Pinus pinea L. (Mediterranean stone pine) is an important forest species not only for its economically relevant kernel production, but also for environmental protection. The detection of genetic variability is an essential issue for Mediterranean forest species for conservation and improvement programs. Based on data collected for several years at three field sites from a P. pinea L. provenance trial established in Portugal in 1993, the present study aimed to evaluate the genetic variability of the adaptive traits of P. pinea L. and to identify a group of provenances with high performance for growth traits to be used in future plantings. Several mixed models were fitted to survival and growth trait data to estimate provenance and provenance × site interaction variability. The empirical best linear unbiased predictors of provenance genotypic effects were used to select a superior group of provenances. The provenance genetic variability of P. pinea L. was successfully detected for survival and height of different planting ages and also for diameter at breast height at age 13 years after planting. For growth traits, the most successful methods used for evaluating provenance genetic variability were based on the linear mixed spatial models. When multi-environmental analysis was performed, provenance × site interaction variability was detected for survival, but not for height at age 6 years. The existence of provenance variability in P. pinea L. permitted the identification of a seed lot composed of a mixture of the best provenances for height and diameter.  相似文献

Antibody against the light-dependent NADPH-protochlorophyllide oxidoreductase of oat was used to detect a protein of the same molecular weight in cotyledons of 40-day-old dark-grown seedlings of Pinus pinea L. Exposure of the seedlings to light resulted in a rapid decrease in protochlorophyllide content without the concomitant decrease in 38 kDa protein which is observed on transfer of dark-grown angiosperm seedlings to light. Pinus pinea L. is widely disseminated all over the Mediterranean Basin. Qualitatively, P. pinea fatty acid seed composition is identical and typical of the genus Pinus. This composition is made of unsaturated oil with several unusual polymethylene-interrupted unsaturated fatty acids. Linoleic acid is the major fatty acid followed by oleic, palmitic and stearic acids. Quantitatively, for all Mediterranean populations, total amounts of fatty acids seem to be fairly constant and independent from their origin. When applying principal component analysis, it seems that there is not a distinct geographical variability. Tunisian populations appear to be integral part of the Mediterranean populations without any particular structuring. Taking into account this research and the data reported in the literature, we can confirm that P. pinea expresses no significant variability. This low genetic diversity revealed by fatty acid composition can be explained by anthropogenetic diffusion of genetically homogeneous reproductive material as early as the first explorations.  相似文献

The PAS reaction was used to stain total insoluble polysaccharides in pollen of two species of Pinus collected in two different years and germinated in vitro at different pH values. PAS-detectable polysaccharides are localized in the intine, cytoplasm and amyloplasts; PAS-undetectable (callose) in the middle of the intine, but only in the distal part of the grain. The total PAS stained polysaccharide content was quantified with a microdensitometer. Values are correlated with germination percentages and pollen tube length. The total polysaccharide content, as well as pollen tube length, varies in each species according to the year, probably due to environmental conditions. Also the various pHs deeply influence pollen germination and tube length, the lowest pH being the most unfavourable condition. At this pH (3.5) the total polysaccharide content is higher than in the control (pH 5.5): this is probably due to a lack of callose synthesis for tube wall or to a depolymerization of this polysaccharide, and a subsequent storage in a PAS-detectable form.  相似文献

Proteins and lipids are the two main storage substances in theendosperm of Pinus pinea L. In imbibed seeds, lipid and proteinbodies almost completely fill the cell volume and only a fewpoorly structured organelles can be detected. During germinationthe storage substances are reabsorbed whilst plastids, roughendoplasmic reticulum, ribosomes and polysomes, microbodies(glyoxysomes, most likely) become evident. Starch is also formed.When the cotyledons emerge completely from the endosperm envelopethe latter shows highly vacuolate cells but still rich in cytoplasmicorganelles, lipids, protein bodies and starch grains. Adventitious shoots were induced on cotyledons of Pinus pinea. Among seven salt formulations, three carbon sources (sucrose, glucose and fructose), and two cytokinins [6-benzyladenine (BA) and thidiazuron (TDZ)] with and without [-naphthaleneacetic acid (NAA)], cotyledons grown on 1/2 MS medium containing sucrose and 30 M BA produced the highest frequency of shoot organogenesis (>90%) and mean number of shoots/explant (>40% of cotyledons produced over 20 shoots/cotyledon). As for the site of shoot organogenesis along the cotyledonary explant, the highest number of shoots per explant was observed along the basal segment of the cotyledon (distal to the hypocotyl) over all cytokinin and auxin treatments tested. Shoots were elongated on a growth regulator-free medium containing activated charcoal.  相似文献

