This is the second of two articles that empirically details the working practices of regulators charged with overseeing biological research. The first article considered how regulators from the Biological Agents Unit of the UK Health and Safety Executive went about implementing the British legislation controlling work with biological agents and genetically modified organisms prior to the terrorist attacks of 11 September 2001 and the ensuing anthrax letters in the U.S. This second article continues the investigation of the Biological Agents Unit's working practices by considering how its activities have changed and adapted to the new, post-9/11 policy and regulatory environment. The main argument put forward in the articles is that an understanding of the implementation processes--that is, the strategies adopted by regulatory agencies and the styles employed by agency regulators in their interactions with those regulated--is critical to developing a coherent policy logic for the emerging regulatory regime around biorisks.  相似文献

Significant advances over the last few years have seen plant-made pharmaceuticals (PMPs) move from the exploratory research phase towards clinical trials, with the first commercial products for human use expected to reach the market by 2009. Europe has yet to witness the commercial application of PMP technology, although at least one product has begun phase II clinical trials with others following close behind. These emerging products are set to challenge the complex and overlapping regulations that currently govern GM plants and ‘conventional’ pharmaceutical production. The areas of responsibility are being mapped out between the different EU regulatory agencies, with specific guidelines currently being drawn up for the regulation of PMPs. This article discusses issues surrounding the development of robust risk-assessment and risk-management practices based on health and environmental impact, while working with EU regulatory authorities to ensure appropriate regulatory oversight.  相似文献

The daily life of photosynthetic plants revolves around sugar production, transport, storage and utilization, and the complex sugar metabolic and signaling networks integrate internal regulators and environmental cues to govern and sustain plant growth and survival. Although diverse sugar signals have emerged as pivotal regulators from embryogenesis to senescence, glucose is the most ancient and conserved regulatory signal that controls gene and protein expression, cell-cycle progression, central and secondary metabolism, as well as growth and developmental programs. Glucose signals are perceived and transduced by two principal mechanisms: direct sensing through glucose sensors and indirect sensing via a variety of energy and metabolite sensors. This review focuses on the comparative and functional analyses of three glucose-modulated master regulators in Arabidopsis thaliana, the hexokinase1 (HXK1) glucose sensor, the energy sensor kinases KIN10/KIN11 inactivated by glucose, and the glucoseactivated target of rapamycin (TOR) kinase. These regulators are evolutionarily conserved, but have evolved universal and unique regulatory wiring and functions in plants and animals. They form protein complexes with multiple partners as regulators or effectors to serve distinct functions in different subcellular locales and organs, and play integrative and complementary roles from cellular signaling and metabolism to development in the plant glucose signaling networks.  相似文献

The successful long‐term use of taxane for cancer therapy is often prevented by the development of drug resistance in clinic. Thus, exploring the mechanisms involved is a first step towards rational strategies to overcome taxane resistance. Taxane resistance‐related microRNA (miRNAs) are under investigation and miRNAs could induce the taxane resistance of tumour cells by regulating cell cycle distribution, survival and/or apoptosis pathways, drug transports, epithelial–mesenchymal transition and cancer stem cell. This article summarizes current research involving miRNAs as regulators of key target genes for tanxanxe chemoresistance and discusses the complex regulatory networks of miRNAs. Also, the authors will envisage future developments towards the potential use of targeting miRNAs as a novel strategy for improving response of tumour patients to taxane. miRNAs play critical roles in taxane chemoresistance and the miRNA‐based therapies will be helpful for overcoming drug resistance and developing more effective personalized anti‐cancer treatment strategies. Further research studies should be performed to promote therapeutic–clinical use of taxane resistance‐related miRNAs in cancer patients, especially in those patients with taxane‐resistant cancers.  相似文献

