Spontaneous running in a wheel has emerged as a useful method of exercise in rodents. We investigated how exercise training with a running wheel affects ambient temperatures (T a) at the onset of thermoregulatory responses in rats. Female rats were allowed to run freely in the wheel for 6 months. Sedentary control rats did not exercise during the same period. After the exercise training period, they were loosely restrained and T a values at the onset of tail skin vasodilation and cold- induced thermogenesis were determined by raising or lowering T a. Resting levels of core temperature and heat production of the exercise-trained rats were significantly higher than those of the controls. T a values at the onset of tail skin vasodilation and cold-induced thermogenesis of the exercise-trained rats were higher than those of the controls. The results suggest that, in rats, exercise training with a running wheel elevates ambient temperatures for heat loss and heat production, which may then contribute to maintaining the core temperature at a high level. This study assesses body composition changes and their time course during the initial days of acclimatization to high altitude (HA). Comparisons were made between gradual and acute induction to HA using 60 male lowlander volunteers (24–28 years of age) divided into two equal groups for inducting them to HA. Thirty subjects were air-lifted from sea level (SL) to 3500 m HA in 1 h. These subjects were air inductees (AI). The other 30 subjects were transported in 4 days by road to the same location at 3500 m. These were road inductees (RI). After remaining for 15 days at 3500 m both groups were inducted to 4200 m by road. All the subjects could not reach the various altitudes at the same time due to logistical problems. Ultimately, data for each altitude (SL, 3500 m and 4200 m) were available for only 26 RI subjects and 10 AI subjects. Skinfold thickness (SKF) measurements for the subscapular, thigh, triceps, biceps, juxtanipple, umbilicus, suprailiac and calf regions were taken in order to calculate fat percentages. Measurements were taken at SL and on days 1 and 9 at both 3500 m and 4200 m. On day 1 at 3500 m, RI showed a significant fall in body weight (BW) with respect to SL but AI maintained it. On subsequent days at HA both groups showed a significant fall in BW and lean body mass but not in percentage fat. SKF in the biceps and triceps regions decreased significantly but in the umbilicus and suprailiac regions it significantly increased at HA in both groups. Body composition, along with other parameters, is discussed determining the acclimatization schedule for sojourners at HA. Possibly, translocation of body fat takes place from the periphery to deep body fat depots in the core/main trunk due to the cold at HA. A mathematical model for predicting human thermal and regulatory responses in cold, cool, neutral, warm, and hot environments has been developed and validated. The multi-segmental passive system, which models the dynamic heat transport within the body and the heat exchange between body parts and the environment, is discussed elsewhere. This paper is concerned with the development of the active system, which simulates the regulatory responses of shivering, sweating, and peripheral vasomotion of unacclimatised subjects. Following a comprehensive literature review, 26 independent experiments were selected that were designed to provoke each of these responses in different circumstances. Regression analysis revealed that skin and head core temperature affect regulatory responses in a non-linear fashion. A further signal, i.e. the rate of change of the mean skin temperature weighted by the skin temperature error signal, was identified as governing the dynamics of thermoregulatory processes in the cold. Verification and validation work was carried out using experimental data obtained from 90 exposures covering a range of steady and transient ambient temperatures between 5°C and 50°C and exercise intensities between 46 W/m2 and 600 W/m2. Good general agreement with measured data was obtained for regulatory responses, internal temperatures, and the mean and local skin temperatures of unacclimatised humans for the whole spectrum of climatic conditions and for different activity levels. Body size affects survival probabilities, reproductive output and individual fitness in many organisms. In freshwater zooplankton, traits ranging from demographic rates to community composition depend on body size, and predation is most often identified as the selection pressure determining body size. We examined the extent to which stage-specific growth trajectories and body sizes of copepod crustaceans are constrained, independent of selection. We used exuviae shed at each molt to quantify the relationship between size at molting and growth during the subsequent instar for two common, herbivorous calanoid copepods, Boeckella triarticulata and Diaptomus leptopus. Individuals of both species were raised under diets of different food quality or quantity, and at different temperatures. Size at molting varied little among individuals of both species, as a consequence of a persistent negative relationship between size at molting and subsequent (absolute) growth increment. Individuals that were small when they molted grew more during the subsequent instar than individuals that were large. This relationship was statistically significant for nearly all instars of both species raised in different food or temperature conditions, and not affected by food quality, food quantity or temperature. Our results indicate that body size is constrained or regulated over much of the copepod life cycle, independent of the effects of environmental conditions (food, temperature, predation). The purpose of this investigation was to examine the effects of aging and aerobic fitness on exercise- and methylcholine-induced
sweating responses during heat acclimation. Five younger [Y group – age: 23±1 (SEM) years; maximal oxygen consumption (V.O2max): 47±3 ml·kg–1·min–1], four highly fit older (HO group – 63±3 years; 48±4 ml·kg–1·min–1) and five normally fit older men (NO group – 67±3 years; 30±1 ml·kg–1·min–1) who were matched for height, body mass and percentage fat, were heat acclimated by daily cycle exercise (≈35% V.O2max for 90 min) in a hot (43°C, 30% RH) environment for 8 days. The heat acclimation regimen increased performance time, lowered
final rectal temperature (T
re) and percentage maximal heart rate (%HRmax), improved thermal comfort and decreased sweat sodium concentration similarly in all groups. Although total body sweating
rates (M.sw) during acclimation were significantly greater in the Y and HO groups than in the NO group (P<0.01) (because of the lower absolute workload in the NO group), the M.sw did not change in all groups with the acclimation sessions. Neither were local sweating rates (m.
sw) on chest, back, forearm and thigh changed in all groups by the acclimation. The HO group presented greater forearm m.
sw (30–90 min) values and the Y group had greater back and thigh m.
sw (early in exercise) values, compared to the other groups (P<0.001). In a methylcholine injection test on days immediately before and after the acclimation, the order of sweat output
per gland (SGO) on chest, back and thigh was Y>HO>NO, and on the forearm Y=HO>NO. No group differences were observed for activated
sweat gland density at any site. The SGO at the respective sites increased in the post-acclimation test regardless of group
(P<0.01), but on the thigh the magnitude of the increase was lower in the NO (P<0.02) and HO (P=0.07) groups than in the Y group. These findings suggest that heat tolerance and the improvement with acclimation are little
impaired not only in highly fit older but also normally fit older men, when the subjects exercised at the same relative exercise
intensity. Furthermore, the changes induced by acclimation appear associated with an age-related decrease in V.O2max. However methylcholine-activated SGO and the magnitude of improvement of SGO with acclimation are related not only to V.O2max but also to aging, suggesting that sensitivity to cholinergic stimulation decreases with aging.

