桃蚜自然种群初级和次级共生菌的分子鉴定   总被引:7,自引:0,他引:7

蚜虫不仅带有专性初级内共生菌,还常常带有不同类群的兼性次级共生菌。本研究采用真细菌16S rDNA的通用引物,从桃蚜Myzus persicae (Sulzer)烟草种群体内扩增出1.5 kb的共迁移目的条带,然后将其克隆,并在阳性克隆筛选时进行了RFLP分析。当采用EcoRⅠ进行单酶切分析时,目的片段被切割成~650 bp和~850 bp两个小片段;当采用EcoRⅠ和HindⅢ进行双酶切分析时,该蚜群共生菌酶切谱带被分成2组,其中1组(GroupⅠ)只具有一个EcoRⅠ酶切位点,而另1组(Group Ⅱ)不仅具有一个EcoRⅠ酶切位点,还具有一个HindⅢ酶切位点,而且GroupⅠ为优势共生菌群。在此基础上分别对2组共生菌的16S rDNA全序列(~1.5 kb)进行了测定,结果表明:GroupⅠ属于泛菌属Pantoea,与成团泛菌Pantoea agglomerans亲缘关系最近(同源性达99.70%),而Group Ⅱ属于蚜虫的专性内共生菌,即Buchnera aphidicola(同源性达99.50%)。这是在蚜虫体内存在泛菌次级共生菌的首次报道。  相似文献

The cleavage of glucosinolates by myrosinase to produce toxic breakdown products is a characteristic insect defense of cruciferous plants. Although green peach aphids ( Myzus persicae ) are able to avoid most contact with myrosinase when feeding from the phloem of Arabidopsis thaliana , indole glucosinolates are nevertheless degraded during passage through the insects. A defensive role for indole glucosinolates is suggested by the observation that atr1D mutant plants, which overproduce indole glucosinolates, are more resistant to M. persicae , whereas cyp79B2 cyp79B3 double mutants, which lack indole glucosinolates, succumb to M. persicae more rapidly. Indole glucosinolate breakdown products, including conjugates formed with ascorbate, glutathione and amino acids, are elevated in the honeydew of M. persicae feeding from atr1D mutant plants, but are absent when the aphids are feeding on cyp79B2 cyp79B3 double mutants. M. persicae feeding from wild-type plants and myrosinase-deficient tgg1 tgg2 double mutants excrete a similar profile of indole glucosinolate-derived metabolites, indicating that the breakdown is independent of these foliar myrosinases. Artificial diet experiments show that the reaction of indole-3-carbinol, a breakdown product of indol-3-ylmethylglucosinolate, with ascorbate, glutathione and cysteine produces diindolylmethylcysteines and other conjugates that have antifeedant effects on M. persicae . Therefore, the post-ingestive breakdown of indole glucosinolates provides a defense against herbivores such as aphids that can avoid glucosinolate activation by plant myrosinases.  相似文献

Cruciferous plants produce a wide variety of glucosinolates as a protection against herbivores and pathogens. However, very little is known about the importance of individual glucosinolates in plant defense and the regulation of their production in response to herbivory. When Myzus persicae (green peach aphid) feeds on Arabidopsis aliphatic glucosinolates pass through the aphid gut intact, but indole glucosinolates are mostly degraded. Although aphid feeding causes an overall decrease in Arabidopsis glucosinolate content, the production of 4-methoxyindol-3-ylmethylglucosinolate is induced. This altered glucosinolate profile is not a systemic plant response, but is limited to the area in which aphids are feeding. Aphid feeding on detached leaves causes a similar change in the glucosinolate profile, demonstrating that glucosinolate transport is not required for the observed changes. Salicylate-mediated signaling has been implicated in other plant responses to aphid feeding. However, analysis of eds5, pad4, npr1 and NahG transgenic Arabidopsis, which are compromised in this pathway, demonstrated that aphid-induced changes in the indole glucosinolate profile were unaffected. The addition of purified indol-3-ylmethylglucosinolate to the petioles of cyp79B2 cyp79B3 mutant leaves, which do not produce indole glucosinolates, showed that this glucosinolate serves as a precursor for the aphid-induced synthesis of 4-methoxyindol-3-ylmethylglucosinolate. In artificial diets, 4-methoxyindol-3-ylmethylglucosinolate is a significantly greater aphid deterrent in the absence of myrosinase than its metabolic precursor indol-3-ylmethylglucosinolate. Together, these results demonstrate that, in response to aphid feeding, Arabidopsis plants convert one indole glucosinolate to another that provides a greater defensive benefit.  相似文献

