The growth and level of xanthophylls of several representative species of green algae were investigated as a possible source of pigmentation for the egg yolk and broiler markets. Chlorella pyrenoidosa 7-11-05 was selected for fermentation studies because of its high level of xanthophylls and wide temperature range for growth. The heterotrophic metabolism was preferred because of the ease of adaptability to present fermentation equipment. When used as the sole carbon source, glucose was the only sugar, among many tested, that gave appreciable growth in illuminated shaken flasks. A dry cell weight of 90 g per liter and total xanthophylls of 450 mg per liter were obtained from 190 g per liter of glucose monohydrate in 168-hr illuminated shaken flasks. Higher levels of glucose decreased yields. In combination with glucose, monosaccharides, such as fructose and galactose, were readily assimilated. The 7-11-05 strain was adapted to galactose as the sole carbon source after six vegetative passages. Light of the proper intensity and duration stimulated total xanthophylls approximately 35%. The effect on dry cell weight and total xanthophylls of seven antibiotics added at various levels in shaken flasks was studied. Erythromycin was essentially stable throughout the fermentation and nontoxic up to 25 μg/ml, with only slight toxicity at higher levels. Both erythromycin and ristocetin were effective in controlling a high incidence of bacterial contamination in 30-liter fermentors. With the higher agitation and aeration rates possible in 30-liter fermentors, dry cell weights in excess of 100 g per liter and total xanthophylls of 467 to 512 mg per liter were readily obtained from 230 to 260 g per liter of glucose in 162-hr illuminated batch-type fermentations. Continuous-feed runs yielded a dry cell weight of 302 g per liter and total xanthophylls of 650 mg per liter from 520 g per liter of glucose. The type of Chlorella cell produced was an important consideration with respect to the availability of the xanthophylls in pigmenting egg yolks and broilers.  相似文献

We previously identified a strong haemagglutination activity in the freshwater unicellular green alga, Chlorella pyrenoidosa. Here, we sought to purify and characterize the haemagglutinin associated with this activity. Ammonium sulfate precipitation, gel filtration on sephacryl S-200 and DEAE-Sepharose ion-exchange chromatography were used to purify the haemagglutinin, which was designated CPH (Chlorella pyrenoidosa haemagglutinin). The molecular weight of CPH was estimated as 58 kDa by SDS-PAGE and 60 kDa by gel filtration of the native protein, indicating that this haemagglutinin exists as a monomer. The haemagglutinin activity of CPH was inhibited by glycoproteins, especially yeast mannan, but not by monosaccharides or disaccharides, indicating that CPH is carbohydrate-specific. In addition to the composition of CPH shown to be rich in glycine and acidic amino acids, heamagglutinating activity of CPH was insensitive to variations in pH or the presence of divalent cations, and atomic force microscopy revealed that the protein is rod-shaped. These results indicate that the characteristics of CPH are consistent with its identification as a haemagglutinin, and suggest that CPH may be a viable candidate for applications in a variety of biomedical fields.  相似文献

A procedure to determine the absolute weight-average molecular weight and molecular weight distribution of agarose and agarose-type polysaccharides by aqueous size-exclusion chromatography coupled with low-angle laser light scattering is described. The molecular weights of the majority of the commercial samples investigated were between 80 000 and 140 000 with a polydispersity lower than 1·7. In contrast, most of the laboratory-extracted agarose-type polysaccharides had lower molecular weights.  相似文献

The lipopolysaccharide (LPS) was isolated from three strains of Salmonella typhimurium, a "smooth" strain, STM 7, the Ra mutant, TV 119, and the Re mutant, SL 1102. The effect of depletion of divalent cations on structure and the effect of deoxycholate on hydrodynamic behavior were studied. The results confirmed previous work by others that divalent cations and hydrophobic forces are important factors influencing LPS size and morphology. The binding of deoxycholate to LPS was measured. When the weight average molecular weights of the deoxycholate-dissociated LPS were determined by sedimentation equilibrium and corrected for bound deoxycholate, the values 5,555, 10,607, and 15,592, respectively, for Re, Ra, and "smooth" LPS were in good agreement with calculated formula weights. Although others have suggested that the basic LPS subunit is a trimer, our results suggest that it exists in the dimeric form.  相似文献

