We determined nonlethal physiological effects of hydroprene on the German cockroach, Blattella germanica L., and the effects of hydroprene exposure on German cockroach susceptibility to traditional insecticides. Live and dry weights of German cockroach adults exposed as nymphs to the juvenile hormone analog hydroprene increased significantly (6-35%) in both sexes. The change in weight was affected by hydroprene dose and time after adult emergence. Hydroprene treatment did not affect total body lipids and carbohydrates consistently. However, total body urates of males and females increased significantly. The susceptible and multiresistant German cockroach strains were significantly more susceptible to propoxur at the lower hydroprene application rate (0.697 mg per tub) than the untreated cockroaches. Hydroprene application also significantly lowered the LD50 for chlorpyrifos of the multiresistant strain at the lower hydroprene application rate. However, hydroprene failed to affect chlorpyrifos susceptibility of the susceptible strain.  相似文献

Formulations used for control of German cockroaches, Blattella germanica (L.), were investigated. The insecticides tested on gravid females were bendiocarb, chlorpyrifos, cyfluthrin, cypermethrin, fenvalerate, hydramethylnon, malathion, propetamphos, propoxur, and pyrethrins. Exposure to each insecticide increased the frequency of oothecal drop and reduced the percentage oothecal hatch. The percentage of oothecae that were dropped increased as a curvilinear function of insecticide concentration. An LD50 of propoxur to female German cockroaches resulted in the greatest oothecal drop (83.50%); fenvalerate caused the least drop (22.60%). LD50's of cypermethrin, propetamphos, and propoxur resulted in 29.60, 31.807, and 37.30% hatch from dropped oothecae, respectively. The smallest percentage hatch from retained oothecae was caused by LD50's of propoxur (1.5%) and cyfluthrin (7.70%). Retained oothecae from females treated with an LD50 of fenvalerate (68.70%) or pyrethrins (68.70%) had the greatest percentage hatch. Total percentage hatch (dropped and retained oothecae) declined exponentially as the insecticide concentration increased. An LD50 of cypermethrin limited total oothecal hatch to 24.50%, whereas LD50's of fenvalerate, malathion, and pyrethrins resulted in 53.60, 52.20, and 58.90% hatch, respectively. Mean time to oothecal hatch increased linearly with increasing insecticide concentration for all insecticides tested.  相似文献

