Spermatozoa of the sea urchin, Glyptocidaris crenularis, use endogenous triglycerides (TG), not phosphatidylcholine, to produce energy for swimming. This study was undertaken to examine ultrastructurally the location of TG available for utilization in energy metabolism in these spermatozoa. Each spermatozoon contained several lipid globules at the bottom of the midpiece. Following incubation with sea water, both the number and size of the lipid globules decreased significantly. The total volume of lipid globules in each spermatozoon was roughly halved after 1 h of incubation. Similarly, about half of the TG was metabolized during the same incubation period. Oxygen consumption by spermatozoa during the incubation indicated the oxidation of fatty acid derived from TG. Thus it appears that G. crenularis spermatozoa obtain energy through oxidation of fatty acid from TG stored in the lipid globules within their midpieces. © 1993 Wiley-Liss, Inc.  相似文献

We have investigated pathways of lipid metabolism in boar spermatozoa sperm cells incubated for up to 3 days with [14C]palmitic acid, [14C]glycerol, [14C]choline, or [14C]arachidonic acid or incorporated these precursors into diglycerides and/or phospholipids. When spermatozoa were incubated with [14C]palmitic acid or [14C]glycerol, there was first an incorporation into phosphatidic acid, followed by labelling of 1,2-diacylglycerol (DAG) and then phosphatidyl-choline (PC). This indicates that the de novo pathway of phospholipid synthesis is active in these cells. However, not all DAG was converted to PC. A pool of di-saturated DAG, which represented a considerable proportion of the high basal levels of DAG, accumulated the majority of label. Another DAG pool, containing saturated fatty acids in position 1 and unsaturated fatty acids in position 2 and representing the remaining basal DAG, was in equilibrium with PC. When spermatozoa were incubated with [14C]arachidonic acid, there was a considerable incorporation of label into PC, which indicates the presence of an active deacylation/reacylation cycle. The behaviour of certain lipid pools varied depending on the temperature at which spermatozoa were incubated. For example, in the presence of [14C]palmitic acid or [14C]arachidonic acid, there was more incorporation of label into PC when spermatozoa were incubated at 25°C than when incubated at 17°C. Taken together, these results indicate that spermatozoa have an active lipid synthetic capacity. It may therefore be possible to design methods to evaluate the metabolic activity of boar spermatozoa based on the incorporation of lipid precursors under standardized conditions. Mol. Reprod. Lipogenesis in the fatty liver of rat, which was induced by feeding an amino acid unbalanced diet containing 8% casein supplemented with 0.3% dl-methionine, has been studied by measuring the incorporation of glycerol-1-14C, palmitate-1-14C, citrate-1,5-14C, pyruvate-1-14C and pyruvate-2-14C into various lipid fractions and 14CO2 during in vitro incubation of liver slices.

The total radioactivity of liver lipid per 100 g of the body and the incorporation of each substrate into triglyceride in the lipid were significantly higher in the imbalance group than the control group. Conversion of each substrate to 14CO2 was not imparied in the imbalance group.

It is evident from these results that the induction of this type of fatty liver is due mainly to the triglyceride synthesis by both the fatty acid synthesis and the transesterification of fatty acid.

These results are considered to support the previous assumption in which acetate-1-14C was used as a precursor.  相似文献

