In this study, the first investigation of population structure in an aquatic angiosperm, I show that populations of a marine angiosperm (eelgrass, Zostera marina) are genetically differentiated at a number of spatial scales. I find also that there is no correspondence between geographic and genetic distances separating subpopulations, an increasingly common result in spatially stratified studies of genetic structure in marine invertebrates. F-statistics, calculated for two years from electrophoretic variation at five polymorphic allozyme loci, indicate significant genetic differentiation among sampling quadrats within each of two bays (θ = 0.064-0.208), between tide zones within a bay (θ = 0.025-0.157) and between bays (θ = 0.079). Spatial autocorrelation analysis was used to explore genetic differentiation at smaller spatial scales; estimated patch sizes (within which genetic individuals are randomly associated) indicated no appeciable genetic structure at scales less than 20 m × 20 m. Calculated values of F-statistics were a function of the spatial scale from which samples were drawn: increasing the size of the “subpopulation” included in calculation of fixation indices for the same “total” sample resulted in an increase in the magnitude of f (e.g., from 0.092 to 0.181) and a decrease in θ (e.g., from 0.186 to 0.025). On the basis of the best estimate of the spatial scale of subpopulations, the effective number of migrants per generation (Nem) ranges from 1.1 to 2.8. Genetic consequences of the disturbance regime in the eelgrass habitat sampled were extreme variation between years in the allele richness and proportion of heterozygotes in a sample and a positive relationship between the extinction probability of patches and the genetic variance among them. The changes in F-statistics as a function of sampling scale and the observation that θ among sampled quadrats was positively associated with the probability of extinction among quadrats indicated that indirect estimates of gene flow (Nem) calculated from θ should be cautiously interpreted in populations that may not yet be in drift-migration equilibrium.  相似文献

In laboratory experiments discs cut from thalli of Ulva fenestrata Postels & Ruprecht grew in diameter and biomass (dry weight and ash-free dry weight) more when dead leaves of Zostera marina L. were present than when absent. A maximum increase in dry weight of 500% in 14 days occurred with Zostera present compared with only a 200% increase with no detritus. When NO?3 and PO3-4 were added the weight of an Ulva disc increased by over 800% when Zostera was present but by only 400% in controls. In general penicillin G (475 units/ml) caused a reduction in algal growth in cultures containing detritus. It is concluded that bacteria on the detritus may compete with algae for nutrients but by releasing unknown substances the bacteria promote the growth of Ulva.  相似文献

The complete assimilatory nitrate reductase (NR) gene from the pennate diatom Phaeodactylum triconutum Bohlin was sequenced from cDNA and compared with NR sequences from fungi, green algae, vascular plants, and the recently sequenced genome of the centric diatom Thalassiosira pseudonana Hasle and Heimdal CCMP1335. In all the major eukaryotic nitrate reductase (Euk‐NR) functional domains, diatom NR gene sequences are generally 50%–60% identical to plant and alga sequences at the amino acid level. In the less conserved N‐terminal, hinge 1, and hinge 2 regions, homology to other NR sequences is weak, generally<30%. Two PCR primer sets capable of amplifying Euk‐NR from plants, algae, and diatoms were designed. One primer set was used to amplify a 750‐base pair (bp) NR fragment from the cDNA of five additional diatom strains. The PCR amplicon spans part of the well‐conserved dimer interface region, the more variable hinge 1 region, and part of the conserved cytochrome b heme binding region. The second primer set, targeted to the dimer region, was used to amplify an approximately 400‐bp fragment of the NR gene from DNA samples collected in Monterey Bay, California and in central New Jersey inner continental shelf (LEO‐15 site) waters. Only diatom‐like NR sequences were recovered from Monterey Bay samples, whereas LEO‐15 samples yielded NR sequences from a range of photosynthetic eukaryotes. The prospect of using DNA‐ and RNA‐based methods to target the NR genes of diatoms specifically is a promising approach for future physiological and ecological experiments.  相似文献

