Hagfish are the most pleisiomorphic extant craniates, and based on the similarity of ionic concentrations between their internal milieu and seawater (SW), they have long been touted as a model for osmo- and ionoconformation. As a result, the lack of direct symmetry between hagfish plasma and the environment with respect to [Na+], [Cl], [Mg2+], and [Ca2+] have been left largely unexplored. In order to determine the capacity of hagfish to regulate their blood compartment, we exposed Pacific hagfish (Eptatretus stoutii) to 24, 32, 40, and 48 g/l salinity for 48 h, as well as to two treatments where a portion of the water [Na+] was replaced with either Mg2+ or Ca2+ at constant salinity for up to 6 days. Following exposure, we measured plasma ion status, pH, and total carbon dioxide (TCO2). As expected, our results indicated that hagfish had no capacity to regulate plasma osmolality, [Na+], or [Cl], but they did maintain plasma [Mg2+] and [Ca2+] nearly constant despite fluctuation of environmental salinity or elevated water [Mg2+] and [Ca2+] (two- and sevenfold, respectively). Furthermore, exposure to elevated water [Mg2+] and [Ca2+] resulted in a large increase of plasma TCO2 with little to no increase of plasma pH. We concluded that hagfish may control plasma [Mg2+] and [Ca2+] at levels below that of their environment via secretion of HCO3 , similar to the mechanisms described in the intestine of teleosts. We speculate that secretion of HCO3 likely evolved to maintain plasma [Mg2+] and [Ca2+] below environmental levels (both of which negatively affect nervous function and muscle contraction if elevated), and was an exaptation for the development of water-absorption mechanisms in the intestine of marine osmoregulators. The ancestors of modern hagfish are thought to have never entered freshwater, thus investigations into their ionoregulatory ability potentially have profound implications regarding the evolution of fishes.  相似文献

Effects of intracellular Mg2+ on a native Ca2+-and voltage-sensitive large-conductance K+ channel in cultured human renal proximal tubule cells were examined with the patch-clamp technique in the inside-out mode. At an intracellular concentration of Ca2+ ([Ca2+]i) of 10−5–10−4 M, addition of 1–10 mM Mg2+ increased the open probability (Po) of the channel, which shifted the Po –membrane potential (Vm) relationship to the negative voltage direction without causing an appreciable change in the gating charge (Boltzmann constant). However, the Mg2+-induced increase in Po was suppressed at a relatively low [Ca2+]i (10−5.5–10−6 M). Dwell-time histograms have revealed that addition of Mg2+ mainly increased Po by extending open times at 10−5 M Ca2+ and extending both open and closed times simultaneously at 10−5.5 M Ca2+. Since our data showed that raising the [Ca2+]i from 10−5 to 10−4 M increased Po mainly by shortening the closed time, extension of the closed time at 10−5.5 M Ca2+ would result from the Mg2+-inhibited Ca2+-dependent activation. At a constant Vm, adding Mg2+ enhanced the sigmoidicity of the Po–[Ca2+]i relationship with an increase in the Hill coefficient. These results suggest that the major action of Mg2+ on this channel is to elevate Po by lengthening the open time, while extension of the closed time at a relatively low [Ca2+]i results from a lowering of the sensitivity to Ca2+ of the channel by Mg2+, which causes the increase in the Hill coefficient. Most inland saline waters in southern Australia predominantly contain Na+ and Cl as major ions. The proportions of Ca2+, Mg2+, SO4 2−, HCO3 and CO3 2− in these waters somewhat vary and might influence salinity tolerance of freshwater organisms. Here the salinity stress of five ionic compositions to the freshwater snail Physa acuta Draparnaud (Gastropoda: Physidae) was compared: commercial sea salt Ocean Nature (ON), synthetic Ocean Nature (ONS) and three saline water types that are common in southern Australia (ONS but without [1]: SO4 2−, HCO3 and CO3 2−, [2]: Ca2+, HCO3 and CO3 2−, [3]: Ca2+, Mg2+), Milli-Q water was used as a negative control. The 96-h LC50 values for all treatments did not differ. However in prolonged sub-lethal exposures, results varied depending on the ionic composition. Growth was negative and shell strength reduced in treatments lacking Ca. Though the content of major cationic elements (Ca, Mg, Na and K) did not differ per unit dry weight of snail across the treatments, the total load of these elements per individual snail varied among treatments. Furthermore, at the sub-lethal salinities tested, 1 and 5 mS cm−1, ionic compositions had more effect on the snail’s growth than salinity. The long-term effects on freshwater animals, especially taxa with calcium-based exoskeletons, from exposure to common saline water types with low calcium concentrations will likely be greater than from exposure to saline waters with an ionic composition similar to seawater.  相似文献

