Russian knapweed (Rhaponticum repens (L.) Hidalgo) is an herbaceous perennial weed that was introduced and has become invasive in the United States, particularly in the semi-arid west. It is characterized by its extensive root system, low seed production, and persistence. The weed has caused serious reductions in yields and crop value and may significantly devalue the land itself. Conventional control strategies have been inadequate because of the size of infestations and economic and environmental costs of control. Biological control has been a sought-after potential solution to this weed problem. In the summer of 2002, diseased R. repens plants were collected near Cankiri, Turkey, and the facultative saprophytic fungus Boeremia exigua isolate FDWSRU 02-059 was isolated from diseased plants. Bayesian analysis of the actin, beta-tubulin, calmodulin, elongation factor, and ITS genes, of 66 isolates, representing the ten species of Boeremia and the 11 varieties of B. exigua, including FDWSRU 02-059, showed that the isolate is a unique genetic entity and was named B. exigua var. rhapontica Berner, Woudenberg & Tunali, var. nov. MycoBank MB809363. Disease incidence and severity data from host-range determination tests conducted at 25 °C, the optimum temperature for growth and sporulation of B. ex. rhapontica, with adequate dew periods, were combined with a genetic distance matrix based on ITS sequences of 66 plant species related to R. repens. The combined disease and genetic data were analyzed by mixed model equations to produce best linear unbiased predictors (BLUPs), standard errors, and P > |t| values, in t-tests against zero, for disease incidence and severity for each species. BLUPs of disease incidence were significantly different from zero only for three Rhaponticum spp. while BLUPs of disease severity rankings were significantly different from zero only for R. repens, Rhaponticum carthamoides, Rhaponticum uniflorum, and Leuzea berardioides. Best linear unbiased predictors for differences in above-ground dry weights between control and inoculated plants of a subset of the species evaluated were not significant. However, above-ground damage by B. ex. rhapontica to R. repens was nearly twice that for any other species, except Rhaponticum species.  相似文献

Several studies have argued that under field conditions plant–soil feedback may be related to the local density of a plant species, but plant–soil feedback is often studied by comparing conspecific and heterospecific soils or by using mixed soil samples collected from different locations and plant densities. We examined whether the growth of the early successional species Jacobaea vulgaris in soil collected from the field is related to the local variation in plant density of this species. In a grassland restoration site, we selected eight 8 m × 8 m plots, four with high and four with low densities of J. vulgaris plants. In 16 subplots in each plot we recorded the density and size of J. vulgaris, and characteristics of the vegetation and the soil chemistry. Soil collected from each subplot was used in a greenhouse pot-experiment to study the growth of J. vulgaris, both in pure field soil and in sterile soil inoculated with a small part of field soil.In the field, flowering J. vulgaris plants were taller, the percentage of rosette plants was higher and seed density was larger in High- than in Low-density plots. In the pot experiment, J. vulgaris had a negative plant–soil feedback, but biomass was also lower in soil collected from High- than from Low-density plots, although only when growing in inoculated soil. Regression analyses showed that J. vulgaris biomass of plants growing in pure soil was related to soil nutrients, but also to J. vulgaris density in the field.We conclude that in the field there is local variation in the negative plant–soil feedback of J. vulgaris and that this variation can be explained by the local density of J. vulgaris, but also by other factors such as nutrient availability.  相似文献