Komodo dragons from hatchlings (≈0.1 kg) to adults (≤80 kg) express the full magnitude of varanid species size distributions. We found that all size groups of dragons regulated a similar preferred body temperature by exploiting a heterogeneous thermal environment within savanna, forest and mangrove habitats. All dragons studied, regardless of size, were able to regulate a daytime active body temperature within the range 34–35.6 °C for 5.1–5.6 h/day. The index of effectiveness of thermoregulation (a numerical rating of thermoregulatory activity) was not different among size groups of dragons. However, the index of closeness of thermoregulation, which rates the variability of body temperature, suggests a greater precision for regulating a preferred body temperature for medium compared to small and large dragons. Reference copper cylinders simulating small, medium and large Komodo dragons heated and cooled at the same rate, whereas actual dragons of all size groups heated faster than they cooled. Larger dragons heated and cooled more slowly than smaller ones. The mean operative environmental temperatures of copper cylinders representing medium sized dragons were 42.5, 32.0 and 29.4° C for savannah, forest and mangrove habitats, respectively. The index for average thermal quality of a habitat as measured by the absolute difference between operative environmental temperature and the dragon’s thermal range suggests the forest habitat offers the highest thermal quality to dragons and the savannah the lowest. The percent of total daytime that the operative environmental temperature was within the central 50% of the body temperatures selected by dragons in a thermal gradient (Phillips, 1984) was 45%, 15%, and 9% for forest, mangrove and savannah, respectively. Forest habitat offers the most suitable thermal environment and provides the greatest number of hours with conditions falling within the dragon’s thermal activity zone. 相似文献

