Morpho‐anatomical and physiological differences between sun and shade leaves in Abies alba Mill. (Pinaceae,Coniferales): a combined approach 下载免费PDF全文
Morphology, anatomy and physiology of sun and shade leaves of Abies alba were investigated and major differences were identified, such as sun leaves being larger, containing a hypodermis and palisade parenchyma as well as possessing more stomata, while shade leaves exhibit a distinct leaf dimorphism. The large size of sun leaves and their arrangement crowded on the upper side of a plagiotropic shoot leads to self‐shading which is explainable as protection from high solar radiation and to reduce the transpiration via the lamina. Sun leaves furthermore contain a higher xanthophyll cycle pigment amount and Non‐Photochemical Quenching (NPQ) capacity, a lower amount of chlorophyll b and a total lower chlorophyll amount per leaf, as well as an increased electron transport rate and an increased photosynthesis light saturation intensity. However, sun leaves switch on their NPQ capacity at rather low light intensities, as exemplified by several parameters newly measured for conifers. Our holistic approach extends previous findings about sun and shade leaves in conifers and demonstrates that both leaf types of A. alba show structural and physiological remarkable similarities to their respective counterparts in angiosperms, but also possess unique characteristics allowing them to cope efficiently with their environmental constraints. 相似文献

Behavior of chloroplasts and chloroplast nuclei during spermatogenesis in the fern,Pteris vittata L.
Summary The fate of the chloroplasts and chloroplast nuclei (cp-nuclei) was followed during spermatogenesis in the fernPteris vittata L. by epifluorescence microscopy after staining with 4-6-diamidino-2-phenylindole (DAPI) and by quantitation of chloroplast DNA (cp-DNA) by fluorimetry using a video intensified microscope photon counting system (VIMPICS). The spores were grown on solid medium that contained antheridiogen (Anptd), and formed an antheridium initial on the protonema cell. The antheridium initial divided and produced 16 spermatocytes and 3 surrounding cells. The chloroplasts in the spermatocytes decreased in volume as cell division was repeated, until finally the volume of each chloroplast was 1/15 of that of the primary chloroplasts. The DNA content of the chloroplasts was also reduced to 1/5 of the original value and when the sperm matured, the fluorescence of cp-DNA disappeared. In the 16-cell spermatocyte, the recognition of the fluorescence of chlorophyll in the chloroplasts with a green excitation filter became difficult. But, the plastids could be observed until the final stage of the sperm. From these observations, it appears that there are two steps in the metamorphosis of chloroplasts during spermatogenesis in the fern. The first step involves the decrease in the volume of chloroplasts, accompanied by reduction of the DNA content, and the second step involves the change of the physical state of chloroplasts to amyloplasts and the disappearance of the cp-DNA from the amyloplasts. 相似文献

The induction of sun and shade leaves of the European beech (Fagus sylvatica L.): anatomical studies
Summary Primordia from buds of sun and shade twigs of European beech (Fagus sylvatica L.) were collected six times a year for anatomical investigations. Differentiation into sun-leaf and shade-leaf primordia was first observed in early August. Sun-leaf primordia had five, and shade-leaf primordia four layers of mesophyll meristem cells. With potted graft unions of beeches possible structural changes of leaf primordia were investigated. Trees adapted to shade develop sun-leaf primordia when put into full daylight, provided the transfer happened before July. Trees adapted to full daylight developed leaf primordia which remained structurally sun-leaf primordia when the plant was kept under shade conditions. Shadeleaf branches of young beech trees cut in February in order to expose the shade buds to full daylight developed either shade leaves or intermediate shade/sun leaves. These experiments show that the subtending leaf may provide the developing axillary bud with photoassimilates, but its character, whether sun or shade leaf, has no influence on the character of the developing leaf primordia. 相似文献

