Discoloring biofilms from Cambodian temples Angkor Wat, Preah Khan, and the Bayon and West Prasat in Angkor Thom contained a microbial community dominated by coccoid cyanobacteria. Molecular analysis identified Chroococcidiopsis as major colonizer, but low similarity values (<95%) suggested a similar genus or species not present in the databases. In only two of the six sites sampled were filamentous cyanobacteria, Microcoleus, Leptolyngbya, and Scytonema, found; the first two detected by sequencing of 16S rRNA gene library clones from samples of a moist green biofilm on internal walls in Preah Khan, where Lyngbya (possibly synonymous with Microcoleus) was seen by direct microscopy as major colonizer. Scytonema was detected also by microscopy on an internal wall in the Bayon. This suggests that filamentous cyanobacteria are more prevalent in internal (high moisture) areas. Heterotrophic bacteria were found in all samples. DNA sequencing of bands from DGGE gels identified Proteobacteria (Stenotrophomonas maltophilia and Methylobacterium radiotolerans) and Firmicutes (Bacillus sp., Bacillus niacini, Bacillus sporothermodurans, Lysinibacillus fusiformis, Paenibacillus sp., Paenibacillus panacisoli, and Paenibacillus zanthoxyli). Some of these bacteria produce organic acids, potentially degrading stone. Actinobacteria, mainly streptomycetes, were present in most samples; algae and fungi were rare. A dark-pigmented filamentous fungus was detected in internal and external Preah Khan samples, while the alga Trentepohlia was found only in samples taken from external, pink-stained stone at Preah Khan. Results show that these microbial biofilms are mature communities whose major constituents are resistant to dehydration and high levels of irradiation and can be involved in deterioration of sandstone. Such analyses are important prerequisites to the application of control strategies.  相似文献

The use of commercial biocides in outdoor environments is increasingly discouraged because of their ecotoxicity, new methods being thus invoked to control patinas of biological origin on the stone cultural heritage. The effects of secondary metabolites (usnic acid, norstictic acid, parietin) produced by saxicolous lichens, natural competitors of rock dwelling microorganisms, were investigated in vitro against microcolonial fungi (MCF: Coniosporium apollinis, Coniosporium perforans, Coniosporium uncinatum, Phaeococcomyces-like sp.), coccoid cyanobacteria (Chroococcus minutus) and green algae (Scenedesmus ecornis) which commonly occur on stonework. An acetone/water 10/90 vol/vol mixture was screened as suitable to solubilise the lichen metabolites and to not affect the bioassay results. Benzalkonium chloride 1% was used as positive control. All the three metabolites (approx. 10−2 mM) inhibited the growth of the assayed MCF species, displaying the same effect of benzalkonium chloride. Chroococcus and Scenedesmus exhibited sensibility to the lichen metabolites when exposed to high incubation temperatures (35 °C), chemicals and temperature synergically yielding percentage decreases of intact cells with red chlorophyll epifluorescence. These findings suggest lichen secondary metabolites as allelopathic agents against rock dwelling microorganisms and as potential natural sources for their control on stone materials in restoration and conservation programmes. In this perspective, the detection of a negligible chromatic alteration (ΔE<0.5) caused by LSM to the white Carrara marble is reported as the first step of the necessary extensive evaluation of the LSM-stone material interactions.  相似文献

microRNAs(miRNAs)have emerged as key components in the eukaryotic gene regulatory network. We and others have previously identified many miRNAs in a unicellular green alga,Chlamydomonas reinhardtii. To investigate whether miRNA-mediated gene regulation is a general mechanism in green algae and how miRNAs have been evolved in the green algal lineage,we examined small RNAs in Volvox carteri,a multicellular species in the same family with Chlamydomonas reinhardtii. We identified 174 miRNAs in Volvox, with many of them being highly enriched in gonidia or somatic cells. The targets of the miRNAs were predicted and many of them were subjected to miRNA-mediated cleavage in vivo, suggesting that miRNAs play regulatory roles in the biology of green algae. Our catalog of miRNAs and their targets provides a resource for further studies on the evolution, biological functions, and genomic properties of miRNAs in green algae.  相似文献