Upland heathland is an internationally important habitat but a large area in the UK has been degraded to acid grassland by intensive livestock grazing. Re-establishment of dwarf shrubs, particularly Calluna vulgaris (L.) Hull, is a key objective for restoring heathland on these sites. A replicated plot-scale experiment was set up to examine effects of disturbance and seed addition on C. vulgaris establishment in a Nardus stricta L. grassland under three grazing regimes: sheep only (1.5 ewes ha?1 for 10 months per year); cattle only (0.5 heifers ha?1 in summer only); and, the cattle regime combined with sheep (1.0 ewes ha?1 for 10 months per year). Early results of the experiment have been reported previously but it was not known if these results were an indication of the longer-term restoration success. Here we evaluate the success of the restoration methods (disturbance, seeding treatments and grazing regime) eight years after the treatments began. In seeded plots, young C. vulgaris plants had greatest above-ground height, dry weight and shoot length if grazing was excluded or the cattle-only regime was applied. C. vulgaris cover was greatest, and increased most, in plots that had been disturbed, seeded and ungrazed or subjected to the cattle-only regime. The vegetation in these plots also became more similar to reference sites with 50% or more cover of C. vulgaris. The invasive Juncus effusus L. was more frequent in disturbed and grazed plots but less frequent in plots with C. vulgaris established from added seed. Previous results that showed the benefits of disturbance and seeding treatments were still valid but changes in the vegetation composition were still occurring and longer-term studies will be needed to determine when grazing regimes including sheep might be reintroduced.  相似文献

Canada thistle (Cirsium arvense) is one of the worst weeds in temperate areas of the world. A rust fungus, Puccinia punctiformis, was first proposed as a biological control agent for C. arvense in 1893. The rust causes systemic disease which ultimately kills C. arvense plants. In 2013 it was demonstrated in four countries, that inoculation of C. arvense rosettes in the fall with ground telia-bearing leaves can initiate epidemics of systemic rust disease with an average of 28% of inoculated rosettes producing a systemically diseased shoot the following spring. Other rosettes that emerged near inoculation points in spring were stunted and appeared diseased. To determine whether other rosettes were diseased, a chemiluminescence western slot blot test, applying polyclonal antibodies raised against P. punctiformis antigens, was developed to detect the fungus in roots. Rosettes were inoculated with telia-bearing leaves in the fall in Maryland, USA and Veroia, Greece. Roots of asymptomatic rosettes that emerged adjacent to inoculation points the following spring were tested for the presence of the fungus with the slot blot test. Rosettes that had diseased shoots were recorded. Based on the slot blot tests, 50–60% of the asymptomatic rosettes adjacent to inoculation points were positive for presence of the rust and likely to be systemically diseased. To demonstrate that systemic disease leads to C. arvense decline, C. arvense shoot densities were measured annually at 10 sites, in three countries, that had been inoculated with telia-bearing leaves in the fall between 2008 and 2012. Changes in C. arvense shoot densities over time were calculated. Average reductions in C. arvense density across the 10 sites were 43.1 ± 10.0% at 18 months after inoculation, 63.8 ± 8.0% at 30 months after inoculation, and 80.9 ± 16.5% at 42 months after inoculation, and 72.9 ± 27.2% at 54 months after inoculation; the 54 month reduction was, however, based on only two sites.  相似文献

Terminalia sericea is widely distributed in the African Savannah bushveld. It is one of the indigenous fruit bearing trees put to multiple uses. Research has focused on the phytochemical composition of its root, bark, and leaf extracts that are used in ethnomedicine neglecting the potential of its seed. This study purposed to determine, by chemical analyses, the nutritive value of T. sericea seed. 78.8% of the seed was found to be crude protein (46.2%) and lipid (32.6%). Ash made up 6.90% of the seed mass. Linoleic and oleic acids constituted 68.63% and 14.05%, respectively, of the seed oil. Phosphorus (1121.75 ± 10.39 mg 100 g 1 DM) and glutamic acid (8.07 ± 0.13 g 100 1 DM) constituted the most concentrated mineral and amino acid, respectively. T. sericea seed could be utilized as a protein source in feeds and foods and could also be exploited as a non-conventional plant oil source of oleic acid and linoleic acid.  相似文献