The ascorbate and glutathione systems have been studied during the first stages of germination in orthodox seeds of the gymnosperm Pinus pinea L. (pine). The results indicate that remarkable changes in the content and redox balance of these metabolites occur in both the embryo and endosperm; even if with different patterns for the two redox pairs. Dry seeds are devoid of the ascorbate reduced form (ASC) and contain only dehydroascorbic acid (DHA). By contrast, glutathione is present both in the reduced (GSH) and in the oxidized (GSSG) forms. During imbibition the increase in ASC seems to be mainly caused by the reactivation of its biosynthesis. On the other hand, the GSH rise occurring during the first 24 h seems to be largely due to GSSG reduction, even if GSH biosynthesis is still active in the seeds. The enzymes of the ascorbate--glutathione cycle also change during germination, but in different ways. ASC peroxidase (EC 1.11.1.11) and glutathione reductase (EC 1.6.4.2) activities progressively rise both in the embryo and in endosperm. These changes are probably required for counteracting production of reactive oxygen species caused by recovery of oxidative metabolism. The two enzymes involved in the ascorbate recycling, ascorbate free radical (AFR) reductase (EC 1.6.5.4) and DHA reductase (EC 1.8.5.1), show different behaviour: the DHA reductase activity decreases, while that of AFR reductase remains unchanged. The relationship between ascorbate and glutathione metabolism and their relevance in the germination of orthodox seeds are also discussed.  相似文献

The origin and natural range of the Stone pine (Pinus pinea L.) has been questioned for more than a century. In this work, we focus the investigation on one of the most important and controversial regions, viz., the Iberian Peninsula and, specifically, the Huelva and Cadiz populations in Andalusia, one of the most representative population cores. Although some authors maintain that it is an autochthonous Iberian species, most of them consider it to be exotic. From this idea, many works have been done and a sintaxonomic scheme has been created, which is accepted by the majority of the scientific community, not including Pinus pinea, nor its formations, since they are considered as man-induced forest crops. However, Stone has been present for several thousand years in the Iberian Peninsula and in the territory studied, as several paleobotanic and historical data show, proving that Pinus pinea is an autochthonous species of this region. This is a clear consequence to the field of geobotany, since – at least – the Stone pine woodlands from the Iberian Southeast must be considered as communities predominated by an autochthonous species that must be included in the sintaxonomichal schemes. Summary The inorganic carbon fixation patterns of Isoetes lacustris and Lobelia dortmanna from an oligotrophic Scottish loch have been examined by following titratable acidity changes in plant sap and light/dark 14CO2 incorporation by roots and shoots. The diurnal pattern of titratable acidity changes in I. lacustris suggests crassulacean acid metabolism (CAM) while the lack of any change in titratable acidity in L. dortmanna suggests C3 metabolism. Of the carbon fixed by L. dortmanna, 99.9% was taken up through the roots and fixation occurred primarily during the day. In Isoetes, CO2 was taken up by both roots and shoots and during both day and night. Regardless of the site of CO2 uptake, fixation occurred only in the shoots of both plants. Analysis of carbon isotope ratios of plant organic material was used to further investigate the photosynthetic mechanisms of these Isoetids. Considering the absence of a nighttime peak in titratable acidity in L. dortmanna, the 13C (=13C plant-13C source) value of the shoots of L. dortmanna (-14.2) is indicative of C3 photosynthesis limited by the rate of CO2 diffusion. The less negative of I. lacustris (-6.0) is consistent with both dark acidification of CAM and CO2 limited C3 photosynthesis. This is in contrast to the terrestrial Isoetes durieui which is shown to have a value which is similar to a terrestrial C3 plant. The carbon fixation patterns of these Isoetids suggest that the CO2 concentration in the loch may be growth limiting, and that root uptake and/or dark acidification are means of optimising CO2 supply. However, in view of the relatively high levels of CO2 in sediment and bulk water, it is suggested that low levels of nutrients may also limit growth in these plants.  相似文献

