Recovery results of Staphylococcus aureus ATCC 6538 treated with phenolics and quaternary ammonium compounds on Dey and Engley (D/E) neutralizing medium at various time intervals were compared by the use of two commonly used media. Two recovery processes were utilized. In one, the chemically treated organisms were plated directly onto an agar medium. In the other, the aliquot was first put in broth and then was plated with agar. By either process, the numbers and the time period for recovery of organism were greater on D/E medium.  相似文献

Letheen broth and Letheen agar have been investigated for their ability to act as neutralising and recovery media for wild-type and envelope mutants exposed to chlorhexidine diacetate and benzalkonium chloride. At high dilutions, untreated cells of the envelope mutant of Pseudomonas aeruginosa 799/61 were unable to produce colonies on Letheen agar. As a result of various procedures, it was concluded that dilution in Letheen broth and plating in Isosensitest agar was a suitable method for quenching cationic bactericides without harming the test strains, and that the increasing use of Gram-negative bacteria with outer membrane defects means that considerable care may be necessary in selecting media for evaluating bactericidal activity.  相似文献

We established a plant regeneration system for Hinoki cypress (Chamaecyparis obtusa) via somatic embryogenesis. Embryogenic tissues were successfully induced on three kinds of Smith media from megagametophyte explants containing pre-cotyledonary embryos of C. obtusa plus-trees. Factors affecting somatic embryo maturation were examined. The concentration of polyethylene glycol 4000 in the medium was a critical factor for embryo maturation and its effective concentration was 150 g/l. The addition of 30 g/l maltose to the medium had a positive effect on embryo maturation, but sucrose was ineffective. The mature somatic embryos germinated at a germination frequency of approximately 60%, and the presence of activated charcoal was effective in stimulating plantlet growth. The plantlets acclimatized successfully in a greenhouse. Suspension culture of Ajuga lobata D. Don cells provides a method of synthesis of the phytoecdysteroid 20-hydroxyecdysone (20E) which can regulate the molting process of larvae. We characterized the culture conditions to optimize 20E production. Growth of A. lobata D. Don cells fits the logistic equation curve with a growth cycle of 19 days. Medium conductivity was negatively correlated with dry cell weight and 20E accumulation, thus could be used to determine the optimal time for cell harvest. Continuous subculture reduced 20E synthesis, but supplementing medium with 20E precursors mevalonic (MVA), α-Pinene, and nitric oxide (NO) can significantly promote cell growth and influence 20E accumulation. Combination of α-Pinene, MVA, and SNP significantly elevated 20E accumulation, thus may synergistically enhance 20E synthesis in A. lobata D. Don. The optimal concentrations of α-Pinene, MVA, and NO donor SNP in suspension culture were 50 μL L?1, 10 mg L?1, and 80 μmol L?1.  相似文献

Two new varieties of Kuepferia pringlei D. Maity & S. K. Dey from north Sikkim (eastern Himalaya) are described, illustrated, and compared with closely related taxa of similar morphology. Details on its habitat, phenology and conservation status are provided. These previously undescribed varieties are most similar to, but distinct from, typical K. pringlei D. Maity & S. K. Dey, known from the eastern Himalaya. Both new taxa are compared with K. leucantha (Harry Sm. ex T.N. Ho & S.W. Liu) Adr.Favre.  相似文献

ABSTRACT. RPMI-1640 is routinely used as the basal medium for the in vitro maintenance of malaria parasites. In this study we tested several commercially available nutritional media in a Plasmodium chabaudi chabaudi erythrocyte invasion assay and showed that three media, BME Basal Medium—modified, Dulbecco's Modified Eagle Medium, and William's Medium E, improved the level of merozoite invasion when compared with RPMI-1640. These media improve the rate of maturation of newly invaded rings to young trophozoites. Radioisotope incorporation by trophozoites maintained in these three media was also improved when compared to trophozoites maintained in RPMI-1640. BME Basal Medium—modified, or a combination of three parts BME Basal Medium—modified with one part William's Medium E, supported higher levels of erythrocyte invasion by merozoites. We suggest that either of these media replace the currently used RPMI-1640 for in vitro studies on P. c. chabaudi.  相似文献