Epilithic and endolithic microorganisms and lichens play an important role in stone biodeterioration. The structural and physiological damage caused by nanosecond pulsed laser of 1064 nm from Nd:YAG laser to Verrucaria nigrescens lichen as well as to endolithic algae and fungi were investigated in the present study. Ultrastructural laser effects on lichen and endolithic microorganisms were study without disturbing the relationship between lichen and lithic substrate by taking lichen-containing rock fragments and processing both together. SEM-BSE, LT-SEM and FM were used to determine cell integrity and ultrastructure, which reflect microorganism viability. Photobiont vitality was determined using a PAM chlorophyll fluorescence technique. The lichen thalli were completely removed by irradiation with 5 ns pulses at a fluence of 2.0 J/cm2 with no stone damage as showed by micro-Raman spectroscopy. The fungal and algal endolithic cells located below were completely destroyed or presented a high plasmolysis degree resulting from heating their microenvironment. The lichen and endolithic mycobiont near the irradiated zone were also damaged. Algal photosynthetic damage prevents fungal survival and lichen viability. This is the first report of laser removal and inactivation of lichen and lithic microorganisms, and thus provide an environmentally friendly and efficient method to control stone biodeterioration.  相似文献

Ocean acidification is expected to alter marine systems, but there is uncertainty about its effects due to the logistical difficulties of testing its large-scale and long-term effects. Responses of biological communities to increases in carbon dioxide can be assessed at CO2 seeps that cause chronic exposure to lower seawater pH over localised areas of seabed. Shifts in macroalgal communities have been described at temperate and tropical pCO2 seeps, but temporal and spatial replication of these observations is needed to strengthen confidence our predictions, especially because very few studies have been replicated between seasons. Here we describe the seawater chemistry and seasonal variability of macroalgal communities at CO2 seeps off Methana (Aegean Sea). Monitoring from 2011 to 2013 showed that seawater pH decreased to levels predicted for the end of this century at the seep site with no confounding gradients in Total Alkalinity, salinity, temperature or wave exposure. Most nutrient levels were similar along the pH gradient; silicate increased significantly with decreasing pH, but it was not limiting for algal growth at all sites. Metal concentrations in seaweed tissues varied between sites but did not consistently increase with pCO2. Our data on the flora are consistent with results from laboratory experiments and observations at Mediterranean CO2 seep sites in that benthic communities decreased in calcifying algal cover and increased in brown algal cover with increasing pCO2. This differs from the typical macroalgal community response to stress, which is a decrease in perennial brown algae and proliferation of opportunistic green algae. Cystoseira corniculata was more abundant in autumn and Sargassum vulgare in spring, whereas the articulated coralline alga Jania rubens was more abundant at reference sites in autumn. Diversity decreased with increasing CO2 regardless of season. Our results show that benthic community responses to ocean acidification are strongly affected by season.  相似文献

Molluscan grazers can have important effects on the abundance, colonization rates, and successional pathways of algal assemblages and the entire intertidal community. In general, early successional algae are more readily consumed than corticated algae and kelps, which usually get established later in the community succession. To generalize, however, the effect of different grazers on algal assemblages must be examined on different coasts and under different scenarios. This information could help us understand the mechanisms of ecosystem processes and situations in which general models do not apply. Along the coast of Chile, humans harvest large keyhole limpets, which seem to be the only invertebrate grazers capable of controlling the dominant corticated alga Mazzaella laminarioides, a canopy-forming species that can cover extensive areas of the mid intertidal zone. In this scenario, where large limpets are harvested, the overall effects of the diverse molluscan assemblage of limpets, chitons and snails on algal succession and on corticated algae in particular are not clear. We conducted a 26-month-long experiment to evaluate the effects of molluscan grazers on mid-intertidal algal succession and to isolate the effects of Chiton granosus, the most conspicuous member of the assemblage at these tidal elevations. At sites heavily impacted by humans the molluscan grazer assemblage had strong negative effects on colonization and abundance of green algae such as ulvoids and Blidingia minima. In doing so, the grazer assemblage had a strong negative indirect effect on the establishments of chironomid fly larvae, which were only observed on green algal mats and rarely on bare rock. No significant effects were detected on epilithic microalgae, and effects on sessile invertebrates were highly variable over space and time. C. granosus also had significant negative effects on green algae but did not account for the total grazing pressure exerted by the guild. Limited foraging excursions (ca. 35 cm) from refuges and moderate site (crevice) fidelity in this species may contribute to the patchiness in green algal distribution observed in the field. Nearly 13 months after rock surface were experimentally cleared, M. laminarioides appeared in all experimental plots, but increased over three times faster in enclosures containing C. granosus than in exclosures plots or controls, suggesting that moderate levels of herbivory could actually facilitate the establishment of this alga in the succession and that the green algal cover found in the absence of grazers may delay its establishment.  相似文献