The aim of this study was to investigate the effects of NaCl-salinity on the physiological attributes in common reed, Phragmites australis (Cav.) Trin. ex Steudel. Plants grew optimally under salinity treatment with standard nutrient solution without added salt and at NaCl concentrations up to 100 mM. Applied for 21 days, NaCl-salinity (300 and 500 mM) caused a significant reduction in growth allocation of all different tissues of P. australis. Shoot growth of reed plants displayed a highly significant correlation with plant–water relations and photosynthetic parameters. The net photosynthetic rate and stomatal conductance of reed plants treated with NaCl-salinity at varying osmotic potential (ψπ) of nutrient solutions were positively correlated, and the former variable also had a strong positive relationship with transpiration rate. Leaf water potential and ψπ followed similar trends and declined significantly as ψπ of watering solutions was lowered. The increase in total inorganic nutrients resulting from increased Na+ and Cl in all tissues and K+, Ca2+ and Mg2+ concentrations were maintained even at the most extreme salt concentration. Common reed exhibited high K+/Na+ and Ca2+/Na+ selectivity ratios over a wide range of salinities under NaCl-salinity. These findings suggest that reed plants were able to adapt well to high salinities by lowering their leaf ψπ and the adjustment of osmotically active solutes in the leaves.  相似文献

Physical, biogeochemical and photosynthetic parameters were measured in sea ice brine and ice core bottom samples in the north-western Weddell Sea during early spring 2006. Sea ice brines collected from sackholes were characterised by cold temperatures (range −7.4 to −3.8°C), high salinities (range 61.4–118.0), and partly elevated dissolved oxygen concentrations (range 159–413 μmol kg−1) when compared to surface seawater. Nitrate (range 0.5–76.3 μmol kg−1), dissolved inorganic phosphate (range 0.2–7.0 μmol kg−1) and silicic acid (range 74–285 μmol kg−1) concentrations in sea ice brines were depleted when compared to surface seawater. In contrast, NH4 + (range 0.3–23.0 μmol kg−1) and dissolved organic carbon (range 140–707 μmol kg−1) were enriched in the sea ice brines. Ice core bottom samples exhibited moderate temperatures and brine salinities, but high algal biomass (4.9–435.5 μg Chl a l−1 brine) and silicic acid depletion. Pulse amplitude modulated fluorometry was used for the determination of the photosynthetic parameters F v/F m, α, rETRmax and E k. The maximum quantum yield of photosystem II, F v/F m, ranged from 0.101 to 0.500 (average 0.284 ± 0.132) and 0.235 to 0.595 (average 0.368 ± 0.127) in the sea ice internal and bottom communities, respectively. The fluorometric measurements indicated medium ice algal photosynthetic activity both in the internal and bottom communities of the sea ice. An observed lack of correlation between biogeochemical and photosynthetic parameters was most likely due to temporally and spatially decoupled physical and biological processes in the sea ice brine channel system, and was also influenced by the temporal and spatial resolution of applied sampling techniques.  相似文献

