We studied the salt stress (100 mM NaCl) effects on the diurnal changes in N metabolism enzymes in tomato seedlings (Lycopersicon esculentum Mill. cv. Chibli F1) that were grown under high nitrogen (HN, 5 mM NO(3)(-)) or low nitrogen (LN, 0.1 mM NO(3)(-)). NaCl stress led to a decrease in plant DW production and leaf surface to higher extent in HN than in LN plants. Total leaf chlorophyll (Chl) content was decreased by salinity in HN plants, but unchanged in LN plants. Soluble protein content was decreased by salt in the leaves from HN and LN plants, but increased in the stems-petioles from LN plants. Nitrate reductase (NR, EC 1.6.1.6) showed an activity peak during first part of the light period, but no diurnal changes were observed for the nitrite reductase (NiR, EC 1.7.7.1) activity. Glutamine synthetase (GS, EC 6.3.1.2) and glutamate synthase (Fd-GOGAT, EC 1.4.7.1) activities increased in HN plant leaves during the second part of the light period, probably when enough ammonium is produced by nitrate reduction. NR and NiR activities in the leaves were more decreased by NaCl in LN than in HN plants, whereas the opposite response was obtained for the GS activity. Fd-GOGAT activity was inhibited by NaCl in HN plant leaves, while salinity did not shift the peak of the NR and Fd-GOGAT activities during a diurnal cycle. The induction by NaCl stress occurred for the NR and GS activities in the roots of both HN and LN plants. Glutamate dehydrogenase (GDH, EC 1.4.1.2) activity shifted from the deaminating activity to the aminating activity in all tissues of HN plants. In LN plants, both aminating and deaminating activities were increased by salinity in the leaves and roots. The differences in the sensitivity to NaCl between HN and LN plants are discussed in relation to the N metabolism status brought on by salt stress.  相似文献

We studied the effect of salinity on the activity of microsomal NADPH:cytochrome P450 reductase (CPR, EC 1.6.2.4) and NADH:ferricytochrome b(5) oxidoreductase (B5R, EC 1.6.2.2) in two dicotyledonous plant species differing in their sensitivity to salt, cotton (Gossypium hirsutum L. cv Ogosta) and common bean (Phaseolus vulgaris L. cv Dobrujanski 7). A significant inhibition of fresh weight of salt-treated bean plants was observed, while cotton was affected to a much lesser degree. NaCl application resulted in a significant increase in the activity of both reductases, but was more pronounced in salt-tolerant cotton. We suppose that alterations in B5R and CPR activities may be targeted to the maintenance of membrane lipids. Most probably, plants use both enzymes (B5R and CPR) and their respective electron donors (NADH and NADPH) to reduce cytochrome b(5), which can donate reducing equivalents to a series of lipid-modification reactions such as desaturation and hydroxylation.  相似文献

The effects of salinity on growth, stomatal conductance, photosynthetic capacity, and carbon isotope discrimination (Δ) of Gossypium hirsutum L. and Phaseolus vulgaris L. were evaluated. Plants were grown at different NaCl concentrations from 10 days old until mature reproductive structures were formed. Plant growth and leaf area development were strongly reduced by salinity, in both cotton and bean. Stomatal conductance also was reduced by salinity. The Δ always declined with increasing external salinity concentration, indicating that stomatal limitation of photosynthesis was increased. In cotton plant dry matter, Δ correlated with the ratio of intercellular to atmospheric CO2 partial pressures (pl/pa) calculated by gas exchange. This correlation was not clear in bean plants, although Δ showed a more pronounced salt induced decline in bean than in cotton. Possible effects of heterogeneity of stomatal aperture and consequent overestimation of pl as determined from gas exchange could explain these results. Significant differences of Δ between leaf and seed material were observed in cotton and bean. This suggests different patterns of carbon allocation between leaves and seeds. The photon yield of O2 evolution determined at rate-limiting photosynthetic photon flux density was insensitive to salinity in both species analyzed. The light- and CO2-saturated rate of CO2 uptake and O2 evolution showed a salt induced decline in both species. Possible explanations of this observation are discussed. O2 hypersensitivity was observed in salt stressed cotton plants. These results clearly demonstrate that the effect of salinity on assimilation rate was mostly due to the reduction of stomatal conductance, and that calculation of pl may be overestimated in salt stressed plants, because of heterogeneity of stomatal aperture over the leaf surface.  相似文献