We have identified resistance mechanisms in the German cockroach, Blattella germanica (L.), for propoxur and chlorpyrifos in strains of cockroaches that display multiresistance to several organophosphate and carbamate insecticides. The resistance mechanisms involve the combined effects of increased oxidative and hydrolytic metabolism and both strains are resistant to chlorpyrifos and propoxur. Experiments designed to test for similarity in metabolic enzymes suggest that, although the mechanisms involve similar processes, the enzymes responsible for insecticide detoxification are different in the two strains. Both resistant strains exhibited enhanced activity toward alpha-naphtholic esters relative to a standard susceptible strain; however, analysis of the progeny from resistant X susceptible crosses suggests that this general esterase activity is inherited differently than propoxur or chlorpyrifos resistance. Hybrids of the propoxur-resistant strain displayed the highest activity of all cockroaches tested, in contrast to hybrids of the chlorpyrifos-resistant strain, which were similar to the susceptible strain. Native gel electrophoresis of cytosolic preparations provided further evidence for differences in the pattern of hydrolytic enzymes and inheritance of resistance in the two strains. Analysis of components of the cytochrome P450-dependent monooxygenase system and activities toward model substrates indicate that the two resistance mechanisms also involve different oxidative processes. The propoxur-resistant strain displayed significantly higher levels of total cytochrome P450, but no other components were correlated with resistance. In contrast with the chlopyrifos-resistant strain, which was similar to the susceptible strain in all parameters measured, activity toward model substrates was higher in the propoxur-resistant strain than in any of the other strains and hybrids tested.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Three strains of German cockroach, Blattella germanica (L.) showed varying levels of resistance to chlorpyrifos, methyl parathion, propoxur, bendiocarb, and cypermethrin. The general esterase activity was at least twofold higher than susceptible strain. The subcellular distribution studies revealed that the majority of the esterase activity is present in the 100,000g cytosolic fraction. Only a small portion of the activity was membrane bound. Using non-denaturing gel electrophoresis, ten isozymes were identified in German cockroaches. These isozymes were isolated individually from the gels and analyzed for differences in activity. The isozymes E5, E6, and E7 of resistant strains had significantly higher specific activities when compared with the susceptible strain. The purification process using various column chromatography and preparative gel electrophoresis resulted in 9–11% of total esterase recovery. About double the amount of E6 was recovered from the resistant strains when compared with the susceptible strain. Kinetic analyses of E6 did not indicate differences in Km and Vmax values between the resistant and susceptible strains. Also, inhibition of esterase activity by paraoxon, chlorpyrifos, and propoxur did not suggest any structural differences in esterase E6 between strains. The results suggest that the increased production of E6 esterase contributes to insecticide resistance in German cockroaches. Toxicity of bendiocarb, chlorpyrifos, cyfluthrin, cypermethrin, fenvalerate, hydramethylnon, malathion, propetamphos, propoxur, and pyrethrins against the adult German cockroaches, Blattella germanica (L.), was investigated. At LD50, cyfluthrin was the most toxic insecticide to adult males (0.53 microgram/g), adult females (1.2 micrograms/g), and gravid females (0.85 microgram/g). Malathion was the least toxic insecticide to adult males (464.83 micrograms/g), adult females (335.83 micrograms/g), and gravid females (275.90 micrograms/g). Males and gravid females were generally more sensitive than nongravid females to the insecticides that we tested. In tests with malathion, however, males were more tolerant. The order of toxicity of the insecticide classes varied among the stages of adult German cockroaches. The order of toxicity for males and nongravid females was pyrethroids greater than pyrethrins = organophosphates (except malathion) greater than carbamates = amidinohydrazone. The order of toxicity for gravid females was pyrethroids greater than pyrethrins = organophosphates (except malathion) greater than carbamates greater than amidinohydrazone. These differences in toxicity suggest that sex differences should be considered when determining insecticide toxicity for German cockroaches.  相似文献

The effects of sublethal doses of deltamethrin and propoxur, applied topically at LD10, LD30 and LD50 on German cockroaches, were studied by reciprocal crossing. Male and female longevities decreased curvilinearly with increasing sublethal doses of deltamethrin, and decreased linearly with increasing sublethal doses of propoxur. Fecundity of females treated with deltamethrin and propoxur was reduced with increasing sublethal doses of both insecticides. Oothecal production, oothecal hatchability and nymphal production also declined with increasing doses of deltamethrin and propoxur. Preoviposition and incubation periods were not affected by sublethal doses of deltamethrin and propoxur, although some significant differences were observed at certain oothecal numbers. Two-way analysis of variance indicated that only treated females showed an effect on oothecal production while oothecal hatch and nymphal production were governed by both treated females and males. Insecticide susceptibility tests on the progeny of parents treated with sublethal doses demonstrated that these doses did not increase insecticidal tolerance in the F1 generation.  相似文献

Effects of the synergists piperonyl butoxide (PBO) and S,S,S-tributyl phosphorotrithioate (DEF) on propoxur pharmacokinetics were examined in the German cockroach, Blattella germanica (L.). Treatment of adult male German cockroaches with the cytochrome P450 monooxygenase inhibitor, PBO, or the esterase inhibitor, DEF, increased propoxur toxicity by 2- and 6.8-fold, respectively, implicating hydrolysis as a major detoxification route of propoxur in the German cockroach. However, significant hydrolytic metabolism could not be demonstrated conclusively in vitro resulting in a conflict between in situ bioassay data and in vitro metabolic studies. In vitro propoxur metabolism with NADPH-fortified microsomes produced at least nine metabolites. Formation of metabolites was NADPH-dependent; no quantifiable metabolism was detected with cytosolic fractions. However, microsomal fractions lacking an NADPH source did produce a low, but detectable, quantity of metabolites (1.6 pmol). PBO inhibited NADPH-dependent propoxur metabolism in a dose-dependent fashion, implicating cytochrome P450 monooxygenases as the enzyme system responsible for the metabolism. Interestingly, DEF also inhibited the NADPH-dependent metabolism of propoxur, albeit to a lower extent. Treatment with PBO or DEF also caused a significant reduction in the cuticular penetration rate of propoxur. The data demonstrate that unanticipated effects are possible with synergists and that caution must be exercised when interpreting synergist results.  相似文献