Divergence time estimates suggest that most clades constituting the fern family Pteridaceae (Polypodiales) were in existence by the Early Cretaceous. However, fossil evidence to corroborate this remains exceedingly rare. Burmese amber is an important source of new information on the radiation of derived fern lineages during the Cretaceous Terrestrial Revolution. This study describes Heinrichsia cheilanthoides gen. et sp. nov., a fern with suggested affinities to Pteridaceae, based on fertile foliage portions preserved in Early Cretaceous (~100 Ma) amber from Myanmar. Heinrichsia cheilanthoides is characterized by a pinnate‐pinnatifid frond that bears apical, marginal sori protected by a pseudoindusium. Sporangia are of the polypod type and contain tetrahedral‐globose, trilete spores with a striate perine. This discovery provides a new calibration point to test and refine molecular clock‐based concepts of the evolutionary history of the Pteridaceae. Heinrichsia cheilanthoides further substantiates the suggestion that the Cretaceous forests of Myanmar were home to a rich fern flora. 相似文献

Plants experience a number of limiting factors, as drought and heat, which are often coinciding stress factors in natural environment. This study evaluated the changes in mesophyll cell ultrastructure in the leaves of two varieties of winter wheat (Triticum aestivum L.), differing in their drought tolerance, under individual or combined drought and heat treatment. Although the individual stress factors affected leaf ultrastructure, the damaging effect of the combined drought and heat was more pronounced and manifested certain differences between genotypes. Chloroplasts and mitochondria were affected in a variety-specific manner under all adverse treatments. The organelles of the drought-tolerant Katya were better preserved than those in the sensitive variety Sadovo. Leaf ultrastructure can be considered as one of the important characteristics in the evaluation of the drought susceptibility of different wheat varieties. 相似文献

The distribution and ultrastructural features of peripheral nerve processes in the extra-retinal layers of the eyes of the zebrafish, Danio rerio (Hamilton), were investigated using light and transmission electron microscopy. A comparative study of the quality of preservation provided by three different fixation procedures revealed no consistently striking general differences. However, somewhat subjectively, the fixative containing Millonig's buffer did consistently provide better fixation of myelin. Overall, nerve processes, depending on the site studied, were distributed as either (1) bundles (in the choroid near the optic nerve head and in the choroid adjacent to the limbus), (2) linear arrays (in the junction between the sclera and cartilage and in the choroid adjacent to the retina) or (3) individual units (in the choroid under the cartilage or in the sclera). Both myelinated and unmyelinated processes were identified in these locations. Myelinated processes usually contained both neurofilaments and neurotubules, but a few apparently contained only neurofilaments. Unmyelinated processes usually contained mainly neurotubules, but a few apparently contained only neurofilaments. Taken together, these findings indicate innervation of extra-retinal structures, as seen in zebrafish, is highly conserved among vertebrates, further supporting the use of zebrafish as a model for the vertebrate visual system. 相似文献

