Destabilization of secondary structure in polyadenylic acid by formaldehyde   总被引:1,自引:0,他引:1

Results of circular dichroism and ultraviolet absorption measurements of poly A in neutral aqueous formaldehyde solution are presented, which show that the hydroxymethylated residues retain the ability to stack, but the stacking tendency is diminished. The midpoint of the thermal denaturation profile falls from about 38°C to about 18°C on complete hydroxymethylation. Hydroxymethylation, however, appears to have only a small effect on the apparent standard enthalpy and entropy of stacking, both of which become slightly more negative. When present at concentrations greater than about 2 g/100 ml, formaldehyde has a further and somewhat more dramatic denaturing effect on poly A. This ability is probably related to the chemical similarity of aqueous formaldehyde (methylene glycol) and ethylene glycol, which is known to be an effective denaturant for nucleic acids. In the presence of 20 g/100 ml formaldehyde, a very broad thermally induced transition of poly A is observed through the loss of ultraviolet hypochromism. Only the low-temperature part of the transition is observed; it appears to be completed only at temperatures well over 100°C where the hypochromism with respect to adenosine has largely disappeared. The transition cannot be identified with the unstacking reaction usually observed through thermal denaturation experiments in the absence of formaldehyde, although the associated structure may be related in some way. The molecular nature of the structure associated with the transition is not known.  相似文献

The structure of yeast tRNAAsp in aqueous solutions has been analyzed in the light of results obtained from Raman spectra recorded at from 5 to 82°C and compared to those of tRNAPhe. Firm evidence is given of a reversible conformation transition for tRNAAsp at 20°C. This transition is observed for the first time in the tRNA series. The low-temperature conformation appears to have a more regular ribose–phosphate backbone and a more effective G base-stacking. This conformational change, which occurs essentially in the D loop, could be connected to the existence of two (A and B) crystal forms obtained depending on crystallization conditions. The melting temperatures, which are different for each base stacking in tRNAAsp, lie in a range of about 70°C, much higher than for tRNAPhe. This fact is interpreted by a higher ratio of G-C base pairs in tRNAAsp.  相似文献

The thermoperiodic time cue is known to be one of several zeitgebers (Zt) that entrain an organism's biological rhythms. This study investigated the adult eclosion rhythm of the Indian meal moth, Plodia interpunctella, (Lepidoptera: Pyralidae) under various thermocycles in constant darkness. The insects were exposed to different thermoperiods (e.g. 25.5°C/24.5°C). Thermophase : cryophase ratios (TC) ranged from 4 h : 20 h to 20 h : 4 h. The peak in adult eclosion rhythm occurred at Zt 18.8 ± 4.4 (mean ± SD) on a given day and at Zt 0.7 ± 3.8 on the subsequent day under TC 4 h : 20 h and TC 20 h : 4 h conditions, respectively (start of temperature increase = Zt 0). Thus, longer thermophase periods delayed the peak in eclosion. The regression line obtained was then compared with that obtained under the temperature cycle of 30°C/20°C, and the two lines were found to lie parallel to each other. The peak in adult eclosion rhythm occurred earlier under the 25.5°C/24.5°C conditions than under the 30°C/20°C conditions. The results strongly suggest that the amplitude of the temperature cycle is an important factor determining the temporal position of the peak in adult eclosion. This is a significant finding among insect species.  相似文献

Abstract. The interactive effects of temperature (20 °C or 25 °C) and photoperiod (LD 12 : 12 h or LD 15 : 9 h) on diapause induction and termination are investigated in the west‐Japan type yellow‐spotted longicorn beetle, Psacothea hilaris (Pascoe) (Coleoptera: Cerambycidae). Larval diapause of P. hilaris is induced under three diapause‐inducing conditions (20 °C–SD, 20 °C–LD and 25 °C–SD), and the diapause larvae are transferred to one of four conditions (20 °C–SD, 20 °C–LD, 25 °C–SD or 25 °C–LD) for observation of pupation, which indicates termination of diapause. The intensity of diapause induced under the three conditions increases in the order 20 °C–SD < 25 °C–SD < 20 °C–LD, when assessed by the time course of pupation after the transfer. On the other hand, the effectiveness of the temperature–photoperiod combinations to terminate diapause is in the order 25 °C–SD (ineffective) < < 20 °C–LD < 25 °C–LD < 20 °C–SD. Among the temperatures (5, 10, 15 and 20 °C) examined, 15 °C is the most effective in terminating diapause under the short day; diapause in most larvae appears to have been completed in 15 days.  相似文献