The biological colonization of mural paintings in rocky habitats is a widely recurring phenomenon and should be considered for restoration activities. In order to plan conservation treatment of the Crypt of the Original Sin (Matera, Italy), biodeterioration problems were analysed taking into account their impact on the substrate and their relationship with environmental factors. The mural paintings showed a phenomenology of alteration varying from brilliant green, dark green, brown, and black duff patinas with powdery aspect, to rosy discoloration. These phenomena were linked to different microbial colonizations by Cyanobacteria (Chlorogloea microcystoides, Chroococcus lithophilus, Gloeocapsa spp., Gloeothece rupestris, Pseudocapsa dubia) and green algae (Apatococcus lobatus, Chlorella vulgaris, Chlorococcum sp., Muriella terrestris). The rosy discoloration was connected to pigment production by Actinobacteria related to Rubrobacter radiotolerans. Grey-green patinas, limited to more ventilated areas, were caused by lichen growth (Dirina massiliensis f. sorediata). Vascular plants (Adiantum capillus-veneris) were also detected in wall areas affected by rising damp. To control the growth of microflora, biocide treatment and intervention to reduce humidity and sunlight exposure of mural paintings were suggested. To choose the correct biocide, different products were tested, evaluating their efficiency on Cyanobacteria and algae as well as possible effects on the calcarenite stone.  相似文献

Microbial processes in a shallow, saline, alkaline Lake Khilganta (Southern Siberia) were studied during the dry season. During the drought, a crust was formed on the lake surface, where low rates of production processes were observed, with predominance of anoxygenic photosynthesis at 2.3 mg C/(dm3 day). The rates of microbial processes increased after short-term rains. During this period, a thin cyanobacterial mat was formed on the bottom, in which filamentous cyanbacteria Geitlerinema spp. predominated and the rate of oxygenic photosynthesis was up to 18 mg C/(dm3 day). Subsequent water evaporation and salinity increase resulted in altered community types and their activity. Red spots emerged on the mat surface, where anoxygenic prototrophic members of the genus Ectothiorhodospira predominated. Anoxygenic photosynthesis became the main production process in microbial mats, with the rate of 60 mg C/(dm3 day). At salinity increase to 200 g/L, the water remained in small depressions on the bottom, where extremophilic green algae Dunaliella sp. predominated, and the rate of oxygenic photosynthesis was 0.877 mg C/(dm3 day). These changes in the type and activity of microbial communities is an example of succession of microbial communities in Southern Siberia saline lakes during drought.