To study the relationships between different species of the Einkorn group, 408 accessions of Triticum monococcum, T. boeoticum, T. boeoticum ssp. thauodar and T. urartu were analyzed electrophoretically for their protein composition at the Gli-1 and Gli-2 loci. In all the species the range of allelic variation at the loci examined is remarkable. The gliadin patterns of T. monococcum and T. boeoticum were very similar to one another but differed substantially from those of T. urartu. Several accessions of T. boeoticum and T. monococcum were shown to share the same alleles at the Gli-1 and Gli-2 loci, confirming the recent nomenclature that considers these wheats as different subspecies of the same species, T. monococcum. The gliadin composition of T. urartu resembled that of the A genome of polyploid wheats more than did T. boeoticum or T. monococcum, supporting the hypothesis that T. urartu, rather than T. boeoticum, is the donor of the A genome in cultivated wheats. Because of their high degree of polymorphism the gliadin markers may
help in selecting breeding parents from diploid wheat germ plasm collections and can be used both to search for valuable genes
linked to the gliadin-coding loci and to monitor the transfer of alien genes into cultivated polyploid wheats.

Daily torpor in the gray mouse lemur (Microcebus murinus) in Madagascar: energetic consequences and biological significance 总被引:5,自引:0,他引:5

Patterns and energetic consequences of spontaneous daily torpor were measured in the gray mouse lemur (Microcebus murinus) under natural conditions of ambient temperature and photoperiod in a dry deciduous forest in western Madagascar. Over a
period of two consecutive dry seasons, oxygen consumption (VO2) and body temperature (T
b) were measured on ten individuals kept in outdoor enclosures. In all animals, spontaneous daily torpor occurred on a daily
basis with torpor bouts lasting from 3.6 to 17.6 h, with a mean torpor bout duration of 9.3 h. On average, body temperatures
in torpor were 17.3±4.9°C with a recorded minimum value of 7.8°C. Torpor was not restricted to the mouse lemurs’ diurnal resting
phase: entries occurred throughout the night and arousals mainly around midday, coinciding with the daily ambient temperature
maximum. Arousal from torpor was a two-phase process with a first passive, exogenous heating where the T
b of animals increased from the torpor T
b minimum to a mean value of 27.1°C before the second, endogenous heat production commenced to further raise T
b to normothermic values. Metabolic rate during torpor (28.6±13.2 ml O2 h–1) was significantly reduced by about 76% compared to resting metabolic rate (132.6±50.5 ml O2 h–1). On average, for all M. murinus individuals measured, hypometabolism during daily torpor reduced daily energy expenditure by about 38%. In conclusion, all
these energy-conserving mechanisms of the nocturnal mouse lemurs, with passive exogenous heating during arousal from torpor,
low minimum torpor T
bs, and extended torpor bouts into the activity phase, comprise an important and highly adapted mechanism to minimize energetic
costs in response to unfavorable environmental conditions and may play a crucial role for individual fitness.

The map positions of five previously described phytochrome genes have been determined in tomato (Lycopersicon esculentum Mill.) The position of the yg-2 gene on chromosome 12 has been confirmed and the classical map revised. The position of the phytochrome A (phy A)-deficient
fri mutants has been refined by revising the classical map of chromosome 10. The position of the PhyA gene is indistinguishable from that of the fri locus. The putative phyB1-deficient tri mutants were mapped by classical and RFLP analysis to chromosome 1. The PhyB1 gene, as predicted, was located at the same position. Several mutants with the high pigment (hp) phenotype, which exaggerates phytochrome responses, have been reported. Allelism tests confirmed that the hp-2 mutant is not allelic to other previously described hp (proposed here to be called hp-1) mutants and a second stronger hp-2 allele (hp-2

) was identified. The hp-2 gene was mapped to the classical, as well as the RFLP, map of chromosome 1.

