Relation between peripheral chemoreceptors stimulation and pulmonary arterial blood pressure in rats

As interactions between peripheral chemoreceptors stimulation (PCS) and pulmonary vasomotor tone remain controversial, experiments were made in rats in order to clear up the effects of PCS on pulmonary arterial pressure (PAP). Different stimulations varying in intensities were used, in rats nervously intact (IR-rats), after vagotomy (XT-rats), after chemodenervation obtained without vagotomy (CDN not XT-rats) or with XT (CDN + XT) and finally after alpha 1-receptors blockade (P-rats = pretreated rats). The observed variations were analysed in view of disentangling reflex part of PCS from a direct activity on the pulmonary vascular bed. Ventilation, PAP and systemic blood pressure (BP) were studied in anaesthetized rats. N2 test, NaCN test, 20 s of 5% O2 inhalation and almitrine bismesylate (ALM) were used as PCS, ranged in the order of their relative intensities, from the ventilatory responses observed in IR-rats. In IR-rats, N2-and CN test produced a similar transient increase of PAP, slightly more extended than the hyperventilation. After XT, the responses were prolonged, but amplified only in CN test. Ventilatory responses disappear after CDN, but as far as pulmonary hypertension is concerned, CDN + XT is more potent than CDN without XT to reduce or even suppress them. This fact is particularly evident with ALM who is the strongest PCS used. Similar reduction of PAP rise was also produced in P-rats in which ventilatory responses remain unchanged. Prolonged hypoxic inhalation induced a progressive fall of systolic BP and of PAP. The relationship between selected zinc (Zn) metabolism parameters, arterial blood pressure, age, and renin-angiotensin-aldosterone
system in subjects of both sexes with mild primary arterial hypertension is presented in this study. The following parameters
were measured: systolic and diastolic arterial blood pressure, total and ouabain-dependent efflux rate constants of Zn from
lymphocytes, serum and lymphocyte Zn concentrations, serum aldosterone, angiotensin-converting enzyme, sodium and potassium
concentrations, body mass index, and plasma rennin activity. When all subjects are taken into account, no significant age-related
differences were found for serum Zn. If divided into men and women, negative (r=−0.39) and positive (r=0.34) correlations are observed, respectively. Lymphocyte Zn correlated negatively with age in the entire group (r=−0.55) and also for men (r=−0.54) and women (r=−0.57).
The renin-agiotensin-aldosterone system parameters correlated with those of Zn metabolism only for women: plasma rennin activity
with total Zn efflux from lymphocytes (r=−0.33) and with lymphocyte Zn (r=0.71); the angiotensin-converting enzyme with total Zn efflux from lymphocytes (r=−0.35), with the oubain-dependent Zn efflux from lymphocytes (r=−0.33) and with lymphocyte Zn (r=0.57); serum aldosterone with oubain-dependent Zn efflux from lymphocytes (r=−0.44) and with lymphocyte Zn (r=0.59). For the men, the only positive correlation was that of serum Zn and aldosterone (r=0.45). In all cases (men and women), there was no negative correlation between serum Zn and angiotensin-converting enzyme.
In women, the diastolic blood pressure correlated negatively with total Zn efflux from lymphocytes (r=−0.39), oubain-dependent Zn efflux from lymphocytes (r=−0.49), and serum Zn (r=−0.46); systolic blood pressure correlated negatively with lymphocyte zinc (r=−0.38). In men, the systolic blood pressure had a negative correlation with lymphocyte zinc (r=−0.32), which was also true for the entire group (r=−0.34). These results clearly show gender-related differences in Zn metabolism and indicate the need for further research
to elucidate the possible causes of this phenomenon not only for Zn but for other elements as well. 相似文献

The aim of this study was to compare zinc (Zn) metabolism parameters in groups of premenopausal and postmenopausal women with moderate and severe primary arterial hypertension. The study included 38 women, of which 15 were premenopausal and 23 were postmenopausal. Postmenopausal women had a positive correlation between total (ERCt-Zn) and oubain-dependent (ERCos-Zn) rate constants of Zn efflux from lymphocyte (k = 0.52). In premenopausal women’s ERCos-Zn was negatively but weakly correlated with serum Zn (Zn-s) (k = 0.35). The Zn ERCt-Zn and ERCos-Zn did not show any correlation with age, as did Zn-s. Lymphocyte Zn correlated negatively with age only in premenopausal women (k = -0.62). The renin-angiotensin-aldosterone system correlated with Zn metabolism parameters. In premenopausal women, plasma renin activity and serum aldosterone showed positive correlations with lymphocyte Zn (Zn-l) (k = 0.63 andk = 0.41, respectively), and in postmenopausal women, it correlated negatively with Zn-s (k = -0.38) and whole aldosterone correlated negatively with ERCos-Zn (k = -0.41). Positive correlations between Zn metabolism parameters and arterial blood pressure in premenopausal women were as follows: ERCt-Zn with diastolic blood pressure (dRR) (k = 0.40) and ERCos-Zn with dRR (k = 0.47). In postmenopausal women, the correlations between ERC-t-Zn and dRR and systolic blood pressure (sRR) were negative (k = -0.53 andk = -0.63, respectively). A similar situation was observed between dRR and sRR and Zn-s (k = -0.40 andk = -0.38, respectively). The body mass index (BMI) was positively correlated with ERCt-Zn in premenopausal women (k = 0.36), whereas in postmenopausal, it was negatively correlated with ERCos-Zn (k = -0.42). For the whole group, negative correlations were seen between ZnS and dRR and sRR (k = -0.36 andk = -0.39, respectively) and between ERCos-Zn and BMI (k = -0.39). The results presented show differences in Zn metabolism in arterial hypertension between premenopausal and post-menopausal women. The role of estrogens in these differences is disscused. 相似文献

