Acute lymphoblastic leukemia (ALL) is a heterogeneous cancer commonly affecting children due to dysregulation of miRNA expression. In the current study, authors investigated the expression profile for miRNA-125b-1 and miRNA-203 among childhood ALL. Blood samples were collected from newly diagnosed childhood ALL and healthy control children. The expression profile for candidate miRNAs was detected using quantitative RT-PCR analysis. Statistical analysis were performed using receiver operating characteristic curve (ROC) to examine the diagnostic efficacy of the two miRNA and their levels among ALL clinicopathological factors and phenotypes. The median expression level for miRNA-125b-1 was significantly high in childhood ALL; while miRNA-203 level was significantly low in childhood ALL as compared to control ones. MiRNA-125-1 reported significant increase in T-ALL as compared to other ALL phenotypes. Median miRNA-203 level was high in T-ALL followed by pre-B-ALL although no significant difference was reported. Clinicopathological factors did not emphasize significance with either detected miRNAs. Using ROC curve the diagnostic efficacy was significant with an area under the curve 0.858 for miRNA-125b-1 (83.72, 100%) and 0.878 for miRNA-203 (97.67, 86.96%). The combination of the two key miRNAs revealed absolute sensitivity (100%). MiRNA-125b-1 and miRNA-203 can be useful molecular markers for diagnosis of ALL. Further studies with large cohort are warranted to validate these results.  相似文献

Background  

In a large number of studies, members of the microRNA (miRNA)-34 family such as miRNA-34a, miRNA-34b, miRNA-34c, as well as miRNA-125b and miRNA-155, have been shown to be regulators of apoptosis. The ability of these miRNAs to perform this function is mainly attributed to their ability to interact with the p53 tumor suppressor, which is a powerful regulator of the teratologic susceptibility of embryos. We chose to explore whether miRNA-34a/b/c, miRNA-125b and miRNA-155 may play a role in teratogenesis by using p53+/- pregnant mice treated with cyclophosphamide (CP) as a model. We evaluated how CP-induced alterations in the expression of these miRNAs in the embryonic limbs correlate with embryonic p53 genotype and CP-induced limb phenotypes.  相似文献

The present study aimed to determine the roles of miRNA-543 in osteoporosis in rats induced by ovariectomy. The osteoporosis rat model was established by ovariectomy induction. MiRNA-543 expression in osteoblasts was measured by quantitative real-time polymerase chain reaction. The cell proliferation and apoptosis were measured by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and flow cytometry assays, respectively. Western blot analysis was conducted to examine the expression of YAF-2 and AKT signaling. TargetScan analysis and dual-luciferase reporter assay were performed to determine the target gene of miRNA-543. MiRNA-543 was significantly upregulated in osteoporosis rat model. Overexpression of miRNA-543 significantly suppressed cell growth and promoted apoptosis in osteoblasts, whereas downregulation of miRNA-543 significantly enhanced cell growth and inhibited apoptosis. MiRNA-543 upregulation significantly inhibited YAF-2 expression and suppressed the phosphorylation and expression of AKT and p38 mitogen-activated protein kinases (MAPK) in osteoblasts. Furthermore, YAF-2 knockdown enhanced the effects of miRNA-543 on apoptosis in osteoblasts. AKT inhibitor MK2206 and p38 MAPK inhibitor SB203580 also enhanced the effects of miRNA-543 on apoptosis in osteoblasts. Our findings revealed that inhibition of miRNA-543 could protect osteoblasts against ovariectomy-induced osteoporosis through AKT/p38 MAPK signaling pathway by targeting YAF2.  相似文献