Two strawberry (Fragaria × ananassa Duch.) cvs Korona and Elsanta differing in their tolerance to NaCl salinity were exposed to 40 and 80 mmol NaCl L?1 for over 4 months in the growing seasons of 2002 and 2003, respectively. However, the osmotic potential, i.e. the NaCl concentration of the root medium, varied during the experiments, because Hoagland solution and demineralized water were added usually once a week in order to push NaCl uptake on the one hand, but to allow leaching the soil after application of demineralized water on the other. Leaching the soil should quickly improve the water relations of the plant, but not affect salt levels within the plant. This strategy was chosen to reduce the effects of water stress and to focus onto the salt-specific impacts of NaCl stress. The salt stress reduced fresh and dry matter of the whole plants and photosynthetically active leaf area, especially in cv. Elsanta. Typical leaf symptoms of Na and Cl stress were detected in both cvs and the combined effects of both toxic ions resulted in the leaf scorching symptoms. Na uptake of both cvs was similar, but distribution of Na within plants was different. Korona was able to protect leaves more efficiently from Na accumulation.Under NaCl stress Korona plants achieved a significant increase of K content in leaves and crowns, while Elsanta showed an increase of K in fruits and petioles. The accumulation of K under evaluated NaCl levels suggests an efficient K uptake system in strawberry plants. Concentrations of Ca were not significantly affected, with the exception of rising levels in roots of Elsanta plants. Concentrations of Mg, Mn and Fe significantly decreased in leaves, while those of Mg and Mn remarkably rose in crowns of both cvs. N content in leaves, petioles, and roots of both cvs increased. In addition it rose in fruits and crowns in cv. Elsanta. A significant limitation of N uptake by competition with Cl did not occur in these plants. Concentrations of P increased in roots and petioles of both cvs, and in fruits of cv. Elsanta. With respect to Zn and Cu, significant concentration changes related to NaCl stress could not be detected.  相似文献

Summary The effects of three levels of N (0, 50 and 100 ppm) and four salinity regimes (0.5, 1.5, 2.5 and 3.5 mmhos/cm) on the growth and mineral composition of dry beans (Phaseolus vulgaris L.) were investigated in a greenhouse experiment. Bean plants treated with N produced more dry weight and contained higher N than the untreated check. Growth and N uptake by bean plants generally decreased with increasing irrigation water salinity at all N levels. High salinity caused severe burning of the margins of older leaves and stunting of growth. At the low salinity levels (0.5 and 1.5 mmhos/cm), N additions had no effect on growth; however, the suppressing effects of higher salinity were alleviated somewhat with N fertilization.The concentration and uptake of Cl and Na increased with increasing salinity; probably the relatively high accumulations of Cl and Na were responsible for growth reductions at high salinity.  相似文献

Melon seedlings (Cucumis melo L. cv.Galia) were grown hydroponically to study the effect of salinity (80 mmol/LNaCl) on phosphate (Pi) uptake and translocation at two levels of Pi (25 μmol/L and 1 mmol/L). Net uptake rates of Pi were determined by depletionof the medium and by plant content. Salinity decreased Pi uptake at low Pi (high affinity uptake mechanism), 25 μmol/L, although no specific competitive inhibition of Pi uptake by Cl was observed. When plants were grown with high Pi (1 mmol/L), the uptake of Pi through the low affinity system was increased by 80 mmol/L NaCl. Salinity also reduced the phosphorus flux, as Pi, through the xylem. It is hypothesised that high levels of NaCl decrease the mobility of Pi stored in vacuoles, and as a result, inhibit export from this storage compartment to other parts of the plant.  相似文献