We have investigated pathways of lipid metabolism in spermatozoa and generation of various metabolites with potential messenger functions during exocytosis stimulated with A23187/Ca2+. Stimulation of boar spermatozoa resulted in a considerable rapid increase in saturated/unsaturated 1,2-diacylglycerol (1,2-SU-DAG) and, concomitantly, a substantial reduction in disaturated 1,2-diacylglycerol (1,2-DS-DAG), and in phosphatidylcholine (PC). These changes preceded the onset of exocytosis. Phosphatidic acid was sometimes generated in parallel, but usually rose later, suggesting that 1,2-SU-DAG may be formed directly by phospholipase C action. Lipid changes observed in stimulated spermatozoa that have been prelabelled with several lipid precursors ([14C]palmitic acid, [14C]glycerol, [14C]choline, or [14C]arachidonic acid) suggested the existence of a unique process involving the utilization of the high basal levels of 1,2-DS-DAG to form 1,2-SU-DAG, with the latter being subsequently employed to replenish the PC pool. An ensuing generation of lysoPC and arachidonic acid, which paralleled the occurrence of exocytosis, revealed that the newly synthesized PC was hydrolyzed by phospholipase A2. The highest levels of 1,2-SU-DAG, minimum levels of 1,2-DS-DAG, and the regeneration of the PC pool were tightly coupled to the beginning of visible exocytosis. These results suggest that changes in these lipid metabolites may be fundamental processes during acrosomal exocytosis occurring in response to physiological agonists. Mol. Reprod. 1. 1. Evidence has been provided for the transfer of phosphatidyl[14C]choline and [3H]cholesterol between bovine serum albumin and cauda epididymal rat spermatozoa in Krebs-Ringer bicarbonate medium, which can promote sperm capacitation.
2. 2. An analysis of the lipid composition in both albumin and spermatozoa revealed that phospholipid levels decreased in the protein and increased by roughly comparable amounts in sperm cells during incubation in vitro.
3. 3. Cholesterol (free + ester) increased in albumin and decreased in spermatozoa. Changes in the amount of esterified cholesterol were solely responsible for the increase associated with albumin, whereas whole sperm cell extracts showed a significant decline in free cholesterol.
4. 4. The composition of albumin-bound fatty acids did not alter appreciably as a result of incubation with spermatozoa.
5. 5. Rates of [14C]palmitic acid utilization by spermatozoa suggest that lipid synthesis accounted for less than 5% of the changes observed under the conditions of this study.
6. 6. These results are interpreted as broadly supporting our previous proposal that lipid exchange between albumin and sperm cells is implicated in sperm capacitation in vitro. Specifically, the results are compatible with the idea that a decreased cholesterol/phospholipid ratio in the sperm plasma membrane facilitates this transformation.

Livers of growing rats fed a 5 or 20 protein calories percent (PC %) diet containing purified whole egg protein for three weeks were perfused in situ and the release of triglycerides (TG) and the oxidation of fatty acid by the liver alone were estimated by infusing palmitic acid-l-14C to the perfusion medium.

The release of TG from the liver of the 5 PC% group was significantly lower in both unfractionated perfusate plasma and perfusate plasma very low density lipoprotein (VLDL) than that of the 20 PC% group, whereas the content of liver TG of the 5 PC% group was higher than that of the 20 PC% group. Significantly lower radioactivity appeared in TG of both unfractionated perfusate plasma and perfusate plasma VLDL of the 5 PC% group than that of the 20 PC% group, while total radioactivity of liver TG was higher in the 5 PC% group than in the 20 PC% group. The 14CO2 production in the perfused liver of the 5 PC% group increased gradually with time rather than decreased in comparison with that of the 20 PC% group.

These findings suggest that a major factor responsible for the liver lipid accumulation in rats fed the low protein diet is not an impaired fatty acid oxidation in the liver but an impaired secretion of TG from the liver.  相似文献

Analyses of brain phospholipid fatty acid profiles reveal a selective deficiency and enrichment in eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), respectively. In order to account for this difference in brain fatty acid levels, we hypothesized that EPA is more rapidly β-oxidized upon its entry into the brain. Wild-type C57BL/6 mice were perfused with either 14C-EPA or 14C-DHA via in situ cerebral perfusion for 40 s, followed by a bicarbonate buffer to wash out the residual radiolabeled polyunsaturated fatty acid (PUFA) in the capillaries. 14C-PUFA-perfused brains were extracted for chemical analyses of neutral lipid and phospholipid fatty acids. Based on the radioactivity in aqueous, total lipid, neutral lipid and phospholipid fractions, volume of distribution (VD, μl/g) was calculated. The VD between 14C-EPA- and 14C-DHA-perfused samples was not statistically different for total lipid, neutral lipids or total phospholipids. However, the VD of 14C-EPA in the aqueous fraction was 2.5 times higher than that of 14C-DHA (p=0.025), suggesting a more extensive β-oxidation than DHA. Furthermore, radiolabeled palmitoleic acid, a fatty acid that can be synthesized de novo, was detected in brain phospholipids from 14C-EPA but not from 14C-DHA-perfused mice suggesting that β-oxidation products of EPA were recycled into endogenous fatty acid biosynthetic pathways. These findings suggest that low levels of EPA in brain phospholipids compared to DHA may be the result of its rapid β-oxidation upon uptake by the brain.  相似文献

