This study aims at the optimisation of a microwave pretreatment for wheat straw solubilisation and anaerobic biodegradability. The maximum yield of methane production was obtained at 150 °C with an improvement of 28% compared to an untreated sample. In addition, at this temperature, the time to reach 80% of the methane volume obtained from untreated straw was about 35%. The study of ramp time and holding time at targeted temperature showed that they had no improvement effect. Thus, the best conditions are the highest heating rate for a final temperature 150 °C without any holding time. The reading of energy consumed by pretreatment and energy overproduced by pretreated samples showed that increasing tVS amount and heating rate led to a saving of energy consumption. Nevertheless, to obtain a positive energy balance, a microwave device should consume less than 2.65 kJ/gtVS.  相似文献

The performance and temporal variation of four hybrid, intermittent loading, pilot-scale vertical flow constructed wetlands (VFCWs) were tested for treating domestic wastewater of three different C/N ratios (2.5:1, 5:1, and 10:1, respectively). Two hybrid systems each consisted of the two identical VFCWs in-series, with up-up or down-down flow. The other two hybrid systems consisted of the first VFCWs (up or down flow) followed by a second VFCWs (down or up flow, respectively). The effects of combination mode, season, load level, and interactions on nutrient removal were studied in synthetic wastewater in the two-stage VFCW systems. With varying C/N ratios for influent water (from 2.5:1, 5:1 to 10:1) average removal efficiencies for the two-bed two-stage systems were as follows: COD 73-93%, TN 46-87%, TP 75-90%, and TOC 40-66%, respectively. All two-bed hybrid VFCWs were efficient in removing organics and total phosphorus, and reached the highest removal rates when the C/N ratios were 10 and 5, respectively. The hybrid systems for different flow direction beds had significantly higher performance (P < 0.05) during the wetlands operational period. Compared to the four types of hybrid VFCWs, the two-stage combination with different flow directions achieved significantly higher TN and TOC reductions (P < 0.05). The highest total nitrogen (P < 0.05) and total phosphorus reductions in down-up flow VFCWs were observed at C/N 5:1. However, for organic matter and total organic carbon, the highest COD and TOC removal rates occurred when C/N ratios were 5-10 for the down-up flow VFCWs. With appropriate control of combined mechanisms in series, the concentrations of carbon and nitrogen sources in the influent can achieve the optimal effects of nutrient removal.  相似文献

【目的】明确雷公藤Tripterygium wilfordii Hook. f.生物碱对粘虫Mythimna separata (Walker)神经系统的影响,为阐明其杀虫作用机制提供依据。【方法】采用载毒叶片法测定粘虫5龄幼虫经雷公藤总生物碱处理后体内乙酰胆碱酯酶(AChE)、Na+, K+-ATPase、Ca2+, Mg2+-ATPase、谷丙转氨酶(GPT)和谷氨酸脱羧酶(GAD)等重要神经系统酶活性及乙酰胆碱(ACh)、谷氨酸(Glu)和γ-氨基丁酸(GABA)等神经递质的含量。【结果】雷公藤总生物碱处理对粘虫5龄幼虫AChE无明显影响,麻醉期处理粘虫幼虫体内ACh相对含量与同期对照无显著差异。处理粘虫幼虫在轻度麻醉期、深度麻醉期和复苏期体内GABA和Glu含量显著升高,GABA含量分别升高了89.86%, 49.28%和20.29%,Glu含量分别升高了24.55%, 23.33%和8.13%。处理粘虫幼虫GPT活性明显受到抑制,而GAD活性无明显变化。处理明显抑制粘虫幼虫头部Na+, K+-ATPase和Ca2+, Mg2+-ATPase活性,但对中肠两种ATPase活性影响不大。【结论】研究结果有助于了解雷公藤生物碱对昆虫神经系统的影响,也为进一步阐明其作用靶标奠定了基础。  相似文献

Polarized attenuated total reflection Fourier transform infrared spectra were recorded on multilayer stacks of native gastric tubulovesicle membranes. The spectral intensity and linear dichroism were measured for average thicknesses ranging between 0 and 100 bilayers. Atomic force microscopy was used to investigate the orientation of the membranes at the top of the stack. Height profiles were obtained along randomly drawn lines and slopes were computed over various distances. Orientation distribution functions were obtained from the slopes and decomposed into Legendre polynomials. It was found that the second Legendre polynomials coefficient characterizing the membrane orientation was always larger than 0.9. It could therefore be concluded that the membrane tilt does not significantly contribute to the infrared dichroism, even for the largest thicknesses tested.  相似文献