Uptake and partitioning through the xylem and phloem of K+,Na+, Mg2+ , Ca2+ and Cl were studied over a 9 d intervalduring late vegetative growth of castor bean (Ricinus communisL.) plants exposed to a mean salinity stress of 128 mol m–3NaCl. Empirically based models of flow and utilization of eachion within the whole plant were constructed using informationon ion increments of plant parts, molar ratios of ions to carbonin phloem sap sampled from petioles and stem internodes andpreviously derived information on carbon flow between plantsparts in xylem and phloem in identical plant material. Salientfeatures of the plant budget for K+ were prominent depositionin leaves, high mobility of K+ in phloem, high rates of cyclingthrough leaves and downward translocation of K+ providing theroot with a large excess of K+ . Corresponding data for Na+showed marked retention in the root, lateral uptake from xylemby hypocotyl, stem internodes and petioles leading to low intakeby young leaf laminae and substantial cycling from older leavesback to the root. The partitioning of the anionic componentof NaCl salinity, Cl, contrasted to that of Na+ in thatit was not substantially retained in the root, but depositedmore or less uniformly in stem, petiole and leaf lamina tissues.The flow pattern for Mg2+ showed relatively even depositionthrough the plant but some preferential uptake by young leaves,generally lesser export than import by leaf laminae, and a returnflow of Mg2+ from shoot to root considerably less than the recordedincrement of the root. Ca2+ partitioning contrasted with thatof the other ions in showing extremely poor phloem mobility,leading to progressive preferential accumulation in leaf laminaeand negligible cycling of the element through leaves or root.Features of the response of Ricinus to salinity shown in thepresent study were discussed with data from similar modellingstudies on white lupin (Lupinus albus L.) and barley (Hordeumvulgare L.) The physiological responses of alfalfa (Medicago sativa L. cv. Gilboa) to salinity (100 mM NaCl) and some inorganic nutrients (K+, Ca2+ and N as NO3-) were investigated. Salinity caused a substantial reduction in biomass, carbon assimilation rate, stomatal conductance, water use efficiency, leaf area, relative growth rate, NO3- content and nitrate reductase activity, whereas, transpiration rate was slightly affected. Inclusion of K+, Ca2+ and N as NO3- in plant nutrient medium in combination or alone brought about a marked stimulation in control plants and moderated the salinity caused reductions in NaCl treated plants. Tomato plants (Lycopersicon esculentum Mill, cv. Chibli F1) grown for 10 days on control medium were exposed to differing concentrations of NaCl (0, 25, 50, and 100mM). Increasing salinity led to a decrease of dry weight (DW) production and protein contents in the leaves and roots. Conversely, the root to shoot (R/S) DW ratio was increased by salinity. Na(+) and Cl(-) accumulation were correlated with a decline of K(+) and NO(3)(-) in the leaves and roots. Under salinity, the activities of nitrate reductase (NR, EC 1.6.6.1) and glutamine synthetase (GS, EC 6.3.1.2) were repressed in the leaves, while they were enhanced in the roots. Nitrite reductase (NiR, EC 1.7.7.1) activity was decreased in both the leaves and roots. Deaminating activity of glutamate dehydrogenase (GDH, EC 1.4.1.2) was inhibited, whereas the aminating function was significantly stimulated by salinity in the leaves and roots. At a high salt concentration, the nicotinamide adenine dinucleotide reduced (NADH)-GDH activity was stimulated concomitantly with the increasing NH(4)(+) contents and proteolysis activity in the leaves and roots. With respect to salt stress, the distinct sensitivity of the enzymes involved in nitrogen assimilation is discussed.  相似文献

