随着转Bt基因作物的大面积推广和应用,其释放的毒蛋白在土壤中的残留及对土壤生态系统的影响等问题已经成为人们关注的热点。国内外学者们通过室内构建大田模拟模型的方法对土壤中残留的Bt毒蛋白进行了研究,并取得了显著的进展。土壤是生态系统中物质循环和能量转化过程的主要场所,转Bt基因植物的外源基因表达的Bt毒蛋白可以通过植株残体、根及根系分泌物和花粉的散播等途径进入土壤生态系统,这些高度特化了的Bt毒素蛋白一旦在土壤中积累,将会导致土壤特异生物功能类群以及土壤多样性发生改变,甚至产生级联效应。大量研究表明,苏云金芽孢杆菌产生的Bt毒蛋白进入土壤后,可与土壤粘粒和腐质酸迅速结合,不易分离,而且较之游离态,更难被土壤微生物降解。纯化的Bt毒蛋白与无菌土壤中活性颗粒紧密结合后,存留时间至少可达234d。虽然结合态的Bt蛋白用酶联免疫法(ELISA)方法检测不到,但生物测定表明其仍保持杀虫活性。对转Bt基因作物的研究表明,Bt棉组织埋入土壤7d内,土壤中可提取的杀虫晶体蛋白浓度快速下降,之后下降速度比较稳定,甚至维持数周不变。而Bt玉米根系分泌物和植株残体释放的杀虫晶体蛋白在土壤中至少保持180d杀虫活性。虽然关于Bt毒蛋白在土壤中存留时间的长短,可能因实验材料、试验方法和条件的不同而不同。但是总之,如果长期种植转Bt基因作物,很可能会造成Bt毒蛋白在土壤中的积累,并最终威胁到整个土壤生态系统的平衡。目前对土壤中Bt毒蛋白定性和定量检测的方法主要有印迹分析法(Western-blotting)、SDS-PAGE法、斑点印迹酶联免疫吸附法(dot-blotELISA)、流式细胞仪法(Flowcytometer,FCM)、ELISA平板试剂盒及试剂条和生物测定法。其中最直接、简易、准确的方法是ELISA平板试剂盒及试纸条快速检测法和生物测定法。但检测土壤残留Bt毒蛋白时,采用的方法不同,检测的结果也有差异。  相似文献

为了评价转Bt稻谷储藏期的生态安全性,通过对在自然条件下不同储藏期Bt稻谷与非Bt稻谷抽样调查。结果表明,随着稻谷储藏期的延长,在储藏17个月后Bt稻谷上的虫蚀率依次为0.70%,1.20%,显著低于另外两种非Bt稻谷上的虫蚀率(汕优63:1.7%,2.0%;9311稻:2.70%,3.00%,P<0.05)。但Bt稻谷和非Bt稻谷中的赤拟谷盗、谷蠹、书虱、麦蛾数量,在19个月储藏期内,大部分调查结果差异不显著。  相似文献

麦套夏播转Bt基因棉R93-6对昆虫群落的影响   总被引:36,自引:6,他引:36  
以转Bt(Bacillusthuringiensis)基因棉品系R93-6为试验材料,以中棉所16号为对照,研究了在麦套夏播条件下转基因棉对昆虫群落的影响。结果表明,转基因棉田昆虫群落、害虫和天敌亚群落的多样性指数和均匀度指数均低于常规棉田,而优势集中性则高于常规棉田,所以转基因棉田昆虫群落、害虫和天敌亚群落的稳定性不如常规棉田,某种害虫大发生的可能性较大。对季节性变化格局的研究表明,转基因棉田昆虫群落可以划分为前期(6月初至7月下旬)、中期(7月底至8月底)和后期(9月份以后)三个发展阶段,根据不同阶段害虫和天敌发生的特点, 提出了害虫综合治理的策略。即前期害虫的防治应以生物生态调控为主;中期以化学防治为主,以生物生态调控为辅,协调好生物防治和化学防治的矛盾;后期应以生物生态调控为主,并加强农业防治。  相似文献

利用TAIL-PCR的方法克隆不同来源的转基因抗虫棉中外源基因插入区的侧翼序列并对其进行序列和结构分析,结果表明,同一个较基因的单构自交得到的不同株系中外源基因插入区的两侧DNA序列完全相同,不同的转基因抗虫棉虫的外源基因插入位置各不相同,不同来源的转基因品种外源基因插入的上游侧翼片段含有一段残留质粒片段,外源基因插入的下游侧翼片段为富含AT碱基结构,其中泗棉3号转基因抗虫品系中下游侧翼片段的AT碱基高达92%,Southern杂交结果显示这些侧翼序列为高AT含量的多拷贝序列,序列中没有发现拓扑异构酶的结合位点。  相似文献