为了解湘西特色食品“蒿菜粑粑”原料植物鼠麴草(Gnaphalium affine)总黄酮提取物体外抗氧化能力,采用DPPH、ABTS自由基清除实验,还原力实验和抑制β-胡萝卜素褪色实验等方法,测定鼠麴草总黄酮抗氧化活性。结果显示,鼠麴草总黄酮母液中总黄酮浓度为7.01 mg·mL–1mg/mL;总黄酮提取物对DPPH、ABTS自由基有较好的清除能力,其半数抑制浓度(IC50)分别为16.30 mg·L–1、30.16 mg·L–1,将胡萝卜素相对吸光度降为50%的时间延长至67.49 min,在还原能力、延缓胡萝卜素褪色和抑制脂质过氧化上也有较好效果。鼠麴草总黄酮提取物具有良好的体外抗氧化活性,可作为优质食用植物资源进一步开发与推广。  相似文献

Redox-induced protonation state changes of the Glu residue in the multicopper oxidases, CueO and bilirubin oxidase (BO), were studied by attenuated total reflectance-Fourier transform infrared spectroscopy. By monitoring IR bands of the carboxylic acid CO stretch in the wild-type and Glu-to-Gln mutant enzymes the Glu506 of CueO (Glu463 of BO) was found to be unprotonated in the oxidised and protonated in the reduced forms. The results provided direct evidence for proton uptake by the Glu, suggesting it plays a key role in the proton donation to the activated oxygen species in the catalytic cycle.  相似文献

Changes in cell shape that occur in many cellular processes are thought to arise from polymerization of actin filaments near the cell membrane. End-to-end annealing of actin filaments is believed to play only a minor role in this process, as annealing in solution was shown to be a slow process, which is not typical for a bimolecular reaction, its rate constant decreasing over time, being inversely proportional to the filament length. Furthermore, in vitro studies on f-actin solutions were found to display an exponential steady-state length distribution. In the cell, many physiologically important parameters, such as mechanical strength or viscoelastic response are a direct function of the physical properties of the underlying actin cytoskeleton, such as actin filament length distribution and dynamics. How the underlying physical parameters of the actin cytoskeleton may be influenced by the cell surface or molecular crowding remains poorly understood. Using total internal reflection fluorescence (TIRF) microscopy we reinvestigated actin end-to-end annealing in vitro in a more realistic environment. We studied the process near a hydrophilic surface together with crowding agents, in order to mimic the physiological media near the cell membrane, which has substantial amounts of macromolecules present. We find that actin end-to-end annealing changes in three ways near a crowded hydrophilic surface as compared to solution. First the annealing rate becomes a factor of 20 faster than in solution. Second the rate of annealing becomes typical of a bimolecular reaction, shows no length dependence and is basically just a function of the square of the concentration of ends. Lastly the length distribution is Gaussian throughout the entire annealing process. This implicates that dynamic rearrangement of actin filaments by annealing near the leading edge of the cell, could change physical parameters like the mechanical response and contribute significantly to cell motility.  相似文献

Sac7d and Sso7d are homologous, hyperthermophile proteins with a high density of charged surface residues and potential ion pairs. To determine the relative importance of specific amino acid side-chains in defining the stability and function of these Archaeal chromatin proteins, pK(a) values were measured for the acidic residues in both proteins using (13)C NMR chemical shifts. The stability of Sso7d enabled titrations to pH 1 under low-salt conditions. Two aspartate residues in Sso7d (D16 and D35) and a single glutamate residue (G54) showed significantly perturbed pK(a) values in low salt, indicating that the observed pH-dependence of stability was primarily due to these three residues. The pH-dependence of backbone amide NMR resonances demonstrated that perturbation of all three pK(a) values was primarily the result of side-chain to backbone amide hydrogen bonds. Few of the significantly perturbed acidic pK(a) values in Sac7d and Sso7d could be attributed to primarily ion pair or electrostatic interactions. A smaller perturbation of E48 (E47 in Sac7d) was ascribed to an ion pair interaction that may be important in defining the DNA binding surface. The small number (three) of significantly altered pK(a) values was in good agreement with a linkage analysis of the temperature, pH, and salt-dependence of folding. The linkage of the ionization of two or more side-chains to protein folding led to apparent cooperativity in the pH-dependence of folding, although each group titrated independently with a Hill coefficient near unity. These results demonstrate that the acid pH-dependence of protein stability in these hyperthermophile proteins is due to independent titration of acidic residues with pK(a) values perturbed primarily by hydrogen bonding of the side-chain to the backbone. This work demonstrates the need for caution in using structural data alone to argue the importance of ion pairs in stabilizing hyperthermophile proteins.  相似文献