Early summer in the Arctic with extensive ice melt and break-up represents a dramatic change for sympagic–pelagic fauna below seasonal sea ice. As part of the International Polar Year-Circumpolar Flaw Lead system study (IPY-CFL), this investigation quantified zooplankton in the meltwater layer below landfast ice and remaining ice fauna below melting ice during June (2008) in Franklin Bay and Darnley Bay, Amundsen Gulf, Canada. The ice was in a state of advanced melt, with fully developed melt ponds. Intense melting resulted in a 0.3- to 0.5-m-thick meltwater layer below the ice, with a strong halocline to the Arctic water below. Zooplankton under the ice, in and below the meltwater layer, was sampled by SCUBA divers. Dense concentrations (max. 1,400 ind. m−3) of Calanus glacialis were associated with the meltwater layer, with dominant copepodid stages CIV and CV and high abundance of nauplii. Less abundant species included Pseudocalanus spp., Oithona similis and C. hyperboreus. The copepods were likely feeding on phytoplankton (0.5–2.3 mg Chl-a m−3) in the meltwater layer. Ice amphipods were present at low abundance (<10 ind. m−2) and wet biomass (<0.2 g m−2). Onisimus glacialis and Apherusa glacialis made up 64 and 51% of the total ice faunal abundance in Darnley Bay and Franklin Bay, respectively. During early summer, the autochthonous ice fauna becomes gradually replaced by allochthonous zooplankton, with an abundance boom near the meltwater layer. The ice amphipod bust occurs during late stages of melting and break-up, when their sympagic habitat is diminished then lost.  相似文献

Okhotsk Sea pack ice from Shiretoko in northern Hokkaido, sampled in March 2007, contained microalgal communities dominated by the centric diatoms Thalassiosira nordenskioeldi and T. punctigera. Domination by this genus is very unusual in sea ice. Communities from nearby fast ice at Saroma-ko lagoon were dominated by Detonula conferavea and Odontella aurita. Average microalgal biomass of the Okhotsk Sea pack ice (surface and bottom) was 1.59 ± 1.09 μg chla l−1 and for fast ice (bottom only) at nearby Saroma-ko lagoon, 16.5 ± 3.2 μg l−1 (=31.1 ± 5.0 mg chla m−2). Maximum quantum yield of the Shiretoko pack ice algal communities was 0.618 ± 0.056 with species-specific data ranging between 0.211 and 0.653. These community values are amongst the highest recorded for sea ice algae. Rapid light curves (RLC) on individual cells indicated maximum relative electron transfer rates (relETR) between 20.8 and 60.6, photosynthetic efficiency values (α) between 0.31 and 0.93 and onset of saturation values (E k) between 33 and 91 μmol photons m−2 s−1. These data imply that the pack ice algal community at Shiretoko was healthy and actively photosynthesising. Maximum quantum yield of the Saroma-ko fast ice community was 0.401 ± 0.086, with values for different species between 0.361 and 0.560. RLC data from individual Saroma-ko fast ice algal cells indicated relETR between 55.3 and 60.6, α values between 0.609 and 0.816 and E k values between 74 and 91 μmol photons m−2 s−1 which are consistent with measurements in previous years.  相似文献

Purinergic signalling in rat GFSHR-17 granulosa cells was characterised by Ca2+-imaging and perforated patch-clamp. We observed a resting intracellular Ca2+-concentration ([Ca2+]i) of 100 nM and a membrane potential of −40 mV. This was consistent with high K+− and Cl permeability and a high intracellular Cl concentration of 40 mM. Application of ATP for 5–15 s every 3 min induced repeated [Ca2+]i increases and a 30 mV hyperpolarization. The phospholipase C inhibitor U73122 or the IP3-receptor antagonist 2-aminoethoethyl diphenyl borate suppressed ATP responses. Further biochemical and pharmacological experiments revealed that ATP responses were related to stimulation of P2Y2 and P2Y4 receptors and that the [Ca2+]i increase was a prerequisite for hyperpolarization. Inhibitors of Ca2+-activated channels or K+ channels did not affect the ATP-evoked responses. Conversely, inhibitors of Cl channels hyperpolarized cells to −70 mV and suppressed further ATP-evoked hyperpolarization. We propose that P2Y2 and P2Y4 receptors in granulosa cells modulate Cl permeability by regulating Ca2+-release.  相似文献

