利用光镜、扫描电镜和透射电镜对水龙骨科(Polypodiaceae)瓦韦(Lepisorus thunbergianus (Kaulf.) Ching)孢子壁的结构和发育进行了研究。研究结果表明瓦韦孢子两侧对称、单裂缝,表面具波纹状纹饰。孢壁从内到外由内壁、外壁和周壁三部分构成。外壁来源于绒毡层物质,由外壁内层和外壁外层构成,外壁外层表面的波纹状纹饰形成孢子表面的纹饰轮廓。周壁薄,紧贴外壁表面,由2层片状结构叠合而成。在外壁外层形成过程中,孢子表面和周围出现较多小球。本文探讨了孢壁各层的结构、来源和发育过程,为蕨类植物系统学和孢粉学研究积累资料。  相似文献

The structure of the pycnidiospores of B. theobromae was investigatedby a combination of light microscope, a scanning electron microscopeand a transmission electron microscope observations. As observedwith the light microscope the hyaline non-septate pycnidiosporeis highly vesiculated while the pigmented septate one exhibitslongitudinal hyaline striations. Investigations with the scanningelectron microscope showed the hyaline non-septate pycnidiosporeto be smooth-walled without any form of ornamentation. The pigmentedseptate pycnidiospore on the other hand had a rough outer surfacewith a transverse groove, which probably indicated the positionof the septum. The longitudinal striations seen on such pycnidiosporeswith the light microscope were not observed under these conditions.Observations with the transmission electron microscope showedthat the outermost layer of the hyaline pycnidiospore was smootherthan that of the pigmented one. The pigment was on the outerlayer of the double-layered wall of the pigmented pycnidiospore.The inner non-pigmented wall layer was continuous with the septum.The septum tapered gradually to a thin layer in the centre whereit was perforated by a simple pore. The observations indicatedthat the pigmented spore was multinucleate while the hyalineone was uninucleate.  相似文献

A new species of Haplosporidium Caullery & Mesnil, 1899 parasitising the pulmonate gastropod Siphonaria lessonii Blainville in Patagonia, Argentina, is described based on morphological (scanning and transmission electron microscopy) and sequence (small subunit ribosomal RNA gene) data. Different stages of sporulation were observed as infections disseminated in the digestive gland. Haplosporidium patagon n. sp. is characterised by oval or slightly subquadrate spores with an operculum that is ornamented with numerous short digitiform projections of regular height, perpendicular to and covering its outer surface. The operculum diameter is slightly larger than the apical diameter of the spore. Neither the immature nor mature spores showed any kind of projections of the exosporoplasm or of the spore wall. Regarding phylogenetic affinities, the new species was recovered as sister to an undescribed species of Haplosporidium Caullery & Mesnil, 1899 from the polychaete family Syllidae Grube from Japanese waters. The morphological characters (ornamentation of the operculum, spore wall structure, shape and size of spores, and the lack of spore wall projections) corroborate it as an as yet undescribed species of Haplosporidium and the first for the phylum in marine gastropods of South America. Siphonaria lessonii is the only known host to date.  相似文献

Aspects of the fine structure of the microconidium of the mycelial phase of the dimorphic fungal pathogenBlastomyces dermatitidis as seen by techniques of scanning and transmission electron microscopy are described and illustrated by electron micrographs. The conidia ofB. dermatitidis undergo changes in the ultrastructural appearance of the cell wall as the spore matures. The cell wall becomes irregular in its thickness and possesses two distinct layers. No discrete or unique surface spines or projections were evident when the conidium ofB. dermatitidis was viewed by scanning electron microscopy. Upon maturity there is a marked deposition of lipid material in large, multiple storage bodies which occupy much of the cytoplasmic area. However, the cytoplasmic organelles appear to retain their structural integrity.  相似文献

本文对山茶属(Camellia L.)18个组27个代表种的花粉进行了光学显微镜、扫描电镜和大部分进行了透射电镜的观察,并对花粉大小、形状、外壁纹饰和超微结构等作了综合的比较。结果表明,其形状和大小差异不很明显。花粉壁的层次、厚度和不同层次的比例,如复盖层与柱状层以及外壁外层与外壁内层的比例等也没有显著的差异。但花粉壁的纹饰特征在扫描电镜下显示出较大的差异,具有较重要的分类学价值。根据其纹饰特征,可以把山茶属花粉大致区分为六个类型:皱疣状纹饰;粗瘤状纹饰;颗粒状至皱-颗粒状纹饰;穴-网状纹饰;皱沟状纹饰及皱波-念珠状纹饰。孢粉学上区分的类型不完全与各个学者从植物形态上划分的组相一致。其中管蕊茶 Camellia lanceolata的纹饰非常特殊(粗瘤状),其外壁表面很象牛心果(Annona reticulata)的外形特征,为其重新分类提供孢粉学依据。  相似文献