Effect of glutamine and its metabolites (amino acids) on Chlorella glutamine synthetase (GS) (E.C.6.3.1.2) in the presence of Mg or Mn was studied. Purified GS preparation was used, isolated from Chlorella grown in the presence of NH as a sole nitrogen source. Glutamate, aspartate, alanine and glycine inhibit GS activity in the presence of both Mg and Mn. Tryptophane and valine (up to 15 mM) activate GS in the presence of Mn. Tryptophane inhibits GS in the system with Mg. Sinergistic inhibition was observed under the combined effect of amino acids on GS in the presence of Mn and aspartate or alanine. The change of GS activity observed is supposed to be due to the inhibitory effect of glutamine and amino acids studied, since the glutamine content is increased (in 2.5 times for 5 min) and that of alanine and dicarbonic amino acids (for the following 15 min) under NH assimilation in Chlorella cells.  相似文献

1. Intermediary metabolites, arising from the glyoxylate by-pass, were tested for their effect on the activity of isocitrate lyase, the first enzyme of the by-pass. 2. Oxaloacetate and pyruvate inhibited the enzyme when present at concentrations similar to that of the substrate. 3. Inhibition was competitive and did not depend on a non-optimum pH. The affinities determined at the pH optimum were: K(i) (oxaloacetate), 3.7x10(-5)m; K(i) (pyruvate), 6.6x10(-5)m. 4. The significance of the inhibitions is discussed with particular reference to the known inhibition by phosphoenolpyruvate.  相似文献

In the presence of a high concentration of carbonyl cyanide m-chlorophenylhydrazone (CCCP) (4-10(-6) M), the S2 and S3 dark decays are accelerated and become biphasic with a first half-time of 0.6 s. The first fast phase of the decays does not correspond to a simple reduction of S2, S3 back to S0, S1 (i.e. to an acceleration of the deactivation reaction), but to a decrease in the number of oxygen-evolving System II centers. 在研究稻草秸秆对蛋白核小球藻(Chlorella pyrenoidosa)的化感作用时,偶然发现一株对蛋白核小球藻具有强烈的取食作用的鞭毛虫,能有效抑制小球藻的大量繁殖。经形态学观察,该种鞭毛虫具有2片淡黄色色素体,片状、周生;虫体呈球形、椭圆形或卵形,能变形,具2根不等长的鞭毛,从细胞顶部伸出;长鞭毛约为虫体的1.5倍,短鞭毛约为虫体的0.5倍;对蛋白核小球藻的吞噬能力极强,每个虫体可以吞噬3~4个小球藻,虫体大小因吞噬物的多少变化较大。初步确定这种鞭毛虫为棕鞭藻(Ochromonas sp.)。在SE、稻草以及麦粒3种培养基中的种群生长表明,这种棕鞭藻在麦粒培养基最易培养,生长速度最快。结果表明,这种棕鞭藻为混合营养类型,但以吞噬营养为主。  相似文献

Structural analysis of Asian lacquer polysaccharides in Vietnam, Myanmar, Cambodia, Taiwan, and Japan was carried out by a combination of chemical and physical methods, and then their structures were compared with that of a Chinese lacquer polysaccharide reported previously. It was found that the structure of polysaccharides in China and Japan, Taiwan and Vietnam, Myanmar and Cambodia, was similar to each other. The polysaccharides in Myanmar and Cambodia had larger amounts of -arabinose and -rhamnose than those in other Asian lacquer polysaccharides. In addition, the degradation process of lacquer polysaccharide was revealed for the first time by the time-course of GPC measurements of polysaccharide in Aizu, Japan. The results suggest that the molecular weight of polysaccharide in lacquer tree had around with narrow molecular weight distribution and then decreased gradually into two molecular weight fractions of and 23×103 in the proportion of 25 and 75 mol% isolated after 3 weeks.  相似文献