How are microphytobenthic biofilms adapted to the high incident irradiances and temperatures, low inorganic nutrient concentrations and high desiccation stresses on intertidal flats present in tropical environments? This study investigated biofilms subject to different environmental conditions in a range of tropical sites in Suva lagoon, Fiji. PAM fluorescence was used to measure photophysiological responses to the light climate. Biofilm colloidal carbohydrate, extracellular polymeric substances (EPS) and low molecular weight (MW) carbohydrate concentrations and diel carbohydrate production patterns were measured. Average biomass (Chl a) ranged from 15 to 36?mg?m?2, and was highest in seagrass bed sediments, but biomass was not correlated with water column or sediment porewater nutrient concentrations. Biofilm photophysiology differed significantly along a combined gradient of light and nutrient availability, with F v/F m, relative ETRmax and E k of biofilms highest in mangrove and intertidal main island sites and lowest in subtidal coral reef flats. Subtidal biofilms showed photoinhibition at irradiances > 1000?µmol?m?2. Significant correlations between Chl a and colloidal carbohydrate concentrations were present (except on intertidal sandflats), and tropical biofilms had higher ratios of colloidal carbohydrate and EPS to Chl a than temperate estuarine biofilms, probably due to a combination of high irradiance and low nutrient availability leading to the production of excess photoassimilates. The percentage of EPS present in the colloidal fraction was highest in coral sand biofilms (42%), which had the lowest nutrient concentrations, compared with other sites (25–32%). Intertidal biofilms predominantly consisted of large motile taxa and showed strong rhythms of vertical migration. During tidal emersion, high sediment temperatures (41?°C), irradiance (>2300?µmol?m?2?s?1) and salinity (49‰) stimulated downward migration. In silty sediments, migration resulted in a reduction in photosynthetic activity during the midday period but, in sands with high light penetration (to a depth of > 1700?µm), high production rates of EPS (18.2?µg carbo. µg Chl a?1 h?1) and low MW carbohydrate exudates (40.2?µg carbo. µg Chl a?1 h?1) occurred. Vertical migration, high E k and high rates of photoassimilate dumping are all adaptations to living in the tropical intertidal zone. Seagrass and reef flat biofilms consisted of a diverse non-migratory flora of motile and non-motile taxa that were not subject to such extreme temperature and irradiance conditions. The development of secondary sexual characters, the petasma, and thelycum growth were studied in Xiphopenaeus kroyeri. In adult females, the thelycum is a single plate and its anterolateral portion is characterized by a reduced hood. The aperture resembles a transverse ridge. In immature stages, the ridge has a space between the plates, which becomes narrower as it reaches the end of development. The female gonopore is ‘comma’ shaped. In adult males, the endopods of the petasma are linked at the dorsomedial margin by a large quantity of cincinnuli. In juveniles, cincinnuli gradually increase in number until they join both endopods. At the end of development the petasma is T-shaped. The male gonopore is C-shaped. The relative growth of the petasma total length versus juvenile body length showed a highly positive allometry, whereas in adults the growth was isometric. For the relationship carapace length versus thelycum width, the juvenile phase of females is characterized by an isometry and the adult phase by a negative allometry. 相似文献

The histology of the digestive tract of the amberjack ( Seriola dumerili , Risso) was studied using light and scanning electron microscopy. The anterior oesophagus mucosa displays primary and secondary folds lined with a stratified squamous epithelium with fingerprint-like microridges which is substituted, on the top of the oesogaster folds, by a simple columnar epithelium with short microvilli. Only primary folds are present in the stomach. The anterior portion is rich in simple tubular glands, whereas the oesogaster and the pyloric region are devoid of them. Pyloric caeca and anterior and middle intestine mucosa display the same pattern of folding. The dominant cell type is the enterocyte, which exhibits larger and thinner microvilli in the caeca than in the intestine. The columnar epithelium of the rectum is replaced, in the anal sphincter, by a stratified flattened epithelium. Goblet cells are numerous throughout the whole length of the tract with the exception of the initial part of the oesophagus, the oesogaster, the stomach and the anal sphincter. Mucosubstances have been shown to vary in the different regions of the gut: acid mucines are found in the oesophagus, pyloric stomach, caeca, intestine and rectum, whereas neutral mucosubstances dominate in the anterior portion of the stomach. The muscularis is well developed throughout the length of the tract: two layers of striated muscle at the oesophageal level; two layers of smooth muscle in the stomach wall and three at the intestinal level. 相似文献

Abstract A simple mechanical method for the rapid isolation of chloroplasts with high rates of photosynthesis from young leaves of oat (Avena sativa L.) was described. The photosynthetic activity of these chloroplasts was stable for at least 2 h with rates of CO2-dependent O2 evolution of 30–40 μmol g 1 Chl s 1. The photosynthetic properties of these chloroplasts were similar to those reported for spinach and pea chloroplasts isolated by mechanical disruption. The pH optimum for photosynthetic O2 evolution was pH 7.6. The induction time was 0.5–2 min. Maximal rates of photosynthetic O2 evolution in these chloroplast preparations were obtained in the absence of both divalent cations and EDTA. Addition of divilent cations strongly inhibited photosynthesis which could be partially restored by the subsequent addition of EDTA. But when these cations were not present in the assay medium the addition of EDTA greater than 1 mol m 3 decreased photosynthetic activity. The optimal orthophosphate concentration required for photosynthesis in these chloroplast preparations was 0.2–0.3 mol m 3. In contrast, the addition of pyrophosphate either in the light or dark inhibited photosynthesis. In a comparative study, chloroplasts were also isolated from oat and wheat (Triticum aestivum L., cultivar Hybrid C306) protoplasts. These chloroplast preparations were found to have properties similar to those determined for oat chloroplasts isolated by the mechanical method reported above. 相似文献