Olive ( Olea europaea L. cv. Frantoio) plants grown hydroponically in a glasshouse were supplied with half-strength Hoagland solutions containing 0, 50, 100, and 200 m M NaCl for 4 weeks and subsequently supplied with the standard solution without NaCl to relieve salinity stress. Two complete stress-relief cycles were repeated on the same plant material during one growing season. Growth was inhibited at all salt levels, but most growth parameters of plants treated with 50 or 100 m M NaCl returned to control levels after 4 weeks of relief. More severely stressed plants (200 m M NaCl) recovered to only 60% of the growth of the controls after 4 weeks. During relief, plants treated with 50 and 100 m M NaCl had net photosynthetic rates and stomatal conductances higher than the controls. Increasing the NaCl concentration of the external solution from 0 to 200 m M decreased both leaf pre-dawn water potential (from -0.3 to -1.0 MPa) and osmotic potential (from -2.1 to -2.7 MPa). The sodium concentration in the leaves of plants treated with 200 m M NaCl reached maximum levels of 211 and 388 m M (expressed on a tissue water basis) at the end of the first salinity and relief periods, respectively. Leaf chloride concentrations were 359 and 223 m M at the same sampling dates. These data indicate that the inhibitory effects of salinization on growth and gas exchange of the salt-tolerant olive cv. Frantoio can be readily reversed when salinity is relieved, despite the marked accumulation of potentially toxic ions (Na+. Cl) in the leaf.  相似文献

研究了盐地碱蓬二型性种子中离子含量与刚萌发幼苗耐盐性之间的关系,以及盐分对砂培盐地碱蓬二型性种子的幼苗生长、离子含量及光合特性的影响.棕色种子中离子含量显著高于黑色种子.与对照相比,100和400 mmol/L NaCl对棕色种子幼苗伸长没有抑制作用,却显著抑制黑色种子幼苗的伸长.NaCl处理下棕色种子的幼苗地上部分干重和主茎一级分枝数比黑色种子幼苗高,但二型性种子的幼苗叶片中离子含量、叶绿素含量及光合放氧速率却没有明显差异.上述结果说明盐地碱蓬棕色种子较高的离子含量可能是棕色种子刚萌发幼苗耐盐性较强的重要原因.棕色种子幼苗较高的生物量可能与其较多的分枝数有关.二型种子的这些特征可能决定了其在群落建成中所起到的不同作用.  相似文献

Increasing salinity of growth medium induced a reduction in growth and transpiration rate. The concentrations of chlorophylls and carotenoids were increased in most cases in broad bean leaves while in pea plants they remained more or less unchanged with the rise of salinization up to 80mM NaCl. Thereabove a significant decrease in these contents was observed. A stimulation of the net photosynthetic rate of pea was observed at the lowest levels of NaCl but at the highest levels inhibitory effect was recorded. In broad bean all salinization levels inhibited photosynthetic activity, but dark respiration of both plant species was stimulated. The content of Na+ in the roots and shoots of both species increased at increasing salinity. In broad bean, Ca2+ concentration in shoots and K+ and Ca2+ contents of roots increased at increasing salinization, while in pea plants, the content of K+ and Ca2+ was almost unaffected by salinity. Salinity induced an increase in the content of these ions in pea roots. Mg2+ content in shoots and roots of both broad bean and pea decreased at increasing salinity except in roots of pea, where it was generally increased.  相似文献

Faba bean (Vicia faba L. var. minor cv. Alborea) and pea (Pisum sativum L. cv. Lincoln) plants, inoculated with Rhizobium leguminosarum biovar. viciae strain GRA19, were treated with salt (100 mM NaCl) and/or nitrate (8 mM KNO3) to test whether plants grown with inorganic-nitrogen are more tolerant to salinity than plants entirely reliant upon fixed nitrogen. According to the growth inhibition recorded, pea plants dependent on dinitrogen fixation proved more tolerant to salt stress than those N-fertilized, in contrast to results obtained for faba bean plants. This study therefore confirms that plants dependent on nitrogen fixation are not always more sensitive to salinity than are N-fertilized plants. Nitrate addition did not reduce the specific nitrogenase activity in pea, but did in faba bean. However, nodulation was inhibited in both legumes. The specific nitrogenase activity was more affected by salt treatment in N-fertilized plants for both legumes. The activity of the enzymes mediating ammonium assimilation in nodules (GS, NADH-GOGAT) was inhibited by salt stress both in N-fixing and in N-fertilized pea and faba bean plants.  相似文献