The polymorphism of the mitochondrial gene cytochrome oxidase III was studied in the Mediterranean octopus, Octopus vulgaris Cuvier, 1797. A total of 202 specimens from seven sampling sites were analysed with the aim of elucidating patterns of genetic structure in the central Mediterranean Sea and to give an insight into the phylogeny of the Octopus genus. Phylogenetic analyses showed that individuals from the central Mediterranean belong to the O. vulgaris species whose limits should nevertheless be clarified. Concerning genetic structure, two high-frequency haplotypes were present in all locations. The overall genetic divergence (ΦST = 0.05, P < 0.05) indicated a significant genetic structuring in the study area and an AMOVA highlighted a significant break between western and eastern Mediterranean basins (ΦCT = 0.094, P < 0.05). Possible explanations for the observed patterns of genetic structuring are discussed with reference to their relevance for fisheries management.  相似文献

This study focuses on three factors that affect the survival of the lilac pyralid, Palpita nigropunctalis (Lepidoptera:Crambidae): (1) the effect of leaf toughness on survival rate to clarify the availability of leaves as food, (2) the effect of temperature on immature development to determine the lower thermal threshold, and (3) the effect of temperature on head capsule width to clarify whether head capsule width can be used to discriminate among field-collected larval instars. Larvae could develop on Osmanthus fragrans var. aurantiacus leaves collected in April, but not on leaves collected in June or September which were too tough to eat. More than 80% of the larvae on the leaves of Ligustrum lucidum, Ligustrum japonicum, Ligustrum obtusifolium and Syringa vulgaris completed development, regardless of the collection time. P. nigropunctalis completed development on L. lucidum at temperatures from 15 to 27.5 °C with a photoperiod of either 15 L:9D or 16 L:8D, but not at 30 °C, at which temperature no eggs hatched. The lower thermal threshold and thermal constant for total development from egg to adult were estimated at about 7 °C and 450–460 degree-days. Most of the larvae were 5-instar type larvae (passed through 5 instars) regardless of the temperature, but a few 6-instar type larvae (4 of 355) were noted at temperatures of 22.5 °C and higher. No overlap of the ranges of head capsule widths was detected for the 5-instar type larvae, indicating that head capsule width can be used to discriminate among field-collected larval instars.  相似文献

In higher plants, osmotic adjustment at the various levels of plant organization is partly achieved through accumulation of a range of osmolytes especially LMW organic solutes often termed as osmotic solutes. A metabolite profiling of crude extracts of mature pear leaves of a range of 8 Pyrus genotypes was performed using current HPLC, UPLC and 1H NMR spectroscopy techniques in order to identify such putative compounds. Using as variables the concentrations of 45 identified substances and those of a restricted number of unknowns, all belonging to LMW carbohydrates, polyols, organic acids, amino acids and phenolics on the one hand, and the varieties investigated as individuals on the other, we generated a set of data analyzed further by PCA. Those varieties were discriminated into three clusters respectively comprised of the four Asian varieties, the European variety Williams grafted onto 4 different rootstocks, and the two other European varieties Conference and Angelys. These metabolic phenotypes were shown to rely more on scion genotypes than on rootstocks. High to very high amounts of sorbitol (average content of 363 μmol g?1 DW) associated with low amounts of mannitol and myo-inositol were found in all genotypes as well as in a local ecotype of P. communis where the hexitol accounted for 7.3% DW. Sorbitol actually represented up to 30-40% of the total osmotically active organic solutes accumulated in the set of pear leaves investigated, and it was shown to be significantly more abundant in the variety Williams than in Asian ones (p < 0.01). In contrast, the other well-known compatible solute glycine betaine, barely detectable using 1H NMR spectroscopy or HPLC, occurred in leaves of all pear varieties at weak levels lower than 2 μmol g?1 DW which suggested a minor role in osmotic adjustment. Its amount does not seem to be altered in response to an osmotic upshift applied to detached leaves or to depend on exogenously supplied ABA. For non-sustantiated reasons, these results are in contrast with those showing elsewhere very high accumulation of GB in the Asian genotype Su li. In this study mature leaves of this genotype collected from the same tree in July 2007 and July 2006 were shown to contain respectively 0.59 ± 0.04 and 0.85 ± 0.04 μmol g?1 DW. Other abundant organic substances like arbutine, quinic acid, malic acid, sucrose as well as chlorogenic acid and other quinic acid adducts, might also behave as osmotically active substances. In addition to arbutine, its derivative hydroquinone, chlorogenic acid and structurally related substances might be involved in protective functions against secondary oxidative stresses induced by abiotic and biotic stresses encountered during the growing season.  相似文献

