The feeding and growth of wild juvenile Japanese flounder Paralichthys olivaceus (Temminck et Shlegel), collected after experimental mass releases of marked hatchery-reared juveniles of the same species, was examined at Wada Beach, Wakasa Bay, the Japan Sea, in 1997 and 1998. The feeding and growth of wild juveniles in the eastern area where the hatchery-reared juveniles were released and recaptured were compared with those in the western area (control area) where no hatchery-reared juveniles were recaptured. The stomach contents of both hatchery-reared and wild juveniles collected in the eastern and western areas consisted mainly of mysids in the early season (late-May to mid-June), and fish larvae in the late season (late-June to early July) in both 1997 and 1998. The stomach content index (SI) of juvenile flounder collected in the eastern and western areas showed that the feeding conditions followed similar patterns. However, the percent of empty stomachs in the wild flounder juveniles collected within 4 days after the mass release in the eastern area was as high as 10-30% in 1997 and 40% in 1998, whereas it was 0% in the western area in both years. Otolith microstructure analysis showed that the growth rate in the eastern area was not significantly different before and after release in both years. These results demonstrate that the mass release of hatchery-reared juvenile flounder temporarily affects the feeding efficiency of the cohabiting wild juveniles, but not to an extent that significantly reduces the growth rate.  相似文献

Aluminium (Al) toxicity decreases plant growth. Secale cereale L. is among the most Al-tolerant crop species. In order to study the response to Al-long term exposure, two rye genotypes with different Al sensitivity (‘D. Zlote’ and ‘Riodeva’) were exposed to 1.11 and 1.85 mM Al and the antioxidant responses were followed for 2 and 3 weeks in roots and leaves. Al toxicity signals, such as a severe decrease in root growth, occurred sooner in ‘Riodeva.’ The antioxidant response was dependent on the genotype, the organ, Al concentration and the exposure period. Al-exposed roots of ‘D. Zlote’ showed earlier enhancements of APX, SOD and G-POX activities than those of ‘Riodeva.’ ‘D. Zlote’ roots showed stimulation of the AsA–GSH cycle after the second week (when root growth inhibition was less severe), while later (when severe root growth inhibition was observed), oxidation of AsA and GSH pools was observed. In leaves of both genotypes, CAT, SOD and G-POX activities increased with Al exposure. In these leaves, the effect of AsA–GSH was time dependent, with maximum oxidation at the second week, followed by recovery. We confirmed that the oxidation state of AsA and GSH pools is involved in the detoxification of Al-induced oxidative stress. Moreover, our data demonstrate that the production of ROS does not correlate with the Al-induced root growth decrease. Finally, the differences observed over time indicate that long term exposure may provide additional information on rye sensitivity to Al, and contribute to a better understanding of this species’ mechanisms of Al tolerance.  相似文献

Microorganisms are essential for maintaining ecosystem balance, and understanding their response to toxic pollutants is important in assessing the potential environmental impacts of such releases. In this study, the response to the heavy metal cadmium and the potential defense or adaptive mechanisms of the widely used white-rot fungus, Phanerochaete chrysosporium, were investigated. The results indicated that cadmium causes plasma membrane damage, including rigidification of lipids, a decrease in H+-ATPase activity, and lipid peroxidation. The cellular death may be mediated by oxidative stress with mitochondria membrane potential (MMP) breakdown and reactive oxygen species (ROS) formation. Parts of the cells were able to survive by activating antioxidant defense systems (antioxidant agents and enzymes). Extracellular synthesis of cadmium crystal particles was observed after exposure to dissolved cadmium ion, which is probably another detoxification mechanism in which the dissolved metal is precipitated, thus reducing its bioavailability and toxicity. These physiological responses of P. chrysosporium to cadmium together with the defense mechanisms can provide useful information for the development of fungal-based technologies to reduce the toxic effects of cadmium.  相似文献

Feeding behaviour of Japanese flounder larvae under laboratory conditions   总被引:6,自引:0,他引:6  
Tank–reared Japanese flounder larvae, Paralichthys olivaceus , had a major feeding peak in the morning and a secondary peak in the afternoon throughout the larval development, with light being the primary factor regulating their feeding activity. The larvae consumed rotifers in preference to Artemia for up to 10 days, after which the food preference shifted to Artemia . Feeding rates of the larvae prior to 10 days post–hatch depended on prey density, but in the old larvae, feeding rates were independent of prey density. Maximum feeding rate occurred at 19° C. The occurrence of the attack posture, after its onset at first feeding (2 days post–hatch), increased up to 25 days, began to decrease when the larvae prepared to settle down, then disappeared after settlement. The occurrence frequency of the attack posture was positively related to fish density, but inversely related to starvation duration, and occurred most frequently at 19° C. This posture depended on prey density in larvae prior to 10 days post–hatch, but became independent of prey density as the larvae developed. It was obvious that, for flounder larvae, attack posture was a behavioural character closely related to feeding and subject to larval development and environmental factors.  相似文献