In January and February 2010, heavy sea ice formed along the coast of the Bohai Sea and the northern Yellow Sea, China. Intertidal organisms were subjected to serious freezing stress. In this study, we investigated the freezing tolerance of the upper intertidal economic seaweed Porphyra yezoensis. The maximum photochemical efficiency of PS II (F v/F m) in undehydrated thalli remained high after 24 h at −2°C and that in dehydrated thalli decreased in a proportion to thallial water loss. F v/F m dropped sharply after 24 h at −20°C, regardless of absolute cellular water content (AWC). The F v/F m in frozen thalli recovered rapidly at 0–20°C. A wide range of water loss in the thalli enhanced their tolerance to freezing. F v/F m values in undehydrated thalli dropped sharply after 3 d at −2°C or 10 d at −20°C while those in dehydrated thalli (20–53% AWCs) remained at high levels after 9 d at −2°C or 30 d at −20°C. These results indicate that P. yezoensis has high freezing tolerance by means of dehydration during the ebb tide and rapid recovery of F v/F m from freezing. A strategy of P. yezoensis industry to avoid heavy loss during freezing season is discussed based on these findings.  相似文献

The physiology of hyper-salinity tolerance in teleost fish: a review   总被引:3,自引:0,他引:3  
Hyper-saline habitats (waters with salinity >35 ppt) are among the harshest aquatic environments. Relatively few species of teleost fish can tolerate salinities much above 50 ppt, because of the challenges to osmoregulation, but those that do, usually estuarine, euryhaline species, show a strong ability to osmoregulate in salinities well over 100 ppt. Typically, plasma Na+ and Cl concentrations rise slowly or not at all up to about 65 ppt. At higher salinities ion levels do rise, but the increase is small relative to the magnitude of increase in concentrations of the surrounding water. A number of adjustments are responsible for such strong osmoregulation. Reduced branchial water permeability is indicated by the observation that with the exposure to hyper-salinities drinking rates rise more slowly than the branchial osmotic gradient. Lower water permeability limits osmotic water loss and greatly reduces the salt load incurred in replacing it. Still, increased gut Na+/K+-ATPase (NAK) activity is necessary to absorb the larger gut salt load and increased HCO3 secretion is required to precipitate Ca2+ and some Mg2+ in the imbibed water to facilitate water absorption. All Na+ and Cl taken up must be excreted and increased branchial salt excreting capacity is indicated by elevated mitochondrion-rich cell density and size, gill NAK activity and expression of chloride channels. Excretion of Na+ and Cl occurs against a larger gradient than in seawater and calculation of the equilibrium potential for Na+ across the gill epithelium indicates that the trans-epithelial potential required for excretion of Na+ climbs with salinity up to about 65 ppt before leveling off due to the increasing plasma Na+ levels. During acute transition to SW or mildly hyper-saline waters, some species have shown the ability to upregulate branchial NAK activity rapidly and this may play an important role in limiting disturbances at higher salinities. It does not appear that the opercular epithelium, which in SW acts in a way that is functionally similar to the gills, continues to do so in hyper-saline waters. Little is know about the hormones involved in acclimation to hyper-salinity, but the few studies available suggest a role for cortisol, but not growth hormone and insulin-like growth factor. Despite the increased transport capacity evident in both the gill and gut in hyper-saline waters there is no clear trend toward increased metabolic rate. These studies provide a general outline of the mechanisms of osmoregulation in these species, but significant questions still remain.  相似文献

Oxygen tensions and osmotic potentials are important physiological factors of plant growth and development. To optimize these variables for cotton (Gossypium hirsutum L.) embryo culture, we quantified dissolved O2 (dO2) tensions, osmotic potentials, and pH at several locations in cotton ovules during embryony. Clark O2 microelectrodes were micromanipulated into intact ovules at an angle lateral to the developing embryo, and dO2 tensions were determined in integuments, nucelli and embryos. Ovular osmotic potentials and pH were determined from extracted ovule sap using vapor pressure osmometers and pH microelectrodes. Dissolved O2 tensions near or in embryos decreased from 104 mmol m−3 at 5 days post-anthesis (DPA) to 83 mmol m−3 at 18 DPA. Osmotic potentials of ovule sap decreased from −0.70 megapascals (MPa) at 2 DPA to −1.12 MPa at 8 DPA but then increased to −0.84 MPa by 17 DPA. Ovule sap pH at 5–17 DPA varied inconsistently and ranged from 5.4 to 6.5. Based on these results, a factorial experiment with two osmotic and three O2 treatments was designed. Immature embryos of cotton cultivar HS-26 were randomly assigned to the treatment combinations and cultured for 33 days. Oxygen treatments did not affect embryo growth, and there were no differences among treatments with regard to percentage of embryos that progressed to a more advanced stage of embryo development. However, cotyledons of embryos grown without osmotic adjustment were abnormally large, and embryos exposed to these treatments were abnormally brown. Browning was less severe for embryos exposed to low O2 tensions. Growth and pigmentation were most normal for embryos simultaneously exposed to O2 tensions and osmotic potentials that best simulated the observed in ovulo conditions.  相似文献

