Suspension cultures of Eucommia ulmoides were developed and shown to accumulate chlorogenic acid. MS medium plus 2.0 mg l–1 2,4-dichlorophenoxy acetic acid was used for the cell suspension cultures of Eucommia ulmoides. The chlorogenic acid content of suspension cells was analyzed by capillary electrophoresis, and the mean content was 2.15%, approximate to that of Eucommia ulmoides leaves.  相似文献

Eucommia ulmoides Oliver is a dioecious plant, which plays an important role in traditional Chinese medicine. However, there has not yet been any research on male and female E. ulmoides. The UPLC fingerprints and OPLS-DA approach were able to quickly and easily identify and quantify E. ulmoides and differentiate between the male and female fingerprints. In this study, we optimized the UPLC conditions and analyzed them to investigate fingerprints of twenty-four extracts of Eucommiae Cortex (EC) and twenty-four extracts of Eucommiae Folium (EF) under optimal conditions. It was demonstrated that thirteen and twelve substances were possible chemical markers for EC and EF male and female discrimination and that the level of these markers – chlorogenic acid and protocatechuic acid – was many times higher in male than in female. This approach offered a reference for quality control and precise treatment of male and female E. ulmoides in the clinic.  相似文献

Pinoresinol diglucoside (PDG) is the important antihypertensive compound in Eucommia ulmoides Oliv., a traditional Chinese herb medicine. The research objective was to certify the possibility of producing PDG through fermentation. PDG-producing endophytic fungi were isolated from E. ulmoides Oliv., and the highest PDG-yielding (11.65 mg/L) isolate, XP-8, was identified as Phomopsis sp. according to the morphological characteristics and the phylogenetic tree constructed on the basis of the gene sequence in the internal transcribed spacers district. The microbial PDG was isolated by using S-8 resin and purified to a purity of 98.7% using preparative high-performance liquid chromatography (HPLC). Information obtained from the UV spectrum (277 and 227 nm, in water solution), infra-red spectrum (3,428; 2,930; 2,877; 1,637; 1,600; and 1,513; 1,460; 1,421; 1,269; 1,223; 1,075; 658 cm−1, in powder), molecular weight (682 Da, measured using HPLC-electrospray ionization mass spectrometry (ESI/MS) and tandem mass spectrometry), and nuclear magnetic resonance analysis show the microbial PDG is (+)-1-pinoresinol 4,4′-di-O-β-d-glucopyranoside, same as the plant-derived PDG. The microbial PDG is stable in pH range from 3 to 11 but less stable at temperature higher than 90 °C and in light exposure. During the fermentation, PDG production outside cells starts at the later stage of cell growth when the residual sugar in the medium was low. The study reveals the possibility for production of PDG by fermentation.  相似文献

Gibberellic acid controlled the key developmental processes of the life cycle of landing plants, and regulated the growth and development of plants. In this study, a novel gibberellin receptor gene EuGID1 was obtained from Eucommia ulmoides Oliver. The cDNA of EuGID1 was 1556 bp, and the open reading frame was 1029 bp, which encoded 343 amino acids. EuGID1 had the homology sequence with the hormone-sensitive lipase family. Amino acid sequence alignment confirmed EuGID1 protein had the highest homology with the GID1 protein of Manihot esculenta. EuGID1 was located in the nucleus and cell membrane and had expression in four plant organs. Overexpression of EuGID1 in transgenic Arabidopsis plants promoted plant elongation and increased siliques yield.  相似文献

为充分开发黄花倒水莲(Polygala fallax)的内生真菌资源,获得具有抗植物病原真菌、抗氧化活性的内生真菌,该文以黄花倒水莲内生真菌为研究对象,使用平板对峙法检测内生真菌对6种植物病原真菌的抑菌活性,测定内生真菌发酵液的DPPH清除自由基能力和总还原能力,评价内生真菌的抗氧化活性,并对具有强抑菌活性和抗氧化活性的菌株进行形态和ITS鉴定。结果表明:(1)黄花倒水莲内生真菌中有2株内生真菌对香蕉专化尖孢镰刀菌、柑橘树脂病菌、叶点霉菌、香蕉具条叶斑病菌、茄病镰刀菌、三七根腐病菌具有明显的抑菌活性,抑菌率在50.3%~91.4%之间,其中HNLF-5对柑橘树脂病菌的抑菌率为73.2%,HNLF-44对香蕉专化尖孢镰刀菌抑菌率为91.4%。(2)内生真菌发酵液具有良好的抗氧化活性,DPPH清除率均在80%以上,总还原能力吸光值范围为0.279 2~0.748 8。(3)HNLF-44菌株为链格孢属真菌。该研究表明,药用植物黄花倒水莲内生真菌具有较好的生物活性,为后续从黄花倒水莲内生真菌中挖掘潜在新型抑菌活性和抗氧化活性物质奠定了基础。  相似文献