Randomly amplified polymorphism DNA (RAPD) analysis was performed in order to evaluate one common population and two selected hatchery populations of flounder Paralichthys olivaceus. A total of 90 individuals belonging to three populations were screened using 12 different RAPD primers. A total of 71 loci were produced in the three studied populations, and 43.5%, 42.4% and 42.2% of these loci were polymorphic over all the genotypes tested in common, susceptible and resistant populations, respectively. The number of loci detected by single primers ranged from four to eight. The average heterozygosites of common, susceptible and resistant populations were 0.112, 0.103 and 0.111, respectively. Compared with the common population, the two selected hatchery populations, susceptible and resistant, showed some genetic differences including a smaller number of loci, a smaller number of polymorphic loci, a smaller number of genotypes and a smaller Shannon index. The present study confirmed that it is important to detect the genetic variability of the selected hatchery population for the conservation of natural flounder resources.  相似文献

Philasterides dicentrarchi is a histophagous scuticociliate causes fatal scuticociliatosis in farmed olive flounder Paralichthys olivaceus. The average monthly prevalence of scuticociliatosis due to P. dicentrarchi infections was increased from May to July (40 ± 3.1% to 79.4 ± 1.7%) and it decreased from August to November (63 ± 2.3% to 30 ± 2.6%) in olive flounder farms at Jeju Island, South Korea during 2000-2006. The prevalence of mixed infection along with Vibrio spp. bacterial infection was 49 ± 7.2% than that of other mixed infection. At present no effective control measure for P. dicentrarchi infection has been described and large production losses continue. In the present study, formalin, hydrogen peroxide and Jenoclean chemotheraputants were used for bath treatment. Among Jenoclean at a low concentration of 50 ppm proved effective. The results were confirmed with in vitro motility assessments and morphological changes scoring system in P. dicentrarchi. On the other hand, similar trend was noted following hydrogen peroxide treatment at this concentration, but formalin was only moderately effective. Either hydrogen peroxide or Jenoclean are the promising compounds effective at low concentrations with short application time for P. dicentrarchi. Therefore, these substances were evaluated on day 10, 20 and 30 for their ability to enhance innate immune response and disease resistance against P. dicentrarchi in olive flounder after chemotheraputants bath treatment with 100 ppm for 30 min per day. All the tested immune parameters were enhanced by treatment with Jenoclean, but not formalin and hydrogen peroxide. These findings suggest that Jenoclean bath treatment can be used for ensuring the heath of cultured marine fish against internal parasites such as P. dicentrarchi.  相似文献

Flounder were exposed to waterborne phenanthrene (0.5, 1 and 2 μM) for 4 weeks to test effects of waterborne phenanthrene on growth and hematological properties of the olive flounder (Paralichthys olivaceus). The average weight gain (WG) of flounder was significantly decreased in fish exposed to phenanthrene at 2.0 μM for 2 weeks, whereas WGs of fish treated by 1.0 and 2.0 μM phenanthrene for 4 weeks were significantly decreased. However, hepatosomatic index (HSI) and condition factor (CF) of flounder were not significantly affected by phenanthrene exposure. Red blood cell (RBC) count, hemoglobin (Hb) concentration, hematocrit (Ht), the mean corpuscular hemoglobin (MCH) and the mean corpuscular hemoglobin concentration (MCHC) mean levels were decreased with an increase in exposure time of phenanthrene to the fish, but the level of the mean corpuscular volume (MCV) was increased. Plasma bilirubin concentrations were significantly increased following exposure to waterborne phenanthrene (2.0 μM) for 2 and 4 weeks; however, there were no significant changes in plasma total cholesterol in fish of all treated groups compared to control. The phenanthrene-exposed groups (≥1.0 μM) showed significantly higher mean plasma lysozyme activity. Kidney lysozyme activity of fish exposed to phenanthrene (≥1.0 μM) was also significantly higher than that of control fish. The central finding from these data is that olive flounder exposed to waterborne phenanthrene at more than 1.0 μM are likely to experience negative impacts on fish health and basic physiological functions.  相似文献