Lichenised fungi are traditionally assumed to form obligate symbioses with algae or cyanobacteria and to be confined to the surface of their growing substratum. However, in a recent 454 pyrosequencing study of fungal communities in Picea abies logs, lichen-forming fungi were detected at a depth of more than 6 cm in dead wood, implying the existence of free-living lichen mycobionts. To determine whether this was the case, we investigated whether Cladonia spp., the most frequently encountered mycobionts, occurred in wood without their photobionts. We detected green algae in all samples with records of Cladonia spp. Hence, we found no evidence for free-living Cladonia mycobionts in wood. We suggest that the detected Cladonia DNA in these logs originates from vegetative propagules or thallus fragments dispersed into the logs by animals or water. However, the occurrence of free-living stages of other lichen-forming fungal taxa in dead wood cannot be excluded.  相似文献

Eight adjacent sub-arctic streams with consistently different temperatures but broadly similar chemistry, other than some differences in conductivity and linked pH, were used to investigate effects of temperature regime on the structure of primary producer communities. Grazing by invertebrates was also taken into account to detect possible effects on the primary producers. The moss species Fontinalis antipyretica Hedw. (Bryophyta) was only observed in the warmer streams and its cover was positively linked with temperature. The liverwort Jungermannia exsertifolia (Steph.) was absent from the warmer streams and scarce in the colder streams. The biomass of F. antipyretica was inversely correlated with concentration of epilithic chlorophyll a, possibly due to shading from the bryophyte and/or competition between algae and bryophytes for nutrients. Epilithic algae and bryophyte biomass were not related to density of the main grazer, Radix peregra Müller (Gastropoda).A high biovolume of green algae (Chlorophyta) occurred in all streams. The biovolume Cyanobacteria was greatest in the coldest stream and in the two intermediately warm streams. Diatom biovolume was low compared with the biovolumes of Cyanobacteria and green algae in all streams, except in the two warmest streams, where the highest biovolume of diatom occurred.Regularized Canonical Correlation Analysis (RCCA) classification based on all primary producer assemblages and invertebrates indicated that warm streams (16.7–22.2 °C) were similar to each other. It also showed that the colder streams (6.6–13.2 °C) were different from the warmer streams and from each other because of differences in conductivity. Temperature and conductivity were the most influential variables in determining overall stream diversity.  相似文献

Foliicolous lichens are formed by diverse, highly specialized fungi that establish themselves and complete their life cycle within the brief duration of their leaf substratum. Over half of these lichen‐forming fungi are members of either the Gomphillaceae or Pilocarpaceae, and associate with Trebouxia‐like green algae whose identities have never been positively determined. We investigated the phylogenetic affinities of these photobionts to better understand their role in lichen establishment on an ephemeral surface. Thallus samples of Gomphillaceae and Pilocarpaceae were collected from foliicolous communities in southwest Florida and processed for sequencing of photobiont marker genes, algal cultivation and/or TEM. Additional specimens from these families and also from Aspidothelium (Thelenellaceae) were collected from a variety of substrates globally. Sequences from rbcL and nuSSU regions were obtained and subjected to Maximum Likelihood and Bayesian analyses. Analysis of 37 rbcL and 7 nuSSU algal sequences placed all photobionts studied within the provisional trebouxiophycean assemblage known as the Watanabea clade. All but three of the sequences showed affinities within Heveochlorella, a genus recently described from tree trunks in East Asia. The photobiont chloroplast showed multiple thylakoid stacks penetrating the pyrenoid centripetally as tubules lined with pyrenoglobuli, similar to the two described species of Heveochlorella. We conclude that Heveochlorella includes algae of potentially major importance as lichen photobionts, particularly within (but not limited to) foliicolous communities in tropical and subtropical regions worldwide. The ease with which they may be cultivated on minimal media suggests their potential to thrive free‐living as well as in lichen symbiosis.  相似文献

