA gene sequencing. No pathogenic species were detected either by 16S rRNA gene analysis or by MALDI-TOF MS except for the opportunistically pathogenic bacterium Pseudomonas aeruginosa. In addition, in house software that calculated the correlation coefficient values (CCV) of the mass spectral raw data and their variation was developed and used to allow the rapid and efficient identification of marine bacteria in ballast water for the first time.  相似文献

The population status of the harbour porpoise (Phocoena phocoena) in the Baltic area has been a continuous matter of debate. Here we present the by far most comprehensive genetic population structure assessment to date for this region, both with regard to geographic coverage and sample size: 497 porpoise samples from North Sea, Skagerrak, Kattegat, Belt Sea, and Inner Baltic Sea were sequenced at the mitochondrial Control Region and 305 of these specimens were typed at 15 polymorphic microsatellite loci. Samples were stratified according to sample type (stranding vs. by-caught), sex, and season (breeding vs. non-breeding season). Our data provide ample evidence for a population split between the Skagerrak and the Belt Sea, with a transition zone in the Kattegat area. Among other measures, this was particularly visible in significant frequency shifts of the most abundant mitochondrial haplotypes. A particular haplotype almost absent in the North Sea was the most abundant in Belt Sea and Inner Baltic Sea. Microsatellites yielded a similar pattern (i.e., turnover in occurrence of clusters identified by STRUCTURE). Moreover, a highly significant association between microsatellite assignment and unlinked mitochondrial haplotypes further indicates a split between North Sea and Baltic porpoises. For the Inner Baltic Sea, we consistently recovered a small, but significant separation from the Belt Sea population. Despite recent arguments that separation should exceed a predefined threshold before populations shall be managed separately, we argue in favour of precautionary acknowledging the Inner Baltic porpoises as a separate management unit, which should receive particular attention, as it is threatened by various factors, in particular local fishery measures.  相似文献

Drivers of population genetic structure are still poorly understood in marine micro‐organisms. We exploited the North Sea–Baltic Sea transition for investigating the seascape genetics of a marine diatom, Skeletonema marinoi. Eight polymorphic microsatellite loci were analysed in 354 individuals from ten locations to analyse population structure of the species along a 1500‐km‐long salinity gradient ranging from 3 to 30 psu. To test for salinity adaptation, salinity reaction norms were determined for sets of strains originating from three different salinity regimes of the gradient. Modelled oceanographic connectivity was compared to directional relative migration by correlation analyses to examine oceanographic drivers. Population genetic analyses showed distinct genetic divergence of a low‐salinity Baltic Sea population and a high‐salinity North Sea population, coinciding with the most evident physical dispersal barrier in the area, the Danish Straits. Baltic Sea populations displayed reduced genetic diversity compared to North Sea populations. Growth optima of low salinity isolates were significantly lower than those of strains from higher native salinities, indicating local salinity adaptation. Although the North Sea–Baltic Sea transition was identified as a barrier to gene flow, migration between Baltic Sea and North Sea populations occurred. However, the presence of differentiated neutral markers on each side of the transition zone suggests that migrants are maladapted. It is concluded that local salinity adaptation, supported by oceanographic connectivity patterns creating an asymmetric migration pattern between the Baltic Sea and the North Sea, determines genetic differentiation patterns in the transition zone.  相似文献

Despite the high sensitivity and relatively high tolerance for contaminants of matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS) there is often a need to purify and concentrate the sample solution, especially after in-gel digestion of proteins separated by two-dimensional gel electrophoresis (2-DE). A silicon microextraction chip (SMEC) for sample clean-up and trace enrichment of peptides was manufactured and investigated. The microchip structure was used to trap reversed-phase chromatography media (POROS R2 beads) that facilitates sample purification/enrichment of contaminated and dilute samples prior to the MALDI-TOF MS analysis. The validity of the SMEC sample preparation technique was successfully investigated by performing analysis on a 10 nM peptide mixture containing 2 m urea in 0.1 m phosphate-buffered saline with MALDI-TOF MS. It is demonstrated that the microchip sample clean-up and enrichment of peptides can facilitate identification of proteins from 2-DE separations. The microchip structure was also used to trap beads immobilized with trypsin, thereby effectively becoming a microreactor for enzymatic digestion of proteins. This microreactor was used to generate a peptide map from a 100 nM bovine serum albumin sample.  相似文献

