Pinus hakkodensis has been considered as a hybrid betweenP. pumila andP. parviflora var.pentaphylla. Chloroplast DNA typing of this putative hybrid and hypothesized parental species in the Tanigawa Mountains, Japan, was conducted by PCR and single-strand conformation polymorphism (SSCP) of the intergenic spacer betweentrnL(UAA)3′ exon andtrnF(GAA) of cpDNA. Each of the hypothesized parental species collected from other mountain regions displayed a diagnostic SSCP pattern, whereas all morphological intermediates sampled in the Tanigawa Mountains had the SSCP pattern ofP. parviflora var.pentaphylla. Furthermore, some individuals classified on the basis of needle morphology as belonging toP. pumila in this mountain region showed the SSCP pattern ofP. parviflora var.pentaphylla. This may suggest that pollen-mediated uni-directional introgression fromP. parviflora var.pentaphylla toP. pumila occurs in the Tanigawa Mountains.  相似文献

Pinus species exhibit paternal chloroplast inheritance and maternal mitochondrial inheritance. This independent inheritance of two cytoplasmic genomes provides an exceptional environment for discriminating female (seeds) and male (pollen) components of gene flow across hybridizing species. We obtained mitochondrial genetic markers diagnostic toP. parviflora var.pentaphylla andP. pumila by PCR amplification of the intron ofnad1 on mtDNA, and examined the spatial-distribution pattern of the mtDNA haplotypes in a hybrid zone betweenP. parviflora var.pentaphylla andP. pumila in the Tanigawa Mountains of Japan. These data, in conjunction with previous information on cpDNA haplotypes and needle morphology, revealed contrastive patterns of introgression of two cytoplasmic genomes. CpDNA introgression has occurred uni-directionally fromP. parviflora var.pentaphylla toP. pumila. Conversely, mtDNA introgression has occurred in the opposite direction, fromP. pumila toP. parviflora var.pentaphylla. Levels of introgression are roughly equivalent for cpDNA and mtDNA. The contrastive spatial distribution pattern of cpDNA and mtDNA haplotypes could be caused by differential movement of seeds and pollen for interspecific genetic exchange.  相似文献

Uredinial and telial states of aMelampsora species occurring on the leaves ofSalix serissaefolia were for the first time recorded. Field observations and inoculation experiments showed that the spermogonial and aecial states of the fungus were formed onChelidonium majus var.asiaticum andCorydalis incisa. The fungus was identified asMelampsora yezoensis based on the morphological observation of all the spore states. Urediniospores were able to infectS. erocarpa andS. pierotii as well asS. serissaefolia andS. jessoensis. Salix serissaefolia, S. eriocarpa, andS. pierotii are new uredinial and telial hosts ofM. yezoensis, andC. majus var.asiaticum andC. incisa are new spermogonial and aecial hosts of the fungus. Aplopsora corni sp. nov. is proposed for a rust fungus whose uredinial and basidial stage occurs onCornus controversa (Cornaceae) in Hokkaido. Six fungi isolated from plant materials in Japan are described. The first isPseudohalonectria aomoriensis sp. nov. (Lasiosphaeriaceae). It differs from other known species of the genusPseudohalonectria in the dimensions of its asci and ascospores. The second,Monodictys abuensis, is newly added to the mycoflora of Japan. Its host,Zelkova serrata, is also newly recorded. Three others found on new host plants areDictyochaeta simplex onQuercus myrsinaefolia andBladhia crispa, Colletotrichum dematium onAucuba japonica var.borealis, andNectria mammoidea var.rubi onCercidiphyllum japonicum. The natural reservoir ofHistoplasma capsulatum var.duboisii, the etiological agent of histoplasmosis duboisii (African histoplasmosis) is not yet known. We report the isolation ofH. capsulatum var.duboisii from soil admixed with bat guano and from the intestinal contents of a bat in a sandstone cave in a rural area, Ogbunike in Anambra State of Nigeria. Eight of 45 samples of soil admixed with bat guano yieldedH. capsulatum var.duboisii. Of the 35 bats belonging to the speciesNycteris hispida andTadirida pumila examined, only one (N. hispida) yielded this fungus from its intestinal contents. Identification of the isolates asHistoplasma was confirmed by exoantigen tests and by mating with tester strains ofH. capsulatum. In vitro conversion to large yeast from suggestive ofH. capsulatum var.duboisii was obtained on brain heart infusion agar supplemented with sheep blood and glutamine or cysteine. Pathogenicity tests with mice for all the isolates confirmed their identity by the demonstration of large yeast forms (8–15 µm in diameter) within giant cells in the infected tissues. Erysiphe cumminsiana andE. galeopsidis, which have immature asci in the current season, have been recorded from Japan, but the ascospores of both fungi have not been described. In the present experiments, some observations before and after overwintering were made on the cleistothecia ofE. cumminsiana on three species ofCacalia and two species ofLigularia, andE. galeopsidis onGeranium thunbergii. After overwintering, the former fungus developed six to eight, rarely four spores in an ascus and the latter fungus always four spores in an ascus. Catinaria grossa is lichenized withDictyochloropsis splendida var.gelatinosa, var. nova. When cultured isolated from the fungus the cells of this variety are covered individually by thick gelatinous envelopes. The phycobionts ofMegalospora gompholoma subsp.gompholoma andM. atrorubicans subsp.australis belong to a new variety ofD. symbiontica, i.e. var.pauciautosporica, which preferentially reproduces by zoo- and aplanospores. The phycobiont ofPseudocyphellaria aurata does not belong toMyrmecia reticulata as formerly thought, but toDictyochloropsis symbiontica. Specimens from one locality in Tenerife and from New Zealand are lichenized with a strain ofD. symbiontica var.symbiontica, those from another locality in Tenerife with a strain ofD. symbiontica var.pauciautosporica. These strains differ in certain characters from other lichenized strains of these varieties.

