High strength slaughterhouse wastewater was treated in four 42 l anaerobic sequencing batch reactors (ASBRs) operated at 30 degrees C, 25 degrees C and 20 degrees C. The wastewater contained between 30% and 53% of its chemical oxygen demand (COD) as suspended solids (SS). The ASBRs could easily support volumetric organic loading rates (OLRs) of 4.93, 2.94 and 2.75 kg/m3/d (biomass OLRs of 0.44, 0.42 and 0.14 g/g volatile SS (VSS)/d) at 30 degrees C, 25 degrees C, and 20 degrees C, respectively. At all operating temperatures, the total COD (TCOD) and soluble COD (SCOD) were reduced by over 92%, while average SS removal varied between 80% and 96%. Over the experimental period, 90.8%, 88.7% and 84.2% of the COD removed was transformed into methane at 30 degrees C, 25 degrees C and 20 degrees C, respectively. The decrease in the conversion of the COD removed into methane as operating temperature was lowered, may be partly explained by a lower degradation of influent SS as temperature was reduced. The reactors showed a high average methanogenic activity of 0.37, 0.34 and 0.12 g CH4-COD/gVSS/d (22.4, 12.7 and 11.8 l/d) at 30 degrees C, 25 degrees C and 20 degrees C, respectively. The average methane content in the biogas increased from 74.7% to 78.2% as temperature was lowered from 30 degrees C to 20 degrees C.  相似文献

The potential of clover (Trifolium repens) and ryegrass (Lolium perenne) mixtures as raw materials for ethanol production was investigated. Wet oxidation, at 175, 185 or 195 degrees C during 10min at two different oxygen pressures and with either addition or no addition of sodium carbonate, was evaluated as pretreatment method for clover-ryegrass mixtures. The enzymatic hydrolysis of cellulose was significantly improved after pretreatment. The highest conversion efficiency, 93.6%, was achieved for the sample pretreated at 195 degrees C, 10min, 1.2MPa and no addition of Na(2)CO(3). For that sample, the overall glucose yield after pretreatment and hydrolysis was 75.5%. No inhibition of cellulose enzymatic conversion by the filtrates was observed. The simultaneous saccharification and fermentation of the pretreated material yielded cellulose conversions of 87.5 and 86.6%, respectively, with Saccharomyces cerevisiae and the filamentous fungus Mucor indicus, and revealed that no addition of nutrients is needed for the fermentation of clover-ryegrass hydrolysates.  相似文献

Seven lignocellulosic materials: corn stover, napier grass, wood grass, newspaper, white fir and wheat straw from two different crops; two pure cellulosics: Solka Floc BW200 and Whatman No. 5 filter paper; and glucose, propionic and acetic acids were subjected to long-term batch methane fermentation. Ninety per cent of the original COD was recovered as methane gas from the two pure cellulosics and glucose. For the lignocellulosics, depending on the material, variations from over 80% conversion efficiency to methane for corn stover to less than 10% for white fir were observed. Generally, herbaceous materials were degraded faster and more extensively than woody biomass. A first-order rate model described well the methane fermentation process for the lignocellulosics tested, but was a poor model for the soluble substrates. It was not possible to predict either the biodegradability or the rate of methane fermentation with a reasonable degree of accuracy based solely on the lignin content of the lignocellulosic materials.  相似文献

Biomass waste, including municipal solid waste (MSW), contains lignocellulosic-containing fiber components that are not readily available as substrates for anaerobic digestion due to the physical shielding of cellulose imparted by the nondigestible lignin. Consequently, a substantial portion of the potentially available carbon is not converted to methane and the incompletely digested residues from anaerobic digestion generally require additional processing prior to their return to the environment. We investigated and developed steam pressure disruption as a treatment step to render lignocellulosic-rich biomass more digestible and as a means for increasing methane energy recovery. The rapid depressurization after steam heating (240 degrees C, 5 min.) of the nondigested residues following a 30-day primary digestion of MSW caused a visible disruption of fibers and release of soluble organic components. The disrupted material, after reinoculation, provided a rapid burst in methane production at rates double those observed in the initial digestion. This secondary digestion proceeded without a lag phase in gas production, provided approximately 40% additional methane yields, and was accompanied by a approximately 40% increase in volatile solids reduction. The secondary digestate was found to be enriched in lignin and significantly depleted in cellulose and hemi-cellulose components when compared to primary digestate. Thus, steam pressure disruption treatment rendered lignocellulosic substrates readily accessible to anaerobic digestion bacteria and improved both the kinetics of biogas production and the overall methane yield from MSW. Steam pressure disruption is central to a new anaerobic digestion process approach including sequential digestion stages and integrated energy recovery, to improve process yields, provide cogenerated energy for process needs, and to provide effective reuse and recycling of waste biomass materials.  相似文献