The green peach aphid (Myzus persicae) is a phloem-feeding insect that causes economic damage on a wide array of crops. Using a luminol-based assay, a superoxide-responsive reporter gene (Zat12::luciferase), and a probe specific to hydrogen peroxide (HyPer), we demonstrated that this aphid induces accumulation of reactive oxygen species (ROS) in Arabidopsis thaliana. Similar to the apoplastic oxidative burst induced by pathogens, this response to aphids was rapid and transient, with two peaks occurring within 1 and 4 hr after infestation. Aphid infestation also induced an oxidative response in the cytosol and peroxisomes, as measured using a redox-sensitive variant of green fluorescent protein (roGFP2). This intracellular response began within minutes of infestation but persisted 20 hr or more after inoculation, and the response of the peroxisomes appeared stronger than the response in the cytosol. Our results suggest that the oxidative response to aphids involves both apoplastic and intracellular sources of ROS, including ROS generation in the peroxisomes, and these different sources of ROS may potentially differ in their impacts on host suitability for aphids.  相似文献

Infectivity of entomopathogenic nematode (EPN) Steinernema carpocapsae Pocheon strain on the green peach aphid Myzus persicae and its parasitic wasps (e.g., Aphidius colemani, Aphidius gifuensis and Diaeretiella rapae) was evaluated under laboratory conditions. Infective juveniles (IJs) of S. carpocapsae Pocheon strain had low infectivity against nymph and adult stages of M. persicae, showing 2% and 6.7% of mortality, respectively. Application of the EPNs had little effect on mummies caused by the three parasitoid species, allowing them to remain intact. No IJ invaded the host, regardless of EPN application rate. The parasitoid emergence from mummies ranged from 80% to 85% in the presence of EPN while 79–86% was recorded in the absence of EPN. However, the presence of the IJs reduced oviposition by the three parasitoid species, decreasing the rate up to 59% when the nematodes were applied before parasitoid release, while little difference in oviposition was observed when nematodes were applied after parasitoid release.  相似文献

蚜虫寄生蜂的广泛分布可能与有翅蚜的迁飞有关。为了证明这一假说, 本研究利用微小昆虫飞行磨系统, 分数十批(≤16头/批)对被烟蚜茧蜂Aphidius gifuensis Ashmead单头寄生后的有翅桃蚜Myzus persicae (Sulzer)进行模拟飞行并系统观察了飞行后有翅桃蚜的单头定殖情况。对实验观察数据按飞行时间和飞行距离进行归类, 采用单因素方差分析方法分析飞行时间和飞行距离对有翅桃蚜定殖后的存活和生殖力、烟蚜茧蜂发育等变量的影响; 采用多项式回归分析对所有变量和归类的数据进行统计和分析。结果表明: 成功飞行并定殖的有翅桃蚜有378头, 其中239头有翅桃蚜定殖后形成僵蚜。形成僵蚜的有翅桃蚜在飞行实验时平均飞行时间和飞行距离分别为2.63 h和2.16 km, 定殖后平均存活了6.11 d, 定殖第6天和第14天平均产若蚜累计数量分别为8.5头和162头。形成僵蚜和未形成僵蚜的有翅蚜的模拟飞行数据和定殖观察数据无显著差异。形成僵蚜的239头有翅桃蚜中, 最终成功育出烟蚜茧蜂205头, 僵蚜出蜂率为85.8%。雌雄性比为1∶5.2。结论认为, 专性寄生蚜虫的烟蚜茧蜂可以利用有翅蚜的迁飞而被携带扩散传播, 有翅蚜迁飞在蚜虫寄生蜂扩散中具有重要作用。  相似文献