Embryo-rescue was used to facilitate interspecific hybridization of Cucumis anguria L. and C. zeyheri Sond. Embryos were excised from developing fruits at one week intervals for six weeks after hand pollination. Medium containing coconut water was the most suitable for initial germination, and a medium with ascorbic acid was the best for embryo development and plant recovery. Viable plants were obtained from embryos and these plants showed morphological characteristics different from both parents. The analysis of the leucine aminopeptidase (LAP) locus revealed three hybrid types, H1.1, H1.2 and H2.  相似文献

Seedlings were grown in vitro from embryos of Dipterocarpus alatus and D. intricatus. The problem of explant browning could be overcome by growing embryos initially on a filter paper bridge in liquid medium with activated charcoal. The best basal medium was Woody Plant Medium without the ammonium nitrate. Cytokinin appeared to stimulate seedling growth, 5×10-5 M 2-isopentenyladenine and 10-4 M 6-benzyladenine (BA) being the optimum concentrations for D. alatus and D. intricatus respectively. Cotyledonary nodes, excised from the seedlings, were induced to form axillary shoots and in the case of D. intricatus these could be multiplied rapidly. D. intricatus shoots elongated by reducing the BA level from 10-5 M to 5×10-7 M. Aims: The equivalence of Oxoid (CM 1046) Brilliance(TM)Ecoli/coliform selective agar to mFC agar, as used in the Australian/New Zealand Standard Method to detect thermotolerant coliforms and Escherichia coli in water samples, was assessed. Methods and Results: A total of 244 water samples were analysed in parallel over a 5‐month period. Sewage effluent samples (n = 131, sites = 43), freshwater (n = 62, sites = 18) and marine/brackish water samples (n = 51, sites = 23) were analysed. The Wilcoxon matched‐pairs signed‐ranks test showed a varying degree of statistical difference between the two methods. All matrices had a higher recovery in the trial method. Enterococci faecalis, Aeromonas spp. and Vibrio spp. did not grow on the CM1046 agar, and Pseudomonas aeruginosa and Enterobacter aerogenes were inhibited. Conclusions: The use of CM 1046 for the detection and enumeration of E. coli and thermotolerant coliforms in water samples is a suitable alternative to the AS/NZS Standard Method. Significance and Impact of the study: The use of CM1046 agar was less labour intensive and time consuming, as no secondary confirmation steps were required. Confirmed results could be reported within 24 h of sample analysis, as compared to 48 h with the reference method. Public health concerns can be addressed in a more efficient manner.  相似文献

AIMS: To examine the effect that starvation and sodium hypochlorite stress have on virulence of Escherichia coli O157:H7 and the influence of conditioned media on recovery of stressed cells. METHODS AND RESULTS: Escherichia coli O157:H7 was starved for 5 days then exposed to 1 microg ml(-1) sodium hypochlorite, suspended in defined media Dulbecco's Modified Eagle's Medium supplemented with conditioned media, sampled over a 12-h period; and assayed for growth, production of Shiga toxin (Stx), and attachment to HCT-8 cells. During recovery, stressed and control cells grown in conditioned media exhibited greater attachment efficiencies to HCT-8 cells then cells in DMEM alone. Production of Stx by treated cells mimicked Stx production by control cells suggesting that components of conditioned media assist in recovery. Results showed that levels of autoinducer-2 fluctuate during recovery and growth suggesting involvement of a quorum sensing mechanism during the recovery of stressed E. coli O157:H7. CONCLUSIONS: The recovery of stressed E. coli O157:H7 exposed to starvation conditions and HOCl is positively affected by the presence of autoinducer-2 thereby influencing virulence factor production. SIGNIFICANCE AND IMPACT OF THE STUDY: Food-borne pathogens in a stressed state prior to ingestion can rapidly recover in the presence of bacterial by-products; exhibiting virulence characteristics and presenting a microbial food safety hazard.  相似文献

