Both autonomic nerve activity and electrical remodeling are important in atrial arrhythmogenesis. Therefore, dogs with sympathetic hyperinnervation, myocardial infarction (MI), and complete atrioventricular block (CAVB) may have a high incidence of atrial arrhythmias. We studied eight dogs (experimental group) with MI, CAVB, and sympathetic hyperinnervation induced either by nerve growth factor infusion (n = 4 dogs) or subthreshold electrical stimulation (n = 4 dogs) of the left stellate ganglion. Cardiac rhythm was continuously monitored by a Data Sciences International transmitter for 48 (SD 27) days. Three normal control dogs were also monitored. Six additional normal dogs were used for histology control. Paroxysmal atrial fibrillation (PAF) and paroxysmal atrial tachycardia (PAT) were documented in all dogs in the experimental group, with an average of 3.8 (SD 3) episodes/day, including 1.3 (SD 1.6) episodes of PAF and 2.5 (SD 2.2) episodes of PAT. The duration averaged 298 (SD 745) s (range, 7-4,000 s). There was a circadian pattern of arrhythmia onset (P < 0.01). Of 576 episodes of PAF and PAT, 236 (41%) episodes occurred during either sustained or nonsustained ventricular tachycardia (VT). Among these 236 episodes, 53% started before VT, whereas 47% started after the onset of VT. Normal dogs did not have either PAF or PAT. The hearts from the experimental group had a higher density of nerve structures immunopositive (P < 0.01) for three different nerve specific markers in both right and left atria than those of the control dogs. We conclude that the induction of nerve sprouting and sympathetic hyperinnervation in dogs with CAVB and MI creates a high yield model of PAF and PAT.  相似文献

Intracranial hypotension (IH) is a pathophysiological condition of reduced intracranial pressure caused by low cerebrospinal fluid (CSF) volume due to dural injuries from lumbar puncture, surgery, or trauma. Understanding the prognosis of IH in small animal models is important to gain insights on the complications associated with it such as orthostatic headache, cerebral venous thrombosis, coma, and so forth. Photoacoustic tomography (PAT) offers a novel and cost‐effective way to perceive and detect IH in small animal models. In this study, a pulsed laser diode (PLD)‐based PAT imaging system was used to examine the changes in the venous sinuses of the rat brain due to IH, induced through CSF extraction. After the CSF extraction, an increase in the sagittal sinus area by ~30% and width by 40% ± 5% was observed. These results provide supportive evidence that the PLD‐PAT can be employed for detecting changes in sagittal sinus due to IH in rat model.  相似文献

An immunoassay method was developed to quantitatively detect phosphinothricin-N-acetyltransferase (PAT) encoded by the Bialaphos resistance (bar) gene in genetically modified (GM) pepper. The histidine-tagged PAT was overexpressed in Escherichia coli M15 (pQE31-bar) and efficiently purified by Ni2+ affinity chromatography. A developed sandwich enzyme-linked immunosorbent assay (S-ELISA) method (detection limit: 0.01 microg/ml) was 100-fold more sensitive than a competitive indirect ELISA (CI-ELISA) method or Western blot analysis in detecting the recombinant PAT. In real sample tests, PAT in genetically modified herbicide-tolerant (GMHT) peppers was successfully quantified [4.9 +/- 0.4 microg/g of sample (n = 6)] by the S-ELISA method. The S-ELISA method developed here could be applied to other GMHT crops and vegetables producing PAT.  相似文献

689 long-term electroencephalographic recordings from 88 rats with chronically implanted electrodes were evaluated with reference to the onset and development of rhythmic spike episodes of 8-9 Hz frequency characteristic of quiet wakefulness. Episodes occurred in the recordings of 88% of the test animals. Recordings completely devoid of episodes came from selected rats highly sensitive to an acoustic epileptogenic stimulus and from 50% of rats of a Wistar-Velaz barrier breed. These two groups differed from the other animals in respect of their greater aggressiveness and restlessness in the chronic experiment. The episodes were not evoked by the surgical procedure--implantation of a cobalt-gelatin pin in the brain--or to the actual implantation of chronic electrodes in the brain. The shortest recorded latent period for the appearance of episodes was one week after the implantation of electrodes and the longest was 6 months. The animals' mean age at the time of the onset of episodes was 3.8 months and the lowest age was 2.5 months (i.e. immediately after implantation of the electrodes). The electrodes were not implanted before the age of 2.5 months. In repeated chronic recordings during the animals' life, both the number and the duration of the episodes increased. In most of the rats, the spike frequency in the episode was 8-9 Hz and was maintained at a stable value characteristic for each rat. Episodic rhythmic activity can thus occur in a state of quiet wakefulness in the electroencephalogram of all the given groups of rats (except highly sensitive animals) from early adulthood up to the age regarded in the rat as senility. The results to date indicate that the main condition for its appearance is the attainment of an optimum state of vigilance--quiet immobile wakefulness. The mechanisms leading to this stage of vigilance display both individual and group differences and are evidently related to the ability to become habituated to the experimental environment.  相似文献

