Synopsis The serratus metapatagialis (SMP) muscle of the pigeon has been studied histochemically and ultrastructurally. At the gross anatomical level the SMP is clearly divisible into a peripheral whitish band and a red portion comprised predominantly of pale and red fibres respectively. The pale fibres possess low succinate dehydrogenase, low mitochondrial content, absence of subsarcolemmal mitochondrial aggregates, low fat, moderate glycogen, high phosphorylase, low-to-moderate regular myofibrillar adenosine triphosphatase (M-ATPase), activation of M-ATPase following acid preincubation and jagged Z bands. On the basis of these characteristics, these physiologically slow muscle fibres have been termed Type I white or slow-twitch glycolytic. The SMP red fibres, however, possess high aerobic as well as glycolytic capacity, high M-ATPase activity which is labile after acid preincubation and thick but straight Z bands; therefore, they are the Type II red or fast-twitch oxidative-glycolytic.Some of data in this paper was presented at the Anatomical Society of Australia and New Zealand, 1978. 相似文献

The influence of muscle geometry on muscle shortening of the gastrocnemius medialis muscle (GM) of the rat was studied. Using cinematography, GM geometry was studied during isokinetic concentric activity at muscle lengths ranging from 85 to 105% of the optimum muscle length. The shortening speed of the distal fibre, the proximal aponeurosis and the muscle were determined, as well as the effect of rotation of the distal fibre and the proximal aponeurosis on the muscle speed of shortening. The results show that, due to the geometrical configuration, muscle shortening speed is not only determined by the speed of the fibre, but also to a large extent by the aponeurosis shortening speed. At optimum muscle length, the fibre and aponeurosis shortening speeds expressed relative to the muscle shortening speed amounted to 84% and 6%, respectively. At shorter muscle length, fibre speed relative to muscle speed decreased to values as low as 35%, whereas that of aponeurosis increased to values as high as 31%. Angular effects on the muscle speed of shortening can explain 10% of the muscle shortening speed at optimum muscle length and up to 34% of the muscle speed at shorter muscle length. In addition, a model was formulated to simulate the geometrical effects on muscle speed. This model, incorporating both fibre and aponeurosis length changes, contains a transfer function relating the shortening speeds of fibre and aponeurosis to muscle speed. Intra- and extracellular action potentials of isolated frog muscle fibres were recorded at different distances to the end of the fibre. The first and second time derivatives of the intracellular action potentials were also recorded. The intracellular action potentials and their first and second time derivatives were almost the same regardless of the place of recording. With the decrease in the axial distance to the end the extracellular action potentials changed gradually in a complicated manner from a shape similar to the second time derivative into a shape similar to the first time derivative. Extracellular potentials, having two negative maxima, were recorded over the terminal taper part of the fibres.These alterations were simulated by a mathematical model. It was shown that the changes in the shape of the extracellular action potentials around the end of the fibres were mainly due to the existence of the fibre end though a better correspondence of the experimentally recorded and the calculated extracellular action potentials was obtained when the morphology of the fibre end was taken into consideration. 相似文献

The purpose of the study was to investigate the neuromuscular state in the preterm infants using surface electromyography (EMG). Ten preterm (gestational age, 31–32 weeks) and ten term infants (gestational age, 38–39 weeks) participated in the study in the second, fourth and sixth postnatal weeks. Linear and novel nonlinear parameters were used for the treatment of the interference EMG (iEMG) in four muscles (mm. bic. br. dext., trie. br. dext, tib. ant. sin., and gastr. sin.). In the preterm infants aged 33–37 weeks, both linear and nonlinear iEMG parameters were significantly lower in comparison to the term infants. Thus, the iEMG of the preterm infants was characterized by a more primitive time-domain structure and lower amplitude and spectrum frequency. In addition, unlike in term newborns, the lifetime dynamics of the iEMG parameters in the preterm infants was retarded. Thus, the motor system of a preterm infant is likely to be less prepared for postnatal life due to the shorter stay in utero. Nonetheless, the iEMG of the preterm infants may be characterized as more complex in comparison to the term newborns of the same postconceptual age. Introduction: Mature skeletal muscles are composed of a complex assembly of slow-twitching, fast-twitching and hybrid fibres. Since muscle fibres exhibit a high degree of cellular plasticity, changed physiological conditions or pathophysiological disturbances have generally a substantial impact on fibre specification. Areas covered: This article reviews the findings from comparative proteomic profiling studies that have focused on neuromuscular diseases and discusses the identified protein changes of fibre type shifting. The reviewed literature on weight loss, obesity, diabetes, cancer cachexia, disuse atrophy, motor neuron disease, myotonia, inflammatory myopathies, myofibrillar myopathies, muscular dystrophies and sarcopenia of old age suggests that proteome-wide alterations occur in the expression of distinct protein families, encompassing especially contractile and regulatory proteins of the acto-myosin apparatus. Expert commentary: The systematic determination of proteome-wide changes in neuromuscular disorders can now be used to design novel diagnostic and therapy-monitoring tools for evaluating fibre transitions in pathological muscles. 相似文献

