A new method has been developed for the continuous measurement of aortic blood velocity in patients following cardiac surgery. Using an extractable Doppler ultrasound probe placed on the ascending aorta, the changes in aortic velocity were recorded up to 24 h postoperatively, in 14 patients undergoing coronary bypass surgery. Volume flow rate is calculated from the mean velocity, the diameter of the aorta and the angle between the ultrasound beam and the direction of the blood flow, by means of an analogue flow calculator. Estimation of aortic flow showed a correlation of r = 0.79 with cardiac output measured by a thermodilution technique. The main advantage of the system is that it allows continuous monitoring of cardiac output, as well as short and long-term trend analyses, during the early postoperative period.  相似文献

Sixteen tooth characters were analyzed in 277 specimens of spinner dolphins ( Stenella longirostris ) from the eastern tropical Pacific to determine whether the characters were associated with size of the animal, species-stock membership or geographic location. The tooth characters included 5 continuous measurements and 11 qualitative variables. Statistical analyses of the characters included two-way ANOVA tests which assessed continuous measurement variables for differences based on stock membership, latitude and longitude of collection or whether the animal was collected north or south of the equator. Principal component analyses combined factors of continuous variables with those of qualitative variables into components which were used as variables in further analyses. Discriminant analyses were performed using continuous variables alone and using principal components as potentially discriminating variables. Results showed that tooth differences were not associated with size but significant differences were found in some tooth variables with stock membership, incremental changes in latitude and whether the animal was collected north or south of the equator. A discriminant function, containing only tooth length, correctly classified 67% of northern and 78% of southern animals according to location of collection north or south of the equator.  相似文献

Genetic diversity and population structure were investigated across the core range of Tasmanian devils (Sarcophilus laniarius; Dasyuridae), a wide-ranging marsupial carnivore restricted to the island of Tasmania. Heterozygosity (0.386-0.467) and allelic diversity (2.7-3.3) were low in all subpopulations and allelic size ranges were small and almost continuous, consistent with a founder effect. Island effects and repeated periods of low population density may also have contributed to the low variation. Within continuous habitat, gene flow appears extensive up to 50 km (high assignment rates to source or close neighbour populations; nonsignificant values of pairwise FST), in agreement with movement data. At larger scales (150-250 km), gene flow is reduced (significant pairwise FST) but there is no evidence for isolation by distance. The most substantial genetic structuring was observed for comparisons spanning unsuitable habitat, implying limited dispersal of devils between the well-connected, eastern populations and a smaller northwestern population. The genetic distinctiveness of the northwestern population was reflected in all analyses: unique alleles; multivariate analyses of gene frequency (multidimensional scaling, minimum spanning tree, nearest neighbour); high self-assignment (95%); two distinct populations for Tasmania were detected in isolation by distance and in Bayesian model-based clustering analyses. Marsupial carnivores appear to have stronger population subdivisions than their placental counterparts.  相似文献

Sensory systems extract behaviorally relevant information from a continuous stream of complex high-dimensional input signals. Understanding the detailed dynamics and precise neural code, even of a single neuron, is therefore a non-trivial task. Automated closed-loop approaches that integrate data analysis in the experimental design ease the investigation of sensory systems in three directions: First, adaptive sampling speeds up the data acquisition and thus increases the yield of an experiment. Second, model-driven stimulus exploration improves the quality of experimental data needed to discriminate between alternative hypotheses. Third, information-theoretic data analyses open up novel ways to search for those stimuli that are most efficient in driving a given neuron in terms of its firing rate or coding quality. Examples from different sensory systems show that, in all three directions, substantial progress can be achieved once rapid online data analysis, adaptive sampling, and computational modeling are tightly integrated into experiments.  相似文献

Cultivated for centuries, the varieties of rose have been selected based on a number of flower traits. Understanding the genetic and molecular basis that contributes to these traits will impact on future improvements for this economically important ornamental plant. In this study, we used scanning electron microscopy and sections of meristems and flowers to establish a precise morphological calendar from early rose flower development stages to senescing flowers. Global gene expression was investigated from floral meristem initiation up to flower senescence in three rose genotypes exhibiting contrasted floral traits including continuous versus once flowering and simple versus double flower architecture, using a newly developed Affymetrix microarray (Rosa1_Affyarray) tool containing sequences representing 4765 unigenes expressed during flower development. Data analyses permitted the identification of genes associated with floral transition, floral organs initiation up to flower senescence. Quantitative real time PCR analyses validated the mRNA accumulation changes observed in microarray hybridizations for a selection of 24 genes expressed at either high or low levels. Our data describe the early flower development stages in Rosa sp, the production of a rose microarray and demonstrate its usefulness and reliability to study gene expression during extensive development phases, from the vegetative meristem to the senescent flower.  相似文献