Summary The toxicity of three antibiotics commonly used to eliminate Agrobacterium tumefaciens from plant tissue during transformation were tested to determine their effect on Pinus pinea L. morphogenesis. Cefotaxime and vancomycin at a dose of 250 μg ml−1, as well as ticarcillin at 300 μg ml−1, were essentially nontoxic to the culture and significantly enhanced regeneration and shoot development. Kanamycin, a widely used selection agent for plant transformation, strongly inhibited regeneration even at very low doses. On the contrary, phosphinothricin and its ammonium salt glufosinate in the commercial formulation Finale? proved to be the best selection agents because they were less toxic to stone pine cotyledons. Thus, they could be applied immediately after transformation to avoid escapes. Other schemes for selection and recovery of transgenic stone pine trees are discussed.  相似文献

The possible mechanism(s) by which supplemental UV-B radiation alleviates the adverse effects of summer drought in Mediterranean pines (Petropoulou et al. 1995) were investigated with seedlings of Pinus pinea. Plants received ambient or ambient plus supplemental UV-B radiation (biologically equivalent to a 15% ozone depletion over Patras, 38.3° N, 29.1° E) and natural precipitation or additional irrigation. Treatments started on 1 February, 1994 and lasted up to the end of the dry period (29 September). In well-watered plants, UV-B radiation had no influence on photosystem II photochemical efficiency and biomass accumulation. Water stressed plants suffered from needle loss and reduced photosystem II photochemical efficiency during the summer. These symptoms, however, were less pronounced in plants receiving supplemental UV-B radiation, resulting in higher total biomass at plant harvest. Laboratory tests showed that enhanced UV-B radiation did not improve the tolerance of photosystem II against drought, high light, high temperature and oxidative stress. Enhanced UV-B radiation, however, improved the water economy of water stressed plants, as judged by measurements of needle relative water content. In addition, it caused an almost two-fold increase of cuticle thickness. No such UV-B radiation effects were observed in well-watered pines. The results indicate that the combination of water stress and UV-B radiation may trigger specific responses, enabling the plants to avoid excessive water loss and, thereby, maintain a more efficient photosynthetic apparatus during the summer. The extent of this apparently positive UV-B radiation effect would depend on the amount of summer precipitation. Viable protoplasts of Vigna sublobata L. were isolated enzymatically from hypocotyls of axenic seedlings. Protoplast yields were dependent upon seedling age, with maximum yields (2.25 ± 0.35 × 106 g fwt–1) from seedlings aged 6 d. Protoplasts regenerated cell walls and underwent sustained divisions when cultured in either agarose-solidified or liquid K8P medium. The plating density affected the division frequency and plating efficiency; the division frequency (68 ±0 6.0%) was maximum at 4.0 × 104 ml–1 while plating efficiency was maximum (1.3 ± 0.1%) at 5.0 × 104 ml–1. Dividing protoplasts developed into microcalli, which produced glossy green compact nodular calli on transfer to 8.0 gl–1 w/v agar-solidified medium containing MS salts, B5 organic components, 30 g l–1 sucrose, NAA (0.2–0.5 mg l–1), zeatin riboside (0.5–2.0 mg l–1) and GA3 (0.5–1.0 mg l–1). These calli, after sub-culture on the same medium, produced shoot buds which underwent elongation following transfer of tissues to 6.0 g l–1 agar-solidified B5 medium containing 30g l–1 sucrose, IBA (0.01 mg l–1) and BAP (1.0 mg l–1). Summary The spatial pattern of recruitment and seedling survival was studied in an ant-dispersed sedge (Carex pilulifera L.) in a forest clearing in Denmark. Seedlings were generally more aggregated than juvenile and adult plants. Recruitment distances were skewed towards larger values. While 72% plants of presumably ant-dispersed origin survived for 5 years, only 13% auto-dispersed plants were still alive, predominantly as persistent seedlings. The survivorship showed a negative exponential decrease (Deevey type II) with an expected half-life of 17 months for auto-, and 10 years for ant-dispersed plants. Growth-rate and fecundity were significantly larger for ant-dispersed plants than for auto-dispersed plants. Computer simulation was used to test three different dispersal models and two mortality types on the recruitment pattern observed in the field. The simulations confirmed the importance of ant dispersal for recruitment, but gave no conclusive evidence for evaluating recruitment from ant-nests. High mortality levels, comparable to observed predation levels, were needed to simulate field conditions. Density-dependent mortality proved more powerful than distance-dependent mortality in the simulations. Results are discussed in the context of current hypotheses of seed dispersal. While the directed dispersal hypothesis could not be rejected, results were generally more favourable to the escape hypothesis.  相似文献