A total of 973 isolates of endophytic fungi were recovered from 1144 tissue fragments of the six medicinal plant species belonging to 4 families collected in the Beijing Botanical Garden. Of these isolates 778 sporulated and were identified into 21 taxa by morphological characteristics. Among the taxa 11 belonged to Coelomycetes, 6 to Ascomycetes, and 4 to Hyphomycetes. Various numbers of endophytic fungi (5–8 taxa) were obtained from each plant. Alternaria alternata was the dominant species in the 6 plants, and Microsphaeropsis conielloides was also dominant in Eucommia ulmoides. There were high colonization rates (47.9%–63.1%) and isolation rates (0.7–0.93) of endophytic fungi, and they were conspicuously higher in twigs than those in leaves in the 6 plants examined. The colonization and isolation rates of endophytic fungi increased with the twig age. The results based on the analyses of cluster and Sorenson’s similarity coefficients indicated that some endophytic fungi showed a certain degree of host and tissue preference.  相似文献

以河南郑州和江苏徐州采集到的杜仲为材料,采用纯培养方法对杜仲内生真菌进行分离。将分离纯化得到的90株真菌通过形态学鉴定并进行多样性分析,90株内生真菌分别属于13个属,其中茎点霉属(Phoma)和链格孢属(Alternaria)为优势菌群,分别占总菌株数的18%和16%;其次为黑孢属(Nigrospora)和枝孢属(Cladosporium),均占总菌株数的10%。用平板对峙法对分离得到的杜仲内生真菌进行抗植物病原真菌实验,发现有18个菌株对至少一种病原真菌具有明显的拮抗作用。通过比色法进行内生真菌产IAA(吲哚乙酸)定性实验,结果显示,有37株真菌具有产IAA能力,通过分光光度法对这37株真菌进行产IAA定量实验,发现有8株有较好的产IAA活性。  相似文献

Endophytic fungi are rich in orchids and have great impacts on their host plants. 53 endophytes (30 isolates from Dendrobium devonianum and 23 endophytic fungi from D. thyrsiflorum) were isolated, respectively, from roots and stems of Dendrobium species. All the fungi were identified by way of morphological and/or molecular biological methods. 30 endophytic fungi in D. devonianum were categorized into 11 taxa and 23 fungal endophytes in D. thyrsiflorum were grouped into 11 genera, respectively. Fusarium was the dominant species of the two Dendrobium species in common. Antimicrobial activity of ethanol extract of fermentation broth of these fungi was explored using agar diffusion test. 10 endophytic fungi in D. devonianum and 11 in D. thyrsiflorum exhibited antimicrobial activity against at least one pathogenic bacterium or fungus among 6 pathogenic microbes (Escherichia coli, Bacillus subtilis, Staphylococcus aureus, Candida albicans, Cryptococcus neoformans, and Aspergillus fumigatus). Out of the fungal endophytes isolated from D. devonianum and D. thyrsiflorum, Phoma displayed strong inhibitory activity (inhibition zones in diameter >20 mm) against pathogens. Epicoccum nigrum from D. thyrsiflorum exhibited antibacterial activity even stronger than ampicillin sodium. Fusarium isolated from the two Dendrobium species was effective against the pathogenic bacterial as well as fungal pathogens. The study reinforced the assumption that endophytic fungi isolated from different Dendrobium species could be of potential antibacterial or antifungal resource.  相似文献

A rapid and efficient method for the regeneration of Eucommia ulmoides Oliver has been developed. The ability of hypocotyl segments to produce adventitious buds varied depending upon their position, seedling age and culture medium. The most adventitious buds were induced from the hypocotyl segments near to the root of 2-week-old seedlings on the Murashige and Skoog (MS) basal medium supplemented with 10 μM benzylaminopurine (BAP). However, the MS medium with 6 μM BAP and 6 μM gibberellic acid (GA3) was most suitable for shoot multiplication and further growth. Shoots longer than 2 cm cultured on the half-strength MS medium supplemented with 0.5 μM naphthaleneacetic acid (NAA) produced a maximum number of roots per shoot. Regenerated plantlets could be successfully acclimatized.  相似文献