Background
This study investigated the effect of hydration differences on body fluid and temperature regulation between tropical and temperate indigenes exercising in the heat.Methods
Ten Japanese and ten Malaysian males with matched physical characteristics (height, body weight, and peak oxygen consumption) participated in this study. Participants performed exercise for 60 min at 55% peak oxygen uptake followed by a 30-min recovery at 32°C and 70% relative air humidity with hydration (4 times each, 3 mL per kg body weight, 37°C) or without hydration. Rectal temperature, skin temperature, heart rate, skin blood flow, and blood pressure were measured continuously. The percentage of body weight loss and total sweat loss were calculated from body weight measurements. The percentage change in plasma volume was estimated from hemoglobin concentration and hematocrit.Results
Malaysian participants had a significantly lower rectal temperature, a smaller reduction in plasma volume, and a lower heart rate in the hydrated condition than in the non-hydrated condition at the end of exercise (P <0.05), whereas Japanese participants showed no difference between the two hydration conditions. Hydration induced a greater total sweat loss in both groups (P <0.05), and the percentage of body weight loss in hydrated Malaysians was significantly less than in hydrated Japanese (P <0.05). A significant interaction between groups and hydration conditions was observed for the percentage of mean cutaneous vascular conductance during exercise relative to baseline (P <0.05).Conclusions
The smaller reduction in plasma volume and percentage body weight loss in hydrated Malaysians indicated an advantage in body fluid regulation. This may enable Malaysians to reserve more blood for circulation and heat dissipation and thereby maintain lower rectal temperatures in a hydrated condition. 相似文献11.

A rapidly growing, long-term suspension culture derived from Triticum aestivum L. (wheat) was synchronized using hydroxyurea and colchicine, and a chromosome suspension with chromosomes was made. After staining with the DNA-specific fluorochromes Hoechst 33258 and Chromomycin univariate and bivariate flow-cytometry histograms showed 15 clearly resolved peaks corresponding to individual chromosome
types or groups of chromosomes with similar DNA contents. The flow karyotype was closely similar to a histogram of DNA content
measurements of Feulgen-stained chromosomes made by microdensitometry. We were able to show the stability of the flow karyotype
of the cell line over a year, while a parallel subculture had a slightly different, stable, karyotype following different
growth conditions. The data indicate that flow cytometric analysis of plant karyotypes enables accurate, statistically precise
chromosome classification and karyotyping of cereals. There was little overlap between individual flow-histogram peaks, so
the method is useful for flow sorting and the construction of chromosome specific-recombinant DNA libraries. Using bivariate
analysis, the AT:GC ratio of all the chromosomes was remarkably similar, in striking contrast to mammalian flow karyotypes.
We speculate about a fundamental difference in organization and homogenization of DNA sequences between chromosomes within
mammalian and plant genomes.

Hair is considered to be a basic mammalian feature that provides protection and insulation, promoting energy conservation and survival. To quantify the functional significance of mammalian pelage, we tested the short-term experimental effects of fur removal in a natural population of the California vole, Microtus californicus, in winter. The daily energy expenditure (DEE) of seven voles was directly measured in the field using stable isotopes, first with the animals in their natural condition and then again after experimental removal of pelage by shaving. The initial mean±SD DEE of 96.0±23.1 kJ/day increased by only about 10%, to 106.3±21.4 kJ/day, following shaving. The voles showed a body mass loss of 5%, about half of which was due to fur removal. Comparing slightly larger samples of all animals whose local survival could be documented, we found 100% survival over the 5 initial days of the experimental manipulation in 16 control animals and nine shaved animals; over the following 3 weeks the survival of shaved mice did not differ significantly from that of controls. We were surprised that the average increase in energy expenditure of voles without fur was so modest, though the range of individual values was great, and likewise we were surprised that shaved voles survived as well as they did. M. californicus survives naturally in winter under conditions of social aggregation that include huddling together of individuals in nests; this situation probably provided our experimentally shaved voles an opportunity to minimize the energetic disadvantages of pelage loss. They may also have employed a variety of compensatory physiological and behavioral responses, including reduction in activity time and food intake, and perhaps a related small decline in body mass. Our limited sample sizes made it difficult to detect subtle differences that may have been biologically significant in the system we studied. Nonetheless, we can reaffirm that fur has an insulative value that promotes energy economy and survival. However, we also conclude that mammalian physiology and behavior are sufficiently complex and flexible that a variety of responses can be deployed to promote survival under unusual circumstances such as those of our experimental test. A method based upon targetting of intro-gressed markers in a Phomopsis-resistant line (R) of cultivated sunflower, issuing
from a H. argophyllus cross was used to mark the Phomopsis resistance regions. Our study was based upon 203 families derived from a cross between an inbred line susceptible to Phomopsis (S1) and the introgressed resistant line (R).
Families were checked for Phomopsis resistance level in a design with replicated plots and natural infection was re-inforced
by pieces of contaminated stems. Thirty four primers were employed for RAPD analysis. Out of 102 polymorphic fragments between
(S1) and H. argophyllus, seven were still present in (R) suggesting that they marked introgressions of H. argophyllus into (R). The plants were scored for the presence or absence of 19 fragments obtained from five primers, and the relationships between
the presence/absence of fragments in plants and Phomopsis resistance/susceptiblity in the progenies was determined by using an analysis of variance. We found that at least two introgressed regions, as well as favourable
factors from sunflower, contributed to the level of Phomopsis resistance in cultivated sunflower.