A new variety of soybean, Agakong, has good nutritious values, thus is expected to be grown widely in the near future. To evaluate Agakong as a new soybean host of Riptortus pedestris (Fabricius) (Hemiptera: Alydidae), we assessed the biological attributes of the stink bug fed on Agakong and its field (Fabricius) occurrence in comparison with other varieties, Cheongjakong and Daewonkong. In laboratory studies, the nymphal development time and fecundity of R. pedestris was unaffected by the soybean varieties. However, nymphal mortality of R. pedestris fed on Agakong was significantly lower than those fed on Daewonkong. Furthermore, adult females become significantly larger when they fed on Agakong. R. pedestris preferred Cheongjakong seeds to Daewonkong or Agakong seeds in Petri dish choice experiments. The population dynamics of eggs, nymphs, and adults on the three soybean varieties planted in a 3 × 3 Latin square design were assessed by direct counting from 13 July to 22 September, 2007 at 10 days intervals. R. pedestris was the most abundant stink bug species on all the tested soybean varieties. The occurrence of R. pedestris eggs and nymphs was not significantly different among the three varieties. However, the seasonal density of R. pedestris adults was lower on Agakong than on Cheongjakong. Although higher R. pedestris populations were not observed in the field, the possibility of R. pedestris becoming a pest on this newly developed soybean variety can't be ruled out since some of its biological attributes were positively affected by feeding on Agakong seeds.  相似文献

The objective was to identify legume shrub species for development of agroforestry technologies based on seed and forage (leaves and twigs < 10 mm diameter) yield, and determinants of forage quality. Ten individual plants of Bituminaria bituminosa ‘Ecotypes 1’, B. bituminosa ‘Ecotypes 2’, Medicago citrina, and M. arborea from Spain; Colutea istria and Onobrychis aurantiaca from Syria; C. istria from Jordan; Chamaecytisus mollis from Morocco; and Coronilla glauca from France were randomly selected from plots established in a non-tropical dryland environment in northwest Syria in 2000. Five individual plants of each species were cut back to 0.5 m above ground in March 2004. Coppice regrowths were pruned in December 2004 and April 2005 to determine forage yield and proportion of forage in the total above ground biomass (PEFB). Forage samples were analyzed for concentrations of crude protein (CP), lignin(sa), acid detergent fibre (ADFom), neutral detergent fibre (aNDFom), in vitro organic matter (OM) digestibility (IVOMD), and in vitro 24 h gas production (IVGP24h). Matured seeds were hand harvested from the remaining five plants of each species to estimate seed yield. Forage (21–250 kg DM/ha) and seed (0–200 kg DM/ha) yields; PEFB (0.22–0.96); and concentrations of CP (85–115 g/kg DM), lignin(sa) (14–42 g/kg DM), ADFom (94–170 g/kg DM), aNDFom (122–217 g/kg DM), IVOMD (456–617 g/kg OM), and IVGP24h (27–42 ml 200 mg/DM) varied (P<0.05) among shrub species. The IVOMD and IVGP24h were positively correlated (r = 0.75, P<0.032), whereas IVOMD and IVGP24h were negatively correlated with ADFom, lignin(sa) and aNDFom. In terms of forage and seed yields and determinants of forage quality, C. istria from Jordan, M. arborea, B. bituminosa ‘Ecotype-2’, C. istria and O. aurantiaca have higher potential than C. mollis, C. glauca and B. Bituminosa ‘Ecotype-1’ for the development of agroforestry technologies in non-tropical dry areas.  相似文献