A new polyclonal antibody was raised against centrin isolated from the flagellate green alga Spermatozopsis similis (Chlorophyta; anti-SSC). It stains by immunofluorescence and immunoelectron microscopy well-known reference systems for centrin like the nucleus–basal body connectors in Chlamydomonas reinhardtii (Chlorophyta) and the system II fibers (rhizoplasts) of Scherffelia dubia (Chlorophyta). In addition, it recognizes in immunoblots a single 20-kDa protein in isolated cytoskeletons of Spermatozopsis similis and Tetraselmis striata (Chlorophyta) as well as purified centrin isolated from Tetraselmis striata. Using this antibody, centrin was localized in whole cells and isolated cytoskeletons of Oxyrrhis marina Dujardin (Dinophyceae) by immunofluorescence and immunogold electron microscopy. In the flagellar apparatus of O. marina, five different structures were antigenic. Four short fibers (connectives 1–4) link the basal bodies to the four major fibrous flagellar roots, which do not cross-react with anti-centrin. The most prominent of the labeled structures (connective 5), a crescent-shaped fiber, extends from the flagellar canal of the transverse flagellum along the base of the tentacle to the flagellar canal of the longitudinal flagellum, interconnecting the distal parts of the microtubular roots/bands in the basal apparatus. For most of its length, it underlies and is connected to a transversely oriented subamphiesmal microtubular band. In immunoblot analyses, anti-SSC recognizes only a single 20-kDa protein in cytoskeletons of O. marina. Functional and phylogenetic aspects of centrin-containing structures in dinoflagellates are discussed.  相似文献

Interactions between algal epiphytes and their grazers can have a significant impact on the structure and function of eelgrass (Zostera marina L.) meadows. In Puget Sound, the herbivorous gastropod Lacuna variegata Carpenter and its congeners appear to remove large quantities of the epiphytic community from eelgrass blades. When snails at typical field densities were used in microcosms, Lacuna significantly reduced epiphytic biomass and areal productivity. Biomass-specific productivity of the epiphytic community showed an increasing trend with increasing snail density. Epiphytic productivity increased nonlinearly with increasing epiphytic biomass. The commonly used logistic population growth formula adequately described this relationship. Grazing rate also increased nonlinearly with increasing epiphytic biomass. The Hailing equation adequately described the relationship between grazing rate and epiphytic biomass. The proportion of the epiphytic biomass found on the oldest blade of an eelgrass shoot was related linearly to epiphytic biomass, suggesting that a constant fraction of the epiphytic community is lost regardless of epiphytic density. Lacuna clearly removed large quantities of epiphytic material from eelgrass blades, significantly altering community function. Modified Lotka-Volterra equations, incorporating the logistic growth form and Hailing grazing equation, should prove useful in modeling the epiphyte–grazer interaction.  相似文献

Enclosures in a small piedmont reservoir in the southeastern United States were used to determine the effects of loading by phosphate and two clay sediments on periphyton community structure and production during the summer growing season. The experimental design included replicated controls and the following treatments added at 2-to 3-day intervals: phosphate (PHOS), kaolinite (KAOL), montmorillonite (MONT), kaolinite with phosphate (K+P), and montmorillonite with phosphate (M+P). Periphyton were sampled from polyethylene strips of the same material as the enclosure walls, suspended at depths of 0.5 m and 2.0 m to assess treatment effects on shallow (epilimnetic) vs. deep (hypolimnetic) communities. Colonization after 40 days was sparse, dominated in biovolume by blue-green algae in all but the epilimnion of controls, in which small gelatinous green algae and chrysophyte flagellates were abundant. The biovolume of shallow periphyton after 79 days was lowest in KAOL and MONT, intermediate in controls and PHOS, and greatest in clay + P. Hence, P enrichment alleviated the deleterious effects of clay on periphyton production, and clay + P appeared to act synergistically in stimulating benthic algal growth. Periphyton biovolume was comparable among shallow and deep controls, PHOS, and treatments with the rapidly settling kaolinite. Lowest biovolume occurred at depth in treatments with montmorillonite under extreme light attenuation imposed by the suspended, finely particulate clay. The relative contribution of blue-green algae to total periphyton production was highest in clay + P treatments and lowest in controls. Heterotrophic dinoflagellates increased in the shallow periphyton under clay loading, and in the deep communities under PHOS and M + P. Periphyton contributed only 20–32% of total mesocosm productivity in controls, PHOS, and KAOL, increasing to about 40% in MONT, and to 72–91% of the total in M + P and K + P. The data indicate that benthic microalgae can represent a major proportion of the primary production in shallow reservoirs under high phosphate and sediment loading.  相似文献