BACKGROUND AND AIMS: Dryopteris crassirhizoma is a semi-evergreen fern growing on the floor of deciduous forests. The present study aimed to clarify the photosynthetic and storage functions of overwintering leaves in this species. METHODS: A 2-year experiment with defoliation and shading of overwintering leaves was conducted. Photosynthetic light response was measured in early spring (for overwintering leaves) and summer (for current-year leaves). KEY RESULTS: No nitrogen limitation of growth was detected in plants subjected to defoliation. The number of leaves, their size, reproductive activity (production of sori) and total leaf mass were not affected by the treatment. The defoliation of overwintering leaves significantly reduced the bulk density of rhizomes and the root weight. The carbohydrates consumed by the rhizomes were assumed to be translocated for leaf production. Photosynthetic products of overwintering leaves were estimated to be small. CONCLUSION: Overwintering leaves served very little as nutrient-storage and photosynthetic organs. They partly functioned as a carbon-storage organ but by contrast to previous studies, their physiological contribution to growth was found to be modest, probably because this species has a large rhizome system. The small contribution of overwintering leaves during the short-term period of this study may be explained by the significant storage ability of rhizomes in this long-living species. Other ecological functions of overwintering leaves, such as suppression of neighbouring plants in spring, are suggested. 相似文献

The tropical rainforest mesocosm within the Biosphere 2 Laboratory, a model system of some 110 species developed over 12 years under controlled environmental conditions, has been subjected to a series of comparable drought experiments during 2000–2002. In each study, the mesocosm was subjected to a 4–6 week drought, with well‐defined rainfall events before and after the treatment. Ecosystem CO2 uptake rate (Aeco) declined 32% in response to the drought, with changes occurring within days and being reversible within weeks, even though the deeper soil layers did not become significantly drier and leaf‐level water status of most large trees was not greatly affected. The reduced Aeco during the drought reflected both morphological and physiological responses. It is estimated that the drought‐induced 32% reduction of Aeco has three principal components: (1) leaf fall increased two‐fold whereas leaf expansion growth of some canopy dominants declined to 60%, leading to a 10% decrease in foliage coverage of the canopy. This might be the main reason for the persistent reduction of Aeco after rewatering. (2) The maximum photosynthetic electron transport rate at high light intensities in remaining leaves was reduced to 71% for three of the four species measured, even though no chronic photo‐inhibition occurred. (3) Stomata closed, leading to a reduced ecosystem water conductance to water vapour (33% of pre‐drought values), which not only reduced ecosystem carbon uptake rate, but may also have implications for water and energy budgets of tropical ecosystems. Additionally, individual rainforest trees responded differently, expressing different levels of stress and stress avoiding mechanisms. This functional diversity renders the individual response heterogeneous and has fundamental implications to scale leaf level responses to ecosystem dynamics. 相似文献

采用盆栽方法研究了旺长期遮光及光照转换对不同肥料条件下烟草叶片光合速率(Pn)与荧光特性的影响。结果表明,遮光促进两种肥料条件下烟草叶片叶绿素(Chl)和类胡萝卜素(Car)含量的积累,却降低它们的Pn。其中施无机肥的烟草叶片Chl含量增加较多,而50%无机肥+50%饼肥配施烟草叶片的Car增幅显著,从而,施无机肥烟草叶片Chl/Car上升,而无机肥+饼肥配施烟草叶片Chl/Car却下降。无论从自然光转至遮荫条件下还是从遮荫条件转至自然光下,两种肥料条件下生长的烟草叶片的Pn、实际光化学效率(ΦPSⅡ)、最大光化学效率(Fv/Fm)及光化学猝灭系数(qP)均急剧下降,但与施无机肥相比,无机肥+饼肥配施烟草叶片保持较高的Pn、ΦPSⅡ、Fv/Fm、及qP,可能是饼肥促进了栽培条件下烟草植株的光生态适应性。 相似文献