目的: 探讨miRNA-130a-3p对脂多糖(LPS)诱导的心肌细胞自噬与凋亡的影响及分子机制。方法: H9C2心肌细胞随机分为5组,即正常对照组,LPS模型组,miRNA阴性对照组(miRNA-negative control组),miRNA-130a-3p mimics组(过表达miRNA-130a-3p),miRNA-130a-3p mimics+LY294002组(过表达miRNA-130a-3p + PI3K抑制)。LPS模型组即终浓度为10 μg/ml的LPS诱导24 h,miRNA阴性对照组与miRNA-130a-3p mimics组是利用lipo3000将阴性对照miRNA及miRNA-130a-3p mimics转染至H9C2细胞,培养24 h后,再将LPS加入培养基中培养24 h。miRNA-130a-3p mimics + LY294002组是利用lipo3000将miRNA-130a-3p mimics转染至H9C2细胞,同时在培养基中加入10 μmol/L(终浓度)的LY294002,培养24 h后,再将浓度为10 μg/ml的LPS加入培养基中培养24 h。所有实验均重复5次以上。利用RT-qPCR检测细胞中miRNA-130a-3p mRNA的表达水平,利用CCK-8实验检测细胞活性,利用ELISA实验检测细胞培养液中肿瘤坏死因子-α(TNF-α),白细胞介素-6(IL-6),白细胞介素-1β (IL-1β)的含量,利用比色法检测细胞培养液中超氧化物歧化酶(SOD)、乳酸脱氢酶(LDH)的含量;利用Western blot检测细胞中p-PI3K蛋白,p-AKT蛋白,Bax蛋白,Bcl-2蛋白,cleaved-caspase-3蛋白,LC3蛋白,p62蛋白的表达水平。结果: 结果显示,与正常组相比较,LPS模型细胞中miRNA-130a-3p mRNA水平,p-PI3K蛋白与p-AKT蛋白的水平显著低于正常对照组(P<0.01);与LPS组相比较,miRNA-130a-3p mimics组细胞中p-PI3K,p-AKT蛋白的表达显著升高(P<0.01,P<0.05);与正常对照组相比较,LPS组细胞活性显著降低,细胞培养液中TNF-α,IL-6,IL-1β及 LDH的含量显著升高(P<0.01), SOD的含量显著降低(P<0.01),细胞中Bax蛋白,cleaved caspase-3蛋白,p62蛋白的表达显著升高(P<0.01),Bcl-2蛋白的表达和LC3II/I的比率显著降低(P<0.01);与LPS组相比较,miRNA-130a-3p mimics可提高细胞活性,降低细胞培养液中TNF-α,IL-6,IL-1β及LDH的含量(P<0.01,P<0.05),提高SOD的含量(P<0.05),降低细胞中Bax蛋白,cleaved caspase-3蛋白,p62蛋白的表达(P<0.01),促进Bcl-2蛋白的表达(P<0.01),提高LC3II/I的比率(P<0.05);与miRNA-130a-3p mimics组相比较,miRNA-130a-3p mimics+LY294002组,可部分逆转miRNA-130a-3p mimics对细胞的作用。结论: 过表达miRNA-130a-3p可部分通过激活PI3K/AKT信号通路促进细胞的自噬与抑制细胞凋亡,减轻LPS诱导的心肌细胞损伤。  相似文献

BackgroundHeritable factors are evidently involved in prostate cancer (PrCa) carcinogenesis, but currently, genetic markers are not routinely used in screening or diagnostics of the disease. More precise information is needed for making treatment decisions to distinguish aggressive cases from indolent disease, for which heritable factors could be a useful tool. The genetic makeup of PrCa has only recently begun to be unravelled through large-scale genome-wide association studies (GWAS). The thus far identified Single Nucleotide Polymorphisms (SNPs) explain, however, only a fraction of familial clustering. Moreover, the known risk SNPs are not associated with the clinical outcome of the disease, such as aggressive or metastasised disease, and therefore cannot be used to predict the prognosis. Annotating the SNPs with deep clinical data together with miRNA expression profiles can improve the understanding of the underlying mechanisms of different phenotypes of prostate cancer.ResultsIn this study microRNA (miRNA) profiles were studied as potential biomarkers to predict the disease outcome. The study subjects were from Finnish high risk prostate cancer families. To identify potential biomarkers we combined a novel non-parametrical test with an importance measure provided from a Random Forest classifier. This combination delivered a set of nine miRNAs that was able to separate cases from controls. The detected miRNA expression profiles could predict the development of the disease years before the actual PrCa diagnosis or detect the existence of other cancers in the studied individuals. Furthermore, using an expression Quantitative Trait Loci (eQTL) analysis, regulatory SNPs for miRNA miR-483-3p that were also directly associated with PrCa were found.ConclusionBased on our findings, we suggest that blood-based miRNA expression profiling can be used in the diagnosis and maybe even prognosis of the disease. In the future, miRNA profiling could possibly be used in targeted screening, together with Prostate Specific Antigene (PSA) testing, to identify men with an elevated PrCa risk.  相似文献