Chitosan was N-permethylated by reaction with formaldehyde and sodium borohydride under controlled conditions (pH 4·0, 15°C, reaction times 12 and 8 h, respectively). The N-permethylated chitosan was reacted with methyl iodide at 35°C and N-trimethyl chitosan iodide with a quaternary nitrogen degree of 60% was obtained. The secondary structure of hemoglobin mRNA (HbmRNA has been investigated by optical rotatory dispersion (ORD), circular dichroism (CD) and ultraviolet absorbance spectrophotometry. The dependence on temperature or reaction with formaldehyde of the CD and absorbance are characteristic of a structure with substantial base pairings at 20°C. The presence of Cotton effects in one of the major ultraviolet absorption bands indicates a highly ordered secondary structure. The UV hyperchromism on thermal denaturation is consistent with a value of 58% double helical content.  相似文献

Abstract

Swertia chirayita, a critically endangered medicinal herb, is being over-harvested in the wild. Understanding seed germination is a pre-requisite to ensure species conservation. The germination of seeds collected from six microhabitats was studied at 20°C, 25°C, and 30°C, both under a 14/10 h light/dark photoperiod and in continuous darkness. Two-way ANOVA indicated that microhabitat and temperature significantly affect seed germination, germination rate, germination recovery (GR), and GR rate. Overall, the seeds collected from under canopy showed a significantly (p < 0.05) higher germination than those from open habitats, at 20°C, 25°C, and 30°C (14/10 h light/dark photoperiod). Germination was negligible in continuous darkness but after transfer to a 14/10 h light/dark photoperiod, the seeds from under canopy significantly recovered at 20°C and at 25°C (p < 0.05), and showed the highest germination percentage compared to seeds collected from tree base, stump base, shrubberies, and grassy slope. Similarly, at 30°C, seeds from under canopy recorded the highest GR percentage. In general, seed germination, mean germination rate, seed GR, and GR rate were significantly greater (p < 0.05) at 25°C. Among the microhabitats tested, variation in GR rate was significant (p < 0.05). Seeds were confirmed to be positively photoblastic.  相似文献

Abstract. Diapause adults of Plautia stali Scott maintained at 20°C under short day conditions (LD 12:12 h) were exposed to four temperatures of 5–20°C to examine the effect on diapause development which was assessed in terms of oviposition. Diapause adults kept at 20°C under short day conditions changed their body colour gradually from brown to green and started egg laying after a prolonged preoviposition period. Those transferred to either 10 or 15°C also showed colour change but did not lay eggs. Bugs exposed to 5°C underwent neither body colour change nor oviposition and died more rapidly than those kept at higher temperatures. When 30-day-old diapause adults were chilled at 5, 10 or 15°C for 30 or 60 days and returned to 20°C and long day conditions (LD 16:8 h), the preoviposition period varied primarily depending on the chilling, but not on the temperature. On the other hand, when 60day-old diapause adults chilled for 30 days were observed at 20°C and long day conditions, their preoviposition period tended to be longer as the chilling temperature was lower In this case, a temperature of 10°C appeared to intensify diapause. Therefore, the effect of chilling on diapause development varied depending on the age at which insects were chilled. When chilled bugs were transferred to short day conditions at 20°C, most females failed to lay any eggs and some turned green, then after a while, some green bugs changed to brown again. These results indicate that bugs remained sensitive to short day conditions even after a 60-day chilling at 10 or 15°C.  相似文献