Heme-copper oxygen reductases catalyze proton translocation across the cellular membrane; this takes place during the reaction of oxygen to water. We demonstrate with attenuated total reflection-Fourier transform infrared (ATR-FTIR) difference spectroscopy that a tyrosine residue of the oxygen reductase from the thermohalophilic Rhodothermus marinus becomes deprotonated in the transition from the oxidized state to the catalytic intermediate ferryl state P(M). This tyrosine residue is most probably Y256, the helix VI tyrosine residue proposed to substitute for the D-channel glutamic acid that is absent in this enzyme. Comparison with the mitochondrial like oxygen reductase from Rhodobacter sphaeroides suggests that proton transfer from a strategically situated donor to the active site is a crucial step in the reaction mechanism of oxygen reductases.  相似文献

Plant responses to water deficit occur in a complex framework of organ interactions, but few studies focus on the effect of drought stress on all organs in a whole-plant. The effects of repeated dehydration and rehydration (DH) on physiological and biochemical responses in various organs of Periploca sepium Bunge (P. sepium) were investigated. The leaf relative water content decreased significantly during drought, but recovered and showed an increase when compared to well-watered control plants. The malondialdehyde (MDA) content increased in mature and old leaves, but decreased in young leaves, new stems and fine roots during drought, indicating that the young and vigorous tissues of a whole-plant are protected preferentially from the oxidative stress. Among all organs, the fine roots showed the highest levels of proline, total free amino acids (TFAA) and Na+, while the leaves showed the highest levels of total soluble sugars (TSS), soluble proteins (SP), Ca2+ and Mg2+. The response to DH differed in different organs, both in magnitude and in the type of solutes involved. Drought stress increased the contents of proline, TFAA, TSS, SP and K+ in all organs of P. sepium plants, while the accumulation amounts were obviously different among the organs. The storage starch in stems and roots plays an important role in providing carbohydrates for growth. Changes in Na+, Ca2+ and Mg2+ under DH presented a high degree of organ specificity. Our data indicates that response strategies are different between different organs; therefore, evidence the needs to integrate all the information in order to better understand plant tolerance mechanisms.  相似文献

The bacterial flagellar motor is a rotary motor driven by the electrochemical potentials of specific ions across the cell membrane. Direct interactions between the rotor protein FliG and the stator protein MotA are thought to generate the rotational torque. Here, we used total internal reflection fluorescent microscopy to observe the localization of green fluorescent protein (GFP)-fused FliG in Escherichia coli cells. We identified three types of fluorescent punctate signals: immobile dots, mobile dots that exhibited simple diffusion, and mobile dots that exhibited restricted diffusion. When GFP-FliG was expressed in a DeltafliG background, most of the cells were not mobile. When the cells were tethered to a glass side, however, rotating cells were commonly observed and a single fluorescent dot was always observed at the rotational center of the tethered cell. These fluorescent dots were likely positions at which functional GFP-FliG had been incorporated into a flagellar motor. Our results suggest that flagellar basal bodies diffuse in the cytoplasmic membrane until the axial structure and/or other structures assemble.  相似文献

Searching for improved indolesulfonamides with higher polarities, 45 new analogues with modifications on the sulfonamide nitrogen, the methoxyaniline, and/or the indole 3-position were synthesised. They show submicromolar to nanomolar antiproliferative IC50 values against four human tumour cell lines and they are not P-glycoprotein substrates as their potencies against HeLa cells did not improve upon cotreatment with multidrug resistance (MDR) inhibitors. The compounds inhibit tubulin polymerisation in vitro and in cells, thus causing a mitotic arrest followed by apoptosis as shown by cell cycle distribution studies. Molecular modelling studies indicate binding at the colchicine site. Methylated sulfonamides were more potent than those with large and polar substitutions. Amide, formyl, or nitrile groups at the indole 3-position provided drug-like properties for reduced toxicity, with Polar Surface Areas (PSA) above a desirable 75 Å2. Nitriles 15 and 16 are potent polar analogues and represent an interesting class of new antimitotics.  相似文献

Objective

To describe the development and first results of a dedicated chronic total occlusion (CTO) programme in a tertiary medical centre.

