The pigments contained in the inner seed coats of Cucurbitapepo, C. maxima and C. moschata were investigated spectroscopicallyusing both intact tissues and their ether extracts. The pigment(s) contained in C. pepo and C. maxima was inferred to be "protochlorophyll"and/or Mg vinyl pheoporphyrin a5 which are indistinguishablefrom each other by mere spectroscopic observations. The coatsof C. moschata were found to contain Mg-free vinyl pheoporphyrina5 which has so far been reported to exist only in some Chlorellamutants. The spectra of these pigments in intact tissues wereremarkably different from those of the ether extracts, indicatingthe profound difference in their physical states in vitro andin vivo. Application of ethephon to field-grown plants of both bush andtrailing forms of Cucurbita maxima and C. pepo caused leaf epinasty,suppression of male flowers and earlier production and increasein numbers of female flowers. This gave rise to an increasein the ratio of female to male flowers per plant and a decreasein the total number of flowers. Observations of C. pepo showed that even at the two true leafstage there are several nodes present in the unexpanded shoot.Each node has one main and several secondary buds. The sex ofthe main bud at the first five to six nodes is usually determinedat this stage but the secondary buds still have bisexual potential.The change in sex expression was brought about by all male flowerbuds that had formed by the spraying time aborting, and allbuds that developed (both main and secondary) for at least 7days after spraying became female flowers. Thus, nodes fiveand six had male flowers in the controls, whereas in ethephon-sprayedplants the presumptive male flowers aborted at the bud stageat these nodes and secondary primordia developed into functionalfemale flowers. Alloteropsis semialata (R.Br.) Hitchcock is a widely distributedgrass species known to show marked morphological, anatomicaland physiological variation. An unusual feature of this grassis that it shows both C3, and C4 photosynthetic pathways withthe respective anatomy related to these pathways in one speciesand within a single environment. This study investigates ultrastructuraldifferences between C3 and C4 forms of A. semialata, and showsdistribution of chlorenchyma in the two forms, at light microscopelevel. Quantitative assessment shows marked differences in mesophylland Kranz sheath cells with regard to size and organelle content;also in distribution of starch grains, lipid droplets and organellesin the mesophyll cells of both forms. These differences arediscussed in relation to the C3/C4 syndrome. Measurements were made using a Digiplan electronic planimeteron transmission electron micrographs of mature leaves sectionedtransversely at mid-lamina, and on drawings of leaf sectionsshowing the distribution of chlorenchymatous tissue. Fry, S. C. 1987. Formation of isodityrosine by peroxidase isozymes.—J.exp. Bot. 38: 853–862. Tyrosine residues of extensin are oxidatively coupled in vivoto form isodityrosine bridges, whereas treatment of purifiedextensin with H2O2+ peroxidase in vitro yields only dityrosine.Two explanations for the correct mode of coupling in vivo weretested. The first, that the pH of the cell wall is lower thanthat (pH 9-0) at which in vitro experiments have been conducted,provided part of the answer since treatment of L-tyrosine withH2O2+peroxidase in vitro at pH 37–5 yielded some isodityrosine.The second, that the wall contains other isozymes of peroxidasethan the basic isozyme usually studied in vitro, appeared unlikelybecause several sharply contrasting isozymes yielded similarisodityrosine: dityrosine ratios from L-tyrosine+ H2O2 at anygiven pH. The isozymes were also similar in their ability tooxidize tyrosine-dimers further to higher polymers. It is concludedthat the formation of isodityrosine in vivo is dictated by neighbouringwall molecules, possibly ionically-bound pectins, which modifythe local environment of the tyrosine residues of extensin. The relationship between high temperature stress injury andtemperature dependence of the transverse relaxation time (T2)of leaf water was examined using NMR in four cultivars of wheatdiffering in their sensitivity to high temperature stress. TheT2declined with increasing temperature between 25 and 35 °C.A comparison of relative injury based on electrolyte leakageand T2, between 40 and 50 °C, indicated that while membranepermeability increased with increasing temperature there wasan increase in T2until 44 and 48 °C in susceptible and tolerantcultivars respectively, followed by a sharp decline. This patternof change in T2with increasing temperature was consistent whetherthe same or different samples were used for