A red algal turf is often found just below the barnacle/limpet zone of many European shores, especially on steep shores of moderate exposure. The hypothesis that grazing by limpets determines the upper limit of distribution of this red algal turf was tested on moderately exposed shores in Portugal and Britain. We also aimed to assess whether the grazing effect is modified at various spatial scales. Grazers were excluded by fences, with half-fenced and unfenced controls. Exclusion plots were rapidly colonised by green ephemeral algae in the months immediately after the beginning of the experiment (summer); these algae were later replaced by perennial algae. The percentage cover of turf-forming macroalgae showed a significant increase at both locations. The upper limit of distribution extended more than 50 cm on most of the shores studied. In contrast, control and half-fenced plots remained devoid of algae. After 2 years, ungrazed plots were mainly colonised by a red algal turf (e.g. Caulacanthus ustulatus, Gelidium spp., Laurencia pinnatifida) in Portugal, while canopy cover (Fucus serratus and Himanthalia elongata) dominated in Britain in marked contrast to the grazed plots. Physical factors acting at both local and geographical scales may explain these differences. However, although physical factors probably have an important influence on the identity, size and abundance of sublittoral fringe macroalgae, grazers play a major role in directly setting their upper limits. The effect of grazing by limpets was not consistent for all of the morphological algal groups and spatial scales considered in the present study. The effect of grazing on the cover of turf algae varied between Portugal and Britain (location scale), while effects on ephemeral and canopy algal cover varied at the shore scale within location.  相似文献

The diversity indices of eukaryotic microalgal groups in the Jeonglyeongchi, Waegok, and Wangdeungjae marshes of Mount Jiri, Korea, were measured using Illumina MiSeq and culture-based analyses. Waegok marsh had the highest species richness, with a Chao1 value of 828.00, and the highest levels of species diversity, with Shannon and Simpson index values of 6.36 and 0.94, respectively, while Wangdeungjae marsh had the lowest values at 2.97 and 0.75, respectively. The predominant species in all communities were Phagocata sibirica (Jeonglyeongchi, 68.64%), Aedes albopictus (Waegok, 34.77%), Chaetonotus cf. (Waegok, 24.43%), Eimeria sp. (Wangdeungjae, 26.17%), and Eumonhystera cf. (Wangdeungjae, 22.27%). Relative abundances of the microalgal groups Bacillariophyta (diatoms) and Chlorophyta (green algae) in each marsh were respectively: Jeonglyeongchi 1.38% and 0.49%, Waegok 7.0% and 0.3%, and Wangdeungjae 10.41% and 4.72%. Illumina MiSeq analyses revealed 34 types of diatoms and 13 types of green algae. Only one diatom (Nitzschia dissipata) and five green algae (Neochloris sp., Chlamydomonas sp., Chlorococcum sp., Chlorella vulgaris, Scenedesmus sp.) were identified by a culture-based analysis. Thus, Illumina MiSeq analysis can be considered an efficient tool for analyzing microbial communities. Overall, our results described the environmental factors associated with geographically isolated mountain marshes and their respective microbial and microalgal communities.  相似文献

Phyllosphere algae are common in tropical rainforests, forming visible biofilms or spots on plant leaf surfaces. However, knowledge of phyllosphere algal diversity and the environmental factors that drive that diversity is limited. The aim of this study is to identify the environmental factors that drive phyllosphere algal community composition and diversity in rainforests. For this purpose, we used single molecule real-time sequencing of full-length 18S rDNA to characterize the composition of phyllosphere microalgal communities growing on four host tree species (Ficus tikoua, Caryota mitis, Arenga pinnata, and Musa acuminata) common to three types of forest over four months at the Xishuangbanna Tropical Botanical Garden, Yunnan Province, China. Environmental 18S rDNA sequences revealed that the green algae orders Watanabeales and Trentepohliales were dominant in almost all algal communities and that phyllosphere algal species richness and biomass were lower in planted forest than in primeval and reserve rainforest. In addition, algal community composition differed significantly between planted forest and primeval rainforest. We also found that algal communities were affected by soluble reactive phosphorous, total nitrogen, and ammonium contents. Our findings indicate that algal community structure is significantly related to forest type and host tree species. Furthermore, this study is the first to identify environmental factors that affect phyllosphere algal communities, significantly contributing to future taxonomic research, especially for the green algae orders Watanabeales and Trentepohliales. This research also serves as an important reference for molecular diversity analysis of algae in other specific habitats, such as epiphytic algae and soil algae.  相似文献