A population of aseptate pycnidiospores of the fungus Botryodiplodia theobromae can be induced to germinate or to form septa delimiting two cells; this developmental process is dependent upon nutritional and environmental factors. Transmission electron microscope investigations indicate that during germination of the aseptate spore, a new inner wall layer is synthesized de novo at the site of germ tube emergence. Formation of the septum also involves the de novo synthesis of an inner wall layer which comprises the majority of the septum and completely surrounds the spore. The wall of the germ tube emerging from the septate spore is a direct extension of this inner layer deposited during the formation of the septum. Although the early stages of spore germination may involve localized enzymatic degradation of the internal layers of the spore wall, transmission and scanning electron micrographs of germinating spores show that the outer wall layers are physically fractured by the emerging germ tube. It is suggested that spore germination and septum formation are initially similar processes regarding cell wall genesis but that some mechanism responsive to environmental and nutritional conditions determines the course of development.  相似文献

Abstract

Preliminary observations on spore ornementation of Russula, as seen in the scanning electron microscope. — The spores of 16 species of Russula have been examined in the scanning electron microscope, as a preliminary attempt to see if an accurate examination of the spore surface at the ultrastructural level could reveal details of ornamentation which might be useful for the classification of the many species of this genus. The examination, carried out both on fixed and unfixed specimens, has demonstrated that the spore shape is always round or slightly elliptic, and that the obnormal forms as previously described are probably artifacts. Five main types of ornamentation have been described: single wart-like and single finger-like Protrusion, wart- or finger-like protrusions interconnected by thin ridges, and thick, short « papillae » together with large ridges that run along large tracts of the spore circumference. The type of ornamentation was a constant character in each species.  相似文献

Spore ornamentation of Haplosporidium nelsoni and Haplosporidium costale was determined by scanning electron microscopy. For H. nelsoni, the spore surface was covered with individual ribbons that were tightly bound together and occurred as a single sheet. In some spores, this layer was overlaid with a network of branching fibers, about 0.05 microm in diameter, which often was dislodged from the spore at the aboral pole. For H. costale, ornamentation consisted of a sparse network of branching fibers on the spore surface. Molecular phylogenetic analysis of the phylum Haplosporidia revealed that Urosporidium, Bonamia, and Minchinia were monophyletic but that Haplosporidium was paraphyletic. All species of Minchinia have ornamentation composed of epispore cytoplasm, supporting the monophyly of this genus. The presence of spores with a hinged operculum and spore wall-derived ornamentation in Bonamia perspora confounds the distinction between Bonamia and Haplosporidium. Species with ornamentation composed of outer spore wall material and attached to the spore wall do not form a monophyletic group in the molecular phylogenetic analysis. These results suggest that the widely accepted practice of assigning all species with spore wall-derived ornamentation to Haplospordium cannot be supported and that additional genera are needed in which to place some species presently assigned to Haplosporidium.  相似文献

The family Hymenophyllaceae is represented in the study area by six species in two genera, Hymenophyllum J. E. Smith and Trichomanes L. The study was based on herbarium material and spores were studied under light microscope (LM), scanning electron microscope (SEM) and transmission electron microscope (TEM). Both genera have trilete spores, 23 to 45 μm in equatorial diameter, with an ornamentation of echinulae and cones in Hymenophyllum and of verrucae, gemmae and granules in Trichomanes. Mature spores have a sporoderm composed of a perispore, an exospore and a fibrillar endospore; the exospore is 0.5 to 2.5 μm thick, compact and with an irregular margin. In some cases radial channels and other channels associated with the middle and inner parts of the laesurae were evident. A series of cavities filled with an opaque content line the inner margin of the exospore. The perispore is 20 to 400 nm thick and unevenly differentiated along the surface of a same spore. Under TEM, two main differentially contrasted portions could be distinguished: a dark massive portion with structural components could not be distinguished, and a light portion with several plates arranged in piles. The inner surface of the perispore exhibit short scales. Globules are immersed within the perispore at some depth from the perispore surface and others connected to it by structural threads. The spore characters observed including shape, ornamentation, laesurae length and wall structure are useful in distinguishing the two genera studied, but less useful in differentiation at the species level.  相似文献