Both whole-body heat exposure and intraperitoneal heating (IPH) result in a body temperature (T
b) fall that occurs once heating is abated (”hyperthermia- induced hypothermia”). This phenomenon involves a decrease in the
threshold T
b (T
b-thresh) for activation of metabolic heat production (cold defense). Whether the T
b-thresh for ear skin vasodilation (heat defense) also changes during hyperthermia-induced hypothermia remains unknown. In experiment
1, we applied IPH to guinea pigs by perfusing water through a preimplanted intraperitoneal thermode and delivered the total
heat load of either approximately 1.5 kJ (”short” IPH; perfusion duration: 14 min) or approximately 3.0 kJ (”long” IPH; 40
min). Short IPH caused skin vasodilation and a 1.1°C rise in T
b; no hypothermia occurred when IPH ceased. Long IPH caused vasodilation and hyperthermia of a comparable magnitude (1.4°C)
that were followed by a T
b fall to 1.9°C below the preheating value. In experiment 2, the T
b-thresh for skin vasodilation was measured twice: at the beginning of long IPH and at the nadir of the post-IPH hypothermia. The
two T
b-thresh values were 39.0 (SEM 0.1)°C and 39.2 (SEM 0.2)°C respectively. In the controls, the T
b-thresh was measured at the beginning and after short IPH; both control values were 39.0 (SEM 0.2)°C. We conclude that the hyperthermia-
induced hypothermia, although previously shown to be coupled with a decrease in the T
b-thresh for cold defense, occurs without any substantial change in the T
b-thresh for heat defense. We speculate that postheating thermoregulatory disorders are associated with threshold dissociation, thus
representing the poikilothermic (wide dead-band) type of T
b control.