Although activity of the enzyme nitrate reductase (NR) can potentially be used to predict the rate of nitrate incorporation in field assemblages of marine phytoplankton, application of this index has met with little success because the relationship between the two rates is not well established under steady-state conditions. To provide a basis for using NR activity measurements, the relationships among NR activity, growth rate, cell composition, and nitrate incorporation rate were examined in cultures of Thalassiosira pseudonana (Hustedt)Hasle and Heimdal, growing a) under steady-state light limitation, b) during transitions between low and high irradiance (15 or 90 μmol quanta.m?2.s?1), and c) under steady-state nitrate limitation. Using a modified assay for NR involving additions of bovine serum albumin to stabilize enzyme activity, NR activity in light-limited cultures was positively and quantitatively related to calculated rates of nitrate incorporation, even in cultures that were apparently starved of selenium. During transitions in irradiance, growth rates acclimated to new conditions within 1 day; through the transition, the relationship between NR activity and nitrate incorporation rate remained quantitative. In nitrate-limited chemostat cultures, NR activity was positively correlated with growth rate and with nitrate incorporation rates, but the relationship was not quantitative. NR activity exceeded nitrate incorporation rates at lower growth rates (<25% of nutrient-replete growth rates), but chemostats operating at such low dilution rates may not represent ecologically relevant conditions for marine diatoms. The strong relationship between NR activity and nitrate incorporation provides support for the idea that NR is rate-limiting for nitrate incorporation or is closely coupled to the rate-limiting step. In an effort to determine a suitable variable for scaling NR activity, relationships between different cell components and growth rate were examined. These relationships differed depending on the limiting factor. For example, under light limitation, cell volume and cell carbon content increased significantly with increased growth rate, while under nitrate limitation cell volume and carbon content decreased as growth rates increased. Despite the differences found between cell composition and growth rate under light and nitrate limitation, the relationships between NR activity scaled to different compositional variables and growth rate did not differ between the limitations. In field situations where cell numbers are not easily determined, scaling NR activity to particulate nitrogen content may be the best alternative. These results establish a strong basis for pursuing NR activity measurements as indices of nitrate incorporation in the field.  相似文献

Anabaena flos-aquae (Lyngb.) Bréb. was grown in varying concentrations of nitrate. Specific growth rates, as estimated in batch culture, were constant and approached the maximum rate at all concentrations of NO3?-N tested bewteen 0 and 400 μ/L. Steady-state biomass, as determined in semicontinuous culture, did not vary with NO3? at slower dilution rates. However at a faster dilution rate, significantly less biomass occurred in intermediate concentrations of NO3? than in either higher or lower concentrations. The results indicate that both growth rate and standing crop are maximized by either N2 fixation or NO3? assimilation, but extracellular NO3? reduces the rate of N2 fixation. Consequently, at very low NO3? concentrations, growth is virtually maximized by N2 fixation alone, and at high concentrations of NO3?, N2 fixation is inhibited but growth is maximized by assimilation of NO3?. At intermediate concentrations of NO3?, growth becomes a function of NO3? assimilation augmented by N2 fixation. In this case, full growth potential is realized only if hydraulic residence time is sufficiently long to compensate for the reduced rate of N2 fixation. Growth rate and standing crop are not diminished in response to the large amount of energy allocated to N2 fixation. Instead, other cellular processes are probably affected negatively during N2 fixation.  相似文献