The main objective of this investigation was to examine physiological changes occurring within the olive flounder, Paralichthys olivaceus, in the presence of continuous stress factors. The experiment gave continuous stress, and each stress step is as follows: water level reduction (S1), selection process (S2), confinement (S3), air exposure (S4), transportation (S5), and storing in volume 0.5-t (S6) and 50.0-t (S7) rearing tanks for 24 h after transportation. The cortisol concentration showed a trend to continuously rise in response to consecutive stress from transportation from 7.4 ± 0.6 ng/ml in an experimental opening. The concentration showed the highest level, 25.3 ± 4.4 ng/ml, after confinement stress. Glucose concentration in S3 and S4 were increased significantly to 71.0 ± 13.0 and 78.7 ± 7.0 mg/dl, respectively (P < 0.05). Lactic acid was increased significantly from 0.5 ± 0.1 mM (S0) to 9.0 ± 1.2 mM (S4), but it did not recover until 24 h (7.0 ± 1.0 mM) after transportation (S6 and S7) (P < 0.05). Some contents displayed recovery within 24 h, but a longer time would be required for normal physical metabolism to resume after fish experienced stress. Therefore, it would be endangering the survival of this species if the transport were repeated within a 24-h period.  相似文献

Summary Histological changes in the pituitary TSH cells and in the thyroid gland of flounder (Paralichthys olivaceus) larvae during spontaneous or artificially induced metamorphosis were studied. Activity of the immunoreactive TSH cells (IrTSH cells) gradually increased during premetamorphosis, reaching the highest level in prometamorphic larvae, and the cells were degranulated in metamorphic climax. The IrTSH cells were most inactive at the post-climax stage. The thyroid gland was morphologically the most active in metamorphic climax when the degranulation occurred in the pituitary IrTSH cells, and appeared inactive at post-climax. A few weeks after metamorphosis, both the IrTSH cells and the thyroid gland appeared to be activated again in the benthic, juvenile flounder. Administration of thyroxine or thiourea revealed negative feedback regulation of the pituitary-thyroid axis in flounder larvae. These results indicate that activation of the pituitary-thyroid axis induces metamorphosis in the flounder.  相似文献

Most aerobic organisms are exposed to oxidative stress. Looking for enzyme activities involved in the bacterial response to this kind of stress, we focused on the btuE-encoded Escherichia coli BtuE, an enzyme that shares homology with the glutathione peroxidase (GPX) family. This work deals with the purification and characterization of the btuE gene product.Purified BtuE decomposes in vitro hydrogen peroxide in a glutathione-dependent manner. BtuE also utilizes preferentially thioredoxin A to decompose hydrogen peroxide as well as cumene-, tert-butyl-, and linoleic acid hydroperoxides, confirming that its active site confers non-specific peroxidase activity. These data suggest that the enzyme may have one or more organic hydroperoxide as its physiological substrate.The btuE gene was induced when cells were exposed to oxidative stress elicitors that included potassium tellurite, menadione and hydrogen peroxide, among others, suggesting that BtuE could participate in the E. coli response to reactive oxygen species. To our knowledge, this is the first report describing a glutathione peroxidase in E. coli.  相似文献

A growing body of evidence has pointed to a relationship between oxidative stress and depression. Thus, the present study was aimed at evaluating the effects of the antioxidants n-acetylcysteine (NAC), deferoxamine (DFX) or their combination on sweet food consumption and oxidative stress parameters in rats submitted to 40 days of exposure to chronic mild stress (CMS). Our results showed that in stressed rats treated with saline, there was a decrease in sweet food intake and treatment with NAC or NAC in combination with DFX reversed this effect. Treatment with NAC and DFX decreased the oxidative damage, which include superoxide and TBARS production in submitochondrial particles, and also thiobarbituric acid reactive substances (TBARS) levels and carbonyl proteins in the prefrontal cortex, amygdala and hippocampus. Treatment with NAC and DFX also increased the activity of the antioxidant enzymes, superoxide dismutase and catalase in the same brain areas. Even so, a combined treatment with NAC and DFX produced a stronger increase of antioxidant activities in the prefrontal cortex, amygdala and hippocampus. The results described here indicate that co-administration may induce a more pronounced antidepressant activity than each treatment alone. In conclusion, these results suggests that treatment with NAC or DFX alone or in combination on oxidative stress parameters could have positive effects against neuronal damage caused by oxidative stress in major depressive disorders.  相似文献

We have previously derived 2 V79 clones resistant to menadione (Md1 cells) and cadmium (Cd1 cells), respectively. They both were shown to be cross-resistant to hydrogen peroxide. There was a modification in the antioxidant repertoire in these cells as compared to the parental cells. Md1 presented an increase in catalase and glutathione peroxidase activities whereas Cd1 cells exhibited an increase in metallothionein and glutathione contents. The susceptibility of the DNA of these cells to the damaging effect of H2O2 was tested using the DNA precipitation assay. Both Md1 and Cd1 DNAs were more resistant to the peroxide action. In the case of Md1 cells it seems clear that the extra resistance is provided by the increase in the two H2O2 scavenger enzymes, catalase and glutathione peroxidase. In the case of Cd1 cells the activities of these enzymes as well as of superoxide dismutases (Cu/Zn and Mn) are unaltered as compared to the parental cells. The facts that parental cells exposed to 100 μM Zn2+ in the medium exhibit an increase in metallothionein but not in glutathione and that these cells become more resistant to the DNA-damaging effect of H2O2 suggest that this protein might play a protective role in vivo against the OH radical attack on DNA.  相似文献