To arrive at an appropriate bioreactor design and in situ recovery of the products, red beet hairy roots were used as a model system where the levels of betalain pigments (betacyanins and betaxanthins) were followed as secondary metabolite and the peroxidase enzyme as primary metabolite. Medium volume and other kinetic parameters were found to play significant roles by way of directly affecting the biomass yield rather than a specific metabolite. The hydrodynamic stress created on the roots by large culture volume could be minimized by pulse‐feeding of medium in shake‐flasks; and by separating the biomass chamber from the aerated medium reservoir in circulatory fed‐batch bioreactor. Accordingly the bioreactor was modified to provide anchorage and air‐enrichment chamber which resulted in higher formation of both the metabolites than in shake‐flasks. Various down‐stream processing aspects such as in situ release of pigments by non‐destructive methods, followed by adsorption through a column and recovery by desorption were optimized for betalains. A strategy for simultaneous recovery of pigment and peroxidase was worked out using aqueous two phase extraction (ATPE).  相似文献

Dietary treatment with three diets differing in vitamin E, Low E (15 mg of vitamin E/kg diet), Medium E (150 mg/kg), or High E (1,500 mg/kg), resulted in guinea pigs with low (but nondeficient), intermediate, or high heart a-tocopherol concentration. Neither the antioxidant enzymes superoxide dismutase, catalase, glutathione peroxidase, and reductase, nor the nonenzymatic antioxidants, GSH, ascorbate, and uric acid were homeostatically depressed by increases in heart a-tocopherol. Protection from both enzymatic (NADPH dependent) and nonenzymatic (ascorbate-Fe2+) lipid peroxidation was strongly increased by vitamin E supplementation from Low to Medium E Whereas no additional gain was obtained from the Medium E to the High E group. The GSH/GSSG and GSH/total glutathione ratios increased as a function of the vitamin E dietary concentration closely resembling the shape of the dependence of heart a-tocopherol on dietary vitamin E. The results show the capacity of dietary vitamin E to increase the global antioxidant capacity of the heart and to improve the heart redox status in both the lipid and water-soluble compartments. This capacity occurred at levels six times higher than the minimum daily requirement of vitamin E, even in the presence of optimum dietary vitamin C concentrations and basal unstressed conditions. The need for vitamin E dietary supplementation seems specially important in this tissue due to the low constitutive levels of endogenous enzymatic and nonenzymatic antioxidants present of the mammalian heart in comparison with those of other internal organs.  相似文献

Eulophia alta (Linnaeus) Fawcett & Rendle seeds collected from the Florida Panther National Wildlife Refuge (Collier County, FL; FPNWR) were used in a screen of five asymbiotic orchid seed germination media to determine their effectiveness in promoting germination and protocorm development. In addition, 10 fungal isolates collected from the roots of E. alta at sites in the FPNWR, Highlands County (FL), and Goethe State Forest (Levy County, FL; GSF), and a fungal isolate from the roots of Spiranthes brevilabris collected from GSF were screened for their effectiveness at promoting in vitro symbiotic germination of E. alta seeds. After 18 weeks asymbiotic culture, seeds sown on PhytoTechnology Orchid Seed Sowing Medium germinated to a higher percentage (87.9%) and had a higher percentage of protocorms with developing protomeristems (32.7%) than seeds cultured on Knudson C, Malmgren Modified Terrestrial Orchid Medium, ?-strength Murashige & Skoog, or Vacin & Went Modified Orchid Medium. Significantly more leaf-bearing protocorms were observed on PhytoTechnology Orchid Seed Sowing Medium (0.8%) and Vacin & Went Modified Orchid Medium (1.3%) than other media tested. Of the fungi tested, one fungal isolate (Ealt-396) promoted germination to 69.0%, two isolates promoted germination to less than 0.75% and did not support further protocorm development, and eight isolates did not support germination. Seeds co-cultured in darkness with Ealt-396 grew more rapidly than asymbiotic seedlings following germination. In addition, co-cultured (=symbiotic) seedlings continued to develop more rapidly than asymbiotic seedlings upon transfer to 16/8 h light/dark photoperiod. Symbiotic seed culture of E. alta may be a more desirable method of propagation since protocorms develop more rapidly than seeds sown on asymbiotic media. Symbiotic seedlings may be more appropriate for reintroduction to natural areas than asymbiotic seedlings since symbiotic seedlings could serve to inoculate soils with a germination promoting mycobiont.  相似文献