A literature review of data on nitrate uptake by phytoplankton suggests that nitrate levels above 20 μmol N·L?1 generally stimulated uptake rates in cultured unicellular algae and natural phytoplankton communities. This phenomenon indicates that phytoplankton cells acclimate to elevated nitrate levels by increasing their uptake capacity in a range of concentrations previously considered to be saturating. Cyanobacteria and flagellates were found to present a considerable capacity for acclimation, with low (0.1–2 μmol N·L?1) half‐saturation values (Ks) at low (5–20 μmol N·L?1) substrate levels and high (1–80 μmol N·L?1) Ks values at high (30–100 μmol N·L?1) substrate levels. However, some diatom genera (Rhizosolenia, Skeletonema, Thalassiosira) also appeared to possess a low affinity nitrate uptake system (Ks between 18 and 120 μmol N·L?1), which can help resolve the paradox of their presence in enriched seas. It follows that present models of nitrate uptake can severely underestimate the effects of high nitrate concentrations on phytoplankton dynamics and development. A more adequate approach would be to consider the possibility of multiphasic uptake involving several phase transitions as nitrate concentrations increased. Because it is a nonlinear phenomenon featuring strong thresholds, this effect appears to override that of other variables, such as irradiance, temperature, and cell size. Within the present context of eutrophication and for a range of concentrations that is becoming more and more ecologically relevant, equations are tentatively presented as a first approach to estimate Ks from ambient nitrate concentrations.  相似文献

The three-dimensional structure of the flagellar apparatus in the dinoflagellate Oxyrrhis marina has been reinvestigated and found to consist of several previously unknown components and component combinations that appear strikingly similar to those of some gymnodinoid taxa. The flagellar apparatus of this dinoflagellate is asymmetric and extremely complex consisting of a longitudinal and a transverse basal body that gives rise to eight structurally different components. The only posteriorly directed component is the large microtubular root that consists of 45–50 microtubules at its origin and is attached proximally to a perpendicularly oriented striated fibrous component. Arising from each basal body, two striated fibrous roots with different periodicities extend to the cell's left. A single stranded microtubular root with associated electron dense material emanates from the transverse basal body and also extends to the cell's left. A striated fibrous connective arises from the longitudinal basal body and extends toward the cell's right ventral surface and terminates near the sub-thecal microtubular system. A compound root consisting of microtubules and electron dense material also originates from the longitudinal basal body and extends ventrally into the anterior region of the tentacle. Structural similarities between the parallel striated fibrous roots of Oxyrrhis and Polykrikos are discussed as are flagellar apparatus similarities among other gymnodinoid dinoflagellates. A diagrammatic reconstruction of the Oxyrrhis flagellar apparatus is also presented.  相似文献

The effects of nitrate, phosphate, and iron starvation and resupply on photosynthetic pigments, selected photosynthetic proteins, and photosystem II (PSII) photochemistry were examined in the diatom Phaeodactylum tricornutum Bohlin (CCMP 1327). Although cell chlorophyll a (chl a) content decreased in nutrient-starved cells, the ratios of light-harvesting accessory pigments (chl c and fucoxanthin) to chl a were unaffected by nutrient starvation. The chl a-specific light absorpition coefficient (a*) and the functional absorption cross-section of PSII (σ) increased during nutrient starvation, consistent with reduction of intracellular self-shading (i.e. a reduction of the “package effect”) as cells became chlorotic. The light-harvesting complex proteins remained a constant proportion of total cell protein during nutrient starvation, indicating that chlorosis mirrored a general reduction in cell protein content. The ratio of the xanthophylls cycle pigments diatoxanthin and diadinoxanthin to chl a increased during nutrient starvation. These pigments are thought to play a photo-protective role by increasing dissipation of excitation energy in the pigment bed upstream from the reaction centers. Despite the increase in diatoxanthin and diadinoxanthin, the efficiency of PSII photochemistry, as measured by the ration of variable to maximum fluorescence (Fv/Fm) of dark-adapted cells, declined markedly under nitrate and iron starvation and moderately under phosphate starvation. Parallel to changes in Fv/Fm were decreases in abundance of the reaction center protein D1 consistent with damage of PSII reaction centers in nutrient-starved cells. The relative abundance of the carboxylating enzyme, ribulose bisphosphate carboxylase/oxygenase (RUBISCO), decreased in response to nitrate and iron starvation but not phosphate starvation. Most marked was the decline in the abundance of the small subunit of RUBISCO in nitrate-starved cells. The changes in pigment content and fluorescence characteristics were typically reversed within 24 h of resupply of the limiting nutrient.  相似文献