The biological rhythms of microalgal growth within a hydraulically integrated serial turbidostat algal reactor (HISTAR) were examined after comparison of a simple mechanistic productivity model with actual data yielded a standard error of prediction (SEP) of 62%. The data used for this study were taken on cultures of Selenastrum capricornutum grown under continuous 400-watt metal halide lighting. Fourier series analysis (up to five harmonics) was used to model the biorhythms and differentiate them from stochastic processes. Regression analyses revealed that the best Fourier series fit for the data was a three harmonic summation. Regression analyses on additional harmonic summations did not increase r 2 by more than 1%. The three harmonics were summed and incorporated into the growth term of the simple model, and the resultant full model was calibrated. The mechanistic HISTAR productivity model was greatly enhanced by the addition of the biological rhythm component, resulting in a SEP of <24.8 %. The period of the first harmonic was 13.4 days, which is very close to a circasemilunar rhythm (14.8 days). In summary, the predictive power of productivity models for continuous microalgal cultures can be dramatically improved with the inclusion of a biorhythm analysis.  相似文献

Aim We surveyed mitochondrial DNA (mtDNA) sequence variation among regionally isolated populations of 10 grassland‐associated butterfly species to determine: (1) the utility of phylogeographic comparisons among multiple species for assessing recent evolutionary patterns, and (2) the respective roles of isolation attributable to range disjunction versus isolation attributable solely to geographic distance in establishing divergence patterns. Location The Peace River grasslands of northern Alberta and British Columbia, Canada, which are isolated by 300+ km from similar communities to the south. Methods We sequenced mtDNA (1420 bp of cytochrome c oxidase subunit I) from five grassland‐restricted butterfly species that have geographically disjunct populations and from five ecologically broader species that have more continuous distributions across the same regions. Using analysis of molecular variance (AMOVA), Mantel and partial Mantel tests, and haplotype networks, we compared population structure within and between species in order to assess the validity of single‐species phylogeographic characterizations. We then contrasted variance components between disjunct and continuously distributed species to assess whether divergences were correlated more with disjunction or with geographic distance. Results Single‐species analyses varied substantially within both the disjunct and the continuous groups. One species in each of these groups had mtDNA with unusually deep intraspecific mitochondrial lineage divergences. On the whole, however, the five species with disjunct ranges exhibited greater divergence between geographically distant populations than did the five species with continuous distributions. Comparison of variance components between disjunct and continuous species indicated that isolation attributable only to geographic distance was responsible for up to half of the total sequence variation between disjunct populations of grassland butterflies. Main conclusions Our findings show that single‐species phylogeographic analyses of post‐Pleistocene butterfly distributions are inadequate for characterizing regional biogeographic divergence histories. However, comparison of mtDNA sequence divergences between groups of disjunct and continuously distributed species can allow isolation attributable to range interruption to be quantitatively distinguished from isolation attributable solely to gene flow attenuation over the same geographic area.  相似文献

An anaerobic down-flow fluidized bed reactor was inoculated with granular sludge and started-up with sulfate containing synthetic wastewater to promote the formation of a biofilm enriched in sulfate-reducing bacteria (SRB), to produce biogenic sulfide. The start-up was done in two stages operating the reactor in batch for 45 days followed by 85 days of continuous operation. Low-density polyethylene was used as support. The biofilm formation was followed up by biochemical and electron microscopy analyses and the composition of the community was examined by 16S rDNA sequence analysis. Maximum immobilized volatile solids (1.2 g IVS/Lsupport) were obtained after 14 days in batch regime. During the 85 days of continuous operation, the reactor removed up to 80% of chemical oxygen demand (COD), up to 28% of the supplied sulfate and acetate was present in the effluent. Sulfate-reducing activity determined in the biofilm with ethanol or lactate as substrate was 11.7 and 15.3 g COD/g IVS per day, respectively. These results suggested the immobilization of sulfate reducers that incompletely oxidize the substrate to acetate; the phylogenetic analysis of the cloned 16S rDNA gene sequences showed high identity to the genus Desulfovibrio that oxidizes the substrates incompletely. In contrast, in the granular sludge used as inoculum a considerable number of clones showed homology to Methanobacterium and just few clones were close to SRB. The starting-up approach allowed the enrichment of SRB within the diverse community developed over the polyethylene support.  相似文献

The development of continuous and stepwise automatic dilution and calibration systems that enable analyte dilution and enhancement in multicomponent analyses are described. The continuous system has been used with a robot and the stepwise configuration integrated into a flow-injection analysis system. All analyses are performed using a UV/Visible diode-array detector and data analysis performed using principal components regression. Results of both systems are presented for the simultaneous determination of caffeine, aspirin and paracetamol in pharmaceutical formulations.  相似文献