The contents of norepinephrine (NE), epinephrine (E), dopamine (DA), normetanephrine (NMN), and 4-hydroxy-3-methoxyphenylethylene glycol (MHPG) were measured in the plasma and cerebrospinal fluid (CSF) of 66 patients with primary hypertension and 24 patients with normal blood pressure and minor neurological disorders. Plasma and CSF NE and NMN concentrations were raised in the hypertensive patients. The plasma and CSF NE levels and arterial blood pressure of a small subset of hypertensive patients were normalized after clonidine therapy. In hypertensive patients the content of DA was lower and the ratio of NE/DA was greater; CSF and plasma NE contents were related to the level of arterial blood pressure; and the content of MHPG in CSF was linked strongly with NE content in plasma and CSF and to the level of arterial blood pressure. Thus both central sympathetic nerve tone and peripheral sympathetic nerve tone were enhanced in young patients with uncomplicated hypertension. The elevated levels of neurohormones and their metabolites in some patients with primary hypertension may be related to increased synthesis and release of neural NE and may be pathogenic in the blood pressure elevation. 相似文献

Research on humans at high-altitudes contributes to understandingthe processes of human adaptation to the environment and evolution.The unique stress at high altitude is hypobaric hypoxia causedby the fall in barometric pressure with increasing altitudeand the consequently fewer oxygen molecules in a breath of air,as compared with sea level. The natural experiment of humancolonization of high-altitude plateaus on three continents hasresulted in twoperhaps threequantitatively differentarterial-oxygen-content phenotypes among indigenous Andean,Tibetan and Ethiopian high-altitude populations. This paperillustrates these contrasting phenotypes by presenting evidencefor higher hemoglobin concentration and percent of oxygen saturationof hemoglobin among Andean highlanders as compared with Tibetansat the same altitude and evidence that Ethiopian highlandersdo not differ from sea-level in these two traits. Evolutionaryprocesses may have acted differently on the colonizing populationsto cause the different patterns of adaptation. Hemoglobin concentrationhas significant heritability in Andean and Tibetan samples.Oxygen saturation has no heritability in the Andean sample,but does among Tibetans where an autosomal dominant major genefor higher oxygen saturation has been detected. Women estimatedwith high probability to have high oxygen saturation genotypeshave more surviving children than women estimated with highprobability to have the low oxygen saturation genotype. Thesefindings suggest the hypothesis that ongoing natural selectionis increasing the frequency of the high saturation allele atthis major gene locus. 相似文献

A negative relationship between water hardness and cardiovascular mortality rate was demonstrated and became a source of interest
regarding minerals and trace metals in the pathogenesis of atherosclerosis, cardiovascular diseases, and arterial hypertension.
Higher incidences of sudden death, cerebrovascular diseases, arterial hypertension, and coronary heart disease have been reported
in soft water areas. A major research effort has been devoted to the problem in an attempt to find a protective factor in
hard water or a detrimental factor or element in soft water. The roles of calcium, magnesium, cobalt, lithium, vanadium, silicon,
manganese, and copper have been considered potentially beneficial, whereas those of cadmium, lead, silver, zinc, and antimony
have been considered potentially harmful. Cobalt and zinc have been attributed both roles. In the present article, the role
of trace quantities of several elements in mineral water in the etiopathogenesis of primary arterial hypertension is reviewed.