We used site-directed mutagenesis to investigate the role of Glu(69), Asp(70), Asp(71), Asp(72), Tyr-sulfate(73), and Asp(75) in the second acidic region (AR2) of the serpin heparin cofactor II (HCII) during formation of the thrombin.HCII complex with and without glycosaminoglycans. E69Q/D70N/D71N recombinant (r)HCII, D72N/Y73F/D75N rHCII, and E69Q/D70N/D71N/D72N/Y73F/D75N rHCII were prepared to localize acidic residues important for thrombin inhibition. Interestingly, D72N/Y73F/D75N rHCII had significantly enhanced thrombin inhibition without glycosaminoglycan (4-fold greater) and with heparin (6-fold greater), showing maximal activity at 2 microg/ml heparin compared with wild-type recombinant HCII (wt-rHCII) with maximal activity at 20 microg/ml heparin. The other rHCII mutants had lesser-enhanced activities, but they all eluted from heparin-Sepharose at significantly higher ionic strengths compared with wt-rHCII. Neutralizing and reversing the charge of Asp(72), Tyr-sulfate(73), and Asp(75) were done to characterize their individual contribution to HCII activity. Only Y73K rHCII and D75K rHCII have significantly increased heparin cofactor activity compared with wt-rHCII; however, all of the individual rHCII mutants required substantially less glycosaminoglycan at maximal inhibition than did wt-rHCII. Inhibition of either alpha-thrombin/hirugen or gamma(T)-thrombin (both with an altered anion-binding exosite-1) by the AR2 rHCII mutants was similar to wt-rHCII. D72N/Y73F/D75N rHCII and D75K rHCII were significantly more active than wt-rHCII in a plasma-based thrombin inhibition assay with glycosaminoglycans. These results indicate that improved thrombin inhibition in the AR2 HCII mutants is mediated by enhanced interactions between the acidic domain and anion-binding exosite-1 of thrombin and that AR2 may be a "molecular rheostat" to promote thrombin inhibition in the presence of glycosaminoglycans.  相似文献

The protein D1 was obtained from nuclei of Drosophila melanogaster embryos and purified by perchloric acid fractionation and preparative gel electrophoresis. In nuclei its amount is approximately 1% of the amount of DNA by weight. D1 is soluble in 5% perchloric acid and extractable from nuclei by solutions of moderate ionic strength (0.35 M NaCl). Amino acid analysis shows that it is rich in both basic (20%) and acidic (27%) aminoacids. In all these properties D1 resembles HMG proteins (high mobility group; Johns et al., 1975) of vertebrates; however, its apparent molecular weight (50,000) is much higher. The distribution of D1 in salivary gland polytene chromosomes was investigated by immunofluorescence. Two levels of fluorescence intensity were observed: 1) Very bright fluorescence at chromosomal positions 81F, 83E, 101F, 102C and 102F; these sites are shown, by double labeling techniques, to coincide with quinacrine bright sites. 2) Medium to low fluorescence at many sites widely distributed throughout all chromosomes. In order to interpret these results and to relate them to the in vivo distribution of D1, we have investigated the pattern of immunofluorescence staining as a function of the methods of chromosome preparation and salivary gland fixation. The immunological specificity of the anti-D1 serum was studied by comparing its reactivity with D. melanogaster and D. virilis chromosome spreads and whole salivary glands, and by using reagents that minimize non-specific antibody interactions. We conclude that Dl is widely distributed throughout cytoplasm and nucleus, present in many chromomeres but most abundant in chromosomal sites that contain the AT-rich satellite DNA of density 1.672. This distribution, together with available evidence about the nucleotide sequences present in this satellite, suggests that D1 binds preferentially to chromatin containing sequences AATAT and/or AATATAT.  相似文献