The accumulation of inorganic and organic osmolytes and their role in osmotic adjustment were investigated in roots and leaves of vetiver grass (Vetiveria zizanioides) seedlings stressed with 100, 200, and 300 mM NaCl for 9 days. The results showed that, although the contents of inorganic (K+, Na+, Ca2+, Mg2+, Cl, NO3, SO42− and H2PO3)) and organic (soluble sugar, organic acids, and free amino acids) osmolytes all increased with NaCl concentration, the contribution of inorganic ions (mainly Na+, K+, and Cl) to osmotic adjustment was higher (71.50–80.56% of total) than that of organic solutes (19.43–28.50%). The contribution of inorganic ions increased and that of organic solutes decreased in roots with the enhanced NaCl concentration, whereas the case in leaves was opposite. On the other hand, the osmotic adjustment was only effective for vetiver grass seedlings under moderate saline stress (less than 200 mM NaCl).  相似文献

Mg2+ in various concentrations was added to purified Rubisco in vitro to gain insight into the mechanism of molecular interactions between Mg2+ and Rubisco. The enzyme activity assays showed that the reaction between Rubisco and Mg2+ was two order, which means that the enhancement of Rubisco activity was accelerated by low concentration of Mg2+ and slowed by high concentration of Mg2+. The kinetics constant (K m) and V max was 1.91 μM and 1.13 μmol CO2 mg−1 protein∙min−1, respectively, at a low concentration of Mg2+, and 3.45 μM and 0.32 μmol CO2∙mg−1 protein∙min−1, respectively, at a high concentration of Mg2+. By UV absorption and fluorescence spectroscopy assays, the Mg2+ was determined to be directly bound to Rubisco; the binding site of Mg2+ to Rubisco was 0.275, the binding constants (K A) of the binding site were 6.33 × 104 and 5.5 × 104 l·mol−1. Based on the analysis of the circular dichroism (CD) spectra, it was concluded that the binding of Mg2+ did not alter the secondary structure of Rubisco, suggesting that the observed enhancement of Rubisco carboxylase activity was caused by a subtle structural change in the active site through the formation of the complex with Mg2+.  相似文献

In traditional in vitro culture, the low CO2 concentration inside the vessels restricts photosynthesis and necessitates the addition of sucrose to the culture medium as the main energy source, thus bringing about changes in the absorption of mineral elements from the culture medium. In this study, we investigated macronutrient absorption and sugar consumption in Actinidia deliciosa Chevalier Liang and Ferguson cv. Hayward (kiwi), cultured on medium supplemented with varying amounts of sucrose (0, 10, and 20 g l−1) under both heterotrophy and autotrophy, flushed with different concentrations of CO2 (non-ventilation, 300, 600, and 2,000 μl l−1). In ventilated systems with 20 g l−1 of sucrose, sucrose absorption was less than under non-ventilation. The lowest rate of sucrose absorption was recorded when the explants were cultured on medium supplemented with 20 g l−1 of sucrose and flushed with 600 μl l−1 CO2. Absorption of NO3 , PO4 3−, and Mg2+ were high (maximum) at the end of the culture period (40 d) in explants flushed with 600 μl l−1 CO2 that have been cultured 20 d in the presence of sucrose and then transferred to a sucrose-free medium. These autotrophic conditions promoted maximum plant growth in terms of both fresh and dry mass as well as the length and number of shoots and leaves. The study shows that to maintain an optimum regime of mineral nutrition for prolonged culture of kiwi in vitro, an increased amount of these three ions should be supplemented in Murashige and Skoog’s medium.  相似文献