People who visit high-altitude areas are exposed to a stressful environment and a good percentage of them suffer from high-altitude-induced diseases, including systemic hypertension. Identification of genetic markers for high-altitude-induced diseases would help to reduce the rate of morbidity/mortality from such diseases. The development of systemic hypertension on exposure to high altitude (3,500 m) for 30 days in otherwise normotensive natives of low-altitudes was investigated. The angiotensin-converting enzyme (ACE) insertion/deletion (I/D) genotypes and renin-angiotensin-aldosterone system were simultaneously studied. In the hypertensives during their stay at high altitude, the ACE D allele frequency was significantly higher than in the normotensives (0.67 versus 0.32 chi(2)(1) = 10.6, P < 0.05). In the normotensives during their stay at high altitude, there was no significant increase in plasma aldosterone levels despite increased plasma renin activity. Results of the present study suggest that environmental changes and pre-existing genetic factors, namely the ACE D allele, might be two of the factors predisposing natives of low altitudes to systemic hypertension, a polygenic disease, at high altitude. 相似文献

Serotonin content and accumulation in platelets and its release from them, as well as changes in thrombus formation in mesenteric arterioles and venules of the small intestine have been investigated in control rats and rats with spontaneous hypertension (SHR). Serotonin accumulation in platelets was determined upon its incubation with platelets. Disodium ADP salt was used as an inductor of release. Laser-induced thrombosis was caused by microvessels exposure to impulse laser irradiation. The control animals revealed a significant difference between the initial serotonin platelet level and serotonin level upon incubation and release; in values, the values of basic thrombus-forming parameters were higher than in arterioles. In SHR there is a decrease in biogenic amine content in platelets, a depression in its accumulation and release, an increase in the time of thrombus growth, its size up to the separation of the first embolus and its length along the vascular wall. It is concluded that spontaneous hypertension is characterized by decreased functional activity of platelets and depressed resistance of arterioles and venules to thrombus formation. 相似文献

It was shown in rat experiments that high-altitude hypoxia inhibits the growth of transplantable tumors and enhances the efficacy of antitumor treatment with chemotherapeutic drugs (thiotepa for Heren's carcinoma, and cyclophosphamide for sarcoma 45). Under high-altitude conditions, the cyclophosphamide toxicity as regards blood cells was modified, manifesting in the absence of anemization and less considerable reduction in the platelet and lymphocyte counts. The animals' adaptation to high-altitude hypoxia was followed by a rise in the blood content of sulfhydryl groups and increase in cancerolytic activity of the blood serum. 相似文献

Regulation of the systemic and peripheral hemodynamics in the conditions of acute nitrite hypoxia (doses of NaNO2 10, 30, and 50 mg/kg of the body mass) were studied on white male rats. It was shown that NaNO2 causes a quick dose-dependent decrease in the blood pressure with an intensification of the parasympathetic tonus and development of bradycardia. The hemodynamics was restored as the oxygen capacity of the blood decreased with an increase in the sympathetic tonus and development of tachycardia. The role of intracardial metasympathetic structures and the renin-angiotensin system in cardiovascular adaptation to hypoxia was established. Adaptation to nitrite hypoxia is accomplished by a coordinated interaction of neurogenic and humoral factors. A combination of pharmacological agents, which include separate links of regulator systems of the organism, leads to failure of the adaptation process. 相似文献

Diseases featuring abnormally low alveolar PO2 are frequently accompanied by systemic effects. The common presence of an underlying inflammatory component suggests that inflammation may contribute to the pathogenesis of the systemic effects of alveolar hypoxia. While the role of alveolar macrophages in the immune and defense functions of the lung has been long known, recent evidence indicates that activation of alveolar macrophages causes inflammatory disturbances in the systemic microcirculation. The purpose of this review is to describe observations in experimental animals showing that alveolar macrophages initiate a systemic inflammatory response to alveolar hypoxia. Evidence obtained in intact animals and in primary cell cultures indicate that alveolar macrophages activated by hypoxia release a mediator(s) into the circulation. This mediator activates perivascular mast cells and initiates a widespread systemic inflammation. The inflammatory cascade includes activation of the local renin-angiotensin system and results in increased leukocyte-endothelial interactions in post-capillary venules, increased microvascular levels of reactive O2 species; and extravasation of albumin. Given the known extrapulmonary responses elicited by activation of alveolar macrophages, this novel phenomenon could contribute to some of the systemic effects of conditions featuring low alveolar PO2. 相似文献

The investigation was intended to study the role ofbaroreceptors ofhemodynamically isolated zone of vertebral arteries in regulation of peripheral veins tonus, arterial pressure and external respiration. Pressure decrease in this vascular reflexogenic zone led to reflex responses of increase in femoral vein tonus, elevation of blood pressure level and stimulation of external respiration. The opposite reflex responses of cardio-respiratory functional system to initial pressure activation of vertebral arteries baroreceptors are observed. Basing on generalization of our own findings and similar physiological and morphological researches of other authors, it is established that afferentation from the vertebral artery zone is a reflexogenic factor of somatic muscles' veins tonus regulation. These reflexes of capacity vessels tonic activity changes are part of cardio-respiratory responses of maintaining the tissue gaseous exchange. 相似文献