Diffusion-ordered NMR spectroscopy (DOSY) experiments have been carried out on dilute aqueous solutions of uncharged saccharide systems and, in particular, on six well characterized pullulan fractions of different molecular weights. The values of diffusion coefficients and hydrodynamic radii determined for the pullulan fractions are in good agreement with the results obtained with other methodologies such as light scattering. Fitting the diffusion coefficients data as a function of the molecular weight allows for the determination of a calibration curve that can be applied to a wide range of mono-, oligo-, and polysaccharides. Therefore, DOSY is proposed as a versatile tool for achieving a simple estimation of the molecular weight of uncharged polysaccharides. Mixtures of homopolymers of different molecular weight can be nicely separated. An advantage of the method is that the same sample used for the NMR characterization can be used for the molecular weight determination without any further manipulation. Other water soluble polymers, such as poly(ethylene oxide) and poly(vinylpyrrolidone), can be roughly characterized using the same calibration curve.  相似文献

Chlorella pyrenoidosa is a unicellular green algae and has been a popular foodstuff worldwide. However, no reports on the antitumor peptides from such a microalgae are available in the literature. In this study, using low‐temperature high‐pressure extraction, enzymatic hydrolysis, ion exchange, and gel filtration chromatography, we separated a polypeptide that exhibited inhibitory activity on human liver cancer HepG2 cells, and named the polypeptide CPAP (C. pyrenoidosa antitumor polypeptide). Furthermore, the micro‐ and nanoencapsulation of CPAP were investigated by using two methods: complex coacervation and ionotropic gelation. The in vitro release tests revealed that CPAP was well preserved against gastric enzymatic degradation after micro/nanoencapsulation and the slowly controlled release in the intestine could be potentially achieved. These results suggest that CPAP may be a useful ingredient in food, nutraceutical, and pharmaceutical applications. © 2013 American Institute of Chemical Engineers Biotechnol. In the unicellular green alga Chlamydomonas reinhardi (strain y-1), synthesis of the enzymes required for urea hydrolysis is under substrate induction control by urea and under end product repression control by ammonia. Hydrolysis of urea if effected by the sequential action of the discrete enzymes urea carboxylase and allophanate lyase, collectively called urea amidolyase. The carboxylase converts urea to allophanate in a reaction requiring biotin, adenosine 5'-triphosphate, and Mg2+. The lyase hydrolzyes allophanate to ammonium ions and bicarbonate. Neither activity is present in more than trace amounts when cultures are grown with ammonia or urea plus ammonia, or when they are starved for nitrogen for 8 h. Urea in the absence of ammonia induces both activities 10 to 100 times the basal levels. Addition of ammonia to an induced culture causes complete cessation of carboxylase accumulation and an 80% depression of lyase accumulation. Ammonia does not reduce urea uptake by repressed cells, so it does not prevent induction by the mechanism of inducer exclusion. The unicellular green alga Chlorella pyrenoidosa (strain 3 Emerson) also has discrete carboxylase and lyase enzymes, but only the carboxylase exhibits metabolic control.  相似文献

Hermetical microwave was used to degrade Porphyridium cruentum polysaccharides from 2918 to 256.2, 60.66 and 6.55 kDa. The antioxidant properties of different molecular weight polysaccharides were evaluated by determining the scavenging ability of free radicals, inhibitory effects on lipid peroxidation in liver homogenates and hemolysis of mouse erythrocytes. Analysis of physicochemical properties confirmed that microwave degradation might not markedly change the chemical components of the polysaccharides. High-molecular-weight polysaccharides from P. cruentum had no obvious antioxidant activity, but low-molecular-weight fragments after degradation exerted an inhibitory effect on oxidative damage. The 6.55-kDa fragment had stronger antioxidant activity than the 60.66 and 256-kDa fragments.  相似文献