Mucosa from the hard and soft palates, molar gingiva, cheek and dorsal surface of the tongue of the rat was examined in the light microscope, following Mallory's triple connective tissue stain, and in the scanning and transmission electron microscopes. The epithelium covering the hard palate, gingiva, the smooth band of mucosa at the junction of the hard and soft palates, intermediate zones of the soft palate, fungiform papilla-like structures in the central zone of the soft palate, the fungiform papillae, and the more superficial part and posterior surfaces of the filiform papillae of the tongue all exhibited complete orthokeratinization. The oral surfaces of the epithelial cells in all these areas had a honeycomb pattern of interconnecting ridges surrounding depressions. Imprints of the overlying cells that had been desquamated were apparent, and the lateral boundaries between the cells were formed by two raised ridges separated by a gap. The epithelium covering the cheek, central zone of the soft palate apart from the fungiform papilla-like structures, lateral zones of the soft palate, gingival crevice, and the mucosa between the fungiform and filiform papillae of the tongue all exhibited incomplete orthokeratinization. The oral surfaces of the epithelial cells in all these areas were relatively smooth and did not exhibit a honeycomb pattern of interconnecting ridges. Imprints of the overlying cells that had been desquamated and the lateral boundaries between the cells were only very occasionally found. In the transmission electron microscope the outlines of the cells were compatible with the surface patterns seen in the scanning electron microscope. The possible relationships between the degree of orthokeratinization and ultrastructure of the various epithelia are discussed. 相似文献

In a study of sexual reproduction in long-lived semelparous plants, we observed Agave macroacantha in the tropical desert of Tehuacán-Cuicatlán, Mexico, describing duration of flowering, flower phenology, and nectar production patterns. We also performed two manipulative experiments evaluating (a) the seed production efficiency of different crossing systems (selfing, cross-pollination, apomixis, and control), and (b) the effect of different pollinators (diurnal exposure to pollinators, nocturnal exposure, exclusion, and control) on the seeds produced. Flowering occurred from early May to late July and had a mean duration of 29 days in the individual rosettes. The flowers were protandrous; anthesis occurred in the afternoon of the third day after floral opening, and the pistils matured in the afternoon of the fifth day. The stigmas remained receptive from dusk to the following morning. Pollination was mostly allogamous. Nectar was produced principally during the night, from the first stages of floral aperture until the stigmas wilted and flowering ceased. The flowers were visited during the day by hymenoptera, butterflies, and hummingbirds and during the night by bats and moths. Only the nocturnal visitors, however, were successful pollinators. Agave macroacantha is extremely dependent on nocturnal pollinators for its reproductive success. 相似文献

The electron transport rates of photosystems II and I, amounts of electron carriers, coupling factor activity and photosynthetic rates were investigated in thylakoids isolated from pea plants grown under a wide range of light intensities (16 h light-8 h dark). The electron transport rates of PS II and PS I, as partial reactions or in whole chain, and coupling factor activity on a unit chlorophyll basis, all increased as the light intensity available for growth was altered from a very low intensity of 10 E m-2s-1 to a high intensity of 840 E m-2s-1. Similarly, there were increases in the amounts of atrazine binding sites, plastoquinine, cytochrome f and P700 per unit chlorophyll; significantly, the amounts of reaction centres of PS II and PS I were not equal at any light intensity. The rate of change of all parameters with respect to light intensity could be represented by two straight lines of different slopes which met at a transition point corresponding to approximately 200 E m-2s-1 during growth. These photoadaptations were similar to those observed for both the relative distribution of chlorophyll in chlorophyll-protein complexes and the chl a/chl b ratios [Leong and Anderson, 1984, Photosynthesis Research 5:117–128]. Since these thylakoid components and functions were affected in the same direction by light intensity during growth and all show linear relationships with chl a/chl b ratios, it indicates that they are closely regulated and markedly well co-ordinated. Plants compensate for the limited amount of low light intensities by drastically increasing the light-harvesting antenna unit size of photosystem II and to a lesser extent that of photosystem I. Changes in the composition of the thylakoid membranes exert a regulatory effect on the overall photosynthetic rate up to approximately 450 E m-2s-1.Abbreviations chl