摘要 目的:探讨MiRNA-125a-5p对于脊髓损伤(SCI)后炎症反应、细胞凋亡及促进神经再生的作用机制。方法:建立6周龄大鼠SCI模型,将60只大鼠随机分为对照组(n=20)、SCI组(n=20)和MiRNA-125a-5p组(n=20),对照组仅行椎板切除术,SCI组大鼠鞘内注射等量生理盐水,MiRNA-125a-5p组大鼠鞘内注射MiRNA-125a-5p agomir。采用定量实时聚合酶链反应(qRT-PCR)检测各组大鼠MiRNA-125a-5p和核因子kB(NF-kB)的相对RNA表达水平。酶联免疫吸附法(ELISA)检测炎症因子白细胞介素1(IL-1)、趋化因子受体-4(CXCR-4)和单核细胞趋化因子-1(MCP-1)水平的表达。采用蛋白免疫印迹试验检测细胞凋亡蛋白和神经调节因子蛋白的表达情况。采用Basso-Beattie-Bresnahan(BBB)运动能力评定量表评价大鼠神经运动功能。结果:与对照组比较,SCI组MiRNA-125a-5p表达均降低,NF-kB表达均升高,差异均有统计学意义(P<0.05);与SCI组比较,MiRNA-125a-5p组MiRNA-125a-5p表达均升高,NF-kB表达均降低,差异均有统计学意义(P<0.05)。与对照组比较,SCI组IL-1、CXCR-4、MCP-1水平均升高,差异有统计学意义(P<0.05)。与SCI组比较,MiRNA-125a-5p组IL-1、CXCR-4、MCP-1水平均降低,差异有统计学意义(P<0.05)。与对照组比较,SCI组半胱氨酸天冬氨酸蛋白酶3(caspase-3)、聚腺苷二磷酸-核糖聚合酶(cleaved PARP)蛋白和原癌基因(bcl-2)蛋白表达均升高,神经细胞粘附分子1(NCAM1)、神经胶质蛋白1(NL1)和神经调节蛋白-1(NRG1)蛋白表达均降低,差异有统计学意义(P<0.05);与SCI组比较,MiRNA-125a-5p组caspase-3和cleaved PARP蛋白表达均降低,bcl-2、NCAM1、NL1和NRG1蛋白表达均升高,差异均有统计学意义(P<0.05)。与对照组比较,SCI组大鼠神经运动功能评分在第7 d以后显著降低,差异有统计学意义(P<0.05);与SCI组比较,MiRNA-125a-5p组大鼠神经运动功能评分在第7 d以后均升高,差异有统计学意义(P<0.05)。结论:MiRNA-125a-5p抑制NF-kB信号通路,降低炎症反应和细胞凋亡,促进神经再生,改善大鼠神经运动功能恢复。  相似文献

Murine Oct4+, very small embryonic-like stem cells (VSELs), are a quiescent stem cell population that requires a supportive co-culture layer to proliferate and/or to differentiate in vitro. Gene expression studies have revealed that the quiescence of these cells is due to changes in expression of parentally imprinted genes, including genes involved in cell cycle regulation and insulin and insulin-like growth factor signaling (IIS). To investigate the role of microRNAs (miRNAs) in VSEL quiescence, we performed miRNA studies in highly purified VSELs and observed a unique miRNA expression pattern in these cells. Specifically, we observed significant differences in the expression of certain miRNA species (relative to a reference cell population), including (i) miRNA-25_1 and miRNA-19 b, whose downregulation has the effect of upregulating cell cycle checkpoint genes and (ii) miRNA-675-3 p and miRNA-675-5 p, miRNA-292-5 p, miRNA-184, and miRNA-125 b, whose upregulation attenuates IIS. These observations are important for understanding the biology of these cells and for developing efficient ex vivo expansion strategies for VSELs isolated from adult tissues.  相似文献

Objective

Metastasis is the major cause of death in colorectal cancer patients. Expression of certain miRNAs in the primary tumors has been shown to be associated with progression of colorectal cancer and the initiation of metastasis. In this study, we compared miRNA expression in primary colorectal cancer and corresponding liver metastases in order to get an idea of the oncogenic importance of the miRNAs in established metastases.

Methods

We analyzed the expression of miRNA-21, miRNA-31 and miRNA-373 in corresponding formalin-fixed paraffin-embedded (FFPE) tissue samples of primary colorectal cancer, liver metastasis and healthy tissues of 29 patients by quantitative real-time PCR.