The ferrous iron oxidation kinetics of Thiobacillus ferrooxidans in batch cultures was examined, using on-line off-gas analyses to measure the oxygen and carbon dioxide consumption rates continuously. A cell suspension from continuous cultures at steady state was used as the inoculum. It was observed that a dynamic phase occurred in the initial phase of the experiment. In this phase the bacterial ferrous iron oxidation and growth were uncoupled. After about 16 h the bacteria were adapted and achieved a pseudo-steady state, in which the specific growth rate and oxygen consumption rate were coupled and their relationship was described by the Pirt equation. In pseudo-steady state, the growth and oxidation kinetics were accurately described by the rate equation for competitive product inhibition. Bacterial substrate consumption is regarded as the primary process, which is described by the equation for competitive product inhibition. Subsequently the kinetic equation for the specific growth rate, μ, is derived by applying the Pirt equation for bacterial substrate consumption and growth. The maximum specific growth rate, μ max, measured in the batch culture agrees with the dilution rate at which washout occurs in continuous cultures. The maximum oxygen consumption rate, q O2,max, of the cell suspension in the batch culture was determined by respiration measurements in a biological oxygen monitor at excess ferrous iron, and showed changes of up to 20% during the course of the experiment. The kinetic constants determined in the batch culture slightly differ from those in continuous cultures, such that, at equal ferric to ferrous iron concentration ratios, biomass-specific rates are up to 1.3 times higher in continuous cultures. Peanut is an important oil crop worldwide and shows considerable adaptability but growth and yield are negatively affected by continuous cropping. Soil micro-organisms are efficient bio-indicators of soil quality and plant health and are critical to the sustainability of soil-based ecosystem function and to successful plant growth. In this study, 18S rRNA gene clone library analyses were employed to study the succession progress of soil eukaryotic micro-organisms under continuous peanut cultivation. Eight libraries were constructed for peanut over three continuous cropping cycles and its representative growth stages. Cluster analyses indicated that soil micro-eukaryotic assemblages obtained from the same peanut cropping cycle were similar, regardless of growth period. Six eukaryotic groups were found and fungi predominated in all libraries. The fungal populations showed significant dynamic change and overall diversity increased over time under continuous peanut cropping. The abundance and/or diversity of clones affiliated with Eurotiales, Hypocreales, Glomerales, Orbiliales, Mucorales and Tremellales showed an increasing trend with continuous cropping but clones affiliated with Agaricales, Cantharellales, Pezizales and Pyxidiophorales decreased in abundance and/or diversity over time. The current data, along with data from previous studies, demonstrated that the soil microbial community was affected by continuous cropping, in particular, the pathogenic and beneficial fungi that were positively selected over time, which is commonplace in agro-ecosystems. The trend towards an increase in fungal pathogens and simplification of the beneficial fungal community could be important factors contributing to the decline in peanut growth and yield over many years of continuous cropping.  相似文献

Highly purified human granulocytes isolated from continuous flow centrifugation leukapheresis concentrates by counterflow centrifugation-elutriation were stored at 4 °C in concentrations of 6 × 106 to 1 × 107 granulocytes per milliliter for up to 14 days. The in vitro physiological function assays of phagocytosis, oxygen consumption associated with phagocytosis, bacterial growth inhibition, chemotaxis, and five enzyme analyses indicated good storage survival for up to 4 days. Stored granulocytes separated from other blood cells have greater storage stability than granulocytes stored as leukapheresis concentrates. After 14 days of storage a small percentage of granulocytes still maintained all physiological functions, with the exception of chemotaxis. Of the five enzymes assayed, only the enzyme activity of leucine aminopeptidase decreased significantly by the 14th day of storage. The storage stability of each physiological function assayed decreased as follows: bacterial growth inhibition (most stable), phagocytosis, oxygen consumption, and chemotaxis (least stable).  相似文献

Utilization of solvent tolerant bacteria as biocatalysts has been suggested to enable or improve bioprocesses for the production of toxic compounds. Here, we studied the relevance of solvent (product) tolerance and inhibition, carbon metabolism, and the stability of biocatalytic activity in such a bioprocess. Styrene degrading Pseudomonas sp. strain VLB120 is shown to be solvent tolerant and was engineered to produce enantiopure (S)-styrene oxide from styrene. Whereas glucose as sole source for carbon and energy allowed efficient styrene epoxidation at rates up to 97 micromol/min/(g cell dry weight), citrate was found to repress epoxidation by the engineered Pseudomonas sp. strain VLB120DeltaC emphasizing that carbon source selection and control is critical. In comparison to recombinant Escherichia coli, the VLB120DeltaC-strain tolerated higher toxic product levels but showed less stable activities during fed-batch cultivation in a two-liquid phase system. Epoxidation activities of the VLB120DeltaC-strain decreased at product concentrations above 130 mM in the organic phase. During continuous two-liquid phase cultivations at organic-phase product concentrations of up to 85 mM, the VLB120DeltaC-strain showed stable activities and, as compared to recombinant E. coli, a more efficient glucose metabolism resulting in a 22% higher volumetric productivity. Kinetic analyses indicated that activities were limited by the styrene concentration and not by other factors such as NADH availability or catabolite repression. In conclusion, the stability of activity of the solvent tolerant VLB120DeltaC-strain can be considered critical at elevated toxic product levels, whereas the efficient carbon and energy metabolism of this Pseudomonas strain augurs well for productive continuous processing.  相似文献