The food production and processing value chain is under pressure from all sides—increasing demand driven by a growing and more affluent population; dwindling resources caused by urbanization, land erosion, pollution and competing agriculture such as biofuels; and increasing constraints on production methods driven by consumers and regulators demanding higher quality, reduced chemical use, and most of all environmentally beneficial practices ‘from farm to fork’. This pressure can only be addressed by developing efficient and sustainable agricultural practices that are harmonized throughout the value chain, so that renewable resources can be exploited without damaging the environment. Bridges must, therefore, be built between the diverse areas within the food production and processing value chain, including bridges between different stages of production, between currently unlinked agronomic practices, and between the different levels and areas of research to achieve joined-up thinking within the industry, so that the wider impact of different technologies, practices and materials on productivity and sustainability is understood at the local, regional, national and global scales. In this article, we consider the challenges at different stages and levels of the value chain and how new technologies and strategies could be used to build bridges and achieve more sustainable food/feed production in the future.  相似文献

In the biomedical field, calls for the generation of new regulations or for the amendment of existing regulations often follow the emergence of apparently new research practices (such as embryonic stem cell research), clinical practices (such as facial transplantation) and entities (such as Avian Influenza/'Bird Flu'). Calls for regulatory responses also arise as a result of controversies which bring to light longstanding practices, such as the call for increased regulation of human tissue collections that followed the discovery of unauthorised post-mortem organ retention. Whilst it seems obvious that new regulations should only be generated if existing regulations are inadequate (a practice referred to in this paper as 'regulatory syncretism'), this does not always occur in practice. This paper examines the conceptual steps involved in generating regulatory responses to emerging phenomena. Two decision points are identified. First, a stance is taken as to whether the emerging phenomenon raises unique ethical or legal issues (exceptionalism versus non-exceptionalism). Second, the decision is made as to whether new regulation should be generated only for truly unique phenomena (syncretism versus asyncretism). It is argued here that it is important to make a careful assessment of novelty, followed by a reflective and deliberative choice of regulatory syncretism or asyncretism, since each type of regulatory response has advantages which need to be harnessed and disadvantages which need to be managed--something that can only occur if regulators are attentive to the choices they are making.  相似文献

The field of non-coding RNA (ncRNA) has expanded over the last decade following the discoveries of several new classes of regulatory ncRNA. A growing amount of evidence now indicates that ncRNAs are involved even in the most fundamental of cellular processes. The heat shock response is no exception as ncRNAs are being identified as integral components of this process. Although this area of research is only in its infancy, this article focuses on several classes of regulatory ncRNA (i.e., miRNA, lncRNA, and circRNA), while summarizing their activities in mammalian heat shock. We also present an updated model integrating the traditional heat shock response with the activities of regulatory ncRNA. Our model expands on the mechanisms for efficient execution of the stress response, while offering a more comprehensive summary of the major regulators and responders in heat shock signaling. It is our hope that much of what is discussed herein may help researchers in integrating the fields of heat shock and ncRNA in mammals.  相似文献

Polyhydroxyalkanoates (PHA) are a promising and sustainable alternative to the petroleum-based synthetic plastics. Regulation of PHA synthesis is receiving considerable importance as engineering the regulatory factors might help developing strains with improved PHA-producing abilities. PHA synthesis is dedicatedly regulated by a number of regulatory networks. They tightly control the PHA content, granule size and their distribution in cells. Most PHA-accumulating microorganisms have multiple regulatory networks that impart a combined effect on PHA metabolism. Among them, several factors ranging from global to specific regulators, have been identified and characterized till now. This review is an attempt to categorically summarize the diverse regulatory circuits that operate in some important PHA-producing microorganisms. However, in several organisms, the detailed mechanisms involved in the regulation of PHA synthesis is not well-explored and hence further research is needed. The information presented in this review might help researcher to identify the prevailing research gaps in PHA regulation.  相似文献