Morphological, histochemical and ultrastructural characteristics of intrafusal fibre types were studied in rat muscle spindles. The existence of three intrafusal fibre types, namely the typical bag, the intermediate bag and the chain fibres was confirmed. Intrafusal fibres differ in diameter, length and number of nuclei in the equatorial zone. Histochemically, typical bag fibres exhibit both alkali- and acid-stable ATPase activity and low SDH activity. Intermediate bag fibres possess low alkali-stable ATPase activity; after acid-preincubation, however, they have low activity only in the juxtaequatorial region, whereas in the polar zones they exhibit high acid-stable ATPase activity. The SDH activity varies from moderate to high. The chain fibres exhibit high alkali-stable and low acid-stable ATPase and high SDH activity in the extensor digitorum longus muscle, whereas in the soleus muscle the acid-stable ATPase activity varies from a low one to a high one, either among individual chain fibres in one spindle, and/or repeatedly along the fibre length. Since there are regional differences in morphological characteristics and in staining properties of intrafusal fibres, a reliable identification of intrafusal fibre types can only be achieved by an analysis of serial sections. 相似文献

Phospholipid compositions were determined in samples of gastrocnemius muscles of individuals from fetal stage to the age of 55 yr. The lipids were separated by thin-layer chromatography. In order to enable characterization of the individual phospholipids, a lipid extract was prepared from pooled samples of gastrocnemius muscles from adult males and separated by ion-exchange chromatography on TEAE-cellulose. The individual phosphoglycerides were purified by thin-layer chromatography and then characterized according to their content of fatty acids, aldehydes, phosphorus, and the identity of the bases. The relative amounts of the major phospholipids, choline and ethanolamine phosphoglycerides, changed little with age, and in adult males they constituted 47% and 24%, respectively. Cardiolipin increased from 3% in the fetal stage to 9% by the end of the first year of life. Sphingomyelin and serine phosphoglyceride decreased with increasing age, while inositol phosphoglyceride increased. In adult males, cardiolipin constituted 10% of total lipid phosphorus, inositol phosphoglyceride constituted 9%, serine phosphoglyceride 3%, and sphingomyelin 7%. 相似文献

目的:探讨臀肌挛缩松解术结合术后康复训练治疗青少年注射性臀肌挛缩症的临床疗效。方法:对97例青少年注射性臀肌挛缩症患者行臀肌挛缩松解术治疗,结合术后康复训练,术后随访10月~46月,平均33个月,临床疗效根据黄耀添评价标准评定。结果:所有患者中:优75例,良13例,可8例,差1例,优良率为90.7%。结论:对于青少年注射性臀肌挛缩患者,手术彻底松解挛缩组织,减少创伤,结合术后有效的康复训练,可获得满意疗效。 相似文献

Fibre type distributions in the vastus lateralis muscles of six male subjects (age 18 to 22 years) have been compared at rest and during exercise. Exercise consisted of one leg cycling at 60% VO2 max (one leg) for 120 min. The increased contractile activity was associated with a 28.8% (p less than 0.05) decrease in the distribution of Type I fibres in the exercised leg. This change in fibre type distribution was manifested early in the exercise (15 min), and was also evident in muscle samples obtained after 60 and 120 min of activity. Reductions in the Type I fibre distribution was accompanied by an increase in the Type II fibres, specifically the Type IIA distribution (p less than 0.05). Comparable alterations in the specific fibre distribution were also found in the non exercising leg. These observations indicate that alterations in the muscle cell induced either directly or indirectly by the increased contractile activity interact with normal pre-incubation conditions to effect changes in the stability of the myofibrillar ATPase reaction. Specifically, it appears that a percentage of the Type I fibre population becomes acid labile and alkali stable. 相似文献

A high proportion of the "intermediate" muscle fibres, which are semi-dark after both acidic and alkaline preincubation in ATP-ase staining, was found in the thigh with heart failure. Summary The fibre type composition of the striated muscle layer of the oesophagus of the cow, sheep, donkey, dog and cat was examined with standard histochemical methods and immunohistochemical staining using type-specific antimyosin sera. The heavy chain and light chain composition of oesophageal myosin was also examined using electrophoretic peptide mapping and 2-dimensional gel electrophoresis respectively. In the ruminants and donkey the oesophagus was composed of fibre types I, IIA and IIC with immunohistochemical characteristics identical to those of the same fibre types found in control skeletal muscle. In the ruminants there was a gradient in the proportion of type I fibres from 1% (at the cervical end) to about 30% (at the caudal end).In the carnivores the oesophageal muscle was composed of a very small percentage of type I and IIC fibres, but the predominant type was very different hisotchemically and immunohistochemically from all the fibre types (I, IIA, IIB, IIC) present in the control muscles. This oesophageal fibre type (IIoes) had an acid- and alkaline-stable m-ATP-ase activity, a moderate histochemical Ca-Mg actomyosin ATPase activity, and reacted weakly with anti-IIA and antiIIB myosin sera. Although the light chains of the IIoes myosin were the same as the light chains of a mixture of IIA and IIB myosins, their respective heavy chains gave different peptide maps. Greater differences were obtained between the heavy chains of IIoes and other striated muscle myosins.These observations lead us to conclude that this predominant fibre type of the carnivore oesophageal striated muscle is of the fast type, and contains a distinct isoform of myosin similr but not identical to the other fast type myosins. 相似文献