Phosphatidylcholine content in the spermatozoa of the sea urchin, Hemicentrotus pulcherrimus, decreased rapidly during incubation with sea water. Sea urchin sperm contained approx. 85% phospholipid in total lipid. Phosphatidylcholine (PC) was the principal lipid. Other phospholipids, however, remained at constant levels during incubation. Although the free fatty acid content gradually increased following dilution of dry sperm in sea water, the amounts of triacylglycerol and cholesterol ester were extremely low. Analysis by gas-liquid chromatography indicated most of fatty acid moieties in PC to be polyenoic. PC composed in part of unsaturated fatty acids was consumed to a greater extent during incubation than that consisting of saturated fatty acids. Furthermore, 1-palmitoyl-2-[1-14C]linoleoylphosphatidylcholine was transformed to 14C-labelled free fatty acid in a subcellular system. Thus, possibly, phospholipase A2 is present in sea urchin sperm. Also, [1-14C]oleic acid was immediately oxidized to 14CO2 by sperm. It is thus concluded that sea urchin sperm use phosphatidylcholine as a substrate for energy metabolism.  相似文献

Lipid components of a glycolipid, formerly designated as spot A, from the cells of Selenomonas ruminantium were investigated. The basic structure of this material had been previously shown to be β-glucosaminyl-l,6-glucosamine. The major component of O- and N-acyl side chains was β-OH C13:0 acid when the cells were grown with added valerate. Approximately 85 % of the total amide linked fatty acids was this compound. A considerable amount of C13:2 acid was also present as a component of O-acyl fatty acids. When the cells were grown in a glucose medium containing caproate, the major fatty acid component of the spot A compound was β-OH myristic and β-OH C13:0: acids. 14C-Valerate or 14C-caproate, supplemented to the glucose medium, was incorporated into O- and N-acyl linked fatty acid moieties of the spot A compound. It was also shown that the spot A compound was the lipid A component of lipopolysaccharides of this organism.  相似文献

In the incorporation of acetate-1-14C into the total lipid per one g of the slices, no significant difference for the imbalance group was observed. However, the total radioactivity of liver lipid per 100 g of the body and the incorporation into triglyceride in the lipids were significantly higher in the imbalance group than in the control group. Conversion of acetate-1-14C to CO2 was not impaired in the imbalance group.

It is evident from these results that the induction of this type of fatty liver is due mainly to the synthesis of triglyceride.  相似文献