Background

Because of the complexity and the increased risk of complications during percutaneous coronary intervention (PCI) for CTO, it is essential that less experienced and evolving CTO centres perform regular quality analyses.

Methods

We therefore performed analyses to describe the results during the first 3 years of a dedicated CTO programme at a high-volume PCI centre. In addition, we discuss the strategies employed to develop such a programme.

Results

A total of 179 consecutive patients undergoing 187 CTO procedures were included in the study. The complexity of the CTO lesions increased from a mean J‑CTO (Japanese Multicentre CTO Registry) score of 1.3 in 2015 to 2.1 in 2017. In the majority of cases, the antegrade wire escalation technique was performed. Final technical success rate was 78.5% in 175 patients with a single CTO and 80.2% of all 187 CTO procedures. No peri-procedural or in-hospital deaths occurred. One peri-procedural myocardial infarction occurred. Cardiac tamponade occurred in 2 cases, both managed by pericardiocentesis. No urgent cardiac surgery was necessary. Survival and revascularisation rates at 30 days and 1 year were excellent.

Conclusion

Following initiation of a dedicated CTO programme, using up-to-date techniques and strategies, procedural and clinical outcome were comparable with current standards in established centres.

Dietary cholesterol and aging are major risk factors to accelerate oxidation process for developing hypercholesterolemia. The major aim of this study is to elucidate the effects of rice protein on cholesterol level and oxidative stress in adult rats fed with and without cholesterol. After 2 weeks of feeding, hepatic and plasma contents of cholesterol, reduced glutathione (GSH), oxidized glutathione (GSSG), malondialdehyde (MDA) and protein carbonyl (PCO) were measured. In liver, total antioxidative capacity (T-AOC), activities of antioxidant enzymes (total superoxide dismutase, T-SOD; catalase, CAT), glutathione metabolizing enzyme activities and gene expression levels (γ-glutamylcysteine synthetase, γ-GCS; glutathione reductase, GR; glutathione peroxidase, GPx) were determined. Under cholesterol-free/enriched dietary condition, T-AOC, activities of T-SOD and CAT, glutathione metabolism related enzymes' activities and mRNA levels (γ-GCS, GR and GPx) were effectively stimulated by rice proteins as compared to caseins. Compared with caseins, rice proteins significantly increased hepatic and plasma GSH contents, whereas hepatic and plasma accumulations of MDA, PCO and GSSG were significantly reduced by rice protein-feedings. As a result, the marked reductions of cholesterol in the plasma and in the liver were observed in adult rats fed rice proteins with and without cholesterol. The present study demonstrates that the hypocholesterolemic effect of rice protein is attributable to inducing antioxidative response and depressing oxidative damage in adult rats fed cholesterol-free/enriched diets. Results suggest that the antioxidant capability involved in the hypocholesterolemic action exerted by rice protein is independent of dietary cholesterol during adult period.  相似文献

为研究牡荆叶指纹图谱与抗氧化活性的谱-效关系,该研究首先建立了18批牡荆叶的高效液相色谱-电化学检测法(HPLC-ECD)指纹图谱,对不同来源牡荆叶药材进行聚类分析,鉴定主要酚类化合物且测定其含量,分析牡荆叶的总酚和总黄酮含量,并采用DPPH自由基清除法、ABTS自由基清除法、氧自由基吸收能力法及铁离子还原能力法考察其体外抗氧化活性,通过皮尔逊相关分析、灰度关联分析及偏最小二乘回归分析法研究牡荆叶的谱-效关系。结果表明:(1)牡荆叶的指纹图谱标定21个共有峰,共指认出10个峰,其含量顺序为绿原酸>异荭草苷>木犀草苷>异牡荆素>异绿原酸A>异绿原酸C>原儿茶酸>荭草苷>异绿原酸B>新绿原酸;不同产地样品间相似性较高,相似度结果为0.816~0.983。(2)系统聚类分析显示,样品含量对分类有一定影响,不同来源样品被分为3类,其中南北方样品存在一定差异。(3)牡荆叶中总酚和总黄酮的含量分别为15.82~61.83 mg·g-1和27.85~157.65 mg·g-1,样品均具不同程度的抗氧化活...  相似文献