 The release of medusae from three hydrozoan fire corals, Millepora dichotoma, M. murrayi and M. platyphylla, was investigated at three sites in southern Taiwan from February 1994 to July 1995. All three species were gonochoristic, and developed and released several batches of medusae between April and May. The duration of open ampulla appearing on the surface of coralla was short, about three months, and could be used to infer the reproductive season of the fire corals between April and May. No obvious lunar cycles of medusa release were found in these species. Medusa release started before dark at approximately 17:00 h and continued for several hours. Males began releasing medusae earlier than females. Synchronization of medusa release between colonies, i.e., the probability of occurring on the same nights, was significantly higher within a species than between different species. Hybridization in nature among the three species is, therefore, unlikely due to segregation in the spawning dates. Moreover, the synchronization within each species was often significantly higher within versus between sites. The free-swimming medusae released gametes within approximately one hour, and the spent medusae lived for a few more hours. Medusae may facilitate fertilization rates as a result of an apparently negatively geotactic swimming response that results in medusa aggregation at the surface. No differences in the sizes of medusae, eggs and sperm were detected among the three species; however, some characteristic differences of medusa nematocysts were found. Populations of the dinoflagellate Dinophysis norvegica in the Baltic Sea and in the adjacent North Sea are infected by the endoparasite Amoebophrya sp. The high diversity recently unveiled within the genus Amoebophrya brings uncertainty about their identities. We applied molecular biology techniques--18S rDNA sequencing and fluorescent in situ hybridization (FISH)--to compare this host-parasite system from both environments. The North Sea Amoebophrya sp. 18S rDNA sequence was 89% identical to the previously described Baltic Sea Amoebophrya sp. sequence, suggesting they are different species. In spite of that, a phylogenetical analysis placed the North Sea parasite sequence in a well-supported cluster with other Amoebophrya sp. sequences. The D. norvegica 18S rDNA sequences from both environments were 100% identical, indicating that the hosts have not evolved independently. A DNA probe designed for the Baltic Sea Amoebophrya sp. 18S rRNA was used in FISH assays on infected D. norvegica populations from both environments. The probe stained all infected cells from the Baltic sample, whereas none from the North Sea were stained. The results indicate that D. norvegica is released from one parasite when entering the Baltic Sea, and become less infected by an alternative parasite species.  相似文献

After the discovery of large densities of Mnemiopsis leidyi in the Baltic Sea near Kiel by Javidpour et al. (First record of Mnemiopsis leidyi A. Agassiz 1865 in the Baltic Sea, 2006) in October 2006, we investigated the gelatinous zooplankton in the North Sea near Helgoland and recorded Mnemiopsis leidyi for the first time in the North Sea, albeit in much lower densities than those recorded in the Baltic Sea.  相似文献

The first finding of the hydromedusa Hydractinia minima (Trinci, 1903) in plankton of Peter the Great Bay (Sea of Japan) is reported. The hydromedusae are 0.24–0.51 mm in bell diameter and 0.25–0.53 mm in height. The size characteristics of the nematocysts of this species (desmonemes and microbasic euryteles) are given. H. minima is present in the plankton of Peter the Great Bay from June through October at water temperatures of 16.4 to 22°C with the highest mean monthly density of 151 ind./m3. A mature medusa of Velella velella (Linnaeus, 1758) is reportedfor the first time from the North Atlantic; previously adultmedusae were known only from the Mediterranean. This specimen,collected by SCUBA divers, is the largest specimen recordedto date. Distinctive features are: two opposite adaxial-axialpairs of perradial capitate tentacles; two marginal bulbs lackingtentacles; conical manubrium with 4 perradial longitudinal gonads;perradial exumbrellar cnidae tracts; cnidome of stenoteles andmacrobasic euryteles; and zooxanthellae within the subumbrella.Since this specimen was collected near the surface and has zooxanthellae,it is likely that V. velella medusae are epipelagic.  相似文献