为了从酸枣中筛选出内生菌并分析其代谢产物中的活性成分,用于开发和生产药物,该研究通过组织块分离法和划线分离法,从陕北野生酸枣植株体内分离得到内生菌,采用平板对峙法测定其对7株供试指示菌的抗菌活性,以心神宁片提取液为对照,对各拮抗菌株的发酵液进行薄层层析和高效液相色谱分析。结果表明:从野生酸枣中共分离得到121株内生菌,其中内生细菌49株,内生放线菌6株,内生真菌66株;通过抗菌试验,发现54株内生菌(细菌33株,真菌21株)对1~7种指示菌具有抗菌活性,占分离菌株总菌数的44.63%,其中A-04、A-05、B-03、C-03、C-06和D-04共6株菌株的抗菌谱较广,对7种供试指示菌均具有抑菌活性;薄层层析检测结果显示菌株B-03发酵产物在R_f值为0.46处有与酸枣提取液层析带迁移率相同的显色带,液相色谱分析结果显示其属于黄酮类物质;通过16S rRNA基因序列分析结果显示菌株B-03与Bacillus axarquiensis的相似性为99%。菌株B-03能发酵产生黄酮类或产生与黄酮类类似的化合物,表明酸枣内生菌具有合成黄酮类药物的潜力。  相似文献

结合菌株的形态学特征和ITS序列分析结果,对1株分离自杜仲茎部的内生真菌菌株DZ05进行鉴定,并对其在PDA液体培养基摇床培养3 d获得的发酵液对多种测试菌进行抗菌活性研究。结果显示:(1)分离自杜仲茎部的内生真菌菌株DZ05经形态学特征和ITS序列分析,被鉴定为淡紫色拟青霉。(2)其发酵液的乙酸乙酯提取物对6种测试细菌和9种测试植物病原菌均具有明显的抑菌活性,抑菌圈直径在13~45 mm之间,其中对番茄灰霉病菌、番茄叶霉病菌和苹果炭疽病菌抑菌圈直径>40 mm。研究表明,杜仲内生真菌DZ05的代谢产物具有广谱的抗菌活性,在植物病原菌的生物防治领域具有较大的应用前景。  相似文献

Gentiana macrophylla is a traditional Chinese medicinal plant. Its dominant active constituents are secoiridoids, mainly gentiopicrin. The objective of this study was to determine whether endophytic fungi isolated from this plant produce the bioactive ingredient gentiopicrin. Primary screening was done by Dragendorff's reaction and the strain re-selection was done with thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC) to identify the fermentation products of the selected strains. In this study, 20 strains of endophytic fungi were isolated from G. macrophylla, and the extracts from five strains had a positive Dragendorff's reaction. Two strains (QJ16 and QJ18) had a component with the same Rf value in TLC as that of authentic gentiopicrin and one ingredient of the QJ18 extract had a retention time identical with that of authentic gentiopicrin in HPLC. Therefore, the fungus appears to produce the bioactive ingredient gentiopicrin, as does its host plant, and could be used for the production of gentiopicrin by fermentation.  相似文献

Among 67 endophytic fungi isolated from Quercus variabilis, 53.7% of endophytic fungal fermentation broths displayed growth inhibition on at least one test microorganism, such as pathogenic fungi (Trichophyton rubrum, Candida albicans, Aspergillus niger, Epidermophyton floccosum, Microsporum canis) and bacteria (Escherichia coli, Bacillus subtilis, Pseudomonas fluorescens). Moreover, 19.4% of strains showed a broader antimicrobial spectrum, such as Aspergillus sp., Penicillium sp., Alternaria sp., 20.9% of strains showed strong inhibition (+++) to pathogenic bacteria, while only 7.5% displayed that to test fungi. The most active antifungal strain I(R)9-2, Cladosporium sp. was selected and fermented. From the broth, a secondary metabolite, brefeldin A was obtained. This is the first report on the antimicrobial potentials of endophytic fungi residing in Q. variabilis and isolation of brefeldin A produced by Cladosporium sp.  相似文献