Resistance level of seven accessions of wild Solanum species (Solanaceae) to Myzus persicae and Macrosiphum euphorbiae (Homoptera: Aphididae) was evaluated by measuring survival and fecundity during sleeve cage experiments and population growth on whole plants in a controlled environment. The survival was lowest on the Solanum circaeifolium spp. capsicibaccatum, Solanum pinnatisectum and Solanum trifidum accessions for M. persicae and on the S. circaeifolium spp. capsicibaccatum, Solanum okadae and S. trifidum, accessions with M. euphorbiae. Plant species significantly influenced the fecundity of both aphid species. Aphid population growth on whole plants was negatively affected by the age of the plant, but generally followed the levels of net reproductive rate on different plant species observed during the sleeve cage experiment. The population of M. persicae varied among the seven wild potato accessions with the lowest growth rates on S. circaeifolium spp. capsicibaccatum, Solanum polyadenium, Solanum tarijense and S. trifidum. The number of M. euphorbiae also varied among accessions but the smallest population was collected from S. polyadenium. The results can be used to identify sources of resistance to aphids within those accessions already known to be resistant to the Colorado potato beetle. This study highlights the difficulties involved in developing a high-throughput screening test for aphid resistance compatible with a potato breeding programme.  相似文献

Mature turnip plants, mechanically infected as seedlings with the semi-persistent, aphid transmitted caulimovirus, cauliflower mosaic (CaMV), were treated by spraying with either a solution of Pymetrozine plus adjuvant oil, adjuvant oil or water only. At the same time turnip seedlings were sprayed for each of the three treatments. Two h after spraying, Myzus persicae were caged onto an infected turnip plant for each of the three treatments. Twenty four h later, groups of 20 aphids were transferred from the infected plants, to seedlings from each of the three treatments. After 24 h, these were removed and seedlings were later recorded for infection. This acquisition/transmission assay was repeated at 3, 7, 14 and 21 days from treatment. Only aphids exposed to the Pymetrozine treated source plants were shown to move off the plant and failed to transmit CaMV effectively to treated or control seedlings during the 0 and 3 day assays. The majority soon died when transferred to test seedlings. Progressively, more aphids were found to survive and transmit CaMV during the 7 day and 14 day assays. By 21 days no significant effect could be recorded between treatments and controls. Aphids transferred from control treated source plants to Pymetrozine treated seedlings were able to transmit CaMV within all the assays, although higher mortality was recorded in the day 0 assessment when compared to those transferred to control treated seedlings. We conclude from this trial, that a single foliar treatment of 100 mg litre1 Pymetrozine to CaMV infected turnip plants, effectively reduces the vectoring capability of M. persicae, that feed on these plants, for up to 7 days. However, Pymetrozine failed to stop virus transmission to treated seedlings from the ingress of viruliferous aphids. Pymetrozine was not shown to cause any phytotoxic responses to plants used in this trial.  相似文献

Abstract The interval timer mechanism was examined in two clones of Myzus persicae (Sulzer) originating from peach in Naousa, North Greece. Overwintering eggs were collected in April 1997 and after egg hatch the morphs of progeny were recorded at LD 10 : 14 h and 17 °C until the appearance of sexuals. In one clone, observations were continued until the 21st generation after egg hatch. In addition, the production of sexual morphs was examined in two other clones originating from Lehonia, Central Greece, when apterae (wingless females) were transferred from long to short days and 17 °C in the second and eighth generation of parthenogenetic rearing after their collection from peach trees. In clones from Naousa, the first gynoparae appear and comprise 10% of alate (winged) females in the second half of the third generation after egg hatch. The percentage of gynoparae increased in each consecutive generation, reaching 100% in fifth and seventh generations in clones Nb and Na, respectively. First males appeared later than gynoparae, in fifth and sixth generation in clones Nb and Na, respectively. In clone Nb, the number of males, after their appearance in fifth generation, was relatively constant from the sixth to the ninth generation (10.8–12.3). Then a significant increase was observed in the 10th generation, thereafter remaining relatively constant until the 21st generation (18.1–19.8). A similar trend in the number of males produced was observed in clones deriving from Lehonia.  相似文献