Genetic population structure of turbot (Scophthalmus maximus L.) in the Northeast Atlantic was investigated using eight highly variable microsatellite loci. In total 706 individuals from eight locations with temporal replicates were assayed, covering an area from the French Bay of Biscay to the Aaland archipelago in the Baltic Sea. In contrast to previous genetic studies of turbot, we found significant genetic differentiation among samples with a maximum pairwise FST of 0.032. Limited or no genetic differentiation was found among samples within the Atlantic/North Sea area and within the Baltic Sea, suggesting high gene flow among populations in these areas. In contrast, there was a sharp cline in genetic differentiation going from the low saline Baltic Sea to the high saline North Sea. The data were explained best by two divergent populations connected by a hybrid zone; however, a mechanical mixing model could not be ruled out. A significant part of the genetic variance could be ascribed to variation among years within locality. Nevertheless, the population structure was relatively stable over time, suggesting that the observed pattern of genetic differentiation is biologically significant. This study suggests that hybrid zones are a common phenomenon for marine fishes in the transition area between the North Sea and the Baltic Sea and highlights the importance of using interspecific comparisons for inferring population structure in high gene flow species such as most marine fishes.  相似文献

Medusae are important members of marine food webs, but are rare in lakes. In one of the largest lakes in the world, Lake Tanganyika, a small medusa (Limnocnida tanganyicae) is a prominent component of zooplankton. We used field and laboratory methods to study the ecological role of Lake Tanganyika medusae, which occasionally reached high local densities in the whole epilimnion. The largest individuals showed low amplitude, diel vertical migration which minimized their exposure to harmful UV radiation and also may be important for picocyanobacteria regularly present in the medusae. The endosymbiotic picocyanobacteria differed morphologically among medusae and were predominantly one Lake Biwa type Cyanobium sp. that typically was abundant in the water column. Under light, some medusae were net primary producers. Although nitrogen stable isotopic ratios indicated that the free-living cyanobacteria were nitrogen-fixers, the picocyanobacteria in medusae obtained nitrogen predominantly from their host. Stable isotopic ratios of carbon and nitrogen further suggested that copepods were the most likely prey for the medusae. Lake Tanganyika medusae apparently base their metabolism both on animal and plant sources, with possible internal cycling of nutrients; however, the role of picocyanobacteria gardening in the Lake Tanganyika ecosystem and its medusae requires quantification.  相似文献

Matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) and ion-pair-reverse-phase high-performance liquid chromatography (IP-RP HPLC) techniques were combined to determine the sequence identity of short single-stranded deoxyoligonucleotides. This methodology is demonstrated using a commercially available multiplex set of eight primer pairs. The primer pairs were separated and collected by IP-RP HPLC. Partial sequence information for IP-RP HPLC fractions was obtained from analyzing exonuclease digestion products by MALDI-TOF MS. IP-RP HPLC, MALDI-TOF MS, exonuclease digests, and a simple computational algorithm provide an integrated strategy for determining the sequence of short nucleic acid oligomers.  相似文献

Proceeding from long-term data on the numbers of eggs of cod in ichthyoplankton, the total annual production of cod eggs at four main spawning grounds of the Baltic Sea was calculated. It was shown that the long-term fluctuations of cod egg production were positively related to the dynamics of the volume of waters coming to the Baltic Sea in years of the North Sea advections. It is suggested that this dependence was determined by a set of adaptations providing the extension of cod reproduction upon the improvement of the environment.  相似文献