each treatment temperature.Loss of temperature dependence of T2after heat killing indicatedirreversible changes in T2, probably due to the loss of membraneintegrity. Heat tolerant varieties, which suffered less membraneinjury, had a higher T2compared to susceptible varieties. Tolerantvarieties also maintained the T2of leaf water protons to highertemperatures than did sensitive varieties. This NMR-based, non-invasive,rapid technique could be used to efficiently detect heat injuryin leaf tissues. Deoxygibberellin C (DGC), a C/D ring-rearranged isomer of GA20,was shown to inhibit the conversion of [2,3-3H2]GA9 to [2-3H]GA4by gibberellin 3ß-hydroxylase from immature seedsof Phaseolus vulgahs. Deoxygibberellin C inhibited the promotionof growth by exogenously applied GA20 of rice (Oryza sativaL.) seedlings. Evidence is also presented that DGC is a competitiveinhibitor of the 3ß-hydroxylase from P. vulgaris.However, DGC only weakly inhibited the conversion catalyzedby the 3ß-hydroxylase from Cucurbita maxima at highconcentrations, and it did not inhibit the promotion of growthby exogenously applied GA9 of cucumber (Cucumis sativus) seedlings.These results suggest that the 3ß-hydroxylases fromP. vulgaris and C. maxima have different structural requirementswith respect to their substrates. 16-Deoxo-DGC also inhibitedcatalysis of the same conversion by 3ß-hydroxylasefrom P. vulgaris, and it slightly inhibited the conversion catalyzedby the enzyme from C. maxima. Glutathione S-transferases (GSTs; EC 2.5.1.18 [EC] ) in sarcocarptissue of pumpkin (Cucurbita maxima Duch.) fruit and in callusinduced from the tissue were examined. The specific activityof GST in the callus was 6.9-fold higher than that in the tissue.The specific activity in the callus remained constant duringcultivation. Column chromatography on DEAE-cellulose, hydroxylapatite,and S-hexylglutathione-agarose was used to fractionate solubleproteins that were precipitated by ammonium sulfate at 30% to70% saturation from homogenates of the sarcocarp tissue of pumpkinfruit and the callus and GST activity was monitored. Two andseven isozymes of GST were identified in the tissue and in thecallus, respectively. Furthermore, column chromatography onSephadex G-200 and SDS-polyacrylamide gel electrophoresis, indicatedthat these GST isozymes were homo- and heterodimers of subunitsof Mr 22,000 (Puga), and 23,000 (Pugb), 24,000 (Pugc) or 24,500(Pugd). Puga and Pugb were predominant in the sarcocarp tissueand in the callus, respectively. Puga, Pugb, Pugc and Pugd hadacidic pI values of 5.45, 5.00, 5.35 and 5.75, respectively.Rabbit antiserum against Pugb did not cross-react with the threeother subunits of GST during immunoblotting. In 4 cultivars of tomato (Lycopersicon esculentum Mill.), theearly detachment of fruits advanced ripening and considerablyreduced the threshold value of endogenous C2H4. This indicatesa supply from the vegetative parts of (a) labile ripening-inhibitingsubstance(s) antagonizing the action of C2H4. The endogenous level of CO2 increased shortly after the risein C2H4, and maximum levels of C2H4 and CO2 occurred almostsimultaneously. The activity of PE showed no connection with ripening, but PGactivity did not occur until the onset of ripening. However,this activity increased at considerably higher C2H4 concentrationsthan the rise in WSP, and was independent of the possible presenceof ripening inhibitor(s). Hence PG is considered not to be involvedin the primary events leading to fruit ripening. Exposure of fruits to different C2H4 concentrations in the ambientatmosphere also showed PG activity to increase only after therise in WSP had started. Other pectin degrading or synthesizingenzymes may be involved. In the non-ripening Rin mutant of cv. The kinetic properties of phosphoenolpyruvate (PEP) carboxylasehave been studied among several Flaveria species: the C3 speciesF. cronquistii, the C3–C4 species F. pubescens and F.linearis, and the C4 species F. trinervia. At either pH 7 or8, the maximum activities (in µmol.mg Chl–1.h–1)for F. pubescens and linearis (187–513) were intermediateto those of the C3 species (12–19) and the C4 species(2,182–2,627). The response curves of velocity versusPEP concentration were hyperbolic for the C3 and C3–C4species at either pH 7 or 8 while they were sigmoidal for theC4 species at pH 7 and hyperbolic at pH 8. The Km values forPEP determined from reciprocal plots were lowest in the C3 species,and of intermediate value in the C3–C4 species comparedto the K' values of the C4 species determined from Hill plotsat either pH 7 or 8. Glucose-6-phosphate (G6P) decreased theKm values for PEP at both pH 7 and 8 in the C3 and C3–C4species. In the C4 species, G6P decreased the K' values at pH8 