Interspecific C × R hybrid (Coffea congensis × Coffea canephora) in India is cultivated as mixed population with male parent C. canephora as this species is an efficient pollen donor for enhanced yield. But distinction of C × R hybrid from C. canephora in old plantation is difficult due to varying plant sizes of C × R hybrid and often resembles with C. canephora. C × R hybrid cultivated under different agroclimatic conditions show distinct vegetative growth pattern with varying yields. Thus development of DNA marker for identification of C × R hybrid is important for clonal propagation and seed preparation from selective individuals. In this study, two DNA bar coding loci of chloroplast genome (rbcL and matK) of parents, F1 hybrid and its back cross progeny were partially sequenced to identify SNPs as DNA marker for distinction of C × R hybrid from C. canephora. Seven SNPs in the matK gene sequence and three nucleotides in the rbcL gene sequence were identified as DNA markers for the genetic identity of C. congensis. These SNPs were found in F1 and advanced progenies of C × R hybrid due to maternal inheritance. Large number of samples of C × R hybrids with varying morphological features revealed no polymorphism among C × R hybrid and C. congensis. Thus, the SNPs in C. congensis can be used as DNA markers for precise identification of C × R hybrid for production of clones besides tagging the chloroplast inheritance in advanced progenies.  相似文献

A thermogradient apparatus was used to investigate the effect of variable dew temperatures on infection of green foxtail by the indigenous pathogen Pyricularia setariae (Ps) and the exotic pathogens Drechslera gigantea (Dg), and Exserohilum rostratum (Er) from the southern USA that showed bioherbicide potential against several grassy weeds. This device is capable of creating multiple diurnal temperature cycles, mimicking daily temperature fluctuations that occur under field conditions. Seven temperature regimes, i.e., 15/10 °C, 20/5 °C, 20/15 °C, 25/10 °C, 25/20 °C, 30/15 °C, and 30/25 °C (maximum/minimum), were used with temperature cycling from maximum to minimum and then back up to maximum in a 24 h period. Ps and Dg were much more virulent than Er on green foxtail, resulting in higher levels of disease and weed control. Dg was little affected by the dew temperatures in terms of plant infection and was more efficacious than Ps under cooler dew temperatures (15/10 °C and 20/5 °C), causing twice as much disease. This greater amount of disease coincided with higher conidial germination, appressorial formation and infection-hypha frequency by Dg at the lower temperatures. The efficacy of Ps improved as dew temperature increased, accompanied by a higher percentage of germination and more frequent appressorial production. Dg caused severe disease 2 d after inoculation whereas Ps required 4 d to initiate disease symptoms. These observations suggest that Dg is a superior candidate than Ps for green foxtail control on the Canadian prairies.  相似文献

A developed consortium-GR, consisting of Proteus vulgaris NCIM-2027 (PV) and Micrococcus glutamicus NCIM-2168 (MG), completely decolorized an azo dye Scarlet R under static anoxic condition with an average decolorization rate of 16,666 μg h?1; which is much faster than that of the pure cultures (PV, 3571 μg h?1; MG, 2500 μg h?1). Consortium-GR gave best decolorization performance with nearly complete mineralization of Scarlet R (over 90% TOC and COD reduction) within 3 h, much shorter relative to the individual strains. Induction in the riboflavin reductase and NADH–DCIP reductase was observed in the consortium, suggesting the involvement of these enzymes during the fast decolorization process. The FTIR and GC–MS analysis showed that 1,4-benzenediamine was formed during decolorization/degradation of Scarlet R by consortium-GR. Phytotoxicity studies revealed no toxicity of the biodegraded products of Scarlet R by consortium-GR. In addition, consortium-GR applied for mixture of industrial dyes showed 88% decolorization under static condition with significant reduction in TOC (62%) and COD (68%) within 72 h, suggesting potential application of this microbial consortium in bioremediation of dye-containing wastewater.  相似文献