As a contribution to the taxonomic study of Asplenium L. for the new "Flora de la República de Cuba", the spore morphology was studied into three complexes of hardly delimited species. Twenty-two collections have been studied by SEM and eighty-nine by LM. Shape, number of spores per sporangium and perispore ornamentation, specially fine details revealed by scanning electron microscopy, were the most valuable characters in the recognition of taxa. Irregularities in spore shape and size, unusual development of perispore wall surface elements, and collapsed protoplast characterize aborted spores and suggest the occurrence of a possible hybrid species.  相似文献

The type strain of Streptomyces torulosus Lyons and Pridham (1971) was studied by scanning- and transmission electron microscope. Spore chains were formed in spirals by aerial mycelium. The spores were connected by nozzles in which small channels could be observed. The knobby ornamentations of the spores arised on a thin fibrous sheath, enveloping the spore chains. These irregular blunt projections, called knobs, had varying diameters of 100 to 250 nm. The base of the knob, consisting of globose to flattened electron dense material, was sitting directly on the sheath. It was covered by several small vesicles of the same material. Each hollow vesicle beared a thin bowlshaped shell of electron transparent material. In general, the cupular bowls and their supporting vesicles became easily depressed on their base, but not detached from the surface of the spores. This type of knobby spore ornamentation was suggested to be designated as a verrucate spore type.  相似文献

采用扫描电镜方法对井冈山地区18种唇形科植物的果实形状与表面微形态特征进行观察比较。结果表明:18种唇形科植物果实形状多样,可分为矩圆形、圆形、卵形、三棱形、球形和椭圆形6类;外果皮表面细胞形状为4~6边形、不规则形和细胞形状不可见;依据外果皮表面凸起度可分为平滑型、凹陷型和凸起型3类;外果皮表面蜡质纹饰复杂多样,有平滑无纹饰、负网状纹饰、网状纹饰兼具疣状或瘤状凸起和网状纹饰兼具次级条纹4类;外果皮表面附属结构主要包括表皮毛和丝状结构,并具有或多或少的鳞片状或颗粒状分泌物。18种唇形科植物果实形状和外果皮表面微形态特征均表现出丰富的多样性,可为唇形科属间和种间亲缘关系研究提供重要的分类学依据。  相似文献

The ultrastructure of the wall layers and ornamentative features of Scenedesmus pannonicus and S. longus are described using carefully correlated freeze-etched replicas, thin sections and scanning electron micrographs. The two species arc enclosed by different types of ornamented layers, S. pannonicus by the tightly filling, “warty” layer and S. longus by the loosely fitting, “reticulate” layer, held off the coenobium by 2 types of tubular propping spikelets and rosettes. The reticulate layer has an intricate substructure, especially when studied with freeze-etching. Its inner and outer surfaces appear different, as is its attachment to the 2 types of spikelets. Whole cells of S. longus subjected to acetolysis lack the cellulose wall and cytoplasm, but all other surface features survive, including the Trilaminar Sheath (TLS); this ornamentation cannot be “pectic.” The cellulose wall and ornamentation is unaffected by boiling water alone. Boiling in 6n NaOH removes the surface ornamentation, but the TLS and wall remain; the possibility that these features contain silica is discussed. The terminal spines of both species consist of closely packed spikelets enclosed within a skin of hexagonally-packed subunits. Similar subunits are seen in the propping spikelets of S. longus, and in the rows or “combs” of laterally fused spikelets of S. pannonicus. The warty layer of S. pannonicus is tightly appressed to the TLS except close to where the cells are joined, where it is suspended free. It is composed of a layer of globular subunits, and small indentations form the warts. Single, evenly distributed warts characterize the freely suspended sections of the warty layer, and the layer that encloses young coenobia soon after they have been formed: in contrast, the warts are clumped over the surface of older and larger colonies. Some of the single warts form characteristic double rows, but these latter remain single even on older cells. The surface structure of the warty layer, TLS, and plasmalemma are revealed by the freeze-etching process.  相似文献