Fluctuating salinities at different sites on the German salt-polluted rivers Werra and Weser were compared with extracellular ion levels of specimens of Gammarus tigrinus (Sexton; Amphipoda, Crustacea), collected at the same sites. G. tigrinus regulated haemolymph concentrations of inorganic anions (Cl, SO2− 4, PO3− 4) and cations (Na+, K+, Mg2+, Ca2+) during fluctuations of salt pollution in the upper Weser. This capacity to regulate varying levels of salt pollution in the upper Weser, correlated well with the distribution of the brackish amphipods in this river ecosystem. G. tigrinus tolerated periods of Na+ and Cl stress (>380 mmol l−1) without compensating these maxima by regulating extracellular Na+ and Cl. However, during such bursts of Na+ and Cl stress in Werra and Weser, the ability to regulate extracellular [K+] at river water K+ stress of ≥6.0 mmol l−1 may explain why this brackish species has been more successful in these rivers than its competitors like Gammarus pulex. The present investigation demonstrates that the water salinity affects the [NO 3] in the haemolymph of G. tigrinus. With increasing hypo-osmotic stress the animals accumulate increasing amounts of NO 3. A simultaneous increase in stream water [NO 3] causes an additional accumulation of NO 3 in the haemolymph. The high extent of accumulation indicates that active ion transport systems may be involved. The accumulation of NO 3 in the haemolymph has low physiological consequences to G. tigrinus, but when hypo-osmotically stressed under anoxic conditions, nitrite formed by the reduction of nitrate may have an adverse affect on the metabolism of G. tigrinus. A stretch-activated (SA) Cl channel in the plasma membrane of the human mast cell line HMC-1 was identified in outside-out patch-clamp experiments. SA currents, induced by pressure applied to the pipette, exhibited voltage dependence with strong outward rectification (55.1 pS at +100 mV and an about tenfold lower conductance at −100 mV). The probability of the SA channel being open (P o) also showed steep outward rectification and pressure dependence. The open-time distribution was fitted with three components with time constants of τ1o = 755.1 ms, τ2o = 166.4 ms, and τ3o = 16.5 ms at +60 mV. The closed-time distribution also required three components with time constants of τ1c = 661.6 ms, τ2c = 253.2 ms, and τ3c = 5.6 ms at +60 mV. Lowering extracellular Cl concentration reduced the conductance, shifted the reversal potential toward chloride reversal potential, and decreased the P o at positive potentials. The SA Cl currents were reversibly blocked by the chloride channel blocker 4,4′-diisothiocyanatostilbene-2,2′-disulfonic acid (DIDS) but not by (Z)-1-(p-dimethylaminoethoxyphenyl)-1,2-diphenyl-1-butene (tamoxifen). Furthermore, in HMC-1 cells swelling due to osmotic stress, DIDS could inhibit the increase in intracellular [Ca2+] and degranulation. We conclude that in the HMC-1 cell line, the SA outward currents are mediated by Cl influx. The SA Cl channel might contribute to mast cell degranulation caused by mechanical stimuli or accelerate membrane fusion during the degranulation process.  相似文献