In this contribution we present the design of an original Attenuated Total Reflection (ATR)-based device designed for an IR microscope coupled to a FPA detector and optimized for in-vivo cell imaging. The optical element has been designed to perform real time experiments of cell biochemical processes. The device includes a manually removable Ge-crystal that guarantees an ease manipulation during the cell culture and a large flat surface to support the cell growth and the required change of the culture wells. This layout will allow performing sequential ATR IR imaging with the crystal immersed in the culture wells, minimizing contributions due to water vapors in the optical system. Using existing brilliant synchrotron radiation sources this ATR device may collect images at the surface of the Ge crystal at a sub-cellular spatial resolution with a penetration depth of the evanescent wave inside the sample of ~ 500 nm within few seconds. A brief summary of the cellular components that should be detected with such optical device is also presented.  相似文献

To explore how the sulfonylurea receptor (SUR1) is involved in docking and fusion of insulin granules, dynamic motion of single insulin secretory granules near the plasma membrane was examined in SUR1 knock-out (Sur1KO) beta-cells by total internal reflection fluorescence microscopy. Sur1KO beta-cells exhibited a marked reduction in the number of fusion events from previously docked granules. However, the number of docked granules declined during stimulation as a consequence of the release of docked granules into the cytoplasm vs. fusion with the plasma membrane. Thus, the impaired docking and fusion results in decreased insulin exocytosis from Sur1KO beta-cells.  相似文献

During fatty acid oxidation by rat liver mitochondria, the rate of β-oxidation is dependent on the relative amounts of substrate and mitochondrial protein, on the energy state of the mitochondria, on the chain length and the number of double bonds of the fatty acid and on the concentration of various compounds in the reaction medium (l-carnitine, CoASH, hexokinase, albumin).2. The rate of β-oxidation of long-chain fatty acids decreases when the ratio of albumin over fatty acid is increased. This effect is most marked in the absence of added carnitine.3. Addition of excess hexokinase decreases the rate of β-oxidation in the presence of added carnitine.4. Maximal rates of β-oxidation are observed with octanoate and decanoate (40–60 nmoles acetyl-CoA/min per mg mitochondrial protein at 25 °C).5. Odd-numbered fatty acids are oxidized at a much lower rate than the even-numbered homologues. In a low-energy state propionyl-CoA accumulates; in a high-energy state in the presence of bicarbonate, Krebs-cycle intermediates accumulate.6. l-Carnitine enhances the rate of β-oxidation of all fatty acids except butyrate. The stimulatory effect is most pronounced with odd-numbered and with long-chain fatty acids.7. In the absence of added carnitine the rate of β-oxidation of long-chain fatty acids decreases with the chain length and increases with the number of double bonds. It is suggested that the solubility of the long-chain fatty acids in the aqueous medium is the rate-limiting factor under these conditions.8. In the presence of carnitine and albumin, palmitate, oleate, linoleate and linolenate are all oxidized at about the same rate (25–30 nmoles/min per mg protein at 25 °C).9. Propionyl-CoA is not formed as an intermediate during oxidation of unsaturated fatty acids.  相似文献

RNAs in biological processes often interconvert between defined structures. These RNA structure conversions are assisted by proteins and are frequently coupled to ATP hydrolysis. It is not well understood how proteins coordinate RNA structure conversions and which role ATP hydrolysis has in these processes. Here, we have investigated in vitro how the DEAD-box ATPase Ded1 facilitates RNA structure conversions in a simple model system. We find that Ded1 assists RNA structure conversions via two distinct pathways. One pathway requires ATP hydrolysis and involves the complete disassembly of the RNA strands. This pathway represents a kinetically controlled steady state between the RNA structures, which allows formation of less stable from more stable RNA conformations and thus RNA structure conversion against thermodynamic equilibrium values. The other pathway is ATP-independent and proceeds via multipartite intermediates that are stabilized by Ded1. Our results provide a basic mechanistic framework for protein-assisted RNA structure conversions that illuminates the role of ATP hydrolysis and reveal an unexpected diversity of pathways.  相似文献