在15℃、20℃、25℃、30℃及35℃条件下分别测定了吡虫啉、丁硫克百威和氰戊菊酯对烟蚜毒力的温度效应。结果显示,吡虫啉和丁硫克百威对烟蚜的毒力呈正温度效应,但随着温度上升,烟蚜对吡虫啉的敏感性变化比丁硫克百威大。毗虫啉毒力在35℃时为15℃时的7.12倍;而丁硫克百威为2.88倍。氰戊菊酯对烟蚜的毒力呈负温度效应,在35℃时的毒力约为15℃时的1/5。鉴于这些药剂的毒力受温度影响的规律,建议在气温较低时选用氰戊菊酯等负温度效应的农药,在气温较高时则选用吡虫啉等正温度效应的农药,以充分发挥农药的性能,减少农药使用量、降低防治成本。  相似文献

为了明确B型烟粉虱Bemisia tabaci取食诱导烟草对烟蚜Myzus persicae的防御反应和时间效应,探讨外来入侵害虫B型烟粉虱取食诱导的烟草防御反应在其与桃蚜种间竞争中的作用,本研究采用小叶笼法和生化分析法,分别研究了B型烟粉虱取食诱导烟草对烟蚜生长发育的影响和烟草主要营养指标的变化。结果表明:B型烟粉虱取食诱导的烟草可对烟蚜产生明显的防御反应,对烟蚜的存活和生长发育有显著的抑制作用和时间效应。在烟粉虱若虫危害烟草至第5天,烟草系统白脉叶片上烟蚜的死亡率较对照叶片上烟蚜的死亡率升高36.13%;至第10天死亡率升高72.78%,且平均相对生长率较对照显著降低;至第15天死亡率升高仍达58.89%。在烟粉虱若虫危害至第10天时,将处理植株上的烟粉虱的若虫虫体去除,然后在烟草系统白脉叶片上接烟蚜1龄若蚜,在去虫后15 d内,烟草系统白脉叶片对烟蚜死亡率及相对生长率的影响较不去虫的对照间均有明显差异(P<0.05)。但至去虫后第20天,对烟蚜的不利影响即消失。研究表明B型烟粉虱取食诱导的烟草系统白脉叶片对烟蚜的抗性能持续表达15 d左右。烟粉虱危害后的局部带虫叶片与对照相比,对烟蚜的生长发育没有明显的不利影响。B型烟粉虱危害的烟草,其可溶性糖和可溶性蛋白的含量降低,这与烟蚜的死亡率表现出显著的负相关性。  相似文献

在室内分别测定了阿维菌素类杀虫剂1% 9512-1乳油和虫螨克对南方小花蝽Orius similis的毒力,研究了无阿维菌素介入后南方小花蝽对烟蚜Myzus persicae捕食效应所受的影响,结果表明:阿维菌素类杀虫剂对南方小花蝽具有一定毒力,不同虫态对阿维菌素的敏感性有差异,卵的敏感性明显低于若虫和成虫;不同的阿维菌素类药剂对南方小花蝽的毒力存在差异,9512-1对南方小花蝽卵、若虫及成虫的LC50依次为11.2743±3.8613、2.6584±1.2139、2.4508±1.3266 mg·L-1,虫螨克则为10.8699±3.7653、6.4422±2.8596、6.4738±3.0124 mg·L-1;阿维菌素显著降低了南方小花蝽对烟蚜的捕食量和捕食能力,也降低了同种个体间的竞争能力;阿维菌素对南方小花蝽若虫的发育速率有显著的抑制作用,但对发育速率与取食量的线性关系无明显影响,而且对存活个体成虫期的产卵量无显著影响。  相似文献