Anaerobic co-digestion is a well established process for treating many types of organic wastes, both solid and liquid. In this study we have investigated, on a laboratory scale, the anaerobic co-digestion of olive mill wastewater (OMW) with olive mill solid waste (OMSW) using semi-continuous, feeding, tubular digesters operated at mesophilic temperatures. Each digester was fed with an influent, composed of OMW and OMSW, at an organic loading rate (OLR) varying between 0.67 and 6.67 g COD/l/d. The hydraulic retention times (HRT) were 12, 24 and 36 days. The TCOD concentrations of OMW used as the main substrate were 24, 56 and 80 g COD/l; the amount of the dry OMSW used as a co-substrate was fixed to approximately 56 g/l of OMW. The results indicated that the best methane production was about 0.95 l/l/day obtained at an OLR = 4.67 g COD/l/d, corresponding to influent TCOD = 56 g COD/l at an HRT = 12d. In contrast, the maximum TCOD removal efficiency (89%) was achieved at an OLR = 0.67 g COD/l/d, corresponding to influent TCOD = 24 g COD/l at an HRT = 36 d. Moreover, the inhibition of biogas production was observed at the highest OLR studied.  相似文献

This work studied the hydrolysis kinetics and the solubilization of waste activated sludge under a medium range temperature (50-90 degrees C) and pH in the alkaline region (8-11), as a pretreatment stage for anaerobic digestion. The hydrolysis rate for the solubilization of volatile suspended solids (VSS) followed a first-order rate. A linear polynomial hydrolysis model was derived from the experimental results leading to a satisfactory correlation between the hydrolysis rate coefficient, pH, and temperature. At pH 11 and a temperature of 90 degrees C the concentration of the VSS was 6.82%, the VSS reduction reached 45% within ten hours and at the same time the soluble COD was 70.000 mg/l and the total efficiency for methane production 0.28 l of CH4 per g of VSS loading.  相似文献

A study of anaerobic digestion of piggery wastewater was carried out in a laboratory-scale sludge bed reactor as a secondary treatment. The effect of organic volumetric loading rates (BV) in the range of 1.0-8.1 g TCOD/ld on the process performance was evaluated. The best results were obtained at BV equal to or lower than 4 g TCOD/ld. At higher BV values, the removal efficiency of the process decreased suddenly. A linear relationship was found between the effluent SCOD and the TVFA/alkalinity ratio (P). A relationship was found among the different operational variables (BV , removal efficiency, effluent soluble COD, soluble COD removal rate (R), retention factor (phi), specific microbial growth rate (mu), methane production rate per volume of reactor and per volume of waste treated--QM and qM, respectively) and the corresponding regression equations were obtained. An increase of BV determined a decrease of removal efficiency, phi and qM and an increase of effluent soluble COD, mu, R and QM. The value of the maximum specific microbial growth rate (muM) determined through the equation that correlated BV and mu was found to be 0.19 d(-1). This value was of the same magnitude as those reported in other works of anaerobic digestion of piggery waste.  相似文献

The effects of temperature (35 and 55 degrees C), influent volatile solids (VS) concentration (S(0) = 43, 64, 82, 100, and 128 kg VS/m(3)) and hydraulic retention time (HRT = 4, 5, 8, 10, 15, and 25 days) on methane (CH(4)) production from cattle waste were evaluated using 3-dm(3) laboratoryscale fermentors. The highest CH(4) production rate achieved was 6.11 m(3) CH(4) m(-3) fermentor day(-1) at 55 degrees C, four days HRT, and S(0) = 100 kg VS/m(3). Batch fermentations showed an ultimate CH(4) yield (B(0)) of 0.42 m(3) CH(4)/kg VS fed. The maximum loading rates for unstressed fermentation were 7 kg VS m(-3) day(-1) at 35 degrees C and 20 kg VS m(-3) day(-1) at 55 degrees C. The kinetic parameter (K, an increasing K indicates inhibition of fermentation) increased exponentially as S(0) increased, and was described by: K = 0.8 + 0.0016 e(0.06S(0) ). Temperature had no significant effect on K for S(0) between 40 and 100 kg VS/m(3). The above equation predicted published K values for cattle waste within a mean standard error of 7%.  相似文献