The aim of this study was to provide direct in vivo information of the physiological and structural characteristics of active muscle fibres from a large part of the upper trapezius muscle. Two-dimensional (2-D) multi-channel surface electromyography recordings were used, with 13 × 10 electrodes covering 6 × 4.5 cm of the skin’s surface. A previously developed method was applied to detect individual propagating motor unit action potentials and to estimate their corresponding muscle fibre conduction velocity (MFCV) and muscle fibre orientation (MFO). Using these estimates, spatial distributions of MFCV and MFO were examined for five male subjects performing isometric shoulder elevation at different force levels. The main results were: (1) the general relationship between MFCV and force generation was non-systematic, with a positive relationship at the inferior part of the muscle, (2) the spatial distribution of MFCV at different force levels and fatigue was inhomogeneous and (3) the MFO was slightly different (6°) of the muscle fibres with origin superior compared to inferior to the C7 vertebra. These findings provide new information of the MFO of contracting muscle fibres and knowledge of the physiological characteristics of a large part of the upper trapezius muscle that previously was based on observations from human cadavers only. 相似文献

A comparison was carried out between the motor unit (MU) firing rate and the characteristics of the twitch and the fibre type composition of anconeus and triceps brachii. Fibre type composition (type I, type II) was determined in whole cross-sections of cadaver specimens. The proportion of type I fibre was 60%-67% in anconeus and 32-40% in the lateral head of triceps brachii. Reflecting these histochemical differences, the contraction time of anconeus and triceps was 92 +/- 9 ms and 68 +/- 9 ms respectively. It follows that anconeus can be classified as a slow muscle, as opposed to the lateral head of triceps. The relationship between MU firing rate and isometric force, tested at 90 degrees elbow flexion, differed between the two muscles for force values below 30% of maximal voluntary contraction. No significant increase in MU firing rate was found in anconeus at forces above 5% of maximal voluntary contraction. It is concluded that even within a single agonistic muscle group acting at a single joint there is an adaptation of MU firing rate to the contractile characteristics of each muscle. 相似文献

In vitro incubation of isolated rodent skeletal muscle is a widely used procedure in metabolic research. One concern with this method is the development of an anoxic state during the incubation period that can cause muscle glycogen depletion. Our aim was to investigate whether in vitro incubation conditions influence glycogen concentration in glycolytic extensor digitorum longus (EDL) and oxidative soleus mouse muscle. Quantitative immunohistochemistry was applied to assess glycogen content in incubated skeletal muscle. Glycogen concentration was depleted, independent of insulin‐stimulation in the incubated skeletal muscle. The extent of glycogen depletion was correlated with the oxidative fibre distribution and with the induction of hypoxia‐induced‐factor‐1‐alpha. Insulin exposure partially prevented glycogen depletion in soleus, but not in EDL muscle, providing evidence that glucose diffusion is not a limiting step to maintain glycogen content. Our results provide evidence to suggest that the anoxic milieu and the intrinsic characteristics of the skeletal muscle fibre type play a major role in inducing glycogen depletion in during in vitro incubations. - 1.1. The influences of age (5, 13 and 25-month-old rats), overload as obtained by denervation of synergists, and training on the metabolic capacity, relative muscle cross-sectional area occupied by each fibre type, capillarization and fatigue resistance of the rat m. plantaris were investigated.
- 2.2. Creatine kinase, phosphorylase and citrate synthase activities were lower in muscles of 25 than in those of 13-month-old rats (P < 0.001).

The occurrence of muscle spindles was studied in the masseter muscle of rabbits by light-microscopy of whole muscle sections. The distribution of the spindles appeared to be heterogeneous. Most spindles lay in the anterior/deep and mid-parts of the masseter. The lateral/superficial part contained only a few muscle spindles. The distribution of the spindles is correlated to the distribution of slow twitch (type I) extrafusal fibres. This means that spindles, like type I fibres might be involved in the control of fine movements and posture. Spindle density and type I fibre density increase with distance from the temporomandibular joint. This could mean that spindles are involved in controlling bite force. 相似文献