A study was conducted for isolation, identification and antibacterial potential of fungal endophytes of Adenocalymma alliaceum Miers., (Bignoniaceae), a medicinal shrub vine plant which has long history for its usages in curing various disorders. A total of 149 isolates of endophytic fungi representing 17 fungal taxa were obtained from 270 segments (90 from each stem, leaf and petiole) of this plant. Hyphomycetes (77.85%) were the most prevalent, followed by Ascomycetes (8.05%) and Coelomycetes (4.03%) respectively. A considerable amount of fungal isolates was kept under (10.07%) Mycelia-Sterilia (MS). Leaf harboured maximum colonization of endophytic fungi (72.22%) which was greater than stem (67.78%) and petiole (25.54%). The Jc similarity index was maximum (0.619) between stem vs leaf followed by leaf vs petiole (0.571) and stem vs petiole (0.428). The dominant endophytic fungi were Alternaria alternata, Aspergillus niger, Stenella agalis, Fusarium oxysporum, Curvularia lunata and Fusarium roseum. Among twelve endophytic fungi tested for antibacterial activity, crude extracts of nine endophytic fungi (75%), showed antibacterial potential against one or more clinical human pathogens. Alternaria alternata, Curvularia lunata, Penicillium sp. and Chaetomium globosum exhibited significant antibacterial activity against 4 of 5 tested pathogens, showing broad spectrum activity. This investigation explains the value of sampling from different tissues of a host plant for the greater species diversity, and additionally, the antibacterial screening of some endophytic fungi from this specific medicinal plant may represent a unique source for many of the useful antibacterial compounds.  相似文献

We investigated the modulation of innate and adaptive immune cell activation by Eucommia ulmoides Oliver extract (EUE) and its ingredient genipin. As an innate immunity indicator, the phagocytic activity of macrophages was determined by measuring engulfed, fluorescently labeled Escherichia coli. As a surrogate marker for the respective activation of cellular and humoral adaptive immunity, concanavalin A (Con A) and lipopolysaccharide (LPS) induction of primary splenocyte proliferation was assayed in in vitro and ex vivo systems. EUE and genipin suppressed the proliferation of primary splenic lymphocytes induced by Con A or LPS, but not macrophage phagocytosis. Oral administration of EUE and genipin to mice decreased splenic lymphocyte proliferation induced by Con A or LPS. These results revealed that E. ulmoides and genipin suppressed cellular and humoral adaptive immunity, and they suggest that E. ulmoides and genipin are promising candidates for immunosuppressive drugs that target diseases that involve excessive activation of adaptive immunity.  相似文献

Eucommia ulmoides Oliver is a natural product widely used as a dietary supplement and medicinal plant. Here, we examined the potential regulatory effects of Eucommia ulmoides Oliver extracts (EUE) on hepatic dyslipidemia and its related mechanisms by in vitro and in vivo studies. EUE and its two active constituents, aucubin and geniposide, inhibited palmitate-induced endoplasmic reticulum (ER) stress, reducing hepatic lipid accumulation through secretion of apolipoprotein B and associated triglycerides and cholesterol in human HepG2 hepatocytes. To determine how EUE diminishes the ER stress response, lysosomal and proteasomal protein degradation activities were analyzed. Although proteasomal activity was not affected, lysosomal enzyme activities including V-ATPase were significantly increased by EUE as well as aucubin and geniposide in HepG2 cells. Treatment with the V-ATPase inhibitor, bafilomycin, reversed the inhibition of ER stress, secretion of apolipoprotein B, and hepatic lipid accumulation induced by EUE or its component, aucubin or geniposide. In addition, EUE was determined to regulate hepatic dyslipidemia by enhancing lysosomal activity and to regulate ER stress in rats fed a high-fat diet. Together, these results suggest that EUE and its active components enhance lysosomal activity, resulting in decreased ER stress and hepatic dyslipidemia.  相似文献

为了评价大面积人工种植杜仲对当地植物多样性的影响,以河南省汝阳县不同植被类型为研究对象,通过群落学调查,运用重要值、Shannon-Wiener物种多样性指数(H)、Simpson物种多样性指数(D)和均匀度指数(JH')等指标,统计分析杜仲人工林、温带落叶阔叶林(以下简称次生林)和撂荒地3种不同植被类型的植物物种多样性,探讨杜仲种植对植物多样性的影响。调查发现,杜仲林样地中出现植物82种,隶属39科63属,草本层为最发达的一层;次生林样地中出现植物70种,隶属32科62属,乔木层为最发达层;撂荒地样地中出现植物84种,隶属35科69属,无乔木层。杜仲林物种丰富度和多度均不亚于次生林和撂荒地,且杜仲林草本层物种丰富度和植株总数均高于次生林。统计分析显示,杜仲林乔木层、灌木层和草本层多样性指数H和D值、均匀度指数JH'值与其它两种植被类型相比无显著性差异(P0.05)。因此,种植杜仲过程中采用合理密度,适当管理,不仅能提供叶、花、果等资源,而且能够丰富草本植物的种类和数量,增加植物物种多样性。  相似文献