Epigenetic mechanisms control gene regulation by writing, reading and erasing specific epigenetic marks. Within the context of multi-disciplinary approaches applied to investigate epigenetic regulation in diverse systems, structural biology techniques have provided insights at the molecular level of key interactions between upstream regulators and downstream effectors. The early structural efforts focused on studies at the single domain-single mark level have been rapidly extended to research at the multiple domain–multiple mark level, thereby providing additional insights into connections within the complicated epigenetic regulatory network. This review focuses on recent results from structural studies on combinatorial readout and crosstalk among epigenetic marks. It starts with an overview of multiple readout of histone marks associated with both single and dual histone tails, as well as the potential crosstalk between them. Next, this review further expands on the simultaneous readout by epigenetic modules of histone and DNA marks, thereby establishing connections between histone lysine methylation and DNA methylation at the nucleosomal level. Finally, the review discusses the role of pre-existing epigenetic marks in directing the writing/erasing of certain epigenetic marks. The increasing diversity of the US coupled with the continuing need for information gathered about race/ethnicity require us to re-examine our practices of collecting and tabulating such data, particularly from individuals of mixed heritage. In the context of Census 2000, which allowed people for the first time to identify with multiple race groups, this article focuses on the context of education and looks at high school students’ self-­identification practices on forms. Survey data gathered from 638 freshmen during 1999–2000 at a diverse, public high school in California indicate: there can be high levels of inconsistency in students’ individual identifications depending on question format and response options provided; and, overall demographic counts can greatly vary depending on how multiple-response data are tabulated. Students’ responses raise questions about whether it is possible to attain a high level of measurement reliability when working with a diverse population that includes individuals of mixed ­heritage.  相似文献

The identification of suppressor T cells as important regulators of basic processes that are designed to maintain tolerance has opened an important area of potential clinical investigation in autoimmunity, graft-versus-host disease and transplantation. However, the field has been limited by an inability to define the antigenic specificities of these cells and by the small numbers of circulating regulatory T cells. Recently, new methods for expanding polyclonal and antigen-specific regulatory T cells have emerged. This article summarizes efforts to exploit regulatory T-cell therapy for the treatment of immunological diseases and poses the question of when and where regulatory T cells will first impact on clinical diseases.  相似文献

Concerns about the negative effects of marine scientific research are in clear juxtaposition to the beneficial role that scientific knowledge plays in enhancing the understanding of the oceans and protecting the marine environment. This presents a regulatory paradox that is examined in this article in light of the legal framework in the 1982 United Nations Convention on the Law of the Sea. The article traces how these general principles in the Convention are elaborated in soft law instruments for the promotion of environmentally sustainable research practices. It also looks at an example of state practice in this area by examining regulatory measures instituted in the Canadian Endeavour Hydrothermal Vent Marine Protected Area.  相似文献

U.S. Environmental Protection Agency wetland regulators appear to use monetary estimates of the societal value of wetlands relatively infrequently in Section 404 permit review, enforcement, and compensatory mitigation-related activities. While 52 % of surveyed EPA regulators had used such estimates at least once, only 3 % reported using them frequently. Forty-eight percent said they never used them. Survey respondents indicated that their use of such estimates is inhibited by a lack of relevant information, uncertainty about the scientific validity of estimates, and concerns about the scientific and legal defensibility of estimate use. Respondents said they would be more apt to use estimates in regulatory activities if they perceived agency approval for such use and had access to training and best practices. The barriers and inducements EPA regulators cited largely accord with the literature on the science-policy divide and with the argument that regulatory use of valuation estimates may be inappropriate because research in this arena is not sufficiently advanced.  相似文献

Summary   The closure and decommissioning of Alcoa's first mine in Western Australia at Jarrahdale was a landmark for the company. This article describes the strategies and research contributing to successful mine rehabilitation in the Jarrah forest. Lessons learnt at Jarrahdale are being applied to other Western Australian mines. Over 30 years of research have resulted in a strong working knowledge of ecosystem function in this environment. The direct linkage of research to operational practices provides two-way benefits for practitioners and researchers. The Jarrahdale mine is the first mine in Western Australia to have received a certificate of completion from the state government.  相似文献