Temporal changes of biomass and dominant species in benthic algal communities were investigated in a littoral sand-beach zone in the north basin of Lake Biwa from December 1999 to September 2000. Chlorophyll-a amounts of benthic algal communities per unit area of the sandy sediments rapidly increased from late April to June. Increases in biomass of the benthic algal communities are considered to result from the propagation of filamentous green algae Oedogonium sp. and Spirogyra sp. The cell numbers of filamentous green algae and chlorophyll-a amounts of benthic algal communities at depths of 30 and 50cm at a station protected by a breakwater in May were significantly higher than those of a station exposed directly to wave activity. Thus, the biomass accumulation of the benthic algal communities seems to be regulated strongly by wave disturbance. The development of filamentous green algae may contribute to the increase in biomass of the benthic algal community and to the changes in seasonal patterns of biomass in the sand-beach zone of Lake Biwa. We consider that the development of the filamentous green algal community in the littoral zone of Lake Biwa is the result of eutrophication.  相似文献

The structure of the associated bacterial community of bipartite cyanolichens of the genus Peltigera from three different environmental contexts in the Karukinka Natural Park, Tierra del Fuego, Chile, was assessed. The sampling sites represent different habitat contexts: mature native forest, young native forest and grassland. Recently it has been determined that the bacterial community associated to lichens could be highly structured according to the mycobiont or photobiont identities, to the environmental context and/or to the geographic scale. However, there are some inconsistencies in defining which of these factors would be the most significant on determining the structure of the microbial communities associated with lichens, mainly because most studies compare the bacterial communities between different lichen species and/or with different photobiont types (algae vs. cyanobacteria). In this work bipartite lichens belonging to the same genus (Peltigera) symbiotically associated with cyanobacteria (Nostoc) were analyzed by TRFLP to determine the structure of the bacterial community intimately associated with the lichen thalli and the one present in the substrate where they grow. The results indicate that the bacterial community intimately associated differs from the one of the substrate, being the former more influenced by the environmental context where the lichen grows.  相似文献

Coastal eutrophication may alter the dominance patterns of marine macroalgae, with potential consequences for the associated fauna and the entire ecosystem. Benthic macroalgae and animals in control and nutrient-enriched mesocosms were monitored to investigate eutrophication-induced changes in rocky shore communities. During a 3-year project, nutrient addition had only minor effects on the community structure, such as increased cover and biomass of green Ulva spp. and increased abundance of certain animal species at high nutrient levels. This study is a 4-year extension of a previously reported project, with 2 extra years of effect studies (altogether 5 years) and a subsequent 2 years for recovery. During the 4th year of nutrient enrichment, the cover of Fucus vesiculosus and Fucus serratus started to decline. In the 5th year, these canopy species crashed and there was an evident take-over by green algae at high nutrient addition levels. The previously observed abundance stimulation for fauna disappeared later in the time series, probably due to the loss of the macroalgal canopy. After less than 2 years on regular seawater, the algal and animal communities had returned to within the range of normal variability. The results indicate that established rocky shore communities of perennial algae with associated fauna are able to persist for several years, even at very high nutrient levels, but that community shifts may suddenly occur if eutrophication continues. They also indicate that rocky shore communities have the ability to return rapidly to natural undisturbed conditions after the termination of nutrient enhancement.  相似文献

Abstract Phototrophic microbial communities present in the Roman Catacombs were characterized and different species of terrestrial epilithic cyanobacteria were found to occur as dominant organisms. Eucapsis, Leptolyngbya, Scytonema, and Fischerella were the most frequently encountered cyanobacterial taxa, while a few species of green algae and the diatom Diadesmis gallica occurred in minor amounts. Streptomyces strains, a few genera of eubacteria, and to a lesser extent fungi were always present in the same microhabitats and contributed to the deterioration of stone surfaces. The combined use of light and electron microscopy evidenced the structural relationships among rod-shaped or filamentous bacteria and cyanobacterial cells, as well as the presence of polysaccharide capsules and sheaths, and of mineral precipitates on S. julianum filaments. The significance of the intimate association among the microorganisms was discussed in relation to the damage caused by the growth of biological patinas on stone surfaces.