Eutrophication of coastal waters often leads to excessive growth of microalgal epiphytes attached to seagrass leaves; however, the effect of increased nutrient levels on sediment microalgae has not been studied within seagrass communities. A slow‐release NPK Osmocote fertilizer was added to sediments within and outside beds of the shoal grass Halodule wrightii, in Big Lagoon, Perdido Key, Florida. Gross primary production (GPP) and biomass (HPLC photopigments) of sediment microalgae within and adjacent to fertilized and control H. wrightii beds were measured following two 4‐week enrichment periods during June and July 2004. There was no effect of position on sediment microalgal GPP or biomass in control and enriched plots. However, nutrient enrichment significantly increased GPP in both June and July. These results suggest that sediment microalgae could fill some of the void in primary production where seagrass beds disappear due to excessive nutrient enrichment. Sedimentary chl a (proxy of total microalgal biomass) significantly increased only during the June enrichment period, whereas fucoxanthin (proxy of total diatom biomass) was not increased by nutrient enrichment even though its concentration doubled in the enriched plots in June.  相似文献

The relationship between the diadinoxanthin cycle and changes in fluorescence yield in the diatom Chaetoceros muelleri Lemm. (clone CH10, Amorient Aquafarm, Inc., Hawaii) was investigated. High-light-induced changes in fluorescence yield and xanthophyll de-epoxidation occurred very rapidly (first order rate constant 1.60 min?1). The observed light-induced changes in diatoxanthin and diadinoxanthin concentration were consistent with a two-pool scheme for diadinoxanthin, one of which does not undergo de-epoxidation. Changes in xanthophyll concentration correlated with changes in in vivo absorbance indicating that diadinoxanthin cycle activity in vivo can be monitored spectrophotometrically. However, changes in cell absorbance were small relative to total optical absorption cross section. Increases in the concentration of diatoxanthin were linearly correlated with increases in the rate constant for thermal de-excitation in the antenna of photosystem II (PSII). Antenna quenching produced or mediated by diatoxanthin may, thus, protect the PSII reaction center in diatoms. Changes in the maximum fluorescence yield suggested that changes in the reaction center also contributed to nonphotochemical quenching of fluorescence. Thus, reaction center quenching affected the relationship between antenna quenching and changes in photochemical efficiency producing the effect of a decrease in fluorescence yield without a decrease in photochemical efficiency.  相似文献

Nitrate concentrations within individual cells of Ethmodiscus, Pyrocystis, and Halosphaera and chains of Rhizosolenia were determined from samples collected in the Sargasso Sea. In all cases, field populations exhibited a wide range of internal nitrate concentrations (INCs) within a single sampling date. Halosphaera INCs reached 100 mM, in contrast to diatom and dinoflagellate INCs, which did not exceed 22 mM. Sinking Rhizosolenia, Ethmodiscus and Pyrocystis had significantly lower internal NO3- pools than did floating cells (P< 0.05). Ethmodiscus incubations in surface seawater resulted in a dramatic reduction in the proportion of high INC cells concurrent with decreases in average INCs and an increased proportion of sinking cells. Population buoyancy was inversely related to INC, and negatively buoyant cells rarely exceeded I mM INC, suggesting that a critical INC threshold may exist. The photosynthetic parameters Pmax and α decreased with time as internal NO3-- Pools were depleted. Internal nitrate depletion rates were consistent with oxygen production rates during this time. Based on the known characteristics of Pyrocystis and Ethmodiscus, we conclude that virtually all of the > 100 μm-sized phytoplankton present in the Sargasso Sea can vertically migrate. However, the appropriate time scale for migrators such as Halosphaera that reproduce by swarmer formation is unclear and may be significantly different than the other taxa studied. Changes in the frequency distributions, buoyancy-internal pool relationships, and general P-1 photosynthesis-irradiance time series data in Ethmodiscus suggest that nutrient limitation is related to these migrations. High INC appears to be a fundamental property of the largest microalgal cells present in oligotrophic seas and suggests that nitrate transport by these nonmotile cells is widespread.  相似文献