We investigated the effects of an OH (Fe2+/H2O2) generator system of erythrocyte membrane, particularly the time-course of lipid peroxidation as estimated by measurement of conjugated dienes, thiobarbituric reactive substances (TBARS), lipofuscin-like pigments, and α-tocopherol. Polyunsaturated fatty acids (PUFAs), especially arachidonic acid (20∶4 ω 6) and docosahexenoic acid (22∶6 ω 3), were also measured. Erythrocyte membranes were suspended in phosphate buffer containing Fe2+ (200 μM) and H2O2 (1.42 mM), and incubated in a shaking water bath at 37°C. Initially, there was an increase in TBARS and lipofuscin-like pigments, two well-known end products of PUFA oxidative degradation, whereas PUFAs remained unchanged (incubation time: 1 h). After two or more hours of incubation, marked lipid peroxidation was noted, with the appearance of conjugated dienes and a decrease of PUFAs, indicating that lipid peroxidation had occurred after a lag phase during which TBARS were not produced from PUFAs. Summary Voluntary uptake of triolein, margarine, and lipid-rich natural food (Tubifex) by fasting dragonfly larvae (Aeshna cyanea) led to heavy accumulations of lipid absorption droplets in the enterocytes within 2 days, while subsequent lipid clearance of the midgut epithelium took several weeks depending on the ingested lipid load. No endocytotic lipid uptake was observed after application of a molecular-dispersed fat dye. The smallest lipid droplets first appeared in the subapical groundplasm of the enterocytes and showed a reversible increase in size on their way towards the base. Lipid droplets were also observed at appropriate intervals after oral administration of oleic acid, after feeding margarine in the cold, and after injection of triolein into the isolated midgut.Comparative biochemical analysis after triolein feeding evidenced release of lipase and hydrolytic liberation of FA from TG in the midgut lumen, as well as time-dependent accumulations of TG in the midgut epithelium and of DG in the hemolymph.Oral injection of [14C] oleic acid was followed by its rapid absorption into the midgut epithelium, where it was utilized for the synthesis of MG and esterification to DG and TG. Energy metabolism in spermatozoa of the sea urchin Glyptocidaris crenularis was examined. The spermatozoa contained not only several kinds of phospholipids and cholesterol but also triacylglycerides (TG). Following dilution of the dry sperm in sea water, the TG content decreased rapidly. Other lipids, however, remained at constant levels, except for an increase in the level of free fatty acid. Oil red-O staining of spermatozoa showed that TG was principally located in part of the sperm midpiece. Also, high lipase activity was demonstrated in the spermatozoa. In both intact cells and a cell-free system, 14C-labeled fatty acids were oxidized to 14CO2. It is thus concluded that G. crenularis spermatozoa use TG as a substrate for energy metabolism.  相似文献

Summary

Isolated oocytes of Perinereis cultrifera have been incubated in culture media with added [3H]glycerol, [14C]butyric acid or [14C]oleic acid. The principal neutral lipid synthesized was triacylglycerol, although incorporation of radioactivity into other lipid categories (sterol, fatty acid, wax ester) was also observed. A more significant percentage of triacylglycerol was labelled after incubation with [3H]glycerol and [14C]oleic acid than with [14C]butyric acid. With this precursor, monoacylglycerol appears to be the class of lipid compartment which initially show the most radioactivity. Electron microscopic autoradiography has revealed that labelling after incorporation of glycerol was mainly localized on the lipid droplets but not on the yolk granules. A second metabolic pathway is represented by phospholipid membrane synthesis.  相似文献

Considering the vast number of scientific reports on various potential uses of fungi, there was an attempt to select the best lipid producer of some fungi at optimized conditions (Aspergillus versicolor, Rhizopus oryzae, Rhizopus arrhizus, Tramates versicolor). The aim was to offer new fields of use to the industries already culturing and using such materials. Aspergillus versicolor mycelia were found to be accumulating the highest amount of lipids. Experiments to improve lipid accumulation and transesterification properties were performed in molasses medium; the first steps were testing the effects of different pH values and different nitrogen sources on lipid accumulation. Various concentrations of KNO3 (0.5, 1.0, 1.5 gL?1) and molasses (6%, 8%, 10%) were tried in order to find the optimum carbon and nitrogen requirements. Maximum lipid content was 22.8% in the samples containing 6% molasses solution and 1.0 gL?1 KNO3 at pH 4 after 10 days of incubation. The highest fatty acid ethyl ester yield of these samples was 77% (5.0 ethanol:oil, 0.4 sulfuric acid:oil at 30°C for 6 hr). Since the crude lipids were rich in C16 and C18 fatty acids, this was considered as suitable feedstock for biodiesel production.  相似文献