A thermophilic anaerobic digestion (TAnD, 55 degrees C) system was adopted to hydrolyze the kitchen waste for 3 days, which was then fermented for a hydraulic retention time (HRT) of 10 days. The TAnD system performed much better than a similar system without thermal pre-treatment. A bubble respirometer was employed to study the effects of thermal pre-treatment, which showed that pre-treatment at 60 degrees C yielded the highest Total COD (TCOD) removal efficiency (79.2%) after 300h reaction. Respirometer results also indicated that oil and grease (O and G) began to inhibit the TAnD system at a concentration of approximately 1000mg/L and the gas production was inhibited by 50% at a concentration of approximately 7500mg/L of sodium.  相似文献

The wet oxidation process of wheat straw has been studied as a pretreatment method to attain our main goal: To break down cellulose to glucose enzymatic, and secondly, to dissolve hemicellulose (e.g., for fermentation) without producing microbial inhibitors. Wet oxidation combined with base addition readily oxidizes lignin from wheat straw facilitating the polysaccharides for enzymatic hydrolysis. By using a specially constructed autoclave system, the wet oxidation process was optimized with respect to both reaction time and temperature. The best conditions (20 g/L straw, 170 degrees C, 5 to 10 min) gave about 85% w/w yield of converting cellulose to glucose. The process water, containing dissolved hemicellulose and carboxylic acids, has proven to be a direct nutrient source for the fungus Aspergillus niger producing exo-beta-xylosidase. Furfural and hydroxymethyl-furfural, known inhibitors of microbial growth when other pretreatment systems have been applied, were not observed following the wet oxidation treatment. The biological transformation of lignocellulose of Achras zapota by white rot fungi, Phanerochaete chrysosporium, in solid state fermentation (SSF) was studied for 28 days. The kinetic transformation of lignocellulose was monitored through the determination of acid soluble and acid insoluble lignin content, total organic carbon (TOC) and chemical oxygen demand (COD). The lignolytic enzymes, lignin peroxidase (LiP) and manganese peroxidase (MnP) were quantified on weekly intervals. The degradation of lignin and other structural moieties of A. zapota lignocellulose were confirmed by high performance liquid chromatography (HPLC), Fourier transform infrared spectroscopy (FTIR) and scanning electron microscopy (SEM). The degradation of lignin was increased after 7 days of fermentation with the release of water soluble and fermentable products. The LiP and MnP activities were increased in the first week of SSF and lignin degradation was also set to increase. This was accompanied with increase in COD by 94.6% and TOC by 80% and lignin content was decreased by 76%. The maximum activities of the enzymes LiP and MnP in extracellular fluid of SSF under nitrogen limitation, at pH 5.0, at temperature 37 degrees C and at 60% humidity were 2100 U/L and 1200 U/L.  相似文献

The effect of three methods (mechanical, chemical, and thermal pretreatment) were tested to improve methane production and anaerobic biodegradability of swine wastes. The first experiment was designed to determine the biodegradability enhancement through the separation of liquid and solid matrix by using a 0.25mm pore size screen (mechanical pretreatment). The second approach was the treatment of swine waste by the addition of a flocculant agent and strong chemicals such as acid (HCl) and alkali (NaOH). The third pretreatment studied was thermal application (170 degrees C provided by vapor). The soluble COD was increased by 57% and 32% during the pretreatment period with alkali and thermal application, respectively. In addition, these two pretreatments gave the highest enhancement on methane production with regard to the untreated sample. Meanwhile, the addition of a flocculant improved the methane production of the liquid fraction but not the solid one. On the other hand, mechanical pretreatment did not show any important enhancement. Biodegradability percentage followed the same trend as methane productivity.  相似文献

Starch is one of the most abundant resources on earth and is suited to serve as a cost-effective feedstock for biological hydrogen production. However, producing hydrogen from direct fermentation of starch is usually inefficient, as the starch hydrolysis is often the rate-limiting step. Therefore, in the present work, enzymatic starch hydrolysis was conducted to enhance the feasibility of using starch feedstock for H2 production. The amylase (with a molecular weight of ca. 112 kDa) used for starch hydrolysis was produced from a recombinant E. coli harboring an amylase gene originating from Caldimonas taiwanensis On1. Using statistical experimental design, the optimal pH and temperature for starch hydrolysis with the recombinant amylase was pH 6.86 and 52.4 degrees C, respectively, at an initial starch concentration of 7 g/L. The hydrolyzed products contained mainly glucose, maltotriose, and maltotetrose, while a tiny amount of maltose was also detected. The enzymatically hydrolyzed products of soluble starch and cassava starch were used as the substrate for dark hydrogen fermentation using Clostridium butyricum CGS2 and Clostridium pasteurianum CH4. The highest H2 production rate (vH2) and yield (YH2) of C. butyricum CGS2 was 124.0 mL/h/L and 6.32 mmol H2/g COD, respectively, both obtained with the hydrolysate of cassava starch. The best H2 production rate (63.0 mL/h/L) of C. pasteurianum CH4 occurred when using hydrolyzed cassava starch as the substrate, whereas the highest yield (9.95 mmol H2/g COD) was obtained with the hydrolyzed soluble starch.  相似文献