Forty-five field-collected strains of German cockroaches, Blattella germanica were tested for resistance to 12 different insecticides by the time-mortality response method in comparison with a known susceptible strain. Only low to moderate resistance to diazinon, chlorpyrifos, and acephate was detected. Resistance to malathion was widespread; about half of the strains tested showed high resistance. High resistance to the carbamates propoxur and bendiocarb also occurred. High resistance was uncommon with propoxur, but about 35 strains were highly resistant to bendiocarb. High resistance to pyrethrins was observed in about of the strains tested. Resistance to the pyrethroids allethrin, permethrin, phenothrin, fenvalerate, and cyfluthrin was detected in some of the strains examined. All of the strains tested were susceptible to one or more of the insecticides used. These results indicate that, although resistance is a serious problem in this species, satisfactory control should be possible by selection of an appropriate insecticide.  相似文献

The response of a susceptible and two pyrethroid-resistant field strains of the German cockroach, Blattella germanica, to four emulsifiable concentrates (ECs)--Tempo (cyfluthrin), Demon (cypermethrin), Dursban (chlorpyrifos) LO, and XRM 5184 (chlorpyrifos)--was investigated. Application rates were 0.4, 0.1, 0.5, and 0.5%, respectively. Susceptible cockroaches avoided dried formulations of the pyrethroids Tempo and Demon, but resistant cockroaches did not; avoidance was greater with Demon than with Tempo. Filter papers freshly impregnated (wet) with Demon and Tempo flushed susceptible cockroaches (Demon greater than Tempo). Resistant cockroaches were flushed only by Demon, but less so than susceptible strain cockroaches. No mortality occurred in experiments with the pyrethroids. Dried formulations of Dursban LO and XRM 5184 had little, if any, repellency to nymphs of either the susceptible or resistant strains. Initially (1-3 h), most cockroaches settled on treated papers. Subsequent movement off treated papers was a step in the process leading to knockdown. At 24 h, mortality of susceptible cockroaches was 100% in the experiment with Dursban LO and 95% in the experiment with XRM 5184. Highest mortality in a resistant strain (40%) was in the contact experiment with Dursban LO. In contact repellency and flushing experiments, mortality was higher with Dursban LO than with XRM 5184. Neither chlorpyrifos formulation flushed cockroaches effectively. Mortality in flushing experiments was less than in the contact repellency experiments.  相似文献

The effect of topically applied chlorpyrifos on acetylcholinesterase and other esterases in heads and decapitated bodies of CSMA and Crawford German cockroaches was examined with spectrophotometric enzyme assay and native polyacrylamide gel electrophoresis. The toxicity of chlorpyrifos was greatly reduced in decapitated CSMA male cockroaches with LD50 value 17.1-fold higher than that of normal CSMA cockroaches. Acetylcholinesterase activity from heads was significantly higher in the Crawford compared with the CSMA strain and did not change until 24 h after chlorpyrifos in vivo treatment in both strains. The p-nitrophenyl butyrate (NPB) esterase activities from both heads and decapitated bodies of the resistant Crawford strain were significantly greater than the susceptible CSMA strain. The p-NPB esterase activity was significantly inhibited by chlorpyrifos in vivo treatment, and total p-NPB esterase activity was significantly reduced in decapitated bodies compared with heads of both strains. Native polyacrylamide gel electrophoresis (PAGE) analysis of extracts solubilized with Triton X-100 from heads and decapitated bodies revealed five major esterase bands and an acetylcholinesterase (AChE) band with a high capability of hydrolyzing alpha-naphthyl butyrate and acetylthiocholine, respectively. In the heads of susceptible CSMA male cockroaches, the activity of mobile isozymes d1 and d2 was completely inhibited at 24 h after chlorpyrifos application, and isozyme e was partially inhibited. In contrast, isozymes c1 and c2 from the decapitated bodies of CSMA cockroaches were mostly affected at 24 h after the topical application of chlorpyrifos. The activities of acetylcholinesterase and esterase isozymes a and b from the decapitated body remained uninhibited in both strains. Inhibition of isozymes d1 and d2 seems to be more important in chlorpyrifos intoxication than acetylcholinesterase.  相似文献