几种病原真菌对二点叶螨、桃蚜的侵染能力研究   总被引:1,自引:0,他引:1  
从兰州市刘家堡乡自然感病的菜豆蚜尸上,经分离、纯化、筛选得到5种病原真菌,即新接霉属、干尸霉属、虫瘴霉属、虫疠霉属、匍柄霉属。以室内盆栽菜豆上饲养供试桃蚜、二点叶螨为供试生物,测定了几种病原真菌的侵染力。结果表明:匍柄霉属、新接霉属、虫瘴霉属、虫疠霉属对桃蚜有较高的侵染力,接种第7 d后校正死亡率分别为74.0%、70.0%、64.0%、62.0%;对二点叶螨雌成螨的侵染力相对较低,干尸霉属、新接霉属、虫瘴霉属、匍柄霉属接种第7 d后校正死亡率分别为48%、45.9%、42.7%和38.8%。几种病原真菌孢子悬浮液处理二点叶螨螨卵后,可延长螨卵的发育历期,降低螨卵孵化率。  相似文献

Abstract 1 The life parameters of one red and one green clone of Myzus persicae reared on 11 Oriental tobacco type varieties (KP7, K63, KZ10/z, MA13/b, BX2/a, N34/4, KP14/a, Kolindros, S79, TA21 and BX81) and on five Virginia ones (VE9, VE3, SPG28‐VE2, Kleio‐KN1 and Niki‐NK3) at 20 °C and LD 16 : 8 h were examined. 2 In general, the clones, performed better on Oriental than on Virginia type varieties. The varieties TA21 and Kolindros could be considered as the most resistant among the Oriental ones. Aphids on Virginia varieties showed longer developmental time (9.1–9.6 days), a lower intrinsic rate of increase (0.2347–0.2647) and higher nymphal mortality (27.9–52.5%) than on Oriental varieties. The corresponding values in the latter were 7.3–9.0 days, 0.2784–0.3302 and 10.0–47.9%. 3 The leaves of Virginia type varieties (188–471 hairs/cm2) and one Oriental (K63) 273 hairs/cm2 were more hairy than the other Oriental type varieties (61–150 hairs/cm2). The Virginia type varieties showed also a lower level of reducing sugars (3.5–7.2%) than the Oriental ones (7.0–9.8%). Trichome density and level of reducing sugars were significantly negatively correlated with developmental time and intrinsic rate of increase. 4 An interclonal variation in performance, possibly related to the colour of aphids, was observed. The red clone performed better than the green one. The same trend was observed when the results of the previous clones on the variety KP14/a were analysed with the data obtained on the same variety from two other clones (one red and one green).  相似文献

Abstract:  The green-peach aphid, Myzus persicae (Sulzer) (Hem., Aphididae), is a major agricultural pest of a wide range of host plants, causing damage by feeding and indirectly by transmitting viruses. In this study we tested the effect of the antimicrobial peptide indolicidin on M. persicae survival and on its essential bacterial endosymbionts. Artificial diet bioassays showed a significant dose-dependent lethal response of indolicidin on M. persicae survival (LD50 of 209 ± 60  μ g/ml). Histological analysis of indolicidin-treated aphids revealed a lower number of distorted mycetocytes, whereas control aphids showed abundant number of rounded and filled mycetocytes. These results suggest that aphid survival could be affected via reduction of its endosymbionts. Thus, aphid control based on antimicrobial substances against endosymbionts could be a promising strategy that needs to be further explored.  相似文献