Results

All three miRNAs were significantly up-regulated in the primary tumor tissues as compared to healthy colon mucosa of the respective patients (p < 0.01). MiRNA-21 and miRNA-31 were also higher expressed in liver metastases as compared to healthy liver tissues (p < 0.01). No significant difference of expression of miRNA-31 and miRNA-373 was observed between primary tumors and metastases. Of note, miRNA-21 expression was significantly reduced in liver metastases as compared to the primary colorectal tumors (p < 0.01).

Conclusion

In the context of previous studies demonstrating increased miRNA-21 expression in metastatic primary tumors, our findings raise the question whether miRNA-21 might be involved in the initiation but not in the perpetuation and growth of metastases.  相似文献

Tight junction proteins play crucial roles in maintaining the integrity of intestinal mucosal barrier. MiRNA-182-5p is capable of targeting claudin-2 which is one of the vital tight junction proteins and the effect and mechanism of miRNA-182-5p was explored here in the DSS-induced colitis model. The pathological conditions were evaluated via hematoxylin and eosin staining. The gene expression level was assessed via PCR. Quantitative immunohistochemistry analysis was performed for the measurement of claudin-2. microRNA.org online tool was used for target gene prediction. Luciferase reporter assay and RNA pull-down assay were performed to detect the target of miRNA-182-5p. The inflammatory and oxidative stress level were measured using corresponding kits. MiRNA-182-5p was highly expressed in colitis model and miRNA-182-5p inhibitor exerted protective effects on colitis induced by DSS in mice. The protective effects includded improvement of pathological changes, increases in anti-inflammation and anti-oxidative genes, and up-regulation of TGF-β1. Claudin-2 mRNA was predicted as the target of miRNA-182-5p, which was validated via luciferase reporter assay and RNA pull-down assay. Claudin-2 overexpression was found in miRNA-182-5p inhibitor group. Consistent with the role of miRNA-182-5p, claudin-2 overexpression also exerted protective effects on DSS-induced colitis in mice. Inhibition of miRNA-182-5p exerted protective effects on colitis via targeting and upregulating claudin-2. The findings in study provide a new therapeutic strategy for colitis treatment and lay the foundation for future study.

Hepatic stellate cells activation (HSCs) plays a crucial role in the pathogenesis of liver fibrosis. Specific microRNAs have been suggested to affect the activation of HSCs via various signalling pathways including TGF-β/smads and Wnt/β-catenin pathways. Dasatinib is a multitarget inhibitor of many tyrosine kinases has recently studied for its anti-fibrotic effects in a variety of fibrous diseases. This study investigated the role of modulation of miRNA-378 and miRNA-17 in the pathogenesis of liver fibrosis through altering Wnt/β-catenin and TGF-β/smads pathways and evaluated the beneficial effect of the tyrosine kinase inhibitor, dasatinib, in thioacetamide-induced liver fibrosis model in mice. Treatment with dasatinib down-regulated miRNA-17 expression, leading to the restoration of WiF-1 and smad-7 which cause the inhibition of both Wnt/β-catenin and TGF-β/smads signalling. In addition, it upregulated miRNA-378 leading to the decrease of Wnt-10 which contributes to the suppression of activated HSCs.  相似文献

microRNAs (miRNAs) are implicated in carcinogenesis and their expression in biological fluids offer great potential as nucleic acid markers for cancer detection and progression. Authors investigated the expression level of miRNAs (miRNA-21, miRNA-126, and miRNA-155) to evaluate their role as diagnostic and prognostic markers for breast cancer compared with other commonly used protein-based markers (CEA and CA15-3). Serum samples from patients with breast cancer (n = 96), patients with benign breast lesion (n = 47), and healthy individuals (n = 39) were enrolled for detection of miRNA expression levels and protein-based tumor markers using fluorescent real-time quantitative polymerase chain reaction and enzyme-linked immunosorbent assay, respectively. Correlation among investigated markers with clinicopathological factors and clinical outcomes were determined. Expression of miRNA-21 and miRNA-155 revealed significant increases in patients with breast cancer compared with both benign and control groups, the same result was reported for tumor markers; on the other hand, miRNA-126 was significantly decreased in breast cancer group as compared with the other two groups. miRNA frequencies were significantly related to clinical staging and histological grading as compared with tumor markers. Patients with breast cancer with increased miRNA-21 and miRNA-155 and decreased miRNA-126 expressions had significantly worse disease-free survival, while only miRNA-21 and miRNA-126 showed poor OS (P< 0.005). In conclusion, investigated miRNAs were superior over tumor markers for the early stage of breast cancer especially those with high-risk factor and their assessment in blood facilitates their role as a potential prognostic molecular marker.  相似文献