An acoustic neural telemetry tag has been developed for recording from free-swimming aquatic animals. Microwire electrodes were implanted into the VIIIth nerve of the toadfish, Opsanus tau, and interfaced to the subdermally implanted tag. The telemetry tag frequency modulates the neural signal, converting it into a varying frequency, which is amplified and transmitted acoustically (centre frequency of 90 kHz and a 20 kHz bandwidth). This acoustic signal is detected by a receiver hydrophone, and the receiver reconstructs the full neural waveform from the acoustic signal. However, due to the multipath environment in the experimental aquarium, the acoustic signal is quickly degraded as the hydrophone is moved away from the source. In order to receive the signal independent of fish position, an anechoic aquarium was designed. Streams of microbubbles (ca. 70 microm diameter) were generated to produce a curtain of sound-absorptive material along the walls and water surface of the aquarium. Microbubble generation significantly reduced the multipath artefacts, and allowed signal discrimination independent of fish and hydrophone position. The anechoic aquarium will allow the recording of neural activity from free-swimming fishes in quasi-natural habitats, thus allowing better understanding of the neural mechanisms of behaviour.  相似文献

A small intraperitoneal radio transmitter was developed and used successfully in California sea otters. The transmitter weighs about 120 g and has an internal antenna. Thirty-five implanted transmitters were located for an average of 526 d. The carcasses of five animals that died were recovered; there were no complications associated with the transmitters in these individuals. Reproductive performance of the adult females with implanted transmitters appeared normal. The main advantages of this transmitter are its reliability and long life. It has enabled the collection of significant new information on California sea otters.  相似文献

Metabolic rate, heart rate and respiratory rate of three adult moose were monitored simultaneously on selected days between January 1982 and October 1983. Heart rate was successfully monitored using two types of implanted radio transmitters. Metabolic rate was significantly (P less than 0.001) correlated with heart rate for individual animals (r = 0.94-0.98). The relationship was best described by an exponential equation. A mean weighted equation predicted metabolic rate within 9% of individual equations. In adult moose cows, heart rate was a more reliable predictor of energy expenditure than respiratory rate.  相似文献

Variations of brain tissue redox state potential (E) of freely-moving white rats (300-350 g) in cycles of wakefulness (W), slow-wave sleep (SWS), and paradoxical sleep (PS) were measured by platinum electrodes symmetrically implanted into the frontal and occipital cortices and hippocampus. In addition, EMG of neck muscles and general motor activity of animals were recorded. The common reference electrode was implanted in the nasal bone. It was shown that in some brain sites (called active), episodes of W and PS were accompanied by a rise of E, and during transitions from W and PS to SWS, E dropped. The value of E varied in the range of 100 mV. It is suggested that transitions from W and PS to SWS are accompanied by shifts in the balance between the main energy sources. Oxidative phosphorylation prevails in W and PS, whereas aerobic glycolysis is the main source of energy during SWS. We think that this suggestion is supported both by a decrease in E in SWS and its oscillations typical of glucolytic processes [Aon et al., 1992]. Recent literature data [Bitter et al., 1996] suggest that astroglia is the main compartment for aerobic glycolysis.  相似文献