Genetically modified (GM) Bt corn, through the pest protection that it confers, has lower levels of mycotoxins: toxic and carcinogenic chemicals produced as secondary metabolites of fungi that colonize crops. In some cases, the reduction of mycotoxins afforded by Bt corn is significant enough to have an economic impact, both in terms of domestic markets and international trade. In less developed countries where certain mycotoxins are significant contaminants of food, Bt corn adoption, by virtue of its mycotoxin reduction, may even improve human and animal health. This paper describes an integrated assessment model that analyzes the economic and health impacts of two mycotoxins in corn: fumonisin and aflatoxin. It was found that excessively strict standards of these two mycotoxins could result in global trade losses in the hundreds of millions $US annually, with the US, China, and Argentina suffering the greatest losses. The paper then discusses the evidence for Bt corn’s lower levels of contamination of fumonisin and aflatoxin, and estimates economic impacts in the United States. A total benefit of Bt corn’s reduction of fumonisin and aflatoxin in the US was estimated at $23 million annually. Finally, the paper examines the potential policy impacts of Bt corn’s mycotoxin reduction, on nations that are making a decision on whether to allow commercialization of this genetically modified crop.  相似文献

通过有性杂交手段培育出聚有不同数目Bt基因的植株,在不同生育期进行抗虫性测定和Bt毒蛋白表达的ELISA检测,旨在揭示聚合不同数目Bt基因的植株抗虫性的互作表达机理。聚合有1-4个Bt基因的植株在整个生育期的抗虫性、毒蛋白表达特性和单价抗虫棉时空表达一致,生育前期抗虫性好、毒蛋白表达量高;生育中、后期抗虫性有所下降,毒蛋白表达量降低。聚合有4个Bt基因的纯合材料并未因Bt基因的增加而起到抗性增强的效果,相反还因同源抑制而有所降低。不同来源的Bt基因处于杂合状态时其抗虫性和Bt毒蛋白量均得到充分表达。  相似文献

European corn borer larvae, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae) that have completed development on Event 176 Bt corn hybrids have survived exposure to sublethal doses of the Cry1Ab Bt toxin or are exploiting plant tissues that do not express the toxin. To evaluate the impact of such exposure, diapausing larvae were collected from Event 176 and conventional hybrids and compared for rates of pupation, parasitism, fitness (pupal weight, longevity, and fecundity) and susceptibility to the Cry1Ab toxin. Larvae completing development on Event 176 corn exhibited approximately 10% higher survival rates and correspondingly lower parasitism rates than larvae completing development on conventional hybrids. No significant differences were detected in pupal weight, fecundity, longevity or susceptibility to the Cry1Ab Bt toxin. These results indicate that survival on Event 176 corn are not adversely affect fitness and does not cause increased tolerance to the Cry1Ab toxin in subsequent generations.  相似文献

Seed storage proteins are a major component of mature seeds. They are utilized as protein sources in foods. We designed seed storage proteins containing bioactive peptides based on their three-dimensional structures. Furthermore, to create crops with enhanced food qualities, we developed transgenic crops producing seed storage proteins with bioactive peptides. This strategy promises to prevent lifestyle-related diseases by simple daily food consumption. In this review, we discuss a strategy to develop transgenic crops to improve human health by advanced utilization of seed storage proteins.  相似文献

Bt toxin genes were the insecticidal genes most widely used in genetic engineering of pest resistant plant, were of important significance to study their expression behavior in transgenic plants. In this work, a plant expression vector, pBinMoBc, was constructed. It contained the Cry IA(c) gene under control of chimeric OM promoter and the Ω factor. The vector was transferred into tobacco (Nicotiana tabacum L.) plant via Agrobacterium-mediated transformation. ELISA assay showed that the expression levels of the Cry IA(c) gene in transgenic tobacco plants were significantly higher than that in wild-type tobacco plants. The highest could be up to 0.255% of total soluble proteins; the expression level of CryIA(c) gene in transgenic tobacco plant was changeable during the development stages of tobacco plant. Bioassay showed that pBinMoBc transgenic tobacco plants had more notable insecticidal activity than the wild-type tobacco plants. The above results indicated that pBinMoBc was an effective pest-resistent plant expression vector. This study would be very helpful in screening transgenic cotton with high resistance to cotton bollworm (Heliothis armigeva Hubner).  相似文献

A mathematical model was constructed to describe the evolution of resistance to the Bacillus thuringiensis toxin (Bt) in an insect pest (European corn borer) population on a transgenic crop (Bt corn). The model comprises a set of partial differential equations of the reaction-diffusion type; local interactions of three competing pest genotypes formed by alleles of Bt resistance and susceptibility are described as in the Kostitzin model, and the spread of insects is modeled as diffusion. The model was used to evaluate the influence of pest characteristics on the efficacy of the high-dose/refuge strategy aiming to prevent or delay the spread of Bt resistance in pest populations. It was shown, by contrast, that a model based on Fisher-Haldane-Wright equations and formally incorporating a diffusion term cannot adequately describe the evolution of Bt resistance in a spatially inhomogeneous pest population. Further development of the proposed demo-genetic model is discussed.  相似文献