Exposure of male German cockroaches, Blattella germanica L., to propoxur produced significant water loss at 2 and 24 h after treatment. This effect was dependent upon the state of hydration before insecticide exposure. Hydrated males lost as much as 10% of their total body mass in the form of water; dehydrated males lost about 4.3%; and untreated controls lost approximately 1.8% during the same time period. No significant compensatory feeding responses were observed 1-4 d after treatment. Water depletion associated with propoxur exposure resulted in increased water consumption for the first 2 d after treatment and then reached a plateau for the duration of the study. Males consumed water daily regardless of insecticide exposure.  相似文献

ABSTRACT The laboratory studies were carried out for evaluating control effects of four commercial insecticidal baits such as two different hydramethylnon [2.0%(AI)] products (DBK® and Combat‐Gold®), fipronil [0.05 %(AI)] (Combat‐Power®) and (0.6% chlorpyrifos [0.6%(AI)] (Raid‐Roachbait®) against German cockroaches (Blattella germanica). The control rates of four kinds of toxic baits were all 100.0% mortality of German cockroaches in 5 days after treatment. The results of chlorpyrifos and fipronil brought 100.0% mortalities in 2 to 3 days after treatment, respectively. After 3 day treatment, there was no significant difference of control effect among the four toxic baits. As a result of this study, fipnonil and chlorpyrifos showed faster killing action against German cockroaches than the two hydramethylnon formulation products. In the choice test, DBK® (hydramethylnon) (average 17.0 ind.) significantly attracted more German cockroaches than Combat‐Gold® (hydramethylnon) (avg. 7.0 ind.), Combat‐Power® (fipronil) (avg. 5.3 ind.) and Raid‐Roachbait® (chlorpyrifos) (avg. 3.3 ind.). The difference in attraction effects came 10 minutes after treatment. In order to evaluate the secondary killing effect of toxic baits by coprophagy against adult Blattella germanica, the comparative test was carried out. The adult mortality rates were significantly different among the four toxic baits showing 86.7% mortality for fipronil, 60.0% for hydramethylnon (DBK®), 30.0% for chlorpyrifos, and below 13.3% for hydramethylnon (Combat‐Gold®) in 6 days. During the first 12 days, fipronil showed the highest mortality rate (90.0%), followed by hydramethylnon (DBK®) (60.0%). The secondary killing effect of toxic baits by coprophagy appeared in all of the baits against adult Blattella germanica.  相似文献

Visual inspections and trapping revealed the presence of German cockroaches, Blattella germanica (L.) in 62 of 100 randomly selected commercial food-handling establishments. From live cockroaches trapped in 55 restaurants, 35 strains were established and tested for chlorpyrifos resistance. Poor sanitation and the likelihood of trapping cockroaches were associated. Twenty strains had resistance ratios (RR) as LD50 greater than 10-fold, a value exceeding the pest management threshold level for this insecticide. Resistance ratios were not associated with numbers of cockroaches trapped, the level of sanitation, numbers of different insecticide products used, or the number of treatments per month. Significantly more categories of insecticides (e.g., carbamate, organophosphate, inorganic) were used in restaurants with RR greater than 10 than in those with RR less than 10.  相似文献