We have monitored the helix-coil transition of the self-complementary d-CpCpGpG and d-GpGpCpC sequences (20mM strand concentration) at the base pairs, sugar rings, and backbone phosphates by 360-MHz proton and 145.7-MHz phosphorus nmr spectroscopy in 0.1M phosphate solution between 5 and 95°C. The guanine 1-imino Watson-Crick hydrogen-bonded protons, characteristic of the duplex state, are observed below 10°C, with solvent exchange occurring by transient opening of the tetranucleotide duplexes. The cytosine 4-amino Watson-Crick hydrogen-bonded protons resonate 1.5 ppm downfield from the exposed protons at the same position in the tetranucleotide duplexes, with slow exchange indicative of restricted rotation about the C-N bond below 15°C. The guanine 2-amino exchangeable protons in the tetranucleotide sequence exhibit very broad resonances at low temperatures and narrow average resonances above 20°C, corresponding to intermediate and fast rotation about the C-N bond, respectively. Solvent exchange is slower at the amino protons compared to the imino protons since the latter broaden out above 10°C. The well-resolved nonexchangeable base proton chemical shifts exhibit helix-coil transition midpoints between 37 and 42°C. The transition midpoints and the temperature dependence of the chemical shifts at low temperatures were utilized to differentiate between resonances located at the terminal and internal base pairs while the H-5 and H-6 doublets of individual cytosines were related by spin decoupling studies. For each tetranucleotide duplex, the cytosine H-5 resonances exhibit the largest chemical shift change associated with the helix-coil transition, a result predicted from calculations based on nearest-neighbor atomic diamagnetic anisotropy and ring current contributions for a B-DNA duplex. There is reasonable agreement between experimental and calculated chemical shift changes for the helix-coil transition at the internal base pairs but the experimental shifts exceed the calculated values at the terminal base pairs due to end-to-end aggregation at low temperatures. Since the guanine H-8 resonances of the CpCpGpG and d-CpCpGpG sequences exhibit upfield shifts of 0.6–0.8 and <0.1 ppm, respectively, on duplex formation, these RNA and DNA tetranucleotides with the same sequence must adopt different base-pair overlap geometries. The large chemical shift changes associated with duplex formation at the sugar H-1′ triplets are not detected at the other sugar protons and emphasize the contribution of the attached base at the 1′ position. The coupling sum between the H-1′ and the H-2′ and H-2″ protons equals 15–17 Hz at all four sugar rings for the d-CpCpGpG and d-GpGpCpC duplexes (25°C), consistent with a C-3′ exo sugar ring pucker for the deoxytetranucleotides in solution. The temperature dependent phosphate chemical shifts monitor changes in the ω,ω′ angles about the O-P backbone bonds, in contrast to the base-pair proton chemical shifts, which monitor stacking interactions.  相似文献

Formaldehyde is usually added to brown algae to avoid microbial growth during storage and to fix polyphenols in the algae before alginate extraction. Since formaldehyde is toxic, allergenic and possibly carcinogenic, dry salting of Ascophyllum nodosum was tested as an alternative. The seaweeds, harvested at locations with a salinity of about 30‰ from late autumn to early spring, were stored at 22±2 °C under compost-like conditions. Untreated samples of seaweed lost their quality as a raw material for alginate production within 14 days. Salted (20–22%) as well as formaldehyde treated seaweed was preserved for at least 46 days. Due to the reduced water activity and oxygen saturation in the dry salted seaweed, microbial growth and brown colouring reactions were suppressed. Economic factors must also be taken into account before large-scale applications are considered. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

The influence of environmental factors on the duration of diapause in Plodia interpunctella larvae reared in short photoperiods at 20 or 25° C was examined, Diapause terminated most rapidly in long photoperiods at high temperatures. Pupation was more delayed, and mortality was higher, in darkness than in the presence of light. At 20° C, LD 16: 8 hastened diapause termination only slightly in unchilled samples. Chilling for 10 weeks at 10° C greatly reduced the duration of diapause at 20 or 25° C in constant darkness, and rendered LD 16:8 effective in terminating diapause at 20° C. In addition, the quite short duration of diapause under LD 16:8 at 25° C was further shortened by holding for 6–10 weeks at 10° C or below, or by holding in an outbuilding during winter. Holding diapausing larvae at 15 or 20° C proved less effective. Temperature rises from 20 to 25 or 30° C proved effective in terminating diapause. In one stock, the temperature at which diapause was induced influenced its subsequent duration. Lighting conditions during induction had less influence on duration than had temperature, and no difference occurred between pupation times of larvae reared at different population densities, Under all conditions tested, diapause lasted longer in a recently collected field stock than in a laboratory stock.  相似文献