Many water utilities are required to monitor source water for the presence of total coliforms, fecal coliforms, or both. The Colilert system, an application of the defined substrate technology, simultaneously detects the presence of both total coliforms and Escherichia coli directly from a water sample. After incubation, the formula becomes yellow if total coliforms are present and fluorescent at 366 nm if E. coli is in the same sample. No confirmatory tests are required. The Colilert system was previously assessed with distribution water in a national evaluation in both most-probably-number and presence-absence formats and found to produce data equivalent to those obtained by using Standard Methods for the Examination of Water and Wastewater (Standard Methods). The Colilert system was now compared with Standard Methods multiple-tube fermentation (MTF) for the enumeration of total coliforms and E. coli from surface water. All MTF tubes were confirmed according to Standard Methods, and subcultures were made to identify isolates to the species level. The Colilert system was found equally sensitive to MTF testing by regression, t test, chi-square, and likelihood fraction analyses. Specificity of the Colilert system was shown by the isolation of a species of total coliform or E. coli after the appropriate color change. The Colilert test can be used for source water samples when enumeration is required, and the benefits previously described for distribution water testing--sensitivity, specificity, less labor, lower cost, faster results, no noncoliform heterotroph interference--are applicable to this type of water analysis.  相似文献

Both obesity and aging increase intrahepatic fat (IHF) content, which leads to nonalcoholic fatty liver disease (NAFLD) and metabolic abnormalities such as insulin resistance. We evaluated the effects of diet and diet in conjunction with exercise on IHF content and associated metabolic abnormalities in obese older adults. Eighteen obese (BMI ≥30 kg/m2) older (≥65 years old) adults completed a 6‐month clinical trial. Participants were randomized to diet (D group; n = 9) or diet + exercise (D+E group; n = 9). Primary outcome was IHF quantified by magnetic resonance spectroscopy (MRS). Secondary outcomes included insulin sensitivity (assessed by oral glucose tolerance), body composition (assessed by dual‐energy X‐ray absorptiometry), physical function (VO2peak and strength), glucose, lipids, and blood pressure (BP). Body weight (D: ?9 ± 1%, D+E: ?10 ± 2%, both P < 0.05) and fat mass (D: ?13 ± 3%, D+E ?16 ± 3%, both P < 0.05) decreased in both groups but there was no difference between groups. IHF decreased to a similar extent in both groups (D: ?46 ± 11%, D+E: ?45 ± 8%, both P < 0.05), which was accompanied by comparable improvements in insulin sensitivity (D: 66 ± 25%, D+E: 68 ± 28%, both P < 0.05). The relative decreases in IHF correlated directly with relative increases in insulin sensitivity index (ISI) (r = ?0.52; P < 0.05). Improvements in VO2peak, strength, plasma triglyceride (TG), and low‐density lipoprotein–cholesterol concentration, and diastolic BP occurred in the D+E group (all P < 0.05) but not in the D group. Diet with or without exercise results in significant decreases in IHF content accompanied by considerable improvements in insulin sensitivity in obese older adults. The addition of exercise to diet therapy improves physical function and other obesity‐ and aging‐related metabolic abnormalities.  相似文献