Biogeographic studies separate the Antarctic Notothenioid fish fauna into high- and low-latitude species. Past studies indicate that some species found in the high-latitude freezing waters of the High-Antarctic Zone have low-serum hysteresis freezing points, while other species restricted to the low-latitude seasonal pack ice zone have higher serum hysteresis freezing points above the freezing point of seawater (−1.9°C), but the relationship has not been systematically investigated. Freeze avoidance was quantified in 11 species of Antarctic icefishes by determining the hysteresis freezing points of their blood serum, in addition, the freezing point depression from serum osmolytes, the antifreeze activity from serum antifreeze glycoproteins (AFGPs), and the antifreeze activity from serum antifreeze potentiating protein were measured for each species. Serum hysteresis freezing point, a proxy for organismal freeze avoidance, decreased as species were distributed at increasing latitude (linear regression r 2 0.66, slope −0.046°C °latitude−1), which appeared largely independent of phylogenetic influences. Greater freeze avoidance at high latitudes was largely a result of higher levels of antifreeze activity from serum AFGPs relative to those in species inhabiting the low-latitude waters. The icefish fauna could be separated into a circum High-Antarctic Group of eight species that maintained serum hysteresis freezing points below −1.9°C even when sampled from less severe habitats. Fermentation of biomass derived synthesis gas to ethanol is a sustainable approach that can provide more usable energy and environmental benefits than food-based biofuels. The effects of various medium components on ethanol production by Clostridium ragsdalei utilizing syngas components (CO:CO2) were investigated, and corn steep liquor (CSL) was used as an inexpensive nutrient source for ethanol production by C. ragsdalei. Elimination of Mg2+, NH4 + and PO4 3− decreased ethanol production from 38 to 3.7, 23 and 5.93 mM, respectively. Eliminating Na+, Ca2+, and K+ or increasing Ca2+, Mg2+, K+, NH4 + and PO4 3− concentrations had no effect on ethanol production. However, increased Na+ concentration (171 mM) inhibited growth and ethanol production. Yeast extract (0.5 g l−1) and trace metals were necessary for growth of C. ragsdalei. CSL alone did not support growth and ethanol production. Nutrients limiting in CSL were trace metals, NH4 + and reducing agent (Cys: cysteine sulfide). Supplementation of trace metals, NH4 + and CyS to CSL (20 g l−1, wet weight basis) yielded better growth and similar ethanol production as compared to control medium. Using 10 g l−1, the nutritional limitation led to reduced ethanol production. Higher concentrations of CSL (50 and 100 g l−1) were inhibitory for cell growth and ethanol production. The CSL could replace yeast extract, vitamins and minerals (excluding NH4 +). The optimized CSL medium produced 120 and 50 mM of ethanol and acetate, respectively. The CSL could provide as an inexpensive source of most of the nutrients required for the syngas fermentation, and thus could improve the economics of ethanol production from biomass derived synthesis gas by C. ragsdalei.  相似文献

Rising concentrations of atmospheric CO2 are changing the carbonate chemistry of the oceans, a process known as ocean acidification (OA). Absorption of this CO2 by the surface oceans is increasing the amount of total dissolved inorganic carbon (DIC) and bicarbonate ion (HCO3 ) available for marine calcification yet is simultaneously lowering the seawater pH and carbonate ion concentration ([CO3 2−]), and thus the saturation state of seawater with respect to aragonite (Ωar). We investigated the relative importance of [HCO3 ] versus [CO3 2−] for early calcification by new recruits (primary polyps settled from zooxanthellate larvae) of two tropical coral species, Favia fragum and Porites astreoides. The polyps were reared over a range of Ωar values, which were manipulated by both acid-addition at constant pCO2 (decreased total [HCO3 ] and [CO3 2−]) and by pCO2 elevation at constant alkalinity (increased [HCO3 ], decreased [CO3 2−]). Calcification after 2 weeks was quantified by weighing the complete skeleton (corallite) accreted by each polyp over the course of the experiment. Both species exhibited the same negative response to decreasing [CO3 2−] whether Ωar was lowered by acid-addition or by pCO2 elevation—calcification did not follow total DIC or [HCO3 ]. Nevertheless, the calcification response to decreasing [CO3 2−] was nonlinear. A statistically significant decrease in calcification was only detected between Ωar = <2.5 and Ωar = 1.1–1.5, where calcification of new recruits was reduced by 22–37% per 1.0 decrease in Ωar. Our results differ from many previous studies that report a linear coral calcification response to OA, and from those showing that calcification increases with increasing [HCO3 ]. Clearly, the coral calcification response to OA is variable and complex. A deeper understanding of the biomineralization mechanisms and environmental conditions underlying these variable responses is needed to support informed predictions about future OA impacts on corals and coral reefs.  相似文献