Thermophilic (55 degrees C) anaerobic enrichment cultures were incubated with [C-lignin]lignocellulose, [C-polysaccharide]lignocellulose, and kraft [C]lignin prepared from slash pine, Pinus elliottii, and C-labeled preparations of synthetic lignin and purified cellulose. Significant but low percentages (2 to 4%) of synthetic and natural pine lignin were recovered as labeled methane and carbon dioxide during 60-day incubations, whereas much greater percentages (13 to 23%) of kraft lignin were recovered as gaseous end products. Percentages of label recovered from lignin-labeled substrates as dissolved degradation products were approximately equal to percentages recovered as gaseous end products. High-pressure liquid chromatographic analyses of CuO oxidation products of sound and degraded pine lignin indicated that no substantial chemical modifications of the remaining lignin polymer, such as demethoxylation and dearomatization, occurred during biodegradation. The polysaccharide components of pine lignocellulose and purified cellulose were relatively rapidly mineralized to methane and carbon dioxide; 31 to 37% of the pine polysaccharides and 56 to 63% of the purified cellulose were recovered as labeled gaseous end products. An additional 10 to 20% of the polysaccharide substrates was recovered as dissolved degradation products. Overall, these results indicate that elevated temperatures can greatly enhance rates of anaerobic degradation of lignin and lignified substrates to methane and low-molecular-weight aromatic compounds.  相似文献

采用Hungate厌氧培养技术分别从厌氧污泥、好氧污泥及河底泥中分离出12株厌氧产氢细菌,并对其中的Enterococcus sp.LG1(注册号:EU258743)进行了研究.结果表明,该株细菌为专性厌氧菌,经革兰氏染色结果为阴性.通过16S rDNA碱基测序和比对证实,该菌株是目前尚未报道过的1个新菌种,初步确定其细菌学上的分类地位.同时,以灭菌预处理的污泥为底物培养基,对该菌的产氢能力及污泥发酵过程中底物性质变化(SCOD、可溶性蛋白质、总糖和pH值等)进行了探讨.实验结果显示,产氢茵Enterococcus sp.LG1的发酵过程中只有H2和CO2产生,无CH4产生.产气量最高为36.48 mL/g TCOD,氢气含量高达73.5%,为已报道文献中以污泥为底物发酵制氢中之最高.根据污泥发酵产物分析得知,该菌的发酵类行为典型的丁酸型发酵.  相似文献

The effects of pretreating wheat straw with gamma-ray irradiation, ammonium hydroxide, and sodium hydroxide on methane yield, fermentation rate constant, and loss of feedstock constituents were evaluated using laboratory-scale batch fermentors. Results showed that methane yield increased as pretreatment alkali concentration increased, with the highest yield being 37% over untreated straw for the pretreatment consisting of sodium hydroxide dosage of 34 g OH(-)/kg volatile solids, at 90 degrees C for 1 h. Gamma-ray irradiation had no significant effect on methane yield. Alkaline pretreatment temperatures above 100 degrees C caused a decrease in methane yield. After more than 100 days of fermentation, all of the hemi-cellulose and more than 80% of the cellulose were degraded. The loss in cellulose and hemicellulose accounted for 100% of the volatile solids lost. No consistent effect of pretreatments on batch fermentation rates was noted. Semicontinuous fermentations of straw-manure mixtures confirmed the relative effectiveness of sodium and ammonium-hydroxide pretreatments.  相似文献