but increased the K' values at pH 7. In all cases, G6P hadits effect by influencing the activity at limiting PEP concentrationswith little or no effect on the maximum activity. At pH 8 andlimiting concentrations of PEP the degree of stimulation ofthe activity by G6P was greatest in the C4 species, intermediatein F. linearis, a C3–C4 species, and lowest in the C3species. Seeds of two lettuce cultivars (Lactuca sativa L., cv. Meikoninginand cv. Grand Rapids) were hermetically stored with constantmoisture contents ranging between 3.6 and 17.9 per cent (freshweight basis) at constant temperatures ranging between 5 and75 °C. The decline with time in percentage germination andpercentage normal seedlings was determined for each storagetreatment. The data were fitted to an equation which containsthe constants: K1, the probit of the initial percentage germinationor normal seedlings; KE, a species constant; CW, the constantof a logarithmic moisture term; CH, the constant of a lineartemperature term and CQ, the constant of a quadratic temperatureterm. Regression analysis of data from storage periods up to5.5 years at temperatures of 5–75 °C and seed moisturecontents of 3.6–13.6 per cent yielded the following values:KE= 8.218, CW=4.797±0.163, CH=0.0489±0.0050 andCQ=0.000365±0.000056. Although this equation consistentlyprovided a better fit, simplified equations, assuming eithera log-linear relationship between seed longevity and temperature,or a log-linear relationship between seed longevity and bothmoisture content and temperature, accounted for more than 94per cent of the variation at the restricted temperature rangeof 5–40 °C. Longevity of the same seed lots at sub-zero temperatures (–5,–10 and –20 °C) was studied in separate tests.Freezing damage, resulting in abnormal seedlings in the germinationtest, occurred at –20 °C when the moisture contentof the seeds exceeded 12 per cent. No decline in percentagenormal seedlings was observed after a storage period of 18 monthsor longer at –20 °C, provided the seed moisture contentdid not exceed 9.5 per cent. For seeds stored at –5 and–10 °C with 9.6–12.5 per cent moisture content,the observed rate of decline of percentage normal seedlingswas adequately predicted by the viability equation, using theabove values for the constants. This suggests that for low moisturecontents the viability equation can be applied to estimate longevityat sub-zero temperatures. Mitochondria were isolated from mesophyll protoplasts and bundlesheath protoplasts or strands which were obtained by enzymaticdigestion of six C4 species: Zea mays, Sorghum bicolor, Panicummiliaceum, Panicum capillare, Panicum maximum and Chloris gayana,representative of three C4 types. Photorespiratory glycine oxidationand related enzyme activities of mesophyll and bundle sheathmitochondria were compared. Mesophyll mitochondria showed good P/O ratios with malate andsuccinate as substrate but lacked the ability to oxidize glycine.On the other hand, mitochondria isolated from bundle sheathprotoplasts of P. miliaceum and bundle sheath strands of Z.mays possessed glycine oxidation activity similar to that ofmitochondria from C3 plant leaves. The two enzymes involvedin glycine metabolism in mitochondria, serine hydroxymethyltransferaseand glycine decarboxylase, were also assayed in the mitochondriaof the two cell types. The activities of the two enzymes inbundle sheath mitochondria were in the range found in C3 mitochondria.In contrast, the activities in mesophyll mitochondria were eithernot detectable or far lower than those in bundle sheath mitochondriaand ascribed to contaminating bundle sheath mitochondria. The present results indicate the deficiency of a complete glycineoxidation system in mesophyll mitochondria and also a differentiationbetween mesophyll and bundle sheath cells of C4 plants withrespect to the photorespiratory activities of the mitochondria. The influence of temperature on photosynthesis and transpirationwas studied in ten varieties of Lolium perenne, L. multiflorum,Dactylis glomerata, and Festuca arundinacea from three climaticorigins grown in three different controlled environments (15?C, 72 W m-2 visible irradiation, 16-h photoperiod; 25 ?C, 72W m-2 visible irradiation, 16-h photoperiod; and 25 ?C, 180W m-2 visible irradiation, 16-h photoperiod) and in the glasshousein July/August. The optimum temperature for photosynthesis was influenced primarilyby growth environment; growth at low temperature (15 ?C) resultedin a low optimum temperature, which differed little from varietyto variety. The maximum CO2-exchange rate was influenced bygrowth environment and by variety. Within a variety, plantsgrown at higher light intensity or lower temperature had a greaterCO2-exchange