This experiment was conducted to study the effect of high ozone concentrations on two cotton (Gossypium hirsutum L.) cultivars. Two cotton cultivars (Romanos and Allegria) were exposed to control (CF < 4 ppb O3) and 100 ppb O3. Plant exposure to ozone began eight days after emergence and was interrupted one day before removing the leaves, to calculate the leaf area. Plants were exposed to ozone 7 h/day, in closed and controlled-environment chambers, during their illumination with artificial visible light.In comparison to control plants, plants exposed to O3 showed chlorotic and necrotic patches on their leaves, increased stomatal or epidermal cell density and yellowness of cotton fibers. Elevated ozone concentration did not have a significant effect on stomatal width, total leaf thickness and thickness of histological components of leaves. Exposure to ozone concentration reduced non-glandular hair density of main leaf veins, plant height, mainstem internode length, chlorophyll content, net photosynthetic rate, stomatal conductance and length and area of bracts and petals. Elevated ozone treatment reduced the maximum length of staminal tube, anther number, pollen grain germination, leaf area, leaf dry weight, boll number, raw cotton weight, total branch length, dry weight of the mainstem–branches–bracts–carpophylls and of root dry weight. Furthermore, exposure to O3 reduced the seed weight, the lint weight, the yield, the ratio of lint weight to seed weight, the fiber strength, the micronaire, the maturity index and the fiber uniformity index values. This study shows that the exposure to high ozone concentrations mainly affected the rate of photosynthesis, raw cotton weight and strength of cotton fibers.  相似文献

Tryptophanins (TRPs) are low molecular weight, tryptophan-rich, basic proteins found in oat (Avena sativa L.) seeds. Like their counterpart puroindolines (PINs) from wheat (Triticum aestivum L.), TRPs are thought to be involved in flour softness as well as disease resistance against phytopathogenic fungi. PINs are known to be the major components of ‘friabilin’ associated with the surface of water washed starch grains and possess lipid binding properties. Two polyclonal antisera against puroindoline-a (PIN-a), and puroindoline-b (PIN-b) respectively; and a monoclonal antiserum raised against ‘friabilin’ were used as primary antibodies in immunoblotting experiments. All antisera detected immunoreactive polypeptides, with approximate relative masses of 15–16 kDa, in oat, wheat, and barley (Hordeum vulgare L.) seed extracts but not in rice (Oryza sativa L.), maize (Zea mays L.), bean (Phaseolus vulgaris L.), pea (Pisum sativum L.) and lentil (Lens culinaris Medic.) seed extracts. Immunoreactive polypeptides were detected in aqueous ethanol [52% (v/v) ethanol] seed extracts. Both anti-‘friabilin’ monoclonal and anti-PIN-b polyclonal antisera recognized 15 as well as 16 kDa tryptophanins in oat seeds from different cultivars. On the other hand, anti-PIN-a polyclonal antiserum strongly cross-reacted with 16 kDa TRP and weakly with 15 kDa TRP. Tryptophanins were found to be associated with the surface of starch grains in oat endosperm tissue using both fluorescence and confocal laser scanning microscopies with anti-‘friabilin’ monoclonal antiserum. SDS-PAGE and immunoblotting assays revealed a gradual synthesis of TRPs as early as milk stage in developing oat seeds. On the other hand, TRPs tend to undergo degradation during seed germination.  相似文献