Tangential flow microfiltration (MF) is a cost‐effective and robust bioprocess separation technique, but successful full scale implementation is hindered by the empirical, trial‐and‐error nature of scale‐up. We present an integrated approach leveraging at‐line process analytical technology (PAT) and mass balance based modeling to de‐risk MF scale‐up. Chromatography‐based PAT was employed to improve the consistency of an MF step that had been a bottleneck in the process used to manufacture a therapeutic protein. A 10‐min reverse phase ultra high performance liquid chromatography (RP‐UPLC) assay was developed to provide at‐line monitoring of protein concentration. The method was successfully validated and method performance was comparable to previously validated methods. The PAT tool revealed areas of divergence from a mass balance‐based model, highlighting specific opportunities for process improvement. Adjustment of appropriate process controls led to improved operability and significantly increased yield, providing a successful example of PAT deployment in the downstream purification of a therapeutic protein. The general approach presented here should be broadly applicable to reduce risk during scale‐up of filtration processes and should be suitable for feed‐forward and feed‐back process control. © 2015 American Institute of Chemical Engineers Biotechnol. 目的:探讨左室主动固定电极在心脏再同步化治疗(cardiac resynchronization therapy,CRT)中的作用并总结其临床经验。方法:收集2008年1月至2013年1月由梅州I市人民医院心内科植入CRT的患者,共56例(男34例,女22例)。根据植入冠状窦电极的不同,分为左室主动固定电极组(17例)和被动电极组(39例),观察和比较两组的手术时间、透视时间和造影剂用量。结果:左室主动固定电极组的手术时间[(61±36)minvs(143+61)min,P=0.035]和透视时间[(10±5)minvs(45+11)min,P=0.042]均较被动电极组显著缩短,而造影剂用量[(36±20)mlvs(87±46)ml,P=0.041]也较被动电极组明显减少。结论:左室主动固定电极用于心脏再同步化治疗可明显缩短手术时间,降低造影剂用量,使患者和术者均获益。  相似文献

EEG activity was recorded from rats with chronic cobalt foci and implanted electrodes. Within this activity, hypersynchronous rhythmic spike episodes (and those of waves) of mean frequency of 8-9/sec were studied. The spike amplitude-interval relationship was assessed, using correlation analysis method. A positive correlation of an exponential nature was found to be growing in line with temporal development of episode.  相似文献

The author studied the effect of diazepam in doses of 1 and 3 mg/kg on rats with a chronic cortical cobalt-gelatin focus and implanted cortical and subcortical electrodes. Focal spike activity localized at the site of the focus and hypersynchronous generalized episodes of spikes (and waves) of 8--9/sec frequency were studied in the electroencephalogram and the main phases of vigilance (waking, telencephalic slow waves/SWS/and REM sleep) after diazepam were evaluated. The effect of diazepam on rats temporarily immobilized with tubocurarine was also evaluated. 1. Focal spike activity during sleep was mildly inhibited by diazepam. If present in the waking state, it was markedly inhibited. 2. The number of episodes diminished significantly after diazepam. The maximum decrease occurred 30--45 minutes after administering diazepam and after that they slowly recovered. 3. Diazepam did not inhibit alteration of the phases of vigilance, but there was an increase in the proportion of telencephalic sleep with large numbers of spidles of 12--14/sec frequency and the incidence of REM phases rose by 250--300%. 4. Diazepam brought no renewal of the episodes which disappeared from the waking EEG recording of rats with a chronic focus temporarily immobilized with tubocurarine. Its administration was followed mostly by sleep activity with spindles. 5. Despite certain effects in common (disappearance of episodes), the action of diazepam differs from that of barbiturates in many respects and is effected by different mechanisms.  相似文献

The structural instability exhibited by IncP-1 plasmids in Pseudomonas aeruginosa strain PAT was shown to be Rec+ dependent and involved interaction with the resident plasmid pVS1. Structural instability resulted from deletion of plasmid deoxyribonucleic acid at a frequency of ca. 10(-2)/cell per generation. Deletants could be stabilized by transduction into P. aeruginosa strain PAO, but in strain PAT deletants had only a transient existence, as continued deletion led eventually to the loss of the entire plasmid. The patterns of markers lost in PAT were used to demonstrate a marker order for R68 similar to that published elsewhere for RP4 (Barth and Grinter, J. Mol. Biol. 113:455-474, 1977), except that only one Tra region was found. R68 also exhibited Rec+-dependent structural instability in PAO(pVS1) derivatives but, unlike the case in PAT, instability was not accompanied by chromosome mobilization. We isolated deletants of pVS1 which were unable to promote structural instability.  相似文献

The involvement of soluble growth-promoting factors in stretch-induced hypertrophy of the Patagialis muscle (PAT) in the chicken wing was investigated. Soluble extracts were prepared from young chicken PAT muscles made hypertrophic by passive stretch and from unstretched contralateral controls. Extracts were tested for their ability to stimulate cell proliferation and creatine phosphokinase (CPK) activity in primary monolayer cultures of chick embryo muscle cells. Factors were present in muscle extracts which showed a dose-dependent stimulation of cell proliferation and CPK activity in vitro. Passive stretch for 5 days produced a rapid hypertrophy of the PAT which was accompanied by a dramatic increase in the activity of the growth factor(s). Release of stretch resulted in an arrest of growth and an immediate fall in growth factor activity. The difference in growth-stimulating activity between control and stretched PAT extracts could be demonstrated in chicken transferrin-sensitive chick myoblast cultures. Stretch thus induces an increase in a class-specific growth factor, possibly Transferrin, in the PAT. Stretched PAT extracts stimulated: (a) chick myoblast proliferation to a greater extent than an optimum concentration of chick embryo extract, and (b) CPK activity in vitro to a greater extent than excess Transferrin. Both control and stretched PAT extracts supported the growth of rat myoblasts. We conclude that PAT muscle extracts also contain unknown growth factor(s) which are different from Transferrin.  相似文献