The susceptibility of American cockroaches, Periplaneta americana (L.); smoky brown cockroaches, P. fuliginosa (Serville); oriental cockroaches, Blatta orientalis L.; German cockroaches, Blattella germanica (L.); and brownbanded cockroaches, Supella longipalpa (F.), to Steinernema carpocapsae Weiser (All strain) was evaluated under laboratory conditions. A 1-ml water suspension containing 500,000 nematodes was placed on filter paper in a petri dish or the pad of a bait station. German, brown-banded, oriental, and smoky-brown cockroaches died within 1 d after placement in the petri dishes. The relative order for the LT50s were American greater than oriental greater than smoky-brown greater than brown-banded = German. All cockroaches actively groomed nematodes from legs and antennae of forced (petri dish) exposure. The LT50s for S. carpocapsae for nonforced (bait station) exposure were significantly greater than those for forced exposure. The LT50s were 3.25, 4.13, 9.86, and 11.38 d for brown-banded, German, oriental, and smoky-brown cockroaches, respectively. The relative order of the LT50s after forced (American greater than oriental greater than smoky-brown greater than German = brown-banded) and nonforced (American greater than smoky-brown greater than oriental greater than German greater than brown-banded) exposure to S. carpocapsae was inversely related to the moisture of their preferred habitats.  相似文献

Bait formulations containing boric acid, chlorpyrifos, or hydramethylnon were evaluated in field and laboratory experiments against the German cockroach, Blattella germanica (L.). In continuous exposure tests, chlorpyrifos formulations produced 100% mortality in less than 24 h, boric acid formulations had LT50's of approximately 5.5 d, and hydramethylnon had an LT50 of 0.86 d. LT50's estimated in Ebeling choice boxes were 1.6 to 10.8 times greater than those from continuous exposure tests for chlorpyrifos formulations and 2.5 times greater for the hydramethylnon formulation. The same LT50's were produced by choice box and continuous exposure tests for boric acid formulations. Relative repellency, measured as the mean percentage of live cockroaches in the light side of the choice box, was greatest for chlorpyrifos formulations, followed by boric acid formulations, then hydramethylnon. A performance index (PI) that combined choice box repellency and mortality data indicated that hydramethylnon (Combat) had the greatest potential for field effectiveness. Only hydramethylnon significantly reduced German cockroach populations in the field. Apartments treated with the IT WORKS formulation of boric acid bait had significantly greater cockroach trap catches. The PI is an accurate predictor of the field performance of toxic baits for German cockroach control.  相似文献

The effect of Metarhizium anisopliae (Metschnikoff) Sorokin strain ESC-1 alone and in combination with sublethal doses of commercial formulations of chlorpyrifos, propetamphos and cyfluthrin on mortality of CSMA strain of German cockroach, Blattella germanica (L.), was determined by conducting in vivo studies that included 3 bioassays. Spores of M. anisopliae cultured on SDAY media had germination of >90%. Based on bioassay 1, doses ranging from 0.5 to 300 ppm of chlorpyrifos and propetamphos, and 0.05 to 40 ppm of cyfluthrin were selected for bioassays 2 and 3. Cockroach mortality ranged from 5 to 20% for insecticides alone and 48 to 70% for insecticides + M. anisopliae in bioassay 2. In bioassay 3, mortality ranged from 15 to 60% for insecticides and 57.5 to 92.5% for insecticides + M. anisopliae. Percentage of cockroach mortality resulting from insecticide + M. anisopliae combinations was significantly higher than insecticide alone. Mortality was also significantly higher in certain insecticide + M. anisopliae combinations than M. anisopliae alone. There was no significant interaction between M. anisopliae and insecticides with their concentrations in bioassay 2, indicating an additive effect. But in bioassay 3, a significant interaction was observed when M. anisopliae was combined with multiple insecticide concentrations. The interaction indicated an additive effect for chlorpyrifos and cyfluthrin, and a synergistic effect for propetamphos. There were significant differences in LT50 among various treatment combinations. M. anisopliae alone or insecticide + M. anisopliae combinations did not affect body weight in treated German cockroaches.  相似文献