The cortical microtubular cytoskeleton of Oxyrrhis marina Dujardin was investigated using indirect immunofluorescence and transmission electron microscopy. The cortical microtubular cytoskeleton is unlike that of other previously examined dinoflagellates because all cortical microtubules are oriented longitudinally and do not attach or abut tranverse microtubular arrays. This difference is considered along with other morphological and cytological variables as indicative of Oxyrrhis's phylogenetic position relative to the Dinophyceae.  相似文献

Three nitrate reductase mutants were independently isolated and characterized in the colonial alga, Eudorina elegans Ehrenberg. nar-1 is a leaky mutant, deficient in the production of nitrate reductase. nar-2 and nar-3 both lack the ability to produce nitrate reductase. However, nar-2 grows and nar-3 does not grow when hypoxanthine is the sole nitrogen source. The specific activity of the next enzyme, in the pathway, nitrite reductase is increased in nar-3 when compared to wild-type, nar-1 and nar-2.  相似文献

Catalase activity increased in Peridinium gatunense (formerly P. cinctum fa. westii) cells during the decline of the seasonal spring bloom period in Lake Kinneret. This was correlated with the low ambient total CO2 concentration. The relationship was confirmed in laboratory experiments where maximum catalase activity occurred under an atmosphere composed of 30% O2 and 0.003% CO2. Conversely, high CO2 concentrations inhibited catalase activity. The rise in catalase activity was not directly due to increasing environmental pH, as in vitro and in vivo measurements showed a characteristic broad pH curve with a constant activity from pH 6–10 for catalase. Photoinhibition of catalase occurred above 250 μmol photons · m?2· s?1. However, at high photoinactivating irradiances, photoinhibition was ameliorated under high pO2/pCO2. Such conditions prevail in the Kinneret at the end of the spring. We propose that the enhancement of photorespiration (under high pO2/pCO2) induces a temporary burst in catalase activity despite the progressively photoinhibitory conditions of early summer.  相似文献

The nitrate uptake capacity of mature blade tissue of the giant kelp, Macrocystis pyrifera (L.) C. Ag., was examined as a function of the availability of light and nitrate. Time course measurements indicated that nitrate uptake rate, as measured by the incorporation of 15N, was significantly increased by N starvation. The response was linear over the first hour of exposure regardless of the N status of the tissue indicating that surge uptake was not responsible for the increase. The Michaelis-Menten parameters Vmax and Ks, however, were not significantly changed by either growth nitrate concentration or growth irradiance as a result of high variability among blades. Similarly, the initial slope (α) of the nitrate uptake kinetics curves was unaffected. Concentration of photosynthetic pigments increased in response to increased nitrate availability but not to increased growth irradiance. Time course and pigment data demonstrated that mature blade tissue responds to increased N availability by decreasing its capacity to take up nitrate and by increasing its investment in photosynthetic pigments, perhaps for N storage or enhanced light-harvesting capabilities and the increase in reducing power available for N assimilation. This study provides evidence for a dynamic regulatory system that responds to changes in nitrate availability in an integrated manner.  相似文献

A polyclonal antibody, raised against nitrate reductase (NR) purified from the photosynthetic dinoflagellate Gonyaulax polyedra Stein, was used as a probe in immunogold-labeling experiments on thin sections prepared from cells harvested both during day and night phases. Previous experiments have shown that both NR activity and the amount of immunoreactive NR in cell extracts is greater when day-phase cells are examined, and this property was exploited as an internal control for the cytochemical labeling. We observed that in day-phase cells, chloroplasts contained approximately three times more gold particles than night-phase cells (highly significant difference; P < 0.0001), whereas cytoplasmic labeling levels remained relatively level between day and night. We conclude from the diurnal difference in labeling that our antibody faithfully reflects the distribution of NR in Gonyaulax cells. Thus, as in to some other higher plants and green algae, Gonyaulax compartmentalizes active NR in its chloroplasts.  相似文献