The diversity and metal tolerance of endophytic fungi from six dominant plant species in a Pb–Zn mine wasteland in Yunnan, China were investigated. Four hundred and ninety-five endophytic fungi were isolated from 690 tissue segments. The endophytic fungal colonization extent and isolation extent ranged from 59 % to 75 %, and 0.42–0.93, respectively, and a positive correlation was detected between them. Stems harboured more endophytic fungi than leaves in each plant species, and the average colonization extent of stems was 82 %, being significantly higher than that of leaves (47 %) (P  0.001, chi-square test). The fungi were identified to 20 taxa in which Phoma, Alternaria and Peyronellaea were the dominant genera and the relative frequencies of them were 39.6 %, 19.0 % and 20.4 %, respectively. Metal tolerance test showed that 3.6 mM Pb2+ or 11.5 mM Zn2+ exhibited the greatest toxicity to some isolates and they did not grow on the metal-amended media. In contrast, some isolates were growth stimulated in the presence of tested metals. The isolates of Phoma were more sensitive to Zn2+ than the isolates of Alternaria and Peyronellaea. However, the sensitivity of isolates to Pb2+ was not significantly different among Phoma, Alternaria, Peyronellaea and other taxa (P > 0.05, chi-square test). Our results suggested that fungal endophyte colonization in Pb–Zn polluted plants is moderately abundant and some isolates have a marked adaptation to Pb2+ and Zn2+ metals, which has a potential application in phytoremediation in this area.  相似文献

The density of Zostera marina L. seeds in bottom sediments was examined to study the reproductive patterns of the Z. marina population in Ago Bay, Mie Prefecture, central Japan.Seeds and seed coats were numerous in Tategami, where the annual type of Z. marina grows. In contrast, seeds were scarce in Hamajima, where the perennial type of Z. marina grows. Bottom sediment was sampled with sediment cores at Tategami in November 2004 and March 2005 to examine density and depth distribution of the seeds. Seeds were found as deep as 8 cm, but no deeper. On the other hand, empty seed coats were found as deep as 16 cm in both months. In the upper layers of the sediment to a depth of 8 cm, the average number of seed coats was 7960 ± 2997 m−2 in November and 16,318 ± 2922 m−2 in March. Deeper than 8 cm, the number of seed coats gradually decreased owing to decomposition, and none was found below 16 cm. We used the density of reproductive shoots and number of seeds per spadix in Tategami to estimate the fate of seeds and seed coats of the annual type of Z. marina in bottom sediments: out of the 6000 seeds m−2 produced annually, 72% disappears from the stand and 28% is buried locally. The density and distribution of Z. marina seeds are among the most important factors in maintenance and propagation of the annual population at Tategami.  相似文献

Systemic disease of Cirsium arvense caused by Puccinia punctiformis depends on teliospores, from telia that are formed from uredinia, on C. arvense leaves. Uredinia result from infection of the leaves by aeciospores which are one main source of dispersal of the fungus. However, factors governing aeciospore spread, germination, infection, and conversion to uredinia and telia have not been extensively investigated. In this study, effective spread of aeciospores from a source area in a field was fitted to an exponential decline model with a predicted maximum distance of spread of 30 m from the source area to observed uredinia on one leaf of one C. arvense shoot. However, the greatest number of shoots bearing leaves with uredinia/telia was observed within 12 m of the source area, and there were no such shoots observed beyond 17 m from the source area. Aeciospore germination under laboratory conditions was low, with a maximum of about 10%. Temperatures between 18 °C and 25 °C were most favorable for germination with maximum germination at 22 °C. Temperature and dew point data collected from the Frederick, MD airport indicated that optimum temperatures for aeciospore germination occurred in late spring from about May 18 to June 20. Dew conditions during this period were favorable for aeciospore germination. A total of 122 lower leaves, 2 per shoot, on 61 C. arvense shoots were individually inoculated in a dew tent in a greenhouse by painting suspensions of aeciospores onto the leaves. Of these inoculated leaves, 47 produced uredinia within an average of 21.2 ± 6.9 days after inoculation. Uredinia were also produced, in the absence of dew, on 17 non-inoculated leaves of 12 shoots. These leaves were up to 4 leaves above leaves on the same shoots that had been individually and separately inoculated. Results of PCR tests for the presence of the fungus in non-inoculated leaves that were not bearing uredinia, showed that 44 leaves above inoculated leaves on 27 shoots were positive for the presence of the fungus. These leaves were up to 5 leaves above inoculated leaves on the same shoot. Uredinia production and positive PCR results on leaves above inoculated leaves on the same shoot indicated that aeciospore infection was weakly systemic. In other tests in which all leaves of plants were spray-inoculated with aeciospores, uredinia were produced by 10 days after inoculation and converted to telia and sole production of teliospores in about 63 days after inoculation. Successful systemic aeciospore infections in late spring would be expected to result in uredinia production in excess of a 1:1 ratio of aeciospore infections to uredinia and ultimately telia production in late summer. In this manner, systemic aeciospore infections would promote increased density of telia that lead to systemic infections of roots in the fall.  相似文献