The effect of temperature and organic loading rate on the rate of methane production from acidic petrochemical wastewater without neutralization was investigated by continuously feeding an anaerobic upflow fixed-film reactor. The temperatures selected for the studies were 25, 37, 45 and 55 degrees C. Organic loading rate (OLR) for each temperature was varied from 3.6 to 21.7 kg COD m(-3) d(-1). Best performance with respect to COD and BOD reduction, total gas production and methane yield was obtained with the reactor operating at 37 degrees C. OLR could be increased to a maximum of 21.7 kg COD m(-3) d(-1) with 90-95% COD and BOD reduction and methane yield of 0.450 m3 kg(-1) COD d(-1) added. The reactor operating at 55 degrees C gave the highest methane yield of 0.666 m3 kg(-1) COD d(-1) at an OLR of 6 kg COD m(-3) d(-1). This decreased to 0.110 m3 kg(-1) COD d(-1) when the OLR was increased to 18.1 kg COD m(-3) d(-1). The reactor operating at 45 degrees C gave a maximum methane yield of 0.416 m3 kg(-1) COD d(-1) added at an OLR of 6 kg COD m(-3) d(-1). On further increasing the OLR to 9 kg COD m(-3) d(-1), COD reduction was 89%, however, methane yield decreased to 0.333 m3 kg(-1) COD d(-1) added. The highest methane yield of 0.333 m3 kg(-1) COD d(-1) added at an OLR of 6 kg COD m(-3) d(-1) was obtained with reactors operating at 25 degrees C. These studies indicate potential rates of methane production from acidic petrochemical wastewater under different temperatures. This provides a guideline for various kinetic analyses and economic evaluation of the potential feasibility of fermenting acidic wastewater to methane.  相似文献

Dried grass silage (GS) was pre-treated at different NaOH loading rates (1%, 2.5%, 5% and 7.5% by volatile solids (VS) mass in grass silage) and temperatures (20 °C, 60 °C, 100 °C and 150 °C) to determine effects on its bio-degradability in terms of the hydrolysis yield and degradation of ligno-cellulosic materials for biogas production. At 100 °C and the four NaOH loadings, up to 45% of the total COD was solubilised and up to 65.6%, 36.1% and 21.2% of lignin, hemicellulose and cellulose were removed, respectively; biological methane production potentials obtained were 359.5, 401.8, 449.5 and 452.5 ml CH?/g VS added, respectively, being improved by 10-38.9% in comparison with untreated GS. VS removals following anaerobic digestion were 67.6%, 76.9%, 85.3%, 95.2% and 96.7% for untreated GS and GS treated at the four NaOH loadings, respectively. 100 °C and the NaOH loading rate of 5% is recommended as a proper GS pre-treatment condition.  相似文献

In this study the comparative destruction of municipal biosolids using thermal hydrolysis (140 or 165 °C) and wet oxidation (220 °C) was followed by biological degradation via mesophilic anaerobic digestion (36 °C). Wet oxidation (WO) destroyed more than 93% of the VSS, while thermal hydrolysis (TH) at 140 and 165 °C destroyed 9% and 22%, respectively. Combined TH and anaerobic digestion resulted in approximately 50% VSS destruction. The ultimate methane potential of the combined fractions from the thermal hydrolysis at 140 and 165 °C improved by 12-13% relative to the untreated control sample. Methane production from the WO material was 53% of the control yield and wholly attributable to soluble organic carbon in the liquid fraction, indicating that the WO destroyed all putrescible carbon from the solids fraction. Point sampling during the BMP assay revealed that methanogenic development, not solids hydrolysis, was the kinetic barrier during anaerobic digestion in this study.  相似文献

Under northern climatic conditions, a temporary decrease in the temperature of anaerobic reactors treating swine manure is likely to happen at the farm. The objective of this study was to evaluate the impact of temperature fluctuations, between 10 and 20 degrees C, on the stability and performance of psychrophilic anaerobic sequencing batch reactors (ASBRs) treating swine manure. Methane yield decreased from 0.266+/-0.014 l/g of total chemical oxygen demand (TCOD) fed to the ASBRs at 20 degrees C to 0.218+/-0.022 and 0.080+/-0.002 l/g TCOD (fed) at 15 and 10 degrees C, respectively. Soluble chemical oxygen demand (SCOD) reduction decreased from 94.2+/-1.1% at 20 degrees C to 78.8+/-3.0% at 15 degrees C and 60.4+/-6.4% at 10 degrees C. Total COD removal also tended to decrease as temperature was lowered, but difference between operating temperatures was not as pronounced. A lower methanogenic activity in the ASBRs operated at 10 degrees C probably favoured quiescent conditions during the settling period, thereby increasing physical removal of the TCOD through sedimentation of the solids with the biomass. When the operating temperature was increased back to 15 and 20 degrees C, methane yield and SCOD reduction improved, but reactor performance remained significantly (P<0.05) lower than that achieved before the cycles at 10 degrees C. Results from this experiment nevertheless suggested that fluctuation in the operating temperature of psychrophilic ASBRs should only have temporary effects on the performance and stability of the process.  相似文献