Introduction. The susceptibility of Anopheles albimanus to organophosphates, carbamates and pyrethroid insecticides was unknown in the Panama communities of Aguas Claras, Pintupo and Puente Bayano, located in the Amerindian Reservation of Madungandi. This region is considered a malaria transmission area, where An. albimanus is the main vector. Objective. The resistance to organophosphate insecticides, carbamates and pyrethroids was evaluated in field populations of the Anopheles albimanus in Panama. Materials and methods. Progeny of An. albimanus collected in three localities in the indigenous Madugandi region were exposed to bioassays of susceptibility to organophosphate insecticides (fenitrothion, malathion and chlorpyrifos), the carbamate (propoxur) and pyrethroids (deltamethrin, lambdacyhalothrin, cyfluthrin and cypermethrin). The protocols were in accordance with those established for adult mosquitoes by World Health Organization. Results. The three strains of the An. albimanus were resistant to the pyrethroid insecticides deltamethrin, lambdacyhalothrin, cyfluthrin and cypermethrin. Susceptibility remained for the organophosphate insecticides fenitrothion, malathion, chlorpyrifos, and the carbamate insecticide propoxur. Conclusion. The results provided important information to the vector control program, contributing to the application of new strategies on the use of insecticides, and thereby lengthening the life of the insecticide in use.  相似文献

The susceptibility values for field‐collected (BG‐BG) and laboratory (KS‐BG) strains of German cockroaches (BS‐BG), Blattella germanica (L.) to five synthetic pyrethroid and three organophosphate insecticides were assessed by topical application. Both KS‐BG and BS‐BG strains demonstrated the highest levels of susceptibility to deltamethrin with LD50 values of 0.0035 μg/female and 1.5744 μg/female, respectively. The KS‐BG strain demonstrated the lowest susceptibility level to fenthion with an LD50 value of 0.0698 μg/female, while the BS‐BG field population showed the lowest susceptibility to bifenthrin with 4.6811 μg/female. The BS‐BG field population showed resistance levels of 50 to 640 fold higher against the eight pesticides evaluated compared to the KS‐BG strain. The resistance ratios (RR) of the eight pesticides were compared using populations of B. germanica that were collected from the same locality during 2007 and 2014. The field population of B. germanica collected during 2014 demonstrated significantly high resistance to an organophosphates and all pyrethroids when compared with those collected during 2007, but RR values were <10 except for chlorpyrifos‐methyl and chlorpyrifos with RR values of 74 and 104, respectively.  相似文献

Multifactored logistic models were developed for chlorpyrifos and cyfluthrin formulations based on mortality data from laboratory studies with the German cockroach, Blattella germanica (L.). Insecticides were applied to stainless steel surfaces and aged at three different temperatures (23, 30, and 37 degrees C) and two levels of relative humidity (40 and 70%). After the insecticides dried, the treated panels were placed opposite plywood panels to simulate a crack and crevice application. At appropriate aging times, treated panels were removed from environmental chambers for bioassay. The combined effects of high temperature, high humidity, and aging of residues caused the greatest decline in cockroach mortality for chlorpyrifos. Increasing temperature and aging of residues resulted in decreased cockroach mortality for cyfluthrin formulations; however, mortality was greater than 87% for all formulations through 84 d. Information from this study can be incorporated into integrated pest management programs for German cockroaches.  相似文献

The effects of sublethal concentrations of chlorpyrifos, cyfluthrin, and hydramethylnon on adult longevity and fecundity of German cockroaches, Blattella germanica (L.), were investigated. Longevity of males declined linearly with increasing doses of insecticide. An LD50 of cyfluthrin decreased male longevity by 52%, whereas an LD50 of hydramethylnon reduced male longevity by 81%. Longevity of females increased linearly with increasing sublethal doses of chlorpyrifos, whereas all doses of both cyfluthrin and hydramethylnon decreased longevity of females. Fecundity increased linearly with increasing sublethal doses of chlorpyrifos. Number of oothecae formed, oothecae hatched, and number of offspring produced in each ootheca increased with increasing sublethal concentrations of chlorpyrifos. In contrast, fecundity declined with increasing sublethal concentrations of cyfluthrin and hydramethylnon. Formation of the first ootheca occurred approximately 8 d after mating for untreated females but generally longer with sublethal concentrations of all insecticides. The period between oothecae hatch and the formation of subsequent oothecae increased with successive oothecae in all treatments.  相似文献