Genome analysis of plants indicated majority of putative protease genes that need characterization at enzymatic and molecular level. Proteases execute important role in seed development but knowledge of dipeptidylpeptidases in seeds is limited. A dipeptidylpeptidase-III that cleaves Arg-Arg from Arg-Arg-4mβNA was purified from germinated Mung bean seeds. Screening of cereals and legumes for DPP-III revealed highest activity in Phaseolus vulgaris and Vigna radiata (variety P.9531) seeds germinated for 48 h. Homogenate (10%) was acid precipitated upto pH 4.2 and then subjected to 0–55% ammonium sulphate precipitation followed by successive chromatographies on cation exchanger, gel filtration and anion exchanger. Enzyme purity was confirmed by native polyacrylamide gel electrophoresis and in situ gel assays. DPP-III is a heterodimer of ~60 kDa with two subunits of 32.8 kDa and 27 kDa on SDS-PAGE. DPP-III worked optimally at pH 8.0 and at 37 °C with pH stability between 7.5 to 9.5. Arg-Arg-4mβNA was preferred substrate and none of endopeptidase and monopeptidase substrates were hydrolysed. Inhibitors studies revealed it as thiol protease with involvement of metals at active site. The enzyme might be involved in plant's development by generating/deactivating bioactive peptides. Further studies on functional characterization and molecular structure are in progress in our laboratory.  相似文献

The frog skin host-defense peptide tigerinin-1R (RVCSAIPLPICH.NH2) is insulinotropic both in vitro and in vivo. This study investigates the effects on insulin release and cytotoxicity of changes in cationicity and hydrophobicity produced by selected substitutions of amino acids by l-arginine, l-lysine and l-tryptophan. The [A5W], [L8W] and [I10W] analogs produced a significant (P < 0.01) increase in the rate of insulin release from BRIN-BD11 rat clonal β cells at concentration of 0.01 nM compared with 0.1 nM for tigerinin-1R. The increase in the rate of insulin release produced by a 3 μM concentration of the [S4R], [H12K], and [I10W] analogs from both BRIN-BD11 cells and mouse islets was significantly greater (P < 0.05) than that produced by tigerinin-1R. No peptide stimulated the release of lactate dehydrogenase at concentrations up to 3 μM indicating that plasma membrane integrity had been preserved. [A5W] tigerinin-1R was the only analog tested that showed cytotoxic activity against human erythrocytes (LC50 = 265 ± 16 μM) and inhibited growth of Escherichia coli (MIC = 500 μM) and Staphylococcus aureus (MIC = 250 μM). The circular dichroism spectra of tigerinin-1R and [A5W] tigerinin-1R indicate that the peptides adopt a mixture of β-sheet, random coil and reverse β-turn conformations in 50% trifluoroethanol/water and methanol/water. Administration of [S4R] tigerinin-1R (75 nmol/kg body weight) to high-fat fed mice with insulin resistance significantly (P < 0.05) enhanced insulin release and improved glucose tolerance over a 60 min period following an intraperitoneal glucose load. The study supports the claim that tigerinin-1R shows potential for development into novel therapeutic agents for treatment of type 2 diabetes mellitus.  相似文献