Rhizanthes lowii (Beccari) Harms (Rafflesia- ceae) is a parasitic plant that grows in the understory of the rainforest in South-East Asia. This plant does not have leaves, stems, or photosynthetic tissue and is characterised by the emission of a strong odour that attracts the natural pollinators, carrion flies. Flowers that volatilise odorous compounds and attract carrion flies, beetles and other insects are often thermogenic. Here we present evidence of both thermogenesis and thermoregulation in R. lowii from microclimate and tissue temperatures measured during different stages of flower development in R. lowii, in natural conditions in Brunei, Borneo. Endothermy was detected in young and mature buds as well as in blooming flowers and even in decaying tissues 3 or more days after blooming. Genetic variation in protoplast-derived rice (Oryza sativa L.) plants was characterized using first and second generation selfed progenies. A total of 133 regenerated plants were obtained from ten protoplasts of the japonica rice cultivar Nipponbare. Sixty two regenerated plants which set enough seeds for the subsequent field tests at the next generation and were derived from five protoplasts were selected, and their selfed seeds were used as the first selfed-seed progeny generation). Fifteen plants were selected from each of the 15 lines, and their selfed seeds were used for tests at the generation. Thirty seven lines (60%) segregated plants with detrimental mutant characters of yellow-green phenotype, dwarf stature, dense and short panicle, or low seed fertility. According to the segregation patterns in the lines having mutated plants among those originated from the same protoplasts, the stages of mutation induction were estimated. Additionally, five quantitative traits were changed in almost all and lines. Varied quantitative traits of heading date, number of spikelets per panicle, and seed fertility, were in a heterozygous state. However, culm and panicle lengths showed high uniformity, whereas reduced culm and panicle lengths were caused by mutational changes in polygenes and/or multiple genes. To assess the role of bone marrow fat in survival during a period of negative energy balance, we investigated the relationship between the time-course of marrow fat mobilization and the metabolic states associated with body fuel utilization during a prolonged fast. In order to mimic the winter fast of the barn owl (Tyto alba), captive birds were subjected to fasts of various durations at 5 °C ambient temperature. Body mass and plasma metabolites were used to determine the metabolic state at the end of fasting. Skeleton composition remained unchanged throughout phase II of fasting, during which the birds essentially rely on lipid fuels. During the following phase III, characterized by an increase in net body protein breakdown, the lipid mass in skeleton marrow decreased sharply by 78%, concomitant with an increase of the bone water content. This marrow fat mobilization occurred in all parts of the skeleton. This observation supports the hypothesis that bone marrow fat is not only involved in local nutrition, but can also be used as a lipid reserve for total energy requirements. However, in contrast to other fat deposits, marrow fat is mobilized only during phase III of the fast, when the last shift from lipid to protein fuel metabolism occurs. Thus, metabolic and/or hormonal changes associated with this transition could be involved in bone marrow fat mobilization. It was shown previously in infant rats (Rattus norvegicus) that the ability to produce heat in the cold using brown adipose tissue (BAT) is closely related to the ability to maintain
cardiac rate. When the limits of BAT thermogenesis were exceeded, interscapular temperature (which reflects the temperature
of the interscapular BAT depot) and cardiac rate fell together. As an extension of this earlier study, the relation between
BAT thermogenesis and cardiac rate was examined here in the golden hamster (Mesocricetus auratus), a species whose young do not exhibit BAT thermogenesis until the end of the 2nd week postpartum. It was found that 3 to
12-day-old hamsters were unable to increase shivering or nonshivering thermogenesis in the cold and exhibited decreases in
cardiac rate that proceeded in lock-step with decreases in interscapular temperature. In contrast, as the thermogenic capability
of hamsters increased after 12 days of age, cardiac rate was maintained within narrow limits across a wide range of air temperatures.
These results support the hypothesis that heat produced by BAT helps to warm the heart and thus aids in the maintenance of
cardiac rate during cold exposure.

Plant breeders would like to predict which biparental populations will have the largest genetic variance. If the population genetic variance could be predicted using coefficient of parentage or genetic distance estimates based on molecular marker data, breeders could choose parents that produced segregating populations with a large genetic variance. Three biparental soybean {Glycine max (L.) Merr.} populations were developed by crossing parents that were closely related, based on pedigree relationships. Three additional biparental populations were developed by crossing parents that were assumed to be unrelated. The genetic variance of each population was estimated for yield, lodging, physiological maturity, and plant height. Coefficient of parentage was calculated for each pair of parents used to develop the segregating populations. Genetic distance was determined, based on the number of random amplified polymorphic markers (RAPD) that were polymorphic for each pair of parents. Genetic distance was not associated with the coefficient of parentage or the magnitude of the genetic variance. The genetic variance pooled across the three closely related populations was smaller than the genetic variance pooled across the three populations derived from crossing unrelated parents for all four traits that were evaluated. Microsatellites consisting of AT repeats are highly polymorphic in rice genomes and can be used to distinguish between even
closely related japonica cultivars in Japan. Polymorphisms of 20 microsatellite loci were determined using 59 japonica cultivars,
including both domestic and modern Japanese cultivars. Although the polymorphisms of these 20 microsatellite loci indicated
that the Japanese cultivars were genetically quite similar, microsatellites consisting of AT repeats showed high gene diversity
even among such closely related cultivars. Combinations of these hypervariable microsatellites can be employed to classify
individual cultivars, since the microsatellites were stable within each cultivar. An identification system based on these
highly polymorphic microsatellites could be used to maintain the purity of rice seeds by eliminating contamination. A parentage
diagnosis using 17 polymorphic microsatellite loci clearly demonstrated that plants which carried desired chromosome regions
had been selected in breeding programs. Thus, these hypervariable microsatellites consisting of AT repeats should promote
the selection of plants which carry desired chromosomes from genetically similar parents. Backcrossing could also help to
eliminate unnecessary chromosome regions with microsatellite polymorphisms at an early stage in breeding programs.