Abstract. The effects of thermoperiods on diapause induction in continuous darkness or under a 12 : 12 h LD photoperiod were investigated in the cabbage beetle, Colaphellus bowringi Baly, a typical short‐day species. The diapause response curves both at different constant temperatures and at the thermocycle of format CT x: (24 ? x) h (16 : 28 °C) under continuously dark rearing conditions showed that the incidence of diapause depended mainly on whether or not the mean temperature was ≤20 °C or >20 °C. If the mean temperature was ≤20 °C, all individuals entered diapause; if >20 °C, the incidence of diapause declined gradually with increasing mean temperatures. The thermocycle (CT 12 : 12 h) with a series of different cryophases (8–22 °C) and thermophases (24–32 °C) under continuous darkness demonstrated a cryophase response threshold temperature of approximately 19 °C and a thermophase response threshold temperature of approximately 31 °C. Thermoperiodic amplitude (temperature difference between cryophase and thermophase) was shown to have a significant influence on diapause induction at the mean temperatures of 22, 23 and 24 °C, but not at ≥25 °C. Thermoperiodic responses under LD 12 : 12 h clearly showed that the incidence of diapause was influenced strongly by the photophase temperature. The thermoperiod under LD 12 : 12 h induced a much lower incidence of diapause than the thermoperiod with the same temperature in continuous darkness. The ecological significance of thermoperiodic induction of diapause in this species is discussed.  相似文献

An algorithm is developed that enables the routine determination of backbone conformations of nucleic acids. All atomic positions including hydrogen are specified in accord with experimental bond lengths and angles but with theoretically determined conformational angles. For two Watson-Crick base pairs at a separation of 3.38 Å, and perpendicular to a common helical axis, minimum energy configurations are found for all 10 combinations at helical angles of α ~ 36°–38°, corresponding to the B-DNA structure with C(2′)-endo sugar puckers. Backbone configurations exist only within the range 35.5° ? α ? 42°, which suggests the origin of the 10-fold helix. Calculated stacking energies for the B-DNA structure increases for each of the clustered groups of base pairs: G·C with G·C, G·C with A·T, and A·T with A·T, and they are in approximate agreement with experimental observations. The counter-clockwise helix is examined, and physically meaningful structures are found only when the helical axes of successive base pairs are disjointed.  相似文献

Feeding behavior of Plutella xylostella under optional to non-optional conditions was studied at 10°C, 15°C, 20°C, and 25°C on Indian mustard, Brassica juncea. The study reveals that the variety Pusa Bahar was significantly less preferred by the larvae as compared with Pusa Bold and Varuna under optional to non-optional conditions. Larvae of P. xylostella consumed more food at 25°C than 20°C, 15°C and 10°C. Larval survival was found to be highest on cabbage (control) as compared with Indian mustard and was found to vary with host plants and temperature. The larval survival decreased to 11.29% on Pusa Bahar at 10°C. Increasing the temperature from 10°C to 20°C, larval mortality resulted more on Varuna than Pusa Bahar and Pusa Bold. Developmental period was prolonged on Pusa Bold at 10°C while it was shortest on cabbage at 25°C. A total of 536.47 degree days were required to complete the development by immature stages on Varuna at 25°C and 421.64 degree days on cabbage.  相似文献

The stacking and hydrogen bonding energies between bases in the B form of DNA were calculated by a perturbation method using the wave functions by the CNDO and the P-P-P methods. The exchange energies were calculated by using the corresponding orbitals. The magnitudes of the sums of the average stacking and hydrogen bonding energies per base pair of double-stranded DNA-like polymers are in good parallel with the melting temperatures of the polymers. The polymers containing I-C pairs are exceptions to this relation. Intrastrand stacking bases have the potential minimum at the distances of 2·8–3·7 Å. The minimum of stacking energy of double-stranded polymer for rotation of base pair around the helix axis exists near 36°. The deviation of the potential minimum from 36° seems to parallel the feature of the X-ray diffraction pattern of the polymer.  相似文献

Abstract Sericinus montelus overwinters as diapausing pupae. In the present study, the effects of photoperiod and temperature on diapause induction and termination of diapause are investigated. The results obtained demonstrate that high temperature can reverse the effect of short day‐lengths on diapause induction. Under an LD 12 : 12 h photoperiod, all pupae enter diapause at 15, 20 and 25 °C, whereas all pupae develop without diapause at 35 °C. No pupae enter diapause under an LD 14 : 10 h photoperiod when the temperature is above 20 °C. Photoperiodic response curves obtained at 25 and 30 °C indicate that S. montelus is a long‐day species and the critical day‐length is approximately 13 h at 25 °C. At 25 °C, the duration of diapause is shortest when the diapausing pupae are maintained under an LD 16 : 8 h photoperiod and increases under LD 14 : 10 h and LD 12 : 12 h photoperiods. Under an LD 16 : 8 h photoperiod, the duration of diapause is shortest when the diapausing pupae are maintained at 25 °C, followed by 20 and 30 °C, and then at 15 °C. These results suggest that a moderate temperature favours diapause development under a diapause‐averting photoperiod in this species. The duration of diapause induced by an LD 12 : 12 h photoperiod is significantly longer at 25 °C than those at 15, 20 and 30 °C, and is shortest at 15 °C. At 25 °C, the duration of diapause induced by LD 6 : 18, LD 12 : 12 and LD 13 : 11 h photoperiods is similar and longer than 90 days. Thus, the diapause‐inducing conditions may affect diapause intensity and a photoperiod close to the critical day‐length has significant influence on diapause intensity in S. montelus.  相似文献