Removal of the shoot above the primary node (detopping) of 3-week-old bean plants ( Phaseolus vulgaris L. cv. Contender) altered the metabolism and development of the remaining leaves. An increase in levels of chlorophyll, protein, stomatal opening, photosynthesis and growth, i.e. rejuvenation of primary leaves, was established within 7 days of detopping. These levels were maintained while the primary leaves of equivalent intact plants senesced.
The flux of xylem solution (mineral ions, cytokinins and water) into leaves is related to the leaf area to be supplied and root supply capacity; it has been suggested that detopping leads to an increased availability of root-supplied solutes and hence rejuvenation of the remaining leaves. This assumes however that root output of solutes is not decreased by the defoliation treatment.
We found that root output of ions (electrical conductivity of passive xylem exudate) in detopped plants was 30% lower than in intact plants after 24 h and 60% lower after 7 days. The output of Ca2+, Mg2+ and K+ were similarly reduced 7 and 14 days after detopping as were fresh and dry weights of roots. Furthermore, neither the calculated xylem flux of ions nor directly measured levels of Ca2+, Mg2+ and K+ were significantly increased in leaves of detopped plants during their rejuvenation. We therefore conclude that root output is tightly coupled to shoot demand and that the apparent rejuvenation of primary leaves caused by detopping bean plants is not a consequence of increased xylem flux of mineral ions into the leaves.  相似文献

Water uptake rate of decapitated root systems of cotton (Gossypium hirsutum L.), tomato (Lycopersicon esculentum L. cv. Rutgers), and kidney bean (Phaseolus vulgaris L.) plants shows an exponential increase with applied suction up to about —1 bar. The water uptake rate was higher on the descending path of applied suction than on the ascending path, indicating a hysteresis effect in the roots. The root resistance in a cotton plant increased between 3-to 5-fold during the photoperiod of 12 hours. The water uptake rate increased with increasing temperature of the root medium up to 30°C in cotton and 25°C in tomato and bean plants.  相似文献

The comparative responses of young olive trees (Olea europaea L. cv “Chemlali”) to different NaCl salinity levels were investigated over 11 months. One-year-old own rooted plants were grown in 10-L pots containing sand and perlite mixture (1:3 v/v). Trees were subjected to three irrigation treatments: CP (control plants that were irrigated with fresh water); SS1 (salt stressed plants irrigated with water containing 100 mM NaCl) and SS2 plants (salt stressed plants irrigated with water containing 200 mM NaCl). Shoot elongation rate, relative water content, leaf water potential and net carbon dioxide exchange rates decreased significantly with increased NaCl salinity level. Under stressed conditions, the increase of Na+ and Cl ions in both leaves and roots was accompanied with that of proline and soluble sugars. The above results show that the accumulation of proline and sugars under stressed conditions could play a role in salt tolerance. The absence of toxicity symptoms under both stress treatments and the superior photosynthetic activity recorded in SS1-treated plants suggest that cv Chemlali is better able to acclimatize to 100 mM NaCl than at 200 mM NaCl. Our findings indicate that saline water containing 100 mM NaCl, the most available water in arid region in Tunisia, can be recommended for the irrigation of cv Chemlali in the arid south of Tunisia.  相似文献