rate. Seven varieties showed a negative correlationbetween the optimum leaf temperature and the maximum CO2-exchangerate. Activation energies for photosynthesis were influencedby growth environment only. There were marked varietal differences in the values of leafresistances (ra + rt) obtained from transpiration data at theoptimum leaf temperature for CO2 exchange. In Lolium, and Dactylisthe Mediterranean varieties had higher leaf resistances thanthe Northern varieties with the maritime varieties intermediate.In general the Dactylis varieties had higher resistances thanthe corresponding Lolium and Festuca varieties. Only at highgrowth temperatures was (ra+rl) insensitive to temperature;otherwise an activation energy of about 10 kcal/mole was observed.A negative correlation was found between mean varietal diffusionresistances (ra+rl), and corresponding maximum CO2-exchangerates.  相似文献

Understanding temporal niche separation between C3 and C4 species(e.g. C3 species flourishing in a cool spring and autumn whileC4 species being more active in a hot summer) is essential forexploring the mechanism for their co-existence. Two parallelpot experiments were conducted, with one focusing on water andthe other on nitrogen (N), to examine growth responses to wateror nitrogen (N) seasonality and competition of two co-existingspecies Leymus chinensis (C3 grass) and Chloris virgata (C4grass) in a grassland. The two species were planted in eithermonoculture (two individuals of one species per pot) or a mixture(two individuals including one L. chinensis and one C. virgataper pot) under three different water or N seasonality regimes,i.e. the average model (AM) with water or N evenly distributedover the growing season, the one-peak model (OPM) with morewater or N in the summer than in the spring and autumn, andthe two-peak model (TPM) with more water or N in the springand autumn than in the summer. Seasonal water regimes significantlyaffected biomass in L. chinensis but not in C. virgata, whileN seasonality impacted biomass and relative growth rate of bothspecies over the growing season. L. chinensis accumulated morebiomass under the AM and TPM than OPM water or N treatments.Final biomass of C. virgata was less impacted by water and Nseasonality than that of L. chinensis. Interspecific competitionsignificantly decreased final biomass in L. chinensis but notin C. virgata, suggesting an asymmetric competition betweenthe two species. The magnitude of interspecific competitionvaried with water and N seasonality. Changes in productivityand competition balance of L. chinensis and C. virgata undershifting seasonal water and N availabilities suggest a contributionof seasonal variability in precipitation and N to the temporalniche separation between C3 and C4 species. In species of Clusia, switching from C3-photosynthesis (C3-PS)to crassulacean acid metabolism (CAM) may be a means of optimizingwater use, plant carbon balance and photon utilization duringperiods of stress. We ask whether, in perennial species of Clusia,the switch from CAM back to C3-PS is also of ecophysiologicalsignificance. Our objective was to investigate the performanceof C. minor L. during a short-term shift from CAM to C3-PS.During the transition from CAM to C3-PS, nocturnal malate andcitrate accumulation decreased whereas CO2uptake increased duringthe daytime. However, after 7 d, marked nocturnal accumulationof citrate and 24 h CO2uptake occurred. Enzymes of the C4, C3 pathway and photorespiration have beenanalyzed for P. hians and P. milioides, which have chlorenchymatousbundle sheath cells in the leaves. On whole leaf extracts thelevels of PEP carboxylase are relatively low compared to C4species, RuDP carboxylase is typical of C3 species, and enzymesof photorespiratory metabolism appear somewhat intermediatebetween C3 and C4. Substantial levels of PEP carboxylase, RuDPcarboxylase, and photorespiratory enzymes were found in bothmesophyll and bundle sheath cells. Low levels of C4-acid decarboxylatingenzymes may limit the capacity for C4 photosynthesis in P. hiansand P. milioides. The peroxidase activities of developing pea seedlings were determinedwith several substrates including three phenolic compounds,eugenol, caffeic acid and ferulic acid, which are possible precursorsin the biosynthesis of lignin. Column chromatography of thereaction products of peroxidase with caffeic and ferulic acidsindicates the formation of larger molecular weight complexesof these substrates. The peroxidase isozymes of peas were shownto be heterogenous both in molecular weight and in substratespecificity. Apparent Km determinations of two isolated isozymesindicate differences in affinities for various substrates. Starchgel zymograms with two different substrates also indicate largedifferences in staining intensities of the different isozymes.The observed pattern of changes in peroxidase level in the developingpea seedling differed according to substrate. For example, whencaffeic acid is the hydrogen donor, a large increase in activitywas observed in the 6th to 8th day of germination. This peakof activity was not observed with other substrates. Photosynthetic and photorespiratoryenzymes in widely divergent plant species with special referenceto the moss Ceratodon purpureus: Properties of ribulose bisphosphatecarboxylase/oxygenase, phosphoenolpyruvate carboxylase and glycolateoxidase.