转Bt基因抗虫棉对亚洲玉米螟抗性的研究   总被引:6,自引:0,他引:6  
对转Bt抗虫棉苏抗103与GK—22对玉米螟抗性的研究结果显示:(1)用苏抗103嫩叶、簧,花、蕾饲曩第三代玉米螟幼虫,最终幼虫校正死亡率分别达100%、95.83%、83.33%与100%。校正化蛹下降率分别达100%、92.31%、50.07%与100%,校正幼虫成蛾减退率分别达100%、90.92%、0%与100%。(2)人工饲料中加入5个不同浓度苏抗103嫩叶饲喂第二代幼虫,除0.4%浓度,幼虫校正死亡率与成蛹下降率均为100%,0.4%浓度的幼虫期延长20.68%,蛹重下降18.24%,蛹期延长19.79%。(3)人工饲料中加入5个浓度的苏抗103或GK22嫩叶分别饲喂第三代幼虫,苏抗103上幼虫校正死亡率较第二代有所下降,分别为3.04%、-16.68%、28.2%、33.33和75%。GK22上幼虫校正死亡率分别为3.13%、26.89%、45.69%、70.816%和68.09%。其幼虫校正死亡率及幼虫期、蛹期、成虫校正减退率均有随浓度增加而上升的趋势。较亲本棚,对蛹重、雌成虫寿命、单雌产卵量、卵孵化率也有一定程度的影响。上述研究结果表明,苏抗103与GK22影响棉田玉米螟幼虫与蛹的存活与生命力,进而影响其成虫繁殖力而表现出良好抗性。  相似文献

Bt水稻对田间非靶标害虫种群动态的影响   总被引:4,自引:0,他引:4  
以转cry1Ab/cry1Ac基因水稻汕优63(以下简称汕优63/Bt)为材料,亲本汕优63(以下简称汕优63/CK)为对照,在田间自然感虫条件下研究汕优63/Bt对稻田几种非靶标害虫种群消长动态的影响。结果表明,汕优63/Bt上稻苞虫Pelopidas mathias(Fabricius)和稻眉眼蝶Mycalesis gotamaMoore幼虫数量都显著低于对照品种,但汕优63/Bt对稻苞虫的毒性强于稻眉眼蝶。汕优63/Bt上稻飞虱(白背飞虱Sogatellafurcifera(Horvath)和褐飞虱Nilaparvatalugens(Stal))混合种群数量通常与对照品种基本一致,在少数调查时间显著高于对照品种。相反,除少数调查时间与对照品种没有显著差异外,稻叶蝉(黑尾叶蝉Nephotettixcincticeps(Uhler)和二点黑尾叶蝉N.viriscens(Distant))混合种群数量通常显著高于对照品种。稻飞虱和叶蝉成为汕优63/Bt上的主要害虫。  相似文献

Detection and monitoring of insect resistance to transgenic Bt crops   总被引:5,自引:0,他引:5  
Transgenic crops expressing Bacillus thuringiensis (Bt) endotoxins havebecome one of the most important tools for managing corn and cotton insect pests in the US and other countries. The widespread adoption of transgenic Bt crops could place a high degree of selection pressure on the target insect populations and accelerate development of resistance, raising concerns about the long-term durability of Bt plants as an effective pest management tool. Conservation of Bt susceptibility in insects has become one of the most active research areas in modern agriculture. One of the key factors for a successful Bt resistance management plan is to have a cost-effective monitoring system that can provide information on. (i) the initial Bt resistance allele frequencies at low levels in field insect populations; and (ii) early shifts in Bt resistance allele frequencies so that proactive measures for managing resistance can be deployed well before field control failures. Developing such a monitoring program has been difficult because: (i) resistance traits that occur at very low frequencies are hard to detect; (ii) many factors affect the sensitivity and accuracy of a Bt resistance monitoring program; and (iii) monitoring resistance is costly. Several novel methods for detecting Bt resistance alleles developed during the last decade have made a cost-effective monitoring system possible. Future studies should focus on how to improve and standardize the methodologies for insect sampling and Bt resistance detection.  相似文献

随着转基因作物的应用和推广 ,转 Bt基因作物释放后对生态环境及其它方面产生的潜在影响越来越受到重视。分别从生物活性杀虫晶体蛋白在土壤中的残留特性、杀虫晶体蛋白对土壤中非目标生物的影响、转 Bt基因玉米植株体成分的变化、转Bt基因玉米花粉中杀虫晶体蛋白的表达特性及其在田间和马力筋叶片上的散积状况、花粉中表达的杀虫晶体蛋白对君主斑蝶的毒性、君主斑蝶幼虫暴露在 Bt花粉中的概率及综合风险评价估算等方面对转 Bt基因玉米产生的杀虫晶体蛋白与土壤生态环境的相互作用、花粉对非目标生物影响的研究现状进行了综述。通过对转 Bt基因作物生态安全性的科学评价和广泛宣传 ,以确保生物技术的健康发展。  相似文献