The effect of salinity (100 mM NaCl) on ethylene metabolism in the early phase of vegetative development of several plant species has been investigated. The effects of saline treatment on shoot and root growth, ranged in sensitivity with respect to species: pepper (Capsicum annum L. cv Pairal) > tomato (Lycopersicon esculentum Mill. cv Malpica) > broccoli (Brassica oleraceae L. var. Italica Plenk. cv Marathon F1) ≅ lettuce (Lactuca sativa var. longifolia Lam. cv Inverna) ≅ melon (Cucumis melo L. cv Ruano F1, Roche type) > bean (Phaseolus vulgaris L. cv. Gator Green 15) ≅ spinach (Spinacia oleracea L. cv Boeing) > beetroot (Beta vulgaris L. var. crassa (Alef.) J. Helm. cv Detroit). After saline treatment, ethylene production increased 4.2-fold in pepper shoots. Significant increases were also found in shoots of tomato, broccoli and bean. In contrast, salinity decreased shoot ethylene production rate in melon, spinach, and beetroot. In roots, the general effect of salinity was a decrease in ethylene production, especially in broccoli and bean, except in tomato root, in which a sharp increase in ethylene production occurred. In general, saline treatment increased total ACC concentration in both shoot and root in most of the plant species examined, which was related to plant sensitivity to salinity. For example, pepper shoot was the most sensitive to saline treatment, showing the highest fresh weight inhibition and the highest increase in total ACC concentration (8.5-fold), while, beetroot was less affected by salinity and showed no effect on total ACC concentration in response to saline treatment.  相似文献

The effect of salt stress (NaCl 85.7 or 110 mmol/L) was investigated in the triticale T300 and its parental species, Triticum dicoccum farrum (Triticum df) and Secale cereale cv. Petkus. Triticum df and T300 were more salt-tolerant than the rye (110 mmol/L NaCl was the highest concentration allowing rye growth to the three-leaf stage). Na+, K+ and Cl- ions accounted for almost half of the osmotic adjustment in Triticum df and T300, and up to 90% in rye. Salinity decreased the net photosynthesis and transpiration rates of the three cereals as compared to control plants, but induced no significant change in chlorophyll a fluorescence parameters. Water-use efficiency (WUE) increased with salinity. In the presence of 110 mmol/L NaCl, the K+/Na+ ratio decreased markedly in rye as compared to the other two cereals. Proline concentration, which increased in Triticum df and T300, could have protected membrane selectivity in favour of K+. Proline content remained low in rye, and increasing soluble sugar content did not appear to prevent competition between Na+ and K+. The salt sensitivity of rye could be due to low K+ uptake in the presence of a high NaCl concentration.  相似文献

To elucidate the osmotic adjustment characteristics of mangrove plants, inorganic ion and organic solute contents of intermediate leaves were investigated in 3-month-old Kandelia candel (L.) Druce seedlings during 45 days of NaCl treatments (0, 200, and 500 mM NaCl). The contents of Na+, Cl, total free amino acids, proline, total soluble sugars, pinitol and mannitol increased to different degree by salinity, whereas, K+ content decreased by salinity compared with control. NaCl treatment induced an increase of inorganic ion contribution while a decrease of organic solute contribution. It was concluded that accumulating a large amount of inorganic ions was used as the main osmotic adjustment mechanism under salinity treatment. However, accumulation of organic osmolytes might be considered to play much more important role in osmoregulation under severe salinity (500 mM NaCl) than under moderate salinity (200 mM NaCl), thus the damage caused by high toxic ions (Na+ and Cl) concentration in K. candel leaves could be avoided.  相似文献

The effects of increasing relative humidity on the growth and salt tolerance of two melon (Cucumis melo L.) cultivars, Revigal C-8 (salt sensitive) and Galia (salt tolerant) was investigated. One month after germination, the plants were exposed for 15 d to 0 (control) and 80 mM NaCl, under relative humidity (RH), 30 and 70 %. The growth of the whole plant, leaf, stem and root of cv. Revigal C-8 was increased with increasing RH. On the other hand, cv. Galia showed an increase in root growth with increasing RH only under the NaCl treatment. Under salinity, most of the Na+ was withheld in the stems. An increase in RH in the NaCl treatment significantly decreased Na+ and Cl concentrations in leaves of cv. Revigal C-8, while it had no effect on their concentrations in cv. Galia. In both cultivars, increasing RH under NaCl condition significantly decreased water contents in leaves and stems, and increased osmotic potential in roots. The amount of the root exudate of cv. Galia was significantly decreased with increasing RH, while it was not affected in cv. Revigal C-8. Under the NaCl treatment, cv. Galia had significantly higher leaf osmotic potential than cv. Revigal C-8 at both relative humidities and higher amount of root exudate at 30 % RH.  相似文献