Higher lipid biosynthesis and accumulation are important to achieve economic viability of biofuel production via microalgae. To enhance lipid content, Chlamydomonas reinhardtii was genetically engineered with a key enzyme diacylglycerol acyltransferase (BnDGAT2) from Brassica napus, responsible for neutral lipid biosynthesis. The transformed colonies harbouring aph7 gene, screened on hygromycin‐supplemented medium, achieved transformation frequency of ~120 ± 10 colonies/1 × 106 cells. Transgene integration and expression were confirmed by PCR, Southern blots, staining lipid droplets, proteins and spectro‐fluorometric analysis of Nile red‐stained cells. The neutral lipid is a major class (over 80% of total lipids) and most significant requirement for biodiesel production; this was remarkably higher in the transformed alga than the untransformed control. The levels of saturated fatty acids in the transformed alga decreased to about 7% while unsaturated fatty acids increased proportionately when compared to wild type cells. Polyunsaturated fatty acids, especially α‐linolenic acid, an essential omega‐3 fatty acid, were enhanced up to 12% in the transformed line. Nile red staining confirmed formation of a large number of lipid globules in the transformed alga. Evaluation of long‐term stability and vitality of the transgenic alga revealed that cryopreservation produced significantly higher quantity of lipid than those maintained continuously over 128 generations on solid medium. The overexpression of BnDGAT2 significantly altered the fatty acids profile in the transformed alga. Results of this study offer a valuable strategy of genetic manipulation for enhancing polyunsaturated fatty acids and neutral lipids for biofuel production in algae.  相似文献

This research aims to examine the effect of cadmium uptake on lipid composition and fatty acid biosynthesis, in young leaves of tomato treated seedlings (Lycopersicon esculentum cv. Ibiza F1). Results in membrane lipids investigations revealed that high cadmium concentrations affect the main lipid classes, leading to strong changes in their composition and fatty acid content. Thus, the exposure of tomato plants to cadmium caused a concentration-related decrease in the unsaturated fatty acid content, resulting in a lower degree of fatty acid unsaturation. The level of lipid peroxides was significantly enhanced at high Cd concentrations. Studies of the lipid metabolism using radioactive labelling with [1-14C]acetate as a major precursor of lipid biosynthesis, showed that levels of radioactivity incorporation in total lipids as well as in all lipid classes were lowered by Cd doses. In total lipid fatty acids, [1-14C]acetate incorporation was reduced in tri-unsaturated fatty acids (C16:3 and C18:3); While it was enhanced in the palmitic (C16:0), palmitoleic (C16:1), stearic (C18:0) and linoleic (C18:2) acids. [1-14C]acetate incorporation into C16:3 and C18:3 of galactolipids [monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG)] and some phospholipids [phosphatidylcholine (PC) and phosphatidylglycerol (PG)] was inhibited by Cd stress. Our results showed that in tomato plants, cadmium stress provoked an inhibition of polar lipid biosynthesis and reduced fatty acid desaturation process.  相似文献

Membrane-free cytosol contained over 4% of both the total lipids and phospholipids present in homogenates of lactating rat mammary gland, and much of this lipid was associated with a high molecular weight complex isolated from cytosol by gel exclusion chromatography or by density gradient centrifugation.
  • 2.2. This complex principally consisted of polypeptides with apparent molecular weights of 220 and 116kDa. Lipids associated with this complex were transferred to endoplasmic reticulum and to intracellular lipid droplet precursors of milk lipid globules upon incubation in a cell-free system.
  • 3.3. This lipoprotein complex was abundant in cytosol from lactating mammary gland, but was diminished in amount in cytosol from involuted mammary glands. The 220 kDa constituent of this complex was identified as the monomer of fatty acid synthase.
  • 4.4. These results suggest that fatty acid synthase complex in lactating mammary gland may function in transfer of lipids necessary for formation or growth of lipid droplet precursors of milk lipid globules.

Stimulation of hepatic triglyceride synthesis and secretion by clofibrate   总被引:2,自引:0,他引:2  
Isolated hepatocytes prepared from rat and squirrel monkey livers were used to explore the mechanism of action of clofibrate, a hypolipidemic agent in current use. Addition of sodium clofibrate to cells suspended in Hanks medium stimulated the conversion of [1-14C]palmitate into esterified lipids and to 14CO2. This agent also promoted the incorporation of [2-3H]glycerol into cellular lipids when fatty acids were present in the incubation medium. Triglycerides were the major lipid class increased by the drug. Sodium clofibrate enhanced the discharge of labeled lipids into the medium from liver cells prelabeled with [2-3H]glycerol. These data suggest that clofibrate does not lower plasma triglyceride levels by interference with hepatic triglyceride production or secretion.  相似文献