烟蚜Myzus persicae是一种世界性的重要农业害虫。为探讨我国不同地理种群烟蚜形态特征变异情况, 本研究测量了我国18个地理种群烟蚜的体长、 体宽、 头宽、 各足腿节长、 各足胫节长等18个形态性状度量特征和Q值, 并进行方差分析和基于欧氏距离与地理距离和海拔差距的Mantel Test, 利用各形态特征与体长的比值构成的比例特征进行系统聚类和主成分分析。结果表明: 部分地理种群烟蚜的形态性状差异显著。系统聚类和主成分分析表明, 18个烟蚜地理种群中贵州贞丰种群(ZF-GZ)与其他种群相比差异最大, 陕西南泥湾(NNW-SX)、 湖南慈利(CL-HN)、 安徽谯城(QC-AH)和重庆武隆(WL-CQ)种群差异最小, 其他种群间的差异程度介于这两者之间。Mantel Test显示欧氏距离与地理距离、 海拔高度差距都不具有相关性。烟蚜种群的形态分化不符合地理隔离模式, 地理气候条件对种群形态有一定影响, 种群形态差异的形成是多种因素综合作用的结果, 但具体作用机制尚不明确。  相似文献

The antibiotic and antixenotic resistance of six commonly produced potato cultivars in Iran including Aozonia, Agria, Cosima, Cosmos, Kondor and Savalan to the green peach aphid, Myzus persicae Sulzer, were investigated under laboratory conditions at 20 ± 2oC, 65 ± 5% RH and 16:8h (L:D) in 2009. Antibiosis experiments showed significant differences in the developmental time, nymphal survivorship, fecundity, adult longevity of the green peach aphid among the potato cultivars. Intrinsic rate of natural increase (r m) for apterous aphids varied significantly with the potato cultivars on which the aphids were reared. This value ranged from 0.225 to 0.293 females/female/day, which was lowest on Cosmos and highest on Aozonia. Additionally, the estimated net reproductive rate (R0) and finite rate of increase (λ) for apterous aphids were the lowest on Cosmos. For the antixenosis experiment, no significant difference was found in aphid's preference to the potato cultivars. However, Aozonia was preferred more than the other five cultivars by the apterous aphids. Therefore, our results demonstrated that among the investigated cultivars the Cosmos cultivar is moderately resistant to the green peach aphid.  相似文献

Female adults of the aphidopagous gall midge, Aphidoletes aphidimyza (Rondani) (Diptera: Cecidomyiidae), showed an olfactory response to honeydew excreted by the green peach aphid, Myzus persicae (Sulzer) (Hemiptera: Aphididae) under laboratory conditions. The response was only elicited by treatments with honeydew, whereas aphids, aphids with pepper plants or leaves, and pepper plants or leaves were not attractive to the midges. Dose‐dependent responses were observed from whole honeydew, honeydew volatiles extracted in pentane, and honeydew volatiles captured on Porapak Q®. When honeydew was eluted with three sequential pentane washes, a positive response was only observed from the midges for the first wash. Female midges laid more eggs on pepper plants infested with higher densities of M. persicae. The olfactory response of midges to honeydew is discussed with respect to prey location.  相似文献

Analysis of the holocentric mitotic chromosomes of the peach-potato aphid, Myzus persicae (Sulzer), from clones labelled 50, 51 and 70 revealed different chromosome numbers, ranging from 12 to 14, even within each embryo, in contrast to the standard karyotype of this species (2n?=?12). Chromosome length measurements, combined with fluorescent in situ hybridization experiments, showed that the observed chromosomal mosaicisms are due to recurrent fragmentations of chromosomes X, 1 and 3. Contrary to what has generally been reported in the literature, X chromosomes were frequently involved in recurrent fragmentations, in particular at their telomeric ends opposite to the nucleolar organizer region. Supernumerary B chromosomes have been also observed in clones 50 and 51. The three aphid clones showed recurrent fissions of the same chromosomes in the same regions, thereby suggesting that the M. persicae genome has fragile sites that are at the basis of the observed changes in chromosome number. Experiments to induce males also revealed that M. persicae clones 50, 51 and 70 are obligately parthenogenetic, arguing that the reproduction by apomictic parthenogenesis favoured the stabilization and inheritance of the observed chromosomal fragments.  相似文献