The aim of this study was to evaluate the health benefits associated with apple consumption following cadmium exposure. A total of 15 Wistar rats were distributed into three groups (n = 5), as follows: control group (non-treated group, CTRL); cadmium group (Cd) and apple juice group (Cd + AJ). The results showed a decrease in the frequency micronucleated cells in bone marrow and hepatocytes in the group exposed to cadmium and treated with apple juice. Apple juice was also able to reduce the 8OHdG levels and to decrease genetic damage in liver and peripheral blood cells. Catalase (CAT) was decreased following apple juice intake. Taken together, our results demonstrate that apple juice seems to be able to prevent genotoxicity and oxidative stress induced by cadmium exposure in multiple organs of Wistar rats.  相似文献

Predator-prey interaction between sandy shore crab, Matuta lunaris (Forskål, 1775), and juvenile Japanese flounder, Paralichthys olivaceus (Temminck et Schlegel), was investigated under controlled laboratory conditions. Possibility of training and conditioning hatchery-reared flounder to avoid predators was also examined. Crabs took over 75% of their daily ration at night when they were given access to prey 24 h a day. Large (64.8±5.4 g)- and medium (30.68±3.33 g)-sized crabs ate ca. 5.5±1.45 and 3.9±1.99 individuals of flounder (TL=4.96±0.23 cm) a day, respectively. When flounder juveniles that have experienced predation pressure by crabs encountered predators again, they exhibited better survival compared to the naive fish. Flounder juveniles were also conditioned either using small and, thus, benign predators, or large crabs over fence. The conditioned fish with either method were better able to avoid capture by crabs than naive fish, revealing that learning process should play an important role in their predator avoidance. Anti-predator performance was also compared between starved and fed flounder juveniles. Fed fish were rarely eaten by predators after 3 h of exposure, whereas starved fish continued to be eaten. Our results suggest that stock-enhancement program of Japanese flounder can be improved by applying proper feeding protocol and conditioning to avoid predators prior to release. Present research supports the idea that behavioural and ecological consideration for the target species is indispensable for the success of stock enhancement.  相似文献

We characterized thioredoxin reductase 1 (TrxR1) from Chironomus riparius (CrTrxR1) and studied its expression under oxidative stress. The full-length cDNA is 1820 bp long and contains an open reading frame (ORF) of 1488 bp. The deduced CrTrxR1 protein has 495 amino acids and a calculated molecular mass of 54.41 kDa and an isoelectric point of 6.15. There was a 71 bp 5’ and a 261 bp 3' untranslated region with a polyadenylation signal site (AATAAA). Homologous alignments showed the presence of conserved catalytic domain Cys-Val-Asn-Val-Gly-Cys (CVNVGC), the C-terminal amino acids ‘CCS’ and conserved amino acids required in catalysis. The expression of CrTrxR1 is measured using quantitative real-time PCR after exposure to 50 and 100 mg/L of paraquat (PQ) and 2, 10 and 20 mg/L of cadmium chloride (Cd). CrTrxR1 mRNA was upregulated after PQ exposure at all conditions tested. The highest level of CrTrxR1 expression was observed after exposure to 10 mg/L of Cd for 24 h followed by 20 mg/L for 48 h. Significant downregulation of CrTrxR1 was observed after exposure to 10 and 20 mg/L of Cd for 72 h. This study shows that the CrTrxR1 could be potentially used as a biomarker of oxidative stress inducing environmental contaminants.  相似文献

Since the double Δgrx1Δgrx2 mutant is hypersensitive to selenite we decided to evaluate mechanisms underlying this phenomenon and establish the roles of other components of yeast glutaredoxin system, in particular glutaredoxin 5 in the selenite resistance. We found elevation in the intracellular and mitochondrial superoxide production in the Δgrx1Δgrx2 and Δgrx5 mutants after Se(IV) treatment. The last effect was more pronounced for cells lacking the mitochondrial Grx5 protein. We also recorded selenite-induced increase in the peroxide production in all strains tested. Nonfermentable carbon sources, glycerol and ethanol, augmented selenite toxicity. Hypo- and anoxia protected against the harmful effects of Se(VI). Augmentation of the intracellular levels of two endogenous antioxidants, erythroascorbic acid and glutathione confers resistance to selenite. We recorded a strain-unspecific, selenite-mediated decrease in the level of acid-soluble thiols. Collectively, our data demonstrate that hypersensitivity to the Δgrx1Δgrx2 and Δgrx5 disruptants to selenite is mediated by altered intracellular redox equilibrium.  相似文献