Ooencyrtus nezarae Ishii (Hymenoptera: Encyrtidae) was reared on eggs ofRiptortus clavatus (Thunberg) (Heteroptera: Alydidae) at various temperatures under long-day (L16:D8) or short-day (L10:D14) conditions. There was no diapause during egg, larval or pupal stages under any set of conditions examined. However, at 15°C under short-day conditions, vitellogenesis was arrested in all adult females and they entered diapause. At 15°C under long-day conditions, or at 20°C under short-day conditions, some adult females entered diapause. Under the latter set of conditions, the adult females laid eggs but they laid fewer eggs than under long-day conditions, Even at 25°C, under short-day conditions, adult females laid fewer eggs than under long-day conditions, and this low rate of oviposition was attributed to the retarded development of ovaries. Diapause adults reared at 15°C were more resistant to low temperature than nondiapause adults reared at 25°C.  相似文献

The influence of different temperatures 10, 15, 20, and 25°C on the food consumption, growth, moulting rate, and respiration of Palaemon pacificus (Stimpson) from Langebaan Lagoon, west coast of South Africa, was studied under laboratory conditions. At 10°C mortality was high so that food consumption and moulting rate could not be determined as these were very low. At higher temperatures, food consumption was found to be temperature dependent, the rate at 25°C being twice that at 15°C. Growth rate was most favourable at 25°C. At 28°C growth rate was lower than at 20°C but higher than at 15°C. The intermoult period was 17 days at 15°C, and 11 and 10 days at 20, and 25°C, respectively. It seems that from an energetic point of view, 25°C is the most favourable temperature for P. pacificus. Several indices of growth efficiency at different temperatures are presented. The appearance of this prawn in South African west coast localities such as Langebaan during the summer and its disappearance during winter, can be explained by its temperature preferences. The possibility that thermal pollution from a nuclear power station may be beneficial to this prawn, is discussed.  相似文献

The effects of temperature on the development and growth of hemp (Cannabis sativa L.) have never been quantified. Therefore, to establish the effect of temperature on leaf appearance and canopy establishment of fibre hemp under controlled and field conditions, plants were grown in growth chambers at 11 regimes with average temperatures between 10°C and 28°C, and three cultivars were sown in the field in March, April and May in 1990, 1991 and 1992. In the field, thermal time (base 0°C) between sowing and emergence ranged from 68°Cd to 109.5°Cd (average 88.3°Cd). Rates of leaf appearance and stem elongation increased linearly with temperature between 10°C and 28°C. The base temperature for leaf appearance was 5.7°C from the growth chamber experiments and 1°C from the field experiments. In the field, the base temperature for the relationship between light interception by the canopy and thermal time was 2.5°C, and thermal time, calculated at the appropriate base temperature, accounted for about 98% of the variance in the number of leaves and for 98.6% of the variance in the proportion of light intercepted by the canopy. Days from emergence accounted for less of the variance in both parameters than thermal time. Interception of 90% of light was attained on average at 465°Cd (base 0°C) after emergence. It is concluded that thermal time is a simple and accurate tool to describe leaf appearance and light interception in fibre hemp.  相似文献

The aim of this study is to evaluate the physiological costs and the consequences on fitness of cold exposure on the parasitoid Aphidius ervi (Hymenoptera: Aphidiinae) under non-lethal conditions. We exposed 1-day-old mummies to different treatments: control at 20°C, 7°C constant, 7°C fluctuating (7°C for 22 h and 20°C for 2 h). Two performances of fitness were particularly affected after two weeks at 7°C constant: female longevity and the sex ratio of the progeny was male biased. In contrast, egg load at emergence, lifetime fecundity, frequency of asymmetric individuals in both sexes, and mating success were not significantly affected under all treatments. The effects of these treatments on trends such as utilisation of fat reserves and fertility of males and females are discussed.  相似文献