—J. exp. Bot. 36: 1677–1684. Km(CO2) values and maximal velocities of ribulose bisphosphatecarboxylase/oxygenase (E.C. 4.1.1.39 [EC] ) were determined for sixplant species growing in the wild, consisting of a moss, a fernand four angiosperms. The maximum velocities of the RuBP carboxylasesvaried from 0.13 to 0.;62 µmol CO2 fixed min–1 mg–1soluble protein and the Km(CO2) values from 15 to 22 mmol m–3CO2. The highest Km(CO2) values found were for the moss, Ceratodonpurpureus, and the grass, Deschampsia flexuosa. These plantsalso had the highest ratios of the activities of RuBP carboxylaseto RuBP oxygenase. Glycolate oxidase (E.C. 1.1.3.1 [EC] ) activitieswere slightly lower in D.flexuosa, but not in C. purpureus,than for typical C3 species. Phosphoenolpyruvate carboxylase(E.C. 4.1.1.31 [EC] ) was not involved in the photosynthetic carboxylationby these two plants. However, another grass, Phragmites australis,was intermediate in PEP carboxylase activity between C3 andC4 plants The properties of RuBP carboxylase/oxygenase are discussedin relation to the activities of PEP carboxylase and glycolateoxidase and to the internal CO2 concentration. The lower limit to the negative logarithmic relation betweenseed longevity and moisture content was determined in threesubspecies of rice (Oryza satwa L.) by storing seeds of fivecultivars at 65 °C with 11 different moisture contents (1.5–15.3%f. wt) for various periods up to 150 d and then testing forgermination. The estimates of the low-moisture-content limit(mc) were 4.3% for subsp. indica, 4.4% for subsp. japonica,and 4.5% for subsp. javanica. These moisture contents were inequilibrium with 10.5—12.0% r.h. No significant effectof moisture content between 1.5% and mc on longevity was detected(P > 0.05), while between mc and 15.3% there were negativelogarithmic relations between longevity and moisture content.There were no significant differences in the relations betweenlongevity and moisture either above or below mc between thetwo japonica cultivars or between the two javanica cultivars(P > 0.10). There was also no significant difference in theslope of the negative logarithmic relation between longevityand moisture above mc among the three subspecies (P > 0.25).However, there were significant differences in the standarddeviation of the frequency distribution of seed deaths in timeat any one moisture content, both above and below mc; this isa measure of seed longevity which is independent of pre-storageenvironment, and the differences observed show that there aregenetically determined differences in longevity among the threesubspecies (P < 0.005), indica being the longest and japonicathe shortest lived. The results provide no evidence for intra-specificvariation in mc and support the view that the maximum seed storagemoisture content which provides the maximum longevity is thatwhich is in equilibrium with about 10–11% r.h. It is concludedthat while the seed viability constant Cw of the seed viabilityequation is species specific and therefore applies to most,if not all, cultivars of rice, variation in the value of KEis the source of the differences in potential longevity of thethree subspecies. High O2 tensions, CO4, C2H4 and high temperatures were effectivenot only in breaking the dormancy of cocklebur (Xanthium pennsylvanicumWallr.) seeds but also in increasing the germination potentialof the nondormant but small seeds. There were few qualitativedifferences in response to these factors between the dormantand impotent seeds. Unlike CO2, however, enriched O2 and C2H4were stimulative even at the low temperature of 13°C. Germination induced by CO2, C2H4 and high temperature treatmentswas lowered when endogenously evolved C2H4 or CO2 was removed,whereas the effect of O2 enrichment was not affected by theirremoval. CO2 and high temperatures remarkably stimulated C2H4production, whereas O2 enrichment had no such effect. C2H4 productivity was lower in the dormant than non-dormantseeds, suggesting that the after-ripening is characterized byincreasing C2H4 production. (Received August 20, 1974; )  相似文献