Extracts of three continuous cell lines from the cabbage looper, Trichoplusia ni, were assayed for the presence of ecdysteroids. While no evidence of ecdysteroids was present in the extracts of the ovarian (TN-368) or embryonic (IPLB-TN-R2) cell lines, radioimmunoassays on extracts of media and extracts of cell pellets from imaginal disc cell cultures (IAL-TND1) were positive. The immunoreactive material from both cells and media co-migrated with a 20-hydroxyecdysone standard on reversed-phase high-performance liquid chromatography (HPLC). The immunoreactive fractions from the cell extract were chromatographed on silica HPLC and subjected to mass spectral analysis. Both of these analyses indicated that the unknown compound was 20-hydroxyecdysone. Radioimmunoassay indicated up to 28 ng of ecdysone equivalents in cells (3.75 x 107 cells) from 50 ml of IAL-TND1 cultures, which is equivalent to 120 ng of 20-hydroxyecdysone based on relative reactivity of the antiserum used in this study. This report presents the first evidence of 20-hydroxyecdysone production by a continuous insect cell line and also the first to show that cells from imaginal discs are capable of ecdysteroid synthesis.  相似文献

Abstract. To identify management treatments suitable for the conservation of extensively managed grasslands, the ‘Fallow experiments in Baden‐Württemberg’ were set up in 1975. In this investigation, species composition of the grazing, mowing, mulching, controlled burning and unmanaged (succession) treatments were analysed after 25 yr of continuous management in Arrhenatherum elatius and Bromus erectus grasslands. Through ordination analyses it was found that species composition is strongly dependent on the management treatment. The first axis, identified by ordination analysis, essentially corresponded to a gradient of decreasing disturbance frequency. Controlled burning resulted in a unique species composition. Grazing, mowing and mulching twice a year were found to be most suitable for the conservation of unimproved, species‐rich grasslands.  相似文献

Displacement chromatography is a predominantly nonlinear mode of chromatography, which has certain advantages over the elution mode for preparative bioseparations. Whereas continuous production (and separation) processes have their theoretical benefits in this context, protein displacement chromatography has up to now only been performed in the batch mode. In this contribution, we demonstrate that the principle of continuous annular chromatography can be adapted to displacement chromatography. Separations of up to three standard proteins (two whey proteins, soybean trypsin inhibitor) were developed and optimized using a small (4 x 250 mm) batch column. These separations were subsequently transferred directly to the continuous system (500-mL column). Separations of similar quality in terms of final product purity and recovery yield were obtained using the continuous system.  相似文献

Quantitative and continuous characters have rarely been included in cladistic analyses of morphological data; when included, they have always been discretized, using a variety of ad hoc methods. As continuous characters are typically additive, they can be optimized with well known algorithms, so that with a proper implementation they could be easily analyzed without discretization. The program TNT has recently incorporated algorithms for analysis of continuous characters. One of the problems that has been pointed out with existing methods for discretization is that they can attribute different states to terminals that do not differ significantly—or vice versa. With the implementation in TNT, this problem is diminished (or avoided entirely) by simply assigning to each terminal a range that goes from the mean minus one (or two) SE to the mean plus one (or two) SE; given normal distributions, terminals that do not overlap thus differ significantly (more significantly if using more than 1 SE). Three real data sets (for scorpions, spiders and lizards) comprising both discrete and quantitative characters are analyzed to study the performance of continuous characters. One of the matrices has a reduced number of continuous characters, and thus continuous characters analyzed by themselves produce only poorly resolved trees; the support for many of the groups supported by the discrete characters alone, however, is increased when the continuous characters are added to the analysis. The other two matrices have larger numbers of continuous characters, so that the results of separate analyses for the discrete and the continuous characters can be more meaningfully compared. In both cases, the continuous characters (analyzed alone) result in trees that are relatively similar to the trees produced by the discrete characters alone. These results suggest that continuous characters carry indeed phylogenetic information, and that (if they have been observed) there is no real reason to exclude them from the analysis. © The Willi Hennig Society 2006.  相似文献