 This study is the first report of a protocol for transfer and expression of foreign chimeric genes into cotyledons excised from Pinus pinea L. embryos. Agrobacterium tumefaciens EHA105 harbouring the plasmid p35SGUSint was more infective than LBA4404 or C58 GV3850, as determined by the percentage of cotyledons showing uidA expression. Factors which significantly affected the T-DNA transfer included: (1) preinduction and concentration of bacteria, (2) days of coculture and (3) the wounding procedure applied. More efficient transfer of the uidA gene was achieved growing the bacteria in YEP medium at pH 7, infecting the cotyledons according to the sonication-assisted Agrobacterium-mediated transformation procedure with a bacterial density of 1 (OD600 nm) for 5 min, and coculture for 72 h. Using this protocol, 49.7% of the cotyledons showed a diffuse blue staining 7 days after infection. However, all were necrotic 30 days after inoculation. Since a decrease in bacterial density to 0.01 allowed the recovery of about 4% of cotyledons forming buds 1 month after inoculation, we conclude that the high mortality associated with the infection may be related to the hypersensitive response of the plant to bacterial infection. The effect of the method of methionine addition, growth-limiting carbon source (glucose vs sucrose), and culture growth rate on cephalosporin C production was investigated in a Cephalosporium acremonium defined medium fed batch fermentation. Batch addition of methionine, at a concentration of 3 g/L, prior to the start of a fed sucrose fermentation was found to interfere with the ability of the culture to utilize this sugar, thus limiting growth and decreasing cephalosporin C production. Batch methionine addition had no effect on glucose-limited cultures. Concurrent exponential feeding of methionine with sucrose improved both culture growth and productivity. Under the control of identical carbon source limiting feed profiles, sucrose was observed to support greater cephalosporin C production than glucose. Optimal cephalosporin C production in a C. acremonium defined medium fed batch fermentation was obtained through controlling culture growth during the rapid growth phase at a relatively low level with respect to mumax (mu approximately 0.036 h-1) until achieving a desired cell mass with a concurrent sucrose and methionine feed, followed by maintaining relatively vigorous growth (mu approximately 0.01 h-1) with sucrose for the duration of the fermentation.  相似文献

The effect of co-inoculation with Pisolithus tinctorius and a PGPR belonging to the genus Bacillus (Bacillus licheniformis CECT 5106 and Bacillus pumilus CECT 5105) in enhancing growth of Pinus pinea plants and the changes that occurred in rhizosphere microbial communities and the degree of mycorrhization were evaluated. Both bacterial strains of Bacillus promote the growth of Pinus pinea seedlings, but this biological effect does not imply a synergic effect with mycorrhizal infection. However, the positive response to mycorrhiza in a longer-term experiment it could be expected. The introduction of both inocula causes an lateration in the microbial rhizosphere composition, despite the low levels of inocula that were found at the end of the assay.  相似文献

Because estuarine nekton are integrators of the environment abiotic and biotic factors can influence or constrain the relative value of estuarine nursery zones. Recent laboratory experiments on young spot, Leiostomus xanthurus, indicate that both water temperature and salinity significantly affect somatic growth. These experimental data contrast with previous work on young Atlantic croaker,Micropogonias undulatus, white trout,Cynoscion arenarius, and mullet,Mugil sp. Together these results suggest that quantifying vital metrics of nekton, such as survival and growth, along realistic environmental gradients through critical laboratory experiments, allows a more accurate definition of constraints on habitat use. Our studies of factors influencing recruitment success in both winter- and spring/summer-spawning fishes illustrate seasonal as well as intrafamilial differences in growth. Effective management of coastal ecosystems must take into account both variability in abiotic conditions and the nested habitat component, both of which can be modified by coastal development, which could lead to reduced productivity and sustainability of these estuarine landscapes.  相似文献