The Asian pine gall rust, which has hitherto been assigned toCronartium quercuum distributed in North America, is described asC. orientale, sp. nov. The spermogonial and aecial states occur on hard pines, and the uredinial and telial states occur on various oaks.Cronartium orientale has globose, almost hyaline basidiospores in contrast to the ellipsoid, yellow-orange ones of North AmericanC. quercuum s.l. Food preferences and nematode parasitism were studied in natural populations of mycophagousDrosophila in and near Sapporo, northern Japan. Species which preferred fresh mushrooms showed species-specific responses toPleurotus mushrooms:D. pirka bred only onPleurotus cornucopiae, D. trivitata onP. cornucopiae andP. ostreatus, D. trilineata on these twoPleurotus mushrooms and some other mushrooms, whileD. sexvittata bred on a wide variety of mushrooms but seldom onPleurotus mushrooms. Species which preferred decayed mushrooms (D. quadrivittata, D. histrioides, D. testacea and species of thequinaria species-group) showed host preferences different from those of the above species. The rate of parasitism by nematodes was generally higher in species which prefer decayed mushrooms than in species which prefer fresh mushrooms. Among species which prefer fresh mushrooms, onlyD. trilineata was parasitized frequently by nematodes. It was not clear what factors determine the rate of parasitism in these mycophagousDrosophila. D. pirka, D. trivittata andD. trilineata passed through three or four generations per year and entered reproductive diapause in early September in and near Sapporo. A taxonomic revision ofPuccinia species causing rust diseases on sugarcane was conducted to clarify their morphological characteristics. Specimens including previously reported species,Puccinia melanocephala, P. kuehnii andPuccinia sp.sensu Muta, 1987, were collected in Japan and the Philippines and borrowed from various herbaria worldwide. Morphological characteristics of these specimens were examined under light and scanning electron microscopes. Comparative morphological studies of the specimens showed that rust fungi infecting sugarcane could be classified into two species,Puccinia melanocephala andP. kuehnii. Puccinia sp.sensu Muta was morphologically identical withP. kuehnii. Results of this study corroborate previous phylogenetic analysis results of D1/D2 regions of LSU rDNA gene. A species ofMycosphaerella with aPseudocercospora anamorph was collected on overwintered fallen leaves of Japanese beech,Fagus crenata. Based on comparison of morphology withMycosphaerella species on Fagaceae, the fungus was newly described asMycosphaerella buna. The genusCryptococcus was found to be heterogeneous on the basis of partial rRNA sequences. The human-pathogenic speciesC. neoformans, comprising 4 serotypes and havingFilobasidiella neoformans andF. bacillispora as teleomorphs, was found at a relatively large distance fromFilobasidium. Serotypes B and C had identical sequences, while in A and D they were different, with D closer to B and C than to A.Filobasidiella depauperata, which lacks a yeast-like anamorph, clustered withF. neoformans.The genusFilobasidium was clearly separated fromFilobasidiella and clustered withC. albidus, C. kuetzingii, C. gastricus, C. lupi, C. vishniaciae, C. bhutanensis, C. aerius, C. terreus andC. ater. The latter may represent the anamorph ofFilobasidium elegans. The organe to red species ofCryptococcus, as well asC. aquaticus andC. yarrowii, were found completely unrelated with these taxa,C. macerans being affiliated toCystofilobasidium capitatum.The genusTrichosporon was found relatively homogeneous; it includesC. humicola, C. curvatus and the filamentous speciesHyalodendron lignicola. Cryptococcus flavus andC. dimennae probably belong to the Tremellales, though distances between these species are large.