目的经颈静脉途径应用心室起搏的方法制备心脏记忆犬模型。方法 8只普通级成年健康Beagle犬经腹腔麻醉后,Seldinger’s法穿刺颈外静脉成功后送入心内膜起搏电极,将电极头端固定于右室心尖部,近端连接于脉冲发生器。起搏频率设置较犬窦性心律时的基础心率快15%,保证起搏器连续起搏。结果连续起搏1周后所有动物均成功制备为心脏记忆模型。建模后犬的心率、呼吸、体重与建模前比较,无明显改变;所有模型组犬起搏前心电图均为窦性心律,起搏1周后出现心脏T波记忆,在下壁导联以及胸前导联均出现T波倒置,停止起搏后,心脏T波记忆逐渐消失;模型组犬与正常组犬心肌病理相比,无明显改变。结论经颈静脉途径应用心室起搏法建立心脏记忆犬模型的方法,具有手术简单,创伤小,诱发方式与临床相似等优点,为深入展开心脏记忆的机制研究奠定基础。  相似文献

Patulin (PAT) is a fungal secondary metabolite commonly present in apples and apple products. In the present study, PAT was evaluated for its genotoxic, cytotoxic and cytostatic effects to human peripheral blood lymphocytes by using the cytokinesis-block micronucleus cytome (CBMN Cyt) assay. Lymphocyte cultures were treated with PAT at the following concentrations, 0.1, 0.3, 0.5, 1.0, 2.5, 5.0, and 7.5 μM, as well as 0.5 μM mitomycin c (MMC) as a positive control and dimethyl sulfoxide (DMSO) as a vehicle control. PAT was found to induce nucleoplasmic bridges (NPBs) at 5.0 and 7.5 μM concentrations (P?<?0.05), apoptotic cells at 0.1, 1.0, 5.0 μM (P?<?0.05), 7.5 μM concentrations (P?<?0.01) and necrotic cells at 0.3 and 2.5 μM (P?<?0.05), 5.0 and 7.5 μM (P?<?0.01) concentrations in human lymphocytes. The 2.5, 5.0, and 7.5 μM PAT concentrations also led to a clear decrease in the nuclear division index (NDI) (P?<?0.05). PAT caused a significant dose-dependent increase in the number cells of NPBs, in the frequency of apoptotic and necrotic cells, and a significant dose-dependent decrease in the NDI values in lymphocytes. These results indicate that PAT at high concentrations is genotoxic, cytotoxic and cytostatic in cultured human lymphocytes.  相似文献

Reabsorption of amino acids is an important function of the renal proximal tubule. pH-dependent amino acid transport has been measured previously using rabbit renal brush-border membrane vesicles (BBMV). The purpose of this investigation was to determine whether this pH-dependent uptake represents H(+)/amino acid cotransport via a PAT1-like transport system. The rabbit PAT1 cDNA was isolated (2296bp including both 5' and 3' untranslated regions and poly(A) tail) and the open reading frame codes for a protein of 475 amino acids (92% identity to human PAT1). Rabbit PAT1 mRNA was found in all tissues investigated including kidney. When expressed heterologously in a mammalian cell line, rabbit PAT1 mediates pH-dependent, Na(+)-independent uptake of proline, glycine, l-alanine and alpha-(methylamino)isobutyric acid. Proline uptake was maximal at pH 5.0 (K(m) 2.2+/-0.7 mM). A transport system with identical characteristics (ion dependency, substrate specificity) was detected in rabbit renal BBMV where an overshoot was observed in the absence of Na+ but in the presence of an inwardly directed H+ gradient. In the presence of Na+ and under conditions in which PAT1 transport function was suppressed, a second proline uptake system was detected that exhibited functional characteristics similar to those of the IMINO system. The functional characteristics of rabbit PAT1 in either mammalian cells or renal BBMV suggest that PAT1 is the low-affinity transporter of proline, glycine and hydroxyproline believed to be defective in patients with iminoglycinuria.  相似文献
